この発明は、アカントアメーバに起因する炎症を治療又は予防するための医薬組成物等に関するものである。
アカントアメーバは、アメーバ型の原虫(原生生物)の一種で、土壌中等に多く存在する微生物である。アカントアメーバは、眼の、特に角膜に感染し、角膜の炎症を引き起こす。アカントアメーバに起因する炎症は特にコンタクトレンズ装着者に多く、例えば水道水を介してコンタクトレンズケースに付着し、ケース内で増殖し、コンタクトレンズを介して角膜における感染を引き起こす。

本発明によれば、アカントアメーバを除去、抗原虫、又は殺原虫する効果、並びにその増殖を抑える効果を奏する。特に、本発明のリゾチームとキトサンとを結合させた複合体は、アカントアメーバの増殖を抑制するのみならず、有意に死滅させる効果も有する。したがって、本発明によれば、アカントアメーバ感染症の治癒及び/又は予防をすることができる。また、リゾチームは、安全性の高い天然由来の食品添加物としても広く利用されており、このリゾチームとキトサンとの複合体を用いた抗原虫組成物は、利用する患者を安心させるとともに、その負担を軽減することができる。
以下に本発明の一実施形態を詳述する。なお、なお、以下で例示する好ましい態様やより好ましい態様等は、「好ましい」や「より好ましい」等の表現にかかわらず適宜相互に組み合わせて使用することができる。また、数値範囲の記載は例示であって、各範囲の上限と下限並びに実施例の数値とを適宜組み合わせた範囲も好ましく使用することができる。さらに、「含有する」又は「含む」等の用語は、「本質的になる」や「のみからなる」と読み替えてもよい。
<医薬組成物>
本発明は、リゾチームとキトサンとを結合させた複合体を含有する、アカントアメーバに起因する炎症を治療又は予防するための医薬組成物に関する。以下、当該医薬組成物を具体的に説明する。
「リゾチームとキトサンとを結合させた複合体」とは、リゾチームとキトサンとを、例えばメイラード反応等により結合させた複合体である(図1参照)。リゾチームと水溶性キトサンとをメイラード反応により結合させることで、リゾチーム中の抗原構造のほとんど又はすべてがマスクされるため、リゾチーム-キトサン複合体をヒトが摂取してもアレルギーを起こしにくいという特徴がある。その他、架橋剤を使用してリゾチームとキトサンとを共有結合させて上記複合体を得ることもできる。
<医薬組成物>
本発明は、リゾチームとキトサンとを結合させた複合体を含有する、アカントアメーバに起因する炎症を治療又は予防するための医薬組成物に関する。以下、当該医薬組成物を具体的に説明する。
「リゾチームとキトサンとを結合させた複合体」とは、リゾチームとキトサンとを、例えばメイラード反応等により結合させた複合体である(図1参照)。リゾチームと水溶性キトサンとをメイラード反応により結合させることで、リゾチーム中の抗原構造のほとんど又はすべてがマスクされるため、リゾチーム-キトサン複合体をヒトが摂取してもアレルギーを起こしにくいという特徴がある。その他、架橋剤を使用してリゾチームとキトサンとを共有結合させて上記複合体を得ることもできる。
ここで「リゾチーム」は、ムコ多糖類を加水分解する酵素であり、ニワトリ由来のリゾチーム、ヒト由来のリゾチームを好ましく用いることができる。
リゾチームの分子量の上限は、例えば、30,000Da以下であり、より好ましくは25,000Da以下、20,000Da以下、18,000Da以下、15,000Da以下であってもよい。またリゾチームの分子量の下限は特に制限しなくてもよいが、例えば、1,000Da以上、好ましくは、5,000Da以上、10,000Da以上、12,000Da以上であってもよい。当該リゾチームの分子量の範囲は、上記いずれかの上限値及び下限値の間の範囲であり得るが、例えば、1000Da~30,000Da、好ましくは5,000Da~20,000Daであり、より好ましくは10,000Da~15,000Daであり得る。
「キトサン」は、以下の化学式(I)で示されるポリ-β1→4-グルコサミンである((C6H11NO4)n、CAS登録番号9012-76-4)。
当該キトサンは水溶性である。キトサンの分子量の上限は、例えば、30,000Da以下であり、より好ましくは20,000Da以下、15,000Da以下、10,000Da以下、7,000Da以下であってもよい。またキトサンの分子量の下限は特に制限しなくてもよいが、例えば、300Da以上、好ましくは、500Da以上、1,000Da以上、3,000Da以上であってもよい。当該キトサンの分子量の範囲は、上記いずれかの上限値及び下限値の間の範囲であり得るが、例えば、300Da~30,000Da、好ましくは500Da~15,000Daであり、より好ましくは1,000Da~10,000Da、特に好ましくは3,000Da~7,000Daであり得る。抗原虫性に着目すると、分子量の大きいキトサンの方が有利となり、製造のしやすさに着目すると、分子量の小さいキトサンの方が溶解性及び安定性が良好となり、低分子量のキトサンの方が有利となる。
なお、ここでキトサンと言うときは、上記キトサンの他、キトサンオリゴ糖やグルコサミンを含む意味とする。キトサンオリゴ糖は、上記式(I)で示される、Dグルコサミンが数個程度連なったもので、低分子のキトサン、又は狭義のキトサンを塩酸または酵素で加水分解したものを意味する。

なお、ここでキトサンと言うときは、上記キトサンの他、キトサンオリゴ糖やグルコサミンを含む意味とする。キトサンオリゴ糖は、上記式(I)で示される、Dグルコサミンが数個程度連なったもので、低分子のキトサン、又は狭義のキトサンを塩酸または酵素で加水分解したものを意味する。
