Abstract
本发明提供了一种益生菌果蔬酵素饮料及其制备方法,通过将果蔬、四合一PPHR乳酸菌菌粉(包含副干酪乳杆菌Lactobacillus paracasei、植物乳杆菌L. plantarum、瑞士乳杆菌 L. helveticus、以及鼠李糖乳杆菌L. rhamnosus)混合、发酵,所得到之发酵产物即为益生菌果蔬酵素饮料。与现有技术相比,本发明可缩短发酵过程、简化发酵流程,并提高果蔬汁的营养价值,赋予果蔬汁较强的抗氧化性,拓展乳酸菌在食品特别是饮料中的应用范围。
Description
技术领域
本发明涉及乳酸菌和果蔬加技术领域,具体涉及一种益生菌果蔬酵素饮料及其制备方法。
背景技术
乳酸菌是一群革兰氏阳性菌,乳酸菌的各种菌株可用于制造发酵食品,包括牛奶、面包、蔬果和其他可食用的植物材料。乳酸菌的膳食和临床应用的优点已被广泛研究,食用乳酸菌对于维持人体肠道内菌群平衡、刺激肠道运动、改善肠道通畅、抑制肠道内有害菌群增殖、减少有害物质生成、缓解腹泻症状、治疗肠道功能紊乱等有显著效果。另外还可缓解乳糖不耐症,增强免疫力,预防生殖系统感染,促进蛋白质、单糖及钙、镁等营养物质的吸收,产生维生素B群等大量有益物质,并协助维持人类和动物的健康,故又称为益生菌。此外,乳酸菌已被用作保存食物的发酵剂,采用其下列优点:低pH值及发酵活性期间产生能抑制腐败菌的生长的发酵产物的作用。因此,乳酸菌已被广泛用于制备各种不同的食品,如奶酪、酸奶和其他发酵制品。
随着人们健康意识的不断增强,消费者趋向于购买营养、健康的食品。果蔬汁饮料具有营养丰富、味道鲜美的特点,含有天然抗氧化剂,能消除人体内有害的自由基。然而普通果蔬汁饮料随着储存时间增长,质量风味发生变化,需添加防腐剂等对人体造成伤害。市面上常见的果蔬汁大多采用压榨的方式制作,果蔬中的部分营养元素被破坏或者被浪费掉,且传统的果蔬酵素饮料工艺是将水果、蔬菜粉碎后,在无氧的条件下发酵,这种发酵方式对于发酵过程中参与发酵的菌种无法控制,而有些参与发酵的菌可能会产生对人体有害之物质,且发酵时间也较常。
发明内容
为解决上述缺陷,本发明的目的是提供一种益生菌果蔬酵素饮料,有效提高果蔬汁的营养价值,增加果蔬汁的抗氧化性,延缓果蔬汁变质,减少防腐剂之使用,拓展乳酸菌在食品特别是饮料中的应用范围。
本发明的另一目的是提供一种益生菌果蔬酵素饮料的制备方法。
为达到上述目的,本发明采用的技术方案是:一种益生菌果蔬酵素饮料,是通过果蔬、赤砂糖及四合一PPAR乳酸菌混合发酵而成,其中所述四合一PPAR乳酸菌包括以下按质量百分比的乳杆菌组成:
副干酪乳杆菌L.paracasei 30%~45%、
植物乳杆菌L.plantarum 5%~20%、
瑞士乳杆菌 L.helveticus 20%~40%、
鼠李糖乳杆菌L.rhamnosus 2%~10%。
优选的是,所述四合一PPAR乳酸菌包括以下按质量百分比的乳杆菌组成:
副干酪乳杆菌L.paracasei 42%、
植物乳杆菌L.plantarum 15%、
瑞士乳杆菌 L.helveticus 38%、
鼠李糖乳杆菌L.rhamnosus 5%。
一种益生菌果蔬酵素饮料的制备方法,其特征在于包括以下步骤:
1)选取成熟、无病害、无腐烂的新鲜果蔬,然后分别经水清洗、干燥、紫外线杀菌、粉碎后,进行发酵;
2)发酵时粉碎后的果蔬按600-1400Kg加入赤砂糖20-60Kg,以及四合一PPAR乳酸菌菌粉0.005-0.035g搅拌混合进行发酵,发酵温度保持在10-40°C之间,监测pH值变化与酒精浓度,当pH值在3.0~4.0,酒精度<0.5%时,发酵完成;
3)将完成发酵的发酵液体进行过滤,并充分的混合;最后将精滤后的发酵液按配比泵吸到静置纯化罐中,盖好罐盖进行纯化,使其充分发酵,每周监控一次发酵液的pH值、酒精度和乳酸菌总数,当酒精度<0.5%,乳酸菌总数≥1×106CFU/ml,pH值稳定在2.0~4.0之间不再变化时表明纯化完成,即得到益生菌果蔬酵素饮料。
优选的是,步骤1)中的所述新鲜果蔬包括水果和蔬菜,其中水果包含苹果、猕猴桃、刺梨、桑椹、红枣、山楂、樱桃、青梅、蓝莓、梨、木瓜、菠萝、香蕉、西番莲、枇杷、荔枝、番石榴、火龙果、葡萄柚、桔子、柳橙、山竹、芒果、草莓、柠檬、橙子、哈密瓜、甜瓜、西瓜、柚子、葡萄、酪梨、李子、水蜜桃、桃子、枣子、百香果、莲雾、杨桃及柿子;蔬菜包含萝卜、胡萝卜、莲藕、芹菜、茼蒿、菠菜、空心菜、大白菜、冬瓜、苦瓜、丝瓜、南瓜、黄瓜、西红柿、山药、生姜、牛蒡、竹笋、荸荠、芦笋、小白菜、芥菜、芥蓝、油菜、青江菜、金针菜、茄子、胡瓜、蒲瓜及辣椒。
步骤2)中的所述发酵温度保持在25-30°C之间。
步骤2)中的发酵时间为7-60天。
步骤2)中发酵时粉碎后的果蔬按1000 Kg加入赤砂糖40Kg,以及四合一PPAR乳酸菌菌粉0.02Kg搅拌混合进行发酵。
本发明将果蔬、四合一PPHR乳酸菌菌粉混合、发酵,所得到之发酵产物即为益生菌果蔬酵素饮料,并可缩短发酵过程、简化发酵流程,并提高果蔬汁的营养价值,赋予果蔬汁较强的抗氧化性;完成发酵时,乳酸菌总菌数≧1×106 CFU/mL,食用后增加肠道益生菌菌数;本发明制作过程中产生的超氧化物岐化酶(SOD)广泛存在于生物体内,含Cu、Zn、Mn、Fe的金属类酶,可做为自由基清除剂及防御生物体氧化损伤;发酵后第2周SOD活性较起始高出4倍。
本发明提供的四合一PPHR乳酸菌配方,配方内包含副干酪乳杆菌L. paracasei、植物乳杆菌L. plantarum、瑞士乳杆菌 L. helveticus、以及鼠李糖乳杆菌L. rhamnosus,在特定组成比例的情况下,可以发酵果蔬得到具有较佳抗氧化力之益生菌果蔬酵素饮料,且活菌数达到106 CFU/mL以上,此活菌数或其分泌的代谢产物可定殖于人体的肠道,有抑制肠道腐败细菌生长的作用,从而起到调节肠道菌群平衡、增强免疫力、延缓衰老等作用,对人体有多项有益功效,而其增强之抗氧化力可以消除人体内自由基,能提高身体的免疫功能,进而达到预防疾病、减缓细胞老化的目的。
本发明发酵用之果蔬,系经紫外线杀菌后,再进行发酵,用以避免无法控制发酵过程中参与发酵的菌种,具有比较高的抗氧化力;水果、蔬菜经益生菌发酵后,能够较大程度的保持营养元素,并利用益生菌的代谢作用,使营养元素更有利于人体吸收,且产生芳香风味和良好的质地,具有独特的风味。
与现有技术相比,本发明利用四合一PPHR乳酸菌菌粉制备益生菌果蔬酵素饮料,不仅有效提高果蔬汁的营养价值,增加果蔬汁的抗氧化性,延缓果蔬汁变质,减少防腐剂之使用,拓展乳酸菌在食品特别是饮料中的应用范围。

下面详细描述本发明的实施例,所述实施例的示例,旨在用于解释本发明,而不能理解为对本发明的限制。实施例中未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件或者按照产品说明书进行。所用试剂或仪器未注明生产厂商者,均为可以通过市购获得的常规广品。为让本发明的上述和其他目的、特征和优点能更明显易懂,下文仅以例示为目的,举出较佳实施例,并配合所附图式,作详细说明如下。发明所属技术领域中技术人员不违背本发明的技术原理及精神下,可轻易完成的改变或修饰,均属本发明所主张的范围。
实施例1:四合一PPHR乳酸菌菌粉制备:
挑选单一菌落L. paracasei接种至2 L培养瓶,培养基成分包含1%蛋白胨、1%牛肉膏、0.5%酵母抽提物、2%葡萄糖、0.1%吐温-80、0.2% K2HPO4、0.5% NaCl、0.2%柠檬酸三铵、0.02% MgSO4、0. 005% MnSO4,于35℃静置培养24 hr后,于200L发酵槽放大培养,于35℃培养24 hr后,再于2吨发酵槽放大培养,同样于35℃培养24-72 hr后,离心收取菌泥,于-40℃冷冻干燥即可得到L. paracasei菌粉。L. plantarum、 L. helveticus及L. rhamnosus菌粉的生产方式与L. paracasei菌粉相同,将四种菌粉以特定比例混和后,即为四合一PPHR乳酸菌菌粉。
