Abstract

基因治疗方法经常受到重组病毒载体对靶细胞的低转导效率的阻碍。逆转录病毒载体,特别是基于人免疫缺陷病毒1(HIV-1)的慢病毒载体(LV)是用于基因疗法的有希望的介质。这些载体目前在临床应用中用于治疗各种疾病,如免疫缺陷、神经退行性疾病或神经系统疾病、贫血或HIV感染。
逆转录病毒载体的一些应用依赖于特异性靶细胞的离体转导,例如表达CD34标志物的淋巴细胞或造血干/祖细胞。LV用于转导T淋巴细胞以产生长寿命的CAR-T细胞,用于治疗癌症,例如难治性慢性淋巴细胞白血病(CLL)(Porter等,Science Transl.Med.7:303ra139 2015)。LV用于转导CD34+细胞用于遗传起源的各种免疫、血液、代谢或神经退行性疾病的自体基因疗法(Wagemaker G.HumGene Ther.2014Oct;25(10):862-5.doi:10.1089/hum.2014)。慢病毒载体是通用工具,其可以用各种病毒包膜糖蛋白假型化,从而赋予颗粒特定的细胞趋向性特征(Buchholz等Trends Biotechnol.2015Dec;33(12):777-90.doi:10.1016/j.tibtech.2015.09.008)。
使用重组慢病毒颗粒的一个限制因素是在重组慢病毒颗粒的生产过程中获得高感染性滴度的能力。避免这种限制的一种方法是浓缩纯化步骤中的病毒上清液。然而,取决于用于假型化病毒颗粒的包膜糖蛋白,对于一些LV难以建立纯化方案。因此,许多慢病毒载体制剂具有低滴度且转导效力有限。
另一个限制因素是难以转导原代细胞,因此它们必须常常被活化以被转导,从而失去其初始状态。
最后,另一个限制因素是慢病毒载体本身感染靶细胞的能力。几种包膜糖蛋白如VSV-G或RD114TR可用于假型化慢病毒载体,并且对靶细胞如CD34+细胞和原代血细胞具有可变的感染性。
合胞素是具有膜融合特性的内源性逆转录病毒(ERV合胞素)包膜糖蛋白(Dupressoir等,2005;Lavialle等,2013)。
专利申请EP2385058已经描述了由ERVW-1基因(ENSG00000242950;也称为合胞素-1或HERV-W)编码的人内源性逆转录病毒包膜糖蛋白的膜融合特性。所述申请描述了其通过形成合胞体在癌症治疗中的用途。
一些研究表明用HERV-W包膜假型化HIV-1病毒粒子(而不是MLV颗粒)的可能性,以获得能够转导293T细胞的感染性病毒。然而,这样的HERV-W颗粒随着浓度不稳定,冷冻和解冻大幅降低了滴度(An等,Journal of Virology,Apr 2001,p.3488-3489)。使用R胞质内区域的缺失来增加HIV-1衍生的基因转移载体的融合和假型滴度(Lavillette等2002)。使用由ERVFRD-1基因编码的糖蛋白(ENSG00000244476;也称为合胞素-2或HERV-FRD)来假型化SIV载体,据报道其作为HIV或MLV假型,但是在非常低的滴度下排除功能研究,并且对于HERV-FRD HIV假型,可获得的信息很少(Blaise等,Journal of Virology,Jan2004,p.1050-1054)。
因此需要提高病毒或病毒载体,特别是稳定的病毒或病毒载体的转导效率的方法,例如用于改进基因向特定靶细胞的递送。具体地,需要一种病毒或病毒载体,它们是稳定的,因此可以以工业规模获得。还需要一种稳定的载体,其可以在体外或体内作为呈现新生物学特性的工具而使用。
还需要一种用病毒或病毒载体选择性转导细胞的方法,以便在体外或体内仅靶向特定细胞。这种病毒或病毒载体必须是完全耐受的,对靶细胞具有特异性,并且适合多次施用。
令人惊讶的,本发明人已经详细描述了获得用特定包膜糖蛋白假型化的慢病毒颗粒的方法,其是稳定的并且可以是冷冻的。所述慢病毒假型化颗粒还改进所选靶细胞,特别是免疫细胞的转导;这扩大了所述颗粒的治疗范围。所述假型化慢病毒颗粒确实有效转导免疫细胞,特别是B细胞、T细胞和树突细胞,并允许功能性校正细胞缺陷。此外,这些慢病毒假型化载体可在体内施用,在脾脏和骨髓中产生可检测的、稳定的和良好耐受的基因转移,其中证据是CD19+脾B细胞转导。
因此,使用所述假型化慢病毒颗粒对于患有胎盘功能障碍、癌症、感染性疾病、免疫缺陷、自身免疫的患者或对于基因疗法或作为疫苗或作为生物技术工程工具是有前景的策略。
发明简述
本发明涉及获得包含异源目标基因的稳定的假型化慢病毒颗粒的方法,包括以下步骤:
a)在合适的细胞系中转染至少一种质粒,其中所述至少一种质粒包含所述异源目标基因、逆转录病毒rev、gag和pol基因和编码ERV合胞素的核酸;
b)孵育a)中获得的转染细胞,使得它们产生分别用ERV合胞素假型化且包装有所述异源目标基因的稳定的慢病毒颗粒;和
c)收获并浓缩b)中获得的稳定的慢病毒颗粒。
根据本发明的方法允许获得高物理滴度以及高感染性滴度的包含目标异源基因的稳定的假型化慢病毒颗粒。
还提供用ERV合胞素假型化且包装有异源目标基因的稳定的慢病毒颗粒。它们通过上述段落中描述的方法获得或可获得。所述稳定的假型化慢病毒颗粒可用作药物。具体地,所述稳定的假型化慢病毒颗粒可用于基因疗法或免疫疗法,尤其是用作疫苗或用于免疫预防。
本发明还涉及用ERV合胞素假型化且包括异源目标基因的慢病毒颗粒,优选用ERV合胞素假型化且包括异源目标基因的稳定的慢病毒颗粒,通过转导细胞用于治疗的用途,所述细胞表达同源受体,例如分别用于合胞素-1(HERV-W)和合胞素-2(HERV-FRD)的ASCT2、ASCT1或MFSD2a,如人绒毛膜癌细胞、人上皮细胞或人免疫细胞如B细胞、T细胞或CD11c+细胞如髓样细胞(例如:粒细胞、单核细胞及其祖细胞)。本发明还涉及用ERV合胞素假型化的慢病毒颗粒用于有效转导鼠源B细胞和T细胞的能力。
本发明还涉及用ERV合胞素假型化且包括目标基因的慢病毒颗粒,优选用ERV合胞素假型化且包括异源目标基因的稳定的慢病毒颗粒,用于治疗免疫缺陷、自身免疫、感染性疾病、胎盘功能障碍(例如先兆子痫)、涉及血脑屏障的疾病,或包括B细胞相关癌症的癌症,或在小鼠中制造这些疾病模型的用途。
用ERV合胞素假型化且包括异源目标基因的稳定的慢病毒颗粒也可用于体外转导免疫细胞,优选B细胞或CD11c+细胞如髓样细胞(例如粒细胞、单核细胞及其祖细胞)。免疫细胞,优选B细胞的转导可以应用于生物技术工程,例如用于生产免疫球蛋白,或用于免疫细胞,优选B细胞工程。
本发明还涉及用于获得经修饰以表达异源目标基因的免疫细胞的离体方法,包括以用ERV合胞素假型化且包括异源目标基因的慢病毒颗粒,优选以用ERV合胞素假型化且包括异源目标基因的稳定的慢病毒颗粒,优选在LAH4肽或其功能性衍生物存在下感染免疫细胞的步骤。
本发明还涉及用ERV合胞素假型化且包括异源目标基因的慢病毒颗粒,优选用ERV合胞素假型化且包括异源目标基因的稳定的慢病毒颗粒用于生物技术工程的用途。特别地,它们可用于制备细胞系(优选B细胞系)以产生特定产物(例如免疫球蛋白)。本发明的稳定颗粒也可用于制备研究模型,特别是在小鼠中(以获得鼠模型)。这可用于探索疾病机制,特别是B细胞疾病或B细胞介导的疾病,制备表达产物用于接种目的的树突细胞和/或髓样细胞,或产生具有特定治疗效果的产品。
根据本发明,ERV合胞素选自HERV-W、HERV-FRD、鼠合胞素-A、鼠合胞素-B、合胞素-Ory1、合胞素-Car1和合胞素-Rum1及其功能性直系同源物,优选ERV合胞素选自HERV-W、HERV-FRD和鼠合胞素-A,甚至更优选ERV合胞素是HERV-W或HERV-FRD。
发明详述
发明人惊讶地发现,具有高促融合特性的ERV合胞素,例如人合胞素-1(HERV-W)、人合胞体素-2(HERV-FRD)和鼠合胞素A,可用于获得重组HIV-1衍生的慢病毒的假型。如在实施例中所清楚证明,结果显示可以产生包含目标基因的稳定的感染性慢病毒颗粒,其对细胞系具有选择性趋向性。令人惊讶的是,合胞素-假型化的慢病毒颗粒能够有效转导人原代B淋巴细胞和CD11c+髓样和/或树突细胞,特别是在Vectofusin-1(一种阳离子转导添加剂)存在下。
因此,发明人已经详细描述了获得用ERV合胞素假型化的慢病毒颗粒的方法,所述慢病毒颗粒是稳定的,具有感染性的并且可以冷冻。慢病毒假型化颗粒改进所选靶细胞,特别是免疫细胞,特别是B细胞、T细胞和树突细胞的转导。
本发明的详细实施方案如下所述。
本发明涉及获得包含异源目标基因的稳定的假型化慢病毒颗粒的方法,包括以下步骤:
a)在合适的细胞系中转染至少一种质粒,其中所述至少一种质粒包含所述异源目标基因、逆转录病毒rev、gag和pol基因和编码ERV合胞素的核酸;
b)孵育a)中获得的转染细胞,使得它们产生分别用ERV合胞素假型化且包装有所述异源目标基因的稳定的慢病毒颗粒;和
c)收获并浓缩b)中获得的稳定的慢病毒颗粒。
优选地,根据本发明的步骤c)包括从上清液收获、浓缩和/或纯化步骤b)中产生的稳定的慢病毒颗粒。因此,优选地,步骤c)的浓缩包括离心和/或纯化步骤b)中获得的收获的稳定的慢病毒颗粒。所述收获可以根据本领域熟知的方法进行。优选地,在与转染细胞融合之前,更优选转染后20-72小时,优选24小时后收获慢病毒载体。优选地,收获步骤是单次慢病毒收获,优选在转染后20-72小时,优选转染后20-30小时,更优选24小时后进行。
根据本发明的ERV合胞素是指来自真哺乳亚纲哺乳动物的高度促融合的包膜糖蛋白,其属于内源性逆转录病毒(ERV)家族。这些蛋白质由在胎盘中显示优先表达并在引入培养细胞时诱导形成合胞体的基因编码(Lavialle等,2013)。
根据本发明的ERV合胞素可以选自人合胞素(例如:HERV-W和HERV-FRD)、鼠合胞素(例如:合胞素-A和合胞素-B)、合胞素-Ory1、合胞素-Car1、合胞素-Rum1或它们的功能性直系同源物(Dupressoir等,2005;Lavialle等,2013)。
功能性直系同源物意欲是指由直系同源基因编码的并且表现出促融合特性的直系同源蛋白。如Dupressoir等(PNAS 2005)所述,可以在融合测定中评估促融合特性。简言之,例如通过使用Lipofectamine(Invitrogen)转染细胞,约1-2μgDNA用于5×105个细胞或磷酸钙沉淀(Invitrogen,5-20μgDNA用于5×105个细胞)。通常在转染后24-48小时检查平板的细胞融合。通过使用May-Grünwald和Giemsa染色(Sigma)可以看到合胞体,并且融合指数计算为[(N-S)/T]×100,其中N是合胞体中的核数,S是合胞体数,T是计数的核总数。
