本发明属于营养食品技术领域,具体涉及一种结肠癌营养食品及其制备方法。所述的结肠癌营养食品中含有蛋白、脂肪、碳水化合物、维生素、矿物质、氨基酸、银杏黄酮、植物甾醇营养元素。本发明的营养食品除了能为机体提供充足、全面的营养外,更重要的是增强了肠道黏膜的屏障功能,更符合代谢生理状态,有利于机体蛋白质合成与代谢调节,促进肠道功能和形态恢复,防止细菌和毒素易位,明显减少肠源性感染的发生,是改善化疗患者营养状况的有效手段。对中晚期结肠癌化疗患者的营养状况能够起到一定的改善作用,降低患者不良反应,能有效提高患者的免疫力。

背景技术
据世界卫生组织国际癌症研究中心(IARC)公布的资料显示,2012年全球结肠癌的新发病例约136万例,占全部新发癌症病例的9.7%;因结肠癌死亡的病例约69.4万例,占因癌症死亡病例的8.5%。中国医学科学院肿瘤医院/国家癌症中心的统计数据显示,我国2015年结肠癌发病率、死亡率在全部恶性肿瘤中均位居前5位。
肿瘤自身的能量消耗,宿主对肿瘤的反应以及各种抗肿瘤治疗的干预等均会导致机体出现异常代谢状态。40%-80%的恶性肿瘤患者伴有不同程度的营养不良,约20%的恶性肿瘤患者直接死于营养不良,营养不良严重影响患者的生存时间及生活质量。营养不良通常是由摄入量减少、生理活动下降、机体代谢紊乱引起,严重影响临床预后,如导致肿瘤病人生活质量下降、预后不良及增加肿瘤病人治疗的不良反应、增加感染的风险和死亡率。因此,改善癌症病人的营养能帮助病人保持体重、减少化疗及放疗的不良反应、提高生活质量、延长肿瘤病人生存期。
化疗是中晚期结肠癌患者综合治疗的重要手段,但化疗药物引起的消化道反应症、免疫力低下、造血系统受抑等不良反应会进一步使患者营养状况恶化;营养不良又会导致肿瘤患者对进一步抗肿瘤治疗的耐受力下降。化疗造成营养不良的原因是多方面的,其直接原因是化疗引起的恶心、呕吐、腹泻、胃肠道黏膜损伤、食欲减退等症状,影响营养物质的摄入,且化疗药物干扰人体对营养物质的吸收与利用。在病人化疗期间,营养不良不仅会影响病人自身的生理和心理状态,而且还会影响后期治疗效果以及病人的生活质量,甚至缩短生存期。早期发现并针对营养不良情况给予适当的治疗十分必要。改善化疗病人的营养状况,不仅有助于机体的恢复,提高免疫功能,而且还能提高病人对化疗的耐受性、改善治疗效果和生存质量,延长生存期。
对于特殊医学用途食品,在美国、欧盟、日本等发达国家都有相应的规定和开发应用;我国GB29922-2013《特殊医学用途配方食品通则》食品安全国家标准中全营养食品明确规定全营养配方食品是指可作为单一营养来源满足患者营养需求的特殊医学用途配方食品。目前市场上尚无针对结肠癌化疗期间的特殊医学用途食品,特别是针对特定年龄阶段,特定生理状况结肠癌患者进行设计的。因此,开发一种结肠癌全营养配方食品,具有重大的经济效益和社会意义。
发明内容
本发明的目的是提供一种结肠癌营养食品,能够为机体提供充足、全面的营养外,更重要的是能够增强肠道黏膜的屏障功能,有利于机体蛋白质合成与代谢调节,促进肠道功能和形态恢复,防止细菌和毒素易位,明显减少肠源性感染的发生;本发明同时提供其制备方法。
