Abstract

本发明提供了一种宫内节育器用吲哚美辛硅橡胶药条的去爆释方法,使用95%乙醇和纯化水,将硅橡胶药条置于95%乙醇溶剂里面浸泡适当的时间,将吲哚美辛硅橡胶药条表层的药粉和浅层的吲哚美辛药物去除后,再用纯化水将乙醇置换出来,建立吲哚美辛药物在硅橡胶缓释系统中的稳定释放通道,最终实现处理后的药条能够平稳缓慢释放吲哚美辛的效果,本发明方案设计新颖,操作安全简单,操作过程中不引入对人体有害的溶剂,经过去爆释处理的吲哚美辛硅橡胶药条的安全性和有效性都有明显地提高。

Description

一种宫内节育器用吲哚美辛硅橡胶药条的去爆释方法
技术领域
本发明技术涉及一种宫内节育器用吲哚美辛硅橡胶药条的去爆释方法,属于医疗器械领域。
背景技术
宫内节育器是一种安全、长效的避孕方式,应用较为广泛。现有的宫内节育器在放置初期一般会产生疼痛流血等情况,为缓解疼痛出血情况的产生,我国自80年代开始在宫内节育器上面加载吲哚美辛等消炎镇痛类药物,吲哚美辛的加入明显降低了疼痛出血率,效果明显。目前国内市场上很多宫内节育器都加载了吲哚美辛,加载模式大多采用将吲哚美辛加入到硅橡胶中硫化制成吲哚美辛硅橡胶药条,然后将药条加载到节育器上的方式进行。吲哚美辛硅橡胶药条靠在体内缓慢释放吲哚美辛药物而起作用。但是,应用中通过测试发现吲哚美辛硅橡胶药条在初期释放量很大,一般持续一周左右后释放量才逐渐降低并在以后相当长的时间内保持稳定,初期释放量很大的情况称为爆释效应。药物的起效是建立在一定浓度范围内的,药物浓度过量会对人体产生一定的毒害作用,因此去除吲哚美辛硅橡胶药条的爆释效应,实现吲哚美辛药物的缓慢稳定释放对提高含吲哚美辛宫内节育器的使用安全性和有效性意义重大。
发明内容
本发明的目的旨在提供一种宫内节育器用吲哚美辛硅橡胶药条的去爆释方法。
本发明的技术方案是:一种宫内节育器用吲哚美辛硅橡胶药条的去爆释方法,包括以下步骤:
所用的原材料:乙醇(纯度95%)、纯化水,步骤如下:
(1)按重量份1︰4的比例将吲哚美辛硅橡胶药条及95%的乙醇放入容器中浸泡,同时用搅拌机搅拌,选择适当的搅拌速度,保持所有的吲哚美辛硅橡胶药条处于悬浮状态,搅拌浸泡时间为7—8小时;
(2)搅拌浸泡完成后将吲哚美辛硅橡胶药条从95%乙醇中滤出,用纯化水冲洗3遍去除残余的95%乙醇,再用纯化水浸泡11—12小时,然后再用纯化水冲洗2-3遍,最后再加入纯化水搅拌清洗30-40分钟后再用纯化水冲洗2-3遍;
(3)将冲洗后的吲哚美辛硅橡胶药条滤出,沥干后在70-80℃条件下鼓风干燥3—4小时即可。
本发明技术的原理是基于吲哚美辛药物的物理性质,利用其在乙醇中微溶而在水中几乎不溶,同时乙醇和水具有互溶的特点进行设计。
吲哚美辛硅橡胶药条是一种用吲哚美辛药物和硅橡胶充分混炼混合后硫化制作而成的缓释系统,硅橡胶在硫化过程中相互交联,将吲哚美辛药物包裹其中,在溶解过程中位于缓释系统表面和浅层的吲哚美辛由于与溶剂的接触面积大而容易溶解并首先溶解,这部分药物溶解后在硅橡胶中形成微小的孔道,溶剂通过这些孔道进入缓释系统内部与药物接触溶解药物,实现药物的缓慢释放。药物的爆释效应就是由于未经处理的吲哚美辛硅橡胶药条进入体内初期,表层和浅层的吲哚美辛药物集中释放引起释放量过大,释放初期过后形成释放通道后,药物与溶剂的接触面基本稳定,释放量也趋于稳定。本发明去爆释的方法就是利用适当的溶剂将药条表面和浅层的吲哚美辛药物去除,形成释放通道后使药条进入体内后直接到达稳定释放的阶段。
具体原理是将硅橡胶药条置于95%乙醇溶剂(无水乙醇溶解性稍大,成本高)里面浸泡适当的时间,将吲哚美辛硅橡胶药条表层的药粉和浅层的吲哚美辛药物去除后,再用水将乙醇置换出来,建立吲哚美辛药物在硅橡胶缓释系统中的稳定释放通道,最后再通过烘干将多余的水分干燥去除,以便于药条的贮存和使用,最终实现处理后的药条能够平稳缓慢释放吲哚美辛的效果。选择纯化水浸泡的另一个原因是利用纯化水浸泡过程等同于模拟吲哚美辛硅橡胶药条在体内(子宫腔内存在宫腔液,大部分成分是水)的使用环境,使吲哚美辛硅橡胶药条中的吲哚美辛在硅橡胶缓释系统中有一个溶解再平衡,待含药条的节育器植入子宫后能继续进行释放,从而达到去除爆释实现平稳释放的效果。本发明方案设计新颖,操作安全简单,操作过程中不引入对人体有害的溶剂,经过去爆释处理的吲哚美辛硅橡胶药条的安全性和有效性都有明显地提高,与已有技术相比具有实质性特点和显著的进步。
下面结合附图和实施例对本发明进行详细地描述。

