The skin care composition is characterized in that the plant components of the witch hazel extract, the aloe leaf extract, the centella extract and the dracaena cochinchinensis leaf extract and the agave stem extract in the skin care composition are mixed and dispersed at a lower temperature (20-50 ℃), so that the inactivation of active ingredients in the plant extracts caused by high temperature is avoided; the mixing and dispersing temperature of the components such as sodium hyaluronate, erythritol and the like can be increased to 70-90 ℃ to ensure full dissolution and dispersion of the components.
Illustratively, the skin care composition is prepared by: sequentially adding sodium hyaluronate, erythritol and water into a vacuum emulsifying pot according to the formula amount, homogenizing and dispersing uniformly, heating to 70-90 ℃, and keeping the temperature and stirring for 5-10 min; and cooling to 20-50 ℃, adding a witch hazel extract, an aloe leaf extract, a centella extract, a dragon blood leaf extract and a agave stem extract, stirring for 10-30 min, and uniformly mixing to obtain the skin care composition.
In another aspect, the present invention provides a skin care product comprising the skin care composition for removing acne marks as described above.

Preferably, the facial essence further comprises at least one of a solvent, a humectant, a thickener, a pH adjuster, a preservative, a solubilizer or a fragrance.

Illustratively, the preparation method of the facial essence comprises the following steps: sequentially adding the thickening agent, the humectant and water into a vacuum emulsifying pot according to the formula amount, homogenizing and dispersing uniformly, heating to 70-90 ℃, and keeping the temperature and stirring for 5-10 min; and cooling to 20-50 ℃, adding the acne and acne removing skin care composition, a pH regulator, a preservative, a solubilizer and an aromatic, stirring for 10-30 min, uniformly mixing, cooling to below 40 ℃, filtering by using filter cloth, and discharging to obtain the facial essence.
Compared with the prior art, the invention has the following beneficial effects:
the skin care composition for removing acnes and lightening spots comprises a witch hazel extract, an aloe leaf extract, a centella extract and a dragon blood leaf extract, and the four natural plant components are cooperated with each other, so that a path formed by acne is used for conditioning and repairing damaged skin, secretion of skin oil is reduced, bacteria and inflammation are inhibited, acne is removed, and immunity and allergy resistance of the skin are improved. The skin care composition for removing acnes and lightening spots provided by the invention can obviously inhibit and kill propionibacterium acnes and staphylococcus aureus, promote the expression of anti-inflammatory cytokine interleukin 10, obviously reduce the expression level of proinflammatory cytokines and inhibit histamine release by synergistically compounding the components in a specific ratio. The skin care composition for removing the acne and lightening the marks has good functions of removing the acne and repairing the acne damaged skin, can reduce the grease secretion of the skin by 30 percent after being used for 4 weeks, reduces the erythema number by 50 percent, has no adverse case and has very obvious treatment effect.

In vitro cell assay testing:
firstly, bacteriostatic experiment:
(1) preparing the drug sensitive tablet: respectively soaking sterile circular filter paper with the diameter of 6mm into the skin care composition for removing acne and lightening marks provided by the examples 1-6 and the comparative examples 1-8, taking out after 30min, and drying in the shade to obtain a drug sensitive tablet;
(2) selecting a propionibacterium acnes colony by using a sterilization inoculating loop, placing the propionibacterium acnes colony in sterile normal saline to prepare a bacterial suspension, and correcting the turbidity of the bacterial suspension to be 0.5 McLee unit; the bacteria solution is uniformly coated on an anaerobic brain-heart infusion flat plate by using a sterile cotton swab, after being dried for 5min, the drug sensitive tablets of examples 1 to 6 and comparative examples 1 to 8 are respectively stuck on the bacteria solution, and then the bacteria solution is put into an anaerobic jar (10% CO)2、10%H2、80%N2) Culturing at 37 ℃ for 48h, taking out, measuring the diameter of the drug sensitive tablet bacteriostatic circle by a cross method, and subtracting 6mm from the measured diameter to obtain the diameter of the bacteriostatic circle;
(3) adding 20mL of nutrient medium into a sterile culture dish, drying, condensing water for solidification, and adhering staphylococcus aureus liquid (about 10%) with a sterile cotton swab5CFU/mL) is uniformly coated on the surface of a flat plate, after no liquid flows on the flat plate, the drug sensitive tablets of examples 1-6 and comparative examples 1-8 are respectively stuck, the culture is carried out at the constant temperature of 37 ℃, and after 24 hours, the diameter of a bacteriostatic circle is measured by a cross method; the zone diameter data is shown in table 2.

