本发明公开了一种仔猪饲用液体复合酶的制备方法,涉及动物营养和饲料技术领域,本发明液体复合酶以脂肪酶、酸性蛋白酶、淀粉酶为主纤维素酶、果胶酶、葡聚糖酶为辅可以有效的弥补仔猪内源酶的不足,增强仔猪的消化能力即可减少断奶后的仔猪因消化不良引起的腹泻,另外本发明液体复合酶中溶菌酶、甘氨酸、茶多酚相互配合有很强的抗菌、消炎、抗病毒的作用对预防病毒和细菌引起的仔猪腹泻起到了明显的效果。

技术领域
本发明涉及动物营养和饲料技术领域,特别是涉及一种仔猪饲用液体复合酶的制备方法。
背景技术
仔猪早期断奶是提高仔猪生产水平的主要措施之一,目前应用已相当普遍,但是,早期断奶仔猪存在消化系统发育不完善、消化酶分泌量不足、胃酸量少及断奶应激等特点,常常造成仔猪消化不良、生长抑制、饲料利用率低、抗病力差等问题。因此如何刺激仔猪消化系统发育提高仔猪对饲料的利用、增强抗病力等一直是人们研究探讨的问题。
近年来,酶制剂行业迅速发展及酶在饲料中的初步应用为解决此问题带来了新的曙光。断奶仔猪日粮中添加酶制剂的目的在于弥补仔猪消化系统发育不成熟导致的内源酶分泌不足,如脂肪酶、蛋白酶、淀粉酶等,内源酶含量低,消化能力就弱,在生产上易导致仔猪营养应激反应和早期断奶综合症,引起消化功能紊乱并出现腹泻。复合酶作为一种有益微生物发酵产生的具有特殊功能的蛋白质,在日粮中添加一定的比例,能有效的改善动物对饲料的消化吸收,从而提高动物的增重和饲料利用率,减少粪尿排出,具有一定的经济效益和环保效益。
酶是一种活性蛋白质,它与所有的蛋白质一样对环境很敏感,在高温时会变性失活。而现在的仔猪饲料基本都要进行制粒的工序,但是高温制粒,会涉及到蛋白质的变性问题。低温制粒的成本较高故采用喷涂酶制剂技术在饲料中加入酶来避免因蛋白质变性而导致的酶活性降低的问题。
液体酶喷涂技术作为一项新型的饲料加工后喷涂工艺,可以有效保护饲料中酶的活性,达到加酶饲料的最佳效果,提高饲料品质。因此,液体酶喷涂技术具有很好的推广应用价值。目前国外许多大型饲料厂已广泛地采用这一工艺。
但是在如今液体酶制剂以单一酶类居多,虽然还有液体复合酶但其使用范围过广包括家禽家畜等,从而没有针对性,具体使用时会因不同种类的畜、禽对酶需求的差异有些酶不需要或需要的含量较少则会造成酶的浪费亦或缺少某些酶需要补加对饲料的使用者带来极大的不便。
发明内容
本发明所要解决的技术问题是提供一种液体复合酶及其制备方法,提高了仔猪对饲料的利用率并对降低仔猪腹泻率具有显著的作用,能够有效地促进断奶仔猪的生长发育。
一种仔猪饲用液体复合酶,由以下步骤制备而成:
步骤一、破碎将固体发酵曲料用破碎机将曲料块破碎至5mm粒径,与水以1:1-1.5的比例混合进行搅拌10-20分钟得到湿润曲料;
步骤二、用压榨机将湿润曲料榨汁得到滤液和渣料;
步骤三、过滤:加入5%硅藻土,用板框过滤机过滤获得滤清液;
步骤四、防腐稳定:滤清液︰稳定剂︰防腐剂=80︰15︰5的比例添加甘油和防腐剂搅匀既得仔猪饲用液体复合酶。
如权利要求 1 所述的一种仔猪饲用液体复合酶的制备方法,其特征在于步骤一中的固体发酵曲料含有丰富的酸性蛋白酶、淀粉酶、脂肪酶、纤维素酶、果胶酶、葡聚糖酶。
