Description
本発明は、安定な偽型化レンチウイルス粒子及び特に治療におけるその使用に関する。本発明はまた、そのような安定な偽型化レンチウイルス粒子を調製するための方法を指す。本発明はまた、ベクトフシンの存在において、そのような偽型化レンチウイルス粒子を使用し、好ましくは最小限又は無の予備活性化を用いてヒト末梢血ナイーブB細胞及び単球細胞に形質導入するインビトロの方法に関する。
遺伝子治療アプローチは、しばしば、組換えウイルスベクターによる標的細胞の低い形質導入効率により妨げられる。レトロウイルスベクター、特にヒト免疫不全ウイルス1(HIV−1)ベースのレンチウイルスベクター(LV)は、遺伝子治療のための有望な媒体である。これらのベクターは、種々の疾患、例えば免疫不全、神経変性疾患もしくは神経学的疾患、貧血、又はHIV感染などを処置するために臨床応用において現在使用されている。
シンシチンは、融合特性を有する内在性レトロイルスウイルス(ERVシンシチン)エンベロープ糖タンパク質である(Dupressoir et al., 2005; Lavialle et al., 2013)。
ERVW−1遺伝子(ENSG00000242950;シンシチン 1又はHERV−Wとしても公知である)によりコードされるヒト内在性レトロイルスエンベロープ糖タンパク質は、特許出願EP2385058においてその融合特性について記載されている。前記出願には、合胞体の形成による、癌処置におけるその使用が記載されている。
一部の試験では、293T細胞を形質転換することが可能な感染性ウイルスを得るために、HERV−Wエンベロープを用いてHIV−1ビリオン(MLV粒子ではない)を偽型化する可能性が示されている。しかし、そのようなHERV−W粒子は、濃縮、凍結、及び解凍によって力価がかなり低下したため、安定ではなかった(An et al, Journal of Virology, Apr 2001, p.3488-3489)。R細胞質内領域の欠失が、HIV−1由来の遺伝子導入ベクターの融合及び偽型力価を増加させるために使用された(Lavillette et al 2002)。ERVFRD−1遺伝子(ENSG00000244476;シンシチン2又はHERV−FRDとしても公知である)によりコードされる糖タンパク質が、SIVベクターを偽型化するために使用され、報告によれば、HIV又はMLV偽型として作用するが、非常に低い力価であり、機能試験を妨げ、HERV−FRD HIV偽型に関する情報はほとんど入手できない(Blaise et al, Journal of Virology, Jan 2004, p.1050-1054)。
本発明は、目的の異種遺伝子を含む安定な偽型化レンチウイルス粒子を得るための方法に関し、以下の工程を含む:
a)適当な細胞株において少なくとも1つのプラスミドをトランスフェクトすることであって、前記の少なくとも1つのプラスミドは目的の異種遺伝子、レトロウイルスのrev、gag、及びpol遺伝子、ならびにERVシンシチンをコードする核酸を含む;
b)ERVシンシチンを用いてそれぞれ偽型化された安定なレンチウイルス粒子を産生するようにa)において得られたトランスフェクト細胞をインキュベートすること、及び目的の異種遺伝子をパッケージングすること;及び
c)b)において得られた安定なレンチウイルス粒子を収集し、濃縮すること。
本発明に従った方法によって、目的の異種遺伝子を含む安定な偽型化レンチウイルス粒子の高い物理力価ならびに高い感染力価を得ることが可能になる。
本発明者らは驚くべきことに、高い融合特性を有するERVシンシチン、例えばヒトシンシチン1(HERV−W)、ヒトシンシチン2(HERV−FRD)、及びマウスシンシチンAなどを、組換えHIV−1由来レンチウイルスについての偽型を得るために使用してもよいことを発見した。実施例において明らかに実証するように、結果は、細胞株に対して選択的指向性を有する、目的の遺伝子を含む安定で感染性のレンチウイルス粒子を産生することが可能であることを示す。驚くべきことに、シンシチン偽型化レンチウイルス粒子によって、注目すべきことに、ベクトフシン1(カチオン性形質導入添加剤)の存在においてヒト初代Bリンパ球ならびにCD11c+骨髄細胞及び/又は樹状細胞を効率的に形質導入することができる。

「高い物理力価」は、200ng p24/mLより高い、好ましくは210ng p24/mLより高い、さらにより好ましくは300ng p24/mLより高い、工程b)の終了時に産生される粒子の力価を意味する。より好ましくは、工程b)の終了時に産生される、ERVシンシチンを用いて偽型化された粒子(例えばHERV−Wなど)の物理力価は、300ng p24/mLより高く、好ましくは400ng p24/mLより高い。より好ましくは、工程b)の終了時に産生される、ERVシンシチンを用いて偽型化された粒子(例えばHERV−FRDなど)の物理力価は、210ng p24/mLより高く、好ましくは300ng p24/mLより高い。
「高い物理力価」はまた、1×105ng p24/mLより高い、好ましくは1.1×105ng p24/mLより高い、より好ましくは1.5×105ng p24/mLより高い、工程c)の終了時に産生される(即ち、濃縮される) 粒子の力価を意味する。より好ましくは、工程c)の終了時に産生される、ERVシンシチンを用いて偽型化された粒子(例えばHERV−Wなど)の物理力価は、2.2×105ng p24/mLより高い、好ましくは3.0×105ng p24/mLより高い、さらにより好ましくは4、0×105ng p24/mLより高い。より好ましくは、工程c)の終了時に産生される、ERVシンシチンを用いて偽型化される粒子(例えばHERV−FRDなど)の物理力価は、1.1×105ng p24/mLより高い、好ましくは1.5×105ng p24/mLより高い。
「高感染力価」は、2E+04TU/mlより高い、好ましくは1E+05TU/mlより高い、より好ましくは1E+06TU/mlより高い、さらにより好ましくは2+06TU/mlより高い、工程c)の終了時に産生される感染性粒子の力価を意味する。より好ましくは、工程c)の終了時に産生される、HERV−Wを用いて偽型化された粒子の感染力価は、1,2E+05TU/mlより高く、好ましくは2E+05TU/mlより高く、より好ましくは 1E+06TU/mlより高く、さらにより好ましくは1.5E+06TU/mlより高い。より好ましくは、工程c)の終了時に産生される、HERV−FRDを用いて偽型化された粒子の感染力価は、2E+04TU/mlより高く、好ましくは2E+05TU/mlより高く、より好ましくは1E+06TU/mlより高く、さらにより好ましくは2.5E+06TU/mlより高い。好ましくは、そのような高感染力価は、293T細胞感染及び本明細書中の実施例に記載する滴定方法を使用して測定する。
そのようなレンチウイルス偽型化粒子は、このように、それらの融合特性及び感染特性、ならびに特にインビボで目的の遺伝子を送達するその能力を維持しながら、遠心分離及び/又は凍結することができる。ここでも、本発明に従った目的の異種遺伝子をパッケージングする安定な偽型化レンチウイルス粒子は、高い物理力価ならびに高い感染力価を提示する。
