Abstract
Description
葉酸受容体1(FOLR1)としても知られる葉酸受容体アルファは、葉酸受容体ファミリーに属し、そのメンバーは、葉酸および/またはその誘導体(例として、5−メチルエトラヒドロ葉酸)に対して高い結合親和性を有する。FOLR1は、卵巣、乳房、腎臓、肺、結腸直腸、および脳など、数多の上皮由来腫瘍で過剰に発現していることが報告されている。したがって、この受容体は、かかる上皮由来腫瘍の処置の標的となり得る。
したがって、がんの処置および診断の両方で使用するための抗FOLR1抗体などの有効なFOLR1アンタゴニストを開発することは非常に重要である。
本開示は、少なくとも部分的に、FOLR1に特異的な数多の抗体の開発に基づいている。かかる抗体は、標的FOLR1抗原に対する高い結合親和性および/またはFOLR1+細胞に対する高い阻害活性を示した。

標的FOLR1抗原への高い結合親和性、および/またはFOLR1+細胞に対する高い阻害活性を含む優れた特徴を示した数多の抗FOLR1抗体が本明細書に開示される。
結果的に、FOLR1に結合可能な抗体、それをコードする核酸、抗体−薬物抱合体(ADC)および抗FOLR1抗体を含むキメラ抗原受容体(CAR)、および治療および診断目的の両方のためのそれらの使用が本明細書に提供される。また、抗体および/またはADCおよびそれを含むCARの治療的および/または診断的使用のためのキット、ならびに抗FOLR1抗体を生産するための方法も本明細書に提供される。

本明細書に開示の方法を実行するために、本明細書に記載の有効量の医薬組成物は、例として、筋肉内、腹腔内、脳脊髄内、皮下、関節内、滑液嚢内、髄腔内、経口、吸入または局所ルートによる、ボーラスとして、またはある期間にわたる連続的な注入によって、静脈内投与などの好適なルートを介して処置を必要とする対象(例として、ヒト)に投与され得る。ジェット噴霧器および超音波噴霧器を含む、液体製剤用の市販の噴霧器は、投与に有用である。液体製剤は直接噴霧することができ、凍結乾燥粉末は再構成後に噴霧することができる。代替的に、本明細書に記載の抗体は、フッ化炭素製剤および定量吸入器を使用してエアロゾル化するか、または凍結乾燥および製粉された粉末として吸入することができる。
一例において、本明細書に記載の抗体の投薬量は、抗体の1以上の投与(単数または複数)を受けた個体において経験的に決定され得る。個体には、アンタゴニストの漸増投薬量が与えられる。アンタゴニストの有効性を評価するために、疾患/障害の指標を追跡することができる。

FOLR1を標的とするCARを発現する免疫細胞を疾患の処置に使用するとき、患者は、Tリンパ球またはNK細胞などの治療的に有効な用量のかかる免疫細胞を体重の1キログラムあたり約105〜1010個(細胞/Kg)またはそれ以上の範囲で注入することによって処置できる。注入は、所望の応答が達成されるまで、患者が耐えることができる限り何度でも繰り返すことができる。適切な注入量およびスケジュールは患者ごとに異なるが、特定の患者の処置を行う医者によって決定され得る。典型的には、およそ106細胞/Kgの初期用量が注入され、108細胞/Kg以上にまで増加する。IL−2は、注入された細胞を拡大するために同時投与され得る。IL−2の量は、体表面積1平方メートルあたり約1〜5x106国際単位であり得る。

本明細書に記載の抗FOLR1抗体のいずれも、試料中のFOLR1+細胞の存在またはレベルを検出するために使用することができる。かかる診断アッセイは、in vitroまたはin vivoで実施され得る。