本发明所述的结肠癌营养食品,每百克营养食品中营养成分如下:蛋白34-58g、脂肪1.3-4g、碳水化合物34-60g、维生素A 159-889μg、维生素D 4-13μg、维生素E 4-50mg、维生素K1 17-185μg、维生素B1 0.33-1mg、维生素B2 0.332-1mg、维生素B6 0.332-1mg、维生素B12 1-20μg、烟酸0.9-9mg、叶酸88-938μg、泛酸2-20mg、维生素C 2.2-7mg、生物素9-85μg、钠60-70mg、钾90-115mg、铜182-230μg、镁73-100mg、铁4-9mg、锌1.2-1.8mg、锰99-125μg、钙45-54mg、磷159-164mg、碘27-50μg、硒13-91μg、铬7-228μg、谷氨酰胺100-160mg、支链氨基酸 60-110mg、精氨酸100-150mg、银杏黄酮100-150mg、植物甾醇300-500mg。

蛋白的原料选自豌豆蛋白与乳清蛋白,或选自豌豆肽与乳清蛋白;
脂肪的原料选自大豆油、菜籽油、花生油、玉米籽油、红花油、紫苏油、翅果油、元宝枫籽油或牡丹籽油中的一种或多种;

本发明豌豆肽的制备方法为:将含水量25wt.%的豌豆蛋白进行挤压,挤压后,将挤出物冷却、干燥、粉碎,再用中性蛋白酶进行酶解反应;

本发明所述的结肠癌营养食品的制备方法如下:以结肠癌营养食品质量100克计,将蛋白、碳水化合物、维生素A、维生素D、维生素E、维生素K1、维生素B1、维生素B2、维生素B6、维生素B12、烟酸、叶酸、泛酸、维生素C、生物素、钠、钾、铜、镁、铁、锌、锰、钙、磷、碘、硒、铬、谷氨酰胺、支链氨基酸、精氨酸、银杏黄酮、植物甾醇营养成分的原料灭菌后,平均分成5份,形成功能袋;
脂肪营养成分的原料灭菌后,平均分成5份,形成能量袋。
其中:灭菌功率为400-500W,灭菌时间为100-120s。
本发明所述的结肠癌营养食品的食用方法如下:
目标人群的年龄为18-64岁,不包括孕妇和哺乳期妇女;
用量为:0.4袋/(kg·d),总量不超过27袋;
按照以下两种方法之一食用:
(1)用100-260mL的开水降温至40-60℃,加入功能袋和能量袋中的营养物质,搅拌、摇匀后,直接饮用或经管饲摄入;
(2)将功能袋中的营养物质添加到不含有食盐的日常饮食中食用,日常饮食中含有的脂肪与能量袋中的脂肪含量一致。
本发明产品的销售方式为直接销售给消费者,不通过医院、超市或药店等场所或途径,减少中间环节,降低费用,实现低价薄利销售的特点,消费者可以更经济、更实惠的食用,满足消费者长期食用的要求。
本发明的有益效果如下:
本发明结肠癌营养食品的效果取决于营养成分的各自作用和彼此间的协同作用。
本发明中,豌豆蛋白或豌豆肽具有显著高效的清除各种自由基的功效,其DPPH清除率、-OH清除率、O2 -清除率分别为95%、88%、45%,而且还具有抗炎症、免疫调节、抑制肾素和ACE酶活性的作用,特别是具有防治癌症引起的与氧化应激有关的并发症作用;另外,豌豆蛋白或豌豆肽具有选择性清除引起结肠癌的ROS物质DPPH和-OH,而对于能杀死结肠癌细胞的O2 -具有较低清除率,从而保证了O2 -的高存在率和高杀癌细胞效果。同时,豌豆商业品种尚无转基因品种,致敏性低,无饲喂不耐受性;豌豆蛋白肽可以针对性清除肾小球肾炎的氧自由基和MDA,来提高患者身体状态,此外与其他蛋白质或肽相比较,它还富含精氨酸和支链氨基酸,降低了额外补充精氨酸和支链氨基酸的量。
