背景技术
视疲劳又称眼疲劳,是目前眼科常见的一种疾病,以病人自觉眼的症状为基础,眼或全身器质性因素与精神(心理)因素相互交织的综合征,又称眼疲劳综合征,如不及时防治,会造成近视。中医认为,“五脏六腑之精皆上注于目而为之睛”,“目得血而能视”,视疲劳与肝肾不足、肝郁气滞、气血亏虚等因素有关,治疗上采用滋肝补肾、益气养血、疏肝理气为主。目前已有很多相关中成药或保健食品问世,但是大多生产工艺简单、剂型落后。
药材粉碎是中药保健食品制备中的关键工艺,与煎煮等提取工艺相比,它能充分保留活性成分,确保药效,同时还能减少药材损失,提高产品收率。然而,大多数中药材质地坚硬,富含纤维、油脂、糖类、蛋白等物质,具有较高的硬度、韧性和粘性,很难通过粉碎一次成型。在实际生产中,往往需要使用大功率粉碎机,并且进行重复多次粉碎,才能使粒径达到要求,从而提高了粉碎周期、能耗和成本,同时还增加了物料损失。
膨化技术在食品领域已得到广泛应用,它是将谷物、豆类、薯类、海带等原料置于膨化机中进行加热、加压处理,随着温度、压强的升高,物料中的水分呈过热状态,而物料本身变得柔软,此时再由高温高压瞬间降至常温常压,物料内部过热状态的水分子急剧汽化,巨大的膨化张力改变了物料的内部组织结构和外部形态,使其变得膨胀疏松,并实现干燥目的。膨化后的食品可使得蛋白变性、淀粉熟化,从而易于消化。
目前,已有将膨化技术用于中药材加工的报导,但由于绝大多数中药材经高温膨化处理后会对其活性成分产生影响,因此该技术在中药保健食品领域尚未大范围推广。
另外,冷冻干燥也是中药材加工中的一种常见工艺,与普通的加热干燥相比,它具有更好的保鲜作用,能够减少易挥发、热敏性成分的损失,但很少有将其应用于粉碎物料的前处理,本发明针对桑葚、枸杞子药材中果胶、多糖、淀粉成分含量高,粉碎时产生的热量容易导致药材粘性增加,从而使粉碎难度加大的技术难题,引入冷冻干燥对其进行前处理,从而减少粘性成分发生变性,并使其能更容易地在低温状态下实现粉碎。

1)将葛根、菊花先粉碎成粗粉,然后用冷水浸泡1~5小时(目的是使药材组织吸收水分,受热膨化时水分在从药材组织向外汽化的过程中留下更多的通道,这些通道将有利于提高药物崩解速度和活性成分向外部渗出),取出后沥干表面水分,再置于膨化机中进行高温膨化处理,接着将膨化处理后的上述药材粉碎成细粉;
2)将桑葚、枸杞子粉碎成粗粉,然后在-30℃以下预冻3~5小时,接着在真空条件下冷冻干燥至含水量低于5%,最后在液氮条件下(使药材中的果胶、多糖等成分被冻结,防止其温度升高导致粘性增加,同时使药材脆性增加而更有利于粉碎)研磨成细粉;
3)将步骤1)和2)中的细粉混合,然后制成颗粒剂或片剂或胶囊剂。
优选地,所述膨化处理的温度为150~250℃,压强为2~6倍标准大气压,时间为5~30分钟。膨化温度越高、压强越大、时间越长,则膨化效果越好,但是温度过高、时间过长会对活性成分的稳定产生不利影响,甚至还会导致药材中的大量淀粉被糊化以及组织细胞碳化,从而影响药效的发挥。本发明进一步优选地,所述膨化处理的物料温度为180℃,压强为3倍标准大气压,时间为15分钟。
优选地,所述浸泡时间为3小时。
优选地,所述冷冻干燥的真空度为5~30pa,温度为-10~20℃,时间为15-30小时。不同于普通干燥的是,药材经低温预冻,组织内的水分形成冰晶,在真空条件下升温后,冰晶不经过液态而是直接升华成气态,从而使水分在向外部渗出的过程中也会在组织内部形成大量孔洞。
