Abstract
本发明公开了一种解热消炎的葛根茶,涉及饮品领域。包括以下组分:葛根30‑40g、荸荠20‑30g、玉米须5‑10g、生姜10‑20g、低聚木糖1‑2g、益生菌粉20‑40g。本发明制得的葛根茶冲饮方便,多种组分科学搭配,解热消炎,同时可有效改善宿主的微生态平衡,提高宿主免疫力,改善宿主健康。

本发明涉及饮品领域,具体涉及一种解热消炎的葛根茶。
背景技术
葛根为豆科葛属植物野葛的根,其味甘、辛、凉,具有解肌退热、生津、除烦止渴等功效,聚营养、药用、美食于一身,素有“江南人参”的美称;我国卫生部2002年正式公布葛根为药食同源的植物,可用于功能性保健食品及新药的研发。我国葛根资源丰富,多被加工成葛根茶、葛根粉等系列产品推向市场。现有技术中,虽然存在葛根与多种营养成分,如天麻、苍术、枸杞、丹参、核桃、荷叶、决明子等复配制茶,但是通常都只考虑是否含有营养元素;或者感官功能,满足人们对色、香、味、形等嗜好的要求;其有效成分如葛根素的利用率和具体功效等还有待考察。
中国专利号CN107348063公开了一种保健葛根茶的制备方法,新鲜葛根经切条、烘干后,热水浸泡去除葛根条中部分淀粉,后加入纯的淀粉酶进行酶解去淀粉,经蒸煮熟化使酶失活;然后再加入益生菌粉发酵,随后制备发酵干粉,再配以甘草粉等制成成品。该发明加入清热解毒的甘草、金银花、夏枯草等与具有改善脑部血液循环作用的葛根黄酮、葛根素配伍,其药理作用不明确;其次,该发明通过先通过热水浸泡去除葛根条中部分淀粉,再淀粉酶降低葛根中的淀粉含量,然后通过菌粉发酵,增加纤维素等有效成分的利用率,经过的步骤较多,大大降低了葛根茶原料中功能性活性成分含量,对药材造成了较大的损失,降低了饮料的营养保健功能;再者,采用的菌粉功能不明,实际发酵效率未知。
发明内容
本发明的目的在于克服上述现有技术的缺陷,提供一种具有解热消炎功能的葛根茶饮品,其以葛根为主要原料,辅以药效相似的荸荠、玉米须等,具有增强葛根解热消炎的作用,还可添加益生菌粉,改善肠道健康,提高免疫力。
具有解热消炎功能的葛根茶,各组分配比为:葛根30-40g、荸荠20-30g、玉米须5-10g、生姜10-20g、低聚木糖1-2g。
进一步的,具有解热消炎功能的葛根茶,还包括益生菌粉20-40g,所述益生菌粉含卵磷脂6-12mg。
进一步的,所述益生菌粉由唾液乳酸菌2-4份、嗜酸乳杆菌2-4份、罗伊氏乳杆菌1-2份、双岐杆菌1-2份混合组成。
进一步的,所述解热消炎功能的葛根茶,各组分配比为:葛根35g、荸荠25g、玉米须8g、生姜15g、低聚木糖2g、益生菌粉30g,所述益生菌粉含卵磷脂9mg。
进一步的,所述的益生菌粉为唾液乳酸菌3份、嗜酸乳杆菌3份、罗伊氏乳杆菌1份、双岐杆菌2份混合组成。
进一步的,所述解热消炎的葛根茶通过下述方法制得:
S1、制备葛根片:a.精选长度为10~50cm的葛根,清洗、去皮,切成约6mm的片状;b.漂烫:采用蒸汽灭菌机,温度95℃,时间1~2min;c.烧烤:设备,烤箱,温度180℃,时间20min;d.炒茶:使用柴火炒茶,炒至茶刚回色,不回锅;e.筛分,选取小于20目,大于50目的葛根片;f.烘干:生产中采用热风进行流化干燥,温度100℃,时间20~30min;g.杀菌,留待备用。
S2、制备茶粉:将经过清洗并烘干的葛根片、荸荠、玉米须和生姜经粉碎机粉碎至200~220目,并按重量比混合均匀。
S3、湿热灭菌:在高压蒸汽灭菌锅中调节温度为115℃,灭菌30min。
