Abstract
本发明涉及一种香菜素的提取工艺,具体涉及一种匀浆法与超声波相结合提取香菜素的方法。其特点是,包括如下步骤:a.取天南星块茎切成1~2cm的小段置于匀浆机内,加入提取溶剂,在匀浆机中混合匀浆;b.匀浆后的物料置于超声装置中,提取2h;c.微滤膜过滤,减压浓缩至原体积的1/4‑6;d.上述提取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙醚萃取,萃取液浓缩结晶,即得香菜素。本发明流程简单、快速、高效的优点,同时整个工艺有机溶剂使用量少,能够降低成本,具有良好的应用前景。

技术领域:
本发明属于植物提取技术领域,涉及一种香菜素的提取工艺,具体涉及一种匀浆法与超声波相结合提取香菜素的方法。
背景技术:
匀浆提取是指组织通过加入萃取溶剂进行组织匀浆或磨浆,以提取组织中有效成分的一种提取方法。应用匀浆法提取植物有效成分,可以直接将物料置于匀浆机内,与提取溶剂在匀浆装置中混合匀浆,通过机械及液力剪切作用将物料撕裂和粉碎,使物料破碎和有效成分的提取同步进行,达到对植物有效成分快速、强化提取的目的。该方法具有提取完全、效率高、能耗低等特点。目前尚未见此法用于提取香菜素的报道。
发明内容:
本发明所要解决的技术问题是:针对现有技术的不足,提供一种从天南星中提取香菜素的工艺。
本发明的技术方案按照下列步骤进行:
a.取天南星块茎切成1~2cm的小段置于匀浆机内,加入提取溶剂70-80%乙醇,在匀浆机中混合匀浆;
b.匀浆后的物料置于超声装置中,提取2h;
c.微滤膜过滤,减压浓缩至原体积的1/4-6;
d.上述提取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙醚萃取,萃取液浓缩结晶,即得香菜素。
步骤a中所述的匀浆机功率为160W,匀浆转速为8000~12000r/min。
步骤c中所述的微滤膜为Al2O3,0.2μm
下面将结合具体实施方式进一步说明本发明,但本发明要求保护的范围并不局限于下列实施方式。
具体实施方式:
实施例1:
取天南星块茎切成1~2cm的小段置于匀浆机内,加入提取溶剂70%乙醇,在匀浆机中混合匀浆,匀浆机功率为160W,匀浆转速为8000r/min,匀浆后的物料置于超声装置中,提取2h,微滤膜过滤,微滤膜为Al2O3,0.2μm,减压浓缩至原体积的1/4,上述提取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙醚萃取,萃取液浓缩结晶,即得香菜素。
实施例2:
取天南星块茎切成1~2cm的小段置于匀浆机内,加入提取溶剂80%乙醇,在匀浆机中混合匀浆,匀浆机功率为160W,匀浆转速为12000r/min,匀浆后的物料置于超声装置中,提取2h,微滤膜过滤,微滤膜为Al2O3,0.2μm,减压浓缩至原体积的1/6,上述提取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙醚萃取,萃取液浓缩结晶,即得香菜素。
实施例3:
取天南星块茎切成1~2cm的小段置于匀浆机内,加入提取溶剂75%乙醇,在匀浆机中混合匀浆,匀浆机功率为160W,匀浆转速为10000r/min,匀浆后的物料置于超声装置中,提取2h,微滤膜过滤,微滤膜为Al2O3,0.2μm,减压浓缩至原体积的1/5,上述提取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙醚萃取,萃取液浓缩结晶,即得香菜素。

1.一种香菜素的提取工艺,其特征在于,包括以下步骤:
a.取天南星块茎切成1~2cm的小段置于匀浆机内,加入提取溶剂70-80%乙醇,在匀浆机中混合匀浆;
b.匀浆后的物料置于超声装置中,提取2h;
c.微滤膜过滤,减压浓缩至原体积的1/4-6;
d.上述提取物用乙醇溶液溶解,加入活性炭回流脱色,脱色液浓缩后用乙醚萃取,萃取液浓缩结晶,即得香菜素。
2.根据权利要求1所述的一种香菜素的提取工艺,其特征在于,步骤a中所述的匀浆机功率为160W,匀浆转速为8000~12000r/min。
3.根据权利要求1所述的一种香菜素的提取工艺,其特征在于,步骤c中所述的微滤膜为Al2O3,0.2μm。
