Abstract
本发明属于保健品技术领域,具体地说,是涉及一种具有辅助降血糖功能的保健品及其制备方法。该保健品是由以下重量份数的中药组成的:蛹虫草5‑9份、黄芪15‑30份、瓜蒌10‑15份、桑叶5‑10份、苦瓜10‑15份、南瓜10‑20份、枸杞子6‑12份、山药15‑30份、菟丝子6‑12份、葛根10‑15份、茯苓10‑15份、山茱萸6‑12份、黄精9‑15份、玉竹6‑12份、麦冬6‑12份、知母6‑12份。将这十六味中药按一定比例进行组方,符合中药的配伍原则,可以增强各成分的原有药效,使各成分相辅相成,能有效弥补单一组分保健品的功效局限性,对人体血糖具有显著的降低作用,具有潜在的社会效益和经济效益。

技术领域
本发明属于保健品技术领域,具体地说,是涉及一种具有辅助降血糖功能的保健品及其制备方法。
背景技术
血液中的糖份称为血糖,绝大多数情况下都是葡萄糖(英文简写Glu)。体内各组织细胞活动所需的能量大部分来自葡萄糖,所以血糖必须保持一定的水平才能维持体内各器官和组织的需要。正常人在空腹血糖浓度为3.9~6.0mmol/L。空腹血糖浓度超过6.0mmol/L称为高血糖。
人体的血糖是由一对矛盾的激素调节的:他们就是胰岛素和胰高血糖素,当感受到血液中的血糖低的时候,胰岛的A细胞会分泌胰高血糖素,动员肝脏的储备糖原,释放入血液,导致血糖上升;当感受到血液中的血糖过高的时候胰岛的B细胞会分泌胰岛素,促进血糖变成肝糖原储备或者促进血糖进入组织细胞。
正常情况下,人体能够通过激素调节和神经调节这两大调节系统确保血糖的来源与去路保持平衡,使血糖维持在一定水平。但是在遗传因素(如糖尿病家族史)与环境因素(如不合理的膳食、肥胖等)的共同作用下,两大调节功能发生紊乱,就会出现血糖水平的升高。长期的高血糖状态对人体多种组织器官,特别是眼、足、神经、心血管等都有长期的损伤。
高血糖是目前世界医学上难以根治的疾病之一,药物不能解决所有的问题,目前营养干预法已成为降血糖的一个广泛应用的方法,市面上出现了众多的提高机体免疫力的保健食品,其组方各异,功效不一,然而能真正有效辅助降血糖的保健品较少,且效果不明显。
发明内容
本发明的目的在于提供一种具有辅助降血糖功能的保健品,可以辅助降血糖、滋养身体,同时兼具清热润肺,益胃生津,滋阴补肾,润燥止渴的功效。
为实现本发明的上述目的,本发明采用如下技术方案。
一种具有辅助降血糖功能的保健品,该保健品是由以下重量份数的中药制成的:蛹虫草5-9份、黄芪15-30份、瓜蒌10-15份、桑叶5-10份、苦瓜10-15份、南瓜10-20份、枸杞子6-12份、山药15-30份、菟丝子6-12份、葛根10-15份、茯苓10-15份、山茱萸6-12份、黄精9-15份、玉竹6-12份、麦冬6-12份、知母6-12份。

一种具有辅助降血糖功能的保健品,具体步骤如下:
步骤1、预处理:凭生产指令领取物料并核对物料代号、品名、规格、编号和重量;
步骤2、提取:按配方比例取蛹虫草、黄芪、瓜蒌、桑叶、苦瓜、南瓜、枸杞子、山药、菟丝子、葛根、茯苓、山茱萸、黄精、玉竹、麦冬、知母,加水煎煮 2 次,每次加药材重量10倍的水量,每次煎煮2小时,过滤,浓缩到65℃时相对密度为 1.15 的浸膏,提取后备用;
