なお本発明において、拡大培養とは所望の細胞を増殖させる培養(細胞数を増加させる培養)を意味する。
本発明は下記工程を包含することを特徴とする、CD62L及び/又はCCR7を発現する細胞集団の製造方法である。
工程(1):T細胞を含有する細胞集団からCD8陽性T細胞を除去、又はCD4陽性T細胞を採取することにより培養に供する細胞集団を調製する工程、及び
工程(2):工程(1)で得られた細胞集団を、下記(A)及び(B)の存在下で培養し、かつ当該培養中及び/又は当該培養後の拡大培養中、全部もしくは一部の期間において下記(C)の存在下で培養する工程。
(A)FN、FNフラグメント及びそれらの混合物からなる群より選択される少なくとも1つのポリペプチド
(B)CD3リガンド、又はCD3リガンド及びCD28リガンド
(C)インターロイキン-4
本発明の製造方法の工程(1)では、T細胞を含有する細胞集団からCD8陽性T細胞を除去、又はCD4陽性T細胞を採取することにより、培養に供する細胞集団を調製することが行われる。CD8陽性T細胞の除去は、細胞表面に発現しているCD8を指標として実施することができ、例えば抗CD8抗体が固定化されたビーズや培養容器などの担体とT細胞を含有する細胞集団とを混合し、前記担体に結合したCD8陽性T細胞を除去する、又は、前記担体と結合しない細胞集団を回収することにより実施される。抗CD8抗体が固定化されたビーズとしては、例えば、Dynabeads M-450 CD8(インビトロジェン社製)、Eligix anti-CD8 mAb coated nickel particles(バイオトランスプラント社製)、CliniMACS System(Miltenyi Biotec社製)などが好適に使用できる。以下、本明細書において、T細胞を含有する細胞集団からCD8陽性T細胞を除去した細胞集団を「CD8除去細胞集団」と称する場合がある。
本発明の製造方法の工程(2)では、工程(1)で得られた細胞集団を、前記(A)成分及び(B)成分の存在下で培養し、かつ当該培養中及び/又は当該培養後の拡大培養中、全部もしくは一部の期間において前記(C)成分の存在下で培養することにより実施される。
態様1:工程(1)で得られた細胞集団を、前記(A)成分及び(B)成分の存在下で培養し、かつ当該培養期間中の全部もしくは一部の期間において(C)成分を共存させる。当該培養により得られた細胞集団を、更に拡大培養し、かつ当該拡大培養中の全部もしくは一部の培養を(C)成分の存在下で実施する。
態様2:工程(1)で得られた細胞集団を、前記(A)成分及び(B)成分の存在下、かつ(C)成分の非存在下で培養する。当該培養により得られた細胞集団を、更に拡大培養し、かつ当該拡大培養中の全部又は一部の期間の培養を(C)成分存在下で実施する。
態様3:工程(1)で得られた細胞集団を、前記(A)成分及び(B)成分存在下で培養し、かつ当該培養中の全部又は一部の期間において(C)成分を共存させる。当該培養により得られた細胞集団を、更に前記(C)成分非存在下で拡大培養する。
本発明の製造方法は、その製造の過程において細胞集団の細胞増殖率が高いことを1つの特徴とする。本明細書において、細胞増殖率は拡大培養率とも記載する。細胞増殖率は、ある一定の培養期間において、培養開始時の細胞数が培養後に何倍に増殖したのかを示す数値である。ここで、細胞増殖率が高いとは、本発明の細胞集団の製造方法の培養条件(以下、本発明の培養条件と記載する)における細胞集団の細胞増殖率が、(A)成分及び(B)成分の存在下で、かつ(C)成分の非存在下での培養、又は(B)成分のみの存在下で、かつ(C)成分の非存在下での培養条件(以下、本発明の培養条件でない場合と記載する)における細胞集団の細胞増殖率と比較して高いことを意味する。好適には本発明の培養条件は、本発明の培養条件ではない場合と比較して、培養開始から7日目以降において、5%以上、より好ましくは10%以上高い細胞増殖率で細胞集団を得ることができる。
