Abstract
本发明涉及保健食品领域,提供了一种提高免疫力的灵芝全活性组分组合微囊的制备方法,包括如下步骤:(1)制备灵芝子实体提取物;(2)制备灵芝孢子油;(3)制备霍山石斛超微粉;(4)以霍山石斛超微粉、食用胶、蛋白粉为壁材,灵芝孢子油为心材,加水和灵芝子实体提取物,高速匀浆乳化,形成水包油型微胶囊乳液,再喷雾干燥。本发明的优点在于:分别从灵芝子实体和灵芝孢子提取其全部活性成分并浓缩;合理配比后,将其配合霍山石斛超微粉等其他辅料加工成抗氧化的微囊颗粒,一方面可有效解决灵芝活性成分不能全效利用的难题,另一方面也有效解决了提取方法复杂,产品易氧化、不耐保存的难题,同时该产品可有效提高人体免疫力、降低血糖。

技术领域
本发明涉及保健食品领域,尤其涉及一种提高免疫力的灵芝全活性组分组合微囊的制备方法。
背景技术
灵芝又称林中灵、琼珍(学名:Ganoderma Lucidum Karst),是多孔菌科真菌灵芝的子实体。具有补气安神、止咳平喘、延年益寿的功效。用于眩晕不眠、心悸气短、神经衰弱、虚劳咳喘。主要分布于中国浙江、黑龙江、吉林、安徽、江西、湖南、贵州、广东、福建等地。
灵芝药用在我国已有2000多年的历史,被历代医药家视为滋补强壮、扶正固本的神奇珍品。2000年,已知灵芝属(Ganoderma)真菌约100余种,分布最广的为赤芝(G.lu-cidum),其次为紫芝(G.japonicum),还有树舌(G.applana-tum)、松杉灵芝(G.tsugae)和簿树芝(G.capense)等均供药用。经过大量临床研究,灵芝对神经衰弱、高脂血症、冠心病心绞痛、心律失常、克山病、高原不适症、肝炎、出血热、消化不良、气管炎等各有不同程度的疗效。
药理研究证明,灵芝有多方面的生物活性,其主含氨基酸、多肽、蛋白质、真菌溶菌酶,以及糖类(还原糖和多糖)、麦角甾醇、三萜类、香豆精甙、挥发油、硬脂酸、苯甲酸、生物碱、维生素B2及C等;孢子还含甘露醇、海藻糖)。这些活性成分分别存在于灵芝子实体和灵芝孢子中。
然而,目前的天然灵芝虽具有较强的保健治疗作用,可有效提高人体的免疫力,但消费者无法有效利用其全部活性成分。市场上的产品或者以破壁灵芝孢子粉为主成份(主要剂型是颗粒和胶囊),或者以灵芝子实体提取物为主成份(主要剂型是软胶囊),二者的混合组分产品成分复杂,没有合适的剂型来包裹产品,以防止其氧化,延长其保存期,所以无法真正高效利用天然灵芝的全部活性成分。
霍山石斛,又称米斛,是石斛中的极品,是中国国家濒临灭绝的珍稀药材,其干燥茎(霍枫斗)和鲜斛均可入药。《本草纲目》记载:霍斛性甘平无毒。主治伤中,除痹下气,补五脏虚劳赢瘦,强阴益精,久服厚肠胃,补内绝不足,平胃气,长肌肉,逐皮肤邪热痱气,脚膝疼冷痹弱。定志除惊,轻身延年,益气除热,治男子腰脚软弱,健阳。逐皮肤风痹,骨中久冷,补肾益力,壮筋骨,暖水脏,益智清气,治发热自汗,痈疽排脓内塞。
霍斛富含多糖、氨基酸和石斛碱、石斛胺碱等十多种生物碱,能有效提高机体免疫功能;对人体眼、咽、肺。胃、肠、肾等器官和血液、心血管等疾病有特殊疗效;能抗白内障,延缓衰老,抗突变,抗肿瘤。霍斛全草入药,加工成超微粉能有效提高其利用率和吸收率,提高药效。