步骤3、干燥:将所述提取后的蛹虫草、黄芪、瓜蒌、桑叶、苦瓜、南瓜、枸杞子、山药、菟丝子、葛根、茯苓、山茱萸、黄精、玉竹、麦冬、知母浸膏喷雾干燥、粉碎后,过60目筛;
步骤4、填充:所述提取物检验合格后,加入适宜的辅料,制成所需剂型;
步骤5、抛光:填充合格的胶囊,置胶囊抛光机中抛光,去除表面药粉;
步骤6、内包装:分装入口服固体药用高密度聚乙烯瓶;
步骤7、外包装:装箱,可以采用瓦楞纸箱,形成成品;
步骤8、入库:所述成品经检验合格后,进入库房的产品合格区。
所述过筛、混合、填充、抛光、及内包装工序的生产环境的空气洁净度要求30万级,符合GB17405要求;所述预处理、外包装、入库工序在一般控制区。
在本发明的组方中,各组分具有如下药理的作用:
蛹虫草补肺益肾,化痰止咳。用于久咳虚喘,产后虚弱、阳痿阴冷等“虚”的病症。冬虫夏草主要含有冬虫夏草素、虫草酸、腺苷和多糖等成分;冬虫夏草素是抗癌的活性物质,对人体的内分泌系统和神经系统有好的调节作用,虫草酸有调节心、脑、血管的作用,促进人体的新陈代谢、改善人体的微循环、降血脂、降血压。
黄芪益气固表、敛汗固脱、托疮生肌、利水消肿。用于治疗气虚乏力,中气下陷,久泻脱肛,便血崩漏,表虚自汗,痈疽难溃,久溃不敛,血虚萎黄,内热消渴,慢性肾炎,蛋白尿,糖尿病等。黄芪对糖代谢呈双向调节作用,能显著降低葡萄糖负荷后小鼠的血糖水平,对抗肾上腺素所致的血糖升高,又能对抗苯乙双胍所致的小鼠血糖降低。黄芪对正常血糖无明显影响。
瓜蒌润肺,化痰,散结,润肠。治痰热咳嗽,胸痹,结胸,肺痿咳血,消渴,黄疸,便秘,痈肿初起。研究表明瓜蒌子油具有降血糖以及改善糖耐量的作用,其降血糖的作用可能与升高血清胰岛素含量,降低血清的NO和NOS水平有关。瓜蒌子原药材及其石油醚提取部位对模型小鼠的血糖升高有一定的抑制作用,并能促进该模型小鼠的体质量增长;此外,瓜萎子石油醚提取部位对糖耐量有一定的改善作用。
桑叶疏散风热,清肺润燥,清肝明目。用于风热感冒,头痛咳嗽;肺热燥咳;肝阳眩晕,目赤昏花;血热妄行之吐血、衄血等症。现代中、西医把桑叶和桑叶生物制剂作为改善糖尿病及其他各种疑难杂症的药物而使用,认为其药效极为广泛。有清肺润燥、止咳、去热、化痰、治盗汗;补肝、清肝明目、治疗头晕眼花、失眠、消除眼部疲劳;消肿、清血、治疗痢疾、腹痛、减肥、除脚气、利大、小肠;抗应激、凉血、降血压、降血脂、预防心肌梗塞、脑溢血、祛头痛、长发;降血糖、抗糖病等。
苦瓜祛暑涤热,明目,解毒。用于暑热烦渴,消渴,赤眼疼痛,痢疾,疮痈肿毒。苦瓜粗提取物含类似胰岛素物质,有明显的降血糖作用。正常的以及患四氧嘧啶性糖尿病的家兔灌服苦瓜浆汁后,可使血糖明显降低。皮下注射脑垂体前叶浸膏引起高血糖的大鼠,灌服苦瓜浆汁的水提取物亦有降低血糖的作用。
南瓜补中益气,消炎止痛,解毒杀虫,降糖止渴。主治久病气虚、脾胃虚弱、化痰排脓、气短倦怠、便溏、糖尿病、蛔虫等病症。现在研究表明南瓜含有丰富的钴,钴能活跃人体的新陈代谢,促进造血功能,并参与人体内维生素B12的合成,是人体胰岛细胞所必需的微量元素,对防治糖尿病、降低血糖有特殊的疗效。
