Abstract
本发明涉及食品及制药技术领域,提供一种消化道凝胶保护剂,包括凝胶剂和酸溶性交联剂,所述凝胶剂为海藻酸盐,所述酸溶性交联剂为在中性条件下不溶于水而在酸性条件下可溶的多价金属盐或碱,此外本发明还提供了该消化道凝胶保护剂的使用方法和应用。该发明具有的优点在于凝胶保护剂能够在口服后迅速在肠胃黏膜表面形成稳定的凝胶保护层,通过“包埋”与“隔离”的方式阻隔或延缓食物成分被消化道黏膜的吸收,尤其是可减少食物中脂肪、胆固醇、葡萄糖、重金属离子、酒精的吸收,达到减肥、紧急解毒、避酒、保护消化道溃疡、降低血脂、保持血糖稳定的效果且无任何毒副作用,该凝胶保护剂可以制成饮料、冲剂、片剂来服用,生产简便,使用方便。

技术领域
本发明属于食品及制药技术领域,尤其是涉及一种消化道凝胶保护剂及其使用方法和应用。
背景技术
消化道黏膜保护剂是用来治疗肠胃疾病的常用药物,比如铋剂(如胶体果胶铋等)以铋盐的形式沉积于胃粘膜,保护溃疡面并发挥抗幽门螺旋杆菌的作用。此外还有硫糖铝、铝碳酸镁、枸橼酸铋钾、蒙脱石散等药物,口服后可迅速附于胃黏膜及病损表面,形成一层保护性薄膜,使局部不再受到胃酸、消化酶、药物等的侵袭,起到隔离作用,被广泛用于胃及十二指肠溃疡的治疗。但以上药物均有副作用,比如引发便秘、铋等金属在人体的毒性积累等,而且这些药物基本只对胃黏膜有保护作用,加上形成的保护性薄膜是黏性胶体,进入肠道后保护性薄膜层会稀释或者重新溶解,无法形成稳定的保护层。
中国专利公告号CN101411477、CN102727514和CN101292994提供了由果胶作为主要成分制成的凝胶保护剂,但果胶形成凝胶的条件比较复杂而且苛刻,不易形成整个消化道内稳定的凝胶层,一旦凝胶层重新溶解或者仅仅形成的是黏性保护层,对消化道的黏膜保护、隔离能力就会大幅度降低。
海藻酸盐是一种天然的植物性食物纤维,已经广泛应用于食品医药领域,由于其具有优于其他材料的离子交联特性,可快速与多价金属离子如Ca2+、Ba2+、Sr2+、Cu2+、Zn2+、Fe2 +、Fe3+、Al3+形成热不可逆稳定的凝胶,常被用来作为凝胶剂使用,其凝胶性能优异,形成凝胶的条件很低,微量的多价金属离子(一般浓度超过千分之一)即可与海藻酸盐形成凝胶,是目前已知的天然性材料中性能较为突出的凝胶剂。中国专利公告号CN101485440和CN101933894使用了海藻酸盐作为消化道凝胶保护剂,但选用的是微溶于水的多价金属离子硫酸钙、磷酸氢钙作为交联剂,一旦接触水或者形成溶液,交联剂少量溶解的量即可与海藻酸盐发生凝胶反应,从而使得海藻酸盐在体外或者进入胃部前就已经形成凝胶。此外,中国专利公告号CN102871117同样使用了海藻酸盐作为消化道凝胶保护剂,但使用了钙的络合剂,这种钙的络合剂与钙源在胃部酸性条件下可以释放出钙离子,从而与海藻酸盐发生凝胶反应,不过,当凝胶进入肠道脱离了酸性环境下,钙的络合剂会重新与钙离子结合,使得凝胶重新溶解,对肠道的保护仅仅是黏性溶胶的保护功能,失去对肠道黏膜的凝胶保护功能。
发明内容
为了解决上述技术问题,本发明提供一种消化道凝胶保护剂及其使用方法和应用,该凝胶保护剂能够在口服后迅速在肠胃黏膜表面形成稳定的凝胶保护层,通过“包埋”与“隔离”的方式阻隔或延缓食物成分被消化道黏膜的吸收。
本发明的第一个目的在于提供一种消化道凝胶保护剂,所述消化道凝胶保护剂包括凝胶剂和酸溶性交联剂。
进一步地,所述凝胶剂为海藻酸盐。
更进一步地,所述海藻酸盐包括但不限于海藻酸钠、海藻酸钾和海藻酸铵。
