Abstract
本发明涉及一种开音利咽的中药制剂口含滴丸的制备方法，所述方法，步骤如下：a.将薄荷、连翘、川芎采用超临界萃取法提取挥发油；b.将药材桔梗、蝉蜕、甘草、胖大海采用水提法，提取并浓缩成清膏；C.将薄荷脑用乙醇溶解后与a中提取的挥发油混合、均制；加入b中提取的清膏，再加入聚乙二醇6000，混合均匀；d.将步骤c所得混合物加入滴丸机中，在60℃下保温15分钟，滴入冷凝剂中，收集成型的滴丸，脱油，即得成品滴丸。

技术领域：
本发明涉及一种开音利咽的中药制剂口含滴丸的制备方法。
背景技术：
急慢性咽喉炎是常见多发疾病，西医治疗咽喉炎主要以片剂内服及针剂注射消炎，这些治疗方法对咽喉局部治疗效果差。
中草药治疗咽喉炎用的含片及喷雾，其疗效缓慢，用药量大，治疗时间长。
黄氏响声丸具有疏风清热，化痰散结，利咽开音之功效。目前市场上主要有丸剂、含片、茶剂。其中药配方为：胖大海、桔梗、蝉蜕、甘草、连翘、大黄、川芎、诃子肉、浙贝母、薄荷、儿茶11味药材，有关制备方法公开在以下中国专利中
现有黄氏响声丸药材种类多，用现有方法制备的活性提取物，其中含有纤维成分，口服尚可，但口含则有不适感，本发明为制备一种口含制剂，特别是口含滴丸制剂，对现有黄氏响声丸的配方和制备方法进行了改进。
本发明采用超临界萃取法提取配方中部分药材的挥发油成分；同时去除掉部分会引起口含不适的中药材如：蝉蜕、大黄、诃子肉、浙贝母、儿茶，意外的发现，其治疗效果和现有技术相同，本发明将其制备成口含滴丸，其中挥发油成分含量达50％以上，吸收迅速，可直达患处快速起效，对失音症状尤为明显。
发明内容：
本发明公开了一种开音利咽的中药制剂口含滴丸的制备方法如下：
a.按配比称取薄荷、连翘、川芎、桔梗、甘草、蝉蜕、胖大海、薄荷脑，备用；将配比的薄荷、连翘、川芎，混合，粉碎成60目的颗粒，CO2超临界萃取，在萃取压力35MPa，萃取温度60℃，CO2流速35L/h条件下萃取3小时，收集挥发油；
b.将桔梗、甘草、蝉蜕、胖大海加水6倍/次，煎煮2次，每次 1.5小时，合并煎液，过滤，浓缩至相对密度1.35的清膏。
c.将薄荷脑用3倍量乙醇溶解，与a中收集的挥发油混合，均质；加入到b中的清膏里，再加入2.5倍量聚乙二醇6000混合，搅拌均匀；
d.将步骤c所得混合物转移至滴丸机内，在65～75℃下保温15分钟，调整滴头到冷凝剂液面距离7-10cm，用60滴/分钟速度滴入冷凝剂12-18℃液体石蜡中，收集成型滴丸，脱油，得成品滴丸。
本发明发现，采用本发明的方法制备的挥发油，收率远大于现有技术，以下通过实验数据进一步说明本发明的有益效果：
现有传统水蒸气提取挥发油方法：称取取薄荷500g、连翘500g、川芎500g，混合，粉碎，加10倍量水，提取6小时，收集挥发油，得到78ml挥发油。
本发明采用CO2超临界萃取法：称取薄荷500g、连翘500g、川芎500g，混合，粉碎成60目的颗粒，CO2超临界萃取，在萃取压力 35MPa，萃取温度60℃，CO2流速35L/h条件下萃取3小时，收集挥发油，得到110ml挥发油，以本方法提取的挥发油量比原有技术提高了40％。
本发明口含滴丸每日、每次服用量少，起效快，直达患处，对改善急性咽喉炎、失声症状有明显效果。
具体实施方式
以下通过实施例进一步说明本发明，但不作为对本发明的限制。
实施例1：
按比例称取薄荷20份、连翘20份、川芎20份，混合，粉碎成 60目的颗粒，CO2超临界萃取，在萃取压力35MPa，萃取温度60℃， CO2流速35L/h条件下萃取3小时，收集挥发油240ml；将桔梗10份、甘草10份、蝉蜕10份、胖大海10份，加水6倍/次，煎煮2次，每次1.5小时，合并煎液，过滤，浓缩至相对密度1.35的清膏。将薄荷脑20份，用3倍量乙醇溶解，提取的挥发油混合，均质；加入到浓缩好的清膏里，再加入8倍量的聚乙二醇6000混合，搅拌均匀；混合物转移至滴丸机内，在65～75℃下保温15分钟，滴入冷凝剂中，收集成型滴丸，脱油，得成品滴丸。
实施例2：
按比例称取薄荷40份、连翘40份、川芎40份，混合，粉碎成 60目的颗粒，CO2超临界萃取，在萃取压力35MPa，萃取温度60℃，CO2流速35L/h条件下萃取3小时，收集挥发油500ml；将桔梗20份、甘草20份、蝉蜕20份、胖大海20份，加水6倍/次，煎煮2次，每次1.5小时，合并煎液，过滤，浓缩至相对密度1.35的清膏。将薄荷脑40份，用3倍量乙醇溶解，提取的挥发油混合，均质；加入到浓缩好的清膏里，再加入8倍量的聚乙二醇6000混合，搅拌均匀；混合物转移至滴丸机内，在65～75℃下保温15分钟，滴入冷凝剂中，收集成型滴丸，脱油，得成品滴丸。

