技术领域
本发明涉及一种含有虾青素的软胶囊,属于药物/食品制剂领域,具体涉及一种内容物中含有55%以上碳酸钙的软胶囊。
背景技术
虾青素(astaxanthin),又名虾黄质、龙虾壳色素,是一种类胡萝卜素,呈深粉红色,不溶于水,在酸、氧、高温及紫外条件下均不稳定,易氧化降解。人工合成虾青素的稳定性和抗氧化活性均比天然虾青素低。而天然虾青素来源中又以雨生红球藻中虾青素含量最高,红花籽油是自雨生红球藻中提取虾青素的优选提取溶剂。
虾青素具有多种生物功能,如抗氧化功能、保护视网膜、增强免疫力、抗癌、抗炎等,以前三种功能应用更为广泛。虾青素在骨健康方面的应用很少见到。其物理性质决定了它的主要服用形式,主要是软胶囊和原料微囊化处理后的其他口服形式,其中,微囊化的原料不但成本更高,因包合物的加入虾青素的含量又进一步降低。
软胶囊可直接包封油性溶液、混悬液、乳液或半固体物质,其中混悬液一般要求固形物含量小于20%,该范围内工艺相对成熟,固形物大于20%特别是35%以上时,容易出现装量不稳定、均一度不合格、成品率低等问题。软胶囊内容物分层也是比较常见的现象,特别碳酸钙,质重,更易分层。常规的解决方案是降低固形物的粒径、增加助悬剂、胶体磨乳化均质等手段,但这并不能解决所有种类及各种用量的固形物沉降分层问题。
发明内容
本发明所要解决的技术问题,是使含有55%以上碳酸钙的虾青素软胶囊的内容物体系更稳定、更均一。
本发明提供了一种含有虾青素的软胶囊,该软胶囊的内容物包括虾青素、碳酸钙、食用油和蜂蜡,重量份数依次为75-85份、295-320份、120-140份和12-15份,其中所述的虾青素为虾青素油;内容物的制备方法为:
1)蜂蜡加入食用油中,加热熔融,混匀,冷却至40-45℃,保温,得基质;
2)将虾青素油加入到碳酸钙中,混匀,浸润后加入到基质中,35℃保温,边加边搅拌至均匀。
虾青素对骨关节炎(OA)的治疗作用已被确认。有研究表明骨关节炎可以诱发骨质疏松症。骨关节炎患者骨丢失率增加是骨折的一个独立危险因素,而多种因素造成的骨关节炎患者骨密度的高估,往往会导致对骨折风险的忽视。而目前尚无兼顾两者的制剂。因此,提供一种治疗骨关节炎同时预防或缓解骨质疏松症的组合物非常必要。所以本发明提供了含有虾青素和碳酸钙的软胶囊剂,填补该项空白。但在研发过程中遇到很多困难。
碳酸钙因其含钙量高、安全性高、价格低廉,无论是在药品、保健食品还是普通食品(作为营养强化剂)领域中,都是应用最广的钙源。虽然碳酸钙在钙源中的含钙量高,但也仅有40%,且不溶于水也不溶于油。软胶囊需要吞服,为提升顺应性,单粒软胶囊最好控制在七八百毫克左右或更低,否则吞咽困难,服用后异物感很强,不适感会降低患者持续服用时间,从而影响疗效。在此前提下,若想达到补充钙质缓解骨质疏松症的目的,内容物的固形物比重便很高,达55%(内容物重量百分百)以上,以常规方法要制成含量均一、质量稳定的软胶囊剂非常困难。比如增加助悬剂用量,当蜂胶采用常规用量的4%时,流动性已无法满足灌装要求;而胶体磨、均质机对混悬液的均一性虽有改善,但主要起辅助功效,若内容物体系不稳定,则胶体磨、均质机的作用时间很有限。
目前市场上没有纯虾青素的原料售卖,均为提取物,以天然的雨生红球藻提取的质量最佳,主要有微囊粉和油状两种,本发明采用纯度及性价比均更高的虾青素油。该青素油是指雨生红球藻提取物的红花籽油溶液,其中虾青素的含量不低于5%。
蜂蜡是软胶囊剂中最为常用的助悬剂,通常的用量范围在3~6%,依据固形物的性质及用量不同,2~12%的范围内均有人尝试使用。但使用2-3%的低用量蜂蜡实现55%的固形物(碳酸钙)稳定混悬的技术方案和效果,未见任何报道或技术启示。
本发明的发明人为实现发明目的,分别对配方、工艺及其组合进行了大量的研究试验,在工艺筛选过程中,惊喜地发现,先用虾青素油将碳酸钙浸润一段时间,再加入到特定温度的基质中,并做保温处理(保温温度在35℃左右),便可仅用2.5%左右的蜂蜡实现约55%碳酸钙在基质中稳定、均一混悬的效果。其中,碳酸钙的浸润时间不少于5分钟。
