Abstract
本发明公开了一种具有广谱活性氧清除功能的微球及其制备方法。其制备方法是将过氧化氢酶和牛血清白蛋白一起溶解到氯化锰溶液中,在搅拌的条件下,迅速加入碳酸钠溶液,得到同时包埋过氧化氢酶(CAT)和牛血清白蛋白(BSA)的碳酸锰粒子,在粒子表面沉积聚多巴胺壳层并组装聚电解质多层膜,通过戊二醛交联稳定粒子结构,加入乙二胺四乙酸钠溶液去除碳酸锰模板,从而得到具有广谱活性氧清除功能的微球,该微球可以利用过氧化氢酶清除双氧水,利用聚多巴胺壳层清除活性氧自由基,从而拥有广谱活性氧清除的性能。本发明制备方法简单,材料来源广泛,生产效率高,得到的具有广谱活性氧清除功能的微球具有良好的稳定性和较高活性,具有良好应用前景。
Description
技术领域
本发明属于抗炎微纳米材料制备领域,涉及一种具有广谱活性氧清除功能的微球及其制备方法。
背景技术
炎症是免疫系统对损伤和感染的自然反应,其可导致炎性物质的释放,如:细胞因子,自由基,激素和其他小分子等,从而有利于保护人体免受这些病理性畸变。然而,有流行病学和临床证据表明过度炎症是有害的并且与许多病理性功能障碍相关,例如肝炎,类风湿性关节炎,糖尿病,以及神经退行性疾病和心血管疾病。现在已经阐明,异常的活性氧产生是炎症发病机制中的关键介质之一,体内过量的活性氧会导致各种炎症相关的病理学异常。抗炎治疗剂已经被广泛开发了数十年,其中清除过量ROS以中断异常炎症反应已被认为是炎症抑制的可行策略。
在炎症部位中,往往存在多种活性氧,如:双氧水、羟基自由基、超氧阴离子自由基等。然而,当前的天然酶和人工纳米酶虽然对某些特定的活性氧有高效的清除能力,但是对疾病过程中产生的多种活性氧的抗氧化作用不足。因此急需开发一种具有广谱活性氧清除能力的材料。
发明内容
针对现有技术中存在的缺陷,本发明提供了一种具有广谱活性氧清除功能的微球及其制备方法。本发明的具有抗炎功能的广谱活性氧清除的微球清除活性氧效果良好,能够减轻体内由活性氧引起的炎症反应,原料细胞的毒性小,有良好的应用前景。
本发明采用以下技术方案实现:
一种具有广谱活性氧清除功能的微球的制备方法,包括以下步骤:
1)配制等浓度的氯化锰溶液和碳酸钠溶液,将过氧化氢酶(CAT)和牛血清白蛋白(BSA)溶于氯化锰溶液中,最终氯化锰溶液中CAT和BSA的总浓度为1~10 mg/mL,CAT和BSA的质量比为4:1~1:4;在高速搅拌下,迅速加入与氯化锰溶液等体积的碳酸钠溶液,30s~2 min后停止搅拌,从而得到包埋有CAT和BSA的碳酸锰粒子,离心去除上清液,并用去离子水洗涤粒子多次;
2)取步骤1)所得的碳酸锰粒子分散于去离子水中制成浓度为1~10 mg/mL的悬浮液,离心去除上清液,加入含有1~3 mg/mL多巴胺的Tris缓冲液,震荡10 min~24 h,离心去除上清液,加入去离子水洗涤粒子多次;然后加入浓度为1~2 mg/mL的聚阳离子溶液,震荡10~20 min,离心去除上清液,用去离子水洗涤粒子多次;再加入浓度为1~2 mg/mL的聚阴离子溶液,震荡10~20 min,离心去除上清液,用去离子水洗涤粒子多次,得到表面为聚阴离子的粒子;
3)将步骤2)所得的粒子分散到质量浓度为0.025~0.05%的戊二醛溶液中,震荡至少1h,以去除步骤3)中残留的戊二醛,然后离心去除上清液,并用去离子水洗涤多次;
4)将步骤3)所得的粒子分散到1~3 mg/mL的甘氨酸溶液中,震荡至少1 h,离心去除上清液,并用去离子水洗涤多次;
5)将步骤4)所得的粒子分散到0.1~0.2 mol/L乙二胺四乙酸钠溶液中,震荡20~30min,离心去除上清液,并用EDTA溶液洗涤多次,再用去离子水洗涤多次,去除碳酸锰模板得到具有广谱活性氧清除功能的微球。
上述技术方案中,进一步地,步骤1)中所述的搅拌条件的转速为600-1000 rpm。
