本发明公开了一种增加骨密度的中药保健食品,包括以下组份及各自质量百分含量:珍珠粉1‑3%、碳酸钙10‑15%、臭梧桐叶2‑6%、D‑氨基葡萄糖盐酸盐15‑20%、板栗壳5‑10%、猫爪草1‑3%、蛤蜊5‑12%和辅料45‑50%。本发明将臭梧桐叶、猫爪草、板栗壳和蛤蜊与现有用于增加骨密度药物中的珍珠粉、碳酸钙、D‑氨基葡萄糖盐酸盐共同配合,得到的中药保健食品可提供丰富的钙元素、氨基酸等可促进骨胶原合成和骨基质形成的营养成分,可有效增加骨密度。

技术领域
本发明属于中药保健食品技术领域,具体涉及一种增加骨密度的中药保健食品及其制备方法。
背景技术
骨质疏松症(osteopsis简称OPS)是一种以低骨量和骨组织的纤维结构退化为特征的常见老年病,主要表现为骨折及侵袭脊椎而引起腰背疼痛与身长短缩等并发症,是一种严重的致残性疾病。随着进入人口老龄化的社会,老龄人口不断增加,骨质疏松症已严重威胁着中老年人的健康,骨质疏松症有“无声杀手”之称,WHO统计表明:目前世界范围内骨质疏松症患者总数超过2亿人口,每年花在治疗和住院上的费用高达250亿美元。中国健康促进基金会发布的《骨质疏松症中国白皮书2008》显示,调查估算2006年全国50岁以上的人群中约有6944万人患有骨质疏松症,有资料预计,从现在算起未来的50年中,有75%的髋骨骨折会出现在发展中国家,这与骨质疏松密切相关。骨质疏松症好发于绝经后妇女和老年男性,现在还有年轻化趋势。
骨密度的测量是评价骨质丢失,诊断骨质疏松的重要手段之一。骨密度指骨单位面积所含的骨矿物质在骨骼沉积的量,骨密度越大,骨骼就越坚实。20岁以后,骨的长度就停止生长了,但还有一个骨量加强的阶段,即骨密度增加,直到30-40岁,骨的致密与坚硬程度才达到最高值,也即达到了峰值骨密度。骨骼达到峰值密度以后,随着年龄增加,成骨细胞功能逐渐下降,破骨细胞对骨过度吸收,造成骨矿物质不断丢失,骨量逐渐减少。
由于骨质疏松引起骨量的减少是不可逆的,因此预防远比治疗更为重要。目前,对于骨质疏松的防治,主要体现在两个方面:一是在青少年时期增加摄取相关的营养物质,使骨成熟期能达到最佳骨峰值;二是在更年期前后,采用适当的措施以尽量减少骨质的丢失。目前已有增加骨密度的保健食品,如CN200610159776.3公开了一种治疗骨质疏松症的中药组合物及其制备方法,组合物由熟地黄、骨碎补、补骨脂、淫羊藿、黄芪、丹参、茯苓、黄瓜子、自然铜、血竭、乳香和没药组成;CN200910260971.9公开了一种用于缓解骨关节病、增强免疫力的制剂,制剂配方由珍珠粉、淫羊藿提取物、D-氨基葡萄糖盐酸盐、硫酸软骨素组成。这些成分通过补钙方式或通过调整内分泌而促进钙吸收的方式达到增加骨密度的目的,但仍存在一定的缺陷。如起效慢,疗效不显著,骨碎补或者D-氨基葡萄糖盐酸盐的用量较大,成本高,并且大量补钙易引起高血钙水平,可引起心血管疾病危险的增加。
发明内容
本发明的目的是克服现有技术的不足,提供一种增加骨密度的中药保健食品及其制备方法。
本发明提供了一种增加骨密度的中药保健食品,包括以下组份及各自质量百分含量:珍珠粉1-3%、碳酸钙10-15%、臭梧桐叶2-6%、D-氨基葡萄糖盐酸盐15-20%、板栗壳5-10%、猫爪草1-3%、蛤蜊5-12%和辅料45-50%。
优选的,所述增加骨密度的中药保健食品包括以下组份及各自质量百分含量:珍珠粉1%、碳酸钙14%、臭梧桐叶3%、D-氨基葡萄糖盐酸盐18%、板栗壳粉7%、猫爪草2%、蛤蜊9%和辅料46%。
优选的,所述辅料包含环糊精、淀粉和甘露醇。
本发明还提供了一种增加骨密度的中药保健食品的制备方法,包括以下步骤:
1)取臭梧桐叶、板栗壳和猫爪草,干燥后粉碎,过筛,得到的粉末加水提取,过滤得到水提液和滤渣;
2)往滤渣中加入乙醇溶液进行回流提取,过滤得到醇提液;