进一步优选地,所述冷冻干燥的真空度为15pa,温度为5℃,时间为20小时。
优选地,所述细粉的粒径为100-180目。
优选地,所述中药材的重量份数为:葛根10份、枸杞子10份、菊花6份、桑葚6份。
与现有技术相比,本发明具有以下有益效果:
(1)本发明通过对葛根、菊花进行高温膨化预处理,使其组织结构变得膨胀疏松,降低了药材的硬度和韧性,从而减少了粉碎次数,降低了能耗和成本,并提高了粉碎回收率。
(2)本发明通过对桑葚、枸杞子进行真空冷冻干燥预处理,降低了药材的粘性,防止药材中果胶、多糖、淀粉类成分变性,实现了药材的低温粉碎。
(3)本发明通过以上预处理,不仅提高了生产效率,同时还意外地发现,所生产的药物中活性成分含量更高,药物崩解、溶出速度更快,能更好地发挥疗效。
具体实施方式
下面结合具体实施例对本发明进行详细说明。
实施例1
处方:
葛根100g、菊花60g、桑葚60g、枸杞子100g
葛根:甘、辛,凉。归脾、胃、肺经。既能升脾胃清阳之气以明目,又能补气生津,以润泽目珠,是调补脾胃,升清明目的代表中药。菊花:甘、苦,微寒。归肺、肝经。既能散风热、清肝火、平肝阳,益肝阴,而且有明目之功,是调理肝火上攻,肝阴不足,肝阳上亢,目失所养,视觉疲劳的佳品。桑葚:归心、肝、肾经。具有补血滋阴、生津润燥之功和护眼明目之效。枸杞子:枸杞性甘、平,归肝肾经,具有滋补肝肾,养肝明目的功效,自古就是滋补养人的上品。
制法:
1)将葛根、菊花先粉碎成粗粉(粉末能全部通过5目筛网),然后用冷水浸泡3小时,取出后沥干表面水分,再置于膨化机中进行高温膨化处理,当膨化机内的温度上升到180℃,压强上升到3倍标准大气压后保持15分钟,然后停止加热并迅速泄压,接着将膨化处理后的上述药材粉碎成150目细粉;
2)将桑葚、枸杞子粉碎成粗粉(粉末能全部通过5目筛网),然后在-50℃预冻3小时,接着在15pa真空条件下冷冻干燥至含水量低于5%(重量),控制冷冻干燥的温度为5℃,时间为20小时,最后通入液氮并在球磨粉碎机中研磨成150目细粉;
3)将步骤1)和2)中的细粉混合,然后加入糖浆,搅拌制软材,制粒,干燥,压制成0.5g/片。
经检测,片剂中葛根素含量为1.22%,总黄酮含量为5.67%。
实施例2
葛根100g、菊花60g、桑葚60g、枸杞子100g
1)将葛根、菊花先粉碎成粗粉,然后用冷水浸泡5小时,取出后沥干表面水分,再置于膨化机中进行高温膨化处理,当膨化机内的温度上升到250℃,压强上升到5倍标准大气压后保持5分钟,然后停止加热并迅速泄压,接着将膨化处理后的上述药材粉碎成180目细粉;
2)将桑葚、枸杞子粉碎成粗粉,然后在-30℃预冻5小时,接着在30pa真空条件下冷冻干燥至含水量低于5%,控制冷冻干燥的温度为20℃,时间为15小时,最后通入液氮并在球磨粉碎机中研磨成180目细粉;
3)将步骤1)和2)中的细粉混合,然后加入糖浆,搅拌制软材,制粒,干燥,压制成0.5g/片。
经检测,片剂中葛根素含量为1.15%,总黄酮含量为5.53%。
实施例3
葛根100g、菊花60g、桑葚60g、枸杞子100g
1)将葛根、菊花先粉碎成粗粉,然后用冷水浸泡1小时,取出后沥干表面水分,再置于膨化机中进行高温膨化处理,当膨化机内的温度上升到150℃,压强上升到2倍标准大气压后保持30分钟,然后停止加热并迅速泄压,接着将膨化处理后的上述药材粉碎成100目细粉;