「目的の遺伝子」は遺伝子の機能的なバージョンを意味する。機能的バージョンとは、前記遺伝子の野生型バージョン、同じ科に属する変異型遺伝子、又はコードされたタンパク質の機能性を保存する切断型バージョンを意味する。
目的の遺伝子は、患者において欠損している又は非機能的である遺伝子、あるいは免疫原性タンパク質(例えばウイルスタンパク質、細菌タンパク質、又は腫瘍抗原など)をコードする遺伝子から由来しうる。目的の異種遺伝子はまた、レポーター遺伝子(診断目的のため、及び標的タンパク質のリガンドを同定するために有用)、自殺遺伝子、又は治療用RNAをコードする(即ち、標的DNA又はRNA配列に相補的なアンチセンスRNA、又はgRNA、RNAi、例えばshRNAなどをコードする)遺伝子でありうる。目的の遺伝子は、コードされたタンパク質、ペプチド、又はRNAを産生することができる。
「偽型」は、以下を含むレンチウイルス粒子を意味する:
−ウイルスから由来するエンベロープ糖タンパク質であって、前記ウイルスは前記レンチウイルス粒子が由来するウイルスとは異なる;
−修飾されたエンベロープ糖タンパク質。そのような場合において、天然ウイルスエンベロープ糖タンパク質は、変異により又は任意の他のアミノ酸修飾により修飾されうる;又は
−キメラ糖タンパク質。そのような糖タンパク質は、ウイルス糖タンパク質の少なくとも一部と別の配列の間での融合タンパク質である。
一実施形態において、適当な細胞株は、目的の異種遺伝子、レトロウイルスのrev、gag、及びpol遺伝子、ならびに好ましくは誘導型でERVシンシチン(例えばHERV−W、HERV−FRD、又はマウスシンシチンAなど)をコードする核酸である5つの配列の少なくとも1つ、最大4つを既に発現している。そのような場合において、工程a)は、前記細胞株においてまだ発現されていない少なくとも1つの配列を含む少なくとも1つのプラスミドを用いて前記細胞株をトランスフェクトする工程を含む。プラスミド混合物、又は単一プラスミド(1つだけのプラスミドが使用される場合)は、工程a)において前記細胞株中にトランスフェクトされた場合、前記細胞株が上記の5つの全ての配列を発現するように選ぶ。
例えば、適当な細胞株がレトロウイルスのrev、gag、及びpol遺伝子を発現する場合、トランスフェクトされるプラスミド又はプラスミドの混合物は、発現される残りの配列、即ち、目的の異種遺伝子及びERVシンシチン(例えばHERV−W、HERV−FRD、又はマウスシンシチンAなど)をコードする核酸を含む。
−目的の異種遺伝子、
−rev、gag、及びpol遺伝子、ならびに
−以前に記載したERVシンシチンをコードする、特にHERV−W、HERV−FRD、又はマウスシンシチンAをコードする核酸を含む。
−第1のプラスミドは目的の遺伝子を含み、
−第2のプラスミドはrev遺伝子を含み、
−第3のプラスミドはgag及びpol遺伝子を含み、ならびに
−第4のプラスミドは、以前に記載したERVシンシチンをコードする、特にHERV−W、HERV−FRD、又はマウスシンシチンAをコードする核酸を含む。
前記四重トランスフェクションは、好ましくは、4つのプラスミド間の特定の比率で実施する。異なるプラスミド間のモル比は、産生のスケールアップを最適化するために適合させることができる。当業者であれば、目的のレンチウイルスを産生するために使用する特定のプラスミドにこのパラメータを適応することができる。特に、第1、第2、第3、第4のプラスミドの重量比は、好ましくは、(0.8−1.2):(0.1−0.4);(0.5−0.8):(0.8−1.2)、より好ましくは約1:0.25;0.65:1である。
ERVシンシチン(例えば以前に定義したERVシンシチンなど)をコードし、より好ましくはHERV−W、HERV−FRD、又はマウスシンシチンAをより優先的にコードする核酸は、DNA又はcDNA配列である。好ましくは、それは、配列番号1、2、又は40にそれぞれ列挙するcDNA配列、あるいは、そのような配列番号1、2、又は40と少なくとも80%、好ましくは少なくとも90%、より好ましくは少なくとも95%、より好ましくは少なくとも99%の同一性をそれぞれ提示する配列に対応する。好ましくは、工程a)は、少なくとも配列番号3又は4に列挙するcDNA配列を含む、好ましくはそれからなる、少なくともプラスミドのトランスフェクションを含む。
用語「同一性」は、2つのポリペプチド分子間又は2つの核酸分子間の配列類似性を指す。両方の比較配列中の位置が同じ塩基又は同じアミノ酸残基により占められる場合、それぞれの分子はその位置で同一である。2つの配列間の同一性のパーセンテージは、2つの配列により共有されるマッチング位置の数を、比較位置の数で除し、100を掛けた数に対応する。一般的に、2つの配列を整列させて最大の同一性を与える場合に比較を行う。同一性は、例えば、GCG(Genetics Computer Group、GCG Package、Version 7(ウィスコンシン州マジソン)用のプログラムマニュアル)パイルアッププログラム、又は配列比較アルゴリズム(例えばBLAST、FASTA、又はCLUSTALWなど)のいずれかを使用したアラインメントにより算出してもよい。
本発明の好ましい実施形態に従い、工程a)のトランスフェクションは、リン酸カルシウムを使用して行う。
トランスフェクション後、トランスフェクト細胞は、このように、トランスフェクション後20〜72時間、特に24時間後に含まれる時間にわたり増殖させる。
特定の実施形態において、細胞を培養するために使用される培地は、糖(例えばグルコースなど)を含む古典的培地(例えばDMEMなど)である。好ましくは、培地は無血清培地である。培養は、懸濁液中の細胞の培養に適応させた多数の培養装置(例えばマルチスタックシステム又はバイオリアクターなど)で行ってもよい。バイオリアクターは、使い捨て(ディスポーザブル)又は再利用可能なバイオリアクターでありうる。バイオリアクターは、例えば、培養容器又はバッグ及びタンクリアクターより選択してもよい。非限定的な代表的なバイオリアクターには、ガラスバイオリアクター(例、B-DCU(登録商標)2L-10L、Sartorius)、ロッキング運動撹拌機、例えばウェーブバイオリアクター(例、Cultibag RM10L-25L、Sartorius)などを利用した使い捨てバイオリアクター、使い捨て撹拌タンクバイオリアクター(Cultibag STR(登録商標)50L、Sartorius)、又はステンレス鋼タンクバイオリアクターが含まれる。
好ましくは、遠心分離は、1時間30分〜2時間30分間2℃〜5℃、好ましくは約4℃の温度で45000〜55000gの間のスピードで実施する。この工程の終了時に、粒子を遠心分離物の形態で濃縮し、これを使用してもよい。
「医薬的に許容可能な担体」は、適宜、哺乳動物、特にヒトに投与した場合、有害な、アレルギー性の、又は他の都合の悪い反応を産生しない賦形剤を指す。医薬的に許容可能な担体又は賦形剤は、任意の型の非毒性固体、半固体もしくは液体充填剤、希釈剤、カプセル化材料、又は製剤補助剤を指す。
好ましくは、医薬組成物は賦形剤を含み、それは、注射することが可能な製剤用に医薬的に許容可能である。これらは、特に、等張性の無菌生理食塩水(リン酸一ナトリウムもしくはリン酸二ナトリウム、塩化ナトリウム、塩化カリウム、塩化カルシウム、もしくは塩化マグネシウムなど、又はそれらの塩の混合物)又は乾燥した、特に凍結乾燥した組成物であってもよく、それによって、場合に依存し、滅菌水又は生理食塩水の添加時に注射溶液の構成が可能になる。
