另一方面,PD-L2表达比PD-L1更受限制。例如,PD-L2在DC、巨噬细胞和骨髓来源的肥大细胞上可诱导地表达。
另外,一些研究显示,PD-L1的受体不依赖于PD-1。B7.1也被确定为PD-L1的结合伴侣。Butte et al., Immunity 27: 111-22 (2007)。化学交联研究表明,PD-L1和B7.1能够通过其IgV样结构域相互作用。B7.1:PD-L1相互作用能够诱导进入T细胞的抑制信号。通过B7.1在CD4+ T细胞上连接PD-L1,或通过PD-L1在CD4+ T细胞上连接B7.1,都产生抑制信号。当受到抗CD3加B7.1包被的珠子刺激时,缺乏CD28和CTLA-4的T细胞显示增殖和细胞因子产生减少。在缺乏B7.1的所有受体(即CD28、CTLA-4和PD-L1)的T细胞中,抗CD3加B7.1包被的珠子不再抑制T细胞增殖和细胞因子产生。这表明在不存在CD28和CTLA-4的情况下,B7.1通过PD-L1特异性地作用于T细胞。同样地,当在抗CD3加PD-L1包被的珠子的存在下进行刺激时,缺乏PD-1的T细胞显示增殖和细胞因子产生减少,证明PD-L1连接对T细胞上的B7.1的抑制作用。当T细胞缺乏PD-L1的所有已知受体(即,没有PD-1和B7.1)时,抗CD3加PD-L1包被的珠子不再损害T细胞增殖。因此,PD-L1能够通过B7.1或PD-1对T细胞发挥抑制作用。
在人疾病的情况下,已经在几种鼠和人癌症(包括人肺癌、卵巢癌和结肠癌以及各种骨髓瘤)中发现了PD-L1表达(Iwai et al. (2002) PNAS 99: 12293-7; Ohigashi etal. (2005) Clin Cancer Res 1 1 :2947-53)。有人已经表明,PD-L1通过增加抗原特异性T细胞克隆的凋亡而在肿瘤免疫中发挥作用(Dong et al. 概述

II.抗PD-L1抗体
除其他原因外,本文提供的抗PD-L1抗体可以被用于治疗癌症等。据信本公开的抗PD-L1抗体通过可归因于PD-L1的至少一种负的共刺激信号的拮抗来增强宿主免疫应答的共刺激。
本公开的抗PD-L1抗体及其功能片段将具有多种用于治疗癌症或恶性疾病的功能性质,包括但不限于具有抗体依赖性细胞毒性(ADCC)活性、具有抗肿瘤活性、抑制PD-L1与PD-1的结合、以及阻止PD-1介导的T细胞活化抑制。

III.药物组合物和制剂
适用于本文所述方法的药物组合物可以包括治疗活性剂(例如抗PD-L1抗体或其功能片段)和药学上可接受的载体或稀释剂。
该组合物可以被配制用于静脉内的、皮下的、腹膜内的、肌肉内的、口腔的、鼻的、肺的、眼的、阴道的、或直肠的施用。在一些实施方案中,抗PD-L1抗体或其功能片段被配制用于静脉内的、皮下的、腹膜内的或肌肉内的施用,例如在溶液、悬浮液、乳液、脂质体制剂等中。使用本领域已知的技术,可以将药物组合物配制成速释组合物、缓释组合物、迟释组合物等。
用于各种剂型的药学上可接受的载体是本领域已知的。例如,已知用于固体制剂的赋形剂、润滑剂、粘合剂和崩解剂;已知用于液体制剂的溶剂、增溶剂、悬浮剂、等渗剂、缓冲剂和舒缓剂。在一些实施方案中,药物组合物包括一种或多种另外的组分,例如一种或多种防腐剂、抗氧化剂、稳定剂等。
另外,本公开的药物组合物可以被配制成溶液、微乳液、脂质体或适合于高药物浓度的其他有序结构。载体可以是溶剂或分散介质(含有例如水、乙醇、多元醇(例如,甘油、丙二醇和液体聚乙二醇等)),以及合适的其混合物。例如,通过使用诸如卵磷脂的涂层、通过在分散的情况下保持所需的粒度和通过使用表面活性剂,可以保持适当的流动性。在一些实施方案中,优选在组合物中包括等渗剂,例如糖、多元醇(如甘露醇、山梨糖醇)或氯化钠。通过在组合物中包括延迟吸收的试剂(例如单硬脂酸盐和明胶),可以实现可注射组合物的延长吸收。