例1:抗FOLR1抗体の生成
試薬および一般的な方法
ハイブリドーマ細胞培養培地(PFHM-II Protein-Free Hybridoma Medium; #12040077)をThermo Fisherから購入した。
RNAは、標準的なプロトコルおよびThermo FisherのTRIzol Reagent(#15596018)を使用して単離した。得られたcDNA分子は、TakaraのcDNA合成キット(PrimeScript II 1st strand cDNA synthesis kit; #6210A)を使用して生成した。TakaraのPremix Taq(#RR901A)を使用して、抗体V領域の増幅を実施した。標準的なPCRプライマーセット(Ig-Primer Sets #TB326)はNovagenから得た。遺伝子は、EcoRI、HindIII、SalI、およびT4リガーゼ(すべてNEBのもの)の使用を含む標準的な技法を使用して、pET28a(Novagen; #69864)にクローン化した。QIAEX II Gel Extraction Kit(QIAgen #20021)を利用して、すべてではないが一部のオリゴヌクレオチド分子を精製した。
SK−OV−3およびDaudi細胞培養は、5%CO2の雰囲気において37℃で単層培養としてin vitroで維持した。必要に応じて、腫瘍細胞を定期的に継代した。
US2015/0153356において以前に記載されているように、モノクローナル抗体の大規模なライブラリー(合計>100,000)をプロテオームおよびペプチド抗原の混合物を使用して生成した。がん腫瘍試料およびFDA Normal Tissue Panelに対してスクリーニングする前に、ライブラリーを一連の高密度抗体アレイに分割した。
個々のハイブリドーマクローンを、10mLハイブリドーマ細胞培養培地(PFHM-II Protein-Free Hybridoma Medium)を有するT25フラスコにおいて培養した。細胞を37℃で80%コンフルエントになるまで増殖させた。次いで培養培地を除去し、細胞を1xPBSで2回洗浄した。TRIzol試薬(1mL体積)をフラスコに直接加え、ピペット混合により細胞を溶解した。次いで、細胞ライセートをT25フラスコから回収し、標準的な方法を使用して全RNAを単離した。続いてRNA濃度をNanodrop 2000(Thermo Fisher)で測定した。次いで、Takara PrimeScript II 1st strand cDNA synthesis kitプロトコルに従って、単離したRNAからcDNA鎖を生成した。次いで、得られたcDNAのハイブリドーマV領域の増幅を、NovagenユーザープロトコルTB326に従って実施した。以下の表3に示すようなプライマーペアを増幅に使用した。
FOLR1を過剰発現するSK−OV−3細胞(FOLR1+SK−OV−3)およびFOLR1の発現が陰性のDaudi細胞(FOLR1−Daudi)を、トリプシン−EDTA部分消化とそれに続く1000rpmで5分間の遠心分離を使用して回収した。細胞を冷PBSに再懸濁し、等分した。抗FOLR1抗体をPBSで希釈し、FOLR1+SK−OV−3細胞またはFOLR1−Daudi細胞のいずれかに加えた。細胞溶液を混合し、暗所で4℃でインキュベートし、PBSで洗浄した後、二次抗体抱合体を加えた(検出目的のため)。インキュベーション後、細胞をPBSで洗浄し、固定液で固定し、次いでFACS分析に供した。図1A〜1Eに示されるように、これらの抗体は、FOLR1+SK−OV−3への飽和性結合を示した。これらの抗体は、FOLR1−Daudiへの飽和性結合を示さなかった。