注射可能な使用のために適切な医薬形態には、滅菌水溶液又は懸濁液が含まれる。この溶液又は懸濁液は、エンベロープを有するウイルスに適合し、標的細胞中へのウイルスの侵入を妨げない添加剤を含みうる。全ての場合において、形態は無菌でなければならず、簡単な注射可能性が存在する程度まで流動性でなければならない。それは、製造及び保存の条件下で安定でなければならず、微生物(例えば細菌及び真菌など)の混入作用に対して保存されなければならない。適当な溶液の例は、緩衝液、例えばリン酸緩衝食塩水(PBS)などである。
実際に、本願において提供する結果は、本発明のシンシチン偽型化レンチウイルスベクターがヒトにおける免疫療法において使用することができ、ヒト疾患におけるB細胞を機能的に補正することができることを明らかに実証する。さらに、本発明のシンシチン偽型化ベクターは、インビボでB細胞を標的とすることができる。本明細書において示すデータは、遺伝子導入が長期間にわたり観察されるため、これらの新たなツールがインビボで非常に良好に許容されることを例証した。インビボでの適用のために、標的細胞(例えばB細胞など)の効率的な形質導入を、ベクトフシン1を必要とすることなく得ることができる。
このように、安定な偽型化レンチウイルス粒子は、遺伝子治療もしくは免疫療法において、又はワクチンとして、又は免疫予防において使用されうる。
遺伝子治療は、遺伝子導入、遺伝子編集、エキソンスキッピング、RNA干渉、トランススプライシング、又は核、ミトコンドリア、もしくは共存DNA(細胞中に含まれるウイルス配列)中に含まれるものを含む、細胞中の任意のコード配列又は調節配列の任意の他の遺伝子改変により実施することができる。
遺伝子治療の主な2つの型は以下である:
−欠損/異常遺伝子の置換を目的とした治療:これは置換遺伝子治療である;
−遺伝子編集を目的とした治療:そのような場合において、目的は、目的とする遺伝子が発現されるように必要なツールを細胞に提供することである:これは遺伝子編集治療である。
−X連鎖性重症複合免疫不全症(SCID−X1)(患者はT−NK−B +表現型を有する)において、又は遺伝子治療により既に処置されているが、B細胞補正が誘発されなかったSCID−X1患者についてガンマ鎖をコードする遺伝子;
−共通の可変免疫不全症(CVID)において変異しているTACI(膜貫通アクチベーター及びカルシウムモジュレーター及びシクロフィリンリガンド相互作用因子)をコードする遺伝子TNFRSF13B;
−共通の可変免疫不全症(CVID)において変異しうるCD19又はBAFFR遺伝子;
−特にCD40をコードする遺伝子の変異により起こされる高免疫グロブリンMに関与する遺伝子のいずれか;
−幹細胞ベースの遺伝子治療により既に処置され、B細胞補正が不完全であるWAS患者を含むWiskott−Aldrich症候群において変異したWAS遺伝子;
−ファンコニ貧血サブタイプのいずれか、例えばFancA又はFancCにおいて変異しているFanc遺伝子;
−高免疫グロブリンEに関与する遺伝子のいずれか。
このように、これらの遺伝子は目的の遺伝子として使用することができた。
遺伝子編集の特定の例は、ワルデンストレームマクログロブリン血症の処置であろう:そのような疾患において、(MYD88)の点変異(L265P)が患者の80〜88%のB細胞中で観察されており、これらの腫瘍細胞の生存及び持続性に寄与する。このように、罹患患者においてこの遺伝子の補正バージョンを遺伝子編集することにより、これは、この疾患に対する効果的な治療に寄与しうる。
形質導入された免疫細胞がマクロファージである場合、次に治療は、抗炎症治療、寛容誘導、又はワクチン接種でありうる。
形質導入された免疫細胞がB細胞である場合、次に治療は、自己免疫疾患を処置するためでありうる。
形質導入された免疫細胞が樹状細胞である場合、次に治療はワクチンとしてでありうる。
例えば、同種異系の臓器ドナーに特異的な主要又は副組織適合抗原を用いて形質導入されたレシピエントの未成熟樹状細胞を使用し、レシピエントにおいて同種異系の臓器移植寛容を誘導してもよい。
また、抗原提示細胞(例えばヒトB細胞、ヒト骨髄性樹状細胞、又は単球など)の遺伝子操作を介した免疫療法の適用は、感染性疾患に対して防御するためのワクチン接種の目的のための抗原提示のために;癌を処置するために;あるいは炎症性免疫成分を用いて疾患(例えば糖尿病又はアルツハイマー病など)を処置するため、又は臓器もしくは骨髄移植を促進するために、細胞特異的又は抗原特異的免疫寛容を誘導する目的のために、これらの細胞中に、免疫調節配列と関連する又はしない抗原性配列を含む組換え遺伝子発現カセットを導入することにより見出されうる。
免疫細胞は、免疫系に含まれる細胞である。それは、特にB細胞、T細胞、NK細胞、マクロファージ、及び樹状細胞を含む。
B細胞(又はBリンパ球)は、抗体の産生に関与する免疫細胞であり、体液性免疫応答に含まれる。
樹状細胞は免疫細胞であり、それはアクセサリーである:それは抗原提示細胞である。その主な機能は、抗原材料を処理し、それをその細胞表面上でT細胞に提示することである。
T細胞(又はTリンパ球)は、細胞媒介性免疫応答に含まれる免疫細胞である。それは、キラーT細胞、ヘルパーT細胞、及びガンマデルタT細胞より選んでもよい。
このように、本発明はまた、免疫細胞、好ましくはB細胞を形質導入することによる治療のための、ERVシンシチン(例えば以前に定義したERVシンシチンなど)を用いて偽型化され、HERV−W、HERV−FRD、又はマウスシンシチンAを用いてより優先的に偽型化され、目的の異種遺伝子をパッケージングする安定なレンチウイルス粒子の使用に関する。
ヒトB細胞における生物工学はまた、遺伝子、例えば抗アポトーシス遺伝子又はB細胞癌遺伝子(例えばBcl6又はBclXなど)などを用いて抗原応答性ヒトB細胞(例えば、CD27 +胚中心B細胞)を、「Kwakkenbos MJ et al. Nat Med. 2010 Jan;16(1):123-8. doi: 10.1038/nm.2071. Generation of stable monoclonal antibody-producing B cell receptor-positive human memory B cells by genetic programming」に記載されているように、病原体に応答するそのような細胞を増殖させ、それらの抗体配列を選択する目的のために、不死化させるための遺伝子導入を使用することからなりうる。
「免疫不全」は、微生物(細菌性、ウイルス性、寄生虫性)感染の数もしくは重症度の増加又は抗腫瘍サーベイランスの低下及び癌の頻度の増加に導く状態を意味する。
この目的のために、目的の異種遺伝子は、自殺遺伝子、「ドミナントネガティブ」遺伝子、標的細胞活性化をブロックする抗体をコードする遺伝子、癌遺伝子の発現を補正するタンパク質をコードする遺伝子、又は B細胞の形質転換体ウイルス(例えばバーキット症候群の場合におけるEBVなど)のウイルスサイクルを低下又は変化させるタンパク質をコードする遺伝子でありうる。