按比例称取薄荷80份、连翘80份、川芎80份，混合，粉碎成 60目的颗粒，CO2超临界萃取，在萃取压力35MPa，萃取温度60℃， CO2流速35L/h条件下萃取3小时，收集挥发油980ml；将桔梗40份、甘草40份、蝉蜕40份、胖大海40份，加水6倍/次，煎煮2次，每次1.5小时，合并煎液，过滤，浓缩至相对密度1.35的清膏。将薄荷脑80份，用3倍量乙醇溶解，提取的挥发油混合，均质；加入到浓缩好的清膏里，再加入8倍量的聚乙二醇6000混合，搅拌均匀；混合物转移至滴丸机内，在65～75℃下保温15分钟，滴入冷凝剂中，收集成型滴丸，脱油，得成品滴丸。

1.一种开音利咽的中药制剂口含滴丸的制备方法，其特征在于，所述方法，步骤如下：
a.按配比称取薄荷、连翘、川芎、桔梗、甘草、蝉蜕、胖大海、薄荷脑，备用；将配比的薄荷、连翘、川芎，混合，粉碎成40～80目的颗粒，CO2超临界萃取，在萃取压力25～45MPa，萃取温度45～65℃，CO2流速25～45L/h条件下萃取1～6小时，收集挥发油；
b.将桔梗、甘草、蝉蜕、胖大海加水2～6倍/次，煎煮2次，每次1.5小时，合并煎液，过滤，浓缩至相对密度1.2～1.4的清膏。
c.将薄荷脑用2～4倍量乙醇溶解，与a中收集的挥发油混合，均质；加入到b中的清膏里，再加入聚乙二醇6000混合，搅拌均匀；

3.根据权利要求1所述的方法，其特征在于，将桔梗、甘草切片，与蝉蜕、胖大海混合后，加水6倍/次，煎煮2次，每次1.5小时，合并煎液，用200目纱绢过滤，浓缩至相对密度1.35的清膏冷却备用。
4.根据权利要求1所述的方法，其特征在于，其中，薄荷脑用3倍量乙醇溶解，与a中收集的挥发油混合，均质机高速15分钟、再低速20分钟；加入到b中的清膏里，再加入8倍量熔融的聚乙二醇6000混合，搅拌均匀。
5.根据权利要求1所述的方法，其特征在于，其中，将步骤c所得混合物转移至滴丸机内，在65～75℃下保温15分钟，适当加压，滴入冷凝剂液体石蜡中，收集成型滴丸，脱油，得成品滴丸。