进一步地,所述酸溶性交联剂为在中性条件下不溶于水而在酸性条件下可溶的多价金属盐或者碱,其中不溶于水是指在室温条件下在纯水中的溶解度不超过0.01g/100g水,所述多价金属盐或者碱包括但不限于多价金属离子的碳酸盐、草酸盐、磷酸盐、酒石酸盐、柠檬酸盐和水不溶性碱。
更进一步地,所述多价金属离子包括但不限于Ca2+、Ba2+、Sr2+、Cu2+、Zn2+、Fe2+、Fe3+、Al3+。
优选地,所述多价盐或者碱包括但不限于碳酸钙、碳酸铁、碳酸亚铁、碳酸铝、碳酸锌、碳酸铜、草酸钙、草酸亚铁、磷酸钙、磷酸铜、磷酸铝、磷酸锌、酒石酸钙、酒石酸钡、酒石酸铜、酒石酸锌、柠檬酸钙、柠檬酸锌、氢氧化铝、氢氧化铁、氢氧化亚铁、氢氧化铜、氢氧化锌。
进一步地,所述凝胶剂与酸溶性交联剂的重量比为1:0.1-1。
本发明的第二个目的在于提供一种消化道凝胶保护剂的使用方法,将所述消化道凝胶保护剂与其他食品及制药工业中常用辅助成分一起制成饮料、冲剂(固体饮料)或片剂来服用,其中片剂主要为可以使凝胶保护剂在口腔溶解的含片、口腔崩解片、泡腾片、咀嚼片等,口服后能在胃部及胃部以下消化道内形成稳定的凝胶保护层,通过“包埋”和“隔离”的方式阻隔或延缓食物成分被消化道黏膜吸收。
本发明的第三个目的在于提供一种消化道凝胶保护剂在减肥、紧急解毒、避酒、治疗消化道溃疡、降低血脂、保持血糖稳定方面的应用。
本发明提供的消化道凝胶保护剂,是通过“包埋”和“隔离”的方式阻隔或延缓食物成分被消化道黏膜的吸收,在口服时,凝胶保护剂在通过食道和刚到达胃部时是黏性溶胶状态,黏性胶体在遇到食糜或者其他食物成分时,能充分混合,凝胶保护剂中的酸溶性交联剂在遇到胃酸后释放出多价金属离子,与凝胶保护剂中的凝胶剂发生离子交联反应,凝胶剂与食糜或者其他食物成分一起固化成凝胶,形成“包埋”的效果,若凝胶剂与食糜或者其他食物成分充分混合前即发生交联反应,则形成的凝胶层在消化道黏膜与食糜或者其他食物成分形成了一道屏障,达到“隔离”的效果。此外,海藻酸盐作为凝胶剂,本身在胃部酸性条件下也会形成不溶性的海藻酸凝胶,同样可以达到“包埋”和“隔离”的效果。在具体的胃部酸性环境中,所形成的海藻酸盐离子交联凝胶与海藻酸凝胶存在一个平衡,即胃酸分泌较多胃部酸性越强时,海藻酸盐羧基质子化程度越高,海藻酸凝胶含量更多,若胃部酸性弱则海藻酸盐羧基更倾向于与多价金属离子发生交联。当食糜或者其他食物成分到达小肠及以下时,接近中性的环境下海藻酸凝胶会有部分的溶解,或者与消化道内离子发生交联或者离子交换反应,但因为在胃部形成的海藻酸盐离子交联凝胶依然处于固化状态,其凝胶也处于稳定状态,不影响其“包埋”和“隔离”的效果。此外,因为海藻酸盐本身不会被人体消化和吸收,其“包埋”和“隔离”的效果可以在胃部及胃部以下消化道内稳定存在。
本发明提供的凝胶保护剂还可以与其他的水溶性凝胶剂、成膜剂、增稠剂混合使用,比如卡拉胶、果胶、黄原胶、淀粉、羧甲基纤维素等,水溶性成膜剂、增稠剂以及其他的凝胶剂能降低凝胶保护剂的成本或者协同增加所形成凝胶的强度。
对于胃酸分泌过少的使用者,可以在凝胶保护剂中添加酸度调节剂,比如柠檬酸、酒石酸、苹果酸、乳酸等。
本发明具有的优点和积极效果是:
1.本发明提供的凝胶保护剂能覆盖胃部及胃部以下的全部消化道,食物成分会随着凝胶保护剂同时在消化道下行,用“隔离”和“包埋”两种方式阻隔或减缓食物成分的吸收;
2.本发明提供的凝胶保护剂服用方便,无论是制成的饮料、冲剂(固体饮料)、片剂,都能在口腔和食道内形成稳定溶液,这样在到达胃部时能充分保护胃黏膜;
3.本发明提供的凝胶保护剂功能性强,凝胶保护剂在胃部及胃部以下的消化道中一直保持一定强度的凝胶,对食物成分的阻隔或减缓效果好;