据此,目前急需一种既可高效利用天然灵芝的全部活性成分,又添加有霍山石斛作为另一有效成分,同时还具备提取方法简单,产品不易氧化、耐保存等优点的灵芝全活性组分组合微囊的制备方法。
发明内容
本发明所要解决的技术问题在于现有灵芝产品无法高效利用天然灵芝全部活性成分,且提取方法复杂,产品易氧化、不耐保存,同时还缺少霍山石斛作为另一有效成分。
本发明采用以下技术方案解决上述技术问题:
一种提高免疫力的灵芝全活性组分组合微囊的制备方法,包括如下步骤:
(1)灵芝子实体提取物的制备
采收生长成熟的灵芝,经流水冲洗后破碎成小块,105-115℃钝化酶处理4-6min,65-75℃烘干至恒重,再依次用2-4倍量的1-2%柠檬酸+90-95%乙醇溶液、1-2%柠檬酸+70-75%乙醇溶液、1-2%柠檬酸+45-50%乙醇溶液、1-2%柠檬酸+20-25%乙醇溶液和1-2%柠檬酸+去离子水溶液,在65-75℃,分别回流提取2-4h;
不同抽提液抽虑后,收集在一起,负压下65-75℃蒸发浓缩成膏状,即灵芝子实体提取物;
(2)灵芝孢子油的制备
收集成熟灵芝孢子粉,过180-220目筛去杂质,先用3-5%乙酸溶液,室温搅动浸泡6-12h,水洗过滤后,再用3-5%氢氧化钠溶液,室温搅动浸泡6-12h,水洗过滤后,负压下55-65℃烘干,得壁脆化的孢子粉;
将壁脆化的孢子粉于10℃以下的温度环境中进行破壁,并收集相应破壁孢子粉,再经过超临界二氧化碳萃取,获得灵芝孢子油脂,即灵芝孢子油提取物;
(3)霍山石斛超微粉的制备
采收霍山石斛鲜条,去叶、清洗、晾干,105-115℃钝化酶处理4-6min,65-75℃烘干至恒重;接着破碎、过95-105目筛,再进行超微粉碎,过195-205目筛后收集,得霍山石斛超微粉;
(4)灵芝全活性组分组合微囊的制备
以霍山石斛超微粉、食用胶、蛋白粉为壁材,以灵芝孢子油为心材,加水和灵芝子实体提取物,经高速匀浆乳化,形成水包油型的微胶囊乳液,再经喷雾干燥加工成微胶囊颗粒。
作为本发明的优选方式之一,所述步骤(1)中灵芝经流水冲洗后破碎成1mm以下大小的小块。
作为本发明的优选方式之一,所述步骤(1)中配合布氏漏斗对不同抽提液进行抽虑。
作为本发明的优选方式之一,所述步骤(1)中膏状的灵芝子实体提取物的含水量为20%-30%。
作为本发明的优选方式之一,所述步骤(1)中膏状的灵芝子实体提取物包括氨基酸、多肽、蛋白质、真菌溶菌酶,以及糖类、麦角甾醇、三萜类、香豆精甙、挥发油、硬脂酸、苯甲酸、生物碱、维生素B2和维生素C。
作为本发明的优选方式之一,所述糖类具体包括还原糖和多糖。
作为本发明的优选方式之一,所述步骤(2)中乙酸和氢氧化钠均为食品级。
作为本发明的优选方式之一,所述步骤(2)中通过脉冲式挤压破壁机对壁脆化的孢子粉进行破壁,破壁率≥99.5%。
作为本发明的优选方式之一,所述步骤(2)中超临界二氧化碳萃取的萃取条件为:萃取压力14-15Mpa,萃取温度34-36℃,萃取时间3.0-4.0h,分离压力4.0-5.0Mpa,分离温度30-36℃,CO2流量为24-26L/h;在此条件下,灵芝孢子油的萃取率为30-34%,灵芝孢子油中的三萜含量为18.0-18.5%。