本発明は、前記方法により製造される細胞集団も提供する。当該細胞集団は、前述のとおりCD4陽性ナイーブT細胞及び/又はCD4陽性セントラルメモリーT細胞の含有率が高い細胞集団である。すなわち、当該細胞集団は、(A)成分及び(B)成分の、又は(B)成分の存在下で、かつ(C)成分の非存在下での培養により得られた細胞集団と比較して、下記(1)~(5)からなる群より少なくとも1つが高比率に含有されている細胞集団である。
(1):CD4陽性CD45RA陽性CCR7陽性T細胞
(2):CD4陽性CD45RA陽性CD62L陽性T細胞
(3):CD4陽性CD45RA陰性CCR7陽性T細胞

当該細胞集団としては、CD4陽性ナイーブT細胞及びCD4陽性セントラルメモリーT細胞をより高含有することが望ましいという観点から、好ましくは前記(1)-(5)のうち少なくとも2つ以上、より好ましくは3つ以上、さらに好ましくは4つ以上、さらにより好ましくはすべてが高比率に含有されている細胞集団であることが例示される。
以下の実験で使用する培養器材に抗ヒトCD3抗体(OKT3、ヤンセンファーマ社製)及び配列表の配列番号8で表されるアミノ酸配列を有するFNフラグメント〔商品名:レトロネクチン(登録商標)、タカラバイオ社製、以下、CH-296と記載〕を固定化した。すなわち12穴細胞培養プレート(ファルコン社製)に終濃度5μg/mLの抗ヒトCD3抗体と終濃度25μg/mLのCH-296を含むACD-A液(テルモ社製)を0.4mL/ウェルずつ添加した後、37℃で5時間インキュベートした。その後、各ウェルからCH-296を含むACD-A液を吸引除去後、各ウェルを0.5mL GT-T503培地(タカラバイオ社製)で2回洗浄し各実験に供した(以下、抗ヒトCD3抗体/CH-296固定化プレートと記載)。
インフォームド・コンセントの得られた健常人ドナーから常法に従い調製したヒト末梢血単核細胞(PBMC)を、2mM EDTA、0.1% BSAを含むPBS(以下、Buffer1と称す)を用いて1×107cells/mLになるように懸濁した。その後、Buffer1で洗浄したCD8ポジティブセレクションビーズ(Dynabeads M-450 CD8:インビトロジェン社製)をPBMC 1×107cellsあたり2×107ビーズとなるように添加し、ローテーターを用いて4℃で30分間緩やかに攪拌した後、磁気分離装置MPC-15(Dynal社製)上に2~3分静置してビーズ非結合細胞を回収した(以下、CD8除去細胞集団と記載)。回収したCD8除去細胞集団を500×gで5分間遠心したのち、1%ヒト自己血清、100IU/mL IL-2(Proleukin、ニプロ社製)、0.2%ヒト血清アルブミン(バクスター社製)、2.5μg/mL fungizone(ブリストルマイヤーズ社製)、及び50U/mL ペニシリン/ストレプトマイシン(インビトロジェン社製)を含むGT-T503培地(タカラバイオ社製、以下、GT-T503CMと称す)を用いて4×105cells/mLになるように懸濁した。
実施例1-(2)で調製したCD8除去細胞集団の培養を行った。なお、培養は10ng/mLのIL-4を含むGT-T503CMで全期間培養する区(D0-12区)及び培養開始4日目以降に上記濃度のIL-4を含むGT-T503CMで培養する区(D4-12区)を設定した。ネガティブコントロールとして、IL-4を添加しない区を設定した(無添加区)。
実施例1-(3)で得られた培養12日目の細胞集団を、0.1%牛血清アルブミン(BSA、シグマ社製)を含むPBS(以下、0.1%BSA/PBSと記載)で洗浄した。0.1%BSA/PBSに細胞集団を懸濁し、抗体反応液1として、FITC標識マウス抗ヒトCD8抗体(ベクトン・ディッキンソン社製)、RD1標識マウス抗ヒトCD45RA抗体(ベックマンコールター社製)、PerCP標識マウス抗ヒトCD3抗体(ベクトン・ディッキンソン社製)、PE‐Cy7標識マウス抗ヒトCD62L抗体(eBioscience社製)、及びAPC‐Cy7標識マウス抗ヒトCD4抗体(ベクトン・ディッキンソン社製)を含む抗体液を、更に抗体反応液2として、FITC標識マウス抗ヒトCD8抗体、PE標識マウス抗ヒトCXCR4抗体(ベクトン・ディッキンソン社製)、PerCP標識マウス抗ヒトCD3抗体、及びAPC‐Cy7標識マウス抗ヒトCD4抗体を含む抗体液をそれぞれ添加し、抗体反応を行った。その後、0.1%BSA/PBSで細胞集団を2回洗浄し、再度0.1%BSA/PBSに懸濁した。この細胞集団をフローサイトメトリー(FACS CantoII:ベクトン・ディッキンソン社製)に供し、各々の細胞集団のCD3陽性CD4陽性細胞中の各種細胞表面マーカーの含有率を算出した。なお、全細胞中のCD3陽性率はすべての試験区で94%以上であった。結果を表2に示す。
