Abstract
本发明公开了一种β‑胡萝卜素纳米级乳剂的制备工艺。将溶解好的水相物料和油相物料同时放料,分别瞬时控制水相和油相的流量,用管线剪切机在线混合剪切乳化、均质,得到纳米级乳剂。此制备工艺使物料的每个区域均能混合均匀,所得的乳剂平均粒径小于100nm、粒径均一、且具有更窄的粒径分布,稳定性强,在使用时可克服普通乳剂稳定性差、易分层的问题;此生产工艺中没有使用有机溶剂,安全性更高;同时,本发明提出的生产工艺可连续性生产,大大提高了生产效率。
Description
技术领域
本发明涉及食品添加剂及功能食品技术领域涉,具体涉及一种食品医药、保健品、化妆品行业中需要使用的一种β-胡萝卜素纳米级乳剂的制备工艺及应用。
背景技术
近年来,类胡萝卜素是国家批准使用的、应用最广泛的食品着色剂,一直处于供不应求的状态,尤其是天然β-胡萝卜素;同时,随着科学研究的深入和人们生活水平的提高,类胡萝卜素具有的许多生理功也逐步被认识和发现,如清除自由基、抗氧化、抗癌、提高机体免疫力、延缓衰老等,因此,它的应用领域也从食品着色逐步扩大到营养食品、保健品、药品、畜牧农业、化妆品等领域。
天然β-胡萝卜素不溶于水,是一种脂溶性色素,它对热稳定,对光和氧气十分敏感,不稳定且易氧化,因此,它非常适合油性产品的开发,却不能应用于水基食品,并且因其极差的稳定性使其在食品、医药等行业的应用受到了很大的限制,使应用范围较窄。例如,通常需要将含β-胡萝卜素的色素、香料、营养素、营养药品、治疗剂或抗氧化剂这样的水不混溶组分掺入到水基组合物中,这些组合物还需要在典型的食品和饮料运输、储藏以及使用条件下呈现出长时期的货架稳定性,由于β-胡萝卜素不溶于水和稳定性差的特性,而很难实施,目前粉剂产品和普通乳剂产品在使用时大都会出现上浮分层和黄色油圈等现象。然而,通过制剂化处理后可大大提高其对环境的适应能力,增强其稳定性。
纳米级乳剂构成胶体系统,基于其物理和化学特性,纳米乳剂属于微乳状液的组类。微乳状液是由被表面活性剂和共表面活性剂单层包围的脂质核组成的均质的、微米级微粒的水分散体。纳米乳剂的特征在于平均粒径(平均直径)小于200nm、经常是小于100nm,以及窄的单分散性粒径分布。它们通常是通过在表面活性剂的存在下使油相在水相中机械破裂而生产的。鉴于纳米级乳剂粒径小、窄的单分散性粒径分布的特点,其具有比普通乳剂更好的稳定性。
传统的β-胡萝卜素乳剂生产都是将油相加入到水相中,然后进行一定时间(10~60分钟)剪切乳化后再均质,传统工艺间歇式生产,效率低,能耗高,不利于大规模连续工业化生产;由于β-胡萝卜素的稳定性较差且乳剂比粉剂对稳定性要求更高的原因,通过传统方法生产的β-胡萝卜乳剂其粒径均一性差,粒径分布广很且难达到纳米级别,产品稳定性差;同时,传统工艺生产乳剂也会使用到一定量的有机溶剂,造成一定安全风险。
专利为98105436.6公开了一种β-胡萝卜素水包油型微滴乳状液,该乳状液的生产过程中没有采用有机溶剂,所用的乳化剂是无毒的,可用于食品、药物和化妆品中,但是生产工艺的局限使该乳状液粒径难以达到纳米级别。
专利为201210035294.2公开了一种类胡萝卜素乳液的生产工艺,该方法首先将油相和水相混合进行剪切乳化,然后再进行一定时间的多次高压均质得到乳液,此生产工艺过程中没有使用有机溶剂,保证了乳剂的安全性,但由于间歇式生产不利于大规模连续工业化生产,且该乳液粒径也未达到纳米级别。
专利为201110399523.4公开了一种无需高压均质的可食用纳米级β-胡萝卜素乳状液及其制备方法,该方法优化了传统上制备纳米乳状液的技术,但是该工艺过程中加入了一定量的有机溶剂,用于食品、药品和化妆品领域时存在一定的安全风险,且生产工艺也是间歇式,不利于大规模工厂生产。
专利为201380038299.3公开了一种类胡萝卜素水包油乳液的生产工艺,利用此生产工艺可以制备粒径小于150nm的乳液,但是此生产工艺为首先将油相和水相混合,然后预乳化一定时间,再进行高压微型流化床进一步乳化,此工艺操作繁琐,不易于进行大规模生产。
发明内容
针对现有技术中存在的技术问题,本发明的目的在于本发明的任务是提供一种在水中就分散稳定性好,对光、热、氧气稳定的β-胡萝卜素纳米级乳剂的制备工艺,本发明的生产工艺简单,工厂规模化生产时成本低;本发明的另一目的在于提供β-胡萝卜素纳米级乳剂的应用。
本发明的技术方案是这样实现的:
一种β-胡萝卜素纳米级乳剂的制备工艺,其特征在于:包括以下步骤:
(1)将10%~20%甘油、1%~25%改性大豆磷脂、1%~10%聚甘油脂肪酸酯、5%~8%高HLB的辅助乳化剂、40%~82.5%水按比例投到水相罐中,所述比例均为质量百分比,同时常压下升温至50~80℃,待用,此为水相物料;
(2)将5%~25%β-胡萝卜素、1%~10%蔗糖脂肪酸脂、5%~10%辛癸酸甘油酯、1%~20%司盘60、0.1%~1%油相抗氧化剂、54%-86.5%的果油或食用植物油按比例投到油相罐中,充分搅拌混合,所述比例均为质量百分比,同时常压升温至120~170℃,待用,此为油相物料;
(3)将溶解好的水相物料和油相物料同时放料,分别控制水相和油相的流量,按水相:油相=25:1~1:1(质量比)用管线剪切机在线混合、均质,得到粒径均一的纳米级β-胡萝卜素乳剂。
进一步地,所述步骤(1)中改性大豆磷脂中磷脂酰胆碱含量为90%~95%,所述比例为重量百分数。
进一步地,所述步骤(1)中高HLB的辅助乳化剂选自聚山梨酸酯80、聚氧脱水山梨醇单月桂酸酯、聚甘油-6-月桂酸酯、十甘油月桂酸酯、十甘油油酸酯中的一种或任意多种混合物。
进一步地,所述步骤(2)中油相抗氧化剂为维生素E、抗坏血酸棕榈酸酯、叔丁基对苯二酚(TBHQ)、2,6-二叔丁基-4-甲基苯酚(BHT)中的一种或或一种以上。
进一步地,所述步骤(2)中的植物油为葵花籽油、大豆油、棕榈油、玉米油、橄榄油或棉籽油中的一种或一种以上。
进一步地,所述步骤(3)中,均质的条件为均质温度35-50℃、均质压力40Mpa~70Mpa、均质1次。
进一步地,所述的乳剂为O/W型。
进一步地,所述纳米级乳剂具有小于100nm的平均粒径。
如上述的一种β-胡萝卜素纳米级乳剂的制备工艺制备的β-胡萝卜素纳米级乳剂,所述乳剂应用于食品、饮料、保健品、医药、化妆品中,作为着色剂、营养增补剂和抗氧化剂使用。
本发明的有益效果:(1)本发明公开的一种β-胡萝卜素纳米级乳剂的制备工艺,将溶解好的水相物料和油相物料同时放料,分别瞬时控制水相和油相的流量,用管线剪切机在线混合剪切乳化、均质得到的乳剂平均粒径小于100nm,且具有更窄的粒径分布;(2)此方法使物料的每个区域均能混合均匀,因此该乳剂粒径均一,稳定性强,在使用时可克服普通乳剂稳定性差、易分层的问题;(3)本发明的生产工艺不使用有机溶剂,无安全风险;(4)此方法可大规模连续生产,提高了生产效率。