枸杞子滋补肝肾,益精明目。用于虚劳精亏,腰膝酸痛,眩晕耳鸣,内热消渴,血虚萎黄,目昏不明。现代药理学研究证实,枸杞子可调节机体免疫功能、能有效抑制肿瘤生长和细胞突变、具有延缓衰老、抗脂肪肝、调节血脂和血糖等方面的作用。枸杞提取物可显著而持久降低大鼠血糖,增加糖耐量,且毒性较小。因此,枸杞子对糖尿病、血脂异常症、肝功能异常、胃炎等都有一定的治疗作用。
山药补脾养胃,生津益肺,补肾涩精。用于脾虚食少,久泻不止,肺虚喘咳,肾虚遗精,带下,尿频,虚热消渴。山药含有皂甙、黏液质,有润滑,滋润的作用;山药含有粘液蛋白,有降低血糖的作用,可用于治疗糖尿病,是糖尿病人的食疗佳品。
菟丝子补肾益精,养肝明目。适用于肝肾不足的腰膝筋骨酸痛,腿脚软弱无力、阳痿遗精、呓语、小便频数、尿有余沥、头晕眼花、视物不清、耳鸣耳聋以及妇女带下、习惯性流产等症。一般情况下被列为上品的可以久服。久服明目轻身延年。研究者采用菟丝子多糖给予糖尿病小鼠灌胃100~600mg/kg,能显著降低血糖、增加体重、肝糖原含量。
葛根解表退热,生津,透疹,升阳止泻。用于外感发热头痛、高血压颈项强痛、口渴、消渴、麻疹不透、热痢、泄泻。现代研究表明葛根素有明显的降低血糖的作用,葛根所含的黄酮类化合物有降血脂作用,能降低血清胆固醇,降低油三酯,用于治疗高血糖,高血脂病症,有显著疗效。
茯苓渗湿利水,健脾和胃,宁心安神。用于小便不利,水肿胀满,痰饮咳逆,呕逆,恶阻,泄泻,遗精,淋浊,惊悸,健忘等症。现代研究表明茯苓酸具有增强免疫力、抗肿瘤以及镇静、降血糖等的作用。茯苓多糖有明显的抗肿瘤作用;有利尿作用,能增加尿中钾、钠、氯等电解质的排出;有镇静及保护肝脏、抑制溃疡的发生、降血糖、抗放射等作用。
山茱萸补益肝肾,收敛固涩,固精缩尿,止带止崩,止汗 ,生津止渴。用于,腰膝酸痛,头晕耳鸣,健忘,遗精滑精,遗尿尿频,崩漏带下,月经不调,大汗虚脱 ,内热消渴。现代研究表明山茱萸在增加人体胰岛素分泌,提高胰岛素含量方面起到重要作用,对于降低糖尿病的发病率有显著影响。
黄精滋肾润肺,补脾益气。用于阴虚肺燥,干咳痰少;消渴多饮;脾胃虚弱,脾气虚或脾阴不足;肾虚精亏,腰膝酸软,须发早白。研究者将兔灌胃黄精浸膏,其血糖含量渐次增高,然后降低。表明黄精浸膏对肾上腺素引起的血糖过高呈显著抑制作用。
玉竹滋阴润肺,养胃生津。用于燥咳;劳嗽;热病阴液耗伤之咽干口渴;内热消渴;阴虚外感;头昏眩晕;筋脉挛痛。玉竹味甘多脂,质柔而润,是一味养阴生津的良药。研究表明以0.5g/kg玉竹浸剂给家兔肌肉注射,可使血糖升高.口服玉竹浸膏,则血糖先升后降,对肾上腺素引起的高血糖有显著的抑制作用, 对葡萄糖、四氧嘧啶引起的大鼠高血糖也有抑制作用。
麦冬养阴生津,润肺清心。用于肺燥干咳,阴虚痨嗽,喉痹咽痛,津伤口渴,内热消渴,心烦失眠,肠燥便秘。现代研究表明麦冬多糖对葡萄糖、肾上腺素、四氧嘧啶 3种高血糖模型小鼠的高血糖均有抑制作用,对正常小鼠也有明显降血糖作用。
知母清热泻火,生津润燥。清热泻火,生津润燥。用于外感热病,高热烦渴,肺热燥咳,骨蒸潮热,内热消渴,肠燥便秘。既能清泄肺胃之火,又能滋养肺肾之阴,故宜用于阴虚消渴,症见口渴、饮多、尿多者。知母水浸提取物, 能降低正常兔的血糖水平,特别是对四氧密啶性糖尿病兔作用更为显著,尿中酮体减少。
与现有技术相比本发明的有益效果。