好ましくは、免疫細胞は、B細胞、T細胞、樹状細胞、及びマクロファージより選ぶ。
好ましくは、免疫細胞はナイーブである。好ましい実施形態に従い、ナイーブ免疫細胞は以前に刺激されていない。
別の実施形態に従い、刺激が使用される場合、刺激工程は最小であり、ナイーブB細胞の生存を保つように、ナイーブ免疫細胞を、IL−7を含む培地中でインキュベートすることにより実施する。
別の実施形態に従い、本発明に従った目的の異種遺伝子を発現するように改変された免疫細胞を得るためのエクスビボのプロセスは、以下の工程も含みうる:
−場合により、IL−7を含む培地中でナイーブ免疫細胞をインキュベートすることによりそれらを刺激し、次に

好ましくは、LAH4ペプチド又はその機能的誘導体、好ましくはベクトフシン1が、特に7〜20μg/ml、より好ましくは10〜15μg/mlの濃度で存在する。
LAH4ペプチド又はその機能的誘導体は、配列番号5〜31に示す配列の間で選択してもよい:
好ましくは、LAH4ペプチドの機能的誘導体を使用する。好ましくは、前記の機能的誘導体はLAH4−A4であり、ベクトフシン1とも呼ばれ、配列番号18を有する。
ヒト胚性腎臓293T細胞及びヒト結腸直腸癌HCT116細胞(CCL−247;ATCC、バージニア州マナサス)を、10%の熱不活化ウシ胎児血清(FCS)(Life Technologies)を添加したダルベッコ改変イーグル培地(DMEM+グルタマックス)(Life Technologies、フランス、サントーバン)中で37℃、5%CO2で培養した。ヒト絨毛癌BeWO細胞(CCL−98;ATCC)を、10%FCSを添加したHam’s F12K培地(Life technologies)中で培養した。Raji細胞及びJurkat細胞をXVivo 20培地中で感染させた。
ヒトシンシチン1(ERVW−1−001)又はヒトシンシチン2(ERVFRD−1)をコードするプラスミドpCDNA3シンシチン1及びpCDNA3シンシチン2を、NHeI及びXhoI制限酵素を使用し、EnsemblゲノムブラウザENST00000493463及びENSG00000244476転写物配列に対応するそれぞれの合成DNA配列(Gencust、ルクセンブルク、デュドランジュ)をpCDNA3プラスミド(Invitrogen、カリフォルニア州カールスバッド)中にクローニングすることにより構築した。プラスミド配列は、2本鎖配列決定により確認した。シンシチン2プラスミド機能性は、シンシチン2であると予想される65〜70Kdのタンパク質を検出する抗HERV−FRD抗体(LS-BIO、フランス、ナンテール)を使用し、293T細胞のトランスフェクションに続いてタンパク質抽出物をイムノブロッティングすることにより確認した。プラスミドを、DNA RNA精製Nucleobondキット(Macherey-Nagel、ドイツ、デューレン)を使用してエンドトキシンフリーで産生した。シンシチン1又はシンシチン2のいずれかを用いて偽型化したレンチウイルス粒子は、リン酸カルシウムを使用し、4つのプラスミドを用いた293T細胞の一過性トランスフェクションにより産生した。各々のT175cm2のフラスコに、HIV−1 gagpol遺伝子を発現する14.6μgのpKLgagpol、HIV−1 rev配列を発現する5.6μgのpKRev、22.5μgの遺伝子導入カセット(ヒトホスホグリセレートキナーゼ(PGK)プロモーターの制御下で増強緑色蛍光タンパク質(GFP)を発現するpCCL PGK GFP(Charrier et al, 2011)又はヒトPGKプロモーターの制御下で切断型の神経成長因子受容体(NGFR)を発現するpCCL PGKデルタNGFR)、及び22μgのpCDNA3シンシチン1プラスミド又はpCDNA3シンシチン2プラスミドのいずれかを用いてトランスフェクトした。培地を翌日に交換し、ペニシリン及びストレプトマイシンならびに10%FCSを添加したDMEM 4.5g/Lグルコースにより置換した。24時間後、ウイルス粒子を含む培地を回収し、低速で遠心分離し、滅菌濾過し(0.22μm)、4℃で2時間にわたり50,000gの超遠心分離(1900RPM、ローターSW28を用いたBeckman超遠心分離機を使用)により濃縮した。ペレットをPBS中に再懸濁し、分注し、−80℃で保存した。VSVg偽型化粒子は、報告されているように(Merten et al, 2011)、一過性トランスフェクションによっても産生した。
ベクトフシン1(VF1)ペプチドは、標準的なフルオレニル−メチルオキシ−塩化カルボニル固相ペプチド合成、それに続くHPLC及び質量分析(Gencust)により合成した。ペプチドをH2O中に可溶化し、分注し、使用まで−80℃で保存した。形質導入実験のために必要である場合、VF1を解凍し、培地中に再懸濁し、ベクター及び細胞に最終濃度12μg/mlで加えた。硫酸プロタミン(PS)(Sigma-Aldrich、ミズーリ州セントルイス)及びポリブレン(PB)(Sigma-Aldrich)も即時にベクター及び細胞と希釈し、形質導入培養中でそれぞれ8μg/ml及び6μg/mlの最終濃度で使用した。
濃縮して凍結保存したベクターを使用前にタイトレーションした。物理的粒子力価を、以前に報告されたように(Charrier et al, 2011)、p24 ELISA(Alliance(c)HIV-1 Elisaキット、Perkin-Elmer、フランス、ヴィルボン=シュル=イヴェット)により決定した。感染力価は、12μg/mlのVF1の存在においてHEK293T細胞にベクターの連続希釈液を加えることにより決定し、3日後、293T細胞をフローサイトメトリーにより分析し、形質導入単位力価(TU/ml)を算出し、又はqPCRにより、標準的な計算を使用して感染性ゲノム(ig/mL)を測定した(Kutner et al, 2009)。
EDTAを用いて回収した末梢血は、Etablissement Francaisdu Sang(フランス、エヴリー)から購入した。Ficoll勾配遠心分離(Eurobio、フランス、レ・ジュリス)により精製した末梢血単核細胞(PBMC)を、形質導入のために、無血清培地X−VIVO 20(Lonza、フランス、ルヴァロワ=ペレ)中に懸濁した。一部の実験において、サイトカインIL−7(10ng/μL)(Miltenyi Biotech、ドイツ、ベルギッシュ・グラートバッハ)を形質導入に先立ち一晩培地に加えた。形質導入のために、LV−Syn1もしくはLV−Syn2ベクター(1.105TU/ml)又はLV−VSVg(1.108TU/ml)を、VF1(12μg/ml)の存在において細胞に加えた。6時間後、培地を、10%FCS及びIL−7(10ng/mL)を添加したXVivo20に交換し、細胞を7日間まで培養した。形質導入したB細胞を増殖させるために、CD40L(2μg/ml)及びIL−4(20ng/mL)を培地に加え、細胞をこれらの条件において2週間まで培養した。骨髄性樹状細胞を増殖させるために、hGM−CSF(50ng/mL)及びIL−4(20ng/mL)を培地に加えた。
PBMCをIL−7と一晩培養し、次にベクターを用いて感染させた。6時間後、細胞を洗浄し、B95.8マーモセット細胞上清の存在において、EBV不死化のための標準手順を使用してシクロスポリンと培養した。培地は週2回交換した。