人合胞素包括HERV-W和HERV-FRD。这些蛋白质的功能性直系同源物可以在人科中发现。HERV-W是指属于人内源性逆转录病毒(HERV)家族的高促融合膜糖蛋白。HERV-W是包膜糖蛋白;它也称为合胞素-1。它具有Ensembl数据库中所示的序列,对应于转录物ERVW-1-001、ENST00000493463。相应的cDNA具有SEQ ID NO:1所示的序列。HERV-FRD也指属于人内源性逆转录病毒(HERV)家族的高促融合膜糖蛋白。HERV-FRD是包膜糖蛋白,也称为合胞素-2。它具有Ensembl数据库中所示的序列,对应于转录物ERVFRD-1、ENSG00000244476。相应的cDNA具有SEQ ID NO:2所示的序列。
鼠合胞素包括鼠合胞素-A(即:家鼠(mus musculus)合胞素-A,synA)和鼠合胞素-B(即:家鼠合胞素-B,synB)。这些蛋白质的功能性直系同源物可以在鼠科家族中找到。鼠合胞素-A由合胞素-A基因编码。合胞素-A具有如Ensembl数据库Syna ENSMUSG00000085957中所示的序列。相应的cDNA具有SEQ ID NO:40所示的序列。鼠合胞素-B由合胞素-B基因编码。合胞素-B具有Ensembl数据库Synb ENSMUSG00000047977中所示的序列。相应的cDNA具有SEQ ID NO:41所示的序列。
合胞素-Ory1由合胞素-Ory1基因编码。合胞素-Ory1的功能性直系同源物可以在兔科(通常是兔子和野兔)中找到。
合胞素-Car1由合胞素-Car1基因编码。合胞素-Car1的功能性直系同源物可以在来自劳亚兽总目的食肉动物哺乳动物中找到(Cornelis等,2012;Lavialle等,2013)。
合胞素-Rum1由合胞素-Rum1基因编码。合胞素Rum-1的功能性直系同源物可以在反刍动物哺乳动物中找到。
在本发明的各种实施方案中,根据本发明的ERV合胞素通常可以选自HERV-W、HERV-FRD、合胞素-A、合胞素-B、合胞素-Ory1、合胞素-Car1和合胞素-Rum1及其功能性直系同源物;优选地,ERV合胞素选自HERV-W、HERV-FRD、鼠合胞素-A及其功能性直系同源物,更优选地,ERV合胞素选自HERV-W、HERV-FRD和鼠合胞素-A,甚至更优选地,ERV合胞素是HERV-W或HERV-FRD。
根据本发明的方法允许获得高物理滴度以及高感染性滴度的包括异源目标基因的稳定的假型慢病毒颗粒。实际上,由于本发明的方法,所述包含异源目标基因的稳定的假型慢病毒颗粒以高物理滴度获得,并且其具有感染性,因此能特别有效用于转导B细胞或髓样细胞。
“物理滴度”是指产生的慢病毒颗粒的数量。物理滴度可以根据本领域已知的经典方法测量,并且通常用于定量慢病毒颗粒,例如通过实施例中说明的p24 ELISA,通过用自动计数器(如来自Malvern的Nanosight NS300仪器)使用制造商的说明直接进行颗粒计数,通过RT(逆转录酶)ELISA(如Ciré等,Plosone,July 2014,Vol.9,Issue 7,Immunizationofmice withlentiviral vectors targeted to MHC class II+cells is due topreferential transductionof dendritic cells in vivo中所述)或通过病毒RNA RT-qPCR。
“高物理滴度”是指在步骤b)结束时产生的颗粒滴度高于200ng p24/mL,优选高于210ng p24/mL,甚至更优选高于300ng p24/mL。更优选地,在步骤b)结束时产生的用ERV合胞素如HERV-W假型化的颗粒的物理滴度高于300ng p24/mL,优选高于400ng p24/mL。更优选地,在步骤b)结束时产生的用ERV合胞素如HERV-FRD假型化的颗粒的物理滴度高于210ngp24/mL,优选高于300ng p24/mL。
“高物理滴度”还表示在步骤c)结束时(即浓缩的)产生的颗粒滴度高于1×105ngp24/mL,优选高于1.1×105ng p24/mL,更优选高于1.5×105ng p24/m。更优选地,在步骤c)结束时产生的用ERV合胞素如HERV-W假型化的颗粒的物理滴度高于2.2×105ng p24/mL,优选高于3.0×105ng p24/mL,甚至优选高于4.0×105ngp24/mL。更优选地,在步骤c)结束时产生的用ERV合胞素如HERV-FRD假型化的颗粒的物理滴度高于1.1×105ng p24/mL,优选高于1.5×105ng p24/mL。
根据Farson等(Hum.Gene Ther.2001),可以假设1飞克(fg)p24对应于12个物理颗粒(pp)的慢病毒。不希望受理论束缚,发明人认为,与通用自动计数器(例如NanosightNS300仪器)直接进行颗粒计数获得的值相比,基于p24获得的这种物理颗粒的计算数量略微过高,因为单独的p24也被考虑在内。通常,可以估计,用p24 ELISA计算的颗粒平均比用自动计数器测量的颗粒多约4倍(参见下面实施例3中的补充表S6)。因此,物理滴度高于1×105ngp24/mL对应物理滴度高于1.2×1012计算的物理颗粒(pp)/mL或高于约3×1011pp/mL,如通过Nanosight计数所测量的。
“感染性滴度”是指功能性慢病毒颗粒的数量。感染性滴度可以根据本领域已知的经典方法测量,例如通过在实施例中说明的流式细胞术计算转导单位滴度(TU/mL)或通过qPCR测量感染性基因组(ig/mL)。描述这种方法的出版物是Kutner RH,Zhang XY,Reiser J(2009).Production,concentration and titration of pseudotyped HIV-1-basedlentiviral vectors.Nature protocols 4:495-505。本发明的慢病毒颗粒的感染性滴度是在感染允许细胞系,优选293T细胞或另一种允许细胞如A20细胞时测量,更优选使用本申请实验性实施例部分所述的滴定方法。
“高感染性滴度”是指在步骤c)结束时产生的感染性颗粒的滴度高于2E+04TU/ml,优选高于1E+05TU/ml,更优选高于1E+06TU/ml,甚至更优选高于2E+06TU/ml。更优选地,在步骤c)结束时产生的用HERV-W假型化的颗粒的感染性滴度高于1.2E+05TU/ml,优选高于2E+05TU/ml,更优选高于1E+06TU/ml,甚至更优选高于1.5E+06TU/ml。更优选地,在步骤c)结束时产生的用HERV-FRD假型化的颗粒的感染性滴度高于2E+04TU/ml,优选高于2E+05TU/ml,更优选高于1E+06TU/ml,甚至更优选高于2.5E+06TU/ml。优选地,使用293T细胞感染和本文实施例中描述的滴定方法测量这种高感染性滴度。
本发明还涉及用HERV-W或HERV-FRD假型化并包装有异源目标基因的稳定的慢病毒颗粒。这通过上述方法获得或可获得。
“稳定”是指感染293T细胞或另一种允许细胞系(例如A20细胞)之后,在两个连续时间点之间的根据本发明的稳定的慢病毒颗粒的感染性滴度最多除以10,所述慢病毒颗粒用ERV合胞素(例如先前定义的ERV合胞素,更优选HERV-W、HERV-FRD或鼠合胞素-A)假型化且包装有异源目标基因。连续两个时间点可表示持续几周,例如1-3周。连续两个时间点也可以表示持续几个月,例如1、2或3个月,或6、9、10个月或甚至12或18个月。根据本发明的慢病毒假型化颗粒可以冷冻,它们保持其稳定性。它们确实能抵抗极低温处理,例如冷冻或冷冻保存。它们也可以抵抗中温处理:一旦它们呈现如上所述的稳定特性,它们就对应于本发明。
因此,可以将这种慢病毒假型化颗粒离心和/或冷冻,同时保持其融合和感染特性,以及它们特别是在体内将目标基因递送至靶细胞的能力。再次,根据本发明的包装有异源目标基因的稳定的假型化慢病毒颗粒呈现高物理滴度以及高感染性滴度。
“异源”基因是指来自与逆转录病毒rev、gag和pol基因之一不同的生物体的基因。
“目标基因”是指基因的功能性形式。功能性形式是指所述基因的野生型形式,属于同一家族的变体基因,或保留编码蛋白功能性的截短形式。
目标基因可以源自在患者中缺乏或无功能的基因,或编码免疫原性蛋白(例如病毒蛋白、细菌蛋白或肿瘤抗原)的基因。异源目标基因也可以是报告基因(用于诊断目的,用于鉴定靶蛋白的配体)、自杀基因或编码治疗性RNA的基因(即编码与靶DNA或RNA序列互补的反义RNA,或gRNA、RNAi如shRNA)。目标基因能够产生编码的蛋白质、肽或RNA。
“假型化”是指包含以下的慢病毒颗粒:
-来自病毒的包膜糖蛋白,所述病毒与衍生所述慢病毒颗粒的病毒不同;
-修饰的包膜糖蛋白。在这种情况下,可以通过突变或通过任何其他氨基酸修饰来修饰天然的病毒包膜糖蛋白;或
-嵌合糖蛋白。这种糖蛋白是至少一部分病毒糖蛋白与另一个序列之间的融合蛋白。
在步骤a)中,用至少一种质粒转染合适的细胞系。优选地,所述转染是瞬时转染。
优选地,用至少一个、两个、三个或四个质粒转染合适的细胞系。这些细胞类型包括支持慢病毒生命周期的任何真核动物细胞。
优选地,合适的细胞系是稳定细胞系或对于合胞素的促融合作用的灾难性后果不起反应的细胞系,从而在产生颗粒的同时继续生长。所述合适的细胞系是哺乳动物细胞系,优选人细胞系。此类细胞的代表性实例包括人胚肾(HEK)293细胞及其衍生物、HEK293 T细胞,以及选择其作为贴壁细胞生长或适于在无血清条件下悬浮生长的细胞子集。这种细胞是高度可转染的。
在一个实施方案中,合适的细胞系已经表达了五个序列中的至少一个,并且至多四个,所述五个序列是异源目标基因、逆转录病毒rev、gag和pol基因,以及编码ERV合胞素(例如HERV-W、HERV-FRD或鼠合胞素A)的核酸,优选为诱导型。在这种情况下,步骤a)包括用至少一种质粒转染所述细胞系,所述质粒包含至少一个尚未在所述细胞系中表达的序列。选择质粒混合物或单个质粒(如果仅使用一种质粒),使得当在步骤a)中转染到所述细胞系中时,所述细胞系表达上述所有五个序列。
例如,如果合适的细胞系表达逆转录病毒rev、gag和pol基因,待转染的质粒或质粒混合物包含待表达的剩余序列,即异源目标基因和编码ERV合胞素(例如HERV-W、HERV-FRD或鼠合胞素A)的核酸。
当使用单个质粒时,它包含所有5个目标序列,即:
-异源目标基因,
-rev、gag和pol基因,和