上記架橋剤としては、例えば、アミン反応性架橋剤(例えば、アルコキシアミン)、カルボニル反応性架橋剤(例えば、ヒドラジン化合物)、及びスルフヒドリル反応性架橋剤等を挙げることができる。
リゾチーム/キトサンの質量比は、例えば、99/1~1/99、好ましくは90/10~10/90、より好ましくは80/20~20/80、さらに好ましくは60/40~40/60、特に好ましくは50/50となることが適当である。
具体的なリゾチームとキトサンとを結合させた複合体の製造方法としては、例えば次のようなものが挙げられる。まず、上記質量比のリゾチームとキトサンとを水に混合溶解し、得られた水溶液中のリゾチーム及びキトサンの合計質量が5~30質量%となるように調製する。得られた水溶液を凍結乾燥して粉末化する。得られた粉末を、例えば50~80℃、好ましくは55~65℃の温度と、例えば50~80%、好ましくは60~70%の相対湿度の条件下で、例えば2~20日、より好ましくは7~14日、メイラード反応させることにより本発明のリゾチームとキトサンとを結合させた複合体を製造することができる。
具体的なリゾチームとキトサンとを結合させた複合体の製造方法としては、例えば次のようなものが挙げられる。まず、上記質量比のリゾチームとキトサンとを水に混合溶解し、得られた水溶液中のリゾチーム及びキトサンの合計質量が5~30質量%となるように調製する。得られた水溶液を凍結乾燥して粉末化する。得られた粉末を、例えば50~80℃、好ましくは55~65℃の温度と、例えば50~80%、好ましくは60~70%の相対湿度の条件下で、例えば2~20日、より好ましくは7~14日、メイラード反応させることにより本発明のリゾチームとキトサンとを結合させた複合体を製造することができる。
本実施形態のリゾチーム-キトサン複合体が生成しているか否かは、種々の公知の方法で確認できるが、例えば、SDS(Sodium dodecyl sulfate)やSDS-PAGE(Sodium dodecyl sulfate-polyacrylamide)ポリアクリルアミド電気泳動により得られるプレートを、染色処理することによって、蛋白質-キトサン複合体である高分子物質の生成を確認することができる。
「アカントアメーバ」(Acanthamoeba polyphaga)は、原生動物であり、原虫、特に根足虫類に分類されるアメーバの一種であり、例えば土壌や淡水中に存在する。上述したように、アカントアメーバは、皮膚や眼、特に眼の角膜に感染し、皮膚の炎症や角膜の炎症を引き起こす。アカントアメーバに起因する炎症は特にコンタクトレンズ装着者に多く、例えば水道水を介してアカントアメーバがコンタクトレンズケースに付着し、ケース内でアカントアメーバが増殖し、コンタクトレンズ保存液やコンタクトレンズを介して角膜にアカントアメーバが感染して炎症を引き起こす。炎症としては皮膚炎や眼の角膜炎が挙げられる。特に、本発明は、角膜充血、角膜混濁、放射状角膜神経炎、円盤状角膜潰瘍、強膜炎、眼内炎等の眼の障害への効果が期待できる。
本発明は、「アカントアメーバ」に対して抗原虫性を有する。「原虫」はアカントアメーバ等アメーバを含む意味である。ここで抗原虫性とは広く原虫の予防、除去、殺原虫、抗原虫等を含め、原虫数が同等又はそれ以下になる場合、少なくとも原虫を増殖させない全ての状態を意味する。なお、原虫の除去は、原虫を取り除くこと全般を意味し、殺原虫を含む意味である。殺原虫は少なくとも一部の原虫を死滅させることを意味する。従って、本発明において抗原虫組成物とは、原虫除去用組成物、殺原虫組成物、原虫除去剤、殺原虫剤、及び抗原虫剤を含む意味である。
特に本発明は、アカントアメーバがコンタクトレンズケースなどの物質に付着することを防止する、抗付着剤としても利用できる。アカントアメーバはコンタクトレンズケースのようなプラスチック表面に付着しやすい性質を有するが、本発明がコンタクトレンズ用洗浄液中等に存在することによって、アカントアメーバの当該付着を抑制することができる。上述した前記アカントアメーバに起因する炎症の治療又は予防は、アカントアメーバの付着抑制によっても達成され得る。本発明のアカントアメーバに対する抗付着性は、コンタクトレンズケースに使用されるポリプロピレンの他、ポリエチレン、ポリウレタン、ポリカーボネート、ポリビニルポリマー、メラミン樹脂、ユリア樹脂、エポキシ樹脂、アクリル樹脂、シリコーン、エラストマー、ゴム等のコンタクトレンズケース、洗浄液用ボトル、未装着コンタクトレンズを封入するブリスターパック、玩具等で使用される高分子樹脂組成物、ヒドロキシエチルメタクリレート、シリコーンハイドロゲル、ポリメチルメタクリレート、ガラス等のコンタクトレンズ用材料、紙、金属、布、木材、セラミックス、皮等の種々の物質表面に対して効果を発揮する。なお、ここで言う物質は、皮膚などの人体又は動物及びそれらの一部分を含まないこととすることができる。
本発明は、さらに「アカントアメーバ」に起因する炎症の治療及び予防に効果的である。ここで、「治療」とは、本発明の対象である炎症を完全に治癒することの他、炎症を抑制して重傷度を低下させることを含む。「予防」とは、本発明の対象である炎症の病歴がない場合の他、本発明の対象である炎症が治癒した後に当該炎症が再発しないように防止することを含む。
本発明の医薬組成物には、上記複合体の他に、1種以上の追加の有効成分を任意に含むことができる。追加の有効成分としては、それ自体が有効成分として作用するものの他、それ自体は有効成分として作用しないものの、本発明の有効成分である上記複合体と併用することによって効果を発揮するもの(補助剤)も含む。追加の有効成分としては、例えば、ピグアニド系殺菌剤、過酸化水素水、ポビドンヨード(1-ビニル-2-ピロリドンの重合物とヨウ素の複合体)、塩化ポリドロニウム(ポリクォッド(登録商標))、塩酸ポリヘキサニド(PHMB)、テルペンアルコール、脂肪酸、及び/又は該脂肪酸の塩を挙げることができる。これらの追加の有効成分を1種又はそれ以上添加することにより、本発明の複合体と相まって、相加効果及び/又は相乗効果を得ることができる。特に好ましくは、過酸化水素水、ポビドンヨード(1-ビニル-2-ピロリドンの重合物とヨウ素の複合体)、塩化ポリドロニウム(ポリクォッド(登録商標))、塩酸ポリヘキサニド(PHMB)である。
