背景技术

实施例
实施例1:具有药物活性小分子的N-(2-羟乙基)甲基丙烯酰胺单体的合成

实施例5:聚合物溶液的制备
使用助溶剂蒸发法制备聚合物溶液。将80mg pHPMA-b-pBMA在烧杯中溶于1.0mL乙醇。然后,将1.0mL 1X磷酸盐缓冲盐水(PBS)加入烧杯中。将溶液剧烈搅拌6小时使乙醇蒸发。在1X PBS中获得80mg/mL的聚合物溶液,并将其保存在4℃下。通过动态光散射测量聚合物制剂的尺寸。此实施例说明了在溶液中组装成颗粒的过程。如此实施例中所体现的,可以使用其他溶液和溶液中的添加剂来制备颗粒。如此实施例中所体现的,可获得10-1000nm的粒径。
实施例6:通过口服管饲法施用聚合物溶液以及粪便中SCFA的增加

实施例8:通过口服管饲法施用丁酸钠溶液以及丁酸钠相比于聚合物在降低过敏反应和过敏症的生理指标方面的劣势
在断奶时,每周一次共5周,按照先前报道的和本领域技术人员已知的程序,通过使用6mg花生蛋白+10μg霍乱毒素(“过敏阳性对照”或“过敏”;N=19)的口服管饲法致敏,在小鼠中诱发对花生的过敏。小鼠每天接受每g小鼠体重2.83mg丁酸钠的剂量,其方式是通过用针将溶于磷酸盐缓冲盐水中的丁酸钠口服管饲至“过敏”小鼠的胃中。此剂量等效于聚合物以每g小鼠体重施用0.8mg聚合物的剂量。每周对小鼠称重一次,并根据新的重量调整溶液量。第5次致敏后1周时(实验第35天),通过腹膜内注射给予小鼠5mg花生蛋白。在注射后90分钟内通过直肠监测核心体温,并在注射后90分钟和24小时内收集小鼠血清。处理血清用于ELISA,并测定花生特异性免疫球蛋白E(IgE)、花生特异性免疫球蛋白G1(IgG1)和小鼠肥大细胞蛋白酶1(mMCP-1)。这些结果说明,当每天给药时,以与等效嵌段共聚物相同的剂量口服施用丁酸钠对降低过敏反应的生理指标和症状没有效果(图17)。
实施例9:通过口服管饲法施用不同的聚合物制剂不能在降低过敏反应和过敏症的生理指标方面产生等效效力
使用实施例3和5中所述的方法制备两种聚合物制剂。通过动态光散射对一种聚合物进行表征并显示其在共聚物中具有的HPMA:BMA嵌段比率为1.01,粒径为329nm(“制剂2”)。通过动态光散射对另一种聚合物进行表征并显示其在共聚物中具有的HPMA:BMA嵌段比率为0.75,粒径为64.6nm(“制剂1”)(图18)。
在断奶时,每周一次共5周,按照先前报道的和本领域技术人员已知的程序,通过使用6mg花生蛋白+10μg霍乱毒素(“过敏阳性对照”或“过敏”)或单独10μg霍乱毒素(“非过敏阴性对照”;N=4)的口服管饲法致敏,在小鼠中诱发对花生的过敏。如实施例3中那样制备80mg/mL的制剂2聚合物的储备溶液,然后稀释至53.3mg/mL。每天通过口服管饲法用针将每g小鼠体重0.8mg聚合物施用至“过敏”小鼠的胃中。另一组“过敏”小鼠接受与实施例7相同的2.83mg丁酸钠。另一组“过敏”小鼠接受每g小鼠体重0.8mg每日剂量的对照聚合物,其不释放丁酸。对于未接受任何剂量聚合物的“过敏”小鼠,使用体积匹配的1X PBS溶液作为对照。对于“非过敏阴性对照”小鼠,每天给药每g小鼠体重0.8mg的对照聚合物作为对照。第5次致敏后1周时(实验第35天),通过腹膜内注射给予小鼠5mg花生蛋白。在注射后90分钟内通过直肠监测核心体温,并在注射后90分钟和24小时内收集小鼠血清(图19)。处理血清用于ELISA,并测定花生特异性免疫球蛋白E(IgE)、花生特异性免疫球蛋白G1(IgG1)和小鼠肥大细胞蛋白酶1(mMCP-1)。这些结果说明,粪便中丁酸盐的增加不足以预测功效,并且聚合物的分子和形态特征不会明显导致治疗效果的差异。具体地,制剂2仅在分子量和嵌段比率方面与制剂1不同,在每天给药时制剂2不会降低过敏反应的生理指标和症状。
实施例10:通过口服管饲法施用聚合物制剂导致粪便丁酸盐浓度的等效变化,但不能在降低过敏反应和过敏症的生理指标方面产生等效效力。等效效力要求严格控制某些聚合物分子特征。
使用实施例3和5中所述的方法制备三种聚合物制剂(图20)。通过动态光散射对一种聚合物进行表征并显示其在共聚物中具有的HPMA:BMA嵌段比率为0.75,分子量为约40,000g/mol,粒径为60-70nm(“制剂1”)。通过动态光散射对第二种聚合物进行表征并显示其在共聚物中具有的HPMA:BMA嵌段比率为0.71,分子量为约52,000g/mol,粒径为120-130nm(“制剂3”)。通过动态光散射对第三种聚合物进行表征并显示其在共聚物中具有的HPMA:BMA嵌段比率为0.77,分子量为约39,000g/mol,粒径为75-82nm(“制剂4”)。
通过GPC、DLS和NMR对这三种制剂的分析如图21-24所示。
通过胃内管饲法以单次剂量口服施用将8mg的制剂1和制剂3施用于各组小鼠(每组N=8),并在24小时内收集合并来自每只小鼠的粪便。将样品进行气相色谱处理,并使用实施例6中所述的方法确定游离丁酸盐的浓度。确定游离丁酸盐与理论释放丁酸盐的比率,并将其转化为释放百分比(图26)。
使用实施例5和7中所述的诱发过敏模型程序比较这三种制剂(图27)。通过腹膜内注射给予小鼠5mg花生蛋白后,在注射后90分钟内通过直肠监测核心体温,并在注射后90分钟和24小时内收集小鼠血清。处理血清用于ELISA,并测定花生特异性免疫球蛋白E(IgE)、花生特异性免疫球蛋白G1(IgG1)和小鼠肥大细胞蛋白酶1(mMCP-1)。这些结果说明,是分子量差异而不是嵌段比率差异导致治疗效果差异。具体地,制剂2与制剂1和3的嵌段比率没有明显不同,但是分子量和形态确实不同,当每天给药时,制剂2不会降低过敏反应的生理指标和症状。制剂1和3只是在分子和形态特征上略有不同,但当每天给药时,确实降低了过敏反应的生理指标和症状。

Claims (39)