乳清蛋白是优质蛋白质,含有β-乳球蛋白、α-乳白蛋白、免疫球蛋白等多种活性成分;乳清蛋白中脂肪、乳糖的含量低,较其它动物性蛋白质中的过量饱和脂肪、胆固醇等有害物质少,必需氨基酸种类和含量齐全,营养价值高,易被消化吸收,能增强免疫,能防止血脂和胆固醇升高,对心血管疾病有益;配方中采用豌豆蛋白和乳清蛋白的混合物的形式,结合了植物蛋白和动物蛋白两者优势,避免了单一蛋白的不足,更重要的是可以改善结肠癌患者的负氮身体状态。
脂肪的原料选自以富含ω-3不饱和脂肪酸为主的大豆油、菜籽油、花生油、玉米籽油、红花油、紫苏油、翅果油、元宝枫籽油或牡丹籽油中的一种或多种,这种搭配能够充分发挥ω-3不饱和脂肪酸改善细胞膜结构,改善细胞膜运输和免疫功能,促进T淋巴细胞免疫成熟的作用;此外,这些油通过合理搭配价格便宜、易得,其脂肪酸组成符合特定医用食品要求。
谷氨酰胺对结肠癌化疗患者有显著的辅助治疗作用,同时可减少化疗的副作用。通过强抗氧化性可以从根源上选择性清除自由基,保留对结肠癌细胞有杀死作用的超氧离子自由基,此外,氧化应激状态得到调整,也可以对改善结肠癌患者血浆中ADA、GSH、PX、CAT、SOD、MDA的表达水平,从而调节患者的身体状态,有利于患者的康复、避免疾病的恶化或复发。
与其他功能性低聚糖相比较,低聚木糖具有良好的理化特性和双歧杆菌增殖特性。另外,低聚木糖还有抗氧化性、促进钙吸收、增强机体免疫力、降低结肠癌等作用。赤藓糖醇、低聚半乳糖等可以改善饮食的口感,提高患者的胃口,降低化疗对患者的厌食影响,食用后可以改善患者的抑郁心情,同时与豌豆膳食纤维、抗性淀粉相结合调节肠胃,降低化疗带来的排便不畅等问题。羟丙基淀粉是良好的助悬剂,助悬效果好,不粘壁,易分散,絮凝颗粒细腻。麦芽糊精具有良好的溶解性,可改善产品的流动性。
精氨酸增强的肠外营养改善晚期结肠癌化疗患者的免疫功能,保护肠黏膜的免疫屏障,效果明显;支链氨基酸有利于维持患者的营养状况、降低并发症发生率、提高患者的治疗顺应性,与未补充的患者相比可提高其生活质量。
本发明高蛋白、高碳水化合物、低脂肪的配方适合结肠癌食用者身体特点,有利于身体康复。本发明的营养食品除了能为机体提供充足、全面的营养外,更重要的是增强了肠道黏膜的屏障功能,更符合代谢生理状态,有利于机体蛋白质合成与代谢调节,促进肠道功能和形态恢复,防止细菌和毒素易位,明显减少肠源性感染的发生,是改善化疗患者营养状况的有效手段。对中晚期结肠癌化疗患者的营养状况能够起到一定的改善作用,降低患者不良反应,能有效提高患者的免疫力。

实施例1