遺伝子治療により処置されたSCID−X1患者のPBMCを 37℃で迅速に解凍し、次にPBSで2回洗浄した。トリパンブルー計数後、細胞を2μMのCFSE(Molecular Probes、英国ケンブリッジ)で標識し、Lv−Syn1 IL2rgベクターを用いて形質導入し、96ウェル丸底プレート中でCD40リガンド [2μg/ml](Miltenyi Biotec)及びIL−21[50ng/mL](Miltenyi Biotec)の非存在又は存在において培養する(3.105/200μL)。3、6、及び7日間の培養後、B細胞形質導入(即ち、NGFR又はIL2Rg発現)、B細胞増殖(即ち、CFSE希釈)、及びCD19+CD27+細胞の頻度をフローサイトメトリーにより決定した。培地中のヒトIgG分泌をELISA(Sigma-Aldrich、ミズーリ州セントルイス)により決定した。
PBMCを、2μMCFSE(Molecular Probes、英国ケンブリッジ)を用いて標識し、96ウェル丸底プレート中でCD40リガンド[2μg/ml](Miltenyi Biotec)及びIL−21[50ng/mL](Miltenyi Biotec)の非存在又は存在において培養した(3.105/200μL)。3、6、及び7日間の培養後、B細胞形質導入(即ち、NGFR又はIL2Rg発現)、B細胞増殖(即ち、CFSE希釈)、及びCD19+CD27++形質芽細胞の頻度をフローサイトメトリーにより決定した。ヒトIgG分泌をELISA(Sigma-Aldrich、ミズーリ州セントルイス)により決定した。
フローサイトメトリーのために使用した抗体は、BD-Biosciences(フランス、ル・ポン=ド=クレ)から購入し、PEコンジュゲート抗CD3及び抗CD1a、Alexa−Fluor 700コンジュゲート抗CD19、APC−H7コンジュゲート抗CD45及び抗IgD、CF594コンジュゲート抗IgM、PeCy7コンジュゲート抗CD27及び抗HLA−DR、APCコンジュゲート抗CD14、V450コンジュゲート抗CD11cを含む。細胞に抗体を加える前に、Fc受容体を、細胞をガンマグロブリン(1mg/mL)(Sigma Aldrich)と4℃で15分間にわたりインキュベートすることによりブロックした。飽和量の抗体を次に、0.1%ウシ血清アルブミン(BSA)(Sigma Aldrich)を伴うPBS中で、4℃で30分間にわたり加えて、細胞を2回洗浄した。手順の終了時に、7−アミノ−アクチノマイシンD(0.3mg/mL)(Sigma-Aldrich)を加えて、死細胞を排除した。受容体試験のために、ウサギポリクローナル抗SLC1A5(ASCT2)(Abcam、フランス、パリ)、抗MFSD2a(Origene、メリーランド州ロックビル)、又はウサギ免疫グロブリン対照(Origene)を使用し、Alexa647コンジュゲートヤギ抗ウサギ抗体(Thermo Fisher Scientific、メリーランド州ウォルサム)により明らかにした。取得は、Divaソフトウェア(BD-Biosciences)を使用してLSRIIで行い、分析は、Kaluzaソフトウェア(Beckman-Coulter、Villepinte、France)を用いて行った。
ヒトIL2Rgの検出は、PEコンジュゲート抗ヒトCD132抗体(BD bioscience)を使用して実施した。
全RNAを、SV全RNA単離物(Promega、フランス、シャルボニエール=レ=バン)を使用して精製細胞から抽出した。残留DNAを、遊離DNAキット(Ambion、フランス、クールタブフ)を使用してサンプルから除去した。cDNAは、逆転写PCR(Invitrogen)用のプロトコールSuperscript II第一鎖合成システムに従って、ランダムヘキサマーを使用して1μgのRNAから合成した。リアルタイムPCRは、プロトコールSybr Green PCR Master Mix(Applied Biosystem、米国カリフォルニア州フォスターシティ)に従い、0.1μMの各々のプライマーを用いて、LightCycler 480システム(Roche、スイス、バーゼル)を使用して実施した。ASCT2又はMFSD2Aのいずれかの増幅のために使用したプライマー対は、以前に記載されている(Cornelis et al, PNAS, 2008及びToufailly et al, placenta, 2013)。ASCT2プライマーの配列は(センス、5’−GGCTTGGTAGTGTTTGCCAT−3’;アンチセンス、5’−GGGCAAAGAGTAAACCCACA−3’)、及びMFSD2Aプライマーの配列は(センス、5’−CTCCTGGCCATCATGCTCTC−3’;アンチセンス、5’−GGCCACCAAGATGAGAAA−3’)である。本発明者らはまた、TFIID(転写最終II D)プライマーを標準化として使用した。TFIIDプライマー配列は(センス、5’−ACGGACAACTGCGTTGATTTT−3’;アンチセンス、5’−ACTTAGCTGGGAAGCCCAAC−3’)である。結果は、以下の式を使用し、倍率変化で表す:相対存在量=ΔCtを伴う2^−ΔΔCt=Ct ASCT2又はMFSD2A−Ct TFIID及びΔΔCt=ΔCtsample−ΔCtcalibrator。
チャールズリバーから購入した7週齢の雌NSGマウス(Nod/Scid/gc−/−)に、100μL容量中の107個のPBMCを眼窩後洞に注射した。24時間後、マウスに、150μLの未希釈のLV−Syn1ベクターを尾静脈に静脈注射した。7日後、マウスを屠殺し、脾臓を回収し、ACKを用いて溶解し、赤血球枯渇画分をFACSにより分析した。
脾臓細胞は、ACKを用いた赤血球溶解に続いて6週齢の雌C57Bl/6マウスから得た。細胞を、1又は5×105TU/mlのΔNGFRをコードするLV−SYn1又はLV−Syn2及びVF1(12μg/ml)の存在において、100μL中106細胞/mLの濃度で、10%FCS及びベータ2メルカプトエタノールを添加したRPMI中で培養した。3日後、ΔNGFRの発現を、生存細胞でのFACSにより測定した。
統計的有意性を、GraphPad Prismソフトウェア(GraphPad Inc、カリフォルニア州ラホヤ)を使用して、一元配置ANOVA分析、マンホイットニー又はスチューデントのt検定により指定通りに評価した。
2つのヒト内在性レトロイルスエンベロープ糖タンパク質であるシンシチン1及びシンシチン2を、組換えHIV−1由来のLVについての可能性のある新たな偽型として探索した。これらの糖タンパク質の全長cDNAを、293T細胞中の一過性LV産生系において発現させた。トランスフェクション工程のためのシンシチン1及びシンシチン2プラスミドの量の最適化によって、培地中のp24レベルに基づくLV粒子の産生が増加した(補足図S1A)。定められた条件(材料及び方法を参照のこと)において、これらのエンベロープのいずれかを用いて偽型化された安定な感染性粒子を産生することが可能であった。異なる導入遺伝子(GFP及びdNGFR)を使用して、平均705ng p24/mL(n=7産生)で滴定されたLV−Syn−1及び平均496ng p24/mL(n=4産生)で滴定されたLV−Syn−2のローストックを収集した(表1)。これらのエンベロープのいずれかを用いて偽型化したレンチウイルス粒子は、VSVg−偽型化粒子に使用したのと同じ条件を使用した超遠心分離により成功裏に濃縮することができた(Charrier et al, 2011)。濃縮ストックを−80℃で凍結保存し、数ヶ月にわたり安定であった。LVストックの解凍時に、4.1±1.7E+05ng p24/mLの力価がLV−Syn1(n=6産生)について、及び1.7±0.6E+05ng p24/mL がLV−Syn2(n=6産生)について得られ、ベクターを、異なる導入遺伝子(GFP又はΔNGFR)をコードするように産生することができた(表1)。