本公开的药物组合物能与其他治疗剂组合施用。例如,组合疗法能包括药物组合物,其包含至少一种本公开的抗PD-L1抗体或其功能片段和至少一种或多种另外的治疗剂,所述另外的治疗剂包括但不限于CAR T细胞(例如,表达抗CD19、抗Her2、抗BCMA、抗CS-1、抗PSCA、抗CAIX,抗IL13R或抗PD-L1 CAR的经修饰的T细胞)、其他肿瘤靶向抗体(例如,抗CAIX抗体)、免疫应答增强方式(例如,抗GITR抗体、抗OX40抗体、抗CD137抗体或TLR激动剂)和小分子药物(例如,BTK抑制剂、EGFR抑制剂、BET抑制剂、PBKδ抑制剂、BRAF抑制剂或PARP抑制剂)。本发明的药物组合物还能与放射疗法联合施用。
IV.治疗癌症的方法
本文提供了用本公开的抗PD-L1抗体治疗癌症、恶性疾病或癌症细胞增殖的方法。更具体地,本公开内容提供了增强T细胞功能和抗肿瘤免疫力的方法,包含施用治疗有效量的任何上述抗PD-L1抗体或组合物。
增强T细胞功能和抗肿瘤免疫力为治疗癌症、恶性疾病或癌症细胞增殖提供了广谱方法。因此,可以通过施用本公开的抗PD-L1抗体及其功能片段来治疗多种类型的癌症。例如,在一些实施方案中,癌症是血液癌症(例如,淋巴瘤、非霍奇金淋巴瘤、慢性淋巴细胞白血病或多发性骨髓瘤)、神经系统癌症、乳腺癌症、胃肠癌症(例如结肠癌)、肾细胞癌(例如透明细胞肾细胞癌)、或泌尿生殖系统癌症(例如卵巢癌)。在一些实施方案中,癌症是黑素瘤、肺癌(例如,非小细胞肺癌)、头颈癌、肝癌、胰腺癌、骨癌、前列腺癌、膀胱癌、或血管癌。
在一些实施方案中,根据本公开的方法治疗的癌症是高度免疫原性癌。并且在一些实施方案中,根据本公开的方法治疗的癌症是表达PD-L1的癌症。
调整剂量方案以提供最佳的所需反应(例如,如肿瘤消退或缓解的治疗反应)。例如,在一些实施方案中,可以施用单次推注,而在一些实施方案中,可以随时间施用几个分开的剂量,或者可以按照情况所示的按比例减少或增加剂量。例如,在一些实施方案中,本公开的抗体或功能片段可以通过皮下或静脉内注射每周施用一次或两次。在一些实施方案中,本公开的抗体或功能片段可以通过皮下注射每月施用一次或两次。在一些实施方案中,本公开的抗体或功能片段可以每周施用一次、每隔一周施用一次、每三周施用一次、每四周施用一次、每隔一个月施用一次、每三个月施用一次、每四个月施用一次、每五个月施用一次、或每六个月施用一次。
示例性剂量可以根据所治疗个体的尺寸和健康状况以及所治疗的条件而变化。例如,在一些实施方案中,本公开的抗体或功能片段可以以1-100 mg/kg的剂量施用。在一些实施方案中,本公开的抗体和功能片段可以以以下剂量施用:1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20、25、30、35、40、45、50、55、60、65、70、75、80、85、90、95、或100mg/kg。