(1)抗ヒトCD3抗体及びCH-296の固定化
実施例1-(1)と同様の方法で、抗ヒトCD3抗体のみを12穴細胞培養プレート(ファルコン社製)への固定化、及び洗浄を行い、各実験に供した(以下、抗ヒトCD3抗体固定化プレートと記載)。更に、抗ヒトCD3抗体/CH-296固定化プレートも調製し、各実験に供した。
インフォームド・コンセントの得られた健常人ドナーTK19及びTK29から常法に従い調製したPBMCを、10μg/mL DNaseを含むGT-T503CMに懸濁した。その後、500×gで5分間遠心し、上清を除去した。得られた細胞集団をBuffer1で1×107cells/80μLになるように懸濁後、human CD4 Microbeads(Miltenyi Biotec社製)を1×107cellsに対して20μL添加し、4℃で15分間放置した。その後、1×107cellsに対して1.8mLのBuffer1を添加し、300×g、10分間遠心後、上清を除去し、500μLのBuffer1に懸濁した。VarioMACS Separator(Miltenyi Biotec社製)にMACS Separation LS Columns(Miltenyi Biotec社製)をセットし、3mL Buffer1を通した後、上記細胞懸濁液を添加した。その後、5mL Bufferで3回洗浄した後、カラムをセパレーターから取り外し、5mLのBuffer1を添加してシリンジにより細胞液を押し出し、CD4陽性T細胞集団を取得した。
実施例2-(2)で調製したCD4陽性T細胞集団の培養を行った。培養は、培養全期間10ng/mL IL-4を含むGT-T503CMを用い、抗ヒトCD3抗体単独刺激を行う区(抗CD3刺激区)及び抗ヒトCD3抗体とCH-296刺激を行う区(抗CD3/CH-296刺激区)を設定した。ネガティブコントロールとしてIL-4を添加しない区を設定した(無添加区)。
実施例2-(3)で得られた培養12日目の細胞集団を0.1%BSA/PBSで洗浄した。0.1%BSA/PBSに細胞を懸濁し、FITC標識マウス抗ヒトCD8抗体、RD1標識マウス抗ヒトCD45RA抗体、PerCP標識マウス抗ヒトCD3抗体、PE‐Cy7標識マウス抗ヒトCD62L抗体、及びAPC‐Cy7標識マウス抗ヒトCD4抗体を含む抗体液を添加し、抗体反応を行った。その後、0.1%BSA/PBSで細胞を2回洗浄し、再度0.1%BSA/PBSに懸濁した。この細胞をフローサイトメトリーに供し、各々の細胞集団のCD3陽性CD4陽性細胞中の各種細胞表面マーカーの含有率を算出した。結果を表4及び表5に示す。なお、表中、+はIL-4を含むGT-T503CMを用いた試験区を示し、-は無添加区を示す。
(1)ΔLNGFR及びMazF遺伝子搭載レトロウイルスベクターの調製
ΔLNGFR及びMazF遺伝子搭載レトロウイルスベクターの調製は国際公開第2008/133137号パンフレットの実施例1及び2と同様の方法で行った。すなわち、HIV-LTR-MazFカセットがレトロウイルスベクターゲノムの転写とは逆方向に挿入され、かつヒトΔLNGFRがヒトPGKプロモーターの下流に順方向に挿入された組換えレトロウイルスベクタープラスミドpMT-MFR-PL2を取得し、当該プラスミドを用いてエコトロピックMT-MFR-PL2ウイルスを作製した後、これをGaLVレトロウイルスパッケージング細胞PG13に感染させ、高力価のウイルス産生細胞をクローニングしてレトロウイルスベクター産生細胞株PG13/MT-MFR-PL2を樹立した。更に当該産生細胞を用いて、常法によりGaLV/MT-MFR-PL2ウイルス液を取得した。
