技术背景
市场上的保健品大体可以分为一般保健食品、保健药品、保健化妆品、保健用品等。保健食品具有食品性质,如茶、酒、蜂制品、饮品、汤品、鲜汁、药膳等,具有色、香、形、质要求,一般在剂量上无要求;保健药品具有营养性、食物性天然药品性质,应配合治疗使用,有用法用量要求,目前带“健”字批号的药品;保健化妆品具有化妆品的性质,不仅有局部小修饰作用,且有透皮吸收、外用内效作用,如保健香水、霜膏、漱口水等;保健用品具有日常生活用品的性质,如健身器、按摩器、磁水器、健香袋、衣服鞋帽、垫毯等。随着大家健康保健意识的不断增强,保健食品市场也空前的火爆起来。火爆的市场中,各种产品的质量也是参差不齐。
稻米也叫稻或水稻是一种可食用的谷物,一年生草本植物,性喜温湿,中国南方俗称其为“稻谷”或“谷子”,脱壳的粮食是大米,煮熟后称米饭或白饭。稻谷是我国的主要粮食作物之一,具有悠久的种植历史,种植面积。稻米不仅是食粮,同时还可以作为酿酒、制造饴糖的原料。全世界有一半的人口食用它。主要在亚洲、欧洲南部和热带美洲及非洲部分地区。总产量占世界粮食作物产量第三位,低于玉米和小麦,但能维持较多人口的生活。
关于蛋白类保健品技术有很多方法,现有技术如申请公众号为CN 101683158 B的中国发明专利,公开了一种核桃蛋白肽保健品组合物,该发明方法中的保健品组合物由核桃蛋白肽、何首乌、枸杞、大枣、菟丝子、当归、甘草组合而成,保健品组合物制备简单,且有食疗效果,但是其组分价格昂贵,提高了保健品的成本,而且仅仅含有蛋白与中草药成分,无任何氨基酸、糖分、微量元素等营养物质,因此其推广价值较低。

本方法的目的在于提供一种基于稻米蛋白的保健品,该保健品中含有丰富的稻米蛋白,同时含有维生素、氨基酸、膳食纤维、糖分、中草药提取物、微量元素,可以综合补充人体的营养物质,其制备方法简单,适于规模化生产。

再酶解:将初步酶解液的pH调节为7.0,温度为58℃,待温度平衡后加入5000U/g的中性蛋白酶、5500U/g的胰蛋白酶、0.22份功能性多肽,充分搅拌,微波辅助水解,微波功率为100W,频率为24kHz,水解过程中保持pH值恒定,恒温水解反应45分钟,然后灭酶得水解液,取酶解液在6000r/min高速离心后再经喷雾干燥即得草鱼氨基酸粉;功能性多肽的氨基酸序列为:SCASRCKSRCRARRCRARRCGFFRARYFGKRCICKCFRC;功能性多肽具有活化蛋白酶活性的作用,可以提高蛋白酶的水解效率,通过微波的辅助作用,将鱼蛋白质进一步的水解为短蛋白、小肽与氨基酸等小分子物质,提高营养物质含量;通过分步水解降解苦味多肽,降低苦涩味的产生,几种不同作用方式的酶配合使用,可以产生协同促进作用,改善风味,从而得到品质良好的产品;
提取:称取新鲜稻米,烘干后粉碎,过230目筛,将稻米粉依次用去离子水、4% NaCl溶液、0.06mol/L NaOH 和70% 乙醇提取,分别制得清蛋白、球蛋白、谷蛋白和醇溶蛋白四种蛋白组分提取液,分别调pH 值至等电点4.10、4.30、4.80和4.62;待蛋白充分沉淀后以-52℃、15.0 Pa冷冻干燥得各蛋白组分;将稻米蛋白分步彻底地提取出来,为复配保健品做好准备;
复配:精确称取蜂蜜32份、膳食纤维24份、茯苓多糖2.5份、香菇多醣1.5份、小麦胚芽油4.0份、草鱼氨基酸粉6.5份、稻米清蛋白5.0份、稻米球蛋白5.5份、稻米谷蛋白6.0份、稻米醇溶蛋白4.0份、柑橘提取物0.6份、虫草提取物0.12份、多维2.0份、多矿2.5份、聚乳酸菌0.015份、植物甾醇0.2份混合均匀后低温干燥粉碎得基于稻米蛋白的保健品;保健品富含稻米蛋白、糖、膳食纤维、维生素、矿物质、益生菌等人体必需的营养物质,可以增强人体抗病抗菌能力,提高免疫能力、抗炎能力,同时还可以清除人体多余的自由基,还具有降血糖、降血脂、免疫调节、抗氧化、抗肿瘤、抗心律失常等食疗效果,提高人体新陈代谢、增加热量消耗、提高能量水平,是一种营养全面、易于吸收、保健效果显著的基于稻米蛋白的保健品。
本发明的操作步骤中的常规操作为本领域技术人员所熟知,在此不进行赘述。

