本发明提供一种小球藻生长因子螺旋藻多糖功能饮品制备方法,属于生物技术领域。该方法将小球藻粉配成水溶液后,加热、离心,将上清液超滤、超滤液冷冻干燥获得小球藻生长因子冻干粉。将提取藻蓝蛋白的螺旋藻渣和水混合,一定温度下搅拌浸出,浸出液离心,上清液减压蒸馏后,向浓缩液中加入无水食用酒精,然后在冰箱中沉淀,再离心,得到黄色螺旋藻粗多糖沉淀,将螺旋藻粗多糖沉淀用无水食用酒精洗涤,沉淀冷冻干燥获得米白色螺旋藻多糖冻干粉。将获得的小球藻生长因子冻干粉、螺旋藻多糖冻干粉、白砂糖等食品添加剂和水混合配制即得饮品,再用0.22μm滤膜过滤除菌后封装。该方法可以经济、有效的工业化生产小球藻生长因子螺旋藻多糖功能饮品。

本发明涉及生物技术领域,特别是指一种小球藻生长因子螺旋藻多糖功能饮品制备方法。
背景技术
螺旋藻含有多种维生素、蛋白质、脂肪、碳水化合物、矿物质和微量元素,比例科学合理,具有广泛的食用、保健和药用价值,可以促进人体新陈代谢,提高机体免疫力,抗辐射,辅助癌症放、化疗,降低放疗、化疗副作用等作用,是一种理想的纯天然保健食品,被联合国粮农组织和联合国世界食品协会推荐为“二十一世纪最理想的食品”,从螺旋藻中提取的活性物质也被认为是无害的。
螺旋藻多糖是螺旋藻具有显著的抗病毒抗辐射抗突变功能、降低化疗副作用、提高机体免疫功能、提高病菌、病毒免疫力等作用。虽然研究者提出了一些螺旋藻多糖的提取方法,但是大多是以新鲜螺旋藻或螺旋藻粉为对象提取,鲜有以提取藻蓝蛋白的螺旋藻废渣为原料提取螺旋藻多糖的研究方法。螺旋藻多糖提取的一个关键就是螺旋藻多糖和蛋白质的分离,螺旋藻多糖在温度低于50℃的溶解度很小,而藻蓝蛋白在40℃以上分解。因此在温度不高于40℃条件下提取藻蓝蛋白,提取过程中绝大部分螺旋藻多糖留在螺旋藻渣中。由于藻蓝蛋白提取过程中已经将水溶性蛋白提取出来,因此采用热水从螺旋藻废渣中提取螺旋藻多糖省去了蛋白质去除步骤,简化了工艺,减少了有机溶剂使用、降低了螺旋藻多糖提取成本,适合螺旋藻多糖工业化提取。
世界上没有任何动植物像小球藻那样能够每20h增长4倍,这主要是由小球藻生长因子决定的。小球藻生长因子(Chlorella Growth Factor,CG F),也叫小球藻精,是细胞活性物质,包括氨基酸、核酸、多糖、多肽、蛋白质、酶、维生素、矿物质,被称为“类荷尔蒙”。小球藻生长因子具有:激活淋巴细胞、增强人机体免疫能力,活化人体细胞,促进儿童生长发育,抵抗外来疾病入侵,含有许多修复人体器官所需要的核蛋白,可治疗有机物、重金属等中毒;防治胃溃疡、高血压和心血管等疾病。因此,小球藻生长因子在食品、饮品、保健品、医药等领域具有广阔的应用前景。
虽然小球藻生长因子和螺旋藻多糖的保健功能研究很多,但是目前市场上尚未小球藻生长因子螺旋藻多糖的功能性饮品,本发明提供了一种制备小球藻生长因子螺旋藻多糖功能饮品的方法,对改善亚健康人群的健康状况和促进我国整体健康水平具有重要意义。
发明内容
本发明要解决的技术问题是提供一种小球藻生长因子螺旋藻多糖功能饮品制备方法。
该方法包括如下步骤:
(1)小球藻生长因子制备:
A称取小球藻粉,加入去离子水,搅拌均匀后配成2%~5%的小球藻液,在110~121℃下加热提取20~60分钟,得到小球藻热提取物;
