Abstract
本发明提供一种新的具有抗真菌活性的组合物,不依赖已有的具有抗念珠菌活性的组合物或这些组合物的组合。具体地说,本发明是包含将溶菌酶与壳聚糖结合得到的复合物的、具有抗真菌活性的组合物。按照本发明,能应用于皮肤和粘膜的念珠菌病,特别是患者数众多的口腔念珠菌病和阴道念珠菌病,能实现所述疾病导致的症状的改善、疾病的治愈、和感染的预防。此外,溶菌酶作为安全性高的天然食品添加剂被广泛利用,使用所述溶菌酶与多糖类的复合物的具有抗真菌活性的组合物,不仅能让利用的患者安心,还能减轻患者的负担。本发明使用作为安全性高的天然食品添加剂溶菌酶和多糖类的复合物,无需考虑所述风险,患者能安心使用。

技术领域
本发明涉及新的具有抗真菌活性的组合物。
背景技术
在HIV和癌症的治疗以及器官移植中面临的问题是微生物导致的感染病。针对细菌的感染病的抗生物质的开发已经得到确立。尽管也存在出现了MRSA和VRE等耐药性菌的问题,但是比其更严重的是真菌导致的感染病。特别是,念珠菌·白色念珠菌导致的念珠菌病和曲霉菌属菌导致的曲霉病,在移植手术中成为很大的障碍。
酵母和丝状菌是真核生物,相对于作为原核生物的细菌,被称为真菌。某种真菌对人和动物显示致病性,成为真菌感染病的致病菌。所述真菌的致病性一般较弱,但是对于处于抵抗力降低状态的患者来说,仍然会导致危重的症状。
真菌导致的各种疾病,由于对人和动物的健康带来非常大的影响,所以期待着开发对真菌导致的各种疾病的治疗有用的新的药物。此外,反映当前的住宅情况的结露等导致的丝状菌向住宅的侵入,给人带来过敏等症状,对人和动物的健康造成不好的影响。作为所述的有效对策,期待新的抗真菌药的开发。
在此,作为阻碍念珠菌菌丝相生长的药物,公开有专利文献1和非专利文献1。
在专利文献1中,公开了针对体外(in vitro)的念珠菌菌丝相生长,将癸酸(decanoic acid)与香叶醇、丁香酚或柠檬醛中的任意一种并用时,确认到了抗念珠菌活性的协同效果。此外,专利文献1公开了在小鼠口腔念珠菌病模型中,癸酸和姜精油的并用对舌头症状的改善效果最高等。
此外,非专利文献1中公开了针对体外的念珠菌菌丝相生长,并用癸酸和萜品烯-4-醇时,确认到了抗念珠菌活性的协同效果。此外,非专利文献1中公开了在小鼠口腔念珠菌病模型中,通过癸酸和萜品烯-4-醇的并用显示出包含活菌数降低的可靠的治疗效果。
可是,在所述的专利文献1和非专利文献1中,仅仅显示了作为具有抗念珠菌活性的已知的癸酸等中链脂肪酸与作为同样具有抗念珠菌活性的已知的萜品烯-4-醇等萜烯醇的并用效果。
另一方面,如专利文献2所示,着眼于溶菌酶与壳聚糖(30kDa)的复合物具有抗菌性,可以考虑将该复合物应用于食品和化妆品。所述复合物的安全性极高,将比产品设计等更看重的稳定性赋予溶菌酶,进一步扩展了抗菌谱。
可是,仅显示了对大肠杆菌K12具有抗细菌活性,而未确认到针对念珠菌属和曲霉菌属等真菌的抗菌活性。