具体实施方式
展示一下实例来具体说明β-胡萝卜素乳剂的生产工艺、粒径及稳定性特征,其中实施例1为采用传统工艺生产,实施例2~4为采用本发明的制备工艺生产,具体实施方式不应解释为限制本发明的范围。对本发明公开的内容可以同时从材料、方法和反应条件进行改进,所有这些改进,均应落入本发明的的精神和范围之内。
实施例1:传统工艺生产β-胡萝卜素乳剂
1、将10公斤的β-胡萝卜素(含量为96%)、大豆油75公斤、中链甘油三酸酯75公斤和维生素E 0.5公斤搅拌均匀成油相,加热至130~140℃使之溶解得到油相物料,待用;
2、将大豆磷脂50公斤、亚硫酸钠0.5公斤、甘油80公斤加入600L水中,常压下升温至70℃充分溶解,待用;
3、将油相加入到水相中,然后剪切乳化20分钟后,在50Mpa-70Mpa均质3次制成最终乳剂。
实施例2:本发明的一种β-胡萝卜素纳米乳剂的制备方法
(1)将140公斤的甘油、15公斤改性大豆磷脂、14.5公斤聚甘油脂肪酸酯、70公斤高HLB辅助乳化剂加入到1000公斤水中,常压下升温至70℃充分溶解,待用;
(2)称取10公斤的β-胡萝卜素(含量为96%)、8公斤蔗糖脂肪酸脂、10公斤辛癸酸甘油酯、20公斤司盘60、油相抗氧化剂维生素E 0.4公斤溶于151.6公斤葵花籽油中,加热至130~140℃时快速搅拌使之溶解,搅拌均匀得到油相物料,待用;
(3)将溶解好的水相物料和油相物料同时放料,分别控制水相和油相的流量,按水相:油相=6.2:1(质量比)用管线剪切机在线混合、均质,得到粒径均一的纳米级β-胡萝卜素乳剂。
实施例3:本发明的一种β-胡萝卜素纳米乳剂的制备方法
(1)将180公斤的甘油、45公斤改性大豆磷脂、15公斤聚甘油脂肪酸酯、75公斤高HLB辅助乳化剂加入到1000公斤水中,常压下升温至70℃充分溶解,待用;
(2)称取35公斤的β-胡萝卜素(含量为96%)、18公斤蔗糖脂肪酸脂、28公斤辛癸酸甘油酯、52公斤司盘60和油相抗氧化剂维生素E 0.8公斤溶于166公斤橄榄油中,加热至130~140℃时快速搅拌使之溶解,搅拌均匀得到油相物料,待用;
(3)将溶解好的水相物料和油相物料同时放料,分别控制水相和油相的流量,按水相:油相=4.4:1(质量比)用管线剪切机在线混合、均质,得到粒径均一的纳米级β-胡萝卜素乳剂。
实施例4:本发明的一种β-胡萝卜素纳米乳剂的制备方法
(1)将220公斤的甘油、150公斤改性大豆磷脂、45公斤聚甘油脂肪酸酯、100公斤高HLB辅助乳化剂加入到1000公斤水中,常压下升温至70℃充分溶解,待用;
(2)称取90公斤的β-胡萝卜素(含量为96%)、50公斤蔗糖脂肪酸脂、辛癸酸甘油酯、97公斤司盘60和油相抗氧化剂维生素E 2.2公斤溶于307公斤玉米油中,加热至130~140℃时快速搅拌使之溶解,搅拌均匀得到油相物料,待用;
(3)将溶解好的水相物料和油相物料同时放料,分别控制水相和油相的流量,按水相:油相=2.78:1(质量比)用管线剪切机在线混合、均质,得到粒径均一的纳米级β-胡萝卜素乳剂。
实验结果分析:β-胡萝卜素纳米乳剂的粒径及其稳定性
观察上述实施例1传统工艺制备的β-胡萝卜素乳剂,流动性较好,无分层、析油、析晶现象。通过光子相关光谱术、PCS评价,结果显示,此种方法得到的β-胡萝卜素乳剂粒径分布范围较宽,具体分布在100μm~600μm之间,如图1所示。将实施例1中的乳剂吸取适量,稀释后,显微镜下观察本品,形态图如图2所示。