VF1の存在において、293T細胞を用量依存的にシンシチン偽型化ベクターのいずれかで形質導入し、BeWo細胞において得られたレベルと同程度のレベルに達することが可能であった(図2A、B)。対照的に、293T細胞のわずか約1〜2%を、この添加剤を用いることなく形質導入した。VF1の増強効果は、LV−Syn1又はLV−Syn2を使用した293T細胞における反復実験にわたり統計的に有意であった(図2C)。シンシチン偽型化ベクターを用いた293T細胞の形質導入によって、2週間続く連続培養において検証したように、経時的な導入遺伝子の安定したゲノム組込みに導かれた(補足図S1B)。遺伝子発現のレベルは、細胞1個当たりのコピー数と一致し、経時的な導入遺伝子のサイレンシングの証拠は示さなかった(図2D)。
LV−Syn1及びLV−Syn2感染性に対するVF1ペプチドの増強効果の発見によって、293T細胞を使用した頑強な感染力滴定アッセイの開発が可能になった。293T細胞株の選択は、実験室においてVSVg偽型化LVを力価測定するために日常的に使用されるHCT116結腸癌細胞の形質導入の失敗により促された。HCT116は、試験した任意の濃度で、及びベクトフシン1の存在においてでさえ、LV−Syn1又はLV−Syn2を用いて形質導入できなかった(補足図S1C)。また、293T細胞が、より一貫した増殖パターン及び反復実験におけるより高い形質導入レベルの傾向のために、この感染力価アッセイについてはBeWo細胞よりも好まれた(補足図S1C)。アッセイのために定められた条件(材料及び方法を参照のこと)において、GFPをコードするLV−Syn1及びLV−Syn2の濃縮及び凍結保存バッチの感染力価は、それぞれGFP導入遺伝子については2.8±6×E+06TU/ml(n=6バッチ)及び2.7±6×E+06TU/ml(n=6バッチ)、ならびにdNGFR導入遺伝子については6.3±7.5×E+06TU/ml(n=3バッチ)及び4.4±1.5×E+06TU/ml(n=2バッチ)であった(表1)。このように、シンシチン1及びシンシチン2糖タンパク質によって、rHIVベクターを偽型化し、感染性であり、安定した遺伝子導入のために有用な安定な粒子を得ることができる。それらの感染性には補因子が要求され、細胞レベルで選択的である。
観察された効果が、非特異的であり、偽形質導入又は凝集時に生じうるベクトフシン1の人為的自己蛍光により起こされることを排除するために(Fenard et al 2013)、本発明者らは、ベクターを用いて処理し、経時的に培養した細胞中での前ウイルス組込みを確認した。ベクターを細胞に加えた後、本発明者らはCD40L及びIL−4を加えて8〜14日間にわたり培養物を活性化及び増殖させた。そのような培養において、本発明者らは、条件が全ての細胞の生存を確実にするのに最適ではないにもかかわらず、ベクターがベクトフシン1と一緒に加えられた場合に(表4)、特異的にベクターコピーを検出した。また、記憶B細胞の発生のために必須であると以前に報告されたCD40リガンド(CD40L)及びIL−21などのB細胞活性化シグナルを使用して、本発明者らは、形質導入されたB細胞が効率的に活性化されうることを確認した(表5)。CFSEマーキング及びフローサイトメトリーを使用してB細胞分裂及び導入遺伝子発現を検出することにより、本発明者らは、CD40L及びIL−21に応答することが可能な細胞のマーキングならびに活性化B細胞(形質芽細胞腫を含む)表現型を確認した(表5)。機能的B細胞の形質導入をさらに確認するために、本発明者らは、LV−Syn1ベクターを用いて感染させた直後にEBVを使用してPBMCを形質転換し、安定に組込まれたベクター配列を含むリンパ芽球細胞を得た(表4)。これらの結果によって、シンシチンベクターが、機能的な増殖能を保持して増殖する初代B細胞を安定的に形質導入できることが確認される。
新たに単離したマウス脾臓細胞を、ΔNGFRをコードするLV−Syn1又はLV−SYn2ベクターを用いて3日間にわたり培養した。補足図S7に示すように、これは、CD19+B細胞の非常に効率的な形質導入をもたらした。これらの培養物中では、ずっと少ないCD19細胞が比例して形質導入され、このように、ヒトと同様にマウスにおいてB細胞についてのシンシチン偽型化ベクターの特定の指向性が確認された。このデータはまた、シンシチン1及びシンシチン2糖タンパク質がマウスの対応物を認識することを示しており、これらの知見によって、これらのベクターを用いた将来の前臨床試験が促進されるであろう。
図3に示すように、CD19−CD3+CD11c+細胞の大きな割合が、事前の活性化を伴わず、ベクトフシン1の存在においてPBMC由来のLV−Syn1及びLV−Syn2で形質導入された。さらなる表現型分析により、そのようなCD11c細胞も高レベルのHLA−DRを発現し、シンシチン偽型化ベクターを用いて効果的に形質導入されることが示された(図5)。細胞はGM−CSF+IL−4において増殖し、CD1a、CD80、CD86を発現し、CD14を欠き、それらが未成熟樹状細胞であることを示唆した(補足図S3A)。また、単球細胞の表現型を有するCD14+HLA−DR+細胞もまた形質導入される(補足図S3B)。このように、LV−Syn1及びLV−Syn2の両方を、抗原提示細胞において導入遺伝子を発現させるために効果的に使用することができる。
末梢血T細胞は、シンシチン偽型化LVにより効率的に形質導入されなかったが、一部の細胞は、活性化時に形質導入されうる。IL−7を用いた事前の活性化に続き、少量の末梢血T細胞が、LV−Syn1又はLV−Syn2ベクターを用いて形質導入されたが、しかし、これらの効果は実験から実験まで一貫していなかった(図3及び表2)。事前の活性化がなければ、形質導入レベルは常に低かった。
このように、血球サブセットの異なる形質導入が、シンシチン偽型化ベクターを用いて観察される。
それぞれシンシチン1及びシンシチン2のエントリー受容体であるASCT2及びMFSD2aは、間接免疫蛍光検出を使用し、血液細胞サブセットの細胞表面上で検出した(図6A)。これらのマーカーを発現する細胞の最も高いパーセンテージはCD19+及びCD11c+細胞であったのに対し、T細胞はこれらの受容体の低レベルを発現した(図6B)。これらの結果はqRT−PCRにより確認し、ヒトB細胞がMFSD2a及びASCT2を発現することを初めて示す(図6−C)。ASCT2及びMFSD2aの発現は293T細胞でも見出されたのに対し、ベクターを用いて形質導入されていないHCT116細胞のずっと低いレベルであった(補足図S4)。試験した細胞では、受容体の発現は、達成された形質導入レベルと十分に相関した。
