随着人们对浆果认识的逐渐加深和保健意识的不断提高,树莓、沙棘、白刺等营养丰富的浆果越来越受到欢迎。本发明利用树莓、沙棘、白刺浆果提取的多糖制备成多糖复合泡腾片,打破了浆果传统食用方式,对于进一步实现浆果的精深加工和高值化利用具有非常重要的意义。
发明内容
本发明的目的在于提供一种基于树莓、沙棘和白刺三种浆果制备所得多糖的复合泡腾片剂。本发明的另一目的在于提供该泡腾片剂剂的用途。
具体地,本发明提供了一种树莓、沙棘和白刺多糖复合泡腾片剂,它是包括如下重量配比的原辅料制备而成:
原料:树莓多糖2~29份、沙棘多糖3~25份、白刺多糖2~35份。
辅料:柠檬酸10~35份,碳酸氢钠10~35份,葡萄糖5~25份,微晶纤维素5~28份,PEG-6000 1~5份,可溶性淀粉0.5~2份。
进一步地,其原料由如下重量配比的组分组成:
树莓多糖3~10份、沙棘多糖3~10份、白刺多糖3~10份。
本发明几个具体实施方式中,使用的配比为:树莓多糖:沙棘多糖:白刺多糖=1:1:1。
其中,树莓多糖、沙棘多糖、白刺多糖的总含量占片剂总质量的10~30%w/w。
多糖是天然有机化合物中的一类大分子物质,由单糖基通过糖苷键连接而成。越来越多的国内外研究证明某些活性多糖具有抗肿瘤、增强免疫力、抗氧化、降血糖、保肝等多种药理活性,具有很高的医疗和保健价值。沙棘多糖是由半乳糖、阿拉伯糖、甘露糖和葡萄糖组成的中性多糖,具有很强的自由基清除能力。此外,沙棘多糖在免疫调节、治疗肝炎、抑菌等方面有较高的临床利用价值。白刺多糖是由葡萄糖、鼠李糖、阿拉伯糖、半乳糖、半乳糖醛酸和甘露糖组成的多糖,具有较强的免疫调节和体外抗氧化活性。树莓多糖是由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖和半乳糖组成的中性多糖,具有清除自由基、抗肿瘤、降血糖、免疫调节、抗疲劳等多种生理活性。
其中,所述的树莓多糖、沙棘多糖、白刺多糖,分别是以树莓、沙棘或唐古特白刺为原料,经过水提或榨汁后,醇沉制备得到;进一步地,醇沉时加入乙醇至含醇量达到70~85%v/v。
更进一步地,树莓、沙棘和白刺多糖复合泡腾片剂含有如下重量配比的辅料:柠檬酸20~30份,碳酸氢钠20~30份,葡萄糖15~20份,微晶纤维素5~20份,PEG-6000 2份,可溶性淀粉1份。
本发明还提供了上述抗氧化功效的复合多糖泡腾片在制备抗氧化的药物、保健品或功能食品中的用途。
进一步地,所述药物、保健品或功能食品是清除ABTS自由基、超氧阴离子自由基或羟自由基的药物、保健品或功能食品。
优选地,所述药物、保健品或功能食品是清除ABTS自由基或超氧阴离子自由基的药物、保健品或功能食品。
本发明研究发现,树莓多糖、沙棘多糖和白刺多糖均具有良好的体外抗氧化活性,可显著清除羟自由基、超氧阴离子自由基及ABTS自由基。但是将三种多糖组合后,在与单用各多糖相同浓度下,组合物抗氧化活性明显增强(尤其是对ABTS自由基或超氧阴离子自由基),这说明三种多糖具有协同增效作用。
本发明将树莓、沙棘和白刺多糖组合后制成复合泡腾片剂剂,具有生物利用度高、方便携带、利于保存、口感佳、适用人群范围广等诸多优点,显示出广阔的市场前景。

具体实施方式
本发明中,各多糖可以按照目前已有的技术进行制备,也可以通过购买市售产品获得。本发明中多糖参照现有技术制备,方法如下:分别取树莓、沙棘或唐古特白刺,经过水提或榨汁后,加入乙醇至70~85%v/v,沉淀制备得到。
试验例1体外抗氧化活性实验
(1)材料:树莓多糖,沙棘多糖、白刺多糖及三者以1:1:1w/w混合的多糖组合物。
(2)实验仪器及试剂:管式离心机(TGL-16G-A型,上海安亭科学仪器厂);可见紫外分光光度计(UV-722N,上海尼柯有限公司);恒温水浴锅(HD型,辽阳市恒温仪器厂)。ABTS购于上海晶纯生化科技股份有限公司(阿拉丁);过硫酸钾、磷酸二氢钠、磷酸氢二钠、氯化钠、三氯乙酸等试剂均购于山东禹王实业有限公司;超氧阴离子试剂盒、羟自由基试剂盒均购于南京建成生物科技有限公司。