观察上述实施例2~4中本发明工艺制备的β-胡萝卜素乳剂,3个实施例中乳剂流动性较好,无挂壁、分层、析油、析晶现象。通过光子相关光谱术、PCS评价,结果显示,此种方法得到的纳米乳剂具有非常一致和可靠的粒径和窄的微粒分布,如图3所示。并且这些特征在长时间和在不同温度下是稳定。将实施例2中的乳剂吸取适量,稀释后,显微镜下观察本品,形态图如图4所示。本发明的工艺生产β-胡萝卜素乳剂的质量评价数据如表1所示。

结果显示,与传统工艺相比,本发明生产工艺生产的纳米乳剂产品质量稳定,表明本发明的处方和制备工艺对于解决β-胡萝卜乳剂其粒径均一性差、粒径分布广很难达到纳米级别、产品稳定性差的技术问题。
Claims (9)
1.一种β-胡萝卜素纳米级乳剂的制备工艺,其特征在于:包括以下步骤:
(1)将10%~20%甘油、1%~25%改性大豆磷脂、1%~10%聚甘油脂肪酸酯、5%~8%高HLB的辅助乳化剂、40%~82.5%水按比例投到水相罐中,所述比例均为质量百分比,同时常压下升温至50~80℃,待用,此为水相物料;
(2)将5%~25%β-胡萝卜素、1%~10%蔗糖脂肪酸脂、5%-10%辛癸酸甘油酯、1%~20%司盘60、0.1%~1%油相抗氧化剂、54%-86.5%的果油或食用植物油按比例投到油相罐中,充分搅拌混合,所述比例均为质量百分比,同时常压升温至120~170℃,待用,此为油相物料;
(3)将溶解好的水相物料和油相物料同时放料,分别控制水相和油相的流量,按水相:油相=25:1~1:1(质量比)用管线剪切机在线混合、均质,得到粒径均一的纳米级β-胡萝卜素乳剂。
2.根据权利要求1所述的一种β-胡萝卜素纳米级乳剂的制备工艺,其特征在于:所述步骤(1)中改性大豆磷脂中磷脂酰胆碱含量为90%~95%,所述比例为质量百分数。
3.根据权利要求1所述的一种β-胡萝卜素纳米级乳剂的制备工艺,其特征在于:所述步骤(1)中高HLB的辅助乳化剂选自聚山梨酸酯80、聚氧脱水山梨醇单月桂酸酯、聚甘油-6-月桂酸酯、十甘油月桂酸酯、十甘油油酸酯中的一种或任意多种混合物。
4.根据权利要求1所述的一种β-胡萝卜素纳米级乳剂的制备工艺,其特征在于:所述步骤(2)中油相抗氧化剂为维生素E、抗坏血酸棕榈酸酯、叔丁基对苯二酚(TBHQ)、2,6-二叔丁基-4-甲基苯酚(BHT)中的一种或一种以上。
5.根据权利要求1所述的一种β-胡萝卜素纳米级乳剂的制备工艺,其特征在于:所述步骤(2)中的植物油为葵花籽油、大豆油、棕榈油、玉米油、橄榄油或棉籽油中的一种或一种以上。
6.根据权利要求1所述的一种β-胡萝卜素纳米级乳剂的制备工艺,其特征在于:所述步骤(3)中,均质的条件为均质温度35~50℃、均质压力40Mpa~70Mpa、均质1次。
7.根据权利要求1所述的一种β-胡萝卜素纳米级乳剂的制备工艺,其特征在于:所述的乳剂为O/W型。
8.根据权利要求1所述的一种β-胡萝卜素纳米级乳剂的制备工艺,其特征在于:所述纳米级乳剂具有小于100nm的平均粒径。
9.如权利要求1~8中所述的一种β-胡萝卜素纳米级乳剂的制备工艺制备的β-胡萝卜素纳米级乳剂,其特征在于,所述乳剂应用于食品、饮料、保健品、医药、化妆品中,作为着色剂、营养增补剂和抗氧化剂使用。