テルペンアルコールとしては、例えば、テルピネン-4-オール、ヒノキチオール、ゲラニオール、及びメントール等を挙げることができる。脂肪酸としては、例えば、炭素数8~12の脂肪酸、好ましくはデカン酸(カプリン酸)又はラウリン酸を挙げることができる。脂肪酸の塩としては、例えば、ナトリウム塩、カリウム塩、マグネシウム塩、及びカルシウム塩等を挙げることができる。その他、アカントアメーバに起因する炎症に効果があることが知られている塩酸ポリヘキサミド、ラクトフェリン、ホウ酸及び/又はその塩、カスボファンギン及び/又はその塩、ポリコナゾール、ミコナゾール、エコナゾール、スルコナゾール、フルコナゾール、第四級アンモニウム系消毒剤、キチナーゼ等を、当該追加の有効成分として加えてもよい。
テルペンアルコールとしては、例えば、テルピネン-4-オール、ヒノキチオール、ゲラニオール、及びメントール等を挙げることができる。脂肪酸としては、例えば、炭素数8~12の脂肪酸、好ましくはデカン酸(カプリン酸)又はラウリン酸を挙げることができる。脂肪酸の塩としては、例えば、ナトリウム塩、カリウム塩、マグネシウム塩、及びカルシウム塩等を挙げることができる。その他、アカントアメーバに起因する炎症に効果があることが知られている塩酸ポリヘキサミド、ラクトフェリン、ホウ酸及び/又はその塩、カスボファンギン及び/又はその塩、ポリコナゾール、ミコナゾール、エコナゾール、スルコナゾール、フルコナゾール、第四級アンモニウム系消毒剤、キチナーゼ等を、当該追加の有効成分として加えてもよい。
本発明の医薬組成物には、上記複合体の他に、1種以上のその他の成分を任意に含むことができる。その他の成分としては、例えば、賦形剤、結合剤、乳化剤、溶剤、粘着剤、崩壊剤、粘稠剤、滑沢剤、着色剤、流動剤、保湿剤等の添加剤を挙げることができる。
保湿剤としては、例えば、グリセリン、ブチレングリコール、コラーゲン、ヒアルロン酸、及びセラミドを挙げることができる。
結合剤としては、例えば、セルロース、メチルセルロース、ヒドロキシエチルセルロース、及びカルボキシメチルセルロースナトリウムを挙げることができる。
乳化剤としては、例えば、レシチン、ポリエチレングリコール(PG)、PG-水添ヒマシ油、グリセリン脂肪酸エステル、ソルビタン脂肪酸エステル、及びセテス等を挙げることができる。
溶剤としては、例えば、水、エタノール、1-ブタノール、2-ブタノール、1-プロパノール、2-プロパノール、1-ペンタノール等を挙げることができる。
保湿剤としては、例えば、グリセリン、ブチレングリコール、コラーゲン、ヒアルロン酸、及びセラミドを挙げることができる。
結合剤としては、例えば、セルロース、メチルセルロース、ヒドロキシエチルセルロース、及びカルボキシメチルセルロースナトリウムを挙げることができる。
乳化剤としては、例えば、レシチン、ポリエチレングリコール(PG)、PG-水添ヒマシ油、グリセリン脂肪酸エステル、ソルビタン脂肪酸エステル、及びセテス等を挙げることができる。
溶剤としては、例えば、水、エタノール、1-ブタノール、2-ブタノール、1-プロパノール、2-プロパノール、1-ペンタノール等を挙げることができる。
本発明のリゾチーム-キトサン複合体は、本発明の医薬組成物1mL中、例えば、0.01μg/mL以上、0.05μg/mL以上、0.1μg/mL以上、0.5μg/mL以上、1μg/mL以上、10μg/mL以上、50μg/mL以上、100μg/mL以上含有することが適当である。また本発明のリゾチーム-キトサン複合体は、本発明の医薬組成物1mL中、例えば、100,000μg/mL以下、50,000μg/mL以下、10,000μg/mL以下、5,000μg/mL以下、1,000μg/mL以下含有することが適当である。本発明のリゾチーム-キトサン複合体の好ましい含有量の範囲は、これらの下限及び上限を適宜組み合わせた範囲であってよいが、例えば、0.1μg/mL~10,000μg/mL、好ましくは0.5μg/mL~5,000μg/mLや50μg/mL~5,000μg/mL、より好ましくは1μg/mL~1,000μg/mLや100μg/mL~1,000μg/mLとすることが適当である。
本発明のリゾチーム-キトサン複合体は、また、本発明の医薬組成物全体の質量に対し、例えば、0.0001質量%以上、0.001質量%以上、0.005質量%以上、0.01質量%以上、0.05質量%以上、0.1質量%以上、0.5質量%以上、又は1質量%以上含有することが適当である。また本発明のリゾチーム-キトサン複合体は、本発明の医薬組成物全体の質量に対し、例えば、5.0質量%以下、1質量%以下、0.5質量%以下、0.1質量%以下0.05質量%以下、0.01質量%以下、0.005質量%以下含有することが適当である。本発明のリゾチーム-キトサン複合体の好ましい含有量の範囲は、これらの下限及び上限を適宜組み合わせた範囲であってよいが、例えば、0.001質量%~5.0質量%、好ましくは0.005質量%~1.0質量%、より好ましくは0.01質量%~0.5質量%とすることが適当である。