当使用两种或三种质粒时(质粒混合物),每一种包含之前段落中所列的一些目标序列,使得质粒混合物包含所有上述目标序列。
优选使用四种质粒,四转染包含以下:
-第一种质粒包含目标基因,
-第二种质粒包含rev基因,
-第三种质粒包含gag和pol基因,和

优选用特定比例的四种质粒进行所述四转染。可以调节不同质粒之间的摩尔比用于优化放大生产。本领域技术人员能够根据其用于生产目标慢病毒的特定质粒调节该参数。具体地,第一、第二、第三、第四质粒的重量比优选为(0.8-1.2):(0.1-0.4):(0.5-0.8):(0.8-1.2),更优选为约1:0.25:0.65:1。
rev、gag和pol基因是逆转录病毒的,优选慢病毒。优选地,它们是HIV基因,优选HIV-1基因,但也可以是EIAV(马传染性贫血病毒)、SIV(猴免疫缺陷病毒)、泡沫病毒或MLV(鼠白血病病毒)的病毒基因。

在一个具体的实施方案中,用于培养细胞的培养基是经典培养基,例如包含糖(如葡萄糖)的DMEM。优选地,培养基是无血清培养基。可以在适于悬浮培养细胞的很多培养装置,例如多层系统或生物反应器中进行培养。生物反应器可以是单次使用(一次性)或可重复使用的生物反应器。生物反应器例如可以选自培养瓶或袋或罐式反应器。非限制性的代表性生物反应器包括玻璃生物反应器(例如B-2L-10L,Sartorius)、使用运动搅拌的单次使用的生物反应器如波式生物反应器(例如Cultibag10L-25L,Sartorius)、单次使用的搅拌罐式生物反应器(Cultibag50L,Sartorius)或不锈钢罐式生物反应器。