具体实施方式
将裁切好的吲哚美辛硅橡胶药条约1kg,加入95%的酒精4kg放入10L的容器中浸泡,同时采用搅拌机搅拌,搅拌速度在1600—1800转/min左右,保持所有的药条处于悬浮状态,避免有药条沉降在底部的情况出现,搅拌浸泡的时间约7—8小时,浸泡时容器应适当进行密封以避免乙醇挥发。药条与溶剂的重量份采用大约1︰4的固液比,其作用是因为吲哚美辛药物在乙醇中微溶,乙醇用量太少,药物溶解到一定程度后形成饱和溶液就不再溶解,不能将药条表面和浅层的吲哚美辛药物充分的溶解去除;乙醇用量过大,容易造成药物的过度溶解,吲哚美辛硅橡胶药条中的药物损失过大,影响药物含量,缩短体内的释放期,不利于药物发挥作用。在浸泡过程中采用边浸泡边搅拌的方式,其作用一是由于药条为固体,搅拌可以保证药条在乙醇溶剂中处于悬浮状态且与溶剂接触均匀,保证药条所有表面药物的去除效果(如果不搅拌,药条会沉底汇聚,致使药条过度集中而出现局部药物溶解度达到饱和,影响浸泡效果);二是搅拌可以加快药物在乙醇中的溶解速度,缩短浸泡时间。搅拌浸泡完成后将药条从乙醇中滤出,药条用纯化水冲洗3遍,其作用是将药条表面的大量残留乙醇去除。再用纯化水(5kg左右)浸泡11—12小时,其作用是将药条内部通道内的乙醇置换稀释。然后再用纯化水冲洗2-3遍,其作用是将表面存在的置换出来的低浓度的乙醇溶液去除。最后再加入纯化水搅拌清洗30—40分钟后再用纯化水冲洗2-3遍,其作用是再次将药条中残留的乙醇再次置换去除。最后将冲洗后的药条取出、沥干后放在不锈钢盘上,用鼓风干燥箱在70-80℃条件下鼓风干燥3—4个小时即可,其中温度不宜过高和过低,过高温度会影响吲哚美辛硅橡胶药条中药物的稳定性,过低的温度影响水分去除速度,同时该温度易于残留酒精的挥发。经过处理后药条可以实现吲哚美辛的平稳缓慢释放。处理前和处理后的药条吲哚美辛的体外释放曲线分别如图1和图2所示。

Claims (1)

1.一种宫内节育器用吲哚美辛硅橡胶药条的去爆释方法,其所用的原材料为:95%乙醇、纯化水,包括以下步骤:
(1)按重量份1︰4的比例将吲哚美辛硅橡胶药条及95%的乙醇放入容器中浸泡,同时用搅拌机搅拌,保持所有的吲哚美辛硅橡胶药条处于悬浮状态,搅拌浸泡时间为7—8小时;
(2)搅拌浸泡完成后将吲哚美辛硅橡胶药条从95%乙醇中滤出,用纯化水冲洗3遍去除残余的95%乙醇,再用纯化水浸泡11—12小时,然后再用纯化水冲洗2-3遍,最后再加入纯化水搅拌清洗30-40分钟后再用纯化水冲洗2-3遍;
(3)将冲洗后的吲哚美辛硅橡胶药条滤出,沥干后在70-80℃条件下鼓风干燥3—4小时即可。
