本发明提供了一种改善肠道屏障的保健食品,所述保健食品含有多种维生素和矿物质,所述矿物质中含有钙离子、镁离子和硫酸根离子,在特定比例下的镁离子、钙离子和硫酸根离子的组合,可以使得紧密连接蛋白呈蜂巢式网络结构,边缘清晰可见发出较强的绿色荧光,紧密包裹细胞核,呈铺路石状,细胞间隙较小,致密程度较高,即可以调节胞间紧密连接蛋白表达,同时还可以提高细胞活力,将其添加至保健食品中,可以使得保健食品具有修复受损肠道、改善肠道屏障的功效,有利于人体健康。

本发明涉及保健食品技术领域,具体涉及一种改善肠道屏障的保健食品。
背景技术
肠道微生物系统是人体最大、最复杂的微生态系统,肠道菌群作为人体微生物组最重要的组成部分,与人体健康的关系重要而复杂。通常正常肠道功能除了消化吸收之外,还有强大的抵御肠道有害物生物及其产生的各类毒素的屏障功能。一般,保证肠道健康主要依靠肠道的三大屏障,分别为肠粘膜上皮屏障、肠道免疫细胞及其分泌物所形成的免疫屏障以及肠道正常微生物群所构成的生物屏障。然而由于饮食变化、年龄或病毒感染等原因,人体的肠道菌群会发生紊乱,导致人体出现如炎症性肠病、结直肠癌、代谢综合征和过敏性疾病等。
有研究表明,饮食是肠道微生物群落结构和功能的关键因素,饮食成分如膳食纤维、脂肪、动物蛋白、植物活性物质可直接或间接影响肠道菌群的组成和功能。与此同时,肠道微生物反过来影响机体对营养物质的摄取和吸收、新陈代谢和储存。
因此,开发出一种改善肠道屏障的保健食品,具有重大的商业价值和现实意义。
发明内容
有鉴于此,本发明的目的在于提供一种改善肠道屏障的保健食品。所述保健食品可以改善肠道屏障和通透性,调节胞间紧密连接蛋白表达,提高细胞活力。
为达到此目的,本发明采用以下技术方案:
本发明提供一种改善肠道屏障的保健食品,每100克保健食品中含有维生素A 0~60000μg,维生素D 10~750μg,维生素E 7.5~7500mg,维生素K 50~5000μg,维生素B1 0.5~1000mg,维生素B2 0.5~1000mg,维生素B6 0.5~500mg,维生素B12 1~500mg,维生素C30~6000mg,叶酸150~25000μg,烟酸5~2500mg,泛酸2~1000mg,生物素15~5000μg,胆碱200~5000mg,镁150~5000mg,锰1.5~200mg,铁7.5~1000mg,锌4~750mg,硒25~5000μg,铜0.5~75mg,钾1250~12000mg,钙600~50000mg,β-胡萝卜素0~350mg,硫酸根1.25~13000mg。
优选地,所述钙、镁和硫酸根的质量比为(1~25)∶1∶(0.01~16),优选为(1.5~22)∶1∶(0.03~10)。

本发明提供的保健食品含有多种维生素和矿物质,所述矿物质中包括镁离子、钙离子和硫酸根离子,而研究表明特定比例下的镁离子、钙离子和硫酸根离子的组合具有改善肠道屏障和通透性、调节胞间紧密连接蛋白表达以及提高细胞活力的作用,将其添加至保健食品中,可以使得保健食品具有同样的功效,有利于人体健康。

考虑到现有技术中缺乏针对性改善肠道屏障、提高细胞活力的保健食品,本发明提供了一种改善肠道屏障的保健食品,每100克保健食品中含有维生素A 0~60000μg,维生素D 10~750μg,维生素E 7.5~7500mg,维生素K50~5000μg,维生素B1 0.5~1000mg,维生素B2 0.5~1000mg,维生素B6 0.5~500mg,维生素B12 1~500mg,维生素C 30~6000mg,叶酸150~25000μg,烟酸5~2500mg,泛酸2~1000mg,生物素15~5000μg,胆碱200~5000mg,镁150~5000mg,锰1.5~200mg,铁7.5~1000mg,锌4~750mg,硒25~5000μg,铜0.5~75mg,钾1250~12000mg,钙600~50000mg,β-胡萝卜素0~350mg,硫酸根1.25~13000mg。
