Abstract
本发明提供了一种青海三刺多糖组合物,其原料由如下重量配比的组份组成:黄刺多糖10~85份、沙棘多糖8~85份、唐古特白刺多糖3~25份。本发明还提供了基于该组合物的片剂。本发明研究发现,黄刺多糖、沙棘多糖、和白刺多糖单独在体外抗氧化实验中均具有良好的抗氧化活性,但将三者联合使用后,在相同浓度下即可显著提高抗羟自由基、超氧阴离子自由基,以及ABTS自由基的能力,其抗氧化活性明显增强,说明将三种多糖联合使用发挥了协同增效作用。
Description
技术领域
本发明属于保健品领域,具体涉及以黄刺、沙棘、白刺果粉制备所得多糖为原料的片剂,及其用途。
技术背景
黄刺、唐古特白刺、沙棘(黑刺)这三刺果实均为蒙、藏、维吾尔等少数民族药用与食用的传统野生浆果,又因其为低矮灌木,且具刺,因此统称为“青海三刺”。上世纪八十年代,有研究对这三种果实成分进行分析,随之也有青海三刺饮料和面霜问世。目前,对于唐古特白刺和黑刺,研究和应用十分透彻,而对于三刺果实,其复合产品的成分及功能作用应用仍较为空白。因此,我们采用青藏高原特色浆果-“青海三刺”为主要原料,开发了三种浆果资源的同时,又解决了浆果资源的销路,具有一定的经济价值。
目前还未见对青海三刺混合多糖活性及应用的报道。
发明内容
本发明的目的在于提供一种基于青海三刺即黄刺、沙棘和白刺三种浆果制备所得多糖的泡腾片剂。本发明的另一目的在于提供该泡腾片剂的制备方法和用途。
具体地,本发明提供了一种青海三刺多糖组合物,其原料由如下重量配比的组份组成:
黄刺多糖10~85份、沙棘多糖8~85份、唐古特白刺多糖3~25份。
进一步地,其原料由如下重量配比的组份组成:
黄刺多糖10~13份、沙棘多糖8~15份、唐古特白刺多糖3~10份。
本发明一个具体实施方式中,使用的配比为:黄刺多糖∶沙棘多糖∶白刺多糖=1∶1∶1。
其中,所述黄刺多糖、沙棘多糖或唐古特白刺多糖,分别是以黄刺、沙棘或唐古特白刺为原料,经过水提或榨汁后,醇沉制备得到;进一步地,醇沉时乙醇浓度达到60~85%v/v。
本发明还提供了一种青海三刺多糖片剂,它包含上述青海三刺多糖组合物,还包含药学上常用的辅料。
进一步地,青海三刺多糖组合物的含量为20~85%w/w,优选为20~30%w/w。
更进一步地,所述片剂由含有如下重量配比的原辅料制备而成:
黄刺多糖10~85份、沙棘多糖8~85份、唐古特白刺多糖3~25份;柠檬酸20~30份,碳酸氢钠20~30份,微晶纤维素8~25份,淀粉1~3份。
进一步地,所述片剂还包括1~6重量份蔗糖,PEG60001~4份。
本发明所述片剂,可以为泡腾片。
本发明还提供了上述组合物或上述片剂在制备抗氧化的药物、保健品或食品中的用途。
进一步地,所述药物、保健品或食品是清除ABTS自由基、超氧阴离子自由基或羟自由基

其中,所述黄刺为小檗科植物直穗小檗BerberisdasystachyaMaxim.,沙棘为胡颓子科沙棘属HippophaerhamnoidesLinn.;唐古特白刺为蒺藜科白刺属NitrariatangutorumBobr.。
本发明研究发现,黄刺多糖、沙棘多糖、和白刺多糖单独在体外抗氧化实验中均具有良好的抗氧化活性,但将三者联合使用后,在相同浓度下即可显著提高抗羟自由基、超氧阴离子自由基,以及ABTS自由基的能力,其抗氧化活性明显增强(尤其是抗超氧阴离子自由基),说明将三种多糖联合使用发挥了协同增效作用。
本发明将青海三刺多糖制成片剂,即便于产品的贮存、运输和包装,同时也提升了口感,便于各类人群使用。
具体实施方式
实施例1本发明多糖组合物的制备
取黄刺多糖10份、沙棘多糖15份、唐古特白刺多糖5份,混匀后,即得本发明组合物。
实施例2本发明多糖组合物的制备
取黄刺多糖12份、沙棘多糖8份、唐古特白刺多糖7份,混匀后,即得本发明组合物。
实施例3本发明多糖组合物的制备
取黄刺多糖13份、沙棘多糖10份、唐古特白刺多糖3份,混匀后,即得本发明组合物。
本发明中,各多糖可以按照目前已有的技术进行制备,也可以通过购买市售产品获得。本发明中多糖参照现有技术制备,方法如下:分别取黄刺、沙棘或唐古特白刺,经过水提或榨汁后,加入乙醇至60~85%v/v,沉淀制备得到。
实施例4本发明片剂的制备

