Abstract

本发明公开了N‑琥珀酰壳聚糖固载乳酸链球菌素、紫苏油膜的制备方法。N‑琥珀壳聚糖通过EDC、NHS活化其羧基,然后与乳酸链球菌素、紫苏油进行共价结合,通过延流法制得薄膜。实验表明,该方法克服乳酸链球菌素中性条件下不稳定,紫苏油易分解的不足,制得的薄膜结构稳定、机械性能较好,无毒性,对金黄色葡萄球菌、埃希氏大肠杆菌、托拉斯假单胞肝菌、肠炎沙门氏菌、单增李斯特菌有不同程度的抑制作用。

N-琥珀酰壳聚糖固载乳酸链球菌素、紫苏油膜的制备方法
技术领域:
本发明属于食品抑菌保鲜领域,具体涉及N-琥珀酰壳聚糖固载乳酸链球菌素、紫苏油抑菌膜的制备方法。
背景技术:
N-琥珀酰壳聚糖(NSC)是壳聚糖在琥珀酸酐体系下形成的,是一种具备一定晶体结构的高分子材料。与壳聚糖(CS)相比,水溶性较好,因为N-琥珀酰壳聚糖是两亲化合物,当引入取代基后,壳聚糖原本规整的空间排列遭到破坏,形成非晶化结构。此外,N-琥珀酰壳聚糖的生物相容性较好、具有较好的吸湿保湿作用、毒性较低,即使在人体内长期滞留都不会影响身体健康。
乳酸链球菌素(Nisin)是乳酸链球菌产生的一种多肽物质,由34个氨基酸残基组成,由于乳酸链球菌素可抑制大多数革兰氏阳性细菌,被作为食品防腐剂广泛应用于食品行业。食用后在人体的生理pH条件和α—胰凝乳蛋白酶作用下很快水解成氨基酸,不会改变人体肠道内正常菌群以及产生如其它抗菌素所出现的抗性问题,更不会与其它抗菌素出现交叉抗性,是一种高效、无毒、安全、无副作用的天然食品防腐剂。但是Nisin在pH值为2时耐热性好,pH值大于5,耐热性下降,很不稳定。
紫苏油是紫苏植株中提取的含亚麻酸等多组分的混合物,是一种优质保健食用油,具有紫苏和琥珀香气。还含有18种氨基酸,具有很高的营养和药用价值。紫苏油具有显著的抑菌活性和广泛的抗菌谱范围,且与化学抑菌剂相比副作用残留毒性小,紫苏油的疏水性成分能够直接作用于微生物的细胞膜,导致细胞膜的流动性增加,随之细胞膜的结构破坏,细胞内重要的內溶物留出,导致细胞死亡。紫苏精油和其他抑菌剂结合起协同作用,在实际应用中,协同作用使得成本降低,保证安全,还赋予食品特殊的香味,但是紫苏油在使用的过程易分解给其使用带来了限制。
基于N-琥珀酰壳聚糖具有-NH2和-COOH的活性基团,易于制备成多孔球状微粒、纳米粒、凝胶、膜状等多种状态,所以将乳酸链球菌素、紫苏油固载在上面制成抑菌膜。
发明内容:
本发明克服乳酸链球菌素中性条件下不稳定,紫苏油易分解的不足,将N-琥珀酰壳聚糖与Nisin、紫苏油共价结合制备出抑菌性能较强、绿色无污染的薄膜。
为实现上述目的,本发明采用下述技术方案:
N-琥珀酰壳聚糖固载乳酸链球菌素、紫苏油抑菌膜的制备包括以下步骤:N-琥珀酰壳聚糖(NSC)的制备参照CN 10651927 A。
称取1-2g NSC溶解在pH 4-9的磷酸柠檬酸盐缓冲溶液,加入EDC、NHS常温下搅拌3h,加入20-100mg Nisin搅拌1h制得所需溶液NNSC,在NNSC溶液中加入0.5%-2%紫苏油搅拌均匀,将上述制的溶液采用延流法制膜,加入2-10%的甘油,在30-70℃的条件下干燥5-12h。
本发明的优点:本发明克服乳酸链球菌素中性条件下不稳定,紫苏油易分解的不足,将N-琥珀酰壳聚糖与Nisin、紫苏油共价结合制备出的抑菌膜机械性能好,并对金黄色葡萄球菌、埃希氏大肠杆菌、托拉斯假单胞肝菌、肠炎沙门氏菌、单增李斯特菌有较好的抑制作用。而且过程操作简单,所制备的薄膜绿色无毒害。
下面结合实例对本发明进行描述,应该说明的是,下述说明仅是为了解释本发明,并不对其内容进行限定。