1.基于稻米蛋白的保健品,其组成及其重量份为:蜂蜜30-35份、膳食纤维20-25份、茯苓多糖2.0-3.0份、香菇多醣1.0-2.0份、小麦胚芽油3.0-5.0份、草鱼氨基酸粉6.0-7.0份、稻米清蛋白4.0-6.0份、稻米球蛋白3.0-6.0份、稻米谷蛋白6.0-7.0份、稻米醇溶蛋白4.0-6.0份、柑橘提取物0.5-0.6份、虫草提取物0.1-0.12份、多维1.0-2.0份、多矿2.0-2.5份、聚乳酸菌0.01-0.015份、植物甾醇0.2-0.6份。
2.根据权利要求1所述的基于稻米蛋白的保健品,其特征在于:所述的保健品的制备方法包括:
1)初步酶解:酶解草鱼蛋白质脱脂粉;
2)再酶解:以蛋白酶进一步酶解;
3)提取:提取稻米蛋白;
4)复配:将营养组分复配后低温干燥得基于稻米蛋白的保健品。
3.根据权利要求2所述的基于稻米蛋白的保健品,其特征在于:所述的保健品的制备方法中的初步酶解步骤为:将100-120份草鱼蛋白质脱脂粉与150-200份蒸馏水混合成鱼蛋白匀浆,在鱼蛋白匀浆中加入3500-4000U/g复合风味蛋白酶,调节pH为6.0-7.0,微波辅助水解,水解过程中保持pH值恒定,在50-52℃条件下恒温水解20-25分钟,然后升温灭酶,获得初步酶解液。
4.根据权利要求3所述的基于稻米蛋白的保健品,其特征在于:所述的保健品的制备方法中的初步酶解步骤中的微波辅助水解的微波功率为100-120W,频率为20-30kHz。
5.根据权利要求2所述的基于稻米蛋白的保健品,其特征在于:所述的保健品的制备方法中的再酶解步骤为:将初步酶解液的pH调节为6.5-7.0,温度为55-58℃,待温度平衡后加入5000-5500U/g的中性蛋白酶、5000-6000U/g的胰蛋白酶、0.20-0.22份功能性多肽,充分搅拌,微波辅助水解,水解过程中保持pH值恒定,恒温水解反应30-45分钟,然后灭酶得水解液,取酶解液在6000-8000r/min高速离心后再经喷雾干燥即得草鱼氨基酸粉。
6.根据权利要求5所述的基于稻米蛋白的保健品,其特征在于:所述的保健品的制备方法中的再酶解步骤中的功能性多肽的氨基酸序列为:SCASRCKSRCRARRCRARRCGFFRARYFGKRCICKCFRC。
7. 根据权利要求2所述的基于稻米蛋白的保健品,其特征在于:所述的保健品的制备方法中的提取步骤为:称取新鲜稻米,烘干后粉碎,过200-230目筛,将稻米粉依次用去离子水、3-5% NaCl溶液、0.05-0.06mol/L NaOH 和65-70% 乙醇提取,分别制得清蛋白、球蛋白、谷蛋白和醇溶蛋白四种蛋白组分提取液,分别调pH 值至等电点4.08-4.10、4.3-4.32、4.80-4.82 和4.60-4.65;待蛋白充分沉淀后以-50~-55℃、13.3-133Pa冷冻干燥得各蛋白组分。
8.根据权利要求2所述的基于稻米蛋白的保健品,其特征在于:所述的保健品的制备方法中的复配步骤为:精确称取蜂蜜30-35份、膳食纤维20-25份、茯苓多糖2.0-3.0份、香菇多醣1.0-2.0份、小麦胚芽油3.0-5.0份、蒸馏水50-60份在40-45℃下混合均匀,然后加入草鱼氨基酸粉6.0-7.0份、稻米清蛋白4.0-6.0份、稻米球蛋白3.0-6.0份、稻米谷蛋白6.0-7.0份、稻米醇溶蛋白4.0-6.0份、柑橘提取物0.5-0.6份、虫草提取物0.1-0.12份、多维1.0-2.0份、多矿2.0-2.5份、聚乳酸菌0.01-0.015份、植物甾醇0.2-0.6份混合均匀后低温干燥粉碎得基于稻米蛋白的保健品。
9.根据权利要求8所述的基于稻米蛋白的保健品,其特征在于:所述的保健品的制备方法中的复配步骤中的多维为VA或VB1或VB2或VB6或VB12或VC或VD或VE或VK或泛酸钙或烟酸或叶酸或生物素或肌醇或微晶纤维素的至少五种的混合物,多矿为MgSO4·7H2O与CuSO4·5H2O与FeSO4·H2O与ZnSO4·H2O与MnSO4·H2O与CoCl2·H2O与Ca(IO3)2与Na2SeO3的混合物。