3)将水提液和醇提液混合,干燥,粉碎,过筛,得到药物粉末;
4)将蛤蜊粉碎,然后与步骤3)得到的药物粉末混匀,再加入珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐和辅料搅拌均匀,制成片剂,即得增加骨密度的中药保健食品。
优选的,步骤1)中所述过筛为过100-150目筛。
优选的,步骤1)中所述粉末的质量与水的质量比1:6-10。
优选的,步骤2)所述乙醇溶液的体积浓度为30-80%。
优选的,步骤2)所述所述滤渣与乙醇溶液的质量比为1:5-8。
目前用于增加骨密度的保健食品有很多,维生素类如维生素D,蛋白质或氨基酸类如酪蛋白磷酸肽,骨碎补,补骨脂,D-氨基葡萄糖盐酸盐,中药活性成分如葛根素,及其他功能成分等相互组合。这些成分通过补钙方式或通过调整内分泌而促进钙吸收的方式达到增加骨密度的目的,但仍存在一定的缺陷,现有的增加骨密度的保健品存在搭配不合理,起效慢,疗效不显著,对骨密度的增加不明显,需要长期服用,并且骨碎补或者D-氨基葡萄糖盐酸盐的用量较大,成本高,并且大量补钙易引起高血钙水平,可引起心血管疾病危险的增加。
本申请的发明人通过研究发现将臭梧桐叶、猫爪草、板栗壳和蛤蜊与现有用于增加骨密度保健品中的珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐共同配合,得到的中药保健食品可提供丰富的钙元素、氨基酸等可促进骨胶原合成和骨基质形成的营养成分,可有效增加骨密度。
臭梧桐叶:臭梧桐叶含海州常山黄酮甙(clerodendrin),内消旋肌醇,生物碱,刺槐素-7-双葡萄糖醛酸甙,植物血凝素,臭梧桐素A和B,海州常山苦素A和B,洋丁得酚甙。可祛风除湿、平肝降压、解毒杀虫。功能主治风湿痹痛、半身不遂、高血压病、偏头痛、疟疾、痢疾、痈疽疮毒、湿疹疥癣。
猫爪草:猫爪草解毒,化痰散结,主治瘰疬,结核,咽炎,疔疮,疟疾,偏头痛,牙痛。
板栗壳:为板栗的外果皮,药性甘、涩、平,具有降逆、止血的功效,主治反胃、鼻衄、便血等症。
本发明通过大量的试验证明,蛤蜊可增加骨密度和骨钙含量,当其与珍珠粉、碳酸钙和D-氨基葡萄糖盐酸盐合用,效果更佳;此外,本发明人意外发现,当臭梧桐叶、猫爪草、板栗壳与上述4种物质合用,可促进骨髓造血功能,分泌骨胶原性物质,刺激骨生长因子,促进骨细胞生成和增殖,提高骨细胞活性和数量,抑制和减少骨吸收,促进骨形成和骨矿化,增加骨密度和骨强度,加速骨折愈合和增强抗折能力。达到活血理气,生精益髓,补髓生骨,祛风除湿,消肿止痛,强壮筋骨,恢复关节功能的临床治疗效果,补钙效果好,营养成分丰富,易于消化和吸收,适宜长期服用,可促进骨密度持续增长,且安全有效,服用方便。
本申请中珍珠粉和D-氨基葡萄糖盐酸盐的用量较少,其质量占除去辅料用量后各原料总质量的50%以下,而现有技术中,如专利申请号CN200610159776.3公开了一种用于治疗骨质疏松的制剂,其采用珍珠粉、淫羊藿提取物、D-氨基葡萄糖盐酸盐、硫酸软骨素为原料,其中珍珠粉和D-氨基葡萄糖盐酸盐的用量达到了总制剂用量的70%以上。本申请的珍珠粉、碳酸钙和D-氨基葡萄糖盐酸盐的用量少,生产成本低,并且与蛤蜊等原料共同配合,补钙效果好,可促进骨密度持续增长,起效快。
本申请的发明人通过研究发现,将增加骨密度的中药保健食品中各组份的质量百分含量控制如下:珍珠粉1%、碳酸钙14%、臭梧桐叶3%、D-氨基葡萄糖盐酸盐18%、板栗壳粉7%、猫爪草2%、蛤蜊9%、环糊精20%、淀粉14%和甘露醇12%,能够显著促进成骨细胞的增殖、可促进骨髓造血功能,分泌骨胶原性物质,刺激骨生长因子,促进骨细胞生成和增殖,提高骨细胞活性和数量。