(3)ABTS清除能力:将ABTS溶解(+H2O)至终浓度为7mmol/L,过硫酸钾配制成2.45mmol/L,将二者溶液混合,放置于黑暗,室温条件下搅拌16h;而后将ABTS混合溶液用PBS溶液(PH=7.0)稀释到其吸光值(于734nm处测)为0.70±0.02;树莓、沙棘、白刺三种多糖及三种多糖组合物分别用蒸馏水溶解后稀释至不同浓度(0.5mg/mL,1mg/mL,2mg/mL,4mg/mL,8mg/mL和16mg/mL);最后将上述不同浓度的样品加入到ABTS混合溶液中,其样品与ABTS混合溶液比例为1:20,混合后于室温下孵育6min,于734nm处测定吸光值。根据公式计算出清除率。ABTS清除率(%)=(1-样品吸光值/空白吸光值)×100%。结果见图1。
(4)超氧阴离子自由基清除能力:采用黄嘌呤/黄嘌呤氧化酶产生超氧阴离子自由基体系测定对超氧自由基的清除能力。将树莓、沙棘、白刺三种多糖及三种多糖组合物分别用蒸馏水溶解后稀释至不同浓度(1mg/mL,2mg/mL,4mg/mL,8mg/mL,16mg/mL)。各组依次加入75mmol/L磷酸盐缓冲液(pH7.8)1ml、样品0.2ml(对照组用蒸馏水代替)、0.1mol/L盐酸羟胺溶液0.1ml、75mmol/L黄嘌呤溶液0.1ml和0.037U/ml黄嘌呤氧化酶溶液0.1ml,混合均匀后27℃孵育30min。再加入2ml显色剂(0.4g/L甲萘胺+4g/L对氨基苯磺酸),混匀后静置5min,530nm下侧吸光值。根据公式计算出清除率。超氧阴离子自由基清除率(%)=1-样品吸光值/空白吸光值×100%。结果见图2。
(5)羟自由基清除能力:采用Fenton反应原理产生羟自由基,用番红染料显色发来测定对羟自由基的清除能力。将树莓、沙棘、白刺三种多糖及三种多糖组合物分别用蒸馏水溶解后稀释至不同浓度(1mg/mL,2mg/mL,4mg/mL,8mg/mL,16mg/mL)。各组依次加入0.15mol/L的Tris-HCl缓冲液(pH7.4)1ml、0.04mg/ml番红溶液1ml、蒸馏水1ml、0.945mmol/L的EDTANa-Fe2+溶液(pH7.4)0.5ml、样品0.5ml和1%H2O2溶液0.5ml,于37℃孵育30min,对照组用蒸馏水代替,空白组EDTANa-Fe2+溶液和样品用蒸馏水代替。在520nm下检测吸光值。根据公式计算出清除率。羟自由基清除率(%)=1-样品吸光值/空白吸光值×100%。结果见图3。

将树莓多糖、沙棘多糖和白刺多糖与碳源(碳酸氢钠)干混,加入无水乙醇润湿,而后制粒;将酸源(柠檬酸)与微晶纤维素和葡萄糖干混,加入无水乙醇润湿并制粒;于室温下挥干乙醇,再加入PEG6000和可溶性淀粉,混匀后压片即得本发明泡腾片剂。
Claims (10)
1.一种抗氧化功效的复合多糖泡腾片,其特征在于:它是包括如下重量配比的原辅料制备而成:
原料:树莓多糖2~29份、沙棘多糖3~25份、白刺多糖2~35份。
辅料:柠檬酸10~35份,碳酸氢钠10~35份,葡萄糖5~25份,微晶纤维素5~28份,PEG-6000 1~5份,可溶性淀粉0.5~2份。
2.根据权利要求1所述的抗氧化功效的复合多糖泡腾片,其特征在于:其原料配比如下:树莓多糖3~10份、沙棘多糖3~10份、白刺多糖3~10份。
3.根据权利要求1~3所述的抗氧化功效的复合多糖泡腾片,其特征在于:树莓多糖:沙棘多糖:白刺多糖=1:1:1。
4.根据权利要求1所述的抗氧化功效的复合多糖泡腾片,其特征在于:树莓多糖、沙棘多糖、白刺多糖的总含量占片剂总质量的10~30%w/w。
5.根据权利要求1所述的抗氧化功效的复合多糖泡腾片,其特征在于:其辅料重量配比如下:柠檬酸20~30份,碳酸氢钠20~30份,葡萄糖15~20份,微晶纤维素5~20份,PEG-60002份,可溶性淀粉1份。
6.根据权利要求1或2所述的抗氧化功效的复合多糖泡腾片,其特征在于:所述树莓多糖、沙棘多糖、白刺多糖,分别是以树莓、沙棘或唐古特白刺为原料,经过水提或榨汁后,醇沉制备得到。
7.根据权利要求6所述的抗氧化功效的复合多糖泡腾片,其特征在于:醇沉时加入乙醇至含醇量达到70~85%v/v。
8.权利要求1~7所述的抗氧化功效的复合多糖泡腾片在制备抗氧化的药物、保健品或功能食品中的用途。
9.根据权利要求8所述的用途,其特征在于:所述药物、保健品或功能食品是清除ABTS自由基、超氧阴离子自由基或羟自由基的药物、保健品或功能食品。
10.根据权利要求9所述的用途,其特征在于:所述药物、保健品或功能食品是清除ABTS自由基或超氧阴离子自由基的药物、保健品或功能食品。