4.本发明提供的凝胶保护剂食用安全性高,无对人体有害成分,长期食用无任何副作用,而且多糖类材料对肠道益生菌具有营养作用以及保持大便湿润状态的功效。
具体实施方式
下面结合具体实施例对本发明作进一步说明,但不限定本发明的保护范围。
实施例1
按照海藻酸钠:碳酸钙=1:0.1(重量比)的比例配制凝胶保护剂,混合均匀后按照下面配方配制成饮料。
凝胶保护剂 10克
果葡糖浆 60克
纯净水 1000克
90℃下溶解成均匀混悬液后冷藏保存,即可制得饮料型消化道凝胶保护剂。
实施例2
按照海藻酸钾:氢氧化铁=1:0.2(重量比)的比例配制凝胶保护剂,混合均匀后按照下面配方配制成冲剂。
凝胶保护剂 10克
白砂糖 40克
全脂乳粉 30克
植脂末 20克
上述成分混合均匀后,得到100克冲剂,用1000克开水冲泡,搅拌均匀后可使用,得到固体饮料型消化道凝胶保护剂。
实施例3
按照海藻酸钠:磷酸钙=1:1(重量比)的比例配制凝胶保护剂,混合均匀后按照下面配方配制成片剂。

实施例4
按照海藻酸铵:磷酸钙=1:1(重量比)的比例配制凝胶保护剂,混合均匀后按照下面配方配制成片剂。
凝胶保护剂 500克
麦芽糊精 250克
β-环状糊精 250克
微晶纤维素 200克
乳糖 50克
硬脂酸镁 5克
聚乙二醇6000 5克
上述成分研细成粉、过筛、加入无水乙醇造粒、干燥、灭菌、压片。每片重量2克,得到咀嚼片型消化道凝胶保护剂。
实施例5
按照海藻酸钠:酒石酸钙=1:0.1(重量比)的比例配制凝胶保护剂,混合均匀后按照下面配方配制成片剂。
凝胶保护剂 500克
蜂蜜 160克
将凝胶保护剂过筛后与蜂蜜混匀,造粒、压片。每片重量2克,得到含片型消化道凝胶保护剂。

Claims (9)
1.一种消化道凝胶保护剂,其特征在于:所述消化道凝胶保护剂包括凝胶剂和酸溶性交联剂。
2.根据权利要求1所述的消化道凝胶保护剂,其特征在于:所述凝胶剂为海藻酸盐。
3.根据权利要求2所述的消化道凝胶保护剂,其特征在于:所述海藻酸盐包括但不限于海藻酸钠、海藻酸钾和海藻酸铵。
4.根据权利要求1所述的消化道凝胶保护剂,其特征在于:所述酸溶性交联剂为在中性条件下不溶于水而在酸性条件下可溶的多价金属盐或者碱,所述多价金属盐或者碱包括但不限于多价金属离子的碳酸盐、草酸盐、磷酸盐、酒石酸盐、柠檬酸盐和水不溶性碱。
5.根据权利要求4所述的消化道凝胶保护剂,其特征在于:所述多价金属离子包括但不限于Ca2+、Ba2+、Sr2+、Cu2+、Zn2+、Fe2+、Fe3+、Al3+。
6.根据权利要求4所述的消化道凝胶保护剂,其特征在于:所述多价盐或者碱包括但不限于碳酸钙、碳酸铁、碳酸亚铁、碳酸铝、碳酸锌、碳酸铜、草酸钙、草酸亚铁、磷酸钙、磷酸铜、磷酸铝、磷酸锌、酒石酸钙、酒石酸钡、酒石酸铜、酒石酸锌、柠檬酸钙、柠檬酸锌、氢氧化铝、氢氧化铁、氢氧化亚铁、氢氧化铜、氢氧化锌。
7.根据权利要求1-6中任一项所述的消化道凝胶保护剂,其特征在于:所述凝胶剂与酸溶性交联剂的重量比为1:0.1-1。
8.权利要求1-7中任一项所述的消化道凝胶保护剂的使用方法,其特征在于:将所述消化道凝胶保护剂与其他食品及制药工业中常用辅助成分一起制成饮料、冲剂或片剂来服用,口服后能在胃部及胃部以下消化道内形成稳定的凝胶保护层,通过“包埋”和“隔离”的方式阻隔或延缓食物成分被消化道黏膜吸收。
9.权利要求1-7中任一项所述的消化道凝胶保护剂在减肥、紧急解毒、避酒、治疗消化道溃疡、降低血脂、保持血糖稳定方面的应用。