2)将桑葚、枸杞子粉碎成粗粉,然后在-40℃预冻4小时,接着在5pa真空条件下冷冻干燥至含水量低于5%,控制冷冻干燥的温度为-10℃,时间为30小时,最后通入液氮并在球磨粉碎机中研磨成100目细粉;
3)将步骤1)和2)中的细粉混合,然后加入糖浆,搅拌制软材,制粒,干燥,压制成0.5g/片。
经检测,片剂中葛根素含量为1.23%,总黄酮含量为5.74%。
对比例
葛根100g、菊花60g、桑葚60g、枸杞子100g
将上述药材置于干燥箱内80℃烘干至含水量低于5%,然后一起粉碎成150目细粉,接着加入糖浆,搅拌制软材,制粒,干燥,压制成0.5g/片。由于药材未进行高温膨化和冷冻干燥处理,导致药材中的大量纤维、果胶、蛋白、多糖类物质使粉碎难度加大,为达到150目粉碎粒径要求,必须增加粉碎次数并加大粉碎机功率,从而大幅提高了粉碎周期、能耗和成本,同时还增加了损耗,降低了粉碎回收率。
经检测,片剂中葛根素含量为1.03%,总黄酮含量为5.26%。
试验例
1.崩解时限测定
按照《中国药典》一部附录——崩解时限检查法考察片剂的崩解时限,结果见表1。

从以上结果可以看出,本发明制备的片剂崩解时限明显低于普通方法制备的片剂。
2.溶出度测定
按照《中国药典》一部附录——溶出度测定法第一法测定片剂中葛根素和总黄酮的溶出度,溶出介质为0.1mol/L盐酸溶液,温度为37℃,转速为50rpm,分别于规定的取样时间吸取溶出液,微孔滤膜过滤后分别检测葛根素和总黄酮物质含量,并根据片剂含量计算溶出度。其中,葛根素的含量测定参照《中国药典》一部葛根项下的葛根素含量测定方法;总黄酮参照文献报导的分光光度法,采用检测波长510nm,芦丁标准品为对照。检测结果见表2和表3。

从以上结果可以看出,本发明制备的片剂活性成分溶出速度明显高于按照普通方法制备的片剂,尤其是实施例1的葛根素溶出度于15min时就已达到70%以上,溶出速度处于最高水平。溶出度的提高有利于药物的快速释放和吸收,从而能更好地发挥疗效。
Claims (6)
1.一种缓解视疲劳的中药保健食品,其特征在于由以下重量份数的中药材制成:葛根5-30份、枸杞子5-30份、菊花1-10份、桑葚1-10份,制备方法包括以下步骤:
1)将葛根、菊花先粉碎成粗粉,然后用冷水浸泡1~5小时,取出后沥干表面水分,再置于膨化机中进行高温膨化处理,接着将膨化处理后的上述药材粉碎成细粉;
2)将桑葚、枸杞子粉碎成粗粉,然后在-30℃以下预冻3~5小时,接着在真空条件下冷冻干燥至含水量低于5%,最后在液氮条件下研磨成细粉;
3)将步骤1)和2)中的细粉混合,然后制成颗粒剂或片剂或胶囊剂。
2.如权利要求1所述的中药保健食品制备方法,其特征在于:所述膨化处理的温度为150~250℃,压强为2~6倍标准大气压,时间为5~30分钟。
3.如权利要求1所述的中药保健食品制备方法,其特征在于:所述浸泡时间为3小时。
4.如权利要求1所述的中药保健食品制备方法,其特征在于:所述冷冻干燥的真空度为5~30pa,温度为-10~20℃,时间为15-30小时。
5.如权利要求1所述的中药保健食品制备方法,其特征在于:所述细粉的粒径为100-180目。
6.如权利要求1所述的中药保健食品制备方法,其特征在于:所述中药材的重量份数为:葛根10份、枸杞子10份、菊花6份、桑葚6份。