本申请的臭梧桐叶、板栗壳和猫爪草先加水进行回流提取,再加乙醇进行回流提取,合并水提液和醇提液,干燥得到药物粉末与蛤蜊、珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐和辅料搅拌均匀即得增加骨密度的中药保健食品。现有技术中对中药物质的提取大都采用单一水提或者单一醇提的方法,而本发明先采用水提,再采用醇提,并且将乙醇的体积浓度控制在30-80%,能够将臭梧桐叶、板栗壳和猫爪草中的有效成分充分提取出来。
本发明的有益效果是:
1、本申请将臭梧桐叶、猫爪草、板栗壳和蛤蜊与现有用于增加骨密度保健品中的珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐共同配合,得到的中药保健食品可提供丰富的钙元素、氨基酸等可促进骨胶原合成和骨基质形成的营养成分,可有效增加骨密度。
2、本申请中珍珠粉、碳酸钙和D-氨基葡萄糖盐酸盐的用量较少,不到除去辅料用量后各原料总质量的一半,生产成本低,并且与蛤蜊等原料共同配合,补钙效果好,可促进骨密度持续增长,起效快。
3、本申请通过研究发现,将增加骨密度的中药保健食品中各组份的质量百分含量控制如下:珍珠粉1%、碳酸钙14%、臭梧桐叶3%、D-氨基葡萄糖盐酸盐18%、板栗壳粉7%、猫爪草2%、蛤蜊9%、环糊精20%、淀粉14%和甘露醇12%,能够显著促进成骨细胞的增殖、可促进骨髓造血功能,分泌骨胶原性物质,刺激骨生长因子,促进骨细胞生成和增殖,提高骨细胞活性和数量。
4、本申请的制备方法简单,先采用水提,再采用醇提,并且将乙醇的体积浓度控制在30-80%,能够将臭梧桐叶、板栗壳和猫爪草中的有效成分充分提取出来。
5、采用本申请的原料、配比以及制备方法得到的增加骨密度的中药保健食品从根本上治疗骨质疏松导致的疾病,经临床验证,本申请的中药保健食品增加骨密度疗效确切,安全性好。

实施例1
增加骨密度的中药保健食品,包括以下组份及各自质量百分含量:珍珠粉3%、碳酸钙15%、臭梧桐叶2%、D-氨基葡萄糖盐酸盐15%、板栗壳5%、猫爪草3%、蛤蜊12%、环糊精15%、淀粉20%和甘露醇10%。
增加骨密度的中药保健食品的制备方法如下:1)取臭梧桐叶、板栗壳和猫爪草,干燥后粉碎,过120目筛,得到的粉末加8倍重量的水提取,提取温度为80℃,提取3次,每次提取45min,过滤并保留滤渣,合并滤液,得水提液;
2)往滤渣中加入体积浓度为50%的乙醇溶液进行回流提取,滤渣与乙醇溶液的质量比为1:5,提取温度为80℃,提取2次,每次提取60min,过滤,合并滤液,得到醇提液;
3)将水提液和醇提液混合,干燥,粉碎,过100目筛,得到药物粉末;
4)将蛤蜊粉碎,过80目筛,然后与步骤3)得到的药物粉末混匀,再加入珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐、环糊精、淀粉和甘露醇搅拌均匀,采用常规方法制成片剂,即得增加骨密度的中药保健食品。
实施例2
增加骨密度的中药保健食品,包括以下组份及各自质量百分含量:珍珠粉1%、碳酸钙14%、臭梧桐叶3%、D-氨基葡萄糖盐酸盐18%、板栗壳粉7%、猫爪草2%、蛤蜊9%、环糊精15%、淀粉15%和甘露醇16%。
增加骨密度的中药保健食品的制备方法如下:1)取臭梧桐叶、板栗壳和猫爪草,干燥后粉碎,过150目筛,得到的粉末加10倍重量的水提取,提取温度为90℃,提取3次,每次提取45min,过滤并保留滤渣,合并滤液,得水提液;
2)往滤渣中加入体积浓度为80%的乙醇溶液进行回流提取,滤渣与乙醇溶液的质量比为1:8,提取温度为80℃,提取2次,每次提取60min,过滤,合并滤液,得到醇提液;
3)将水提液和醇提液混合,干燥,粉碎,过90目筛,得到药物粉末;
4)将蛤蜊粉碎,过80目筛,然后与步骤3)得到的药物粉末混匀,再加入珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐、环糊精、淀粉和甘露醇搅拌均匀,采用常规方法制成片剂,即得增加骨密度的中药保健食品。