作为本发明的优选方式之一,所述步骤(2)中灵芝孢子油脂包括灵芝酸、孢子酸、赤芝孢子内脂等四环、五环三萜和不饱和脂肪酸、灵芝多糖。
作为本发明的优选方式之一,所述步骤(3)中选取两年生的霍山石斛鲜条进行采收,去叶、清洗、晾干、钝化酶处理、烘干,采用刀片式粉碎机破碎;过筛后,再采用脉冲振动磨进行超微粉碎。
作为本发明的优选方式之一,所述步骤(4)中灵芝全活性组分组合微囊的具体制备工艺如下:
a.取霍山石斛超微粉、灵芝子实体提取物、蛋白粉和水加入容器,室温下,搅拌均匀;同时,取食用胶和水加入另一容器,室温下,搅拌溶解;最后,将两者相混合,搅拌均匀;
b.冷却上述混合溶液至8-12℃,加入预冷保存的灵芝孢子油,在高速匀浆设备中,以10000-12000r/min,匀浆20min以上;
c.将匀质后的乳液连接到高温高压喷雾干燥装置上,在进风温度为100-120℃、出风温度为60-70℃条件下进行喷雾干燥,制成微胶囊颗粒。
作为本发明的优选方式之一,所述步骤(4)中灵芝全活性组分组合微囊的制备原料的质量百分比如下:食用胶1-4%、蛋白粉4-8%、霍山石斛超微粉5-6%、灵芝孢子油5-8%、灵芝子实体提取物5-8%,其余为饮用水。
作为本发明的优选方式之一,所述步骤(4)中食用胶具体为黄原胶、结冷胶、阿拉伯胶,卡拉胶、瓜尔豆胶、琼脂或明胶。
作为本发明的优选方式之一,所述步骤(4)中蛋白粉具体为无致敏性的植物性蛋白粉。
作为本发明的优选方式之一,所述步骤(4)中蛋白粉为大米蛋白粉或小麦蛋白粉。
本发明相比现有技术的优点在于:
(1)本发明通过科学的工艺分别从灵芝子实体和灵芝孢子提取其全部活性成分并浓缩;经合理配比后,将其配合霍山石斛超微粉等其他辅料加工成抗氧化的微囊颗粒,一方面可有效解决灵芝活性成分不能全效利用的难题,另一方面也有效解决了提取成分复杂、易氧化、不耐保存的难题,同时还解决了目前灵芝产品的有效成分单一,缺少霍山石斛作为另一有效成分的难题;
(2)本发明提供的制备工艺简单可行,制备的灵芝全活性组分加霍山石斛超微粉组合微囊是一种粉末,不易粘结,有很好的流动性,能够将其中的孢子油与外界空气相隔离,从而有效防止孢子油和其他活性成分的氧化,大大延长产品中活性成分的保存时间;微囊采用食用原料作为壁材,可以直接食用,也方便包装成其他形式,如胶囊装、袋装、瓶装等,也可方便的添加到其他保健品中增强保健效果;
(3)本发明采用的灵芝补气安神、止咳平喘,性平,味甘,归心经、肺经、肝经、肾经,属补虚药下属分类的补气药;采用的霍山石斛补五脏虚劳赢瘦,强阴益精,性平(炮制后),味甘,归胃经、肾经,属补虚药下属分类的补阴药;两者配伍,“气、阴”同补,能显著提升药效和弥补单味药的不足;本发明产品具有提高人体免疫力、降低血糖等保健功能,为享受大自然赋予人类的保健佳品提供了更佳的选择。

具体实施方式
下面对本发明的实施例作详细说明,本实施例在以本发明技术方案为前提下进行实施,给出了详细的实施方式和具体的操作过程,但本发明的保护范围不限于下述的实施例。
实施例1
如图1所示,本实施例的一种提高免疫力的灵芝全活性组分组合微囊的制备方法,包括如下步骤:
(1)灵芝子实体提取物的制备