将青海三刺浆果多糖、与碳源(碳酸氢钠)干混,后加入乙醇湿混,而后制粒;将酸源与微晶纤维素干混,后加入乙醇湿混,并制粒;常温下鼓风挥干乙醇;再加入PEG6000和淀粉,混匀后压片,即得本发明泡腾片。
体外抗氧化活性实验
(1)材料:青海三多糖:取黄刺、沙棘、白刺果实(干果或鲜果)任意一种,干燥后,粉碎,经过采用水提醇沉法制备得到,由本发明实施例制备而成,所得三种多糖以重量比黄刺多糖∶沙棘多糖∶白刺多糖=1∶1∶1的比例进行一系列浓度的配制。
(2)实验仪器及试剂:TGL-16G-A型管式离心机,上海安亭科学仪器厂;UV-722N可见紫外分光光度计,上海尼柯有限公司;HD型恒温水浴锅,辽阳市恒温仪器厂。DPPH、ABTS购于上海晶纯生化科技股份有限公司(阿拉丁);过硫酸钾、磷酸二氢钠、磷酸氢二钠、氯化钠、三氯乙酸等试剂均购于山东禹王实业有限公司;超氧阴离子试剂盒、羟自由基试剂盒均购于南京建成生物科技有限公司。
(3)ABTS清除能力:将ABTS溶解(+H2O)至终浓度为7mmol/L,过硫酸钾配制成2.45mmol/L,将二者溶液混合,放置于黑暗,室温条件下搅拌16h;而后将ABTS混合溶液用PBS溶液(PH=7.0)稀释到其吸光值(于734nm处测)为0.70±0.02;配制好的青海三刺浆果多糖用蒸馏水溶解后稀释至不同浓度;最后将上述不同浓度的样品加入到ABTS混合溶液中,其样品与ABTS混合溶液比例为1∶20,混合后于室温下孵育6min,于734nm处测定吸光值。
计算公式:ABTS清除率(%)=(1-Asample/Acontrol)×100%
其中Acontrol为ABTS混合溶液的吸光值;Asample为ABTS混合溶液中加入样品后测定的吸光值。
(4)超氧阴离子自由基清除能力和羟自由基均采用南京建成试剂盒检测。配制好的青海三刺浆果多糖用蒸馏水溶解后稀释至不同浓度。结果见表2~4.

黄刺多糖10~85份、沙棘多糖8~85份、唐古特白刺多糖3~25份。
2.根据权利要求1所述的青海三刺多糖组合物,其特征在于:其原料由如下重量配比的组份组成:
黄刺多糖10~13份、沙棘多糖8~15份、唐古特白刺多糖3~10份。
3.根据权利要求2所述的青海三刺多糖组合物,其特征在于:黄刺多糖∶沙棘多糖∶白刺多糖=1∶1∶1。
4.根据权利要求1或2所述的青海三刺多糖组合物,其特征在于:所述黄刺多糖、沙棘多糖或唐古特白刺多糖,分别是以黄刺、沙棘或唐古特白刺为原料,经过水提或榨汁后,醇沉制备得到;进一步地,醇沉时乙醇浓度达到60~85%v/v。

7.根据权利要求5或6所述的青海三刺多糖片剂,其特征在于:所述片剂由含有如下重量配比的原辅料制备而成:
黄刺多糖10~85份、沙棘多糖8~85份、唐古特白刺多糖3~25份;柠檬酸20~30份,碳酸氢钠20~30份,微晶纤维素8~25份,淀粉1~3份。
8.根据权利要求7所述的青海三刺多糖片剂,其特征在于:所述片剂还包括1~6重量份蔗糖,PEG60001~4份。
9.权利要求1~3所述组合物或权利要求5~8所述片剂在制备抗氧化的药物、保健品或食品中的用途。
10.根据权利要求9所述的用途,其特征在于:所述药物、保健品或食品是清除ABTS自由基、超氧阴离子自由基或羟自由基的药物、保健品或食品。