本发明所提供的保健食品中的上述组分和比例在功能上彼此互相支持,存在相互作用关系,只要满足上述特定的组分和比例,均能够实现本发明的技术效果。上述保健食品中含有多种维生素和矿物质,所述矿物质中含有硫酸根、钙离子和镁离子,本发明优选采用无钙培养基培养Caco-2细胞构建肠道屏障受损模型,经研究发现在特定比例下的镁离子、钙离子和硫酸根离子的组合,可以使得紧密连接蛋白呈蜂巢式网络结构,边缘清晰可见发出较强的绿色荧光,紧密包裹细胞核,呈铺路石状,细胞间隙较小,致密程度较高,即可以调节胞间紧密连接蛋白表达,同时还可以提高细胞活力,将其添加至保健食品中,可以使得保健食品具有修复受损肠道、改善肠道屏障的功效。在本发明中,所述钙离子、镁离子和硫酸根离子的质量比为(1~25)∶1∶(0.01~16),优选为(1.5~22)∶1∶(0.03~10),在一个具体的实施方案中,三者的质量比为(1.5~22)∶1∶(0.03~10),在一个具体的实施方案中,三者的质量比为(1.5~21.5)∶1∶(0.04~8,在一个具体的实施方案中,三者的质量比为(1.7~21.5)∶1∶(0.04~6),在一个具体的实施方案中,三者的质量比为(1.88~21.5)∶1∶(0.04~5)。本发明中,所述钙离子的浓度为0.9~14.4mmol/L,优选为0.9~12mmol/L,更优选为0.9~10mmol/L,最优选为0.9~3.6mmol/L;所述镁离子的浓度为0.1~1.6mmol/L,所述硫酸根离子的浓度为0.1~1.6mmol/L。在一个具体的实施方案中,钙离子的浓度为0.9~14.4mmol/L,镁离子的浓度为0.1~1.6mmol/L,硫酸根离子的浓度为0.1~1.6mmol/L;在一个具体的实施方案中,钙离子的浓度为0.9~3.6mmol/L,镁离子的浓度为0.1~1.6mmol/L,硫酸根离子的浓度为0.1~1.6mmol/L;在一个具体的实施方案中,钙离子的浓度为1.8mmol/L,镁离子的浓度为0.4mmol/L,硫酸根离子的浓度为0.4mmol/L;在一个具体的实施方案中,钙离子的浓度为1.8mmol/L,镁离子的浓度为1.6mmol/L,硫酸根离子的浓度为1.6mmol/L。
在本发明中,所述钙离子的来源包括碳酸钙、醋酸钙、酪蛋白钙、氯化钙、柠檬酸钙、柠檬酸钙、柠檬酸苹果酸钙、葡萄糖酸钙、乳酸钙、苹果酸钙、磷酸氢钙、磷酸二氢钙、磷酸钙、硫酸钙、抗坏血酸钙、甘油磷酸钙或乳钙中的任意一种或多种。所述镁离子的来源包括硫酸镁、氯化镁、碳酸镁、碳酸氢镁或葡萄糖酸镁中的任意一种或多种。所述硫酸根离子的来源包括硫酸锌、硫酸亚铁、硫酸铜或硫酸锰中的任意一种或多种。

本实施例提供一种改善肠道屏障的粉剂保健食品,食品配方如下:每100g产品中,含有原料维生素A 300μg,维生素D 10μg,维生素E 7.5mg,维生素K 50μg,维生素B1 1.0mg,维生素B2 1.0mg,维生素B6 1.0mg,维生素B121.0mg,维生素C 50mg,叶酸200μg,烟酸7.5mg,泛酸2.5mg,生物素20μg,胆碱250mg,镁150mg(来源是硫酸镁),锰1.5mg,铁10mg,锌5mg,硒25μg,铜0.5mg,钾1250mg,钙750mg,硫酸根600mg;
含有辅料乳粉50g、赤藓糖醇20g、麦芽糊精20g。
上述保健食品中,钙离子的摩尔浓度为3.75mmol/L,镁离子的摩尔浓度为1.25mmol/L,硫酸根的摩尔浓度为1.25mmol/L;
