Description

Blackhead-derived plant composition, blackhead-derived liquid and preparation method
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a blackhead-derived plant composition, a blackhead-derived liquid and a preparation method thereof.
Background
Acne is one of the most common diseases in the skin-care department, easily occurs on the face, is easy to generate scars after improper treatment, and brings trouble to people. Blackhead comedones, also known as blackheads, are open comedones that appear well on the face and back and are characterized by a dark spot in an enlarged pore that appears as a bug after extrusion, with a blackened tip. The etiology is as follows: under the action of androgen, the pilosebaceous canal generates hyperkeratosis, the follicular wall is thickened, the excretion of the sebaceous gland is blocked, and the increase of epithelial cells cast off from the follicular wall and the excretion of the sebaceous gland are blocked. The mixture of the epithelial cells on the hair follicle wall and the sebum blocks the hair follicle opening to form acne, and the acne is exposed at the top end outside the hair follicle opening. Through the oxidation of air, the deposition of melanin, the pollution of dust and the radiation of ultraviolet rays, the blackhead is changed into the comedone.
The existing solutions in the market for such situations have poor effects, lack basic research on blackhead causes, and are compounded by blindly using some plant ingredients, so that although the solutions have a good moistening effect on skin, the current situation of blackheads cannot be improved, and even more blackheads are used, the more the situation is obvious. The reasons are that they cannot deeply dredge pores, cannot inhibit the secretion of sebaceous glands, the root cause of hair follicle obstruction is not solved, comedones can quickly recur, and the phenomenon of lying one behind the other occurs. The blackhead removing cosmetics capable of controlling oil are mostly chemical synthetic products, blackheads are removed through violence of a chemical adsorbent, the blackhead removing cosmetics are extremely harmful to skin after being used, although the blackhead removing cosmetics can temporarily solve the blackhead problem, the blackhead removing cosmetics cause extremely large damage and harm to pores, elastic fibers are extremely prone to losing energy, the skin is loose, and the pores are large.
CN108567634A discloses a pure natural blackhead export repair composition and a preparation method thereof, and the composition is prepared from 0.1 to 10 weight parts of red melon fruit, 0.1 to 10 weight parts of rambutan and 0.1 to 10 weight parts of Uncaria tomentosa. Although the blackhead export repair composition can temporarily solve the blackhead problem, the root cause of hair follicle blockage is not solved, and the current situation of blackheads cannot be fundamentally improved.
CN105662914A discloses a blackhead discharge liquid and a blackhead discharge film for reducing stimulation, wherein the blackhead discharge liquid comprises organic acid and chitosan; the blackhead removing film consists of a blackhead removing solution and a chitosan film, the chitosan film is completely soaked in the blackhead removing solution, and the blackhead removing solution contains organic acid as a main effective component. The main effective component in the blackhead removing solution is organic acid, and the blackhead removing solution can cause great damage and harm to pores after long-time use, so that elastic fibers of the skin lose vitality, the skin is loose, and the pores are large.
CN105078796A discloses a blackhead-derived essence film, which adopts traditional Chinese medicinal materials such as radix angelicae, rhizoma typhonii, cortex phellodendri, radix scutellariae and chamomile, and although the blackhead-derived essence film can temporarily solve the blackhead problem, the blackhead-derived essence film has poor capability of inhibiting grease generation, and cannot fundamentally solve the blackhead generation.
Therefore, the development of a care product capable of effectively and mildly inducing blackheads to inhibit blackhead regeneration is the focus of current research in the field.

A blank control group of medium and cells was set up simultaneously. After the plate was incubated in an incubator for 72 hours, the supernatant was discarded, washed twice with PBS (phosphate buffered saline), and then 0.5mL of trypsinized cells were added to each well for 3min, and 2mL of a maintenance solution was added to each well to terminate the digestion. After mixing, 0.5mL of each concentration was taken out and counted. Centrifuging the rest cell suspension at 2500r/min for 5min, discarding supernatant, adding NaOH solution into the precipitate, heating to dissolve melanin, and measuring absorbance at 490nm under enzyme-linked immunosorbent assay.
The formula for calculating the melanin synthesis inhibition (%) of the sample is shown as follows:
Figure BDA0002254408870000221
wherein, A1Is the absorbance value of the drug well, P1Cell density of drug well, A2Absorbance values for control wells; p2In order to control the cell density of the wells, I' is the melanin synthesis inhibition (%) of the corresponding samples, and the specific test results are shown in table 4.
TABLE 4
Figure BDA0002254408870000222
Figure BDA0002254408870000231
From the above test results, it can be seen that the blackhead-derived liquid prepared in examples 1 to 10 of the present invention can effectively inhibit melanin synthesis, and the inhibition rate of melanin synthesis is above 50%, while the blackhead-derived liquid prepared in comparative examples 1 to 5 has far lower inhibition effect on melanin synthesis than the blackhead-derived liquid prepared in the present application, which indicates that the blackhead-derived plant composition of the present invention selects four plant extracts, i.e., aloe barbadensis leaf extract, pomegranate fruit extract, tea leaf extract and agave stem extract, and makes them cooperate with each other to achieve synergistic effect, so that not only blackhead and whitehead can be removed, but also melanin can be effectively inhibited from being generated, thereby being helpful to fundamentally solve the blackhead problem and preventing the blackhead from being regenerated again.
Test example 4

The test results are shown in table 5:
TABLE 5
Sample(s) 5 alpha-reductase inhibitory rate
Example 1 51.15%
Example 2 49.84%

Comparative example 1 8.45%
Comparative example 2 20.63%
Comparative example 3 22.35%
Comparative example 4 29.47%
Comparative example 5 25.48%
From the above test results, it is understood that the blackhead discharge liquids prepared in examples 1 to 10 have higher inhibition rates against 5 α -reductase than the blackhead discharge liquids prepared in comparative examples 1 to 5, and that the inhibition rate against 5 α -reductase is 48% or more, and the highest inhibition rate can be 58.06%. The blackhead-derived plant composition selects four plant extracts, namely the aloe vera leaf extract, the pomegranate fruit extract, the tea leaf extract and the agave stem extract, and the four plant extracts are matched with each other to realize synergistic interaction, so that the blackhead and the white head can be removed, and the generation of grease can be effectively controlled, thereby solving the problems of the obstructed sebaceous gland excretion, the increase of epithelial cells falling off from the hair follicle wall and the obstructed sebaceous gland excretion, solving the root problem generated by the blackhead and fundamentally avoiding the regeneration of the blackhead.