Claims (51)
- FOLR1に結合する単離された抗体であって、参照抗体と同じヒトFOLR1のエピトープに結合し、FOLR1−Ab1、FOLR1−Ab4、FOLR1−Ab14、FOLR1−Ab20、およびFOLR1−Ab23からなる群から選択される、前記抗体。
- 以下:
(a)X1YTFTX2YX3として示される重鎖相補性決定領域1(HC CDR1)、ここで、X1は、GまたはIであり、X2は、DまたはSであり、およびX3は、W、N、またはSである;
(b)INX1X2X3X4X5X6として示される重鎖相補性決定領域2(HC CDR2)、ここで、X1は、PまたはTであり、X2は、N、Y、またはEであり、X3は、N、D、またはTであり、X4は、GまたはSであり、X5は、GまたはEであり、およびX6は、TまたはPである;および
(c)ARX1X2X3YX4X5X6X7X8X9X10として示される重鎖相補性決定領域3(HC CDR3)、ここで、X1は、S、KまたはMであり、X2は、GまたはPであり、X3は、GまたはYであり、X4は、Gまたは不在であり、X5は、Pまたは不在であり、X6は、A、R、またはKであり、X7は、W、YまたはIであり、X8は、FまたはMであり、X9は、DまたはAであり、およびX10は、YまたはVである、
の1以上を含む重鎖可変領域(VH)を含む、請求項1に記載の単離された抗体。 - HC CDR1、HC CDR2、およびHC CDR3が、集合的に、参照抗体のHC CDR1、HC CDR2、およびHC CDR3と少なくとも85%同一であるVHを含む、請求項1または2に記載の単離された抗体。
- VHが、参照抗体と同じHC CDR1、HC CDR2、およびHC CDR3を含む、請求項3に記載の単離された抗体。
- VHが、参照抗体のHC CDR1、HC CDR2、およびHC CDR3に対して相対的な5、4、3、2、または1までの変異を集合的に含む、HC CDR1、HC CDR2、およびHC CDR3を含む、請求項3に記載の単離された抗体。
- 以下:
(a)ESVDNYGISFまたはQSLLYSSSQKNYとして示される軽鎖相補性決定領域1(LC CDR1);
(b)X1ASとして示される軽鎖相補性決定領域2(LC CDR2)、ここで、X1は、V、A、またはWである;および
(c)QQX1X2X3X4PX5Tとして示される軽鎖相補性決定領域3(LC CDR3)、ここで、X1は、YまたはSであり、X2は、YまたはKであり、X3は、EまたはSであり、X4は、YまたはVであり、およびX5は、W、Y、または不在である、
の1以上を含む軽鎖可変領域(VL)を含む、請求項1〜5のいずれか一項に記載の単離された抗体。 - LC CDR1、LC CDR2、およびLC CDR3が、集合的に、参照抗体のLC CDR1、LC CDR2、およびLC CDR3と少なくとも85%同一であるVLを含む、請求項1〜6のいずれか一項に記載の単離された抗体。
- VLが、参照抗体と同じLC CDR1、LC CDR2、およびLC CDR3を含む、請求項7に記載の単離された抗体。
- VLが、参照抗体のLC CDR1、LC CDR2、およびLC CDR3に対して相対的な5、4、3、2、または1までの変異を集合的に含む、LC CDR1、LC CDR2、およびLC CDR3を含む、請求項7に記載の単離された抗体。
- ヒトFOLR1に特異的に結合する、請求項1〜9のいずれか一項に記載の抗体。
- ヒトFOLR1および非ヒトFOLR1と交差反応する、請求項1〜9のいずれか一項に記載の抗体。
- 非ヒトFOLR1が、齧歯類の動物のFOLR1または霊長目の動物のFOLR1である、請求項11に記載の抗体。
- 全長抗体またはその抗原結合フラグメント、または単鎖抗体である、請求項1〜12のいずれか一項に記載の抗体。
- IgG分子である、請求項13に記載の抗体。