抗体生产和纯化
使酵母克隆生长至饱和,然后在30℃下振荡诱导48小时。诱导之后,沉淀酵母细胞,收获上清液用于纯化。使用蛋白质A柱纯化IgG,并用pH 2.0的乙酸洗脱。通过木瓜蛋白酶消化来产生Fab片段,并通过KappaSelect(GE Healthcare LifeSciences)进行纯化。
实施例3-具有PD-L1抗体的竞争性FACS
用PD-L1表达载体转染中国仓鼠卵巢(CHO)细胞,随后选择用于蛋白质的表达(PD-L1+细胞)。CHO细胞与1 μg/ml生物素标记的PD-1温育1小时。
在与生物素标记的PD-1温育之后,将抗PD-L1抗体以从10 μl/ml开始的4倍稀释添加至上清液,并孵育1小时。洗涤细胞,然后与链霉亲和素-PE接触。通过流式细胞术来分析链霉亲和素-PE染色,以确定抗PD-L1抗体对PD-1结合的抑制百分比。

计算几种抗体的IC50值,包括临床对照mAb(由SEQ ID NO:73代表的VH结构域和SEQID NO:74代表的VL结构域定义)、CTI-09、CTI-48、CTI-50和CTI-58,并且可以在表3中找到。图1显示了这项研究的结果。
实施例4-抗体结合动力学、特异性和选择性
Octet数据分析用于确定亲和力测量以评估抗体结合动力学。在动力学缓冲液中以20ug/mL(默认浓度)制备2 mL上样样品。将至少200 uL等分至黑色96孔板中。样品的浓度范围基于相互作用的估计的KD(如果可用)。通常,连续稀释在0.1 KD至10 KD的范围内。使用动力学缓冲液作为样品稀释剂,将7点稀释液制成样品柱。样品柱的最后一个孔在之后数据分析中用作参考,应仅含有动力学缓冲液。
将生物传感器在动力学缓冲液(lx PBS、0.1% BSA、0.02% Tween20、0.05% 叠氮化钠)中在室温下水合10分钟。

实施例5-抗体结合亲和力
通常如前所述进行ForteBio亲和力测量(参见,例如,Estep et al., 2013)。简而言之,通过将IgG在线加载到AHQ传感器上来进行ForteBio亲和力测量。将传感器在测定缓冲液中离线平衡30分钟,然后在线监测60秒以进行基线建立。将装载有IgG的传感器暴露于100 nM抗原5分钟,然后将它们转移到测定缓冲液中5分钟以进行解离速率测量。使用1:1结合模型来分析动力学。