优选地,于2-5℃(优选约4℃)下以45000-55000g的速度进行离心1.5-2.5小时。在该步骤结束后,以离心物的形式浓缩颗粒,其可以使用。

优选地,药物组合物含有对于能够注射的制剂而言是药学上可接受的介质。这些可以特别是等渗无菌盐水溶液(磷酸二氢钠或磷酸二钠、氯化钠、氯化钾、氯化钙或氯化镁等或这些盐的混合物),或干燥的尤其是冷冻干燥的组合物,其在添加无菌水或生理盐水(取决于情况)时,允许构成可注射溶液。
适于注射使用的药物形式包括无菌水溶液或分散体。溶液或悬浮液可以包含添加剂,其与包膜病毒相容且不阻止病毒进入靶细胞。在所有情况下,该形式必须是无菌的并且必须是以易于注射的程度流动的。它必须在制造和储存条件下稳定,并且必须防止微生物如细菌和真菌的污染作用。合适的溶液的实例是缓冲液,例如磷酸盐缓冲盐水(PBS)。
由于其包装有异源目标基因,本发明的稳定的假型化慢病毒颗粒可以用作药物。
实际上,本申请提供的结果清楚地证明,本发明的合胞素假型化的慢病毒载体可以用于人的免疫治疗,并且可以在人疾病中功能性校正B细胞。此外,本发明的合胞素假型化的载体能够在体内靶向B细胞。本文所示的数据表明,这些新工具在体内耐受非常好,因为长期观察到基因转移。对于体内应用,可以获得靶细胞(例如B细胞)的有效转染,而不需要vectofusin1。
因此,稳定的假型化慢病毒颗粒可以用于基因疗法或免疫疗法或用作疫苗或用于免疫预防。
基因疗法可以通过以下进行:基因转移、基因编辑、外显子跳跃、RNA干扰、顺式剪切,或细胞中任何编码或调控序列,包括在细胞核、线粒体中含有的那些或作为共生DNA(细胞中含有的病毒序列)的任何其他遗传修饰。
两种主要的基因疗法是以下:
-旨在替代缺陷/异常基因的疗法:这是替代基因疗法;
-旨在基因编辑的疗法:这种情况下,目的是为细胞提供必要工具,使得目标基因被表达:这是基因编辑疗法。
在替代基因疗法中,目标基因可以是在患者中缺陷或突变的基因的正确形式,例如在遗传疾病中的情况。在这种情况下,目标基因将恢复缺陷或突变基因的表达。特别感兴趣的是表现出疾病的患者的缺陷或突变基因,一旦在免疫细胞(优选B细胞、T细胞、单核细胞或树突细胞,更优选B细胞)中校正,其改善患者的疾病或症状。遗传缺陷B细胞中的突变基因的实例如下:
-在X连锁的严重联合免疫缺陷病(SCID-X1)中编码γ链的基因(患者具有T-NK-B+表型),或已经通过基因疗法治疗但在其中没有诱导B细胞校正的SCID-X1患者;
-在常见的可变免疫缺陷(CVID)中突变的编码TACI(跨膜活化剂和钙调节剂和亲环蛋白配体相互作用物)的基因TNFRSF13B;
-在常见的可变免疫缺陷(CVID)中可能突变的CD19或BAFFR基因;
-任何负责超免疫球蛋白M的基因,特别是由编码CD40的基因突变引起的;

在吞噬细胞功能的遗传疾病中,特别是那些导致产生非功能性或异常单核细胞或树突细胞并且可以通过表达或校正所述单核细胞或树突细胞中的致病基因来治疗的遗传疾病中的突变基因的实例如下:Wiskott Aldrich综合征(WAS基因)、慢性肉芽肿病(CGD、CYBB或p47基因)、白细胞粘附缺陷(LAD)、IL-12/23缺陷(IL12、p40、p70、IL23链),由缺乏HLA分子引起的裸淋巴细胞综合征、自身炎症性疾病,如家族性地中海热(pyrin基因)或cryopyrin相关疾病(NALP3突变)。
替代基因疗法也可以用于治疗癌症。癌症中的目标基因可以调控肿瘤细胞的细胞周期或代谢或迁移,或诱导肿瘤细胞死亡。例如,可以在B细胞白血病或B细胞淋巴瘤中表达可诱导的半胱天冬酶-9以触发细胞死亡,优选在联合疗法中以引发可持续的抗肿瘤免疫应答。
涉及表达ERV合胞素(例如合胞素-1或合胞素-2)受体的细胞的疾病也可以通过将基因转移至这些细胞来治疗。例如,可以引用可以治疗胎盘细胞的妊娠病理学;可以治疗恶性细胞的癌症,如白血病、淋巴瘤或实体瘤细胞(包括已知表达ASCT2或MFSD2a的乳房或肺肿瘤细胞);可以治疗血脑屏障内皮细胞的神经系统病症;或任何涉及这些细胞的遗传或获得性病症。
在基因疗法中,有可能通过转导所述免疫细胞,将本发明的稳定的假型化慢病毒颗粒用于免疫细胞工程,优选B细胞工程的治疗。通过在B细胞中表达免疫抑制蛋白(例如IL-10)或诱导免疫调节蛋白(如抗体或融合蛋白)的产生,可能产生调控性B细胞。
也可以插入有利于基因剪切、表达或调控或基因编辑的序列。诸如CRISPR/Cas9的工具可用于此目的。这可用于修饰B细胞中的基因表达(在自身免疫或癌症的情况下),或扰乱B细胞中病毒的循环。在这种情况下,优选地,异源目标基因选自gRNA、核酸酶、DNA模板和RNAi组分,例如shRNA。
基因编辑的一个具体实例是巨球蛋白血症的治疗:在这种疾病中,在80-88%的患者的B细胞中观察到(MYD88)的点突变(L265P),所述点突变有助于这些肿瘤细胞的生存和持久性。因此,通过基因编辑受折磨的患者中该基因的正确形式,这可以有助于针对该疾病的有效治疗。
本发明的稳定的假型化慢病毒颗粒可以一起使用或依次使用,以通过不同的受体靶向相同的细胞。这在诸如基因编辑(其中需要向细胞添加基因编辑平台的多个组分)的策略中可能是有利的。
在基因编辑疗法中,还可以用本发明的稳定的假型化慢病毒颗粒转导免疫细胞,优选B细胞。然后优选地,如果异源目标基因编码免疫球蛋白,则转导的B细胞能够产生相应的免疫球蛋白。一个实例是将可变免疫球蛋白区域的已知序列工程化,所述可变免疫球蛋白区域可以对病原体例如病毒(狂犬病、HIV、埃博拉病毒、Flu、CMV、Zika)或寄生虫病毒(克氏锥虫、恶性疟原虫)进行特异性识别,其可用于预防性或治疗性用途或用于诊断。

当转导的免疫细胞是B细胞时,则治疗可以用于治疗自身免疫疾病。
当转导的免疫细胞是树突细胞时,则治疗可以是疫苗。
例如,用同种异体器官供体特异性的主要或次要组织相容性抗原转导的受体未成熟树突细胞可用于在受体中诱导同种异体器官移植耐受性。
同样,通过抗原呈递细胞如人B细胞、人髓样树突细胞或单核细胞的基因工程的免疫疗法应用可以通过向这些细胞引入用于抗原呈递的含有与免疫调节序列相关或不相关的抗原序列的重组基因表达盒来找到,用于接种疫苗以预防感染性疾病;用于治疗癌症;或用于诱导细胞特异性或抗原特异性免疫耐受,以治疗具有炎性免疫组分的疾病(例如糖尿病或阿尔茨海默氏病),或促进器官或骨髓移植。
在基因疗法中,靶组织不仅是免疫细胞,还可以是胎盘以及发生合胞素介导的异源融合的任何组织,或表达合胞素受体的任何组织或任何细胞。实例包括治疗胎盘以预防先兆子痫的可能性。先兆子痫是产妇死亡的主要原因,并且一些研究已经在先兆子痫胎盘中检测到差异性表达的microRNA解除了对TGF-β2反应的调控(Zhou等Sci Rep.2016Jan29;6:19910.doi:10.1038/srep19910)。可能地,通过使用合胞素-假型化载体在胎盘细胞中进行基因转移或基因编辑可以纠正这个问题。靶组织可能是骨骼肌,因为最近显示合胞素在成肌细胞中表达并且可能有助于成肌细胞之间的融合(Frese等,2015)。可能地,可识别骨骼肌细胞上的受体的合胞素-假型化载体可用于肌肉基因疗法。