初代細胞で得られた結果を延長し、シンシチン偽型化ベクターの細胞特異性をさらに評価するために、本発明者らはヒト細胞株のパネルを形質導入しようと試みた。バーキットリンパ腫Raji B細胞及びJurkat T細胞を試験した。Raji B細胞は、シンシチン偽型化ベクターくぉ用いて形質導入することができた(補足図S5)。2×106TU/mlのLV−Syn2ベクターを使用してRaji細胞において得られた最高レベルは16%に達した(図4に見られるように、初代B細胞において106TU/mlで>90%に達したのに対し)。本発明者らは、Raji細胞がLVを用いた形質導入に許容性であることを確認したが、なぜなら、Raji細胞の88%形質導入が108TU/mlのLV−VSVgを用いて形質導入されたためである(補足図S5の対照、示さず)。1つの実験では、形質導入されたRaji細胞をフローサイトメトリーにより選別し、49日間にわたり培養し、長期間にわたる導入遺伝子の安定な組込みを実証した(補足表S2)。Jurkat T細胞は、シンシチン偽型化ベクターを用いて部分的に形質導入した(補足図S5)。このように、シンシチン偽型化LVは、一部の細胞株用の遺伝子導入ツールとして使用することができる。B細胞において、形質導入は、確立されたB細胞株よりも初代細胞においてより効果的であるように見える。
重症複合免疫不全症1型(SCIDX1)は、インターロイキン2受容体ガンマ鎖(IL2Rg)遺伝子中の変異により起こされるX連鎖性原発性免疫不全症である。同種異系骨髄移植によりこの疾患を治癒することができるが、しかし、一部の患者はB細胞における部分的キメラ化のみを伴い不完全に補正されたままである(Recher et al 2011)。5%未満のB細胞キメラ性を伴う患者は、免疫グロブリンを成熟させ、産生することが不可能なままである。それらのB細胞は、成熟した記憶IgG分泌B細胞になることが不可能なままである。実際に、IL2RgもB細胞の成熟のための必須シグナルを提供するIL−21のための受容体であることが最近発見された(Recher et al 2011)。いくつかの遺伝子治療治験が、造血幹細胞及び前駆細胞におけるIL2Rg遺伝子導入を使用してこの疾患を処置するために試みられている。しかし、患者の条件付けなしでは、遺伝子補正された細胞は、最新の遺伝子治療の治験(Hacein-Bey-Abina et al 2014)において示されるように、補正されたT細胞のみを産生し、B細胞は未補正のままである。結果として、そのような患者のB細胞は非機能的であり、患者は感染を予防するために免疫グロブリン注入に依存したままである。これにより、本発明者らは、遺伝子治療により完全に補正されていない患者において末梢B細胞にIL2Rgを移入するためにシンシチン偽型化ベクターを使用できるか否かを問うた。患者からの細胞を得た。患者の血液単核細胞はCD3+IL2Rg+T細胞を含むが、健常な個体とは対照的に、患者の血液CD19+B細胞は成熟マーカーCD27を発現せず、IL2Rgを発現せず(図7A及びB)、IgGを産生しなかった(表6)。IL2RgをコードするLVシンシチン1ベクターを用いた遺伝子導入時に、患者細胞は、恐らくは、培養中でのこれらの脆弱な細胞の高レベルの死亡率のために、活性化マーカーが細胞上で検出されなかったにもかかわらず、培地中でIgGを産生することによりCD40L+IL−21に応答することができた。これらの結果によって、末梢血B細胞が遺伝子導入により標的化され、SCIDX1において何らかの機能的補正を提供することができること、及びシンシチン偽型化LVがこの適用のための有用なツールであることが実証される。
LV−Syn1による初代ヒトB細胞の効率的な形質導入によって、このベクターがインビボで機能するか否かを試験することが促された。免疫不全のNSGマウスを、最初にマウス1匹当たり107のPBMCを用いて移植し、翌日にLV−Syn1の単回ボーラスを静脈内投与した。7日後、マウスを屠殺し、異なる組織を検証した。3匹中2匹のマウスにおいて、本発明者らは、GFPを少量(3〜15%)発現した脾臓中でのCD45+CD19+ヒトB細胞の存在を見出し、それらがインビボで形質導入されたことを示唆する(補足図S6)。これらの予備的結果は、インビボでヒトB細胞を標的化するためにシンシチン偽型化ベクターを使用する可能性を示唆する。
a.マウスシンシチンAを発現するプラスミドの生成。
pcDNA3.1真核生物発現プラスミドのHindIII及びXbaI部位中へのマウスシンシチンA cDNAのクローニングは、Genecust(ルクセンブルク、エランジュ)により生成され、以下のプライマーを使用してSyn−Aマウス遺伝子全長cDNA(EnsemblレファレンスENSMUSG00000085957)(配列番号33)(Tm=65℃;GC%=54%;Sigma)を含み、ライゲーションによりpUC−SynAプラスミドからのPCR増幅断片を挿入することによって実施した:フォワード5’AGCAAGCTTATGGTTCGTCCTTGG3’(配列番号32)(Tm=69,8℃;%GC=50%;Sigma、米国セントルイス)及びリバース5’AGCTCTAGACTAGACGGCATCCTC3’(配列番号33)(Tm=65℃;%GC=54%;Sigma)。プラスミドを配列決定(Beckman Coulter Genomics、英国テイクリー)により確認した。
b.Syn−A偽型化レンチウイルスベクターの産生。
DMEM+10%ウシ胎仔血清(FCS)においてT175cm2フラスコ中に播種したHEK293T細胞を以下の4つのプラスミド(フラスコ当たりの量)で同時トランスフェクトした:pKLgagpol(14.6μg)、pKRev(5.6μg)、pcDNA3.1−SynA(20μg)、及び導入プラスミドPRRL−SFFV LucII又はpRRL−SFFV−LucII−2A−ΔNGFR−WPRE(22.5μg)。24時間後、細胞を洗浄し、新鮮な培地を加える。翌日、培地を収集し、1500rpmで5分間にわたり遠心分離して清澄化し、0.45μmでろ過し、次に50000gの超遠心分離により12℃で2時間にわたり濃縮し、使用するまで−80℃で保存する。
c.シンシチンA偽型化LVの滴定。
物理力価を、他の型のLVと同様に、p24 ELISAにより決定した。感染力価を、マウスリンパ腫細胞系A20を使用して感染性ゲノム力価(IG/mL)として決定した。ベクターの連続希釈液を、ベクトフシン1(登録商標)(12μg/μL)の存在においてA20細胞に6時間にわたり加える。培地を新しくし、細胞を7日間にわたりインキュベートし、ゲノムDNAを得て、プライマー:PSIフォワード5’CAGGACTCGGCTTGCTGAAG3’(配列番号34)、PSIリバース5’TCCCCCGCTTAATACTGACG3’(配列番号35)、FAM(6カルボキシフルオレセイン)(配列番号36)を用いて標識されたPSIプローブ5’CGCACGGCAAGAGGCGAGG3’、Titinフォワード5’AAAACGAGCAGTGACGTGAGC3’(配列番号37)、Titinリバース5’TTCAGTCATGCTGCTAGCGC3’(配列番号38)、及びVICを用いて標識されたTitinプローブ5’TGCACGGAAGCGTCTCGTCTCAGTC3’(配列番号39)を用いて、iCycler 7900HT(Applied Biosystems)で二本鎖qPCRを使用して細胞1個当たりのベクターコピー数を測定する。