钙、镁和硫酸根的质量比为5:1:4。
制备方法如下:
将上述原理和辅料直接倒入干混机(转速20rpm/min)中进行混合,25min后,混合均匀,密封包装备用。
实施例2
本实施例提供一种改善肠道屏障的片剂保健食品,食品配方如下:每100g产品中,含有原料维生素D 750μg,维生素E 7500mg,维生素K 5000μg,维生素B1 1000mg,维生素B21000mg,维生素B6 500mg,维生素B12 500mg,维生素C 5000mg,叶酸25000μg,烟酸2500mg,泛酸1000mg,生物素5000μg,胆碱5000mg,镁3250mg(来源于硫酸镁),锰200mg,铁1000mg,锌750mg,硒5000μg,铜75mg,钾12000mg,钙45000mg,β-胡萝卜素250mg,硫酸根13000mg;
含有辅料羧甲基纤维素钠10g、二氧化硅10g、硬脂酸镁5.64g。
上述保健食品中,钙离子的摩尔浓度为11.25mmol/L,镁离子的摩尔浓度为1.35mmol/L,硫酸根的摩尔浓度为1.35mmol/L;
钙、镁和硫酸根的质量比为13.85∶1∶4。
制备方法如下:
将上述原理和辅料直接倒入干混机(转速20rpm/min)中进行混合,25min后,混合均匀,放入压片机中冲压成片,密封包装后备用。
对比例1
本对比例提供一种片剂保健食品,食品配方如下:每100g产品中,含有原料维生素A 40000μg,维生素D 750μg,维生素E 7500mg,维生素K 5000μg,维生素B1 1000mg,维生素B21000mg,维生素B6 500mg,维生素B12 500mg,维生素C 5000mg,叶酸25000μg,烟酰胺2500mg,泛酸1000mg,生物素5000μg,胆碱5000mg,锰200mg,铁1000mg,锌750mg,硒5000μg,铜75mg,钙45000mg;
含有辅料羧甲基纤维素钠13g、二氧化硅10g、硬脂酸镁5.89g。
制备方法与实施例2保持一致。
钙离子、镁离子及硫酸根离子的组合对肠道屏障损伤模型的作用
采用无钙培养基培养Caco-2细胞,从而构建肠道屏障受损模型。在复苏Caco-2细胞后,传代培养使其长成致密的单层细胞。当单层细胞跨膜电阻值(Trans EpithelialElectrical Resistance,TEER)超过400Ω·cm2,则认为肠道屏障模型建立成功。在Caco-2模型中TEER值超过400Ω·cm2后,正常组换液(正常DMEM培养基)继续培养,无钙组采用无钙培养基培养至肠道屏障受损状态,各组进行相关指标检测。每组设置3个平行孔。培养基信息如下:DMEM高糖培养基、DMEM无钙离子培养基、无钙离子磷酸盐缓冲液(PBS)、胎牛血清(FBS):美国Gibco公司;0.25%胰蛋白酶消化液(含EDTA):北京索莱宝科技有限公司。
模型构建完成后,观测Caco-2模型的细胞形态,并使用Millicell-ERS 2电阻仪测定单层细胞的跨膜电阻值。随后使用西格玛奥德里奇公司的荧光素(游离酸)测定模型中的表观渗透系数(Papp)值。
利用CC-8试剂盒分别检测受试物质(钙、镁及硫酸根)对Caco-2细胞存活率的影响。在96孔板中接种浓度为1.0×105个/mL得细胞悬液放置培养箱培养,待细胞生长致密后,加入不同浓度的Ca2+继续培养4天,随后加入CCK-8溶液,避光孵育2h,测定OD450nm,计算细胞存活率。
构建Caco-2肠道屏障模型后,使用无Ca2+培养基培养Caco-2细胞建立损伤模型后,设置模型组(继续无钙培养基培养),其余组加入对应受试物浓度的培养液,培养4d后进行相关指标测定。