実施例1-(1)と同様の方法で、抗ヒトCD3抗体/CH-296固定化プレートを調製し、各実験に供した。
実施例2-(2)と同様の方法で、インフォームド・コンセントの得られた健常人ドナーからCD4陽性T細胞集団を取得した。
実施例3-(3)で調製した細胞集団を遠心して上清を除去後、IL-4を含むGT-T503CM又はIL-4を含まないGT-T503CMでそれぞれ懸濁し、2×105cells/mLに調製した。実施例3-(2)で調製した抗ヒトCD3抗体/CH-296固定化プレートに、上記調製した細胞を2.6mL/ウェルで添加した。これらのプレートを37℃、5%CO2インキュベータ内で培養した(培養0日目)。
実施例3-(4)で得られた培養開始12日目の細胞集団を0.1%BSA/PBSで洗浄した。0.1%BSA/PBSに細胞を懸濁し、抗体反応液1として、FITC標識マウス抗ヒトCCR7(R&D systems社)、RD1標識マウス抗ヒトCD45RA抗体、PerCP標識マウス抗ヒトCD3抗体、PE‐Cy7標識マウス抗ヒトCD62L抗体、APC標識マウス抗ヒトLNGFR抗体(Miltenyi Biotec社製)、及びAPC‐Cy7標識マウス抗ヒトCD4抗体を含む抗体液1、更に抗体反応液2として、PE標識マウス抗ヒトCXCR4抗体(PerCP標識マウス抗ヒトCD3抗体、及びAPC‐Cy7標識マウス抗ヒトCD4抗体を含む抗体液を添加し、抗体反応を行った。その後、0.1%BSA/PBSで細胞を2回洗浄し、再度0.1%BSA/PBSに懸濁した。この細胞をフローサイトメトリーに供し、各々の細胞集団のCD3陽性CD4陽性細胞中の各種細胞表面マーカーの含有率を算出した。結果を表7~10に示す。表中、+はIL-4を含むGT-T503CMを用いた場合を示し、-は無添加の場合を示す。

Claims (10)
- 下記工程を包含することを特徴とする、CD62L及び/又はCCR7を発現する細胞集団の製造方法。
工程(1):T細胞を含有する細胞集団からCD8陽性T細胞を除去、又はCD4陽性T細胞を採取する工程、及び
工程(2):工程(1)で得られた細胞集団を、下記(A)及び(B)の存在下で培養し、かつ当該培養中及び/又は当該培養後の拡大培養中、全部又は一部の期間において、下記(C)の存在下で培養する工程。
(A)フィブロネクチン、フィブロネクチンフラグメント及びそれらの混合物からなる群より選択される少なくとも1つのポリペプチド
(B)CD3リガンド、又はCD3リガンド及びCD28リガンド
(C)インターロイキン-4 - 工程(2)が、請求項1記載の(A)及び(B)の存在下で、かつ(C)の存在下で培養し、得られた細胞集団を(C)の存在下で拡大培養する工程である、請求項1記載の製造方法。
- 工程(2)が、請求項1記載の(A)及び(B)の存在下で、かつ(C)の非存在下で培養し、得られた細胞集団を(C)の存在下で拡大培養する工程である、請求項1記載の製造方法。
- 工程(2)が、請求項1記載の(A)及び(B)の存在下で、かつ(C)の存在下で培養し、得られた細胞集団を(C)の非存在下で拡大培養する工程である、請求項1記載の製造方法。
- 更に外来遺伝子を導入する工程を包含する、請求項1~4のいずれか1項に記載の製造方法。
- 請求項1記載の(A)及び(B)の、又は(B)の存在下で、かつ(C)の非存在下での培養により得られた細胞集団と比較して、下記(1)~(5)からなる群より選択される少なくとも1つの細胞が高比率に含有されている、請求項1~5のいずれか1項に記載の製造方法で得られる細胞集団。
(1):CD4陽性CD45RA陽性CCR7陽性T細胞
(2):CD4陽性CD45RA陽性CD62L陽性T細胞
(3):CD4陽性CD45RA陰性CCR7陽性T細胞
(4):CD4陽性CD45RA陰性CD62L陽性T細胞
(5):CD4陽性CXCR4陽性細胞 - 医薬に使用するための請求項6記載の細胞集団。
- 医薬の製造に用いる、請求項6記載の細胞集団。
- 請求項6記載の細胞集団を有効成分として含有する医薬。
- 対象に有効量の請求項6記載の細胞集団を投与する工程を含む、疾患の治療方法又は予防方法。