“免疫缺陷”是指导致微生物(细菌、病毒、寄生虫)感染的数量或严重程度增加或抗肿瘤监测减少并导致癌症频率增加的病症。

材料和方法
细胞系
在37℃,5%CO2下,在补充有10%热灭活胎牛血清(FCS)(Life Technologies)的Dulbecco改良的Eagle培养基(DMEM+glutamax)(Life Technologies,St-Aubin,France)中培养人胚胎肾293T细胞和人结肠直肠癌HCT116细胞(CCL-247;ATCC,Manassas,VA)。在补充有10%FCS的Ham's F12K培养基(Life technologies)中培养人绒毛膜癌BeWO细胞(CCL-98;ATCC)。在XVivo 20培养基中感染Raji细胞和Jurkat细胞。
克隆表达合胞素的质粒并制备载体
通过使用NHeI和XhoI限制酶,将分别对应于Ensembl基因组浏览器ENST00000493463和ENSG00000244476转录物序列的合成的DNA序列(Gencust,Dudelange,Luxembourg)克隆入pCDNA3质粒(Invitrogen,Carlsbad,California)来构建编码人合胞素-1(ERVW-1-001)或人合胞素-2(ERVFRD-1)的质粒pCDNA3-合胞素-1和pCDNA3-合胞素-2。通过双链测序验证质粒序列。通过免疫印迹蛋白质提取物,随后用检测预期为合胞素-2的65-70Kd蛋白质的抗-HERV-FRD抗体(LS-BIO,Nanterre,France)转染293T细胞来验证合胞素-2质粒的功能性。使用DNA RNA纯化Nucleobond试剂盒(Macherey-Nagel,Duren,Germany),在无内毒素情况下产生质粒。使用磷酸钙,用4种质粒瞬时转染293T细胞来制备用合胞素-1或合胞素-2假型化的慢病毒颗粒。用以下转染每个T175cm2烧瓶:14.6μg表达HIV-1gagpol基因的pKLgagpol、5.6μg表达HIV-1rev序列的pKRev、22.5μg基因转移盒(在人磷酸甘油酸激酶(PGK)启动子控制下表达增强的绿色荧光蛋白(GFP)的pCCL PGK GFP(Charrier等,2011)或在人PGK启动子控制下表达截短形式的神经生长因子受体(NGFR)的pCCL PGK delta-NGFR)和22μg pCDNA3-合胞素-1或pCDNA3-合胞素-2质粒。第二天更换培养基,并用补充有青霉素和链霉素和10%FCS的DMEM 4.5g/L葡萄糖代替。24小时后,收集含有病毒颗粒的培养基,低速离心,无菌过滤(0.22μm)并通过在4℃下超速离心50,000g(19500RPM使用具有转子SW28的Beckman超速离心机)浓缩2小时。将沉淀重悬于PBS,等分并储存在-80℃。如报道的那样,还通过瞬时转染产生VSVg-假型化颗粒(Merten等,2011)。
转导添加剂
通过标准的芴基-甲氧基-碳酰氯固相肽合成法合成Vectofusin-1(VF1)肽,然后用HPLC和质谱纯化(Gencust)。将肽溶解在H2O中,等分并在-80℃下储存直至使用。当转导实验需要时,将VF1解冻,重悬于培养基中并以12μg/mL的终浓度添加至载体和细胞中。硫酸鱼精蛋白(PS)(Sigma-Aldrich,St Louis,MO)和聚凝胺(PB)(Sigma-Aldrich)也用载体和细胞临时稀释,在转导培养中分别以8μg/mL和6μg/mL的终浓度使用。
载体的滴定
使用前滴定浓缩且冷冻保存的载体。如先前报道的(Charrier等,2011),通过p24ELISA(HIV-1Elisa试剂盒,Perkin-Elmer,Villebon/Yvette,France)测定物理颗粒滴度。通过以下测定感染性滴度:在12μg/mL VF1存在下,向HEK293 T细胞添加连续稀释的载体,并且在3天后,通过流式细胞术分析293T细胞以计算转导单位滴度(TU/mL),或通过qPCR使用标准计算测量感染性基因组(ig/mL)(Kutner等,2009)。
培养并转导外周血单核细胞
用EDTA收集的外周血购自Etablissement du Sang(Evry,France)。将通过Ficoll梯度离心(Eurobio,les Ulis,France)纯化的外周血单核细胞(PBMC)悬浮于无血清培养基X-VIVO 20(Lonza,Levallois-Perret,France)中用于转导。在一些实验中,在转导之前将细胞因子IL-7(10ng/μL)(Miltenyi Biotech,Bergisch Gladbach,Germany)加入培养基过夜。对于转导,在VF1(12μg/mL)存在下,将LV-Syn1或LV-Syn2载体(1.105TU/mL)或LV-VSVg(1.108TU/mL)加入细胞。6小时后,将培养基更换为补充有10%FCS和IL-7(10ng/mL)的XVivo20,并将细胞培养长达7天。为了扩增转导的B细胞,向培养基中加入CD40L(2μg/mL)和IL-4(20ng/mL),并在这些条件下培养细胞长达2周。为了扩增髓样树突细胞,向培养基中加入hGM-CSF(50ng/ml)和IL-4(20ng/ml)。
EBV的永生化
用IL-7培养PBMC过夜,然后用载体感染。6小时后,洗涤细胞并在B95.8狨猴细胞上清液和环孢菌素存在下使用EBV永生化的标准方法进行培养。培养基每周更换两次。
转导SCID-X1患者细胞
将通过基因疗法治疗的SCID-X1患者的PBMC在37℃快速解冻,然后用PBS洗涤两次。在台盼蓝计数后,用2μM CFSE(Molecular Probes,Cambridge,UK)标记细胞,用Lv-Syn1IL2rg载体转导并在不存在或存在CD40配体[2μg/mL](Miltenyi Biotec)和IL-21[50ng/mL](Miltenyi Biotec)的情况下,在96孔圆底板中培养(3.105/200μL)。培养3、6和7天后,通过流式细胞术测定B细胞转导(即NGFR或IL2Rg表达)、B细胞增殖(即CFSE稀释)和CD19+CD27+细胞的频率。通过ELISA(Sigma-Aldrich,St Louis,MO)测定培养基中的人IgG分泌。
体外B细胞活化
用2μM CFSE(Molecular Probes,Cambridge,UK)标记PBMC,并在不存在或存在CD40配体[2μg/mL](Miltenyi Biotec)和IL-21[50ng/mL](Miltenyi Biotec)的情况下,在96孔圆底板中培养(3.105/200μL)培养。培养3、6和7天后,通过流式细胞术测定B细胞转导(即NGFR或IL2Rg表达)、B细胞增殖(即CFSE稀释)和CD19+CD27+细胞的频率。通过ELISA(Sigma-Aldrich,St Louis,MO)测定培养基中的人IgG分泌。
流式细胞术
用于流式细胞术的抗体购自BD-Biosciences(Le Pont de Claix,France),包括PE偶联的抗CD3和抗CD1a、Alexa-Fluor 700偶联的抗CD19、APC-H7偶联的抗CD45和抗IgD、CF594偶联的抗IgM、PeCy7偶联的抗CD27和抗HLA-DR、APC偶联的抗CD14、V450偶联的抗CD11c。在向细胞添加抗体之前,通过将细胞与γ-球蛋白(1mg/mL)(Sigma Aldrich)在4℃孵育15分钟来阻断Fc受体。然后在4℃下,在含有0.1%牛血清白蛋白(BSA)(SigmaAldrich)的PBS中加入饱和量的抗体30分钟,并将细胞洗涤两次。在该过程结束时,加入7-氨基-放线菌素D(0.3mg/mL)(Sigma-Aldrich)以排除死细胞。对于受体研究,使用兔多克隆抗SLC1A5(ASCT2)(Abcam,Paris,France)、抗MFSD2a(Origene,Rockville,MD)或兔免疫球蛋白对照(Origene),并通过Alexa 647-偶联的山羊抗兔抗体(Thermo FisherScientific,Waltham,MD)显示。使用Diva软件(BD-Biosciences)在LSRII上进行采集,并使用Kaluza软件(Beckman-Coulter,Villepinte,France)进行分析。
用PE偶联的抗人CD132抗体(BD bioscience)检测人IL2Rg。
通过RT-qPCR测量ASCT2和MFSD2A表达
使用SV总RNA分离(Promega,Charbonnièrelesbains,France)从纯化的细胞中提取总RNA。使用游离DNA试剂盒(Ambion,Courtaboeuf,France)从样品中除去残留的DNA。根据用于逆转录-PCR(Invitrogen)的Superscript II第一链合成系统的方案,使用随机六聚体从1μg RNA合成cDNA。根据Sybr Green PCR Master Mix(Applied Biosystem,Fostercity,CA,USA)的方案,使用LightCycler 480系统(Roche,Bale,Switzerland)和0.1μM的每种引物进行实时PCR。先前描述了用于扩增ASCT2或MFSD2A的引物对(Cornelis等,PNAS,2008和Toufailly等,placenta,2013)。ASCT2引物的序列为(有义,5’-GGCTTGGTAGTGTTTGCCAT-3’;反义,5’-GGGCAAAGAGTAAACCCACA-3’)和MFSD2A的引物为(有义,5’-CTCCTGGCCATCATGCTCTC-3’;反义,5’-GGCCACCAAGATGAGAAA-3’)。我们还使用TFIID(Transcription Final II D)引物作为标准化。TFIID引物序列为(有义,5’-ACGGACAACTGCGTTGATTTT-3’;反义,5’-ACTTAGCTGGGAAGCCCAAC-3’)。使用以下公式将结果表示为倍数变化:相对丰度=2^-ΔΔCt,其中Ct=Ct ASCT2或MFSD2A–Ct TFIID,ΔΔCt=ΔCt样品-ΔCt校准物。
LV-Syn1载体的体内转导
向从Charles River购买的7周龄雌性NSG小鼠(Nod/Scid/gc-/-)的眼眶后窦中注射100μL体积的107个PBMC。24小时后,在小鼠尾静脉中通过静脉注射150μL未稀释的LV-Syn1载体。7天后,处死小鼠并收集脾脏,用ACK裂解并通过FACS分析红细胞耗尽的级分。
用LV-Syn1或lV-Syn2载体离体转导鼠细胞
用ACK裂解红细胞后,从6周龄雌性C57Bl/6小鼠获得脾细胞。在1或5×105TU/mL的编码ΔNGFR的LV-SYn1或LV-Syn2和VF1(12μg/mL)存在下,在100μL补充有10%FCS和β2巯基乙醇的RPMI中,以106个细胞/mL的浓度培养细胞。3天后,通过FACS测量活细胞上的ΔNGFR表达。
统计学分析
使用GraphPad Prism软件(GraphPad Inc,La Jolla,CA),按照说明,通过单向ANOVA分析、Mann-Whitney或Student's t检验评估统计学显著性。
结果
合胞素-1和合胞素-2假型化的LV可制备为稳定的感染性颗粒
研究了两种人内源性逆转录病毒包膜糖蛋白,合胞素-1和合胞素-2作为重组HIV-1衍生的LV的可能的新假型。在293T细胞中的瞬时LV生产系统中表达这些糖蛋白的全长cDNA。用于转染步骤的合胞素-1和合胞素-2质粒的量的优化增加了基于培养基中p24水平的LV颗粒的产生(图11A)。在所定义的条件下(参见材料和方法),可以产生用这些包膜中的任何一种假型化的稳定的感染性颗粒。使用不同转基因(GFP和dNGFR),收获的LV-Syn-1母液平均滴度为705ng p24/mL(n=7产品),LV-Syn-2平均滴度为496ngp24/mL(n=4产品)(表1)。使用与VSVg假型化颗粒相同的条件,通过超速离心可以成功地浓缩用这些包膜中的任一种假型化的慢病毒颗粒(Charrier等,2011)。将浓缩的母液在-80℃下冷冻保存并稳定数月。解冻LV母液后,LV-Syn1(n=6产品)的滴度为4.1±1.7E+05ng p24/mL,LV-Syn2的滴度为1.7±0.6E+05ng p24/mL(n=6产品),载体可制备为编码不同的转基因(GFP或ΔNGFR)(表1)。