每百克结肠癌营养食品中营养成分如下:蛋白45g、脂肪4g、碳水化合物46.5g、维生素A249.42μg、维生素D 5μg、维生素E 4mg、维生素K1 19μg、维生素B1 0.4mg、维生素B2 0.4mg、维生素B6 0.45mg、维生素B12 1μg、烟酸0.915mg、叶酸205μg、泛酸2.43mg、维生素C 2.3mg、生物素9μg、钠61.33mg、钾99.41mg、铜200μg、镁73.2mg、铁4.28mg、锌1.36mg、锰100.13μg、钙47.12mg、磷160.01mg、碘29.67μg、硒15.07μg、铬7.82μg、谷氨酰胺100mg、支链氨基酸100mg、 精氨酸100mg、银杏黄酮100mg、植物甾醇300mg。
制备每百克营养食品中的原料如下:
豌豆肽25g、乳清蛋白20g、大豆油1 g、菜籽油1 g、红花油0.5g、紫苏油0.5g、牡丹籽油1.0g、果糖15g、豌豆膳食纤维15g、羟丙基淀粉10g、低聚木糖5g、赤藓糖醇1.5g、醋酸视黄酯260μg、维生素D2 5μg、D-α-醋酸生育三烯酚16mg、维生素K119μg、盐酸硫胺素400μg、核黄素400μg、盐酸吡哆醇450μg、氰钴胺1μg、烟酸915μg、叶酸205μg、D-泛酸钙2.7mg、L-抗坏血酸2.3mg、D-生物素9μg、氯化钠156mg、柠檬酸钾260mg、硫酸铜500μg、氧化镁122mg、富马酸亚铁13mg、氧化锌1.7mg、硫酸锰170μg、柠檬酸钙 270mg、磷酸二氢钾702mg、碘酸钾50μg、亚硒酸钠33μg、氯化铬40μg、谷氨酰胺100mg、支链氨基酸100mg、精氨酸100mg、银杏黄酮100mg、植物甾醇300mg。
制备方法如下:
将豌豆肽、乳清蛋白、果糖、豌豆膳食纤维、羟丙基淀粉、低聚木糖、赤藓糖醇、醋酸视黄酯、维生素D2、D-α-醋酸生育三烯酚、维生素K1、盐酸硫胺素、核黄素、盐酸吡哆醇、氰钴胺、烟酸、叶酸、D-泛酸钙、L-抗坏血酸、D-生物素、氯化钠、柠檬酸钾、硫酸铜、氧化镁、富马酸亚铁、氧化锌、硫酸锰、柠檬酸钙、磷酸二氢钾、碘酸钾、亚硒酸钠、氯化铬、谷氨酰胺、支链氨基酸、精氨酸、银杏黄酮、植物甾醇,于500W的灭菌器中灭菌100s,灭菌后,平均分成5份,形成功能袋;
将大豆油、菜籽油、红花油、紫苏油、牡丹籽油于500W的灭菌器中灭菌100s,灭菌后,平均分成5份,形成能量袋;
豌豆肽的制备方法为:将含水量25wt.%的豌豆蛋白进行挤压,挤压后,将挤出物冷却、干燥、粉碎,再用中性蛋白酶进行酶解反应,酶解反应后,豌豆肽的DPPH清除率、-OH清除率、O2 -清除率分别为95%、88%、45%。
其中挤压时的温度为60℃,螺杆转速为165r/min,模孔孔径为12mm;酶解反应温度为38℃,酶解反应的pH值为7,酶解反应中豌豆蛋白浓度为8.1g/mL,中性蛋白酶的用量为豌豆蛋白用量的12%。
实施例2
每百克结肠癌营养食品中营养成分如下:蛋白52g、脂肪2g、碳水化合物56g、维生素A287.79μg、维生素D 10μg、维生素E 5mg、维生素K1 22μg、维生素B1 0.45mg、维生素B20.5mg、维生素B6 0.6mg、维生素B12 2μg、烟酸1mg、叶酸230μg、泛酸2.52mg、维生素C 2.2mg、生物素12μg、钠64.87mg、钾99.97mg、铜190.31μg、镁80mg、铁5.8mg、锌1.35mg、锰100.04μg、钙46.10mg、磷160.01mg、碘29.67μg、硒15μg、铬8.21μg、谷氨酰胺160mg、支链氨基酸 60mg、精氨酸150mg、银杏黄酮150mg、植物甾醇500mg。