また、追加の有効成分は、本発明の医薬組成物中、例えば、0.00001質量%~20質量%、好ましくは0.00005質量%~10質量%、より好ましくは0.0001質量%~5質量%、さらに好ましくは0.0005質量%~3質量%、特に好ましくは0.001質量%~1質量%含有することが適当である。本発明のリゾチーム-キトサン複合体と、当該追加の有効成分との質量比は、例えば、99/1~1/99、好ましくは90/10~10/90、より好ましくは80/20~20/80、さらに好ましくは60/40~40/60、特に好ましくは50/50となることが適当である。
さらに、その他の成分は、各成分によって適切な含有量は適宜調節する必要があるが、例えば、0.1質量%~90質量%、好ましくは1質量%~70質量%、より好ましくは10質量%~50質量%含有することが適当である。なお、本発明の医薬組成物は、そのほとんどは水などの溶媒であり得、例えば、本発明の医薬組成物中、溶媒を95質量%以上、好ましくは98質量%以上、より好ましくは99質量%以上、さらに好ましくは99.5質量%以上含有することが適当である。
さらに、その他の成分は、各成分によって適切な含有量は適宜調節する必要があるが、例えば、0.1質量%~90質量%、好ましくは1質量%~70質量%、より好ましくは10質量%~50質量%含有することが適当である。なお、本発明の医薬組成物は、そのほとんどは水などの溶媒であり得、例えば、本発明の医薬組成物中、溶媒を95質量%以上、好ましくは98質量%以上、より好ましくは99質量%以上、さらに好ましくは99.5質量%以上含有することが適当である。
<抗原虫組成物>
本発明は、さらに、リゾチームとキトサンとを結合させた複合体を含有する抗原虫組成物に関する。抗原虫組成物中に含まれ得るリゾチームとキトサンとを結合させた複合体の含有量等の詳細、追加の有効成分、その他の成分等の詳細は、すべて上述した医薬組成物の説明の通りである。
本発明は、さらに、リゾチームとキトサンとを結合させた複合体を含有する抗原虫組成物に関する。抗原虫組成物中に含まれ得るリゾチームとキトサンとを結合させた複合体の含有量等の詳細、追加の有効成分、その他の成分等の詳細は、すべて上述した医薬組成物の説明の通りである。
<抗付着剤>
本発明は、さらに、リゾチームとキトサンとを結合させた複合体を含有する抗付着剤に関する。抗付着剤中に含まれ得るリゾチームとキトサンとを結合させた複合体の含有量等の詳細、追加の有効成分、その他の成分等の詳細は、すべて上述した医薬組成物の説明の通りである。理論に縛られるものではないが、この抗付着性は、リゾチームとキトサンとを結合させた複合体が静電的機序によりアカントアメーバに結合し、もってプラスチック等の表面へのアカントアメーバの付着を抑制するものと考えられる。リゾチームとキトサンとを結合させた複合体は正に帯電する一方、アカントアメーバは負に帯電するので、アカントアメーバの物質表面への付着と当該複合体の付着とが競合し、抗付着作用が得られるものと考えられる。
なお、抗付着剤は、通常コンタクトレンズの洗浄液や保存液中に含められる。洗浄液や保存液中における抗付着剤の含有量は、抗付着剤に含まれるリゾチームとキトサンとを結合させた複合体の割合で規定することができ、洗浄液や保存液1mL中、当該リゾチームとキトサンとを結合させた複合体が、例えば、0.001μg/mL以上、0.01μg/mL以上、0.1μg/mL以上、0.5μg/mL以上、1μg/mL以上、10μg/mL以上、50μg/mL以上、100μg/mL以上含有することが適当である。また本発明のリゾチーム-キトサン複合体は、本発明の洗浄液や保存液1mL中、例えば、100,000μg/mL以下、50,000μg/mL以下、10,000μg/mL以下、5,000μg/mL、1,000μg/mL以下含有することが適当である。本発明のリゾチーム-キトサン複合体の好ましい含有量の範囲は、これらの下限及び上限を適宜組み合わせた範囲であってよいが、例えば、0.01μg/mL~100,000μg/mL、好ましくは0.5μg/mL~10,000μg/mL、より好ましくは0.1μg/mL~1,000μg/mLとすることが適当である。
ここで、コンタクトレンズとしては、一般に使用されるソフトコンタクトレンズであってもハードコンタクトレンズであってもよい。洗浄液や保存液としては、生理食塩水をはじめ、界面活性剤、増粘剤、湿潤剤などの成分を含む精製水など、通常コンタクトレンズの洗浄や保存のために使用されるものであってもよく、また、新品の未装着コンタクトレンズに使用されるブリスターパックの保存液であってもよい。
本発明は、さらに、リゾチームとキトサンとを結合させた複合体を含有する抗付着剤に関する。抗付着剤中に含まれ得るリゾチームとキトサンとを結合させた複合体の含有量等の詳細、追加の有効成分、その他の成分等の詳細は、すべて上述した医薬組成物の説明の通りである。理論に縛られるものではないが、この抗付着性は、リゾチームとキトサンとを結合させた複合体が静電的機序によりアカントアメーバに結合し、もってプラスチック等の表面へのアカントアメーバの付着を抑制するものと考えられる。リゾチームとキトサンとを結合させた複合体は正に帯電する一方、アカントアメーバは負に帯電するので、アカントアメーバの物質表面への付着と当該複合体の付着とが競合し、抗付着作用が得られるものと考えられる。

<治療方法、予防方法>
本発明は、リゾチームとキトサンとを結合させた複合体を含有する医薬組成物を対象者に投与して、アカントアメーバに起因する炎症を治療又は予防するための方法を含み得る。当該治療又は予防は、例えば、皮膚や眼、特に眼の角膜に対する感染、角膜の炎症及び/又は疾患を治療または予防することに向けられている。理論に縛られないが、当該治療又は予防効果は、リゾチームとキトサンとを結合させた複合体の上記抗原虫性及び/又は抗付着性に起因すると言われている。