备注:通过瞬时转染、澄清、无菌过滤、浓缩或不浓缩、冷冻保存、解冻和滴定产生独立批次的具有指定包膜和转基因的LV(n=测试的批次数)。通过超速离心(19500rpm(50000g)2小时)进行浓缩。通过ELISA测量p24水平的物理滴度,并使用流式细胞术在Vectofusin-1存在下测量作为293T细胞转导单位(TU)的感染性滴度。
为了测定这些颗粒的感染性,本发明人首先感染已知表达合胞素-1的ASCT2受体和更受限制的合胞素-2的MFSD2a受体的人BeWo绒毛膜癌细胞系(Esnault等,2008)。用LV-Syn1或LV-Syn2未能有效转导BeWo细胞,而用VSVg假型化LV能很好转导。然而,如LV-Syn2所示,添加阳离子试剂如硫酸鱼精蛋白(PS)、聚凝胺(PB)或Vectofusin-1(VF1)增强了BeWo细胞的转导(图1)。在这些试剂中,用VF1获得最大的转导水平增强。Vectofusin-1是一种短的富含组氨酸的两亲性肽,其增强了用各种包膜糖蛋白(包括RD114-TR)假型化的慢病毒和-逆转录病毒载体的感染性(Fenard等,2015和Majdoul等,2015)。之前尚未测试Vectofusin-1与合胞素一起的作用,并且该短肽揭示了合胞素假型化的LV颗粒的基因转移潜力。
在VF1存在下,可以以剂量依赖性方式用任一合胞素假型化的载体转导293T细胞,并达到与BeWo细胞中获得的水平相当的水平(图2A,B)。相反,只有约1-2%的293T细胞在没有这种添加剂的情况下被转导。在使用LV-Syn1或LV-Syn2的293T细胞中的重复实验中,VF1的增强作用具有统计学显著性(图2C)。用合胞素假型化的载体转导293T细胞导致转基因随时间的稳定基因组整合,如在持续2周的连续培养中所检测的(图11B)。基因表达水平与每个细胞的拷贝数一致,未显示随时间转基因沉默的证据(图2D)。
发现VF1肽对LV-Syn1和LV-Syn2感染性的增强作用使得能够使用293T细胞开发稳健的感染性滴定测定。293T细胞系的选择是由于未能转导HCT116结肠癌细胞而导致的,这些细胞通常用于在实验室中滴定VSVg假型化的LV。甚至在Vectofusin-1存在下,用任何测试浓度的LV-Syn1或LV-Syn2都不能转导HCT116(图11C)。此外,因为在重复实验中更一致的生长模式和更高转导水平的趋势,对于这种感染性滴度测定,293T细胞优于BeWo细胞(图11C)。在测定定义的条件下(参见材料和方法),浓缩且冷冻保存的编码GFP的LV-Syn1和LV-Syn2批次的感染性滴度分别为:对于GFP转基因,2.8±6x E+06TU/mL(n=6批次)和2.7±6xE+06TU/mL(n=6批次),对于dNGFR转基因,6.3±7.5x E+06TU/mL(n=3批次)和4.4±1.5xE+06TU/mL(n=2批次)(表1)。因此,合胞素-1和合胞素-2糖蛋白可以假型化rHIV载体,以获得具有感染性并且可用于稳定的基因转移的稳定颗粒。它们的感染性需要辅助因子,并且在细胞水平上具有选择性。
LY-Syn1和LV-Syn2载体有效转导初始血细胞
合胞素是细胞蛋白质,其可用于衍生用于体内应用的免疫耐受载体。因此,为了评估合胞素假型化的载体是否会与血细胞相互作用,本发明人向体外的外周血单核细胞(PBMC)中加入载体。在体外分离后立即感染PBMC,或在IL-7存在下(目的是维持初始淋巴细胞的活力)过夜培养后感染PBMC(图12A)。在感染步骤后,在IL-7存在下培养细胞以支持细胞活力至少3天。总体上,转基因表达在表达CD45的总有核细胞群中较低(补充表S1)。然而,与不含载体的模拟对照相比,在VF1存在下转导的细胞量显著更高(补充表S1)。LV-Syn1的作用与VSVg的作用相当。已经用IL-7预活化的细胞比分离后立即感染的细胞更好地被转导。
补充表S1:在Vectofusin-1存在下用LV-Syn1和LV-Syn2转导完整PBMC

PBMC包含不同比例的多个细胞子集。如图3和表2所示,在大部分CD19+B淋巴细胞和CD11c+髓样/树突细胞(PBMC中是少数)中发现转基因表达,而只有一部分CD3+T细胞(在PBMC中更丰富)与CD45+细胞中获得的总体低转导结果显著一致。在用IL-7预活化后,通过LV-Syn1转导了平均59%的每个CD19+细胞或CD11c+细胞(表2)。如用293T和BeWo细胞所观察到的,添加VF1对于实现PBMC转导是必需的,因为在没有该添加剂的情况下很少细胞被转导。VF1肽还增强通过LV-VSVg的PBMC的转导。

值得注意的是,有可能转导未活化的CD19+细胞,这表明合胞素可以靶向真正的初始细胞。已经表征了各种CD19+B细胞子集(Kaminski等,2012),包括初始CD19+CD27-IgD+细胞(群体的50-70%);记忆非转换CD19+CD27+IgD+细胞(群体的15-20%);记忆转换CD19+CD27+IgD-IgM-细胞(群体的2-5%);记忆转换仅IgM CD19+CD27+IgD-IgM+细胞(群体的2-5%)和浆母细胞CD19+CD27hi(群体的1-2%)。图12B示出了感染PBMC后的FACS门控策略。总体而言,无论预活化如何,用合胞素假型化的载体均有效转导了初始和记忆B细胞子集(图3)。更详细地,发现所有记忆B细胞子集在没有预活化的情况下被转导(表3)。在这种非活化条件下,合胞素假型化的载体比LV-VSVg更有效地转导B细胞,表明合胞素对于这些细胞的特殊趋向性。发现LV-Syn1和LV-Syn2的B细胞转导作用依赖于载体浓度(图4),且与转基因无关。几乎所有外周血CD19+细胞都可以用106TU/mL的编码GFP或ΔNGFR的LV-Syn1或LV-Syn2载体转导,从而提供一种非常有效的系统以在这些细胞中表达各种类型的异源蛋白质。