B将步骤A得到的小球藻热提取物冷却至室温,在4℃、6 000~10 000g下离心15~30分钟,得到小球藻生长因子溶液;
C将步骤B中得到的小球藻生长因子溶液超滤,超滤液冷冻干燥得到小球藻生长因子冻干粉;
(2)螺旋藻多糖制备:
D称取提取藻蓝蛋白的螺旋藻藻渣,将藻渣按照1:5~20的质量比加入去离子水混合均匀,在60~100℃、搅拌转速为100~400rpm条件下搅拌浸出2~4小时,得到浸出液;
E将步骤D中得到的浸出液在4℃、6 000~10 000g离心15~30分钟,得到的上清液在60~80℃减压蒸馏至1/4~1/5体积,得到浓缩液;
F向步骤E中得到的浓缩液中加入4~5倍体积的无水食用酒精,放在冰箱4℃下沉淀24小时,得到沉淀液;
G将步骤F中得到的沉淀液在4℃、6 000~10 000g离心15~30分钟,将得到的沉淀用无水食用酒精洗涤2~3次,然后离心获得米白色螺旋藻粗多糖沉淀;
H将步骤G中得到的螺旋藻粗多糖沉淀在-20℃冷冻,然后冷冻干燥获得米白色螺旋藻多糖冻干粉;
(3)小球藻生长因子螺旋藻多糖功能饮品制备:
称取步骤(1)制备的小球藻生长因子冻干粉和步骤(2)制备的螺旋藻多糖冻干粉加入去离子水中,制成小球藻生长因子螺旋藻多糖功能饮品,同时加入食品添加剂来调节功能饮品的口味,最后用0.22μm滤膜过滤除菌后封装。
其中,步骤C中小球藻生长因子溶液超滤的截留分子量为8 000~15000Da。
步骤A中采用高压锅或蒸汽加热的密闭式机械搅拌罐在110~121℃提取小球藻生长因子。
步骤(3)小球藻生长因子螺旋藻多糖功能饮品中小球藻生长因子含量为0.6mg/L~1.0mg/L,螺旋藻多糖含量为1mg/L~2.0mg/L。
步骤(3)中食品添加剂包括白砂糖、苯甲酸钠、柠檬酸等。
上述方法中,步骤(2)中所用的原料由于已经去除了可溶性蛋白,因此纯化螺旋藻多糖过程中不需要去除蛋白质。

上述方案中,采用热提法提取小球藻生长因子,再通过超滤纯化小球藻生长因子,冷冻干燥获得小球藻生长因子冻干粉;采用提取了藻蓝蛋白的螺旋藻渣热水提取螺旋藻多糖,由于提取藻蓝蛋白过程中去除了可溶性蛋白,因此提取多糖过程中不需要多糖和蛋白分离过程,显著降低了螺旋藻多糖制备成本;采用本方法可以经济、有效的工业化生产小球藻生长因子螺旋藻多糖功能饮品。

具体实施方式
为使本发明要解决的技术问题、技术方案和优点更加清楚,下面将结合附图及具体实施例进行详细描述。
本发明提供一种小球藻生长因子螺旋藻多糖功能饮品制备方法,其工艺流程如图1所示。
实施例1
(1)称取小球藻粉50克,加入至含有1200毫升去离子水的2L三角瓶中搅拌均匀,在高压灭菌锅中121℃条件下浸出40分钟。
(2)浸出液用冷冻离心机在4℃、离心力为8000g条件下离心15分钟,上清液即小球藻生长因子溶液。
(3)将小球藻生长因子溶液用截留分子量为10 000Da的超滤膜超滤,即得小球生长因子溶液。
(4)将小球藻生长因子溶液减压蒸馏至约1/4体积,浓缩液放在冰箱中-20℃冷冻,然后在冷冻干燥机中冷冻干燥即得小球藻生长因子冻干粉。
(5)称取提取藻蓝蛋白的螺旋藻渣80克,加入到含有1 000毫升去离子水的2升烧杯中搅拌均匀,在90℃、200rpm条件下搅拌浸出2小时。
(6)将冷却至室温的浸出液在4℃、离心力为8 000g条件下离心20分钟得到上清液。
(7)将上清液在50℃条件下减压蒸馏至约1/4体积。
(8)向浓缩液中加入约5倍体积的无水食用酒精,在4℃冷藏沉淀24小时。
(9)将螺旋藻多糖沉淀液在4℃、离心力为8 000g条件下离心20分钟,用无水食用酒精将螺旋藻多糖沉淀洗涤3次,然后离心获得脱色的螺旋藻多糖沉淀。
(10)将(9)得到的沉淀在冰箱中-20℃冷冻,然后在真空冷冻干燥机中冷冻干燥获得米白色的螺旋藻多糖冻干粉。
(11)称取小球藻生长因子冻干粉1mg、螺旋藻多糖冻干粉1mg、白砂糖16g、苯甲酸钠0.8mg、柠檬酸1.5g,加入2升去离子水配成小球藻精生长因子螺旋藻多糖功能饮品。
(12)配置的小球藻生长因子螺旋藻多糖功能饮品用0.22μm滤膜过滤除菌后封装。
实施例2
(1)称取小球藻粉60克,加入到含有1200毫升去离子水2L三角瓶中,搅拌均匀后在高压灭菌锅中115℃条件下浸出50分钟。
(2)浸出液用冷冻离心机在4℃、离心力为10 000g条件下离心20分钟,上清液即是小球藻生长因子溶液。
(3)将小球藻生长因子溶液用截留分子量为8 000Da的超滤膜超滤,即得小球藻生长因子溶液。
(4)将小球藻生长因子溶液减压蒸馏至约1/5体积,浓缩液放在冰箱中-20℃冷冻,然后在冷冻干燥机中冷冻干燥即得小球藻生长因子冻干粉。
(5)称取提取藻蓝蛋白的螺旋藻渣60克,加入到含有1 000毫升去离子水的2升烧杯中搅拌均匀,在80℃、200rpm条件下搅拌浸出1小时。
(6)将冷却至室温的浸出液在4℃、离心力为8 000g条件下离心20分钟得到上清液。
(7)将上清液在60℃条件下减压蒸馏至约1/4体积。
(8)向浓缩液中加入约5倍体积的无水食用酒精,在4℃冷藏沉淀24小时。
(9)将螺旋藻多糖沉淀液在4℃、离心力为8 000g条件下离心20分钟,用无水食用酒精将螺旋藻多糖沉淀洗涤3次,然后离心获得脱色的螺旋藻多糖沉淀。
(10)将(9)得到的沉淀在冰箱中-20℃冷冻,然后在真空冷冻干燥机中冷冻干燥获得米白色的螺旋藻多糖冻干粉。
(11)称取小球藻生长因子冻干粉0.8mg、螺旋藻多糖冻干粉0.9mg、白砂糖8g、苯甲酸钠0.6mg、柠檬酸0.8g,加入1升去离子水配成小球藻生长因子螺旋藻多糖功能饮品。
(12)配置的小球藻生长因子螺旋藻多糖功能饮品用0.22μm滤膜过滤除菌后封装。
实施例3
(1)称取小球藻粉30克,加入到含有1000毫升去离子水2L三角瓶中,搅拌均匀后在高压灭菌锅中121℃条件下浸出30分钟。
(2)浸出液用冷冻离心机在4℃、离心力为10 000g条件下离心20分钟,上清液即是小球藻生长因子溶液。
(3)将小球藻生长因子溶液用截留分子量为10 000Da的超滤膜超滤,即得小球藻生长因子溶液。
(4)将小球藻生长因子溶液减压蒸馏至约1/5体积,浓缩液放在冰箱中-20℃冷冻,然后在冷冻干燥机中冷冻干燥即得小球藻生长因子冻干粉。
(5)称取提取藻蓝蛋白的螺旋藻渣50克,加入到含有1 000毫升去离子水的2升烧杯中搅拌均匀,在80℃、200rpm条件下搅拌浸出1小时。
(6)将冷却至室温的浸出液在4℃、离心力为8 000g条件下离心20分钟得到上清液。
(7)将上清液在60℃条件下减压蒸馏至约1/4体积。
(8)向浓缩液中加入约5倍体积的无水食用酒精,在4℃冷藏沉淀24小时。