采收生长成熟的灵芝,经流水冲洗后破碎成0.3mm大小的小块,105℃钝化酶处理4min,65℃烘干至恒重,再依次用2倍量的1%柠檬酸+90%乙醇溶液、1%柠檬酸+70%乙醇溶液、1%柠檬酸+45%乙醇溶液、1%柠檬酸+20%乙醇溶液和1%柠檬酸+去离子水溶液,在65℃,分别回流提取2h;
不同抽提液经布氏漏斗抽虑后,收集在一起,负压下65℃蒸发浓缩成膏状(含水量为20%),即灵芝子实体提取物,主要成分包括氨基酸、多肽、蛋白质、真菌溶菌酶,以及糖类(包括还原糖和多糖)、麦角甾醇、三萜类、香豆精甙、挥发油、硬脂酸、苯甲酸、生物碱、维生素B2和维生素C;
(2)灵芝孢子油的制备
收集成熟灵芝孢子粉,过180目筛去杂质,先用3%乙酸(食品级)溶液,室温搅动浸泡6h,水洗过滤后,再用3%氢氧化钠(食品级)溶液,室温搅动浸泡6h,水洗过滤后,负压下55℃烘干,得壁脆化的孢子粉;
将壁脆化的孢子粉于10℃以下的温度环境中通过脉冲式挤压破壁机进行破壁(破壁率≥99.5%),并收集相应破壁孢子粉,再经过超临界二氧化碳萃取(萃取条件:萃取压力14Mpa,萃取温度34℃,萃取时间3.0h,分离压力4.0Mpa,分离温度30℃,CO2流量为24L/h;在此条件下,灵芝孢子油的萃取率为30%,灵芝孢子油中的三萜含量为18.0%),获得灵芝孢子油脂,即灵芝孢子油提取物,包括灵芝酸、孢子酸、赤芝孢子内脂等四环、五环三萜和不饱和脂肪酸、灵芝多糖;
(3)霍山石斛超微粉的制备
采收两年生的霍山石斛鲜条,去叶、清洗、晾干,105℃钝化酶处理4min,65℃烘干至恒重;接着破碎、过95目筛,再采用脉冲振动磨进行超微粉碎,过195目筛后收集,得霍山石斛超微粉;
(4)灵芝全活性组分组合微囊的制备
以霍山石斛超微粉、食用胶、蛋白粉(大米蛋白粉、小麦蛋白粉或其他无致敏性的植物性蛋白粉)为壁材,以灵芝孢子油为心材,加水和灵芝子实体提取物,经高速匀浆乳化,形成水包油型的微胶囊乳液,再经喷雾干燥加工成微胶囊颗粒;具体工艺如下:
a.取霍山石斛超微粉、灵芝子实体提取物、蛋白粉和水加入容器,室温下,搅拌均匀;同时,取食用胶(黄原胶、结冷胶、阿拉伯胶,卡拉胶、瓜尔豆胶、琼脂或明胶)和水加入另一容器,室温下,搅拌溶解;最后,将两者相混合,搅拌均匀;
b.冷却上述混合溶液至8℃,加入预冷保存的灵芝孢子油,在高速匀浆设备中,以10000r/min,匀浆20min以上;
c.将匀质后的乳液连接到高温高压喷雾干燥装置上,在进风温度为100℃、出风温度为60℃条件下进行喷雾干燥,制成微胶囊颗粒;
上述各原料配方的质量百分比如下:食用胶1%、蛋白粉4%、霍山石斛超微粉5%、灵芝孢子油5%、灵芝子实体提取物5%,其余为饮用水。
实施例2
如图1所示,本实施例的一种提高免疫力的灵芝全活性组分组合微囊的制备方法,包括如下步骤:
(1)灵芝子实体提取物的制备
采收生长成熟的灵芝,经流水冲洗后破碎成1mm大小的小块,115℃钝化酶处理6min,75℃烘干至恒重,再依次用4倍量的2%柠檬酸+95%乙醇溶液、2%柠檬酸+75%乙醇溶液、2%柠檬酸+50%乙醇溶液、2%柠檬酸+25%乙醇溶液和2%柠檬酸+去离子水溶液,在75℃,分别回流提取4h;