蜂胶与食用油加热熔融后,通常冷却至室温或30℃左右,然后加入润湿剂、固形物等混合,从而起到助悬作用。本发明人试验中发现,在上述温度时加入碳酸钙分散非常困难,部分粉末结成小球,搅拌45分钟后碳酸钙的均一性仍不符合要求。随着加入时基质温度的提高,分散情况得到改善,但当基质温度达到50℃以上时,混悬稳定性下降。虾青素油浸润10分钟后的碳酸钙加入40-45℃的基质,混合20分钟即可,而且气泡少,灌装前过胶体磨或均质机所需时间短,35℃左右保温灌装时流动性良好,经长期(12个月)稳定性考察证实混悬的内容物稳定,无分层现象。而灌装内容物的温度偏低时,如30℃,流动性差,装量浮动过大,废丸率过高。
推测,有可能是因为40-45℃时蜂蜡尚未完全凝固,此时混入油浸润过的碳酸钙分散过程中逐渐冷却凝固,分散与凝固的速度适当,从而使碳酸钙混悬的状态稳定。而更高温度时,分散速度更快,相应的沉降速度也快,但凝固过程过长、不同部位冷却速度不同,蜂蜡的作用尚未完全发挥出来,导致混悬体现不稳定。
上述的虾青素油是指雨生红球藻提取物的红花籽油溶液,其中虾青素的含量不低于5%。
本发明所述的食用油为可食用油,包括:大豆油、橄榄油、玉米油,优选为大豆油。
在一优选实施例中,所述虾青素油、碳酸钙、食用油和蜂蜡的重量份数分别为80份、300份、137份和13份。
碳酸钙有轻质和重质两种,二者的堆密度和沉降体积有所区别,而这两点均对软胶囊内容物混悬液的流动性和沉降稳定性有影响,经过试验对比,本发明优选堆密度更大的重质碳酸钙。进一步优选400-600目的碳酸钙。
所述制备方法的步骤⑵为:将虾青素油加入到碳酸钙中,混匀,浸润10分钟。
为进一步提高虾青素的稳定性,本发明所述的软胶囊还含有有效剂量的稳定剂,如,维生素E。
本发明该软胶囊还可以含有其他剂量小的营养素,如精氨酸、维生素B6、维生素B12和
叶酸中的一种或组合,以实现不同的综合效果。
本发明所述软胶囊的囊皮包括重量比为1:0.4:1的明胶、甘油和水。在另一优选实施例中还含有遮蔽剂,在又一实施例中还有山梨醇。
发明人进一步通过试验来验证本发明的技术效果。
再次重申:以下实验只是本发明研制过程中众多实验中的举例性实验,并未涵盖和穷尽发明人为本发明所做的所有实验,目的仅仅在于用那些数据来阐述不同配方及工艺对软胶囊内容物均一性及物理稳定性的影响。
试验一:基质温度的选择
内容物配方:虾青素油(百奥斯汀虾青素油)80份碳酸钙(重质,400目)300份
大豆油139.4份蜂蜡10.6份
实验方法:蜂蜡与大豆油一起加热至约70℃,搅拌溶解混合,冷却至55℃后加入虾青素油,混合,在表1所列不同温度时加入碳酸钙,边加边搅拌,加完继续搅拌45分钟。
1、分别于搅拌的第15、30、45分钟时在不同位置取样,测定碳酸钙含量;
2、取搅拌好的样品经离心机2000rmp离心3min,观察是否有分层现象;
3、取搅拌好样品,置倾斜45度角的玻璃上端,自然下流,观察流动性。

注:1、取样点发现有明显没混匀的碳酸钙时,不取样测定。
2、“-”代表无分层现象,“+”有分层现象
实验结果表明,通常选择的基质温度下,加入56.6%的碳酸钙很难混合均匀,流动性差。随着基质温度升高,碳酸钙的分散情况得到改善,但当基质温度达到50度以上时,混悬体系稳定性降低,离心后,上部明显变稀。
试验二:碳酸钙虾青素油浸润时间选择
内容物配方:虾青素油(百奥斯汀虾青素油)80份 碳酸钙(重质,400目)300份 大豆油137份 蜂蜡13份
实验方法:蜂蜡与大豆油一起加热至约70℃,搅拌溶解混合,冷却至45℃,保温备用;将虾青素油加入到碳酸钙中,混匀,放置不同时间,加入到上述基质中,35℃保温,边加边搅拌至均匀。
1、分别于搅拌的第15、30、45分钟时在不同位置取样,测定碳酸钙含量;
2、取搅拌好的样品经离心机2000rmp离心3min,观察是否有分层现象;
3、取搅拌好样品,置倾斜45度角的玻璃上端,自然下流,观察流动性。

上述实验结果,表明在特定基质温度基础上,选择用虾青素油浸润碳酸钙一段时间后在与基质混合的方法,对碳酸钙与基质的混合过程大大改善,流动性与混悬稳定性均良好。浸润5分钟即可显著提高碳酸钙的分散性,混合30分钟均匀度达到要求,浸润10分钟效率最高;该实验方法条件下的内容物混悬液物理稳定性高,流动性好。
除上述实验外,发明人还进行了大量的重复性实验,包括基质温度在40-45℃之间浮动,更换不同食用油的种类,配方中添加稳定剂等,均可实现本发明目的。丸重低、有效成分高,内容物的均一性及物理稳定性更好,在为骨关节炎患者治疗主要病灶的同时可预防并发症——骨折的发生。
具体实施方式
实施例1
虾青素软胶囊(1000粒)
内容物配方:虾青素油80g碳酸钙300g大豆油137g蜂蜡13g
工艺:1、蜂蜡加入大豆油中,加热70℃熔融,混匀,冷却至40℃,保温,得基质;
2、将虾青素油加入到碳酸钙中,混匀,浸润5分钟,加入到基质中,35℃保温,边加边搅拌至均匀,即得内容物。
3、取比例为1:0.4:1的明胶、甘油、水,按常规工艺制备囊材胶液、压制软胶囊。
实施例2
虾青素软胶囊内容物配方(1000粒):
虾青素油75g碳酸钙320g橄榄油120g蜂蜡15g
工艺:同实施例1。

虾青素油85g碳酸钙300g玉米油133g蜂蜡12g

虾青素油80g碳酸钙295g大豆油142g蜂蜡13g
工艺:同实施例1。
实施例5
虾青素软胶囊内容物配方(1000粒):
虾青素油80g 碳酸钙295g 大豆油142g 蜂蜡13g 精氨酸0.5mg 维生素B6 3mg 叶酸0.2mg
工艺:同实施例1,精氨酸、维生素B6、叶酸同碳酸钙一同浸润。
实施例6
虾青素软胶囊内容物配方(1000粒):
虾青素油75g 碳酸钙300g 橄榄油142g 蜂蜡13g
工艺:1、蜂蜡加入大豆油中,加热65℃熔融,混匀,冷却至45℃,保温,得基质;
2、将虾青素油加入到碳酸钙中,混匀,浸润10分钟,加入到基质中,34℃保温,边加边搅拌至均匀,即得内容物。
实施例7
虾青素软胶囊内容物配方(1000粒):
虾青素油80g 碳酸钙300g 大豆油125g 蜂蜡15g 维生素E 10g
工艺:1、蜂蜡加入大豆油中,加热65℃熔融,加入维生素E,混匀,冷却至42℃,保温,得基质;
2、将虾青素油加入到碳酸钙中,混匀,浸润15分钟,加入到基质中,36℃保温,边加边搅拌至均匀,即得内容物。
以上仅为本发明的部分实施例,旨在对本发明进行进一步地说明,并不对本发明保护范围加以限定。任何人对上述实施方案进行并不偏离本发明范畴和精神的改进和变化,均应涵盖在本发明权利保护范围内。
Claims (9)
1.一种含有虾青素的软胶囊,该软胶囊的内容物包括虾青素、碳酸钙、食用油和蜂蜡,重量份数依次为75-85份、295-320份、120-140份和12-15份,其中所述的虾青素为虾青素油,所述的碳酸钙为重质碳酸钙;内容物的制备方法为:
⑴蜂蜡加入食用油中,加热熔融,混匀,冷却至40-45℃,保温,得基质;
⑵将虾青素油加入到碳酸钙中,混匀,浸润不少于5分钟,然后加入到基质中,35℃保温,边加边搅拌至均匀。
2.如权利要求1所述的软胶囊,其特征在于,所述的虾青素油是指雨生红球藻提取物的红花籽油溶液。
3.如权利要求2所述的软胶囊,其特征在于,所述的虾青素油中虾青素的含量不低于5%。
4.如权利要求1所述的软胶囊,其特征在于,所述的食用油包括:大豆油、橄榄油、玉米油。
5.如权利要求1所述的软胶囊,其特征在于,所述虾青素油、碳酸钙、食用油和蜂蜡的重量份数分别为80份、300份、137份和13份。
6.如权利要求1所述的软胶囊,其特征在于,所述制备方法的步骤⑵为:将虾青素油加入到碳酸钙中,混匀,浸润10分钟。
7.如权利要求1所述的软胶囊,其特征在于,该软胶囊还含有稳定剂。
8.如权利要求1所述的软胶囊,其特征在于,该软胶囊还含有精氨酸、维生素B6、维生素B12和叶酸中的一种或组合。
9.如权利要求1所述的软胶囊,其特征在于,该软胶囊的囊皮包括重量比为1:0.4:1的明胶、甘油和水。