(9)将螺旋藻多糖沉淀液在4℃、离心力为8 000g条件下离心20分钟,用无水食用酒精将螺旋藻多糖沉淀洗涤3次,然后离心获得脱色的螺旋藻多糖沉淀。
(10)将(9)得到的沉淀在冰箱中-20℃冷冻,然后在真空冷冻干燥机中冷冻干燥获得米白色的螺旋藻多糖冻干粉。
(11)称取小球藻生长因子冻干粉0.6mg、螺旋藻多糖冻干粉0.8mg、苯甲酸钠0.4mg、白砂糖9g、柠檬酸1.0g,加入1升去离子水配成小球藻生长因子螺旋藻多糖功能饮品。
(12)配置的小球藻生长因子螺旋藻多糖功能饮品用0.22μm滤膜过滤除菌后封装。
以上所述是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明所述原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (5)
1.一种小球藻生长因子螺旋藻多糖功能饮品制备方法,其特征在于:包括如下步骤:
(1)小球藻生长因子制备:
A称取小球藻粉,加入去离子水,搅拌均匀后配成2%~5%的小球藻液,在110~121℃下加热提取20~60分钟,得到小球藻热提取物;
B将步骤A得到的小球藻热提取物冷却至室温,在4℃、6 000~10 000g下离心15~30分钟,得到小球藻生长因子溶液;
C将步骤B中得到的小球藻生长因子溶液超滤,超滤液冷冻干燥得到小球藻生长因子冻干粉;
(2)螺旋藻多糖制备:
D称取提取藻蓝蛋白的螺旋藻藻渣,将藻渣按照1:5~20的质量比加入去离子水混合均匀,在60~100℃、搅拌转速为100~400rpm条件下搅拌浸出2~4小时,得到浸出液;
E将步骤D中得到的浸出液在4℃、6 000~10 000g离心15~30分钟,得到的上清液在60~80℃减压蒸馏至1/4~1/5体积,得到浓缩液;
F向步骤E中得到的浓缩液中加入4~5倍体积的无水食用酒精,放在冰箱4℃下沉淀24小时,得到沉淀液;
G将步骤F中得到的沉淀液在4℃、6 000~10 000g离心15~30分钟,将得到的沉淀用无水食用酒精洗涤2~3次,然后离心获得米白色螺旋藻粗多糖沉淀;
H将步骤G中得到的螺旋藻粗多糖沉淀在-20℃冷冻,然后冷冻干燥获得米白色螺旋藻多糖冻干粉;
(3)小球藻生长因子螺旋藻多糖功能饮品制备:
称取步骤(1)制备的小球藻生长因子冻干粉和步骤(2)制备的螺旋藻多糖冻干粉加入去离子水中,制成小球藻生长因子螺旋藻多糖功能饮品,同时加入食品添加剂来调节功能饮品的口味,最后用0.22μm滤膜过滤除菌后封装。
2.根据权利要求1所述的小球藻生长因子螺旋藻多糖功能饮品制备方法,其特征在于:所述步骤C中小球藻生长因子溶液超滤的截留分子量为8 000~15 000Da。
3.根据权利要求1所述的小球藻生长因子螺旋藻多糖功能饮品制备方法,其特征在于:所述步骤A中采用高压锅或蒸汽加热的密闭式机械搅拌罐在110~121℃提取小球藻生长因子。
4.根据权利要求1所述的小球藻生长因子螺旋藻多糖功能饮品制备方法,其特征在于:所述步骤(3)小球藻生长因子螺旋藻多糖功能饮品中小球藻生长因子含量为0.6mg/L~1.0mg/L,螺旋藻多糖含量为1mg/L~2.0mg/L。
5.根据权利要求1所述的小球藻生长因子螺旋藻多糖功能饮品制备方法,其特征在于:所述步骤(3)中食品添加剂包括白砂糖、苯甲酸钠、柠檬酸。