进一步地,步骤2)中所述的Tris缓冲液为10~25 mmol/L、pH 8~9的Tris缓冲液。
进一步地,步骤2)中所述的聚阳离子是为聚烯丙基胺盐酸盐、聚二烯丙基二甲基季铵盐、壳聚糖、聚赖氨酸或聚乙烯亚胺。
进一步地,步骤2)中所述的聚阴离子是为聚苯乙烯磺酸钠、聚丙烯酸、硫酸肝素、透明质酸或硫酸葡聚糖。
进一步地,步骤2)中所述的碳酸锰粒子悬浮液、多巴胺溶液、聚阳离子溶液及聚阴离子溶液的体积相等。
本发明还提供一种具有广谱活性氧清除功能的微球,所述的微球采用上述方法制备得到。
本发明的原理是:
本发明通过模板法和表面组装将过氧化氢酶、多巴胺和聚电解质相结合构建具有广谱活性氧清除能力的微球。微球表面修改聚电解质可以增强微球在生理条件下的稳定性并对内部的酶起到保护作用。其中,多巴胺是一种天然多酚,其酚羟基具有优异的还原性,可以与多种活性氧反应。并且多巴胺在碱性条件下会很快发生自聚现象,可以在各种基质表面沉积聚多巴胺壳层。自聚后聚多巴胺中部分酚羟基被保留,所以聚多巴胺仍可以清除多种活性氧。过氧化氢酶是一种天然酶,广泛存在于动植物体内,可以将双氧水快速分解为水和氧气,维持体内的双氧水平衡。
某些疾病和炎症部位会存在比正常部位更高浓度的活性氧,如双氧水、羟基自由基、超氧阴离子自由基等。过氧化氢酶可以将双氧水分解成水和氧气,多巴胺在碱性条件下自聚形成聚多巴胺沉积在微球表面,多巴胺中的部分酚羟基在自聚后仍然被保留下来,并且其酚羟基具有优秀的还原性,可以清除多种活性氧。通过模板法和表面组装将过氧化氢酶、聚多巴胺和聚电解质结合制备得到在生理条件下稳定分散且具有广谱活性氧清除功能的微球。
本发明的有益效果在于:
原料来源广泛,制备过程简单可控,可以放大规模;选择不同的聚电解质组装到微球表面可以调控微球的表面性质;具有广谱活性氧清除功能,对多种活性氧均有良好的清除效果;将自由的酶固定,可以长时间保持酶活性,能够在4℃长久保存。

具体实施方式
以下结合实例进一步说明本发明,但这些实例并不用来限制本发明。
实施例1
1)配制0.25 mol/L氯化锰溶液和0.25 mol/L碳酸钠溶液,将CAT和BSA溶于2 mL氯化锰溶液中,CAT与BSA的质量比为1:2,二者在氯化锰溶液中的总浓度为7.5 mg/mL。在1000rpm的搅拌下,迅速加入2 mL的碳酸钠溶液,1 min后停止搅拌,从而得到同时包埋有CAT和BSA的碳酸锰粒子,离心去除上清,并用水洗涤3次。
2)将步骤1)所得的碳酸锰粒子分散于4 ml去离子水中,加入8 mL含有3 mg/mL多巴胺的Tris缓冲液(25 mmol/L,pH8.35),震荡10 min,离心去除上清,加入去离子水洗涤粒子3次;分散到8 ml去离子水中,加入4 ml浓度为6 mg/ml聚烯丙基胺盐酸盐(PAH)溶液,震荡10 min,离心去除上清,加入去离子水洗涤粒子3次;分散到8 ml去离子水中,加入4 ml浓度为6 mg/ml硫酸葡聚糖(DS)溶液,震荡10 min,离心去除上清,加入去离子水洗涤粒子3次,得到表面为DS的粒子。
3)将步骤2)所得的粒子分散到10 ml 0.025%的戊二醛溶液中,震荡1 h,离心去除上清,并用去离子水洗涤3次。
4)将步骤3)所得的粒子分散到10 mL 1 mg/mL的甘氨酸溶液中,震荡1 h,离心去除上清,并用去离子水洗涤3次。
5)将步骤4)所得的粒子分散到10 mL 0.2 mol/L乙二胺四乙酸钠溶液中,震荡30min,离心去除上清,并用乙二胺四乙酸钠溶液洗涤2次,离心去除上清,并用去离子水洗涤3次,最终得到具有广谱活性氧清除能力的微球。
实施例2
步骤同实施例1,但在步骤2)中用聚乙烯亚胺代替PAH,用聚丙烯酸代替DS。最终得到具有广谱活性氧清除能力的微球。
实施例3
步骤同实施例1,但在步骤2)中用聚赖氨酸代替PAH,用透明质酸代替DS。最终得到具有广谱活性氧清除能力的微球。
