Abstract
本发明提供了一种树莓、沙棘、白刺复合多糖组合物,其原料由如下重量配比的组份组成:树莓多糖3~18份、沙棘多糖4~16份、白刺多糖3~20份。本发明还提供了该组合物的用途。本发明研究发现,树莓多糖、沙棘多糖和白刺多糖均具有良好的体外抗氧化活性,可显著清除羟自由基、超氧阴离子自由基及ABTS自由基。三种多糖组合后,其体外抗氧化活性明显增强,说明三种多糖具有协同增效作用。
Description
技术领域
本发明属于保健品领域,具体涉及以一种树莓、沙棘、白刺多糖为原料制备所得的复合多糖组合物及其用途。
技术背景
随着现代生活节奏的加快,社会竞争的加剧,人们的体力、脑力、心理等疲劳长期积累,加之饮食不健康、环境辐射增多等因素,导致人体内自由基过多,使越来越多的人处于“亚健康”状态。自由基是指那些游离存在的,含有1个或1个以上不配对电子的分子、离子、原子或原子团,具有很大的能量。它们是机体正常代谢的产物,在体内有很强的氧化反应能力。正常情况下,机体的氧化与抗氧化处于一种动态平衡。平衡状态下,自由基作为机体的正常代谢产物,是机体不可缺少的活性物质。一旦平衡被打破,自由基便会产生强大的伤害作用,造成生物膜的脂质过氧化损伤、引起酶、氨基酸、蛋白质的氧化破坏,对内脏器官、免疫系统的形态功能产生影响,从而引起动脉粥样硬化、肿瘤、帕金森综合征等多种由于自由基水平升高而导致的疾病。因此,清除过剩的自由基对于维持机体健康,预防多种疾病都有极为重要的意义。
随着人们对浆果认识的逐渐加深和保健意识的不断提高,树莓、沙棘、白刺等营养丰富的浆果越来越受到欢迎。本发明拟利用树莓、沙棘、白刺浆果提取的多糖制备成组合物,对于进一步实现浆果的精深加工和高值化利用具有非常重要的意义。目前还未见将三者多糖联合使用的报道。
发明内容
本发明的目的在于提供一种基于树莓、沙棘和白刺三种浆果制备所得多糖的复合含片。本发明的另一目的在于提供该片剂的用途。
具体地,本发明提供了一种树莓、沙棘、白刺复合多糖组合物,其原料由如下重量配比的组份组成:
树莓多糖3~18份、沙棘多糖4~16份、白刺多糖3~20份。
进一步地,其原料由如下重量配比的组份组成:
树莓多糖5~15份、沙棘多糖5~15份、白刺多糖5~15份。
更进一步地,树莓多糖:沙棘多糖:白刺多糖=1:1:1。
多糖是天然有机化合物中的一类大分子物质,由单糖基通过糖苷键连接而成。越来越多的国内外研究证明某些活性多糖具有抗肿瘤、增强免疫力、抗氧化、降血糖、保肝等多种药理活性,具有很高的医疗和保健价值。沙棘多糖是由半乳糖、阿拉伯糖、甘露糖和葡萄糖组成的中性多糖,具有很强的自由基清除能力。此外,沙棘多糖在免疫调节、治疗肝炎、抑菌等方面有较高的临床利用价值。白刺多糖是由葡萄糖、鼠李糖、阿拉伯糖、半乳糖、半乳糖醛酸和甘露糖组成的多糖,具有较强的免疫调节和体外抗氧化活性。树莓多糖是由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖和半乳糖组成的中性多糖,具有清除自由基、抗肿瘤、降血糖、免疫调节、抗疲劳等多种生理活性。
其中,所述树莓多糖、沙棘多糖或白刺多糖是分别以树莓、沙棘或白刺为原料,经水提醇沉法制备得到。
进一步地,所述水提醇沉法中,醇沉时加乙醇至含醇量为70~85%v/v。加入的乙醇形式不限于95%乙醇、无水乙醇。
其中,所述组合物是以所述配比的原料为活性成分,加上食品、保健品或药品上常用的辅料或辅助性成分制备得到的制剂。实际使用过程中,可以根据市场需求,制备不同的剂型。
