发明详述
在消化后,当暴露于低pH的胃液时,包含果胶和藻酸盐的液体营养补充剂将在胃中形成凝胶。凝胶形成将导致胃中糖的缓慢释放(图1、图2和图3),避免不需要的胃肠道症状。
果胶
果胶,也被称为果胶多糖,是一种包含在陆生植物的细胞壁中的结构性杂多糖,并且富含半乳糖醛酸。已经在果胶组内识别和表征了若干种不同的多糖。同聚半乳糖醛酸(homogalacturonan)是α-(1-4)-连接的D-半乳糖醛酸的线性链。鼠李半乳糖醛酸聚糖(rhamnogalacturonan)I果胶(RG-I)包含重复二糖的骨架:→4)-α-D-半乳糖醛酸-(1,2)-α-L-鼠李糖-(1→。从许多鼠李糖残基中,分叉出(branch off)多种中性糖的侧链。中性糖主要是D-半乳糖、L-阿拉伯糖和D-木糖,其中中性糖的类型和比率随果胶的来源变化。果胶的另一种结构类型是鼠李半乳糖醛酸聚糖II(RG-II),这是一种不太常见的、复杂的、高度支化的多糖。鼠李半乳糖醛酸聚糖II可以被分类在被取代的聚半乳糖醛酸的组内,因为鼠李半乳糖醛酸聚糖II骨架由D-半乳糖醛酸单元排他地制成。
分离的果胶具有典型地60g/mol-150,000g/mol的分子量,随来源和提取条件变化。
同聚半乳糖醛酸的羧基基团本质上被甲基基团高度酯化(典型地~70%)。甲基化的羧基基团和非甲基化的羧基基团之间的比率,或者甲基化的程度(DM)取决于植物的成熟度及其他方面。在同聚半乳糖醛酸从植物中被提取后,DM常规地变化,在酸性条件下经由在甲醇中分散而增加或经由酸处理、碱处理或酶处理降低。酯化的半乳糖醛酸与非酯化的半乳糖醛酸的比率决定了果胶的胶凝行为。具有高甲酯程度(>50%)的果胶被表示为高甲氧基果胶(HM果胶)或高酯果胶(HE果胶),并且具有低甲氧基含量(<50%)的果胶被表示为低甲氧基果胶(LM果胶)或低酯果胶(LE果胶)。
非酯化的半乳糖醛酸单元可以是游离酸(羧基基团)或与钠、钾或钙的盐。部分地酯化的果胶的盐被称为果胶酯酸盐,如果酯化的程度低于5%,则盐被称为果胶酸盐,不溶性酸形式的果胶酸。根据本发明,可以使用所有形式的果胶,包括HM-果胶、LM-果胶和酰胺化的LM-果胶。优选的果胶是HM-果胶。
藻酸盐
藻酸盐,也被称为藻胶(algin)或藻酸(alginic acid),是一种广泛地分布在褐藻的细胞壁中的阴离子多糖。藻酸(alginate acid)是一种线性共聚物,具有分别以不同的顺序或嵌段共价连接在一起的(1-4)连接的β-D-甘露糖醛(M)及其C-5差向异构体α-L-古洛糖醛酸(G)残基的均聚物嵌段。单体可以出现在连续的G残基(G-嵌段)、连续的M残基(M-嵌段)或交替的M残基和G残基(MG-嵌段)的均聚物嵌段中。根据本发明,可以使用所有形式的藻酸盐,包括高G藻酸盐(>50%G)和高M藻酸盐(>50%M)。优选的藻酸盐是高G藻酸盐。抗衡离子可以是例如钠(藻酸钠)、钾(藻酸钾)、铵(藻酸铵)或其他合适的单价阳离子、或其混合物。
根据本发明可以使用的糖是但不限于单糖如葡萄糖、果糖、半乳糖,二糖如乳糖、麦芽糖、蔗糖、乳果糖、海藻糖、纤维二糖。
复合碳水化合物
根据本发明可以使用的复合碳水化合物是但不限于淀粉如直链淀粉和支链淀粉的组分、麦芽糖糊精、葡萄糖糖浆和果糖糖浆。
实施例
材料和方法
实验1.将葡萄糖(12g)、果糖(6g)、藻酸盐(0.4g)和果胶(0.6g)与水(总重量100g)在小瓶中通过摇动混合。在至少1min之后,将15ml的混合物倒入在60ml圆柱形容器(内径26mm,长度120mm)中的40ml的模拟的胃液(SGF)中,缓慢地旋转该圆柱形容器(16rpm)。在0.5min、1min、2min、3min、6min、9min、12min、15min、18min和21min采集SGF的样品(0.20ml)。作为参照,使用包含所有成分(包括SGF),但排除藻酸盐和果胶的水溶液。光密度和相应的BRIX浓度通过AT AGO PAL-3折射计测量。
实验2.与实验1相同,但仅用0.18g藻酸盐+果胶。
结果
制剂与SGF接触形成凝胶。
使用包含0.4wt%藻酸盐和0.6wt%果胶的制剂,在21min之后SGF中的游离蔗糖浓度小于参照的50%(图1)。释放的速率取决于胶凝剂藻酸盐和果胶的量。(图2和图3)。