本发明提供一种具有辅助降血糖功能的保健品。高血糖患者的常见症状为“三多一少”,多饮,多食,多尿及体重减轻,即中医所谓的“消渴”,该病以阴虚为本,燥热为标。其病变脏腑主要在肺、胃、肾三脏,又以肾最为重要。故方中重用蛹虫草益肺补肾,着重补肾;黄芪健脾,瓜蒌润肺清热为君,三药兼顾肺胃肾三脏,共奏养阴清热之功。臣以桑叶、苦瓜、葛根清热,山药、茯苓健脾和胃,枸杞子、菟丝子、山茱萸、黄精益肾,麦冬、知母、玉竹养阴,南瓜调和诸药为使。根据上述中医药理论,将这十六味中药按一定比例进行组方,符合中药的配伍原则,可以增强各成分的原有药效,使各成分相辅相成,能有效弥补单一组分保健品的功效局限性,对人体血糖具有显著的降低作用,具有潜在的社会效益和经济效益。
具体实施方式
实施例1:
本实施例提供一种具有辅助降血糖功能的保健品,是由如下重量份数的原料组成的:蛹虫草5份、黄芪15份、瓜蒌10份、桑叶5份、苦瓜10份、南瓜10份、枸杞子6份、山药15份、菟丝子6份、葛根10份、茯苓10份、山茱萸6份、黄精9份、玉竹6份、麦冬6份、知母6份。
具体制备方法如下:
步骤1、预处理:凭生产指令领取物料并核对物料代号、品名、规格、编号和重量等;

实施例4:
小鼠尾静脉注射四氧嘧啶(100mg/kg)建立糖尿病小鼠模型,给予不同剂量的实施例1所制的保健品提取物灌胃,分为低剂量组、中剂量组和高剂量。另设正常对照组、模型对照组和阳性药对照组,正常对照及模型对照组给予等体积食用色拉油,阳性药对照组给予盐酸二甲双胍片研细,用蒸馏水溶解后灌胃给药,连续给药4周,每周监测小鼠血糖,如见表1所示。
与正常对照组比较,*P﹤0.05;与模型对照组比较,#P﹤0.05 。
实验结果:给予保健品提取物不同剂量的各组小鼠血糖值呈剂量依赖性下降。
实验结论:本发明制得的保健品提取物具有降血糖作用。
实施例5:本发明君药组分的筛选试验。
小鼠尾静脉注射四氧嘧啶(100mg/kg)建立糖尿病小鼠模型,实验组给予中剂量的实施例1制得的保健品提取物灌胃。
设对照组3组,制备方法同实施例1,使用剂量同实验组,具体组方如下:
对照组1:熟地黄5份、黄芪15份、瓜蒌10份、桑叶5份、苦瓜10份、南瓜10份、枸杞子6份、山药15份、菟丝子6份、葛根10份、茯苓10份、山茱萸6份、黄精9份、玉竹6份、麦冬6份、知母6份。
对照组2:蛹虫草5份、党参15份、瓜蒌、桑叶5份、苦瓜10份、南瓜10份、枸杞子6份、山药15份、菟丝子6份、葛根10份、茯苓10份、山茱萸6份、黄精9份、玉竹6份、麦冬6份、知母6份。
对照组3:蛹虫草5份、黄芪15份、浙贝母10份、桑叶5份、苦瓜10份、南瓜10份、枸杞子6份、山药15份、菟丝子6份、葛根10份、茯苓10份、山茱萸6份、黄精9份、玉竹6份、麦冬6份、知母6份。
另设正常对照组、模型对照组和阳性药对照组,正常对照及模型对照组给予等体积食用色拉油,阳性药对照组给予盐酸二甲双胍片研细,用蒸馏水溶解后灌胃给药,连续给药4周,每周监测小鼠血糖。
实验结果见表2:给予本发明保健品提取物1、2、4周的小鼠血糖值与模型对照组比较,差异均有统计学意义,给予对照组1、2提取物4周的小鼠血糖值与模型对照组比较,差异有统计学意义,给予对照组3提取物2、4周的小鼠血糖值与模型对照组比较,差异有统计学意义,三个对照组小鼠的血糖值均高于实验组。三个对照组血糖值比较,结果为:对照组1>对照组2>对照组3。
