背景技术
现有技术中乳酸菌饮料提取方法,工艺粗糙、落后,杂质多,有效成分含量低,导致患者用量过大,不方便服用,严重影响了本品应用。
发明内容
发明目的:为了解决上述问题,本发明的目的在于提供一种乳酸菌饮料。
技术方案:本发明的目的是通过如下的方案实现的:
一种乳酸菌饮料,由下列原料药重量份配比提取制备而成:党参20g、黄芪15g、茯苓10g、山药6g、丹参8g、玉竹20g、当归30g、麦冬10g、瓜子金15g,制备方法为:取药材加药材10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置,过滤,滤液浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,使用硅胶精制,预先将选定的硅胶装入树脂柱中,同时将所述浓缩液经纯化水稀释后,通过反相硅胶柱,去掉液体中的杂质,收集全部流出液,加入低聚果糖和水,接种乳酸菌进行发酵;调配、过滤、灌装,得乳酸菌饮料

所述源自植物的乳酸菌包括乳秆菌属、链球菌属、双歧杆菌属、德氏乳杆菌、保加利亚乳杆菌、瑞士乳秆菌、嗜酸乳秆菌、干酪乳杆菌、短乳秆菌、发酵乳杆菌、乳酸链球菌、丁二酮乳酸链球菌、乳酪链球菌、嗜热乳链球菌、两歧双歧秆菌、长双歧杆菌、短双歧秆菌、婴儿双歧秆菌、青春双歧杆菌、乳酸片球菌、戊糖片球菌中单一菌种或其组合物。
所述乳酸菌饮料在制备抑制提高免疫力食品中的应用。
现有技术中,乳酸菌饮料原来提取方法,工艺粗糙、落后,杂质多,导致患者用量过大,不方便服用,本发明制备的阿魏酸含量增加,含量高。本发明中的浓缩设备结构简单、操作便捷,其设有震动板和震动器,而还设有刮板和与所述刮板相连接的伸缩气缸,可有效防止罐体底部结块,一定程度上可大大提高加热效率,有效解决了传统技术中药浓缩设备中容易出现底部结块而影响加热效率的技术不足,使用稳定性好且适用性强,药物的有效成分稳定不被破坏。

具体实施方式
以下通过实施例形式,对本发明的上述内容再作进一步的详细说明,但不应将此理解为本发明上述主题的范围仅限于以下的实例,凡基于本发明上述内容所实现的技术均属于本发明的范围。
实施例1
取党参20g、黄芪15g、茯苓10g、山药6g、丹参8g、玉竹20g、当归30g、麦冬10g、瓜子金15g,加药材10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置,过滤,滤液浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,使用硅胶精制,预先将选定的硅胶装入树脂柱中,同时将所述浓缩液经纯化水稀释后,通过反相硅胶柱,去掉液体中的杂质,收集全部流出液,加入低聚果糖和水,接种乳酸菌进行发酵;调配、过滤、灌装,得乳酸菌饮料。
所述乳酸菌饮料,所述硅胶精制方法为:预先将选定的硅胶装入树脂柱中,同时将所述浓缩液经纯化水稀释后,通过反相硅胶柱,去掉液体中的杂质,收集全部流出液。
所述乳酸菌饮料,所述硅胶比表面为300m2/g,孔容为0.90ml/g,所述稀释后浓缩液通过反相硅胶柱分离、洗脱,至出水无色止。
所述乳酸菌为双歧杆菌属。
见图1:所述乳酸菌饮料,上述三次浓缩使用装置,包括罐体1,设置在所述罐体1上的进料口2和设置在所述罐体1底部的出料管3,在所述出料管3上设有出料阀门4和过滤器5,在所述罐体1内设有搅拌轴6,在所述搅拌轴6上设有搅拌桨7,在所述罐体1的顶部设有与所述搅拌轴6相连接的搅拌电机8,在所述罐体1内的底壁设有震动腔9,在所述震动腔9内设有震动板10,在所述震动板10与所述震动腔9之间设有震动器11,通过震动器可以使震动板震动,从而可有效防止药物颗粒集结在震动板上,从而可防止药物颗粒在震动板上结块,在所述罐体1内的侧壁上设有凹腔12,在所述震动板10的顶面设有刮板13,在所述刮板13与所述凹腔12之间设有伸缩气缸14,可以根据需要控制伸缩气缸,使刮板在震动板的表面刮动,从而将部分集结在震动板表面的药物颗粒刮动,所述刮板13在所述伸缩气缸14的作用下实现对震动板10表面的刮擦,在所述震动板10的中心设有与所述出料管3相连通的出料口15。
在所述震动板10与所述震动腔9的连接处设有密封金属条16,在所述罐体上设有与所述震动器相连接的震动控制器17。
在所述刮板的底部设有金属刮丝层18,所述金属刮丝层抵压在所述震动板上,在所述罐体上设有与所述伸缩气缸相连接的伸缩控制按钮19。
在所述罐体内设有水份测试仪21,在所述出料管内设有冷却器20。
实施例2

实施例3

实施例4:本发明乳酸菌饮料中阿魏酸含量测定实验研究资料
1.1实验药物:本发明乳酸菌饮料:按实施例1-3方法制备。对照组为参照实施例1,采用普通浓缩方法制备,取上述方法制备得到的乳酸菌饮料。
1.2仪器:高效液相色谱仪系统包括高效液相色谱仪(Waters 515);P200高压泵;Waters2487紫外检测器及GJ605型高压六通进样阀;色谱柱为BDS HYPERSIL-C18(4.6mm×250mm,5μm);数据采集及处理采用HS色谱数据工作站V4.0+(杭州英谱科技)
1.3测定条件:照高效液相色谱法(通则0512)测定。色谱条件与系统适用性试验以十八烷基硅烷键合硅胶为填充剂;以甲醇-水(60:40)为流动相;检测波长为250nm;理论板数按阿魏酸峰计算应不低于2000。取阿魏酸对照品适量,精密称定,加甲醇制成每lm l含5ug的溶液,即得。供试品溶液的制备取装量差异项下的本品,研细,取约0.5g,精密称定,置25ml量瓶中,加甲醇20ml,超声处理(功率250W,频率40kHz)30分钟,放冷,加甲醇至刻度,摇匀,滤过,取续滤液,即得。分别精密吸取对照品溶液与供试品溶液各20ul注入液相色谱仪,测定,即得。
1.4实验结果