实施例3

增加骨密度的中药保健食品的制备方法如下:1)取臭梧桐叶、板栗壳和猫爪草,干燥后粉碎,过100目筛,得到的粉末加6倍重量的水提取,提取温度为70℃,提取3次,每次提取45min,过滤并保留滤渣,合并滤液,得水提液;
2)往滤渣中加入体积浓度为30%的乙醇溶液进行回流提取,滤渣与乙醇溶液的质量比为1:6,提取温度为80℃,提取2次,每次提取60min,过滤,合并滤液,得到醇提液;

增加骨密度的中药保健食品,包括以下组份及各自质量百分含量:珍珠粉3%、碳酸钙20%、蛤蜊9%、D-氨基葡萄糖盐酸盐23%、环糊精15%、淀粉15%和甘露醇15%。
增加骨密度的中药保健食品的制备方法如下:将珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐、环糊精、淀粉和甘露醇搅拌均匀,采用常规方法制成片剂,即得增加骨密度的中药保健食品。
对比例2
增加骨密度的中药保健食品,包括以下组份及各自质量百分含量:珍珠粉1%、碳酸钙14%、臭梧桐叶3%、D-氨基葡萄糖盐酸盐18%、板栗壳粉7%、猫爪草2%、蛤蜊9%、环糊精15%、淀粉15%和甘露醇16%。
增加骨密度的中药保健食品的制备方法如下:1)取臭梧桐叶、板栗壳和猫爪草,干燥后粉碎,过150目筛,得到的粉末加入体积浓度为80%的乙醇溶液进行回流提取,粉末与乙醇溶液的质量比为1:8,提取温度为80℃,提取2次,每次提取60min,过滤,合并滤液,得到醇提液;
2)将醇提液干燥,粉碎,过90目筛,得到药物粉末;
3)将蛤蜊粉碎,过80目筛,然后与步骤3)得到的药物粉末混匀,再加入珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐、环糊精、淀粉和甘露醇搅拌均匀,采用常规方法制成片剂,即得增加骨密度的中药保健食品。
对比例3
增加骨密度的中药保健食品,包括以下组份及各自质量百分含量:珍珠粉1%、碳酸钙14%、臭梧桐叶3%、D-氨基葡萄糖盐酸盐18%、板栗壳粉7%、猫爪草2%、蛤蜊9%、环糊精15%、淀粉15%和甘露醇16%。
增加骨密度的中药保健食品的制备方法如下:1)取臭梧桐叶、板栗壳和猫爪草,干燥后粉碎,过150目筛,得到的粉末加10倍重量的水提取,提取温度为90℃,提取3次,每次提取45min,过滤并保留滤渣,合并滤液,得水提液;
2)将水提液干燥,粉碎,过90目筛,得到药物粉末;
3)将蛤蜊粉碎,过80目筛,然后与步骤2)得到的药物粉末混匀,再加入珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐、环糊精、淀粉和甘露醇搅拌均匀,采用常规方法制成片剂,即得增加骨密度的中药保健食品。
上述制备成片剂的制备方法如下:(1)用体积浓度为90%的乙醇溶液对加入辅料混合均匀的混合物进行搅拌软化,以手轻握成团,捏之即散为度,得到物料待用;(2)制粒:利用高效湿法混合制粒机(YR-160,江阴通达机械,将步骤(1)制得的物料,制成粒并过16目筛,得湿颗粒物待用;(3)干燥:将湿颗粒物放入热风循环干燥箱(CT-C-Ⅱ,无锡华星药化)内干燥,在温度为50~60℃下干燥,使得物料颗粒的含水量<5%,得干燥颗粒;(4)整粒:将步骤(3)制得的干燥颗粒,过14目筛,留下细颗粒,将粗料返回步骤(1),制成软材;(5)制总混颗粒:将步骤(4)中制得的细颗粒放入搅拌机(SYH-600,常州品正干燥设备)内,进行10分钟的充分搅拌,得总混颗粒;(6)压片:将经步骤(5)得到的总混颗粒放入高速压片机(GZPL,北京国药龙立)中压片,得到增加骨密度的中药保健食品。
通过以下试验证明本发明药物的药理药性:
药效试验:
选取90只SPF级SD雌性大鼠,体重250~320g。取80只进行卵巢切除手术,剩余10只进行假手术。大鼠卵巢切除后5天,进行阴道图片检查,确认大鼠卵巢完全切除。将进行卵巢切除的80只大鼠随机分为8组,每组10只,分别为实施例1组、实施例2组、实施例3组、对比例1-3组、模型组和对照组。进行假手术的10只大鼠设为假手术组。其中,假手术组和模型组均灌胃给蒸馏水,实施例1-3组和对比例1-3组分别灌胃给实施例1-3和对比例1-3制得的中药保健食品,灌胃剂量为2g/kg,每日灌胃2次。对照组灌胃给市售雪域骨宝胶囊(西藏阳光生物股份有限公司供应的),灌胃剂量为2g/kg,每日灌胃2次。各组大鼠连续灌胃90天。采用DPX-L双能X线骨密度仪测定股骨骨密度,并采用原子吸收分光光度法测定股骨骨钙含量,结果见下表1。

从上述数据可知,本发明实施例1-3得到的中药保健食品可提高大鼠的股骨骨密度和骨钙含量,其效果要优于市售增加骨密度胶囊(雪域骨宝胶囊)的效果。实施例2制得的中药保健食品增加大鼠的股骨骨密度和骨钙含量的效果最佳。对比例1制得的中药保健食品缺乏臭梧桐叶、板栗壳和猫爪草,其增加大鼠的股骨骨密度和骨钙含量的效果较实施例1-3减弱,说明缺少这几种物质,得到的中药保健食品增加骨密度的效果较差。对比例2采用的是水提法提取臭梧桐叶、板栗壳和猫爪草中的有效成分,其增加大鼠的股骨骨密度和骨钙含量的效果较实施例2减弱,与对照组的效果相当。对比例3采用的是醇提法提取臭梧桐叶、板栗壳和猫爪草中的有效成分,其增加大鼠的股骨骨密度和骨钙含量的效果较实施例2减弱,但仍优于对照组的效果。以上结果表明,将臭梧桐叶、猫爪草、板栗壳和蛤蜊与现有用于增加骨密度保健品中的珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐共同配合,可有效增加骨密度和骨钙含量;对于臭梧桐叶、板栗壳和猫爪草先采用水提,再采用醇提,相较于现有技术中采用单一水提或者单一醇提的方法,本发明能够将臭梧桐叶、板栗壳和猫爪草中的有效成分充分提取出来,得到的中药保健食品能显著增加骨密度和骨钙含量。
Claims (8)
1.一种增加骨密度的中药保健食品,其特征在于,包括以下组份及各自质量百分含量:珍珠粉1-3%、碳酸钙10-15%、臭梧桐叶2-6%、D-氨基葡萄糖盐酸盐15-20%、板栗壳5-10%、猫爪草1-3%、蛤蜊5-12%和辅料45-50%。
2.如权利要求1所述增加骨密度的中药保健食品,其特征在于,包括以下组份及各自质量百分含量:珍珠粉1%、碳酸钙14%、臭梧桐叶3%、D-氨基葡萄糖盐酸盐18%、板栗壳粉7%、猫爪草2%、蛤蜊9%和辅料46%。
3.如权利要求1所述增加骨密度的中药保健食品,其特征在于,所述辅料包含环糊精、淀粉和甘露醇。
4.一种如权利要求1所述增加骨密度的中药保健食品的制备方法,其特征在于,包括以下步骤:
1)取臭梧桐叶、板栗壳和猫爪草,干燥后粉碎,过筛,得到的粉末加水提取,过滤得到水提液和滤渣;
2)往滤渣中加入乙醇溶液进行回流提取,过滤得到醇提液;
3)将水提液和醇提液混合,干燥,粉碎,过筛,得到药物粉末;
4)将蛤蜊粉碎,然后与步骤3)得到的药物粉末混匀,再加入珍珠粉、碳酸钙、D-氨基葡萄糖盐酸盐和辅料搅拌均匀,制成片剂,即得增加骨密度的中药保健食品。
5.如权利要求4所述增加骨密度的中药保健食品的制备方法,其特征在于,步骤1)中所述过筛为过100-150目筛。
6.如权利要求4所述增加骨密度的中药保健食品的制备方法,其特征在于,步骤1)中所述粉末的质量与水的质量比1:6-10。
7.如权利要求4或5所述增加骨密度的中药保健食品的制备方法,其特征在于,步骤2)所述乙醇溶液的体积浓度为30-80%。
8.如权利要求7所述增加骨密度的中药保健食品的制备方法,其特征在于,步骤2)所述所述滤渣与乙醇溶液的质量比为1:5-8。