用合胞素假型化的载体稳定转导功能性B细胞
为了排除所观察到的效应是非特异性的(可能在聚集时发生的由伪转导或Vectofusin-1的人工自发荧光引起(Fenard等人2013)),发明人证实了随时间的用载体处理并培养的细胞中的前病毒整合(proviral integration)。在向细胞中加入载体后,本发明人加入CD40L和IL-4以活化和扩增培养物8-14天。在这样的培养物中,发明人特别检测了当载体与Vectofusin-1(表4)一起加入时的载体拷贝,即使条件对于确保所有细胞存活而言并非最佳。此外,使用B细胞活化信号如CD40配体(CD40L)和先前报道的对于记忆B细胞发育所必需的IL-21(Recher等2011),发明人证实了转导的B细胞可以被有效活化(表5)。使用CFSE标记和流式细胞术检测B细胞分裂和转基因表达,发明人证实了能够响应CD40L和IL-21以及具有活化B细胞(包括浆母细胞)表型的细胞的标记(表5)。为了进一步证实功能性B细胞的转导,本发明人使用EBV,在用LV-Syn1载体感染PBMC后立即转化它们,并获得含有稳定整合的载体序列的淋巴母细胞(表4)。这些结果证实,合胞素载体可以稳定转导保留增殖和扩增功能性能力的原代B细胞。

转导鼠原代B细胞
将新鲜分离的鼠脾细胞与编码ΔNGFR的LV-Syn1或LV-SYn2载体一起培养3天。如图17所示,这导致CD19+B细胞的非常有效的转导。在这些培养物中,按比例转导的CD19-细胞少得多,从而证实了小鼠中合胞素假型化的载体对B细胞的特定趋向性,与在人中一样。数据还显示合胞素-1和合胞素-2糖蛋白识别鼠对应物,这些发现将有助于未来这些载体的临床前研究。
转导CD11c+细胞
如图3所示,在Vectofusin-1存在下,用来自没有预活化的PBMC的LV-Syn1和LV-Syn2转导了大部分的CD19-CD3-CD11c+细胞。进一步的表型分析表明,这种CD11c细胞也表达高水平的HLA-DR,并被合胞素假型化的载体有效转导(图5)。细胞在GM-CSF+IL-4中生长,表达CD1a、CD80、CD86且缺乏CD14,表明它们是未成熟的树突细胞(图13A)。另外,还转导了具有单核细胞表型的CD14+HLA-DR+细胞(图13B)。因此,LV-Syn1和LV-Syn2均可以有效地用于在抗原呈递细胞中表达转基因。
LV-Syn1和LV-Syn2载体不能有效转导原代人T细胞
合胞素假型化的LV不能有效转导外周血T细胞,尽管一些细胞可以在活化后被转导。用IL-7预活化后,用LV-Syn1或LV-Syn2载体转导了小部分外周血T细胞,但是这些效果在实验与实验之间不一致(图3和表2)。没有预活化的情况下,转导水平总是很低。
因此,用合胞素假型化的载体观察血细胞子集的差异性转导。
血细胞子集和细胞系上ASCT2和MFSD2a受体的表达
使用间接免疫荧光检测在血细胞子集的细胞表面上检测ASCT2和MFSD2a,它们分别是合胞素-1和合胞素-2的进入受体(图6A)。表达这些标志物的最高百分比的细胞是CD19+和CD11c+细胞,而T细胞表达的这些受体水平较低(图6B)。qRT-PCR证实了这些结果,其首次显示人B细胞表达MFSD2a和ASCT2(图6-C)。还在293T细胞上发现ASCT2和MFSD2a的表达,而所述表达在未用载体转导的HCT116细胞中水平低得多(图14)。在测试的细胞上,受体的表达与实现的转导水平很好地相关。
用合胞素假型化的载体转导人B和T细胞系
为了扩展在原代细胞上获得的结果并进一步评估合胞素假型化的载体的细胞特异性,本发明人试图转导一组人细胞系。测试了Burkitt淋巴瘤Raji B细胞和Jurkat T细胞。可以用合胞素假型化的载体转导Raji B细胞(图15)。使用2×106TU/mL的LV-Syn2载体在Raji细胞中获得的最高水平达到16%(与在原代B细胞中用106TU/mL达到>90%相反,如图4所示)。本发明人证实,Raji细胞允许用LV转导,因为用108TU/mL的LV-VSVg转导了88%的Raji细胞(图15的对照,未显示)。在一个实验中,通过流式细胞术分选转导的Raji细胞并培养49天,证明转基因在延长的时间段内稳定整合(补充表S2)。用合胞素假型化的载体部分转导了Jurkat T细胞(图15)。因此,合胞素假型化的LV可用作某些细胞系的基因转移工具。在B细胞中,转导似乎在原代细胞中比在已建立的B细胞系中更有效。
补充表S2:用合胞素假型化的载体稳定转导Raji细胞
备注:在Vectofusin-1(12μg/mL)存在下,用浓度为1.2×105TU/mL的三种不同的GFP编码载体感染Raji细胞。2周后,LV-Syn1的GFP表达为2.5%,LV-Syn2的GFP表达为5%,LVV-VSVg载体的GFP表达为100%。30天后,通过流式细胞术分选表达(GFP+)或不表达GFP(GFP-)的细胞,并将这些分选的细胞培养至第49天,以通过FACS测量GFP表达,并通过qPCR测量每个细胞的平均载体拷贝数(VCN)。
原发性免疫缺陷的功能矫正
严重联合免疫缺陷型-1(SCIDX1)是X连锁的原发性免疫缺陷,其由白细胞介素2受体γ链(IL2Rg)基因的突变引起。同种异体骨髓移植可以治愈这种疾病,但是一些患者仍然校正不完全,其中B细胞只有部分嵌合状态(Recher等2011)。B细胞嵌合低于5%的患者仍然无法成熟并产生免疫球蛋白。它们的B细胞仍然不能成为分泌B细胞的成熟记忆IgG。实际上,最近发现IL2Rg也是IL-21的受体,为B细胞的成熟提供必需的信号(Recher等2011)。已经尝试了几种基因治疗试验,在造血干细胞和祖细胞中使用IL2Rg基因转移来治疗该疾病。然而,在没有对患者进行调理的情况下,基因校正的细胞仅产生校正的T细胞,并且B细胞仍然未经校正,如最近的基因治疗试验中所示(Hacein-Bey-Abina等2014)。结果,这种患者B细胞是无功能的,并且患者仍然依赖于免疫球蛋白输注以预防感染。这促使发明人询问是否可以使用合胞素假型化的载体将IL2Rg转移至已经通过基因疗法不完全校正的患者的外周B细胞。从患者获得细胞。发现患者血液单核细胞含有CD3+IL2Rg+T细胞,但与健康个体相反,患者血液CD19+B细胞不表达成熟标志物CD27,不表达IL2Rg(图7A和B)并且不产生Ig。用编码IL2Rg的LV-合胞素1载体进行基因转移后,患者细胞能够通过在培养基中产生IgG来响应CD40L+IL-21,即使在细胞上未检测到活化标志物,可能是由于这些脆弱的细胞在培养物中的高水平死亡率。这些结果表明,外周血B细胞可以通过基因转移进行靶向以在SCIDX1中提供一些功能性校正,并且合胞素假型化的LV是用于该应用的有用工具。
体内实验
通过LV-Syn1有效转导原代人B细胞促使测试该载体是否在体内起作用。首先以每只小鼠107个PBMC植入免疫缺陷的NSG小鼠,第二天静脉内接受单次推注的LV-Syn1。7天后,处死小鼠并检查不同的组织。在3只中的2只小鼠中,发明人在脾中发现存在CD45+CD19+人B细胞,其中一小部分(3-15%)表达GFP,表明它们已在体内转导(图16)。这些初步结果表明可以使用合胞素假型化的载体在体内靶向人B细胞。
实施例2:鼠合胞素A和LV-SynA在小鼠中的体内基因递送
制备稳定的感染性LV-SynA颗粒
材料和方法(也参见实施例1的材料和方法)
克隆合胞素A并产生LV-SynA
a.制备表达鼠合胞素A的质粒
通过插入使用以下引物:正向5’AGCAAGCTTATGGTTCGTCCTTGG 3’(SEQ ID NO:32)(Tm=69,8℃;%GC=50%;Sigma,Saint-Louis,USA)和反向5’AGCTCTAGACTAGACGGCATCCTC3’(SEQ ID NO:33)(Tm=65℃;%GC=54%;Sigma),从Genecust(Ellange,Luxembourg)制备的pUC-SynA质粒PCR扩增的且包含SynA鼠基因全长cDNA(Ensembl参考号ENSMUSG00000085957)的片段和连接将鼠合胞素A的cDNA克隆入pcDNA3.1真核表达质粒的HindIII和XbaI位点。通过测序验证该质粒(Beckman Coulter Genomics,Takeley,UK)。
b.产生Syn-A假型化的慢病毒载体
用以下4种质粒(每瓶的量)共转染在T175cm2烧瓶的DMEM+10%胎牛血清(FCS)中的HEK293T细胞:pKLgagpol(14.6μg)、pKRev(5.6μg)、pcDNA3.1-SynA(20μg)和转移质粒PRRL-SFFV LucII或pRRL-SFFV-LucII-2A-ΔNGFR-WPRE(22.5μg)。24小时后,洗涤细胞并加入新鲜培养基。第二天,收获培养基,通过1500rpm离心5分钟澄清并0.45μm过滤,然后通过在12℃下50000g超速离心2小时来浓缩,并在-80℃下储存直至使用。
c.滴定合胞素A假型化的LV
如其他类型的LV一样,通过p24 ELISA测定物理滴度。使用鼠淋巴瘤细胞系A20,将感染性滴度测定为感染性基因组滴度(IG/mL)。在Vectofusin-(12μg/μL)存在下,将系列稀释液的载体加入A20细胞中6小时。更新培养基并培养细胞7天,获得基因组DNA,以使用iCycler7900HT(Applied Biosystems)上的双重qPCR,用以下引物测量每个细胞的载体拷贝数:PSI正向5'CAGGACTCGGCTTGCTGAAG3'(SEQ ID NO:34),PSI反向5'TCCCCCGCTTAATACTGACG3'(SEQ ID NO:35)和用FAM(6-羧基荧光素)标记的PSI探针5'CGCACGGCAAGAGGCGAGG3'(SEQ ID NO:36)、Titin正向5'AAAACGAGCAGTGACGTGAGC3'(SEQ IDNO:37)、Titin反向5'TTCAGTCATGCTGCTAGCGC3'(SEQ ID NO:38)和用VIC标记的Titin探针5'TGCACGGAAGCGTCTCGTCTCAGTC3'(SEQ ID NO:39)。
小鼠中的体内生物发光基因转移
产生编码LucII的LV-SynA载体,并将5×105TU的载体静脉内注射到白化病C57Bl/6小鼠中。对照包括编码LucII的LV-VSVg和PBS。为了检测生物发光,用氯胺酮(100mg/kg)/木聚糖(10mg/kg)麻醉小鼠,腹膜内施用100μL的150μg/mL D-荧光素,10分钟后用CCD照相机ISO14N4191成像(IVIS Lumina,Xenogen,MA,USA)。使用15cm视野、像素组合(分辨率)因子4、1/f停止和开放式滤镜获得3分钟的生物发光图像。手动定义目标区域(ROI)(在每种情况下使用标准区域),使用活体成像3.0软件(Xenogen)计算信号强度,并表示为光子/秒。背景光子通量由未施用载体的对照小鼠或对照肌肉上绘制的ROI定义。
结果
研究了小鼠合胞素的体内应用。合胞素A是非直系同源物,但是与人合胞素-1和-2功能相似的鼠对应物(Dupressoir等2005)。我们将鼠SynA克隆到表达质粒中并用它来产生慢病毒载体颗粒。我们发现,合胞素A可以成功地将rHIV衍生的LV假型化。我们使用已经定义的相同条件来生产人合胞素假型化的LV(每个平板22Ug DNA,仅一次收获)以制备稳定的LV-SynA。在VF1存在下,LV-Syn A转导鼠A20 B淋巴瘤细胞系非常有效(图18)(补充表S4)。我们发现,SynA在体外转导原代非活化的鼠B细胞方面与其人类对应物一样有效。如图8和图9所示,LV-SynA能够在没有预处理来自脾(图8)和来自骨髓(图9)的鼠B细胞的情况下进行转导。这些实验还表明也可以转导鼠T细胞。B220低细胞的转导表明,未成熟的B细胞被转导并且可以被靶向用于生物学应用。
有趣的是,可以用鼠合胞素A假型化的LV转导人CD19+B细胞(补充表S5)。
证明了LV-Syn A对小鼠的体内基因递送有效。图10显示注射编码LucII的LV-SynA载体16天后转基因的生物发光检测。通过小鼠尾静脉注射载体,发明人为此没有使用vectofusin。通过流式细胞术细胞分选纯化脾细胞并通过q-PCR测量转导水平证实了脾CD45+CD19+细胞中载体的存在(补充表S3)。因此,合胞素是在体外和体内介导进入人和小鼠B细胞的独特工具。