进一步的、在步骤一中搅拌时间为15分钟可以使固体发酵曲料中的酶更充分的溶解在水中。
进一步的、步骤四中稳定剂是甘油与黄原胶以3:1的比例混合而成。
进一步的、步骤四中防腐剂是溶菌酶、甘氨酸、茶多酚以5:1:4的比例配合而成。
进一步的、本发明所述仔猪饲用液体复合酶的指标为:酸性蛋白酶30000-70000U/ml,淀粉酶60000-120000U/ml,脂肪酶50000-110000U/ml,纤维素酶2000-5000U/ml,果胶酶18000-50000U/ml,葡聚糖酶70000-130000U/ml。
本发明发酵曲料可以用黑曲霉、枯草芽孢杆菌、地衣芽孢杆菌等其产物含有酸性蛋白酶、淀粉酶、脂肪酶、纤维素酶、果胶酶、葡聚糖酶的菌种一种或多种经固态发酵得到的成品混合而成。
断奶仔猪日粮中添加酶制剂的目的在于弥补仔猪消化系统发育不成熟导致的内源酶分泌不足,如脂肪酶、蛋白酶、淀粉酶等,内源酶含量低,消化能力就弱,在生产上易导致仔猪营养应激反应和早期断奶综合症,引起消化功能紊乱并出现腹泻,外加现有仔猪饲料中一般以精饲料为主所以对于纤维素酶、木聚糖酶、甘露糖酶等外源酶的需求量并不大所以在配置仔猪饲用液体复合酶时应以内源酶的添加为主这样才有利于仔猪的生长发育。本发明液体复合酶以添加脂肪酶、酸性蛋白酶、淀粉酶为主纤维素酶、果胶酶、葡聚糖酶为辅可以增强仔猪的消化能力即可减少断奶后的仔猪因消化不良引起的腹泻,大大提高断奶后的抗病力,亦可减少断奶时肠黏膜损伤从而提高断奶猪免疫功能。
本发明中使用的蛋白酶为酸性蛋白酶而本发明液体复合酶的PH接近于7外加甘油与黄原胶对酶有稳定的作用,可以有效的防止蛋白酶对其它酶的分解。
甘氨酸与茶多酚都可以抑制细菌的生长并有很强的抗氧化能力与溶菌酶配伍不仅对液体复合酶的防腐效果有很大的帮助而且对防止仔猪腹泻也能起到良好的效果。
溶菌酶的抗菌谱较广,主要通过破坏细胞壁中的N-乙酰胞壁酸和N-乙酰氨基葡糖之间的β-1,4糖苷键,使细胞壁不溶性黏多糖分解成可溶性糖肽,导致细胞壁破裂内容物逸出而使细菌溶解。溶菌酶还可与带负电荷的病毒蛋白直接结合,与DNA、RNA、脱辅基蛋白形成复盐,使病毒失活。该酶具有抗菌、消炎、抗病毒等作用,与甘氨酸与茶多酚配伍可以起到更好的效果甘氨酸与茶多酚有抑制细菌繁殖的能力可以减缓细菌的生长而溶菌酶的效果可以减少细菌的数量在两者的相互作用下可以在很大程度上增加复合酶制剂的保存时间,并对预防病毒和细菌引起的仔猪腹泻起到了了良好的效果。
与现有技术相比,本发明的有益效果是:
1、本发明的液体复合酶用喷涂的方法加入饲料中避免了因制粒产生的高温对复合酶活力的影响节约了饲料的生产成本。
2、本发明液体复合酶针对仔猪消化能力差腹泻率高的特点进行配制解决了市面上液体复合酶使用范围过广而导致其针对性过差的缺点,本发明可以解决仔猪消化能力差腹泻率高对饲料利用率低的问题。
3、本发明液体复合酶使用溶菌酶、甘氨酸、茶多酚进行防腐处理有效的避免了市面上常用的防腐剂山梨酸钾等只能抑制细菌繁殖不能消灭细菌的缺陷,在很大程度上延长了液体复合酶的储存时间。