不同抽提液经布氏漏斗抽虑后,收集在一起,负压下75℃蒸发浓缩成膏状(含水量为30%),即灵芝子实体提取物,主要成分包括氨基酸、多肽、蛋白质、真菌溶菌酶,以及糖类(包括还原糖和多糖)、麦角甾醇、三萜类、香豆精甙、挥发油、硬脂酸、苯甲酸、生物碱、维生素B2和维生素C;
(2)灵芝孢子油的制备
收集成熟灵芝孢子粉,过220目筛去杂质,先用5%乙酸(食品级)溶液,室温搅动浸泡12h,水洗过滤后,再用5%氢氧化钠(食品级)溶液,室温搅动浸泡12h,水洗过滤后,负压下65℃烘干,得壁脆化的孢子粉;
将壁脆化的孢子粉于10℃以下的温度环境中通过脉冲式挤压破壁机进行破壁(破壁率≥99.5%),并收集相应破壁孢子粉,再经过超临界二氧化碳萃取(萃取条件:萃取压力15Mpa,萃取温度36℃,萃取时间4.0h,分离压力5.0Mpa,分离温度36℃,CO2流量为26L/h;在此条件下,灵芝孢子油的萃取率为34%,灵芝孢子油中的三萜含量为18.5%),获得灵芝孢子油脂,即灵芝孢子油提取物,包括灵芝酸、孢子酸、赤芝孢子内脂等四环、五环三萜和不饱和脂肪酸、灵芝多糖;
(3)霍山石斛超微粉的制备
采收两年生的霍山石斛鲜条,去叶、清洗、晾干,115℃钝化酶处理6min,75℃烘干至恒重;接着破碎、过105目筛,再采用脉冲振动磨进行超微粉碎,过205目筛后收集,得霍山石斛超微粉;
(4)灵芝全活性组分组合微囊的制备
以霍山石斛超微粉、食用胶、蛋白粉(大米蛋白粉、小麦蛋白粉或其他无致敏性的植物性蛋白粉)为壁材,以灵芝孢子油为心材,加水和灵芝子实体提取物,经高速匀浆乳化,形成水包油型的微胶囊乳液,再经喷雾干燥加工成微胶囊颗粒;具体工艺如下:
a.取霍山石斛超微粉、灵芝子实体提取物、蛋白粉和水加入容器,室温下,搅拌均匀;同时,取食用胶(黄原胶、结冷胶、阿拉伯胶,卡拉胶、瓜尔豆胶、琼脂或明胶)和水加入另一容器,室温下,搅拌溶解;最后,将两者相混合,搅拌均匀;
b.冷却上述混合溶液至12℃,加入预冷保存的灵芝孢子油,在高速匀浆设备中,以12000r/min,匀浆20min以上;
c.将匀质后的乳液连接到高温高压喷雾干燥装置上,在进风温度为120℃、出风温度为70℃条件下进行喷雾干燥,制成微胶囊颗粒;
上述各原料配方的质量百分比如下:食用胶4%、蛋白粉8%、霍山石斛超微粉6%、灵芝孢子油8%、灵芝子实体提取物8%,其余为饮用水。
实施例3
如图1所示,本实施例的一种提高免疫力的灵芝全活性组分组合微囊的制备方法,包括如下步骤:
(1)灵芝子实体提取物的制备
采收生长成熟的灵芝,经流水冲洗后破碎成0.5mm大小的小块,110℃钝化酶处理5min,70℃烘干至恒重,再依次用3倍量的1.5%柠檬酸+93%乙醇溶液、1.5%柠檬酸+73%乙醇溶液、1.5%柠檬酸+48%乙醇溶液、1.5%柠檬酸+23%乙醇溶液和1.5%柠檬酸+去离子水溶液,在70℃,分别回流提取3h;
不同抽提液经布氏漏斗抽虑后,收集在一起,负压下70℃蒸发浓缩成膏状(含水量为25%),即灵芝子实体提取物,主要成分包括氨基酸、多肽、蛋白质、真菌溶菌酶,以及糖类(包括还原糖和多糖)、麦角甾醇、三萜类、香豆精甙、挥发油、硬脂酸、苯甲酸、生物碱、维生素B2和维生素C;
(2)灵芝孢子油的制备