实施例3:物理滴度测定:通过p24RT ELISA获得的物理滴度和用自动计数器获得
的直接颗粒计数之间的对应关系
通过2种不同方法测定HIV-1衍生的慢病毒载体(LV)的物理滴度,所述HIV-1衍生的慢病毒载体(LV)用合胞素-1(S1)、合胞素-2(S2)、合胞素A(SA)或VSVg假型化,并编码截短形式的神经生长因子受体(ΔNGFR)或绿色荧光蛋白(GFP)。通过瞬时转染产生LV,通过超速离心浓缩并在滴定前在-80℃冷冻保存。
该方法包括(a)ELISA测量样品中的p24浓度,然后计算作为物理颗粒(pp)的滴度,假设1fg的p24对应于12pp的LV(Farson等,2001)或(b)使用Malvern Instruments(UK)的NS300 Nanosight颗粒计数器,该计数器使用自动显微镜直接测量样品中的颗粒浓度。

Claims (14)
1.一种浓缩稳定的慢病毒颗粒制剂,所述慢病毒颗粒用选自HERV-W、HERV-FRD和鼠合胞素-A的内源性逆转录病毒合胞素(ERV合胞素)假型化且包装有异源目标基因,其在制剂的冷冻和解冻之前或之后包含高于1x105ng p24/mL的物理颗粒的滴度和/或高于2x104TU/mL的感染性颗粒的滴度。
2.获得权利要求1所述的浓缩稳定的假型化慢病毒颗粒制剂的方法,包括以下步骤:
a)在合适的细胞系中转染至少一种质粒,其中所述至少一种质粒包含所述异源目标基因、慢病毒rev、gag和pol基因和编码选自HERV-W、HERV-FRD和鼠合胞素-A的ERV合胞素的序列;
b)孵育a)中获得的转染细胞,使得它们产生用ERV合胞素假型化且包装有所述异源目标基因的所述稳定的慢病毒颗粒;和
c)收获并浓缩b)中获得的稳定的慢病毒颗粒,以获得浓缩稳定的慢病毒颗粒制剂,其在制剂的冷冻和解冻之前或之后包含高于1x105ng p24/mL的物理颗粒的滴度和/或高于2x104TU/mL的感染性颗粒的滴度。
3.权利要求2所述的方法,其中所述慢病毒rev、gag和pol基因是HIV-1rev、gag和pol基因。
4.通过权利要求2或3任一项所述的方法获得的颗粒制剂。
5.权利要求1或4所述的颗粒制剂,其中感染性颗粒的滴度高于1x105TU/mL,和/或其中物理颗粒的滴度高于1.1x105ng p24/mL。
6.权利要求2或3所述的方法,其中感染性颗粒的滴度高于1x105TU/mL,和/或其中物理颗粒的滴度高于1.1x105ng p24/mL。
7.一种药物组合物,包含药学可接受的介质中的权利要求1、4或5任一项所述的颗粒制剂。
8.权利要求1、4或5任一项所述的颗粒制剂用于转导免疫细胞的非治疗体外用途。
9.权利要求1、4或5任一项所述的颗粒制剂用于工程化免疫细胞的非治疗体外用途。
10.权利要求1、4或5任一项所述的颗粒制剂在制备用于治疗免疫缺陷、自身免疫、感染性疾病或B细胞相关癌症的药物中的用途。
11.一种药物组合物,包含药学可接受的介质中的用权利要求1、4或5任一项所述的颗粒制剂感染的B细胞或树突细胞。
12.获得经修饰以表达异源目标基因的免疫细胞的非治疗离体方法,包括用权利要求1、4或5任一项所述的颗粒制剂感染免疫细胞,任选地之前刺激的步骤。
13.权利要求12所述的非治疗离体方法,其中它是在LAH4肽或其功能性衍生物存在下进行,和/或其中所述免疫细胞选自B细胞、树突细胞、单核细胞和巨噬细胞。
14.权利要求12或13任一项所述的非治疗离体方法,其中包括以下步骤:
-任选地,通过在包含IL-7的培养基中培养初始免疫细胞来刺激它们,然后
-在LAH4肽或其功能性衍生物存在下,用权利要求1、4或5任一项所述的颗粒制剂感染刺激或未刺激的初始免疫细胞。