制备每百克营养食品中的原料如下:
豌豆蛋白30g、乳清蛋白22g、花生油0.5g、玉米籽油0.5g、翅果油0.5g、元宝枫籽油0.5g、果糖10g、豌豆膳食纤维10g、海带多糖5g、羟丙基淀粉10g、豌豆抗性淀粉16g、赤藓糖醇5g、醋酸视黄酯300μg、维生素D2 10μg、D-α-醋酸生育三烯酚20mg、维生素K1 22μg、盐酸硫胺素450μg、核黄素500μg、盐酸吡哆醇600μg、氰钴胺2μg、烟酸1000μg、叶酸230μg、D-泛酸钙2.8mg、L-抗坏血酸2.2mg、D-生物素12μg、氯化钠165mg、磷酸氢二钾446 mg、葡萄糖酸铜1.35mg、硫酸镁400mg、富马酸亚铁12mg、硫酸亚铁5mg、乳酸锌4mg、葡萄糖酸锌2mg、硫酸锰350μg、葡萄糖酸锰120ug、L-乳酸钙355mg、磷酸二氢钾702mg、碘酸钾50μg、富硒酵母50mg、氯化铬42μg、谷氨酰胺160mg、支链氨基酸60mg、精氨酸150mg、银杏黄酮150mg、植物甾醇500mg。
制备方法如下:
将豌豆蛋白、乳清蛋白、果糖、豌豆膳食纤维、海带多糖、羟丙基淀粉、豌豆抗性淀粉、赤藓糖醇、醋酸视黄酯、维生素D2、D-α-生育三烯酚、维生素K1、盐酸硫胺素、核黄素、盐酸吡哆醇、氰钴胺、烟酸、叶酸、D-泛酸钙、抗坏血酸棕榈酸酯、D-生物素、氯化钠、磷酸氢二钾、葡萄糖酸铜、硫酸镁、富马酸亚铁、硫酸亚铁、乳酸锌、葡萄糖酸锌、硫酸锰、葡萄糖酸锰、L-乳酸钙、磷酸二氢钾、碘酸钾、富硒酵母、氯化铬、谷氨酰胺、支链氨基酸、精氨酸、银杏黄酮、植物甾醇,于425W的灭菌器中灭菌120s,灭菌后,平均分成5份,形成功能袋;
将花生油、玉米籽油、翅果油、元宝枫籽油于425W的灭菌器中灭菌120s,灭菌后,平均分成5份,形成能量袋。

1.一种结肠癌营养食品,其特征在于每百克营养食品中营养成分如下:蛋白34-58g、脂肪1.3-4g、碳水化合物34-60g、维生素A 159-889μg、维生素D 4-13μg、维生素E 4-50mg、维生素K1 17-185μg、维生素B1 0.33-1mg、维生素B2 0.332-1mg、维生素B6 0.332-1mg、维生素B12 1-20μg、烟酸0.9-9mg、叶酸88-938μg、泛酸2-20mg、维生素C 2.2-7mg、生物素9-85μg、钠60-70mg、钾90-115mg、铜182-230μg、镁73-100mg、铁4-9mg、锌1.2-1.8mg、锰99-125μg、钙45-54mg、磷159-164mg、碘27-50μg、硒13-91μg、铬7-228μg、谷氨酰胺100-160mg、支链氨基酸60-110mg、精氨酸100-150mg、银杏黄酮100-150mg、植物甾醇300-500mg。
2.根据权利要求1所述的结肠癌营养食品,其特征在于:
蛋白的原料选自豌豆蛋白与乳清蛋白,或选自豌豆肽与乳清蛋白;
脂肪的原料选自大豆油、菜籽油、花生油、玉米籽油、红花油、紫苏油、翅果油、元宝枫籽油或牡丹籽油中的一种或多种;