收集成熟灵芝孢子粉,过200目筛去杂质,先用4%乙酸(食品级)溶液,室温搅动浸泡9h,水洗过滤后,再用4%氢氧化钠(食品级)溶液,室温搅动浸泡9h,水洗过滤后,负压下60℃烘干,得壁脆化的孢子粉;
将壁脆化的孢子粉于10℃以下的温度环境中通过脉冲式挤压破壁机进行破壁(破壁率≥99.5%),并收集相应破壁孢子粉,再经过超临界二氧化碳萃取(萃取条件:萃取压力14.5Mpa,萃取温度35℃,萃取时间3.5h,分离压力4.5Mpa,分离温度33℃,CO2流量为25L/h;在此条件下,灵芝孢子油的萃取率为32%,灵芝孢子油中的三萜含量为18.2%),获得灵芝孢子油脂,即灵芝孢子油提取物,包括灵芝酸、孢子酸、赤芝孢子内脂等四环、五环三萜和不饱和脂肪酸、灵芝多糖;
(3)霍山石斛超微粉的制备
采收两年生的霍山石斛鲜条,去叶、清洗、晾干,110℃钝化酶处理5min,70℃烘干至恒重;接着破碎、过100目筛,再采用脉冲振动磨进行超微粉碎,过200目筛后收集,得霍山石斛超微粉;
(4)灵芝全活性组分组合微囊的制备
以霍山石斛超微粉、食用胶、蛋白粉为壁材,以灵芝孢子油为心材,加水和灵芝子实体提取物,经高速匀浆乳化,形成水包油型的微胶囊乳液,再经喷雾干燥加工成微胶囊颗粒;具体工艺如下:
a.取霍山石斛超微粉、灵芝子实体提取物、蛋白粉(大米蛋白粉、小麦蛋白粉或其他无致敏性的植物性蛋白粉)和水加入容器,室温下,搅拌均匀;同时,取食用胶(黄原胶、结冷胶、阿拉伯胶,卡拉胶、瓜尔豆胶、琼脂或明胶)和水加入另一容器,室温下,搅拌溶解;最后,将两者相混合,搅拌均匀;
b.冷却上述混合溶液至10℃,加入预冷保存的灵芝孢子油,在高速匀浆设备中,以11000r/min,匀浆20min以上;
c.将匀质后的乳液连接到高温高压喷雾干燥装置上,在进风温度为110℃、出风温度为65℃条件下进行喷雾干燥,制成微胶囊颗粒;
上述各原料配方的质量百分比如下:食用胶2%、蛋白粉6%、霍山石斛超微粉5.5%、灵芝孢子油7%、灵芝子实体提取物7%,其余为饮用水。
以上所述仅为本发明的较佳实施例而已,并不用以限制本发明,凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,均应包含在本发明的保护范围之内。
Claims (9)
1.一种提高免疫力的灵芝全活性组分组合微囊的制备方法,其特征在于,包括如下步骤:
(1)灵芝子实体提取物的制备
采收生长成熟的灵芝,经流水冲洗后破碎成小块,105-115℃钝化酶处理4-6min,65-75℃烘干至恒重,再依次用2-4倍量的1-2%柠檬酸+90-95%乙醇溶液、1-2%柠檬酸+70-75%乙醇溶液、1-2%柠檬酸+45-50%乙醇溶液、1-2%柠檬酸+20-25%乙醇溶液和1-2%柠檬酸+去离子水溶液,在65-75℃,分别回流提取2-4h;
不同抽提液抽滤 后,收集在一起,负压下65-75℃蒸发浓缩成膏状,即灵芝子实体提取物;
所述灵芝子实体提取物包括氨基酸、多肽、蛋白质、真菌溶菌酶,以及糖类、麦角甾醇、三萜类、香豆精甙、挥发油、硬脂酸、苯甲酸、生物碱、维生素B2和维生素C;
(2)灵芝孢子油的制备
收集成熟灵芝孢子粉,过180-220目筛去杂质,先用3-5%乙酸溶液,室温搅动浸泡6-12h,水洗过滤后,再用3-5%氢氧化钠溶液,室温搅动浸泡6-12h,水洗过滤后,负压下55-65℃烘干,获得壁脆化的孢子粉;