本发明要解决的技术问题
另一方面,本发明人为了不依赖已有的具有抗念珠菌活性的组合物或这些组合物的组合,提供新的具有抗真菌活性的组合物,进行了专心研究。其中,本发明人发现了,将溶菌酶与壳聚糖结合得到的复合物,对念珠菌等真菌显示了抗真菌活性。
解决技术问题的技术方案
即,本发明的具有抗真菌活性的组合物,其包含将溶菌酶与壳聚糖结合得到的复合物。所述抗真菌活性包含念珠菌增殖抑制作用或马拉色菌增殖抑制作用。即,所述组合物包含念珠菌增殖抑制组合物、马拉色菌增殖抑制组合物或毛癣菌增殖抑制组合物。所述的具有抗真菌活性的组合物的效果将在后面进行描述。
此外,优选的是,具有抗真菌活性的组合物还包含萜烯醇或脂肪酸。
作为萜烯醇,可以考虑香叶醇、薄荷醇、萜品烯-4-醇。
作为脂肪酸,优选的是碳原子数8~12的脂肪酸,具体地说,可以考虑辛酸(碳原子数8)、癸酸(碳原子数10)、月桂酸(碳原子数12)等。
发明效果
按照本发明,能够取得抑制皮肤和粘膜的念珠菌感染的效果。因此,作为本发明的效果,能够应用于皮肤和粘膜的念珠菌病,特别是患者数众多的口腔念珠菌病和阴道念珠菌病,并能够实现所述疾病导致的症状的改善、疾病的治愈、感染的预防。此外,溶菌酶作为安全性高的天然的食品添加剂被广泛利用,使用所述溶菌酶和多糖类的复合物的、具有抗真菌活性的组合物,能够使利用的患者安心,并且能够减轻患者的负担。
此外,口腔念珠菌病和阴道念珠菌病容易复发,而且由于副作用和出现耐药菌的风险,抗真菌药的应用存在极限。本发明使用作为安全性高的天然食品添加剂的溶菌酶与多糖类的复合物,无需考虑这样的风险,患者能够安心利用。
此外,通过将所述具有抗真菌活性的组合物用作马拉色菌增殖抑制组合物,能够预防并改善人和动物的皮肤产生的脂溢性皮炎。此外,通过将所述具有抗真菌活性的组合物用作毛癣菌增殖抑制组合物,能够预防并改善人的皮肤产生的足癣等毛癣菌感染病。

具体实施方式
以下具体说明本发明。
1.第一实施方式
第一实施方式的具有抗真菌活性的组合物,包含将溶菌酶与壳聚糖(水溶性物质,分子量为25000Da以下)结合得到的复合物。另外,在本实施方式中,使用分子量14000Da的壳聚糖。此外,可以使用鸡来源和人来源的溶菌酶。
本发明的溶菌酶-壳聚糖复合物,可以通过利用美拉德反应将溶菌酶与壳聚糖结合来制造。通过利用美拉德反应将溶菌酶与壳聚糖结合,由于溶菌酶中的抗原结构的几乎全部或全部被掩盖,所以即使摄取了溶菌酶-壳聚糖复合物,也不易引起过敏。
具体的制造方法如下所述。
将溶菌酶/壳聚糖的质量比更优选成为60/40~40/60的量的溶菌酶和壳聚糖混合溶解在水中,并以使水溶液中的溶菌酶和壳聚糖的合计含量成为5~30质量%的方式进行制备。通过冻结干燥,将得到的水溶液粉末化。将得到的粉末在温度50~80℃、更优选55~65℃,相对湿度50~80%、更优选60~70%的条件下,进行2~20天、更优选7~14天的美拉德反应,由此可以制造本发明的溶菌酶-壳聚糖复合物。
关于本发明的溶菌酶-壳聚糖复合物的生成,可以通过把利用SDS(Sodiumdodecyl sulfate-polyacrylamide)电泳得到的板进行染色处理,来确认作为蛋白质-壳聚糖复合物的高分子物质的生成。
接着,说明本发明的具有抗真菌活性的组合物。
为了更好地保持抗菌活性,本发明的具有抗真菌活性的组合物,优选包含10~100质量%的溶菌酶-壳聚糖复合物,更优选包含30~100质量%的溶菌酶-壳聚糖复合物,最优选包含50~100质量%的溶菌酶-壳聚糖复合物。
另外,作为具有抗真菌活性的组合物中的溶菌酶-壳聚糖复合物以外的成分,可以使用糊精、卵磷脂等。具有抗真菌活性的组合物中的溶菌酶-壳聚糖复合物以外的成分的含量,优选的是0~90质量%,更优选的是0~70质量%,最优选的是0~50质量%。
2.第二实施方式
第二实施方式的具有抗真菌活性的组合物,包含通过美拉德反应将溶菌酶与壳聚糖结合而得到的复合物以及萜烯醇。另外,作为萜烯醇,可以列举香叶醇、薄荷醇、萜品烯-4-醇等。此外,这些萜烯醇是植物精油成分,可以使用包含这些物质的植物精油。
3.第三实施方式
第三实施方式的具有抗真菌活性的组合物,包含通过美拉德反应将溶菌酶与壳聚糖结合而得到的复合物以及脂肪酸。另外,作为脂肪酸,可以使用辛酸(碳原子数8)、癸酸(碳原子数10)、月桂酸(碳原子数12)等。
实施例
以下举出实施例更具体地说明本发明,但是本发明不限于这些实施例。另外,在以下的实施例中,溶菌酶-壳聚糖复合物也被称为LYZOX(和興フィルタテクノロジー株式会社(和兴滤清器科技株式会社)的注册商标)。
[实施例1]LYZOX、萜品烯-4-醇和癸酸对念珠菌菌丝相生长的抑制活性
针对念珠菌的酵母相和菌丝相生长的体外抑制试验的方法已被确立,因此,可以测定各种材料的MIC(最小增殖抑制浓度,最低抑菌浓度)。
念珠菌使用帝京大学医真菌研究中心保存的临床分离株Candida albicansTIMM1768,用沙氏葡萄糖琼脂培养基平板在37℃下培养了20小时。回收生长了的菌体并使其悬浮在无菌水中,调整成菌数5×103cells/ml的念珠菌液。将各试样以与最终浓度配合的方式预先溶解在DMSO(二甲亚砜)中,加入到包含2%小牛血清的1/3RPMI-1640培养基中,由此制成试样液。将(培养基中的DMSO浓度0.5%)的试样液和念珠菌菌液各100μL分别放入96孔微孔板(住友ベークライト,东京)的各孔中,在5%二氧化碳气体存在下,在37℃下培养了3小时。3小时后,将各孔内的液体废弃并清洗孔后,重新向各孔分别加入200μL的1/3RPMI-1640,在5%二氧化碳气体存在下,在37℃下培养了16小时。培养结束后,将各孔中的液体吸除,用生理盐水清洗,放入200μL的70%乙醇杀灭了念珠菌。除去乙醇并用自来水清洗后,放入100μL的染色液(溶解在0.1M磷酸缓冲液中的0.01%结晶紫溶液)后静置15分钟,对附着在孔的表面的念珠菌的菌丝进行了染色。
用自来水清洗并除去多余的染色液后,放入包含0.04NHCl的3-异丙醇150μL和0.25%十二烷基硫酸钠溶液50μL,使附着在菌体上的色素脱离。使色素脱离后,将板放在多扫描光度计(Multiskan FCサーモフィッシャーサイエンティフィック株式会社)上,测定了各孔的OD620nm。利用以下的式子求出了增殖抑制率。

如图1和图2所示,LYZOX、萜品烯-4-醇和癸酸的各IC70(70%增殖抑制浓度)如下,

[实施例2]针对念珠菌菌丝相生长的LYZOX与、萜品烯-4-醇或癸酸并用的抗念珠菌活性的协同效果
通常,针对微生物的并用抗菌物质的情况下的协同效果的定义,是指比两药的单独效果的和明显显示出更大效果的情况。此外针对协同效果的评价如下所述,可以通过棋盘法利用从两药的MIC的值求出的FIC系数(Fractional Inhibitory ConcentrationIndex)进行协同效果的评价。即,当FIC系数小于0.5时,判断为存在协同效果。