LV−SynAをコードするLucIIベクターを産生し、5×105TUのベクターをアルビノースC57Bl/6マウス中に静脈注射した。対照には、LV−VSVgをコードするLucII及びPBSが含まれた。生物発光の検出のために、マウスをケタミン(100mg/kg)/キシラシン(10mg/kg)を用いて麻酔し、100μLの150μg/ml Dルシフェリンを腹腔内投与し、10分後にCCDカメラISO14N4191(IVISルミナ、Xenogen、米国マサチューセッツ州)を用いて撮影した。3分間の生物発光画像を、15cmの視野、ビニング(分解能)係数4、1/fストップ及びオープンフィルターを使用して得た。目的の領域(ROI)を手作業で(各々の場合における標準領域を使用して)定義し、シグナル強度を、リビングイメージ3.0ソフトウェア(Xenogen)を使用して算出し、1秒当たりの光子として表した。バックグラウンドの光子束を、ベクターが投与されていない対照マウス又は対照筋肉上に描かれたROIから定義した。
マウスシンキチンをインビボ適用のために探索した。シンシチンAは非オルソログであるが、ヒトシンシチン1及び2に対する機能的に類似したマウスの対応物である(Dupressoir et al 2005)。本発明者らは、マウスSynAを発現プラスミド中にクローニングし、それを使用してレンチウイルスベクター粒子を産生した。本発明者らは、シンシチンAが、rHIV由来LVを成功裏に偽型化できることを見出した。本発明者らは、安定したLV−SynAを生成するためにヒトシンシチン偽型化LV(プレート当たり22μgのDNA、1回の収集のみ)の産生のために既に定められたのと同じ条件を使用した。LV−Syn Aは、VF1の存在においてマウスA20 Bリンパ腫細胞株を形質導入することにおいて非常に効率的であった(図S8)(補足表S4)。本発明者らは、SynAが、ヒト初代非活性化マウスB細胞をインビトロで形質導入するために、そのヒト対応物と同様に効率的であることを見出した。図8及び図9に見られるように、LV−SynAは、脾臓からの(図8)及び骨髄(図9)からの前処理マウスB細胞なしで形質導入することが可能であった。これらの実験はまた、マウスT細胞もまた形質導入することができることを示す。B220低細胞の形質導入によって、未成熟B細胞が形質導入され、生物学的適用のための標的となりうることが示唆される。
興味深いことに、マウスシンシチンAで偽型化されたLVを用いてヒトCD19+B細胞を形質導入することが可能であった(補足表S5)
Claims (15)
- 内在性レトロウイルスのシンシチン(ERVシンシチン)を用いて偽型化され、目的の異種遺伝子をパッケージングし、高い物理的及び/又は感染力価を提示する、安定なレンチウイルス粒子。
- 請求項1記載の安定な偽型化レンチウイルス粒子を得るための方法であって、以下の工程を含む:
a)適当な細胞株において少なくとも1つのプラスミドをトランスフェクトすることであって、前記の少なくとも1つのプラスミドは目的の異種遺伝子、レトロウイルスのrev、gag、及びpol遺伝子、ならびにERVシンシチンをコードする核酸を含む;
b)a)において得られたトランスフェクト細胞をインキュベートし、ERVシンシチンを用いてそれぞれ偽型化された安定なレンチウイルス粒子を産生すること、及び目的の異種遺伝子をパッケージングすること;及び
c)b)において得られた安定なレンチウイルス粒子を収集し、濃縮すること、好ましくは、工程c)の濃縮が、b)において得られた、収集された安定なレンチウイルス粒子を遠心分離及び/又は精製することを含む。 - レトロウイルスのrev、gag、及びpol遺伝子が、レンチウイルスのrev、gag、及びpol遺伝子、 好ましくはHIV−1のrev、gag、及びpol遺伝子である、請求項2記載の方法。
- 請求項2又は3のいずれか一項記載の方法により得られた粒子。
- ERVシンシチンを、HERV−W、HERV−FRD、マウスシンシチンA、マウスシンシチンB、シンシチンOry1、シンシチンCar1、及びシンシチンRum1からなる群より選択し、好ましくは、ERVシンシチンを、HERV−W、HERV−FRD、及びマウスシンシチンAからなる群より選択し、さらにより好ましくは、ERVシンシチンは、HERV−W又はHERV−FRDである、請求項1又は4記載の粒子、あるいは請求項2〜4のいずれか一項記載の方法。
- 高い感染力価が、2E+04TU/mlより高い、好ましくは1E+05TU/mlより高い、好ましくは1E+06TU/mlより高い、好ましくは2E+06TU/mlより高い、工程c)の終了時に産生された感染性粒子の力価であり、及び/又は高い物理力価が、0.7×105ng p24/mLより高い、特に1×105ng p24/mLより高い、好ましくは1.1×105ng p24/mLより高い、より好ましくは1.5×105ng p24/mLより高い、工程c)の終了時に産生された粒子の力価である、請求項1又は5記載の粒子、あるいは請求項2〜4のいずれか一項記載の方法。
- 医薬としての使用のための、好ましくは遺伝子療法もしくは免疫療法における、又はワクチンとしての、又は免疫予防における使用のための、請求項1、5、又は6のいずれか一項記載の粒子。
- 免疫細胞、好ましくはB細胞又は骨髄細胞の形質導入のための、請求項1、5、又は6のいずれか一項記載の粒子のインビトロでの使用。
- バイオテクノロジー工学のための、好ましくは免疫グロブリンを産生するための、請求項1、5、6、又は8のいずれか一項記載の粒子のインビトロでの使用。
- 免疫細胞を形質導入することによる治療のための使用のための、好ましくは免疫不全、自己免疫、感染性疾患、又はB細胞関連癌を処置するための使用のための、請求項1又は5〜7のいずれか一項記載の粒子。
- 医薬としての使用のための、又は診断目的のための、請求項1、5、又は6のいずれか一項記載の粒子を用いて感染させた免疫細胞のサンプル。
- 請求項1、5、又は6のいずれか一項記載の粒子を用いて、免疫細胞、好ましくは、場合により以前に刺激された、ナイーブ免疫細胞を感染させる工程を含む、改変され目的の異種遺伝子を発現する免疫細胞を得るためのエクスビボの方法。
- LAH4ペプチド又はその機能的誘導体、好ましくはLAH4−A4ベクトフシン1の存在において実施する、及び/又は免疫細胞が、B細胞、T細胞、樹状細胞、単球、及びマクロファージより選ばれる、好ましくはB細胞である、請求項12記載のエクスビボでの方法。
- 請求項12又は13のいずれか一項記載のエクスビボの方法であって、以下の工程を含む:
−場合により、IL−7を含む培地中でナイーブ免疫細胞をインキュベートすることによりそれらを刺激すること、及び
−LAH4ペプチド又はその機能的誘導体の存在において、請求項1、5、又は6のいずれか一項記載の前記粒子を用いて刺激されたか又はされていない、ナイーブ免疫細胞を感染させること。 - 請求項12〜14のいずれか一項記載のエクスビボの方法により入手可能な、目的の前記異種遺伝子を含む免疫細胞、好ましくはB細胞、より好ましくはナイーブB細胞。