根据上表的试验结果可知,本发明实施例1-3制备的乳酸菌饮料中阿魏酸含量明显高于对照组。
实施例5:本发明提高免疫力的药效学研究
实验目的:用本发明实施例1,灌胃小鼠30d,比较本发明和正常组小鼠受ConA诱导的脾脏淋巴细胞增殖的差别,来判断本发明是否增强脾脏淋巴细胞增殖。
实验动物:昆明种小鼠,雄性,体重18-22g,上海斯莱克实验动物有限公司提供,生产许可证:SCXK(沪)2007-0005。
实验药物:按上述实施例1的制备方法制备。
实验步骤:小鼠,按体重分层随机分组,每组12只。如下表所示,正常组灌胃给水;盐酸左旋咪唑组灌胃盐酸左旋咪唑;实施例1灌胃2g·kg-1,各组连续灌胃给药30d。无菌取脾,按照常规制成单个细胞悬液,调整细胞浓度为5×105个/mL。将脾细胞混悬液加在24孔板中,每孔1mL,每只动物设二个复孔,一孔正常培养(加50uL PRMI1640培养液);一孔加50uL刀豆蛋白A(ConA)液(终浓度为4.76ug/mL)。置于5%CO2,37℃CO2孵箱中培养48h。实验结束前4h,每孔吸去上清液700uL,再加入700uL不含胎牛血清的PRMI1640培养液,同时每孔加入50uL 5mg/mL的MTT液。培养结束后,每孔加1mL酸性异丙醇,小心吹打均匀至紫色的结晶全部溶解。检测前分装到96孔培养板中,每孔作4个复孔,在570nm波长处测定其光密度值。
实验结果:
本发明对脾淋巴细胞增殖的影响
注:与正常组相比P<0.01,P<0.05。
结论:本发明实施例1小鼠脾脏淋巴细胞受ConA诱导后的增殖率转化值高于正常组小鼠,说明具有提高免疫力作用。
Claims (7)
1.一种乳酸菌饮料,其特征在于,由下列原料药重量份配比提取制备而成:党参20g、黄芪15g、茯苓10g、山药6g、丹参8g、玉竹20g、当归30g、麦冬10g、瓜子金15g,制备方法为:取药材加药材10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置,过滤,滤液浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,使用硅胶精制,预先将选定的硅胶装入树脂柱中,同时将所述浓缩液经纯化水稀释后,通过反相硅胶柱,去掉液体中的杂质,收集全部流出液,加入低聚果糖和水,接种乳酸菌进行发酵;调配、过滤、灌装,得乳酸菌饮料。
2.根据权利要求1所述乳酸菌饮料,其特征在于制备方法中所述硅胶比表面为300~500m2/g,孔容为0.70~0.90ml/g,所述稀释后浓缩液通过反相硅胶柱分离、洗脱,至出水无色止。
3.根据权利要求1所述乳酸菌饮料,其特征在于上述提取方法中浓缩使用装置,包括罐体,设置在所述罐体上的进料口和设置在所述罐体底部的出料管,在所述出料管上设有出料阀门和过滤器,在所述罐体内设有搅拌轴,在所述搅拌轴上设有搅拌桨,在所述罐体的顶部设有与所述搅拌轴相连接的搅拌电机,在所述罐体内的底壁设有震动腔,在所述震动腔内设有震动板,在所述震动板与所述震动腔之间设有震动器,在所述罐体内的侧壁上设有凹腔,在所述震动板的顶面设有刮板,在所述刮板与所述凹腔之间设有伸缩气缸,所述刮板在所述伸缩气缸的作用下实现对震动板表面的刮擦,在所述震动板的中心设有与所述出料管相连通的出料口。
4.根据权利要求3所述乳酸菌饮料,其特征在于上述提取方法中浓缩装置所述震动板与所述震动腔的连接处设有密封金属条,在所述罐体上设有与所述震动器相连接的震动控制器,浓缩装置所述刮板的底部设有金属刮丝层,所述金属刮丝层抵压在所述震动板上,在所述罐体上设有与所述伸缩气缸相连接的伸缩控制按钮,浓缩装置所述罐体内设有水份测试仪,在所述出料管内设有冷却器。
5.根据权利要求1所述乳酸菌饮料,其特征在于所述乳酸菌为源自植物的乳酸菌。
6.根据权利要求1所述乳酸菌饮料,其特征在于所述源自植物的乳酸菌包括乳秆菌属、链球菌属、双歧杆菌属、德氏乳杆菌、保加利亚乳杆菌、瑞士乳秆菌、嗜酸乳秆菌、干酪乳杆菌、短乳秆菌、发酵乳杆菌、乳酸链球菌、丁二酮乳酸链球菌、乳酪链球菌、嗜热乳链球菌、两歧双歧秆菌、长双歧杆菌、短双歧秆菌、婴儿双歧秆菌、青春双歧杆菌、乳酸片球菌、戊糖片球菌中单一菌种或其组合物。
7.根据权利要求1所述乳酸菌饮料在制备提高免疫力食品中的应用。