<利用FIC系数的并用效果的评价>
关于并用带来的抗念珠菌活性的协同效果,利用棋盘法进行评价。在96孔微孔板上假定棋盘,将萜品烯-4-醇或癸酸与LYZOX各自的浓度系列交叉。试验条件和测定方法等与针对菌丝相生长的试验(实施例1)相同。
如图1所示,在将LYZOX与萜品烯-4-醇并用的情况下,LYZOX和萜品烯-4-醇的IC(70%增殖抑制浓度)如下所述。
LYZOX(并用时) 60μg/ml
萜品烯-4-醇(并用时) 280μg/ml
即,在将LYZOX与萜品烯-4-醇并用的情况下,针对念珠菌的菌丝相生长的FIC系数为0.447,判断为存在协同效果。
此外,如图2所示,在LYZOX与癸酸(decanoic acid)并用的情况下,LYZOX和癸酸(decanoic acid)的IC(70%增殖抑制浓度)如下所述。

即,在将LYZOX与癸酸并用的情况下,针对念珠菌的菌丝相生长的FIC系数为0.208,判断为存在协同效果。
[实施例3]小鼠口腔念珠菌病模型的LYZOX与萜品烯-4-醇的并用的治疗效果
试验动物使用ICR系小鼠(雌性,6周龄,日本チャールスリバー),在念珠菌接种前一天,以免疫抑制为目的,皮下注射了泼尼松龙100mg/kg。此外,该接种前一天使小鼠开始自由摄取包含盐酸金霉素15mg/ml的自来水。接种当天,为了使小鼠保持安静状态,预先肌内注射了氯丙嗪盐酸盐(和光纯药工业)14.4mg/kg。使用的菌株和培养方法与前述的实施例相同,使生长了的菌体悬浮在包含2%小牛血清的RPMI-1640培养基中,将菌数调整成2×108cells/ml,由此得到菌液。将棉棒浸渍于所述菌液,并将浸渍了菌液的棉棒向变得安静的小鼠的口腔擦拭,由此接种了念珠菌。
针对所述口腔念珠菌病小鼠,预先使用吐温80(最终浓度1%)将下表的给与试样悬浮在蒸留水中,在念珠菌接种3、24和27小时后,使用小鼠用胃管将试样滴下给与到口腔内的舌背上。接种2天后使小鼠安乐死,按照基准,评价了舌头症状评分。随后用棉棒擦拭小鼠口腔内,将所述棉棒上回收的念珠菌菌体悬浮在生理盐水中。将一定量的该悬浮液涂布在念珠菌GS平板培养基上,在37℃培养了20小时后,计测了出现的菌落数。根据菌落数计算出从所述个体回收的念珠菌活菌数CFU(Colony Forming Unit)。

试验的结果如图3所示,关于从口腔回收的活菌数,仅在萜品烯-4-醇的组中,确认了活菌数的降低。此外,如图4所示,关于舌头症状评分,仅在LYZOX的组中确认到了显著的改善效果。
[实施例4]针对念珠菌菌丝相生长的溶菌酶单一成分、壳聚糖单一成分、溶菌酶和壳聚糖的混合物或LYZOX与、萜品烯-4-醇的并用带来的抗念珠菌活性的效果
关于并用带来的抗念珠菌活性的协同效果,与所述实施例2同样,利用棋盘法进行了评价。在96孔微孔板上假定棋盘,使溶菌酶单一成分、壳聚糖单一成分、溶菌酶和壳聚糖的混合物或LYZOX与萜品烯-4-醇各自的浓度系列交叉。试验条件和测定方法等与针对菌丝相生长的试验(实施例1)相同。
试验的结果如图5~图7所示,在溶菌酶单一成分与萜品烯-4-醇并用的情况下、壳聚糖单一成分与萜品烯-4-醇并用的情况下、溶菌酶和壳聚糖的混合物与萜品烯-4-醇并用的情况下,都未观察到针对念珠菌的菌丝相生长的并用带来的抗念珠菌活性的改善效果。另一方面如图1所示,与所述三个并用模式相比,在将LYZOX与萜品烯-4-醇并用的情况下,确认到了显著的改善效果。
[实施例5]针对念珠菌菌丝相生长的溶菌酶单一成分、壳聚糖单一成分、溶菌酶和壳聚糖的混合物或LYZOX与、癸酸的并用带来的抗念珠菌活性的效果
关于并用带来的抗念珠菌活性的协同效果,与所述实施例2同样,利用棋盘法进行了评价。在96孔微孔板上假定棋盘,使溶菌酶单一成分、壳聚糖单一成分、溶菌酶和壳聚糖的混合物或LYZOX与癸酸各自的浓度系列交叉。试验条件和测定方法等与针对菌丝相生长的试验(实施例1)相同。
试验的结果如图8~图10所示,在溶菌酶单一成分与癸酸并用的情况下、壳聚糖单一成分与癸酸并用的情况下、溶菌酶和壳聚糖的混合物与癸酸并用的情况下,都未观察到针对念珠菌的菌丝相生长的并用带来的抗念珠菌活性的改善效果。另一方面如图2所示,与所述三个并用模式相比,在LYZOX与癸酸并用的情况下,确认到了显著的改善效果。
[实施例6]针对念珠菌菌丝相生长的溶菌酶单一成分、壳聚糖单一成分、溶菌酶和壳聚糖的混合物、以及LYZOX带来的抗念珠菌活性的效果
关于各试剂的抗念珠菌活性的效果,与所述实施例2同样,利用棋盘法进行了评价。在96孔微孔板上假定棋盘,分别制作了溶菌酶单一成分、壳聚糖单一成分、溶菌酶和壳聚糖的混合物以及LYZOX各自的浓度系列。试验条件和测定方法等与针对菌丝相生长的试验(实施例1)相同。
试验的结果如图11所示,在溶菌酶单一成分的情况和壳聚糖单一成分的情况下,都未观察到针对念珠菌的菌丝相生长的抗念珠菌活性的改善效果。此外,判明了,在溶菌酶和壳聚糖的混合物(MIX)的情况下,尽管观察到针对念珠菌的菌丝相生长的抗念珠菌活性的改善效果,但是随着浓度上升,抗念珠菌活性的效果变小。另一方面,判明了,在LYZOX的情况下,观察到了针对念珠菌的菌丝相生长的抗念珠菌活性的改善效果,并且所述改善效果随着浓度上升而增加。特别是,在LYZOX浓度为250μg/ml的情况下,生长率基本成为零(0.9%)。
[实施例7]针对马拉色菌(Malassezia pachydermatis)的酮康唑(ketoconazole)和溶菌酶-壳聚糖复合物带来的抗马拉色菌活性的效果
作为马拉色菌的药敏试验,将调整为McFarland 1.0的受检菌液100μL分别涂布在MLNA培养基(组成如表2所示)的琼脂平板上并铺开,在培养皿上放置厚的8mm的抗生物质检定用纸的无菌圆形纸片(paper disk),滴下各受检试样50μL并在32℃下培养7天后,进行了观察和判断。此外,测定了抑菌圈直径。另外,作为受检试样,使用了0.5%DMSO、酮康唑10μg/ml、酮康唑100μg/ml、LYZOX5mg/ml、LYZOX10mg/ml、LYZOX20mg/ml、LYZOX40mg/ml。

各受检试样的马拉色菌抑菌圈(mm)的结果如下所示。

在实验1中,如图12所示,在滴下50μl的LYZOX10mg/ml的圆形纸片周围,出现了16×16mm(圆形纸片8mm)的抑菌圈。同样,在滴下50μl的LYZOX20mg/ml的圆形纸片的周围,出现了21×21mm的抑菌圈,在滴下50μl的LYZOX40mg/ml的圆形纸片的周围,出现了24×24mm的抑菌圈。在对照的无菌水中未观察到抑菌圈。
在实验2中,在滴下50μl的LYZOX10mg/ml的圆形纸片周围,出现了14×15mm的抑菌圈,在滴下50μl的LYZOX5mg/ml的圆形纸片周围,出现了10×11mm的抑菌圈。
[实施例8]
使用琼脂稀释法评价了针对马拉色菌(Malassezia pachydermatis)的LYZOX的抗马拉色菌活性。具体地说,制作了以LYZOX及其他的试样分别成为以上的表2的浓度(最终浓度)的方式添加的MLNA培养基的琼脂平板(组成和所述的表2相同),在所述平板上涂布调整为McFarland1.0的受检菌液100μL并铺开,在32℃下培养了7天。随后,将菌的生长率与对照(无试样添加的MLNA培养基)比较,进行了判断。
各受检试样对菌的阴性·阳性的结果如下所示。另外,将对照的菌丝的生长率设为100%,当受检试样的菌丝生长率为0%时判断为“-”,当菌丝生长率为1~20%时判断为“+”,当菌丝生长率为21~90%时判断为“+++”。此外,各受检试样中的菌的菌落形成的状态如图13和图14所示。另外,图13和图14是表示各试样中的马拉色菌的菌落形成的7天后的状态的照片。

