Abstract

本发明提供一种新型K血清型副溶血弧菌及其应用。通过与69种已知K血清进行凝集反应和全基因组测序方法,确定新型K血清型副溶血弧菌,保藏编号为CGMCC No.17249。本发明制备的新型K血清,特异性强,可方便快捷地用于检测一种全新的且近几年逐渐上升的新型K血清型副溶血弧菌,为感染性腹泻病原诊断、监测和防控提供重要的检测技术。

Description

一种新型K血清型副溶血弧菌及其应用
技术领域
本发明属于生物技术领域,涉及一种新型K血清型副溶血弧菌及其应用。
背景技术
副溶血弧菌(Vibrio parahaemolyticus,VP)是食源性疾病的主要病原菌,人类主要通过海 水暴露,生食或半生食海产品而感染,通常引起急性胃肠炎,偶尔也会引起伤口感染和败血 症,严重者可导致死亡。在我国,食源性疾病暴发监测网数据显示,副溶血性弧菌引发的食 源性疾病已超过沙门菌和金黄色葡萄球菌,成为食源性疾病暴发中最重要的生物性致病因素。
血清学分型常用于细菌的溯源和遗传变异研究。副溶血弧菌可根据热稳定的菌体抗原(O) 和热不稳定的荚膜抗原(K)进行血清分型。目前,日本副溶血弧菌分型委员会已确认了11 种O和69种K血清型。1996年以前,副溶血弧菌感染并没有呈现明显的优势血清型。直 至1996年2月在印度加尔各答市引起群体暴发性食物中毒事件的新型O3:K6血清型菌株出现。此新发菌株随后迅速在全球多个沿海国家及地区的人群中发生了广泛的跨洲际传播, 并导致了多起大规模暴发事件,成为了副溶血弧菌致病的最主要亚种。然而,自2013年起, 一种与O4血清凝集但与现有69种K血清均不凝集(KUT)的副溶血弧菌明显增多,在短期内形成跨地区、跨人群感染的优势传播,极有可能成为一种重要的流行株。全基因组测序发现该新型菌株的K抗原决定簇与目前NCBI数据库中副溶血弧菌K抗原决定簇的序列均不同。由此认为,该副溶血弧菌是一种新型K血清型菌株,命名为O4:KUT-recAin。已于2019年2 月20日保藏在中国微生物菌种保藏管理委员会普通微生物中心(简称为CGMCC),保藏编号 为CGMCC No.17249。待相关文章发表后,提交材料至副溶血弧菌血清分型委员会(日本) 进行新血清型命名。本发明涉及一种新型K血清型副溶血弧菌及制备其特异性血清用于腹泻 相关病原体的检测。制备的血清能有效地诊断和监测该血清型副溶血弧菌的暴发,对感染性腹泻病原的防控有着重要的意义。
发明内容
本发明的目的在于提供一种新型K血清型副溶血弧菌,该新型K血清型副溶血弧菌的保 藏编号为CGMCC No.17249,分类命名为副溶血弧菌Vibrio parahaemolyticus,保藏日期为2019 年2月20日,保藏于中国微生物菌种保藏管理委员会普通微生物中心(北京市朝阳区北辰西 路1号院3号,中国科学院微生物研究所)。该副溶血弧菌株的K抗原决定簇的氨基酸序列 SEQ ID No:1所示。
本发明的另一个目的是提供上述副溶血弧菌在制备新型K血清中的应用。
本发明所述的新型K血清,是用上述所述的副溶血弧菌作为抗原,免疫动物制备的,并 经过吸附作用,得到的特异性免疫血清。
优选的,所述的抗原为灭活抗原。
优选的,所述的动物为新西兰大白兔。
优选的,所述的血清是通过与其他不同K血清型菌株的吸附作用,实现特异性。
本发明所述新型K血清的制备方法,通过以下步骤实现:
(1)抗原准备:将副溶血弧菌接种于3%NaCl的BHI培养基中,37℃过夜培养后,收集菌体,用生理盐水清洗2次后,用含0.5%甲醛的生理盐水溶液灭活菌体24h,之后将灭活的菌体溶于生理盐水中,作为抗原。
(2)收集免疫血清:
挑选健康的12周大小的新西兰大白兔,将灭活的副溶血弧菌O4:KUT-recAin株溶于0.85% NaCl中,浓度为109CFU/ml,每只兔子免疫4个部位,每个部位250μl,往后间隔2周加强 免疫1次,免疫量与首次相同,共免疫5次后,通过心脏采血法采集全血,采集的全血在37℃ 放置2h后,转移到4℃过夜,之后离心收集血清,该血清即为免疫血清。
(3)特异性免疫血清的制备:
将制备得到的免疫血清与权利要求1所述的副溶血弧菌进行凝集反应,凝集现象明显, 与已知K血清型菌株中的K24、K59、K61血清型菌株有交叉反应,依次用0.5%甲醛灭活的 浓度为109CFU/ml的K24菌株、K61菌株、K59菌株4℃吸附免疫血清,之后离心得到的上清,即为所述的副溶血弧菌特异性免疫血清,也即本发明的新型K血清。
(4)新型K血清的性能:
新型K血清与副溶血弧菌O4:KUT-recAin株发生明显凝集,与33种已知K血清型副溶 血弧菌均不发生凝集,与其它肠道致病菌霍乱弧菌、河流弧菌、空肠弯曲菌、结肠弯曲菌、 大肠埃希菌、鼠伤寒沙门菌、伤寒沙门菌、痢疾志贺菌和福氏志贺菌均不发生凝集。用生理 盐水倍比稀释新型K血清至64倍,新型K血清与副溶血弧菌O4:KUT-recAin株仍有明显凝 集。新型K血清具有很好的特异性和灵敏度。
本发明的优点和效果:
本发明的优点是提供了一种新型K血清型副溶血弧菌,制备其特异性血清用于腹泻相关 病原体的检测。制备的新型K血清特异性强,可方便快捷地用于检测一种全新的且近几年感 染率逐渐上升的新型K血清型副溶血弧菌(O4:KUT-recAin)。为感染性腹泻病原诊断、监测和 防控提供重要的检测技术。

