Abstract
本发明公开了一种促花青素吸收的超微粉碎工艺应用于黑枸杞含片制备,具体包括以下步骤:(1)对原料进行筛选和清洗;(2)将黑枸杞在‑40℃至~‑10℃下冷冻干燥2~16小时;(3)将干燥后的黑枸杞加入到超微粉碎设备中,在‑20℃至0℃温度下进行粉碎,粉碎粒度为100目~300目;(4)将上述黑枸杞微粉中按重量百分比加入黑枸杞重量0.5%~2.0%的硬脂酸镁及15~30%的木糖醇以等量递增式混合搅拌均匀;(5)将所述的黑枸杞和木糖醇以及硬脂酸镁的混合微粉直接进行压片得到黑枸杞含片,压力为15~40KN;(6)紫外辐照进行黑枸杞含片的灭菌;通过冷冻干燥、低温超微粉碎及直接粉末压力的方法,在制备过程中对花青素不产生破坏,保证了花青素溶出率和高的生物利用度,同时,也最大程度的保留了黑枸杞中其他营养物质的活性。

背景技术
黑枸杞属于茄科枸杞属。分布于高山沙林、盐化沙地、河湖沿岸、干河床、荒漠河岸林中,为我国西部特有的沙漠植物品种。黑枸杞味甘、性平,富含蛋白质、脂肪、糖类、游离氨基酸、有机酸、矿物质、微量元素、生物碱、维生素C、B1、B2、钙、镁、铜、锌、锰、铁、铅、镍、镉、钴、铬、钾、钠等各种营养成分;
黑枸杞中具有高含量的花青素,同时是强有力的天然抗氧化剂和自由基的清除剂,其功效是VC的20倍、VE的50倍,享有“软黄金”的美誉。具有保护心血管、预防高血压、抗肿瘤、抗辐射、抗疲劳、抗突变及美容等作用,并能增强视力、提高夜视能力、消除眼睛疲劳,是一种应用前景十分广阔的植物功能成分;
花青素作为一种安全无毒的纯天然的抗衰老的营养补充剂,在保健食品、化妆品、医药、食品添加剂等领域都有着较大的应用。但由于花青素的高活性,其在温度、氧气等因素条件下会对花青素的稳定性产生影响。故此,在其应用中提高花青素的稳定性有着重要的意义;
中国申请专利CN 103431327A公开了一种黑果枸杞含片及其生产方法,该专利先通过抗坏血酸和柠檬酸混合水溶液对黑果枸杞进行提取,而后进行分离、浓缩等过程后,最后加入淀粉、柠檬酸等辅料进行制粒、压片得到黑果枸杞含片;
中国申请专利 CN 105816571 A公开了一种黑枸杞原花青素咀嚼片及其制备方法,该专利首先对黑枸杞用石油醚进行脱脂处理,而后又加入乙醇进行回流、抽滤、洗脱、浓缩等过程得到枸杞浸膏,而后加入乳粉、粘合剂、甜味剂等其它原料压片制备成咀嚼片;
中国申请专利 CN 105105123 A公开了一种黑果枸杞保健片及其制备方法,该专利首先对黑果枸杞用蒸馏水进行提取,经浓缩得到的提取液用乙醇进行沉淀、并用石油醚和丙酮进行反复洗涤,最终干燥的到其提取物,提取物加入人参皂甙、山药粉、脱脂奶粉等替他物料进行压片,制得黑果枸杞保健片;
在目前已公开的技术文献中,黑枸杞片剂产品一般是先对黑枸杞进行其营养成分的提取,再经过一系列复杂的工艺程序制成片剂。在整个过程中提取时加入的一些化学试剂有可能带入其不安全的物质,而且提取工艺复杂、过程中容易破坏黑枸杞中的活性成分——花青素,同时黑枸杞中的不溶物所含的营养成分也被浪费掉。如何在生产过程中保障黑枸杞中花青素的稳定性及高的利用度具有重要的现实意义。
发明内容
为了解决一般方法生产黑枸杞含片时原花青素易损失及不稳定等因素,同时保证黑枸杞花青素的溶出率及高的生物利用度,本发明的目的在于提供一种促花青素吸收的超微粉碎工艺应用于黑枸杞含片制备;
为达到上述目的,本发明所述的一种促花青素吸收的超微粉碎工艺应用于黑枸杞含片制备,具体包括以下步骤:
(1)对原料进行筛选和清洗;
(2)将黑枸杞在-40℃至~-10℃下冷冻干燥2~16小时;
(3)将干燥后的黑枸杞加入到超微粉碎设备中,在-20℃至0℃温度下进行粉碎,粉碎粒度为100目~300目;
(4)将上述黑枸杞微粉中按重量百分比加入黑枸杞重量0.1%~2.0%的硬脂酸镁及15~35%的木糖醇以等量递增式混合搅拌均匀;
(5)将所述的黑枸杞和木糖醇以及硬脂酸镁的混合微粉直接进行压片得到黑枸杞含片,压力为15~40KN;
(6)紫外辐照进行黑枸杞含片的灭菌;
优选地,对原料进行筛选和清洗的具体步骤(1)为剔除黑枸杞中泥沙、石头等杂质,采用水浴式气泡清洗技术;将清洗后的黑枸杞原料放入转速为3000r/min的离心分离机内进行脱水1~4分钟;
优选地,在步骤(4)中,所述加入的硬脂酸镁为枸杞重量百分比的0.5%~1.5%;
优选地,在步骤(4)中,所述加入的木塘村为枸杞重量百分比的18%~30%;
本发明的优点是:
(1)采用低温超微粉碎技术对黑枸杞原料进行处理,细胞的破壁率高,有利于活性成分的释放和吸收,提高了有效组分的溶出率,增加了生物利用度,节省原料,降低成本;同时在超微粉碎过程中,经控制可不产生热现象,甚至可在低温下进行,并且粉碎速度快,有利于保留不耐高温的生物活性成分和各种营养成分,继而防止花青素的损失,保证了花青素的溶出率和高的生物利用度;
(2)对黑枸杞进行冷冻干燥,避免了高温烘干等工艺造成对原料中花青素的破坏损失,确保花青素的活性;
(3)采用直接粉末压片技术,明显的优势在于同传统的湿法制粒后再压片相比,避免了传统工艺制粒过程中可能造成的产品质量的不稳定等因素,提高了花青素的稳定性;另外降低了设备和运作成本,工艺简单化,更具经济性。