S4、培养复合益生菌:将纯化的单菌按1%的接种量接种到相应菌种培养液中,在28-30℃的恒温摇床震荡培养12-24h,使其处于对数生长期,此时菌的活性高,且生长量大,随后将培养的菌液转接至含有1%茶粉的培养液中,驯化培养一周后,转接至相同培养基,在相同条件下继续培养,传代驯化过程中茶粉的含量逐步提高到5%,将传代第5次的菌液置于冷冻离心机中离心,并用无菌的PBS洗涤菌体3次,以去掉培养液,接着将菌体低温冻干,得到益生菌粉,并在每100g益生菌粉加入30mg的卵磷脂以保护微生物,按重量份计,得到混合复合益生菌。分离获得的菌种虽然具备一定的生物降解作用,但是由于环境中多种微生物的存在,以及底物的限制,使得菌种生物降解能力较低,因此需要对菌种进行进一步的驯化,提高菌体的生物作用。通过在培养过程中,逐步增加茶粉的含量,让本发明的益生菌循序渐进的适应这个繁殖环境,从而驯化出对茶粉耐受或降解能力更强的细菌;经过传代驯化的益生菌性能更稳定,功能更持久。
S5、各组分按重量比将葛根、荸荠、玉米须、生姜、低聚木糖、益生菌粉进行混合。
所述配方中各组分功效如下:
葛根:为豆科植物野葛的干燥根,性凉,味甘、辛;其主要成分为黄酮类物质,包括黄豆甙、黄豆甙元、葛根素及其它活性成分,有解肌退热、发表透疹、生津止渴、升阳止泻等多种功能。现代药理学研究表明,葛根具有解热,缓解心绞痛,增强机体免疫力,降低血糖,温和的降压作用及扩张血管等作用,用于治疗外感发热、项背强痛,麻疹透发不畅,脾虚泄泻、湿热泻痢及消渴等症。现有研究还表明,葛根的有效成分葛根素还具有抗炎、抗氧化等多种功能,葛根素可能通过提高对外来刺激因子的耐受性,从而减弱炎症相关细胞因子应答水平,如降低IL-1、IL-6、CRP、TNF-α等炎性细胞因子的产生,还可减少MMP-2、MMP-9等炎症介质的表达,降低ICAM-1细胞黏附分子的产生等,既可从一方面发挥作用,也可从多方面共同作用以达到抗炎效果。
荸荠,俗称马蹄、地栗、乌芋、菩荠等,为莎草科荸荠属多年生浅水性草本植物,性味甘寒,是一种优良的药食兼用的果蔬类食物,其肉质细嫩,汁多味甜,清脆爽口,自古有“地下雪梨”之美誉。现代医学研究表明,荸荠具有清热解毒、凉血生津、明目清音、消食醒酒利尿排淋的功效,可用于治疗黄疸、痢疾、小儿麻痹、便秘等疾病。荸荠中含有的黄酮类化合物的生理活性作用广泛,具有生物抗氧化性、清除自由基、抗衰老、治疗心脑血管疾病、降血脂降血压、降低血糖、抗癌、镇痛、免疫、抗炎、对消化性溃疡的保护作用等药用保健功能。荸荠中还含有一种抗菌成分-荸荠英,对金黄色葡萄球菌、大肠杆菌、产气肠杆菌、绿脓杆菌等有一定的抑制作用。
玉米须,又名玉米缨为禾本科植物玉蜀黍的花柱和柱头。性平、味甘、淡,其营养丰富,含有脂肪油、挥发油、黄酮类、树胶、生物碱、维生素K、苹果酸、草酸等多种有机物。中医认为,玉米须可降血脂、血压、血糖,还能利水、消肿、泄热,可治疗肾炎、肝炎、胆囊炎、胆结石等病症。且玉米须来源丰富、价格低廉、易于采集收集,具有广阔的研究与开发前景。
生姜:姜属植物的块根茎,性温,其特有的“姜辣素”能刺激胃肠黏膜,使胃肠道充血,有效地治疗吃寒凉食物过多而引起的腹胀、腹痛、腹泻、呕吐等。葛根和荸荠都属于性寒之物,加入生姜可中和寒性,即使脾胃虚寒之人也可饮食本发明的葛根茶。其次,生姜组织中的植物抑菌素-姜油醇、姜醇、姜烯、姜酚等对金黄色葡萄球菌、白色葡萄球菌、伤寒杆菌、宋内痢疾杆菌、绿脓杆菌均有明显抑制作用,具有广谱的抑菌效果。