进一步地,所述制剂为经胃肠道吸收制剂。
本发明所述辅料,是指除活性成分以外包含在剂型中的物质,包括但不仅限于填充剂(稀释剂)、润滑剂(助流剂或抗粘着剂)、分散剂、湿润剂、粘合剂、调节剂、增溶剂、抗氧剂、抑菌剂、乳化剂、崩解剂等。粘合剂包含糖浆、阿拉伯胶、明胶、山梨醇、黄芪胶、纤维素及其衍生物(如微晶纤维素、羧甲基纤维素钠、乙基纤维素或羟丙甲基纤维素等)、明胶浆、糖浆、淀粉浆或聚乙烯吡咯烷酮等;填充剂包含乳糖、糖粉、糊精、淀粉及其衍生物、纤维素及其衍生物、无机钙盐(如硫酸钙、磷酸钙、磷酸氢钙、沉降碳酸钙等)、山梨醇或甘氨酸等;润滑剂包含微粉硅胶、硬脂酸镁、滑石粉、氢氧化铝、硼酸、氢化植物油、聚乙二醇等;崩解剂包含淀粉及其衍生物(如羧甲基淀粉钠、淀粉乙醇酸钠、预胶化淀粉、改良淀粉、羟丙基淀粉、玉米淀粉等)、聚乙烯吡咯烷酮或微晶纤维素等;湿润剂包含十二烷基硫酸钠、水或醇等;抗氧剂包含亚硫酸钠、亚硫酸氢钠、焦亚硫酸钠、二丁基苯酸等;抑菌剂包含0.5%苯酚、0.3%甲酚、0.5%三氯叔丁醇等;调节剂包含盐酸、枸橼酸、氢氧化钾(钠)、枸橼酸钠及缓冲剂(包括磷酸二氧钠和磷酸氢二钠)等;乳化剂包含聚山梨酯-80、脂肪酸山梨坦、普流罗尼克F-68,卵磷酯、豆磷脂等;增溶剂包含吐温-80、胆汁、甘油等。
所述辅助性成分,它具有一定生理活性,但该成分的加入不会改变上述提取物在疾病治疗过程中的主导地位,而仅仅发挥辅助功效,这些辅助功效仅仅是对该成分已知活性的利用,是医药领域惯用的辅助治疗方式。若将上述辅助性成分与本发明组合物配合使用,仍然应属于本发明保护的范围。
本发明还提供了上述组合物在制备抗氧化的药物、保健品或功能食品中的用途。
进一步地,所述药物、保健品或功能食品是清除ABTS自由基、超氧阴离子自由基或羟自由基的药物、保健品或功能食品。
优选地,所述药物、保健品或功能食品是清除ABTS自由基或超氧阴离子自由基的药物、保健品或功能食品。
本发明研究发现,树莓多糖、沙棘多糖和白刺多糖均具有良好的体外抗氧化活性,可显著清除羟自由基、超氧阴离子自由基及ABTS自由基。但是将三种多糖组合后,在与单用各多糖相同浓度下,组合物抗氧化活性明显增强(尤其是对ABTS自由基或超氧阴离子自由基),这说明三种多糖具有协同增效作用。

具体实施方式
本发明中,各多糖可以按照目前已有的技术进行制备,也可以通过购买市售产品获得。本发明中多糖参照现有技术制备,方法如下:分别取树莓、沙棘或唐古特白刺,经过水提或榨汁后,加入乙醇至70~85%v/v,沉淀制备得到。
实施例1本发明多糖组合物的制备

取上述原料,混合均匀,即得本发明组合物。亦可在此基础上,加入合适的辅料,制备相应的剂型,如片剂、胶囊剂、颗粒剂等。
体外抗氧化活性实验
(1)材料:树莓多糖,沙棘多糖、白刺多糖及三者以1:1:1w/w混合的多糖组合物。
(2)实验仪器及试剂:管式离心机(TGL-16G-A型,上海安亭科学仪器厂);可见紫外分光光度计(UV-722N,上海尼柯有限公司);恒温水浴锅(HD型,辽阳市恒温仪器厂)。ABTS购于上海晶纯生化科技股份有限公司(阿拉丁);过硫酸钾、磷酸二氢钠、磷酸氢二钠、氯化钠、三氯乙酸等试剂均购于山东禹王实业有限公司;超氧阴离子试剂盒、羟自由基试剂盒均购于南京建成生物科技有限公司。