实施例6-抗PD-L1抗体的ADCC活性
进行报道分子生物测定从而确定本公开的抗PD-L1抗体的抗体依赖性细胞介导细胞毒性(ADCC)。该测定使用SUDHL-1淋巴瘤细胞和供体PBMC。以1或3 ug/ml的浓度测试各种抗体。
对于图3中的示例性抗-PD-L1抗体(CTI-48)显示了该研究的结果,其由与本公开的抗体孵育4小时后的细胞毒性百分比表示。
实施例7-免疫封锁报道分子测定
在96孔板中使用PD1/PD-L1封锁测定试剂盒(Promega,CS187111)进行免疫封锁测定。在分析中发生三个主要事件。事件1:当通过TCR/TCR激活剂相互作用使工程化的JurkatPD-1效应细胞和aAPC(人工抗原递呈细胞)PD-L1细胞参与时,发生TCR介导的NFAT活化。事件2:当没有封锁抗体存在时,PDAT信号的抑制通过PD-1:PD-L1连接。事件3:通过添加抗PD-1或抗PD-L1封锁抗体回收NFAT信号。
在测定前一天,通过向22.5 mL F-12中加入2.5 mL FBS,在50 mL锥形管中制备25mL细胞回收培养基(10%FBS F-12)用于解冻和使用的PD-L1细胞。从冷冻储存器中取出一小瓶解冻和使用的PD-L1细胞(CS187103)并在干冰上转移到工作台。将病毒在37℃水浴中解冻,直至细胞解冻(约3-4分钟)。通过上下吸移以将细胞悬浮液在小瓶中温和混合,并将所有细胞(0.5 mL)转移至含有14.5 mL细胞回收培养基的标记为“PD-L1细胞”的管中,然后温和倒置。将细胞悬浮液转移至无菌试剂储蓄库。立即使用多通道移液管,每孔加入100 μl细胞回收培养基至测定板的外部孔。将板在CO2培养箱中于37℃温育过夜。
在测定当天,制备新鲜的测定缓冲液(RPMI 1640 + 1% FBS),并在测定缓冲液中对于每种测试抗体以2X的最终浓度制备七点三倍连续的稀释液。从测定板上的所有孔中取出95 μl培养基,并将40 μl测试抗体的连续的稀释液加入到含有PD-L1细胞的孔中。将每孔80 μl测定缓冲液加入每个板的外部孔中。
将解冻和使用的PD-1效应细胞(CS187105)转移到测定板中,并将板在37℃下、在CO2培养箱中温育6小时。诱导6小时后,将测定板从CO2培养箱中取出并在环境温度下平衡5-10分钟。向每个测试孔中加入80 μl的Bio-GloTM试剂,并将板在环境温度下再孵育5-10分钟。在POLARstar Omega读板仪中以0.5秒积分测量发光。
测试了以下抗体:CTI-2、临床对照mAb、CTI-09、CTI-48、CTI-50、CTI-07和CTI-58,终浓度为10 μg/mL、3.33 μg/mL、1.11 μg/mL、0.37 μg/mL、0.123 μg/mL、0.041 μg/mL和0.014 μg/mL。
包括临床对照mAb、CTI-48和CTI-49的示例性抗体的结果显示在下表6中。

实施例8-PD-L1/B7.1抑制剂筛选测定
使用市售的可用的测定试剂盒,筛选和分析本公开的抗体的相互作用以及PD-L1/B7.1相互作用。该试剂盒以96孔形式存在,具有生物素标记的B7-1(CD80)、纯化的PD-L1、链霉亲和素标记的HRP、以及用于100个结合反应的测定缓冲液。该试剂盒用于通过链霉亲和素-HRP检测生物素标记的B7.1。
首先,将PD-L1涂覆在96孔板上。接下来,向每个孔中加入本公开的抗体之一、阳性对照、底物对照或空白,并在温育后加入B7.1-生物素。最后,用链霉亲和素-HRP处理平板,然后加入HRP底物以产生化学发光,然后可以使用化学发光读数器测量。
测试了以下抗体在30 μg/mL、10 μg/mL、3.33 μg/mL、1.11 μg/mL、0.37 μg/mL和0.123 μg/mL的浓度之下对PD-L1和B7.1的结合的抑制作用:CTI-1、CTI-2、CTI-33、CTI-48和CTI-55。
示例性结果表明,本公开的抗体的结合抑制的IC50在0.1816和0.5056 μg/mL之间。例如,计算得CTI-48的IC50为0.1816 μg/mL。CTI-48和临床对照mAb的活性的比较显示在图5中。
实施例9-抗体对IFN-γ生产的影响
将抗体给予进混合的淋巴细胞反应(MLR)培养物,从而确定本公开的抗体对IFN-γ生产的影响。在用浓度为10 μg/mL的抗体进行4天MLR培养后,测定IFN-γ生产的倍数变化。示例性结果,包括适当的同种型对照(hIgG1)的结果。如图6所示,CTI-48诱导了对临床对照mAb的可比较的反应,并且测试的抗体中的许多引起IFN-γ生产的统计学上的显著增加,包括CTI-33和CTI-55比对照水平增加约10倍。