炎症是机体对于刺激的一种防御反应,多以红、肿、热、痛和功能障碍等为标,本发明以葛根为主,配以解热抗炎的荸荠和玉米须,并加入可中和寒性的生姜,一能增强葛根的清热泄火的力量,在解热的同时,生津而不留湿,养阴而不恋邪,燥润相济,二能增强葛根消炎的功效,葛根、荸荠和玉米须中的黄酮类物质,具有消炎解毒的作用,且荸荠和生姜中还含有抗菌类成分,因此对微生物感染引起的炎症具有良好的药效,一方面通过葛根、荸荠和玉米须中的功能活性成分降低炎症反应,另一方面,通过抗菌类成分抑制感染源-微生物的生长繁殖,加速恢复人体健康。
低聚木糖:又称木寡糖,是由2-7个木糖分子以β-1,4糖苷键结合而成的功能性聚合糖。一方面低聚木糖作为甜味剂调节葛根茶的口味,另一方面低聚木糖对多种益生菌具有增效作用,尤其是双歧杆菌,它对双歧杆菌的增效作用是其他聚合糖类的近20倍。人体胃肠道内没有水解低聚木糖的酶,其可直接进入大肠内,优先被双歧杆菌利用,促进双歧杆菌增殖同时产生多种有机酸,降低肠道pH值,抑制有害菌的生长,从而使益生菌在肠道大量增殖,增强人体免疫力,改善人体健康。其次低聚木糖可竞争性的和病原微生物,如大肠杆菌、肠炎门氏菌、肺炎克雷伯氏菌、嗜水气单胞菌等表面凝集素结合,阻碍病原菌在上皮黏附,促使其随粪便排泄,减少对人体的危害。
益生菌是一类对宿主有益的活性微生物,是定植于人体肠道、生殖系统内,能产生确切健康功效从而改善宿主微生态平衡、发挥有益作用的活性有益微生物的总称。本发明选用的双歧杆菌和乳杆菌等有益菌,具有多种功能:
1)唾液乳酸菌,能耐受胆盐和酸性环境,因而能够通过低pH值的胃酸、胆汁酸及胰液等的作用,到达肠道发挥作用;唾液乳酸菌能够分泌纤维素酶,从而能帮助人体和肠道微生物破坏葛根的细胞壁,加速葛根、荸荠细胞内容物如葛根素、荸荠英等的释放,并将纤维素水解为多糖或单糖,如葡萄糖、纤维二糖和寡糖,利于人体吸收;其次,唾液乳酸菌能释放有益双歧杆菌等其他益生菌生长的物质,增加大肠内益生菌的数量和生命力,从而抑制各种有害菌,减少体内毒素生成,使肠胃更加健康。
2)嗜酸乳杆菌,主要定殖在人体小肠内,与肠粘膜上皮细胞相互作用,密切结合构成了生物学屏障,阻止了病原菌的定植与入侵,拮抗致病菌和有害微生物的生长及其毒素的粘附。嗜酸乳杆菌还可以分泌β-葡萄糖苷酶(纤维素酶复合体中的一种),它水解纤维二糖产生两分子的葡萄糖。纤维素酶在水解纤维素产葡萄糖过程中,葡萄糖和纤维二糖对纤维素酶有强烈的反馈抑制作用,这就影响了纤维素的水解速度和程度。因此,将分解纤维素的唾液乳酸菌与能利用纤维二糖的嗜酸乳杆菌混合,在发酵过程中,由于纤维二糖被嗜酸乳杆菌吸收利用,从而大大减弱了酶的反馈抑制,进一步加速了葛根、荸荠和玉米须等细胞内容物的释放。
3)罗伊氏乳杆菌是人胃肠道中天然存在的乳杆菌之一,可承受胃酸和胆汁的作用而到达小肠的上部并黏附于小肠壁。罗伊氏乳杆菌对肠黏膜具有很强的黏附能力,可改善肠道菌群分布,拮抗有害菌定植,避免罹患肠道疾病;罗伊氏乳杆菌能产生一种被称为“罗伊氏菌素”的非蛋白质类广谱抗菌物质,广泛抑制革兰氏阳性菌、革兰氏阴性菌、酵母、真菌和病原虫等的生长。
4)双歧杆菌,主要定殖在人体大肠内,能发酵能产生多种有机酸,降低肠道pH值,抑制肠道腐生菌的繁殖;可以调整复发性感染等引起的肠道菌群失衡;其次,刺激肠道免疫细胞的产生,提高其产生抗体的能力,起到防止疾病的效果。
本发明选用的双歧杆菌和乳杆菌是互利共生的关系,乳杆菌在生长中需要醋酸盐,维生素B2和叶酸等营养物质,而双歧杆菌在发酵中除能生成醋酸外,还能合成维生素B1、B2、B12和叶酸等,刚好供给乳杆菌的需要,促进乳杆菌的生长增殖;同时乳杆菌提供各种已分解的产物,促进双歧杆菌的生长。二者协同还可加快发酵产酸,加速致病菌和有害微生物的死亡。因此,本发明的选用的益生菌相互搭配,各有所长,使解热消炎的葛根茶保健功效发挥到最大。