ここで当該治療又は予防するための方法に使用される医薬組成物や各種定義の詳細は上述した通りであり、追加の有効成分等を含み得る点、その他、リゾチームやキトサンなどの各定義、追加の有効成分やその他の成分の含有量並びに含有率等は、すべて上述の医薬組成物の説明の通りである。
ここで「対象者」は、ヒトの他、ネコ、イヌ、サル、ウシ、ウマなどの哺乳動物を含む。
本発明は、リゾチームとキトサンとを結合させた複合体を含有する医薬組成物を対象者に投与して、アカントアメーバに起因する炎症を治療又は予防するための方法を含み得る。当該治療又は予防は、例えば、皮膚や眼、特に眼の角膜に対する感染、角膜の炎症及び/又は疾患を治療または予防することに向けられている。理論に縛られないが、当該治療又は予防効果は、リゾチームとキトサンとを結合させた複合体の上記抗原虫性及び/又は抗付着性に起因すると言われている。
ここで当該治療又は予防するための方法に使用される医薬組成物や各種定義の詳細は上述した通りであり、追加の有効成分等を含み得る点、その他、リゾチームやキトサンなどの各定義、追加の有効成分やその他の成分の含有量並びに含有率等は、すべて上述の医薬組成物の説明の通りである。
ここで「対象者」は、ヒトの他、ネコ、イヌ、サル、ウシ、ウマなどの哺乳動物を含む。
<投与量、投与形態、投与方法>
本発明の医薬組成物は、その剤型、投与対象、投与ルート、対象疾患、症状等によっても異なるが、例えば、ヒトの体、手、指等の皮膚に噴霧するスプレー剤の形態の場合、一日投与量は、例えば、0.1~20mg/kg体重、好ましくは0.2~10mg/kg体重、さらに好ましくは0.5~10mg/kg体重であり、この量を1日1回~数回(例、2回、3回、4回又は8回)投与するのが望ましい。この投与量は、スプレー剤以外にも以下で説明するクリーム等の他の製剤においても好ましく適用できる。
本発明の医薬組成物は、その剤型、投与対象、投与ルート、対象疾患、症状等によっても異なるが、例えば、ヒトの体、手、指等の皮膚に噴霧するスプレー剤の形態の場合、一日投与量は、例えば、0.1~20mg/kg体重、好ましくは0.2~10mg/kg体重、さらに好ましくは0.5~10mg/kg体重であり、この量を1日1回~数回(例、2回、3回、4回又は8回)投与するのが望ましい。この投与量は、スプレー剤以外にも以下で説明するクリーム等の他の製剤においても好ましく適用できる。
本発明の医薬組成物は、経口でも非経口でも投与することができ、これらの製剤化には特別な技術は必要なく、汎用される技術を用いて製剤化をすることができる。投与剤型としては、クリーム、軟膏、ハップ剤、液剤、スプレー剤、ゲル剤、注射剤、錠剤、坐剤、カプセル剤、顆粒剤、散剤、点眼剤、眼軟膏などが挙げられ、クリーム、軟膏、ハップ剤、液剤、スプレー剤が特に好ましい。
本発明の医薬組成物は、上記投与形態に合わせて適宜投与方法を選択し得る。投与方法は公知の投与方法でよく、例えば、クリームの場合、本発明の医薬組成物を炎症箇所に上記投与量で塗布すればよい。

<抗原虫方法、使用量、使用形態、使用方法>
本発明は、リゾチームとキトサンとを結合させた複合体を含有する抗原虫組成物を対象に使用して、アカントアメーバ等の原虫の予防、除去、殺原虫、抗原虫等を行うことができる。抗原虫性の意味は上述したとおりである。対象がヒト等哺乳動物の場合、上記医薬組成物に関する治療方法、予防方法、投与量、投与形態、投与方法の説明がこの抗原虫方法に適用できる。対象が非ヒトの場合も適宜上述した医薬組成物に関する治療方法、予防方法、投与量、投与形態、投与方法に準じてその使用量、使用形態、使用方法を選択することができる。例えば、対象がコンタクトレンズケースの場合、抗原虫組成物を液剤やスプレー剤としてコンタクトレンズケースに散布してもよい。

本発明は、リゾチームとキトサンとを結合させた複合体を含有する抗付着剤を対象に使用して、アカントアメーバ等の原虫の付着の予防及び除去等を行う。抗付着性の意味及び付着対象は上述したとおりである。抗付着剤の使用量、使用形態、使用方法は、上記医薬組成物に関する治療方法、予防方法、投与量、投与形態、投与方法の説明を適用できる。例えば、抗付着の対象がコンタクトレンズケースの場合、コンタクトレンズの洗浄液や保存液中に当該抗付着剤を液剤として混合してもよい。
本発明は、リゾチームとキトサンとを結合させた複合体を含有する抗付着剤を対象に使用して、アカントアメーバ等の原虫の付着の予防及び除去等を行う。抗付着性の意味及び付着対象は上述したとおりである。抗付着剤の使用量、使用形態、使用方法は、上記医薬組成物に関する治療方法、予防方法、投与量、投与形態、投与方法の説明を適用できる。例えば、抗付着の対象がコンタクトレンズケースの場合、コンタクトレンズの洗浄液や保存液中に当該抗付着剤を液剤として混合してもよい。
<使用>
本発明は、また、リゾチームとキトサンとを結合させた複合体を含有する、アカントアメーバに起因する炎症を治療又は予防するための医薬組成物、抗原虫組成物、並びに抗付着剤の製造における、リゾチームとキトサンとを結合させた複合体の使用を含み得る。
当該医薬組成物、抗原虫組成物、並びに抗付着剤の製造において、リゾチームとキトサンとを結合させた複合体は、複合体以外の追加の有効成分やその他の成分と共に任意に混合され、当該医薬組成物等としてもよい。当該混合方法としては、公知の方法を用いることができるが、例えば液剤の場合、水などの溶剤に、リゾチームとキトサンとを結合させた複合体及び上記任意のその他の成分等を加え、混合し、必要に応じて乳化剤を加えて混合して分散剤又はエマルジョンとし、液剤が調製される。
本発明は、また、リゾチームとキトサンとを結合させた複合体を含有する、アカントアメーバに起因する炎症を治療又は予防するための医薬組成物、抗原虫組成物、並びに抗付着剤の製造における、リゾチームとキトサンとを結合させた複合体の使用を含み得る。