具体实施方式
下面结合附图和具体实施例,进一步阐述本发明。应理解这些实施例仅用于说明本发明, 不是限定性的,不能以下述举例说明来限定本发明的保护范围。
实施例1一种新型K血清型副溶血弧菌(O4:KUT-recAin)的分离、鉴定和毒力试验:
急性腹泻患者粪便标本接种于碱性蛋白胨水,37℃增菌6小时,转种到TCBS培养基。 37℃孵育18小时,选取单个菌落,经布鲁克基质辅助激光解吸电离-飞行时间(MALDITOF) 质谱仪鉴定为副溶血弧菌。使用日本生研的副溶血弧菌诊断血清(包含11种O和69种K), 以0.85%生理盐水为阴性对照,通过玻片凝集法确定副溶血弧菌血清型。与现有69种K血清 均不凝集的副溶血弧菌标记为KUT。
将KUT菌株进行全基因组测序,分析其K抗原决定簇,进一步确定是副溶血弧菌的一 种或几种新型K血清型菌株。本发明提供一种与O4血清凝集但与现有69种K血清均不凝集的新型K血清型副溶血弧菌,命名为O4:KUT-recAin株,已于2019年2月20日保藏在中 国微生物菌种保藏管理委员会普通微生物中心(简称为CGMCC),保藏编号为CGMCC No.17249,保藏地点中国北京。
全基因组测序分析副溶血弧菌O4:KUT-recAin株的K抗原决定簇,发现该菌株的K抗原 决定簇(见序列1)与目前NCBI数据库中副溶血弧菌K抗原决定簇的序列均不同。进一步 证明,该副溶血弧菌是一种新型K血清型菌株。副溶血弧菌O4:KUT-recAin株K抗原决定簇 序列与37种已知K抗原决定簇序列相似性比对结果见图1。
副溶血弧菌O4:KUT-recAin株在平板上的生长形态如图2所示,挑取生长良好的菌落, 接种于3%NaCl的BHI培养基(pH=8.5)中37℃培养,生长曲线见图4。图4中O4:KUT-recAin 株的培养特性显示,在接种2h左右,溶液的OD600nm值开始上升,在培养14-16h之间溶液的 OD600nm值达到最大,以后随着时间延长,溶液OD600nm值反而下降。
将1×1010CFUs的副溶血弧菌O4:KUT-recAin株喂服给出生2天的乳兔,20h后,乳兔出 现腹泻症状,严重的出现体重下降,甚至死亡。图5是5窝共37只乳兔喂服副溶血弧菌O4:KUT-recAin株的致泻情况和死亡情况。2天后乳兔的腹泻率为41%,5天后乳兔的腹泻率达到60%。感染8天后,乳兔死亡率为35%。
实施例2副溶血弧菌的一种新型K血清的制备:
抗原准备:将新型K血清型副溶血弧菌O4:KUT-recAin株接种于3%NaCl的BHI培养基 中,37℃过夜培养后,3000×g离心10min收集菌体,用生理盐水清洗2次后,用含0.5%甲醛的生理盐水灭活菌体24h,3000×g离心10min收集灭活的菌体,最后将灭活的菌体溶于生理盐水中,用于免疫兔子。
免疫兔子:挑选健康的12周大小的新西兰大白兔3只,使其适应环境3天后,将灭活的新型K血清型副溶血弧菌溶于0.85%NaCl中(浓度为109CFU/ml)免疫兔子,每只兔子免疫4个部位(背部和大腿根部的左右两侧),每个部位250μl,间隔2周加强免疫1次,免疫 量与首次相同,免疫5次后,通过心脏采血法采集全血。
收集免疫血清:采集的全血在37℃放置2h后,转移到4℃过夜,次日12000×g离心10min 收集上清,该上清即为免疫血清。
制备特异性免疫血清:将收集得到的免疫血清与新型K血清型菌株进行玻片凝集反应, 玻片凝集具体操作步骤如下:使用记号笔对载玻片进行分区划线,每区内滴加制备的新型K 血清1滴(约5μl),其中一个区域滴加生理盐水代替新型K血清作为对照以排除自身凝集, 之后每区加入5μl菌悬液,充分混合反应1min后,观察是否有凝集现象。制备得到的免疫血 清与新型K血清型菌株凝集现象非常明显。新型K血清型菌株免疫血清与已知K血清型菌 株中的K24、K59、K61血清型菌株有交叉反应(图6)。依次用0.5%甲醛灭活的浓度为109CFU/ml的K24菌株、K61菌株、K59菌株4℃吸附免疫血清2-24h,12000×g离心10min 得到的上清,即为新型K血清型菌株特异性的免疫血清(图7),称为新型K血清。
实施例3新型K血清的性能:
制备得到的新型K血清与KUT菌株能够快速的通过凝集现象检测出新型K血清型菌株 (O4:KUT-recAin)(图8)。新型K血清与33种已知K血清型副溶血弧菌均不发生凝集(表1), 与其它肠道致病菌霍乱弧菌、河流弧菌、空肠弯曲菌、结肠弯曲菌、大肠埃希菌、鼠伤寒沙 门菌、伤寒沙门菌、痢疾志贺菌和福氏志贺菌均不发生凝集(表2)。用生理盐水倍比稀释的 方法,将新型K血清稀释,稀释至64倍时,与新型K血清型菌株(O4:KUT-recAin)仍有明显的凝集现象(表3)。新型K血清具有很好的特异性和灵敏度。