ii.少なくとも1の治療剤;
を含み、ここで、前記抗体は、少なくとも1の治療剤に共有結合的に抱合されている、前記抗体−薬物抱合体。 - 抗体および治療剤が、リンカーを通じて抱合されている、請求項21に記載の抗体−薬物抱合体。
- リンカーが、切断可能なまたは切断不可能なリンカーである、請求項21に記載の抗体−薬物抱合体。
- 切断可能なリンカーが、プロテアーゼ感受性リンカー、pH感受性リンカー、またはグルタチオン感受性リンカーである、請求項23に記載の抗体−薬物抱合体。
- リンカーが、2〜5アミノ酸のペプチド配列を含むプロテアーゼ感受性リンカーである、請求項24に記載の抗体−薬物抱合体。
- 2〜5アミノ酸が、天然に存在するアミノ酸残基、天然に存在しないアミノ酸残基、またはそれらの組み合わせを含む、請求項25に記載の抗体−薬物抱合体。
- ペプチド配列が、バリン−シトルリンを含む、請求項25に記載の抗体−薬物抱合体。
- リンカーが、任意に置換されていてもよいアルカンまたはチオエーテルを含む切断不可能なリンカーである、請求項23に記載の抗体−薬物抱合体。

(i)FOLR1に結合する抗原結合フラグメントを含む細胞外ドメイン、
(ii)膜貫通ドメイン、および
(iii)1以上の細胞内刺激ドメイン
を含む、キメラ抗原受容体。 - 抗原結合フラグメントが、FOLR1−Ab217、FOLR1−Ab218、FOLR1−Ab220、FOLR1−Ab221、およびFOLR1−Ab222からなる群から選択される参照抗体と同じヒトFOLR1のエピトープに結合する、請求項29に記載のキメラ抗原受容体。
- 抗原結合フラグメントが、請求項2〜9のいずれか一項に記載のHC CDRおよび/またはLC CDRを含む、請求項35に記載のキメラ抗原受容体。
- 抗原結合フラグメントが、参照抗体と同じVHおよびVLを含む、請求項35に記載のキメラ抗原受容体。
- 抗原結合フラグメントが、単鎖抗体(scFv)である、請求項34〜37のいずれか一項に記載のキメラ抗原受容体。
- 膜貫通ドメインが、CD28またはCD8受容体に由来する膜貫通ドメインを含む、請求項33〜38のいずれか一項に記載のキメラ抗原受容体。
- (iii)が、CD3ζからのシグナル伝達ドメインを含む、請求項33〜39のいずれか一項に記載のキメラ抗原受容体。
- (iii)が、共刺激シグナル伝達ドメインを含む、請求項33〜40のいずれか一項に記載のキメラ抗原受容体。
- 共刺激シグナル伝達ドメインが、4−1BB、CD7、CD27、CD28、CD40、OX40、ICOS、GITR、HVEM、TIM1、またはLFA−1受容体からのものである、請求項41に記載のキメラ抗原受容体。
- 請求項33〜42のいずれか一項に記載のキメラ抗原受容体をコードするヌクレオチド配列を含む、核酸。
- 請求項43に記載の核酸を含む、ベクター。
- 請求項33〜42のいずれか一項に記載のキメラ受容体を発現する、宿主細胞。
- 免疫細胞である、請求項45に記載の宿主細胞。
- 免疫細胞が、T細胞である、請求項46に記載の宿主細胞。
- (i)請求項1〜16のいずれか一項に記載の抗体、請求項17〜19のいずれか一項に記載の核酸または核酸のセット、請求項21〜33のいずれか一項に記載の抗体−薬物抱合体、または請求項46または47に記載の宿主細胞;および(ii)薬学的に許容し得る担体を含む、医薬組成物。
- 請求項48に記載の医薬組成物の有効量をそれを必要とする対象へ投与することを含む、FOLR1+細胞の数を減少させる方法。
- 対象が、がんを有するか、またはがんを有すると疑われる、請求項49に記載の方法。
- FOLR1+細胞の存在を検出する方法であって、以下:
i.FOLR1+細胞を有すると疑われる試料を請求項1〜16のいずれか一項に記載の抗体と接触させること、抗体は、標識化剤と抱合されている;および
ii.試料中のFOLR1+細胞の存在を試料中の細胞に対する抗体の結合に基づき検出すること
を含む、前記方法。