当該医薬組成物、抗原虫組成物、並びに抗付着剤の製造において、リゾチームとキトサンとを結合させた複合体は、複合体以外の追加の有効成分やその他の成分と共に任意に混合され、当該医薬組成物等としてもよい。当該混合方法としては、公知の方法を用いることができるが、例えば液剤の場合、水などの溶剤に、リゾチームとキトサンとを結合させた複合体及び上記任意のその他の成分等を加え、混合し、必要に応じて乳化剤を加えて混合して分散剤又はエマルジョンとし、液剤が調製される。
この医薬組成物、抗原虫組成物、並びに抗付着剤は、ネブライザーに用いられる洗浄水とすることができる。これにより医薬組成物、抗原虫組成物、並びに抗付着剤を必要な場所に噴霧することで、アカントアメーバ等の原虫の増殖や付着を抑制し、かつ当該原虫を死滅させることができる。
また、本実施形態の医薬組成物、抗原虫組成物、並びに抗付着剤は、例えば、アカントアメーバに対して抗原虫効果や抗付着効果等を有し、及びこれらに起因する炎症に対して治療、抑制、予防効果を有する。
以下に実施例を掲げて本発明を更に詳細に説明するが、本発明はこれら実施例のみに限定されるものではない。なお、本明細書及び図面において、リゾチームとキトサンとを結合させた複合体は、リゾチーム-キトサン複合体、リゾックス、又はLYZOX(和興フィルタテクノロジー株式会社の登録商標)ともいう。
[試料の調製]
・リゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))溶液
リゾチームとキトサンとを結合させた複合体(リゾチーム-キトサン複合体)の溶液として、市販のLYZOX(登録商標、和興フィルタテクノロジー株式会社製)を使用した。市販のLYZOX(登録商標)は、ニワトリ由来のリゾチーム(約14,000Da)と、約5,000Daの水溶性キトサン(キトサンオリゴ糖)とを1:1の質量比で含む粉末である。具体的には、上記リゾチームと水溶性キトサンとを、水に混合溶解し、その後凍結乾燥させることによって、粉末状とし、さらにメイラード反応が完全に終了するのに十分な温度、湿度及び日数の条件下でメイラード反応させることによって本発明において使用するリゾチーム-キトサン複合体(LYZOX(登録商標))を得ている。当該リゾチーム-キトサン複合体(LYZOX(登録商標))を、各実験例において当該リゾチーム-キトサン複合体が所望の濃度となるように滅菌水と混合し、目的のリゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))溶液とした。
・リゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))溶液
リゾチームとキトサンとを結合させた複合体(リゾチーム-キトサン複合体)の溶液として、市販のLYZOX(登録商標、和興フィルタテクノロジー株式会社製)を使用した。市販のLYZOX(登録商標)は、ニワトリ由来のリゾチーム(約14,000Da)と、約5,000Daの水溶性キトサン(キトサンオリゴ糖)とを1:1の質量比で含む粉末である。具体的には、上記リゾチームと水溶性キトサンとを、水に混合溶解し、その後凍結乾燥させることによって、粉末状とし、さらにメイラード反応が完全に終了するのに十分な温度、湿度及び日数の条件下でメイラード反応させることによって本発明において使用するリゾチーム-キトサン複合体(LYZOX(登録商標))を得ている。当該リゾチーム-キトサン複合体(LYZOX(登録商標))を、各実験例において当該リゾチーム-キトサン複合体が所望の濃度となるように滅菌水と混合し、目的のリゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))溶液とした。
・リゾチーム単体の溶液
リゾチーム単体の溶液は、卵白リゾチーム 分子量約14300(日本バイオコン(株)製、愛知県)を、各実験例において当該リゾチームが所望の濃度となるように滅菌水と混合し、目的のリゾチーム単体の溶液とした。
・キトサン単体の溶液
キトサン単体の溶液は、キトサンオリゴ糖((株)キミカ社製キミカキトサンオリゴ糖 COS-A)をキトサンとして用い、各実験例において当該キトサンオリゴ糖が所望の濃度となるように滅菌水と混合し、目的のキトサン単体の溶液とした。
リゾチーム単体の溶液は、卵白リゾチーム 分子量約14300(日本バイオコン(株)製、愛知県)を、各実験例において当該リゾチームが所望の濃度となるように滅菌水と混合し、目的のリゾチーム単体の溶液とした。
・キトサン単体の溶液
キトサン単体の溶液は、キトサンオリゴ糖((株)キミカ社製キミカキトサンオリゴ糖 COS-A)をキトサンとして用い、各実験例において当該キトサンオリゴ糖が所望の濃度となるように滅菌水と混合し、目的のキトサン単体の溶液とした。
・リゾチーム及びキトサンの混合物の溶液
リゾチーム及びキトサンの混合物は、上記リゾチーム単体:キトサン単体の質量比が1:1となるように両者を混合し、さらに当該混合物、リゾチーム、及びキトサンが所望の濃度となるように滅菌水を混合し、調製した。

アカントアメーバ(Acanthamoeba polyphaga、入手先American Type Culture Collection (ACC)、30872TM)を溶媒:リン酸緩衝生理食塩水に分散してアカントアメーバを溶液1mL当たり1×105個含有するアカントアメーバ溶液を調製した。
・試験片
後述する接着性試験で使用したポリプロピレン製試験片は、入手先:ジョーホク(株)、製品番号:PP_A3のポリプロピレンであって、縦320mm×横440mm×厚み0.25mmの寸法のものから,縦15mm、横15mm,厚み0.25mmに切断したものを各実験に用いた。