实施例4新型K血清的应用:
于2013-2017年收集浙江、江苏、辽宁、深圳和上海等地急性腹泻患者来源的副溶血弧 菌2368株,用制备的新型K血清进行玻片凝集实验,共鉴定出329株新型K血清型菌株(O4:KUT-recAin)。对其中的161株进行全基因组测序,分析K抗原决定簇,结果均与副溶血弧菌O4:KUT-recAin株K抗原决定簇相同,即都为新型K血清型副溶血弧菌(O4:KUT-recAin)。 利用制备的新型K血清采用玻片凝集法鉴定新型K血清型副溶血弧菌(O4:KUT-recAin)的 准确性为100%。因此,新型K血清能够方便快捷的鉴定出新型K血清型副溶血弧菌 (O4:KUT-recAin)。

Claims (7)

1.一种新型K血清型副溶血弧菌,其特征在于,该新型K血清型副溶血弧菌的保藏编号为CGMCC No.17249,分类命名为Vibrio parahaemolyticus。该副溶血弧菌株的K抗原决定簇的核苷酸序列SEQ ID No:1所示。
2.权利要求1所述的副溶血弧菌在制备新型K血清中的应用。
3.根据权利要求2所述的应用,其特征在于,所述的新型K血清是用权利要求1所述的副溶血弧菌作为抗原,免疫动物制备的,并经过吸附作用,得到的特异性免疫血清。
4.根据权利要求3所述的应用,其特征在于,所述的抗原为灭活抗原。
5.根据权利要求3所述的应用,其特征在于,所述的动物为新西兰大白兔。
6.根据权利要求3所述的应用,其特征在于,所述的血清是通过与其他不同K血清型菌株的吸附作用,实现特异性。
7.根据权利要求2所述的应用,其特征在于,所述新型K血清的制备方法,通过以下步骤实现:
(1)抗原准备:将副溶血弧菌接种于3%NaCl的BHI培养基中,37℃过夜培养后,收集菌体,用生理盐水清洗后,用含0.5%甲醛的生理盐水溶液灭活菌体24h,之后将灭活的菌体溶于生理盐水中,作为抗原;
(2)收集免疫血清:
挑选健康的新西兰大白兔,将灭活的副溶血弧菌溶于0.85%NaCl中,浓度为109CFU/ml,免疫4个部位,每个部位250μl,往后间隔2周加强免疫1次,免疫量与首次相同,共免疫5次后,采集全血,在37℃放置2h后,转移到4℃过夜,之后离心收集血清,该血清即为免疫血清;
(3)特异性免疫血清的制备:
将制备得到的免疫血清与权利要求1所述的副溶血弧菌进行凝集反应,凝集现象明显,与已知K血清型菌株中的K24、K59、K61血清型菌株有交叉反应,依次用0.5%甲醛灭活的浓度为109CFU/ml的K24菌株、K61菌株、K59菌株4℃吸附免疫血清,之后离心得到的上清,即为本发明的新型K血清。