As can be seen from the data in table 2, the skin care composition for removing acne and lightening marks provided in embodiments 1 to 6 of the present invention has an obvious growth inhibition and killing effect on propionibacterium acnes and staphylococcus aureus, forms an inhibition zone with a diameter greater than 15mm, can effectively inhibit bacteria and achieve an acne treatment effect by conditioning the micro-ecological environment of damaged skin. The antibacterial effect of the skin care composition in the embodiments 1 to 4 is better than that of the embodiments 5 to 6, which shows that the mass ratio of the dragon tree leaf extract to the centella asiatica extract is more than the preferable 1 (0.8 to 1.5) in the invention, so that the antibacterial effect of the skin care composition is reduced.
The skin care composition for removing acne and lightening marks has the antibacterial and antibacterial effects which are realized by mutually matching four plant components including a witch hazel extract, an aloe leaf extract, a centella asiatica extract and a dragon blood leaf extract, and the diameter of an antibacterial ring is reduced and the antibacterial and antibacterial effects are weakened due to the absence of any one plant component (comparative examples 1-4). In addition, the four plant extracts have a synergistic effect, and particularly, the antibacterial effect achieved by the synergistic combination of the centella asiatica extract and the dragon blood leaf extract is obviously superior to that of an excessive single component (comparative examples 5-6), and the antibacterial effect achieved by the synergistic combination of the witch hazel extract and the aloe leaf extract is obviously superior to that of an excessive aloe leaf extract or an excessive witch hazel extract (comparative examples 7-8).
II, anti-inflammatory experiment:
(1) cell collection: mixing the blood sample with the blood cell separation solution in equal proportion, centrifuging at 20 deg.C for 40min, collecting human peripheral blood mononuclear cell layer, washing cells with buffer solution, and suspending the cells with culture medium RPMI-1640; CD14+ monocytes in human peripheral blood mononuclear cells were then purified with CD14+ microspheres (MiniMACS system); the differentiation of cells into dendritic cells is stimulated by the cytokine IL-4 and the growth hormone GM-CSF at 37 ℃ with 5% CO2Culturing for 6 days, collecting differentiated dendritic cells and inoculating the dendritic cells into a pore plate;
(2) the skin care compositions for removing acne and lightening marks provided in example 1 and comparative examples 5 to 8 were added to the well plate at a concentration of 2 μ g/mL (calculated as the content of plant extract in the skin care composition), and the wells to which the medium of the same volume was added were set as blank control at 37 ℃ with 5% CO2Co-culturing for 48 hours; the cell culture supernatant was collected. The enzyme-linked immunosorbent assay (ELISA) is used for detecting the content of the interleukins IL-10 and IL-12 in the supernatant, the content of the anti-inflammatory cytokine IL-10 in the blank control group is defined as 0, the content of the proinflammatory cytokine IL-12 is defined as 100%, and the test results are shown in Table 3.

According to the data in table 3, the skin care composition for removing acne and lightening marks provided in example 1 of the present invention can significantly increase the level of anti-inflammatory cytokine interleukin IL-10, and inhibit the expression level of proinflammatory cytokine interleukin IL-12, which proves that the skin care composition of the present invention has a good anti-inflammatory effect.
Comparing the data of example 1 with comparative examples 5-8, it can be seen that the anti-inflammatory effect of the skin care composition is obtained based on the synergistic cooperation of four natural plant components, namely, the witch hazel extract, the aloe leaf extract, the centella asiatica extract and the dragon blood leaf extract, and the components have synergistic effect and non-substitutability, the synergistic combination effect of the centella asiatica extract and the dragon blood leaf extract is obviously superior to the anti-inflammatory effect of excessive single component (comparative examples 5-6), and the synergistic combination effect of the witch hazel extract and the aloe leaf extract is obviously superior to the anti-inflammatory effect of excessive aloe leaf extract or excessive witch hazel extract (comparative examples 7-8). In addition, the anti-inflammatory effects of comparative examples 7-8 were better than comparative examples 5-6, demonstrating that centella asiatica extract and dragon's leaves extract contributed more to the anti-inflammatory efficacy of the skin care composition.
Thirdly, antiallergic experiment:
histamine is often stored in mast cells of human tissues, and when a human body is stimulated by external stimuli to trigger an antigen-antibody reaction, the permeability of cell membranes of the mast cells is changed to release histamine, and the histamine and a histamine receptor generate pathophysiological effects, so that the histamine release concentration is one of indexes for evaluating allergic reactions.
KU812 cells have high similarity to mast cells, and an antiallergic experiment was performed using KU812 cells as model cells: KU812 cells were cultured at 2.0X 105The density of each well is inoculated in 12-well plate, 37 ℃, 5% CO2Cells adhere to the wall after 24h of culture; the skin care compositions for removing acne and lightening marks provided in example 1 and comparative examples 5 to 8 were added at a concentration of 5. mu.g/mL per well (calculated as the content of plant extract in the skin care composition), at 37 ℃ and 5% CO2Co-culturing for 48 h; discarding the culture medium and adding 1mL of Tyrode solution to balance for 10 min; 50 μ L C48/80(10 μ g/mL) was added to each well, a blank control group was set and 50 μ L of Tyrode solution was added, the reaction was terminated after 30min, the supernatant and cell pellet were separated, the histamine content was measured by histamine fluorescence, and the calculated histamine inhibition rate is shown in Table 4.