总的来说,将具有解热消炎抗菌功能的葛根茶与益生菌复配饮用,不仅能从药理上起到解热生津,抗菌消炎,防治疾病的目的,益生菌还能促进葛根茶中有效成分葛根素、荸荠英等的浸出和利用,起到增强药效,加强营养物质的吸收作用。同时,荸荠和生姜中抗菌类成分的释出,能抑制人体有害菌如金黄色葡萄球菌、大肠杆菌等的生长,为益生菌的生长提供一个良好的环境;而本发明的益生菌在肠道内的定植,可起到拮抗致病菌和腐生菌的粘附、抑制有害微生物的生长繁殖,改善肠道健康,增强人体免疫力的作用,从改善肠道微生态的角度进一步地增强药效,对细菌性肠炎具有较好的防治效果。其次益生菌与药物不同,“己病治病,未病防病,无病保健”,重点是无病保健,健康人食用,可增进健康水平,同时会产生治疗与防病的目的。
本发明的有益效果是:
1.本发明的解热消炎的葛根茶添加有荸荠和玉米须,都具有解热消炎的作用,与清热生津的葛根茶药效相似,进行配伍后,相互协同,一能增强葛根清热泄火的力量,燥润相济;二能增强葛根消炎解毒的功效;同时辅以益生菌类,利用益生菌分泌多种活性酶类,如纤维素酶、纤维二糖酶等,促进葛根、荸荠、玉米须等中纤维素水解,加速葛根素、荸荠英等功能活性成分的浸出,进一步增强葛根茶的药效。
2.本发明通过添加可分泌纤维素酶等的益生菌类,因此食用该益生菌后,可在人体肠道内水解植物细胞壁中的纤维素,将葛根素、荸荠英等功能活性成分直接释放到小肠中,被人体吸收利用,最大程度保留了药材的营养成分。
3.本发明添加的益生菌之间相互搭配,各有所长,双歧杆菌和嗜酸乳杆菌互利共生,协同加快发酵产酸,加速致病菌和有害微生物的死亡;唾液乳酸菌和嗜酸乳杆菌协同降解纤维素,加速葛根茶中抗菌类成分的浸出,罗伊氏乳杆菌对肠黏膜具有很强的黏附能力,具有广谱抗菌能力,四者同为益生菌的生存繁殖提供了一个良好的环境;肠道内益生菌的含量和种类增加将有效改善宿主的微生态平衡,从而提高宿主免疫力,改善宿主健康,尤其是对微生物感染性炎症具有良好的预防作用。
4.本发明所制备的解热消炎的葛根茶不含任何添加剂,所选用的益生菌类也均为国家允许用于保健食品的菌类,安全健康,无毒副作用。

二、试验例
1.解热消炎的葛根茶对发热小鼠体温的影响
实验分组:将110只健康小鼠(平均体重300-320g)随机分为11组,其中,对比1为对照组,对比2为模型组,对比3为灌胃阿司匹林组,对比4为仅灌胃葛根片组,对比5为仅灌胃荸荠组,对比6为仅灌胃玉米须组,对比7为灌胃茶粉组,剩余5组为灌胃本发明实施例1~3所述的葛根茶组。所有小鼠实验前禁食12h,但不禁水。
小鼠造模:用干酵母致大鼠发热作为动物发热模型。取干酵母粉15g置钵体中,逐渐加蒸馏水磨为均匀的悬液,定容为75mL(20%),临用前配制。除对照组外,各鼠于背部皮下注射20%的酵母混悬液10mL/kg,对照组注射等量无菌生理盐水(大鼠皮下注射干酵母悬液后,大鼠体温先有一个迅速而明显的体温降低时期,至4h左右接近正常,随后体温逐渐升高,至6~8h左右达高峰,持续12h以上)。在造模6h时灌胃给药,根据人与大鼠体表面积换算法计算每次的给药剂量,本发明的解热消炎的葛根茶剂为10g/kg(约2mL/只),阿司匹林为0.059g/kg(约2mL/只)。对照组、模型组给予等量的无菌生理盐水灌胃。
体温检测:灌胃后每1h测量肛温1次,计算体温变化值,体温变化值=各时点测量体温一基础体温。