4、本发明液体复合酶中溶菌酶、甘氨酸、茶多酚相互配合有很强的抗菌、消炎、抗病毒的作用对预防病毒和细菌引起的仔猪腹泻起到了良好的效果。
5、本发明以脂肪酶、酸性蛋白酶、淀粉酶为主纤维素酶、果胶酶、葡聚糖酶为辅可以增强仔猪的消化能力即可减少断奶后的仔猪因消化不良引起的腹泻,大大提高断奶后的抗病力,亦可减少断奶时肠黏膜损伤从而提高断奶猪免疫功能。
具体实施方式
下面结合本发明的较佳实施例进行详细阐述,以使本发明的优点和特征能更易于被本领域技术人员理解,从而对本发明的保护范围做出更为清楚明确的界定。
实施例一、一种仔猪饲用液体复合酶,由以下步骤制备而成:
步骤一、破碎:取含有酸性蛋白酶、淀粉酶、脂肪酶、纤维素酶、果胶酶、葡聚糖酶的固体发酵曲料100kg放入破碎机中将曲料块破碎过5目标准筛;
步骤二、混合:将步骤一中经破碎的固体发酵曲料放入搅拌容器中再加入100kg水进行搅拌15分钟得到湿润曲料;
步骤三、压榨:用压榨机将湿润曲料榨汁得到103.7kg滤液和96.3kg渣料;
步骤四、过滤:加入5%硅藻土,用板框过滤机过滤获得滤清液102.3kg;
步骤五、防腐稳定:把溶菌酶、甘氨酸、茶多酚以5:1:4的比例配合制得防腐剂,甘油与黄原胶以3:1的比例混合制得稳定剂,将滤清液︰稳定剂︰防腐剂以80︰15︰5的比例添加甘油和防腐剂搅匀既得仔猪饲用液体复合酶。
使用方法:将液体复合酶以喷淋的方式均匀覆盖在仔猪制粒饲料即可。
实施例二、一种仔猪饲用液体复合酶,由以下步骤制备而成:
步骤一、破碎:取含有酸性蛋白酶、淀粉酶、脂肪酶、纤维素酶、果胶酶、葡聚糖酶的固体发酵曲料200kg放入破碎机中将曲料块破碎过5目标准筛;
步骤二、混合:将步骤一中经破碎的固体发酵曲料放入搅拌容器中再加入300kg水进行搅拌10分钟得到湿润曲料;
步骤三、压榨:用压榨机将湿润曲料榨汁得到307.8kg滤液和192.2kg渣料;
步骤四、过滤:加入5%硅藻土,用板框过滤机过滤获得滤清液305.1kg;
步骤五、防腐稳定:把溶菌酶、甘氨酸、茶多酚以5:1:4的比例配合制得防腐剂,甘油与黄原胶以3:1的比例混合制得稳定剂,将滤清液︰稳定剂︰防腐剂以80︰15︰5的比例添加甘油和防腐剂搅匀既得仔猪饲用液体复合酶。
使用方法:将液体复合酶以喷淋的方式均匀覆盖在仔猪制粒饲料即可。
实施例三、一种仔猪饲用液体复合酶,由以下步骤制备而成:
步骤一、破碎:取含有酸性蛋白酶、淀粉酶、脂肪酶、纤维素酶、果胶酶、葡聚糖酶的固体发酵曲料150kg放入破碎机中将曲料块破碎过5目标准筛;
步骤二、混合:将步骤一中经破碎的固体发酵曲料放入搅拌容器中再加入150kg水进行搅拌20分钟得到湿润曲料;
步骤三、压榨:用压榨机将湿润曲料榨汁得到154.1滤液和145.9kg渣料;
步骤四、过滤:加入5%硅藻土,用板框过滤机过滤获得滤清液153.5kg;
步骤五、防腐稳定:把溶菌酶、甘氨酸、茶多酚以5:1:4的比例配合制得防腐剂,甘油与黄原胶以3:1的比例混合制得稳定剂,将滤清液︰稳定剂︰防腐剂以80︰15︰5的比例添加甘油和防腐剂搅匀既得仔猪饲用液体复合酶。
使用方法:将液体复合酶以喷淋的方式均匀覆盖在仔猪制粒饲料即可。
检测;