具体的な各実験例の詳細は、下記に示すとおりである。
アカントアメーバ(Acanthamoeba polyphaga、入手先American Type Culture Collection (ACC)、30872TM)を溶媒:リン酸緩衝生理食塩水に分散してアカントアメーバを溶液1mL当たり1×105個含有するアカントアメーバ溶液を調製した。
・試験片
後述する接着性試験で使用したポリプロピレン製試験片は、入手先:ジョーホク(株)、製品番号:PP_A3のポリプロピレンであって、縦320mm×横440mm×厚み0.25mmの寸法のものから,縦15mm、横15mm,厚み0.25mmに切断したものを各実験に用いた。
具体的な各実験例の詳細は、下記に示すとおりである。
実験例1(接着性試験及び細胞死試験)
リゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))の抗付着性並びに抗原虫性を検証するため、接着性試験及び細胞死試験を行った。
具体的には、上述したリゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))溶液に、上述したアカントアメーバ溶液を加え、混合溶液1mL中、リゾチーム-キトサン複合体が0.1μg、1μg、10μg、100μg、及び1000μgの濃度となるように混合溶液を調製した。これとは別にリゾチーム-キトサン複合体を含まない0μg/mLの濃度のコントロール溶液を調製した。濃度の調節には希釈液としてリゾチーム-キトサン複合体の調製の際に使用したものと同じ滅菌水を使用した。これら6種類の溶液には、いずれも溶液1mL当たり1×105個のアカントアメーバが含まれている。
調製した混合溶液に上述したポリプロピレン製試験片を浸漬して静置し、浸漬から60分経過後に混合溶液から取り出して、該試験片をリン酸緩衝生理食塩水(PBS、入手先:富士フィルム和光純薬、製品番号:166-23555)で洗浄し乾燥した。得られた試験片の表面を生物顕微鏡(入手先:ニコン、製品番号:ECLIPSEE-200)で観測し、試験片表面の5×5mmの観測範囲に接着したアカントアメーバをカウントした。その後、当該観測範囲にトリパンブルーを適用し、死亡したアカントアメーバを染色して死亡したアカントアメーバの数をカウントした。結果を以下の表1及び図2並びに図3に示す。
リゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))の抗付着性並びに抗原虫性を検証するため、接着性試験及び細胞死試験を行った。
具体的には、上述したリゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))溶液に、上述したアカントアメーバ溶液を加え、混合溶液1mL中、リゾチーム-キトサン複合体が0.1μg、1μg、10μg、100μg、及び1000μgの濃度となるように混合溶液を調製した。これとは別にリゾチーム-キトサン複合体を含まない0μg/mLの濃度のコントロール溶液を調製した。濃度の調節には希釈液としてリゾチーム-キトサン複合体の調製の際に使用したものと同じ滅菌水を使用した。これら6種類の溶液には、いずれも溶液1mL当たり1×105個のアカントアメーバが含まれている。

表1に示すように、リゾチーム-キトサン複合体は、濃度依存的にアカントアメーバの細胞死を引き起こした。リゾチーム-キトサン複合体によるアカントアメーバの接着抑制効果は0.1μg/mLから観察され、特に1μg/mL及びそれ以上の濃度において顕著な接着抑制効果が観察された。100μg/mL及び1000μg/mLではアカントアメーバの接着をほとんど抑制することができ、非常に優れた接着抑制効果が得られた。リゾチーム-キトサン複合体によるアカントアメーバの抑制効果(抗原虫効果)につき、表1中の死亡したアカントアメーバの数及びアカントアメーバの死亡率を見ると、特に100μg/mL及び1000μg/mLにおいてほぼ100%の死亡率を達成しており、顕著な抗原虫作用を有することがわかる。
実験例2(接着性試験及び細胞死試験の比較試験)
上記実験例1のリゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))溶液に代えて、上述したリゾチーム及びキトサンの混合物の溶液(表2)、リゾチーム単体(表3)、キトサン単体(表4)を使用した以外は、実験例1と同様にしてアカントアメーバをカウントし、全アカントアメーバの接着数、死亡したアカントアメーバの数、及びアカントアメーバの死亡率を測定した。結果を以下の表2~4、図4及び図5に示す。
上記実験例1のリゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))溶液に代えて、上述したリゾチーム及びキトサンの混合物の溶液(表2)、リゾチーム単体(表3)、キトサン単体(表4)を使用した以外は、実験例1と同様にしてアカントアメーバをカウントし、全アカントアメーバの接着数、死亡したアカントアメーバの数、及びアカントアメーバの死亡率を測定した。結果を以下の表2~4、図4及び図5に示す。