本发明实施例1~3的解热消炎的葛根茶有减轻小鼠烦热症状,相比于模型组体温一直居高不下,本发明实施例1~3的葛根茶能明显降低小鼠体温,减轻发热症状。
2.葛根茶对脂多糖诱导的小鼠肺部炎症因子的影响
实验分组:将110只健康小鼠(平均体重300-320g)随机分为11组,其中,对比1为对照组,对比2为模型组,对比3为灌胃阿司匹林组,对比4为仅灌胃葛根片组,对比5为仅灌胃荸荠组,对比6为仅灌胃玉米须组,对比7为灌胃茶粉组,剩余5组为灌胃本发明实施例1-5所述的葛根茶组。所有小鼠实验前连续灌胃3d,空白组和模型组则连续灌胃3d生理盐水。
小鼠造模:用LPS致小鼠导致的炎症反应作为动物发炎模型。3%水合氯醛(0.3g/kg)腹腔注射麻醉小鼠,颈部正中切口5mm,分离气管,微量注射器由气管向肺方向滴注LPS溶液(2.5mg/kg),1min内滴注完毕,消毒缝合切口,小鼠保温护理。
炎症因子检测:24h后麻醉小鼠,分离气管,结扎远端支气管,小心将消毒处理后腰麻导管插入气管内,无菌生理盐水灌洗支气管,0.5ml/次,连续抽吸5次,精确回收BALF2.5ml,-80℃保存备用。再使用商业化的ELISA试剂盒检测IL-6、TNF-α、IL-10,即按照ELISA试剂盒说明操作,最后在酶标仪450nm处读取IL-6、TNF-α、IL-10光密度值。

本发明实施例1~3的解热消炎的葛根茶能降低小鼠炎症相关细胞因子IL-10、IL-6、TNF-α的产生,相比于模型组,IL-10的含量降低了1.31~1.41倍,IL-6的含量降低了1.94~2.19倍,TNF-α的含量降低了1.69~1.83倍,可见本实施例1~3的葛根茶能明显减弱炎症小鼠的炎症相关细胞因子的量,具有消炎的功效。
Claims (6)
1.一种解热消炎的葛根茶,其特征在于,包括以下组分:葛根30-40g、荸荠20-30g、玉米须5-10g、生姜10-20g、低聚木糖1-2g。
2.根据权利要求1所述的解热消炎的葛根茶,其特征在于,还包括益生菌粉20-40g,所述益生菌粉含卵磷脂6-12mg。
3.根据权利要求2所述的解热消炎的葛根茶,其特征在于,所述益生菌粉由唾液乳酸菌2-4份、嗜酸乳杆菌2-4份、罗伊氏乳杆菌1-2份、双岐杆菌1-2份混合组成。
4.根据权利要求2所述的解热消炎的葛根茶,其特征在于,包括以下组分:葛根35g、荸荠25g、玉米须8g、生姜15g、低聚木糖2g、益生菌粉30g,所述益生菌粉含卵磷脂9mg。
5.根据权利要求2所述的解热消炎的葛根茶,其特征在于,所述益生菌粉由唾液乳酸菌3份、嗜酸乳杆菌3份、罗伊氏乳杆菌1份、双岐杆菌2份混合组成。
6.根据权利要求2~5任一项所述的解热消炎的葛根茶,其特征在于,通过下述方法制得:
S1、制备葛根片,灭菌,留待备用;
S2、制备茶粉:将经过清洗并烘干的葛根片、荸荠、玉米须和生姜经粉碎机粉碎至200~220目,按重量比混合均匀;
S3、湿热灭菌:在高压蒸汽灭菌锅中调节温度为115℃,灭菌30min;
S4、培养复合益生菌:将纯化的单菌按1%的接种量接种到相应菌种培养液中,在28-30℃的恒温摇床震荡培养12-24h,使其处于对数生长期,随后将培养的菌液转接至含有1%茶粉的培养液中,驯化培养一周后,转接至相同培养基,在相同条件下继续培养,传代驯化过程中茶粉的含量逐步提高到5%,将传代第5次的菌液置于冷冻离心机中离心,并用无菌的PBS洗涤菌体3次,以去掉培养液,接着将菌体低温冻干,得到益生菌粉,并在每100g益生菌粉加入30mg的卵磷脂;
S5、各组分按重量比将葛根、荸荠、玉米须、生姜、低聚木糖、益生菌粉进行混合。