According to the data in table 4, the inhibition rate of the skin care composition for removing acne and pale spots provided in embodiment 1 of the invention on histamine is more than 40%, which is obviously higher than the inhibition rate of the skin care composition in comparative examples 5 to 8 on histamine, and the synergistic combination of the four active ingredients of the witch hazel extract, the aloe leaf extract, the centella asiatica extract and the dragon's blood leaves extract in the invention is proved to be capable of effectively inhibiting histamine release and realizing the anti-allergic effect.
Application example 1
The facial essence comprises the following components in percentage by mass: example 1 provides a skin care composition that removes acne marks 40%, butylene glycol 2%, propylene glycol 5%, glycerin 10%, triethanolamine 0.3%, methylparaben 0.3%, phenoxyethanol 0.1%, ethylhexylglycerin 0.1%, PEG-40 hydrogenated castor oil 0.08%, carbomer 3%, perfume 0.2%, and balance water.
The preparation method comprises the following steps: sequentially adding carbomer, butanediol, propylene glycol, glycerol and water into a vacuum emulsifying pot according to the formula amount, homogenizing and dispersing uniformly, heating to 85 ℃, and keeping the temperature and stirring for 10 min; cooling to 40 deg.C, adding the skin care composition, triethanolamine, methyl hydroxybenzoate, phenoxyethanol, ethylhexyl glycerol, PEG-40 hydrogenated castor oil and essence, stirring for 20min, mixing, cooling to below 35 deg.C, filtering with 200 mesh filter cloth, and discharging to obtain the facial essence.
And (3) performance testing:
30 acne patients (half of men and women) aged 18-45 years are used as test volunteers and are randomly divided into 15 treatment groups and 15 control groups. After the face of the tested volunteer is cleaned, a VISIA facial image analyzer is used for carrying out facial shooting, the facial skin damage condition and the range of an acne area are analyzed, the skin oil content is detected at the cheek, the forehead and the chin, and the average value obtained by 3 times of testing of each part is used as an initial value. The treatment group applied the facial essence provided in application example 1 to the acne affected part 2 times a day (1 time each morning and evening), and the control group applied a placebo containing no skin care composition of the present invention to the acne affected part 2 times a day (1 time each morning and evening), for 4 weeks as a treatment course. The information obtained by the VISIA facial image analyzer after the end of a treatment period was compared with the information before treatment, and the skin lesion improvement, erythema number and oil fat rate of the test volunteers were analyzed, and the statistical data are shown in table 5.

Compared with the results of the VISIA test before and after treatment, no adverse reaction cases occur during the treatment, the total skin feel is good and fresh, the skin damage condition and the color of the affected part of the tested volunteer are obviously improved, the erythema condition left by acne is obviously reduced, the proportion of the reduction of the erythema number can reach more than 50 percent, the skin oil content is obviously reduced, the reduction proportion reaches 30 percent, and the curative effect evaluation is effective.
The applicant states that the present invention is illustrated by the above examples to the acne and acne mark removing skin care composition of the present invention and the preparation method and application thereof, but the present invention is not limited to the above examples, i.e. it does not mean that the present invention must be implemented by the above examples. It should be understood by those skilled in the art that any modification of the present invention, equivalent substitutions of the raw materials of the product of the present invention, addition of auxiliary components, selection of specific modes, etc., are within the scope and disclosure of the present invention.