将壁脆化的孢子粉于10℃以下的温度环境中通过脉冲式挤压破壁机对壁脆化的孢子粉进行破壁,破壁率≥99.5%,并收集相应破壁孢子粉,再经过超临界二氧化碳萃取,获得灵芝孢子油脂,即灵芝孢子油提取物;
(3)霍山石斛超微粉的制备
采收霍山石斛鲜条,去叶、清洗、晾干,105-115℃钝化酶处理4-6min,65-75℃烘干至恒重;接着破碎、过95-105目筛,再进行超微粉碎,过195-205目筛后收集,得霍山石斛超微粉;
(4)灵芝全活性组分组合微囊的制备
以霍山石斛超微粉、食用胶、蛋白粉为壁材,以灵芝孢子油为心材,加水和灵芝子实体提取物,经高速匀浆乳化,形成水包油型的微胶囊乳液,再经喷雾干燥加工成微胶囊颗粒;
蛋白粉具体为无致敏性的植物性蛋白粉。
2.根据权利要求1所述的提高免疫力的灵芝全活性组分组合微囊的制备方法,其特征在于,所述步骤(1)中灵芝经流水冲洗后破碎成1mm以下大小的小块。
3.根据权利要求1所述的提高免疫力的灵芝全活性组分组合微囊的制备方法,其特征在于,所述步骤(1)中配合布氏漏斗对不同抽提液进行抽滤 。
4.根据权利要求1所述的提高免疫力的灵芝全活性组分组合微囊的制备方法,其特征在于,所述步骤(1)中膏状的灵芝子实体提取物的含水量为20%-30%。
5.根据权利要求1所述的提高免疫力的灵芝全活性组分组合微囊的制备方法,其特征在于,所述步骤(2)中乙酸和氢氧化钠均为食品级。
6.根据权利要求1所述的提高免疫力的灵芝全活性组分组合微囊的制备方法,其特征在于,所述步骤(2)中超临界二氧化碳萃取的萃取条件为:萃取压力14-15Mpa,萃取温度34-36℃,萃取时间3.0-4.0h,分离压力4.0-5.0Mpa,分离温度30-36℃,CO2流量为24-26L/h;在此条件下,灵芝孢子油的萃取率为30-34%,灵芝孢子油中的三萜含量为18.0-18.5%。
7.根据权利要求1所述的提高免疫力的灵芝全活性组分组合微囊的制备方法,其特征在于,所述步骤(3)中选取两年生的霍山石斛鲜条进行采收,去叶、清洗、晾干、钝化酶处理、烘干,采用刀片式粉碎机破碎;过筛后,再采用脉冲振动磨进行超微粉碎。
8.根据权利要求1所述的提高免疫力的灵芝全活性组分组合微囊的制备方法,其特征在于,所述步骤(4)中灵芝全活性组分组合微囊的具体制备工艺如下:
a.取霍山石斛超微粉、灵芝子实体提取物、蛋白粉和水加入容器,室温下,搅拌均匀;同时,取食用胶和水加入另一容器,室温下,搅拌溶解;最后,将两者相混合,搅拌均匀;
b.冷却上述混合溶液至8-12℃,加入预冷保存的灵芝孢子油,在高速匀浆设备中,以10000-12000r/min,匀浆20min以上;
c.将匀质后的乳液连接到高温高压喷雾干燥装置上,在进风温度为100-120℃、出风温度为60-70℃条件下进行喷雾干燥,制成微胶囊颗粒。
9.根据权利要求1所述的提高免疫力的灵芝全活性组分组合微囊的制备方法,其特征在于,所述步骤(4)中灵芝全活性组分组合微囊的制备原料的质量百分比如下:食用胶1-4%、蛋白粉4-8%、霍山石斛超微粉5-6%、灵芝孢子油5-8%、灵芝子实体提取物5-8%,其余为饮用水。