通过以上的试验判明了,针对马拉色菌的LYZOX单一成分显示了抗马拉色菌活性。此外判明了,通过并用LYZOX和癸酸,也显示了抗马拉色菌活性,但是未发现相对于LYZOX单一成分的改善效果。认为马拉色菌是完全酵母相的真菌,因此与其他真菌的抗菌效果产生了不同。
[实施例9]针对毛癣菌的LYZOX和癸酸的抗毛癣菌活性、以及通过并用它们带来的抗毛癣菌活性的协同效果
与针对念珠菌的试验同样,针对毛癣菌的体外抑制试验的方法也已确立了,因此可以测定各种材料的MIC(最少增殖抑制浓度)。
毛癣菌使用了帝京大学医真菌研究中心保存的临床分离株Trichophytonmentagrophytes TIMM2789株。作为预培养,在试验日的1周前从低温石英原种(stock)取出,在包含50μg/ml的氯霉素和500μg/ml的放线酮的、(将成分稀释到1/10)沙氏葡萄糖琼脂培养基平板上植菌。将该平板在28℃下培养1周,在试验当天收集并使用。以毛癣菌的菌数的设定的最终浓度成为103cells/ml的方式混合稀释到培养基中。此外,各试样也分别以成为以下的表中的最终浓度的方式混合稀释到培养基中。包含1.5%琼脂和调整的菌液的RPMI培养基(最终浓度稀释到1/3),也以在制备中不凝固的方式被保温在37℃。制备时将各试样溶液1ml、RPMI-1640培养基24ml混合,在无菌塑料培养皿中制备。将这样制备的毛癣菌接种培养基在28℃下培养4天,对培养后的菌丝的增殖抑制效果进行了观察、判断。判断通过目视和混浊时用显微镜观察(40倍),将菌丝的生长程度与对照(没有添加试样)比较,由此进行了评价。
各受检试样的菌的阴性·阳性的结果如以下表6所示。另外,将对照的菌丝的生长率设为100%,当受检试样的菌丝生长率为0%时判断为“-”,当受检试样的菌丝生长率为1~20%时判断为“+”,当受检试样的菌丝生长率为21~90%时判断为“+++”。此外,各受检试样中的菌的菌落形成的状态如图15和图16所示。另外,图15和图16是显示各试样中的毛癣菌的菌落形成7天后的状态的照片。

此外,在将LYZOX和癸酸并用的情况下,LYZOX和癸酸的MIC(最小增殖抑制浓度)如下所述。

癸酸(并用时) 0.2mg/ml
即,在将LYZOX和癸酸并用的情况下,针对毛癣菌的增殖抑制的FIC系数为0.143,判断为存在协同效果。
工业实用性
按照本发明,不依赖已有的具有包含抗念珠菌的抗真菌活性的组合物或这些组合物的组合,能够提供新的具有抗真菌活性的组合物。
Claims (5)
1.使溶菌酶与壳聚糖结合而得到的复合物和癸酸在制造具有抗真菌活性的组合物中的应用,所述抗真菌活性是念珠菌增殖抑制作用或毛癣菌增殖抑制作用,所述壳聚糖的分子量为25000Da以下。
2.根据权利要求1所述的应用,其中,所述壳聚糖的分子量为14000Da以下。
3.根据权利要求1所述的应用,其中,所述抗真菌活性是念珠菌增殖抑制作用。
4.根据权利要求1所述的应用,其中,所述抗真菌活性是毛癣菌增殖抑制作用。
5.根据权利要求1所述的应用,其中,所述组合物包含10质量%以上且小于100质量%的所述复合物。