将实施例一到实施例三所制得的仔猪饲用液体复合酶检测酶活结果如下;
对照实验;试验所用液体复合酶分别为康地恩液体饲用复合酶与本发明仔猪饲用液体复合酶,其用量为每吨饲料100ml。
本实验所用康地恩液体饲用复合酶其含木聚糖酶、纤维素酶、甘露聚糖酶、果胶酶、β-葡聚糖酶、蛋白酶、淀粉酶。将28日龄的断奶仔猪460头,完全随机分成4 个处理组,分为对照组和3个试验组。每组4个重复(阳面和阴面猪舍各2重复),每个重复10头仔猪。试验时间共计15天,实验之前仔猪预饲7 天,从预饲期开始,对照组与试验组饲喂基础日粮,预饲期过后对照组饲喂康地恩液体饲用复合酶加基础日粮其余三组分别用实施例一、二、三加基础日粮进行饲喂,饲喂方法采用颗粒料直接投喂的方法,不限量、自动饮水,钢筋网床。温度、湿度正常。按常规的程序与方法进行编号、驱虫、去势和免疫等管理。各组饲喂密度相同。试验开始和试验结束时,以栏为单位,均于第二天清晨空腹对每个重复的猪只进行称重。以栏为单位记录初始投料量与试验结束时剩料量,并详细记录各组猪在试验期间的腹泻数量。实验结果如下表所示。

从上表可以看出本发明饲用液体复合酶与在市面上常用的液体复合酶进行对比,可以看出本发明对于仔猪腹泻的预防有明显的效果,从料肉比可以看出本发明与一般液体复合酶相比料肉比更低可以看出添加本发明的液体复合酶饲料利用率更高更有利与仔猪的生长发育。
综上所述,本发明液体复合酶对预防仔猪腹泻,补充仔猪内源酶增强仔猪的消化能力,提高仔猪的饲料利用率有明显的作用与市面上现有液体复合酶对比,对与其适用性广泛但对于特定禽畜针对性较差的特点,发明了一种针对仔猪饲料利用率差、腹泻的特点的仔猪饲用液体复合酶,使其更有利于断奶仔猪的饲养。
本发明的实施例仅是本发明的较佳实施例而已,并非是对本发明作其它形式的限制,任何本领域的技术人员可能在上述揭示的技术内容基础之上加以变更或改型的实施例。在未脱离本发明技术方案内容的前提下,均应落入本发明技术方案的保护范围。
Claims (5)
1.一种仔猪饲用液体复合酶的制备方法,其特征在于所述仔猪饲用液体复合酶由以下步骤制备而成:
步骤一、破碎将固体发酵曲料用破碎机将曲料块破碎至5mm粒径,与水以1:1-1.5的比例混合进行搅拌10-20分钟得到湿润曲料;
步骤二、用压榨机将湿润曲料榨汁得到滤液和渣料;
步骤三、过滤:加入5%硅藻土,用板框过滤机过滤获得滤清液;
步骤四、防腐稳定:滤清液︰稳定剂︰防腐剂=80︰15︰5的比例添加甘油和防腐剂搅匀既得仔猪饲用液体复合酶。
2.如权利要求 1 所述的一种仔猪饲用液体复合酶的制备方法,其特征在于步骤一中的固体发酵曲料含有丰富的酸性蛋白酶、淀粉酶、脂肪酶、纤维素酶、果胶酶、葡聚糖酶。
3.如权利要求 1 所述的一种仔猪饲用液体复合酶的制备方法,其特征在于步骤一中搅拌时间为15分钟。
4.如权利要求 1 所述的一种仔猪饲用液体复合酶的制备方法,其特征在于步骤四中稳定剂是甘油与黄原胶以3:1的比例混合而成。
5.如权利要求 1 所述的一种仔猪饲用液体复合酶的制备方法,其特征在于步骤四中防腐剂是溶菌酶、甘氨酸、茶多酚以5:1:4的比例配合而成。