实施例2:果蔬使用前处理方法
选取成熟、无病害、无腐烂的新鲜果蔬,然后分别经水清洗、干燥、紫外线杀菌、粉碎后,进行发酵。处理步骤分述如下:
原料验收:采购的水果(包含但不限于菠萝、葡萄、苹果、木瓜、柠檬、橙子、枇杷、梨、猕猴桃、西瓜)和蔬菜(包含但不限于萝卜、生姜、苦瓜、丝瓜、南瓜、黄瓜),进厂后由化验室取样,取样合格后入库。
清理:对采购的水果(包含但不限于菠萝、葡萄、苹果、木瓜、柠檬、橙子、枇杷、梨、猕猴桃、西瓜)和蔬菜(包含但不限于萝卜、生姜、苦瓜、丝瓜、南瓜、黄瓜)进行拣选,拣选完成后,在进行对水果和蔬菜进行清洗、风干和紫外线杀菌。
粉碎:完成清理的水果和蔬菜,再对这些水果和蔬菜进行粉碎处理。
实施例3:乳酸菌发酵果蔬:
发酵时将粉碎后的果蔬按每吨加入赤砂糖40Kg,以及四合一PPHR乳酸菌菌粉20g发酵,在25-30°C温度下,监测pH值变化与酒精浓度,当pH值在2.0~4.0,酒精度<0.5%时,为发酵完成。
将完成发酵的发酵液体进行过滤,并充分的混合;最后将精滤后的发酵液按配比泵吸到静置纯化罐中,盖好罐盖进行纯化,使其充分发酵,每周监控一次发酵液的pH值、酒精度和乳酸菌总数,当酒精度<0.5%,乳酸菌总数≥1×106CFU/ml,pH值稳定在2.0~4.0之间不再变化时为纯化完成,即得到益生菌果蔬酵素饮料。
实施例4:蔬果酵素抗氧化力测试之一:
依据中华人民共和国国家标准GB/T 5009.171-2003第一法修改的Marklund方法。超氧化物岐化酶(Superoxide dismutase,简称 SOD)广泛存在于生物体内的含Cu、Zn、Mn、Fe的金属类酶。可作为生物体内重要的自由基清除剂,亦可清除体内多余的超氧阴离子,在防御生物体氧化损伤方面起着重要作用。具体分析步骤如下:
1)澄清液体样品可取原液进行适当稀释后测定,混浊液体样品经4000 r/min离心15min后再取上清液适当稀释测定;
2)取适当稀释样品100 mL;
3)加Tris–HCl buffer(pH 7.4, 0.05M Tris–HCl buffer containing 1mM EDTA)2850ml;
4)再加入60 mM邻苯三酚(pyrogallol)50 ml,剧烈摇荡混均;
5)以分光亮度计于325 nm测量吸光值 (A),每隔30秒测量一次,纪录溶液5分钟吸亮度之变化值△A325;
单位时间内抑制邻苯三酚自动氧化速率达50%时之酵素量,定为一单位(U);样品SOD活性,以每mL所含超氧歧化酶活性单位表示(U/ml),再依所配制浓度计算样品中超氧歧化酶活性(U/mg)。

N:稀释倍数;
0.5:达50%之氧化速率。
四合一PPHR乳酸菌蔬果发酵液后期抗氧化测试,结果如图一所示。由结果可知蔬果发酵液SOD活性则于一个月内达最高,之后微幅下降并持平(见图1)。
实施例5:蔬果酵素抗氧化力测试之二:
本实施例是将四合一PPHR乳酸菌蔬果发酵液于起始至1个月发酵过程中,侦测抗氧化之变化。
其抗氧化针测方法同实施例4中中华人民共和国国家标准GB/T 5009.171-2003第一法修改的Marklund方法所侦测。
四合一PPHR乳酸菌蔬果发酵前其抗氧化测试,结果如图二所示。由结果可得知,约1周后四合一PPHR乳酸菌蔬果发酵液SOD为最高值,是起始的4倍左右(见图2)。

1.一种益生菌果蔬酵素饮料,其特征在于:是通过果蔬、赤砂糖及四合一PPAR乳酸菌混合发酵而成,其中所述四合一PPAR乳酸菌包括以下按质量百分比的乳杆菌组成:
副干酪乳杆菌L.paracasei 30%~45%、
植物乳杆菌L.plantarum 5%~20%、
瑞士乳杆菌 L.helveticus 20%~40%、
鼠李糖乳杆菌L.rhamnosus 2%~10%。