具体实施方式
实施例1:
N-琥珀酰壳聚糖(NSC)的制备参照CN 10651927 A;称取1g NSC溶解在30mL pH为5的磷酸柠檬酸盐缓冲溶液,加入EDC 20mg、NHS 20mg常温搅拌3h,加入Nisin20mg搅拌1h制得所需溶液NNSC;在NNSC溶液中加入0.5%紫苏油搅拌均匀;将上述制的溶液采用延流法制膜,加入2%的甘油,在30℃的条件下干燥8h,即得到抑菌膜。
实施例2:
N-琥珀酰壳聚糖(NSC)的制备参照CN 10651927 A;称取2g NSC溶解在30mL pH为6的磷酸柠檬酸盐缓冲溶液,加入EDC 20mg、NHS 20mg常温搅拌3h,加入Nisin 50mg搅拌1h制得所需溶液NNSC;在NNSC溶液中加入1.5%紫苏油搅拌均匀;将上述制的溶液采用延流法制膜,加入8%的甘油,在50℃的条件下干燥6h,即得到抑菌膜。
实施例3:
N-琥珀酰壳聚糖(NSC)的制备参照CN 10651927 A;称取2g NSC溶解在40mL pH为9的磷酸柠檬酸盐缓冲溶液,加入EDC 20mg、NHS 20mg常温搅拌3h;加入Nisin 80mg搅拌1h制得所需溶液NNSC;在NNSC溶液中加入1%紫苏油搅拌均匀;将上述制的溶液采用延流法制膜,加入5%的甘油,在70℃的条件下干燥5h,即得到抑菌膜。
实施例4:
按照牛津杯法对实施例1、实施例2、实施例3进行抑菌性能的测定,按照GB/T228-2010进行机械性能检测,结果见表1。

由表一可知,本发明提供的方法制备的薄膜不仅机械性能优良,抑菌性能也很好。

Claims (4)

1.N-琥珀酰壳聚糖固载乳酸链球菌素、紫苏油膜的制备方法,其特征在于:包括以下步骤:
(1)N-琥珀壳聚糖(NSC)的制备参照CN 10651927A;
(2)称取1-2g NSC溶解在pH 4-9的磷酸柠檬酸盐缓冲溶液,加入EDC、NHS常温搅拌3h,加入20-100mg乳酸链球菌素(Nisin)搅拌1h制得固载乳酸链球菌素的N-琥珀酰壳聚糖,NNSC;
(3)在NNSC溶液中加入0.5%-1.5%紫苏油搅拌均匀;
(4)将上述制的溶液采用延流法制膜,加入2-10%的甘油,在30-70℃的条件下干燥5-12h。
2.根据权利要求1所述的N-琥珀酰壳聚糖固载乳酸链球菌素、紫苏油膜的制备方法,其特征在于,步骤(2)中NSC固载Nisin,磷酸盐缓冲溶液的pH是6,Nisin的含量是50mg,NSC和缓冲溶液的比例是1:30(mg/mL),EDC、NHS的比例是1:1。
3.根据权利要求1所述的N-琥珀酰壳聚糖固载乳酸链球菌素、紫苏油膜的制备方法,其特征在于,步骤(3)加入紫苏油为1.5%。
4.根据权利要求1所述的N-琥珀酰壳聚糖固载乳酸链球菌素、紫苏油膜的制备方法,其特征在于,步骤(4)中制膜时加入8%的甘油,在50℃条件下干燥6h。