细胞分组:模型组、钙组(Ca2+浓度:1.8mmol/L)、钙-低镁-低硫酸根组(Ca2+浓度:1.8mmol/L、Mg2+浓度:0.4mmol/L、硫酸根离子浓度:0.4mmol/L)和钙-高镁-高硫酸根组(Ca2 +浓度:1.8mmol/L、Mg2+浓度1.6mmol/L、硫酸根离子浓度为1.6mmol/L)
随后测定各组的跨膜电阻TEER值和Papp值。最后采用免疫荧光法测定各组胞间紧密连接蛋白ZO-1、Occludin、Claudin-1蛋白形态及免疫因子的表达情况。
Caco-2细胞形态学观察
对上述采用无钙培养基培养的Caco-2细胞进行表面形态测试,结果如图1所示,可以看出Caco-2细胞呈光滑的鹅卵石状,少数细胞中间有液泡,在第7d即可观察到紧密相连的细胞群,在第14d时细胞增殖较快,有少许死细胞脱落漂浮,培养基逐渐变浑浊,第21d时,细胞呈现不规则的多边形,并且逐渐融合成单层细胞,致密性增强,结果表明此时已经形成了单层肠道屏障模型。
正常组和模型组TEER值和Papp值
正常组和模型组构建方法如上文所述,追踪正常组和模型组TEER和Papp值的变化,从而确定肠道屏障损伤模型构建完成。图2(A)为正常组和模型组TEER值随时间变化的数据图;(B)为正常组和模型组Papp值的数据图。可以看出在屏障形成后开始建立损伤模型的起初2天,模型组与正常组的TEER值随时间变化趋势相近并呈上升趋势。但第3天,模型组的TEER值骤降,低于100Ω·cm2,并继续下降;而正常组的TEER仍平稳上升。在第4d,正常组细胞的Papp值小于1.0×10-6cm/s,而无钙组的Papp值显著高于正常组(P<0.01)。
各组的跨膜电阻TEER值和Papp值
图3为不同组别的Papp值的数据图,结果发现模型组的Papp值最高,说明肠道通透性最强,致密性最差,而与模型组相比,其余3组的Papp均显著降低,且钙-高镁-高硫酸根组和钙-低镁-低硫酸根组Papp值与正常组相比未见统计学差异(P>0.05)。实验结果表明两个测试比例均可以修复受损肠道受损,改善肠道屏障功能。
各组的胞间紧密连接蛋白情况
采用荧光免疫的方法测定不同组别间紧密连接蛋白表达情况,结果如图4所示,其中,A为模型组,B为正常组,C为钙-低镁-低硫酸根组,D为钙-高镁-高硫酸根组。由图4可知,模型组的细胞核皱缩变小,三种紧密连接蛋白也皱缩呈小环,散在分布并发出微弱的绿色荧光,部分紧密连接蛋白断裂或缺失,细胞间有明显可见的较大空隙,表明细胞屏障受损程度较高,致密性低。而其他各组的细胞核大而圆润,边界清晰,发出较强的深蓝色荧光;三种紧密连接蛋白呈蜂巢式网络结构,边缘清晰可见,发出较强的绿色荧光,紧密包裹细胞核,呈铺路石状,细胞间隙较小,致密程度较高。
钙离子、镁离子及硫酸根离子对细胞活力的影响
为确定钙离子和镁离子是否对细胞活性有影响,实验中也测定了不同浓度的钙离子、镁离子和硫酸根离子对细胞活力的影响。其中,表1为不同浓度钙离子对细胞活性的影响,表2为不同浓度钙离子对细胞活性的影响,表2为不同浓度钙离子对细胞活性的影响。由表1可以看出随着Ca2+浓度梯度的升高,Caco-2细胞的存活率呈先上升后下降的趋势。0.9、1.8和3.6mmol/L三个较低浓度Ca2+环境下,细胞存活率显著高于其余浓度组(P<0.05),但这此3种浓度下,细胞的存活率尚无明显差异(P>0.05)。根据此结果,将1.8mmol/L(细胞存活率75.88%)Ca2+浓度作为与后续与Mg2+和硫酸根离子的配伍浓度。随后1.8mmol/L Ca2+浓度与不同浓度Mg2+及硫酸根离子配伍,所有剂量组的细胞存活率均超过100%,Caco-2细胞的存活率呈先上升后下降的趋势,其中在0.4mmol/L时细胞存活率与0.1mmol/L相似,Mg2+和硫酸根离子浓度均为1.6mmol/L时细胞存活率最低但仍旧超过100%。依据此结果,将0.4mmol/L和1.6mmol/L这两种浓度分别设置为钙-低镁-低硫酸根组和钙-高镁-高硫酸根组中镁离子和硫酸根的浓度。

