Abstract
本发明公开了一种具有降血糖功效的固体饮料及其制备方法。以重量份计,所述固体饮料包括桑叶提取物10‑25份、桑椹提取物5‑15份、玉竹提取物5‑10份和茯苓提取物5‑15份,其中,所述桑叶提取物中1‑脱氧野尻霉素含量大于50%,所述桑椹提取物、玉竹提取物、茯苓提取物为醇提物。本发明还提供了所述固体饮料的制备方法。本发明中,桑叶、桑椹、茯苓和玉竹均为药食同源物质,几种物质在功效上叠加增效,在味道上又可达到互补和平衡,对糖尿病患者症状有显著的减缓作用,因而是食疗降血糖的创新产物,对治疗糖尿病起到积极作用,且食用安全放心。
Description
技术领域
本发明涉及固体饮料,具体的涉及一种具有降血糖功效的固体饮料及其制备方法。
背景技术
当血糖值高于正常范围即为高血糖;空腹血糖>6.1mmol/L,餐后两小时血糖>7.8mmol/L时,称为高血糖。高糖饮食会改变血液酸碱度,降低机体免疫力,影响体内脂肪消耗,造成脂肪堆积,最终导致动脉粥样硬化、糖尿病、心脑血管病、冠心病和微血管病变等不同程度的各种疾病。因此,控制高血糖势在必行。
桑叶含有多种功能性成分,某些成分的含量与鲜茶或某些蔬菜中的含量相当甚至更高。桑叶的功能性成分包括大量矿物质、维生素、食物纤维、氨基酸和黄酮类化合物,桑叶还含有另一种生物碱:1-脱氧野尻霉素(DNJ),具有特殊的功能。DNJ是所有哺乳动物α-葡萄糖苷酶的有效的抑制剂,它能通过降低碳水化合物的消化和葡萄糖的吸收来降低食后高血糖。由于这一特性,DNJ可用于治疗糖尿病及糖尿病并发症,肥胖症和相关的机能紊乱。
桑椹为桑科植物桑Morus alba L的干燥果穗,又名桑果。桑椹古来就是百姓常采用的一种利尿、保健、消暑的鲜果,在本草纲目中有记载其特殊功效。因桑树特殊的生长环境使桑果具有天然生长,无任何污染的特点,所以桑葚又被称为“民间圣果”。桑椹中含有脂肪酸,主要由亚油酸、硬脂肪酸及油酸组成,具有分解脂肪、降低血脂、防止血管硬化或动脉硬化、高血压等,特别有益于控制血糖。
玉竹又名葳蕤,乃百合科多年生草本植物,耐寒、耐阴湿,忌强光直射与多风。玉竹味美甘甜,玉竹多糖是玉竹中含量最高的成分,经研究表明,玉竹多糖具有明显的降血糖作用,同时玉竹还有抗衰老,润燥润肺,镇静神经,治疗上火,改善睡眠质量,增强身体免疫力等作用。
茯苓是一种寄生于松科植物赤松或马尾松等树根上的菌种,为多孔菌科植物茯苓的干燥菌核,是一种药食同源物质。现代研究发现茯苓不仅有利尿、抗菌等药用价值,而且有很高的营养价值。茯苓干重中93%为β-茯苓聚糖,此外还有蛋白质、脂肪、卵磷脂、葡萄糖等成分。茯苓中膳食纤维含量相当丰富,占80.9%。研究证实,膳食纤维是糖尿病饮食控制中极为有利的一种营养素。因此,茯苓是糖尿病患者的食疗佳品。
公开号为CN103876228A的中国专利文献公开了一种降血糖玉米须饮料,它是由下述重量份的原料组成的:玉米须100-107、杭白菊30-40、山楂16-20、陈皮35-40、枸杞子35-40、甘草20-23、决明子10-13、罗汉果40-50、荞麦10-20、南瓜20-30、桑叶10-20、桑甚汁10-12、黄皮叶1-2、甜菊叶1-2、葛花1-2、竹茹2-3。但是其存在着原料复杂、工艺复杂、降血糖效果不理想等问题。
发明内容
发明目的:为克服现有技术的缺陷,本发明提供了一种具有降血糖功效的固体饮料,桑叶、桑椹、茯苓和玉竹均为药食同源物质,几种物质在功效上叠加增效,在味道上又可达到互补和平衡,对糖尿病患者症状有显著的减缓作用,因而是食疗降血糖的创新产物,对治疗糖尿病起到积极作用,且食用安全放心。本发明还提供了其制备方法。
技术方案:本发明所述的具有降血糖功效的固体饮料,以重量份计,包括:
其中,所述桑叶提取物中1-脱氧野尻霉素(DNJ)含量大于50%,所述桑椹提取物、玉竹提取物、茯苓提取物为醇提物。
所述桑叶提取物的制备方法包括:
(1)桑叶粉碎后用6-10倍重量的沸水提取2-3次,每次提取时间为1-3h,提取结束后,合并提取液,浓缩干燥后得桑叶水提物;
(2)用4-8倍重量的80-90%乙醇于75-80℃提取桑叶水提物提取2-3次,合并提取液,得到1-脱氧野尻霉素提取液;
(3)对所述的1-脱氧野尻霉素提取液进行纯化、浓缩,得到所述的桑叶提取物。
步骤(3)包括:1-脱氧野尻霉素提取液过酸性氧化铝柱,用20-30%乙醇洗脱,收集洗脱液,干燥得到粉末;将粉末溶于甲醇,采用丙酮进行重结晶,得到含DNJ含量大于50%的桑叶提取物。
具体的,所述桑叶提取物的制备方法为:将桑叶粉碎成粗粉,第一次用8倍重量的沸水提取1.5h,第二次用6倍重量的沸水提取1h,合并两次提取液,减压、浓缩、干燥得到桑叶水提物;再用6倍重量的90%乙醇80℃提取桑叶水提物2h,提取两次,合并提取液,得到1-脱氧野尻霉素提取液;提取液过酸性氧化铝柱,用20-30%乙醇洗脱,收集洗脱液,干燥得到粉末;纯化后的粉末溶于甲醇,采用丙酮进行重结晶,得到含DNJ含量大于50%的桑叶提取物。
所述桑椹提取物、玉竹提取物、茯苓提取物的制备方法包括:将桑椹、玉竹、茯苓分别用8-10倍重量的70-75%乙醇在75-80℃温度下回流提取2-3次,每次提取时间为1-1.5h,合并提取液,回收乙醇,减压浓缩,水洗,干燥,粉碎,最终得到对应的桑椹提取物、玉竹提取物和茯苓提取物。
具体的,所述桑椹提取物、玉竹提取物、茯苓提取物的制备方法包括:将桑椹、玉竹、茯苓分别用10倍重量的75%乙醇在80℃温度下回流提取两次,第一次提取1.5h,第二次提取1h;合并两次提取液,回收乙醇,减压浓缩,3倍重量水水洗,干燥,粉碎,最终得到除桑叶提取物之外的桑椹提取物、玉竹提取物和茯苓提取物。
优选的,所述的具有降血糖功效的固体饮料,以重量份计,包括:
本发明的固体饮料还包括30-50重量份的填充剂,填充剂的含量优选为30-45份。
所述的填充剂为膳食纤维、麦芽糊精、变性淀粉和植脂末中的一种或几种。
本发明还提供了所述固体饮料的制备方法,包括:按照配比称取各组分,混合均匀,灭菌,按规定称量包装。
与现有技术相比,本发明的有益效果为:
本发明具有降血糖功效的固体饮料配比合理,功效突出,风味俱佳,活性成分包括桑叶提取物、桑椹提取物、玉竹提取物和茯苓提取物,与公开号为CN103876228A的专利相比,其原料成分大大减少,原料处理工艺较为简单,但效果上桑叶提取物脱氧野尻霉素是一种高效的α-葡糖苷酶抑制剂,对糖尿病、肿瘤、病毒感染等显示出良好的生物活性;玉竹多糖具有明显的降血糖作用,降血糖功效突出;桑椹中含有的脂肪酸具有分解脂肪、降低血脂、防止血管硬化或动脉硬化、高血压等,特别有益于控制血糖,并且桑椹口感酸甜,形成独特的风味;茯苓中的膳食纤维对糖尿病饮食控制极为有利;桑叶、桑椹、玉竹、茯苓共同产生协同作用、增强作用,降血糖的效果更为明显,且本发明的饮料原料均为药食同源,不仅有良好的降血糖功效,口感风味好,而且食用安全放心。

下面结合具体实施例,进一步阐明本发明,应理解这些实施例仅用于说明本发明而不用于限制本发明的范围,在阅读了本发明之后,本领域技术人员对本发明的各种等价形式的修改均落于本申请所附权利要求所限定的范围。
实施例1-5:
一种具有降血糖功效的固体饮料,以质量份数计算,表1为制备原料配方组成:

上述实施例1-5的一种降血糖固体饮料的制备方法,包括如下步骤:
(1)桑叶提取物的制备:将桑叶粉碎成粗粉,第一次用8倍重量的水提取1.5h,第二次用6倍重量的沸水提取1h,合并两次提取液,减压、浓缩、干燥得到桑叶水提物;再用6倍重量的90%乙醇80℃提取桑叶水提物,提取两次,每次提取2h,合并提取液,得到1-脱氧野尻霉素提取液;1-脱氧野尻霉素提取液过酸性氧化铝柱(使用与桑叶原料重量比1:2~1:5的酸性氧化铝填料,柱子径高比为1:6~1:12),用稀醇(20~35%乙醇)洗脱,收集洗脱液,干燥得到粉末;纯化后的粉末溶于甲醇,采用丙酮进行重结晶,得到含DNJ含量大于50%(质量分数)的桑叶提取物。
(2)除桑叶提取物之外的其他提取物的制备:
将桑椹、玉竹、茯苓分别用10倍重量的75%乙醇在80℃温度下回流提取两次,第一次提取1.5h,第二次提取1h;合并两次提取液,回收乙醇,减压浓缩,3倍重量水水洗,干燥,粉碎,最终得到除桑叶提取物之外的桑椹提取物、玉竹提取物和茯苓提取物。
(3)按照配比称取各组分,混合均匀,灭菌,按规定称量包装即可得到降血糖固体饮料。
实施例6
为了观察固体饮料的降血糖疗效,我们观察了该具有降血糖功效的固体饮料对小鼠血清血糖水平的影响。具体实验实施方法如下:健康雄性小鼠60只,实验前于实验室预饲养1周。随机选取8只作为空白对照组,其余小鼠采用单次高剂量(200mg·kg-1)腹腔注射四氧嘧啶生理盐水溶液,1周后,小鼠尾静脉微量取血测定血清血糖水平,血糖水平大于16.7mmol·L-1的小鼠视为高血糖模型造模成功。将高血糖模型成功的小鼠随机分成7组,每组8只,分别为:高血糖模型对照组、阳性对照组、实施例1—5给药组。模型成功后便开始灌胃给药,持续4周。空白对照组和高血糖模型对照组灌胃给予生理盐水,阳性对照组灌胃二甲双胍(50mg·kg-1),5个实施例给药组分别以相当于提取物(混合物)1.5g·kg-1的剂量灌胃给药,实验期间每隔4天记录各组小鼠进食量和进水量。最后一次给药后禁食12h,待糖耐受量实验结束后,麻醉小鼠,摘眼球取血,离心分离血清,记录实验数据。

通过表2结果表明,实施例1-5组与模型对照组相比,均有明显的降血糖功效,实施例3-5组与实施例2组对比,降血糖功效显著,说明桑叶提取物和玉竹提取物按照一定配比结合使用后,协同增效,对小鼠降低血糖的效果更加明显,并且实施例4配方中的配比降血糖效果最佳。
Claims (8)
1.一种具有降血糖功效的固体饮料,其特征在于,以重量份计,包括:
其中,所述桑叶提取物中1-脱氧野尻霉素含量大于50%,所述桑椹提取物、玉竹提取物、茯苓提取物为醇提物。
2.根据权利要求1所述的具有降血糖功效的固体饮料,其特征在于,所述桑叶提取物的制备方法包括:
(1)桑叶粉碎后用6-10倍重量的沸水提取2-3次,每次提取时间为1-3h,提取结束后,合并提取液,浓缩干燥后得桑叶水提物;
(2)用4-8倍重量的80-90%乙醇于75-80℃提取桑叶水提物提取2-3次,合并提取液,得到1-脱氧野尻霉素提取液;
(3)对所述的1-脱氧野尻霉素提取液进行纯化、浓缩,得到所述的桑叶提取物。
3.根据权利要求2所述的具有降血糖功效的固体饮料,其特征在于,步骤(3)包括:1-脱氧野尻霉素提取液过酸性氧化铝柱,用20-30%乙醇洗脱,收集洗脱液,干燥得到粉末;将粉末溶于甲醇,采用丙酮进行重结晶,得到含DNJ含量大于50%的桑叶提取物。
4.根据权利要求1所述的具有降血糖功效的固体饮料,其特征在于,所述桑椹提取物、玉竹提取物、茯苓提取物的制备方法包括:将桑椹、玉竹、茯苓分别用8-10倍重量的70-75%乙醇在75-80℃温度下回流提取2-3次,每次提取时间为1-1.5h,合并提取液,回收乙醇,减压浓缩,水洗,干燥,粉碎,最终得到对应的桑椹提取物、玉竹提取物和茯苓提取物。

8.根据权利要求1所述的具有降血糖功效的固体饮料的制备方法,其特征在于,包括:按照配比称取各组分,混合均匀,灭菌,按规定称量包装。