以下所述的是本发明的优选实施方式,本发明所保护的不限于以下优选实施方式。应当指出,对于本领域的技术人员来说在此发明创造构思的基础上,做出的若干变形和改进,都属于本发明的保护范围,为了进一步描述本发明,下面通过具体实施例来说明;
实施例1
(1)对原料进行筛选与水浴式气泡清洗,将清洗后的黑枸杞原料放入转速为3000r/min的离心分离机内进行脱水1分钟;
(2)将脱水后的黑枸杞在-40℃下冷冻干燥2小时;
(3)将干燥后的黑枸杞加入到超微粉碎设备中,在-20℃温度下进行粉碎,粉碎粒度为100目;
(4)将上述黑枸杞微粉中按重量百分比加入黑枸杞重量0.5%的硬脂酸镁及15%的木糖醇以等量递增式混合搅拌均匀;;
(5)将所述的黑枸杞和木糖醇以及硬脂酸镁的混合微粉直接进行压片得到黑枸杞含片,压力为15KN;
(6)紫外辐照进行黑枸杞含片的灭菌;
实施例2
(1)对原料进行筛选与水浴式气泡清洗,将清洗后的黑枸杞原料放入转速为3000r/min的离心分离机内进行脱水4分钟;
(2)将脱水后的黑枸杞在-40℃至~-10℃下冷冻干燥16小时;
(3)将干燥后的黑枸杞加入到超微粉碎设备中,在0℃温度下进行粉碎,粉碎粒度为300目;
(4)将上述黑枸杞微粉中按重量百分比加入黑枸杞重量2.0%的硬脂酸镁及35%的木糖醇以等量递增式混合搅拌均匀;
(5)将所述的黑枸杞和木糖醇以及硬脂酸镁的混合微粉直接进行压片得到黑枸杞含片压力为40KN;
(6)紫外辐照进行黑枸杞含片的灭菌;
实施例3
(1)对原料进行筛选与水浴式气泡清洗,将清洗后的黑枸杞原料放入转速为3000r/min的离心分离机内进行脱水2分钟;
(2)将黑枸杞在-25℃下冷冻干燥8小时;
(3)将干燥后的黑枸杞加入到超微粉碎设备中,在-10℃温度下进行粉碎,粉碎粒度为200目;
(4)将上述黑枸杞微粉中按重量百分比加入黑枸杞重量1.0%的硬脂酸镁及20%的木糖醇以等量递增式混合搅拌均匀;;
(5)将所述的黑枸杞和木糖醇以及硬脂酸镁的混合微粉直接进行压片得到黑枸杞含片,压力为20KN;
(6)紫外辐照进行黑枸杞含片的灭菌;
将随机获取的实施例1~3中制备的黑枸杞压片糖果各80片置于白色磁盘中,用肉眼直接观察形态,色泽及品尝其味道,结果如下表所示:

从上表可以看出,本发明的实施例1~3中的黑枸杞压片糖果完整性好,无碎裂现象,表面光洁。且本发明通过冷冻干燥、低温超微粉碎及直接粉末压片的方法,在制备过程中对花青素不产生破坏,保证了花青素溶出率和高的生物利用度,同时,也最大程度的保留了黑枸杞中其他营养物质的活性。
Claims (4)
1.一种促花青素吸收的超微粉碎工艺应用于黑枸杞含片制备,其特征在于,包括以下步骤:
(1)对原料进行筛选和清洗;
(2)将黑枸杞在-40℃至~-10℃下冷冻干燥2~16小时;
(3)将干燥后的黑枸杞加入到超微粉碎设备中,在-20℃至0℃温度下进行粉碎,粉碎粒度为100目~300目;
(4)将上述黑枸杞微粉中按重量百分比加入黑枸杞重量0.5%~2.0%的硬脂酸镁及15~30%的木糖醇以等量递增式混合搅拌均匀;
(5)将所述的黑枸杞和木糖醇以及硬脂酸镁的混合微粉直接进行压片得到黑枸杞含片,压力为15~40KN;
(6)紫外辐照进行黑枸杞含片的灭菌。
2.根据权利要求1所述的一种促花青素吸收的超微粉碎工艺应用于黑枸杞含片制备,其特征在于,对原料进行筛选和清洗的具体步骤为:剔除除黑枸杞中泥沙、石头等杂质,采用水浴式气泡清洗技术;将清洗后的黑枸杞原料放入转速为3000r/min的离心分离机内进行脱水1~4分钟。
3.根据权利要求1所述的一种促花青素吸收的超微粉碎工艺应用于黑枸杞含片制备,其特征在于在步骤(4)中,所述加入的硬脂酸镁为枸杞重量百分比的0.5%~1.5%。
4.根据权利要求1所述的一种促花青素吸收的超微粉碎工艺应用于黑枸杞含片制备,其特征在于在步骤,在步骤(4)中,所述加入的木塘村为枸杞重量百分比的18%~30%。