图1a)是包埋有过氧化氢酶和牛血清白蛋白的具有广谱活性氧清除功能微球的扫描电镜照片,b)是包埋有过氧化氢酶和牛白蛋白的碳酸锰粒子的透射电镜照片,c)是去除碳酸锰模板后得到的具有广谱活性氧清除功能微球的透射电镜照片。从图中可知,在组装完成后粒子在水溶液中分散性良好,表面粗糙。从透射电镜结果可观察到除去碳酸锰模板后,微球呈现类似于核壳结构,微球的核为过氧化氢酶和牛血清白蛋白,壳为沉积的聚多巴胺和聚电解质。
制备微球过程中表面电位变化见图2,包埋过氧化氢酶和牛血清白蛋白后的碳酸锰粒子表面为负电,在表面沉积聚多巴胺后微粒的电负性增加,接着在表面分别组装聚阳离子PAH和聚阴离子DS,表面电位分别由负变正和由正变负。可以证明聚多巴胺、PAH、DS已经被成功组装。
微球清除双氧水能力检测见图3。图3a)是包埋和未包埋过氧化氢酶的微球与2mmol/L双氧水反应后的紫外吸收光谱。微球浓度为100 μg/ml,双氧水浓度2mmol/L,溶液pH7.4,反应时间5 min。240 nm处的紫外吸收是双氧水特征吸收值,从图中可知,包埋了过氧化氢酶的微球在5 min反应时间内可以将2 mM的双氧水全部清除,但是未包埋过氧化氢酶的微球在5 min反应后未能使240 nm处吸收值降低,证明过氧化氢酶已经被成功包埋,并且制得的微球仍能有效清除双氧水。图3b)是不同浓度的微球与溶液中被清除双氧水占初始双氧水总量比例的关系。其中,初始双氧水总浓度为100 μmol/L,微球浓度为5、10、20、50、100 μg/ml,溶液pH 7.4,微球与双氧水反应时间为30 min。从图中可知,在30 min反应时间内,随着微球浓度增加清除的双氧水呈线性增加,浓度从50增加到100 μg/ml后,清除双氧水达到饱和,微球表现出优秀的双氧水清除能力。
利用一种稳定的自由基ABTS·检测微球清除活性氧自由基的能力见图4。图4a)是沉积和未沉积聚多巴胺微球清除ABTS·情况。其中,沉积聚多巴胺时间0 min~24 h,ABTS·浓度为0.42 mmol/L,溶液pH 7.4。从图中可知,沉积聚多巴胺时间为0 min的微球几乎不能清除ABTS·,沉积时间为10 min~24 h的微球清除ABTS·的能力相仿。故可证明微球表面成功沉积了聚多巴胺,并且表现出良好的ABTS·清除能力。图4b)是不同浓度微球清除ABTS·。其中,微球浓度为5、10、20、50、100 µg/mL,ABTS·浓度为0.42 mmol/L,溶液pH7.4,反应时间为10 min。从图中可知,在10 min的反应时间内,随着微球浓度增加,被清除的ABTS·也增加,微球表现出良好的ABTS·清除能力。
利用芬顿反应产生羟基自由基,用ABTS来捕获羟基自由基来定性检测不同浓度的沉积和未沉积聚多巴胺微球清除羟基自由基能力见图5。不同浓度沉积聚多巴胺的微球作为实验组,不同浓度未沉积聚多巴胺的微球作为对照组,微球浓度依次为10、20、50、100 μg/ml,Fe3+浓度为1 μg/ml,H2O2浓度为100 μmol/L,溶液pH 4,反应时间30 min。30 min后将微球过滤去除,测量溶液在415 nm吸收峰。从图中可知,对同浓度的微球,沉积聚多巴胺微球与未沉积相比,反应后的溶液在415 nm处吸收值更低,即溶液中的羟基自由基更少,可证明微球沉积聚多巴胺后能够高效清除羟基自由基。
用超氧阴离子试剂盒检测微球清除超氧阴离子自由基能力见图6。图6a)是沉积和未沉积聚多巴胺微球清除超氧阴离子自由基。其中沉积聚多巴胺时间为0 min~24 h。从图中可知,沉积聚多巴胺时间为0 min的微球几乎不能清除超氧阴离子自由基,沉积时间为10min~24 h的微球清除超氧阴离子自由基的能力远优于0 min。证明表面沉积聚多巴胺微球表现出良好的超氧阴离子自由基清除能力。图6b)是不同浓度微球清除超氧阴离子自由基。其中,微粒浓度为10、50、100、350、700 μg/ml,37℃水浴反应30 min,gress显色剂室温显色10 min。从图中可知,随微粒浓度升高,溶液的吸收值降低,即被清除的超氧阴离子自由基增加,证明沉积聚多巴胺微球能够清除超氧阴离子自由基。