Claims (10)
1.一种改善肠道屏障的保健食品,其特征在于,每100克保健食品中含有维生素A 0~60000μg,维生素D 10~750μg,维生素E 7.5~7500mg,维生素K 50~5000μg,维生素B1 0.5~1000mg,维生素B2 0.5~1000mg,维生素B6 0.5~500mg,维生素B12 1~500mg,维生素C30~6000mg,叶酸150~25000μg,烟酸5~2500mg,泛酸2~1000mg,生物素15~5000μg,胆碱200~5000mg,镁150~5000mg,锰1.5~200mg,铁7.5~1000mg,锌4~750mg,硒25~5000μg,铜0.5~75mg,钾1250~12000mg,钙600~50000mg,β-胡萝卜素0~350mg,硫酸根1.25~13000mg。
2.根据权利要求1所述的保健食品,其特征在于,所述钙、镁和硫酸根的质量比为(1~25)∶1∶(0.01~16)。
3.根据权利要求2所述的保健食品,其特征在于,所述钙、镁和硫酸根的质量比为(1.5~22)∶1∶(0.03~10)。
4.根据权利要求1所述的保健食品,其特征在于,所述钙的来源包括碳酸钙、醋酸钙、酪蛋白钙、氯化钙、柠檬酸钙、柠檬酸钙、柠檬酸苹果酸钙、葡萄糖酸钙、乳酸钙、苹果酸钙、磷酸氢钙、磷酸二氢钙、磷酸钙、硫酸钙、抗坏血酸钙、甘油磷酸钙或乳钙中的任意一种或多种。
5.根据权利要求1所述的保健食品,其特征在于,所述镁的来源包括硫酸镁、氯化镁、碳酸镁、碳酸氢镁或葡萄糖酸镁中的任意一种或多种。
6.根据权利要求1所述的保健食品,其特征在于,所述硫酸根的来源包括硫酸锌、硫酸亚铁、硫酸铜或硫酸锰中的任意一种或多种。
7.根据权利要求1所述的保健食品,其特征在于,所述维生素A的来源包括醋酸视黄酯、棕榈酸视黄酯或β-胡萝卜素中的一种或多种;
所述维生素D的来源包括维生素D2和/或维生素D3;
所述维生素E的来源包括D-α-生育酚、D-α-醋酸生育酚、D-α-琥珀酸生育酚、d1-α-醋酸生育酚、dl-α-生育酚或维生素E琥珀酸钙中的一种或多种;
所述维生素K的来源包括维生素K1和/或维生素K2;
所述维生素B1的来源包括盐酸硫胺素和/或硝酸硫胺素;
所述维生素B2的来源包括核黄素和/或核黄素5′-磷酸钠;
所述维生素B6的来源包括盐酸吡哆醇;
所述维生素B12的来源包括氰钴胺;
所述维生素C的来源包括L-抗坏血酸、L-抗坏血酸钠、L-抗坏血酸钙或抗坏血酸棕榈酸酯中的一种或多种。
8.根据权利要求1所述的保健食品,其特征在于,所述烟酸的来源包括烟酸和/或烟酰胺;
所述泛酸的来源包括D-泛酸钙和/或D-泛酸钠;
所述生物素的来源包括D-生物素;
所述胆碱的来源包括酒石酸胆碱和/或氯化胆碱。
9.根据权利要求1所述的保健食品,其特征在于,所述锰的来源包括硫酸锰和/或葡萄糖酸锰;
所述铁的来源包括葡萄糖酸亚铁、富马酸亚铁、乳酸亚铁、硫酸亚铁、琥珀酸亚铁、焦磷酸铁、柠檬酸铁或柠檬酸亚铁钠中的一种或多种;
所述锌的来源包括硫酸锌、柠檬酸锌、三水柠檬酸锌、葡萄糖酸锌、氧化锌、乳酸锌、乙酸锌或氯化锌中的一种或多种;
所述铜的来源包括硫酸铜和/或葡萄糖酸铜;
所述硒的来源包括亚硒酸钠、富硒酵母、L-硒-甲基、硒代半胱氨酸、硒化卡拉胶或硒蛋白中的一种或多种;
所述钾的来源包括磷酸氢二钾、磷酸二氢钾、氯化钾、柠檬酸钾、碳酸钾或葡萄糖酸钾中的一种或多种。
10.根据权利要求1所述的保健食品,其特征在于,所述保健食品的剂型包括片剂、硬胶囊、软胶囊、颗粒剂、凝胶糖果、粉剂或口服溶液。