(3)ABTS清除能力:将ABTS溶解(+H2O)至终浓度为7mmol/L,过硫酸钾配制成2.45mmol/L,将二者溶液混合,放置于黑暗,室温条件下搅拌16h;而后将ABTS混合溶液用PBS溶液(PH=7.0)稀释到其吸光值(于734nm处测)为0.70±0.02;树莓、沙棘、白刺三种多糖及三种多糖组合物分别用蒸馏水溶解后稀释至不同浓度(0.5mg/mL,1mg/mL,2mg/mL,4mg/mL,8mg/mL和16mg/mL);最后将上述不同浓度的样品加入到ABTS混合溶液中,其样品与ABTS混合溶液比例为1:20,混合后于室温下孵育6min,于734nm处测定吸光值。根据公式计算出清除率。ABTS清除率(%)=(1-样品吸光值/空白吸光值)×100%。结果见图1。
(4)超氧阴离子自由基清除能力:采用黄嘌呤/黄嘌呤氧化酶产生超氧阴离子自由基体系测定对超氧自由基的清除能力。将树莓、沙棘、白刺三种多糖及三种多糖组合物分别用蒸馏水溶解后稀释至不同浓度(1mg/mL,2mg/mL,4mg/mL,8mg/mL,16mg/mL)。各组依次加入75mmol/L磷酸盐缓冲液(pH7.8)1ml、样品0.2ml(对照组用蒸馏水代替)、0.1mol/L盐酸羟胺溶液0.1ml、75mmol/L黄嘌呤溶液0.1ml和0.037U/ml黄嘌呤氧化酶溶液0.1ml,混合均匀后27℃孵育30min。再加入2ml显色剂(0.4g/L甲萘胺+4g/L对氨基苯磺酸),混匀后静置5min,530nm下侧吸光值。根据公式计算出清除率。超氧阴离子自由基清除率(%)=1-样品吸光值/空白吸光值×100%。结果见图2。
(5)羟自由基清除能力:采用Fenton反应原理产生羟自由基,用番红染料显色发来测定对羟自由基的清除能力。将树莓、沙棘、白刺三种多糖及三种多糖组合物分别用蒸馏水溶解后稀释至不同浓度(1mg/mL,2mg/mL,4mg/mL,8mg/mL,16mg/mL)。各组依次加入0.15mol/L的Tris-HCl缓冲液(pH7.4)1ml、0.04mg/ml番红溶液1ml、蒸馏水1ml、0.945mmol/L的EDTANa-Fe2+溶液(pH7.4)0.5ml、样品0.5ml和1%H2O2溶液0.5ml,于37℃孵育30min,对照组用蒸馏水代替,空白组EDTANa-Fe2+溶液和样品用蒸馏水代替。在520nm下检测吸光值。根据公式计算出清除率。羟自由基清除率(%)=1-样品吸光值/空白吸光值×100%。结果见图3。

1.一种树莓、沙棘、白刺复合多糖组合物,其特征在于:其原料由如下重量配比的组份组成:
树莓多糖3~18份、沙棘多糖4~16份、白刺多糖3~20份。
2.根据权利要求1所述的树莓、沙棘、白刺复合多糖组合物,其特征在于:其原料由如下重量配比的组份组成:
树莓多糖5~15份、沙棘多糖5~15份、白刺多糖5~15份。
3.根据权利要求1或2所述的树莓、沙棘、白刺复合多糖组合物,其特征在于:树莓多糖:沙棘多糖:白刺多糖=1:1:1。
4.根据权利要求1~3任意一项所述的组合物,其特征在于:所述树莓多糖、沙棘多糖或白刺多糖是分别以树莓、沙棘或白刺为原料,经水提醇沉法制备得到。
5.根据权利要求4所述的组合物,其特征在于:所述水提醇沉法中,醇沉时加乙醇至含醇量为70~85%v/v。
6.根据权利要求1~3任意一项所述的组合物,其特征在于:所述组合物是以所述配比的原料为活性成分,加上食品、保健品或药品上常用的辅料或辅助性成分制备得到的制剂。
7.根据权利要求6所述的组合物,其特征在于:所述制剂为经胃肠道吸收制剂。
8.权利要求1~7所述组合物在制备抗氧化的药物、保健品或功能食品中的用途。
9.根据权利要求8所述的用途,其特征在于:所述药物、保健品或功能食品是清除ABTS自由基、超氧阴离子自由基或羟自由基的药物、保健品或功能食品。