表1~4及び図4から見て取れるように、本発明のリゾチーム-キトサン複合体は、同質量のリゾチーム及びキトサンを混合した混合物、本発明のリゾチーム-キトサン複合体中のリゾチームと同質量のリゾチーム単体、及び本発明のリゾチーム-キトサン複合体中のキトサンと同質量のキトサン単体に比べて、優れたアカントアメーバの接着抑制効果を示した。特に1μg/mL及びそれ以上の濃度においてリゾチーム及びキトサンの混合物、リゾチーム単体、及びキトサン単体と比較した顕著な接着抑制効果が観察された。また、表2~4及び図5から見て取れるように、リゾチーム-キトサン複合体は、同質量のリゾチーム及びキトサンを混合した混合物、本発明のリゾチーム-キトサン複合体中のリゾチームと同質量のリゾチーム単体、及び本発明のリゾチーム-キトサン複合体中のキトサンと同質量のキトサン単体に比べて、優れたアカントアメーバの死亡率、即ちアカントアメーバの抑制効果(抗原虫効果)を示していた。特に100μg/mL及び1000μg/mLにおいて本発明のリゾチーム-キトサン複合体がほぼ100%の死亡率を達成しているのに対し、リゾチーム及びキトサンの混合物、リゾチーム単体、及びキトサン単体はアカントアメーバの抑制効果が不完全であった。
実験例3(接着性試験及び細胞死試験の比較試験)
リゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))と追加の有効成分とを使用した場合の効果、特に相乗的抗付着性並びに抗原虫性を検証するため、接着性試験及び細胞死試験を行った。
まず、アカントアメーバ含有コンタクトレンズ保存液を準備した。具体的には、アカントアメーバ(Acanthamoeba polyphaga、入手先American Type Culture Collection (ACC)、30872TM)を表5に記載された市販の各コンタクトレンズ保存液に溶解して当該保存液1mL当たりアカントアメーバを1×106個含有するアカントアメーバ含有コンタクトレンズ保存液を調製した。
このように調製したアカントアメーバ含有コンタクトレンズ保存液に、上述したリゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))溶液を加え、混合溶液1mL中、リゾチーム-キトサン複合体が100μg(質量0.01%)の濃度の混合溶液を調製した。これとは別に蒸留水をコントロールとして使用した。濃度の調節には希釈液としてリゾチーム-キトサン複合体の調整の際に使用したものと同じ滅菌水を使用した。
調製した混合溶液に実験例1と同様にしてポリプロピレン製試験片を浸漬し、試験片に接着したアカントアメーバをカウントし、かつ、死亡したアカントアメーバの数をカウントした。結果を以下の表5、図6並びに図7に示す。
リゾチーム-キトサン複合体(リゾックス/LYZOX(登録商標))と追加の有効成分とを使用した場合の効果、特に相乗的抗付着性並びに抗原虫性を検証するため、接着性試験及び細胞死試験を行った。
まず、アカントアメーバ含有コンタクトレンズ保存液を準備した。具体的には、アカントアメーバ(Acanthamoeba polyphaga、入手先American Type Culture Collection (ACC)、30872TM)を表5に記載された市販の各コンタクトレンズ保存液に溶解して当該保存液1mL当たりアカントアメーバを1×106個含有するアカントアメーバ含有コンタクトレンズ保存液を調製した。

調製した混合溶液に実験例1と同様にしてポリプロピレン製試験片を浸漬し、試験片に接着したアカントアメーバをカウントし、かつ、死亡したアカントアメーバの数をカウントした。結果を以下の表5、図6並びに図7に示す。

表5及び図6に示すように、各コンタクトレンズ保存液のみの場合ではアカントアメーバの接着抑制効果は限定的であるが、コンタクトレンズ保存液とリゾチーム-キトサン複合体とを組み合わせた場合は、顕著な接着抑制効果が観察された。また、リゾチーム-キトサン複合体によるアカントアメーバの抑制効果(抗原虫効果)につき、表5中の死亡したアカントアメーバの数及びアカントアメーバの死亡率及び図7を見ると、追加の有効成分として、特にポビドンヨード及び塩酸ポリヘキサニドとリゾチーム-キトサン複合体とを併用した場合、コンタクトレンズ保存液のみの場合に比べて、良好なアカントアメーバの抑制効果(抗原虫効果)が得られた。
本発明によれば、アカントアメーバに対して新規な医薬組成物、抗原虫組成物及び抗付着剤等を提供することができる。
Claims (13)
- リゾチームとキトサンとを結合させた複合体を含有する、アカントアメーバに起因する炎症を治療又は予防するための医薬組成物。
- 前記キトサンが、分子量1000Da~30,000Daの水溶性キトサンである、請求項1に記載の医薬組成物。
- 前記医薬組成物中、前記複合体を、0.01μg/mL~100,000μg/mL含有する、請求項1に記載の医薬組成物。
- 前記医薬組成物中、前記複合体を、0.0001質量%~5.0質量%含有する、請求項1に記載の医薬組成物。
- さらに、過酸化水素水、ポビドンヨード、塩化ポリドロニウム、及び塩酸ポリヘキサニドからなる群から選択される追加の有効成分を含有する、請求項1に記載の医薬組成物。
- アカントアメーバに起因する炎症が、皮膚炎又は角膜炎である、請求項1~5のいずれか1項に記載の医薬組成物。
- 前記アカントアメーバに起因する炎症の治療又は予防が、アカントアメーバの付着抑制によって達成される、請求項6に記載の医薬組成物。
- アカントアメーバに起因する炎症を治療又は予防するための医薬組成物の製造における、リゾチームとキトサンとを結合させた複合体の使用。
- リゾチームとキトサンとを結合させた複合体を含有する、抗原虫組成物。
- 前記原虫がアカントアメーバである、請求項9に記載の抗原虫組成物。
- リゾチームとキトサンとを結合させた複合体を含有する、アカントアメーバの抗付着剤。
- リゾチームとキトサンとを結合させた複合体を物質表面に適用することを含む、物質表面に対するアカントアメーバの付着を防止する方法。
- 前記物質がポルエチレン、ポリプロピレン、ポリウレタン及びポリカーボネートからなる群から選択される、請求項12に記載のアカントアメーバの付着を防止する方法。