实验结论:通过实验结果可知,以蛹虫草、黄芪、瓜蒌配伍,协同作用,作为本发明保健品的主药(君药),降血糖效果最佳。这三个主药相比,蛹虫草的降血糖作用最为显著。
与正常对照组比较,*P﹤0.05;与模型对照组比较,#P﹤0.05 。
实施例6 典型病例
病例1:
患者王某,男,37岁,以“口渴多饮3年,加重伴乏力1天”为主诉,现症见:口可多饮,疲乏无力,视物模糊,大便干,尿频量多,睡眠欠佳,饮食尚可。平素口服二甲双胍片1.0g日三次,症状未见明显好转。既往史:高血压病史半个月,血压最高达150/90mmHg。查体:双足背动脉搏动减弱,随机空腹血糖:7.4mmol/L。予患者口服该实施例1的保健品,观察4周,并监测血糖,一周后,测空腹血糖:6.9mmol/L,一个月后,口渴多饮,尿频量多症状明显减轻,测空腹血糖:6.1 mmol/L。目前患者仍坚持服用。
病例2:
患者魏某,男,45岁,以“口渴多饮1年,加重半个月”为主诉,现症见:口可多饮,消谷善饥,大便干,尿频量多,体重减轻,睡眠尚可。未系统用药。既往史:高脂血症。查体:随机空腹血糖:8.5mmol/L。予患者口服实施例2保健品,观察4周,并监测血糖,一周后,测空腹血糖:7.2mmol/L,一个月后,口渴多饮,尿频量多症状明显减轻,测空腹血糖:5.9 mmol/L。目前患者仍坚持服用。
病例3:
患者刘某,男,68岁,以“口渴多饮10余年,加重伴双下肢麻木半个月”为主诉,现症见:口可多饮,视物模糊,双下肢麻木,时有蚁行感,尿频量多,尿中泡沫增多,体重减轻,饮食睡眠尚可。平素规律注射胰岛素(诺和灵R早15μ,午10μ,晚10μ),症状控制不佳。既往史:高血压5年,血压最高达160/90mmHg。查体:双下肢浮肿(++),随机空腹血糖:9.5mmol/L。予患者口服该实施例3保健品,观察4周,并监测血糖,一周后,测空腹血糖:8.6mmol/L,一个月后,口渴多饮,尿频量多,双下肢麻木感减轻,双下肢浮肿(+),测空腹血糖:6.9mmol/L。目前患者仍坚持服用。

5.根据权利要求1所述的具有辅助降血糖功能的保健品,其特征在于,该保健品能够制成各种药学治疗学上可接受的口服制剂。
6.根据权利要求5所述的具有辅助降血糖功能的保健品,其特征在于,所述口服制剂为片剂、颗粒剂、丸剂、胶囊剂。
7.如权利要求1所述的一种具有辅助降血糖功能的保健品的制备方法,其特征在于该方法包括如下步骤:
步骤1、预处理:凭生产指令领取物料并核对物料代号、品名、规格、编号和重量;
步骤2、提取:按配方比例取蛹虫草、黄芪、瓜蒌、桑叶、苦瓜、南瓜、枸杞子、山药、菟丝子、葛根、茯苓、山茱萸、黄精、玉竹、麦冬、知母,加水煎煮 2 次,每次加药材重量10倍的水量,每次煎煮2小时,过滤,浓缩到65℃时相对密度为 1.15 的浸膏,提取后备用;
步骤3、干燥:将所述提取后的蛹虫草、黄芪、瓜蒌、桑叶、苦瓜、南瓜、枸杞子、山药、菟丝子、葛根、茯苓、山茱萸、黄精、玉竹、麦冬、知母浸膏喷雾干燥、粉碎后,过60目筛;
步骤4:所述混合粉检验合格后,加入适宜的辅料,制成所需剂型;
步骤5:分装,密封。
8.根据权利要求7所述的一种具有辅助降血糖功能的保健品的制备方法,其特征在于:所述制备方法的环境的空气洁净度要求30万级,符合GB17405要求。