从图3、4、5、6中可以看出,本发明方法制得的微球能够高效清除多种活性氧,包括双氧水、羟基自由基、超氧阴离子自基,并用ABTS·代替其他活性氧自由基进行检测,同样具有高效清除能力,可以证明微球具有广谱的活性氧清除功能。
检测该微球在Raw264.7细胞中清除活性氧能力见图7,将微球与细胞共培养6 h后,用200 μmol/L的双氧水处理30 min作为实验组。只用培养基培养细胞作为空白对照组,只加微球与细胞共培养而不用双氧水处理作为阴性对照组,不加微球与细胞共培养只用双氧水处理作为阳性对照组,所有组再用活性氧探针(DCFH-DA)检测细胞内活性氧。其中,培养基中微球浓度为16 μg/ml,活性氧探针(DCFH-DA)用不加血清的培养基1:1000稀释,避光处理细胞30 min。Raw264.7在96孔板中10000/孔。从图中可知,双氧水刺激细胞30 min再用活性氧探针检测细胞内活性氧水平,可以看到细胞呈现出明显的绿色荧光,即双氧水刺激细胞30 min可以使细胞内的活性氧水平上升,若先加入微球与细胞共培养6 h再加入双氧水刺激,细胞内并不会产生明显的活性氧水平上升,证明在细胞层面上,微球可以有效降低细胞内的活性氧水平。
Claims (3)
1.一种具有广谱活性氧清除功能的微球的制备方法,其特征在于,包括以下步骤:
1)配制相同浓度的氯化锰溶液和碳酸钠溶液,将过氧化氢酶和牛血清白蛋白溶于氯化锰溶液中,使氯化锰溶液中过氧化氢酶和牛血清白蛋白的总浓度为1~10 mg/mL,且过氧化氢酶和牛血清白蛋白的质量比为4:1到1:4;在搅拌条件下,迅速加入与氯化锰溶液等体积的碳酸钠溶液,30 s~2 min后停止搅拌,从而得到包埋有过氧化氢酶和牛血清白蛋白的碳酸锰粒子,离心去除上清液,并用去离子水洗涤粒子;
2)取步骤1)所得的碳酸锰粒子分散于去离子水中制成浓度为1~10 mg/mL的悬浮液,离心去除上清液,加入含有1~3 mg/mL多巴胺的Tris缓冲液,震荡10 min~24 h,离心去除上清液,加入去离子水洗涤粒子;然后加入浓度为1~2 mg/mL的聚阳离子溶液,震荡10~20 min,离心去除上清液,用去离子水洗涤粒子;再加入浓度为1~2 mg/mL的聚阴离子溶液,震荡10~20 min,离心去除上清液,用去离子水洗涤粒子,得到表面为聚阴离子的粒子;
3)将步骤2)所得的粒子分散到质量浓度为0.025~0.05%的戊二醛溶液中,震荡至少1h,离心去除上清液,并用去离子水洗涤;
4)将步骤3)所得的粒子分散到1~3 mg/mL的甘氨酸溶液中,震荡至少1 h,离心去除上清液,并用去离子水洗涤;
5)将步骤4)所得的粒子分散到0.1~0.2 mol/L乙二胺四乙酸钠溶液中,震荡20~30min,离心去除上清液,先用EDTA溶液洗涤,再用去离子水洗涤,得到具有广谱活性氧清除功能的微球;
步骤2)中所述的Tris缓冲液为10~25 mmol/L、pH 8~9的Tris缓冲液;
步骤2)中所述的聚阳离子是为聚烯丙基胺盐酸盐、聚赖氨酸或聚乙烯亚胺;
步骤2)中所述的聚阴离子是为聚丙烯酸、透明质酸或硫酸葡聚糖;
步骤2)中所述的碳酸锰粒子悬浮液、多巴胺溶液、聚阳离子溶液及聚阴离子溶液的体积相等。
2.根据权利要求1所述的一种具有广谱活性氧清除功能的微球的制备方法,其特征在于,步骤1)中所述的搅拌条件的转速为600-1000 rpm。
3.一种具有广谱活性氧清除功能的微球,其特征在于,采用如权利要求1-2任一项所述的方法制备而成。