碳水化合物的原料选自低聚木糖、低聚半乳糖、果糖、赤藓糖醇、海带多糖、麦芽糊精、羟丙基淀粉、膳食纤维或抗性淀粉中的一种或多种。
3.根据权利要求1所述的结肠癌营养食品,其特征在于:
维生素A的原料选自醋酸视黄酯、棕榈酸视黄酯或β-胡萝卜素中的一种或多种;
维生素D的原料选自维生素D2、维生素D3中的一种或两种;
维生素E的原料选自D-α-生育三烯酚、dl-α-生育三烯酚、维生素E琥珀酸钙中的一种或多种;
维生素B1的原料选自盐酸硫胺素、硝酸硫胺素中的一种或两种;
维生素B2的原料选自核黄素或核黄素5’-磷酸钠中的一种或两种;
维生素B6的原料选自盐酸吡哆醇;
维生素B12的原料选自氰钴胺;
烟酸的原料选自烟酸或烟酰胺中的一种或两种;
泛酸的原料选自D-泛酸钙;
维生素C的原料选自L-抗坏血酸、L-抗坏血酸钠、L-抗坏血酸钙或抗坏血酸棕榈酸酯中的一种或多种;
生物素的原料选自D-生物素。
4.根据权利要求1所述的结肠癌营养食品,其特征在于:
钠的原料选自氯化钠;
钾的原料选自磷酸氢二钾、磷酸二氢钾、氯化钾或柠檬酸钾中的一种或多种;
铜的原料选自硫酸铜或葡萄糖酸铜中的一种或两种;
镁的原料选自碳酸镁、硫酸镁、氧化镁、氯化镁或L-苏糖酸镁中的一种或多种;
铁的原料选自葡萄糖酸亚铁、富马酸亚铁、硫酸亚铁、乳酸亚铁或琥珀酸亚铁中的一种或多种;
锌的原料选自硫酸锌、柠檬酸锌、葡萄糖酸锌、氧化锌或乳酸锌中的一种或多种;
锰的原料选自硫酸锰或葡萄糖酸锰中的一种或两种;
钙的原料选自碳酸钙、醋酸钙、氯化钙、柠檬酸钙、葡萄糖酸钙、乳酸钙、磷酸氢钙、磷酸二氢钙、磷酸三钙、硫酸钙、L-乳酸钙或甘油磷酸钙中的一种或多种;
磷的原料选自磷酸二氢钾;
碘的原料选自碘酸钾;
硒的原料选自亚硒酸钠、富硒酵母或L-硒-甲基硒代半胱氨酸中的一种或多种;
铬的原料选自氯化铬。
5.根据权利要求2所述的结肠癌营养食品,其特征在于:豌豆肽的制备方法为:将含水量25wt.%的豌豆蛋白进行挤压,挤压后,将挤出物冷却、干燥、粉碎,再用中性蛋白酶进行酶解反应。
6.根据权利要求5所述的结肠癌营养食品,其特征在于:酶解反应温度为38℃,酶解反应的pH值为7,酶解反应中豌豆蛋白浓度为8.1g/mL,中性蛋白酶的用量为豌豆蛋白用量的12%;酶解反应后,豌豆肽的DPPH清除率、.OH清除率、O2 .-清除率分别为95%、88%、45%。
7.一种权利要求1所述的结肠癌营养食品的制备方法,其特征在于:以结肠癌营养食品质量100克计,将蛋白、碳水化合物、维生素A、维生素D、维生素E、维生素K1、维生素B1、维生素B2、维生素B6、维生素B12、烟酸、叶酸、泛酸、维生素C、生物素、钠、钾、铜、镁、铁、锌、锰、钙、磷、碘、硒、铬、谷氨酰胺、支链氨基酸、精氨酸、银杏黄酮、植物甾醇营养成分的原料灭菌后,平均分成5份,形成功能袋;
脂肪营养成分的原料灭菌后,平均分成5份,形成能量袋。
8.根据权利要求7所述的结肠癌营养食品的制备方法,其特征在于:灭菌功率为400-500W,灭菌时间为100-120s。
9.一种权利要求1所述的结肠癌营养食品的食用方法,其特征在于:
