Abstract
本发明提供一种中西药复方戒毒药,由中药提取物和西药组成,中药提取物的制备方法为:取石草鞋20g、海州常山18g、姜味草15g、南天竹根20g、延胡索28g、青风藤15g、当归10g、丹参15g、川芎20g、莪术10g、五加皮20g、柏子仁15g、全蝎10g、甘草5g,并与和美沙酮、洛非西丁配伍,制备成戒毒药效果良好。

本发明涉及中西药复方技术领域,具体涉及一种中西药复方戒毒药及其制备方法。
背景技术
现有技术中中药戒毒药提取方法,工艺粗糙、落后,杂质多,有效成分含量低,导致患者用量过大,不方便服用,严重影响了本品在临床上应用。
发明内容
发明目的:为了解决上述问题,本发明的目的在于提供一种中西药复方戒毒药及其制备方法。
技术方案:本发明的目的是通过如下的方案实现的:
一种中西药复方戒毒药,由中药提取物和西药组成,中药提取物的制备方法为:取石草鞋20g、海州常山18g、姜味草15g、南天竹根20g、延胡索28g、青风藤15g、当归10g、丹参15g、川芎20g、莪术10g、五加皮20g、柏子仁15g、全蝎10g、甘草5g,加药材10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置,过滤,滤液浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,使用硅胶精制,预先将选定的硅胶装入树脂柱中,同时将所述浓缩液经纯化水稀释后,通过反相硅胶柱,去掉液体中的杂质,收集全部流出液,继续浓缩,将浓缩液进行干燥,制粒,得中药提取物。
所述中西药复方戒毒药,制备方法中所述硅胶比表面为300~500m2/g,孔容为0.70~0.90ml/g,所述稀释后浓缩液通过反相硅胶柱分离、洗脱,至出水无色止。
所述中西药复方戒毒药,上述中药提取物的提取方法中浓缩使用装置,包括罐体,设置在所述罐体上的进料口和设置在所述罐体底部的出料管,在所述出料管上设有出料阀门和过滤器,在所述罐体内设有搅拌轴,在所述搅拌轴上设有搅拌桨,在所述罐体的顶部设有与所述搅拌轴相连接的搅拌电机,在所述罐体内的底壁设有震动腔,在所述震动腔内设有震动板,在所述震动板与所述震动腔之间设有震动器,在所述罐体内的侧壁上设有凹腔,在所述震动板的顶面设有刮板,在所述刮板与所述凹腔之间设有伸缩气缸,所述刮板在所述伸缩气缸的作用下实现对震动板表面的刮擦,在所述震动板的中心设有与所述出料管相连通的出料口。

石草鞋为萝藦科香花球兰Hoya lyi Levl.,以叶入药。活血祛瘀,祛风除湿。治跌打损伤,风湿关节痛。
海州常山,拉丁名为Clerodendrum trichotomum Thunb.根、茎、叶、花入药,祛除风湿,降血压。根、叶:风湿性关节炎,高血压病,痢疾,疟疾。叶:外用治手癣,水田皮炎,湿疹,痔疮。本发明以全草入药。
姜味草为姜味草Micromeria biflora Benth.的全草。春夏采收,洗净晒干。祛风解表,温中除湿。用于感冒咳嗽,胃痛,急性胃肠炎,消化不良,腹胀,腹痛,疝气痛,预防痢疾。
南天竹根为小檗科植物南天竹的根,祛风,清热,除湿,化痰。治风热头痛,肺热咳嗽,湿热黄疸,风湿痹痛,火眼,疮疡,瘰疬。其余中药均符合药典标准。
现有技术中,戒毒中药原来的提取方法,工艺粗糙、落后,杂质多,导致患者用量过大,不方便服用,本发明制备的延胡索乙素含量增加,含量高。本发明中的浓缩设备结构简单、操作便捷,其设有震动板和震动器,而还设有刮板和与所述刮板相连接的伸缩气缸,可有效防止罐体底部结块,一定程度上可大大提高加热效率,有效解决了传统技术中药浓缩设备中容易出现底部结块而影响加热效率的技术不足,使用稳定性好且适用性强,药物的有效成分稳定不被破坏。本干燥设备结构简单、操作简单快捷,其采用箱体侧壁进行送风,并增设了电加热块,可以根据箱内的温度进行调节,从而保证了干燥温度的稳定性,而且设有旋转电机和震动装置,可以在加热的同时对盛放板进行旋转和震动,从而可防止药局部干燥,从而一定程度上可以提高干燥的效率,解决了传统技术的干燥装置干燥效率低的技术不足。石草鞋、海州常山、姜味草、南天竹根活血祛瘀,祛风除湿为君药,延胡索、青风藤、当归、丹参、川芎、莪术、五加皮、柏子仁、全蝎清热解毒,扶正祛邪为臣药,甘草为佐使药,全方与西药配伍,相得益彰,戒毒效果好。

实施例1
取石草鞋20g、海州常山18g、姜味草15g、南天竹根20g、延胡索28g、青风藤15g、当归10g、丹参15g、川芎20g、莪术10g、五加皮20g、柏子仁15g、全蝎10g、甘草5g,加10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置,过滤,滤液浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,使用硅胶精制,得流出液,继续浓缩,将浓缩液进行干燥,制粒,得中药提取物。
所述中西药复方戒毒药,所述硅胶精制方法为:预先将选定的硅胶装入树脂柱中,同时将所述浓缩液经纯化水稀释后,通过反相硅胶柱,去掉液体中的杂质,收集全部流出液。
所述中西药复方戒毒药,所述硅胶比表面为300m2/g,孔容为0.90ml/g,所述稀释后浓缩液通过反相硅胶柱分离、洗脱,至出水无色止。

在所述震动板10与所述震动腔9的连接处设有密封金属条16,在所述罐体上设有与所述震动器相连接的震动控制器17。
在所述刮板的底部设有金属刮丝层18,所述金属刮丝层抵压在所述震动板上,在所述罐体上设有与所述伸缩气缸相连接的伸缩控制按钮19。
在所述罐体内设有水份测试仪21,在所述出料管内设有冷却器20。
见图2:所述中西药复方戒毒药,上述干燥使用装置,包括箱体1,设置在所述箱体1内的物料架2和设置在所述箱体1顶部的进风管3和出风管4,在所述箱体1的侧壁内设有与所述进风管3相连通的进风通道5,在所述箱体1的侧壁上设有与所述进风通道5相连通的风孔6,进风孔设置在侧壁上,可以保证通风的均匀性,而且可以大大提高与药物的接触面积,所述出风管4穿过所述箱体1的顶壁连通箱体1内与箱体1外部,在所述箱体1的侧壁内还设有调温腔7和灭菌腔8,在所述调温腔7内设有电加热块9,设有电加热块可以根据需要对进风管的温度进行调节,保证干燥温度的稳定性,所述进风通道5穿过所述调节腔7和灭菌腔8设置,在所述箱体1的底壁内设有旋转腔10,在所述旋转腔10内设有旋转电机11,所述旋转电机11的主轴与所述物料架2相连接,在所述物料架2内设有震动凹槽12,在所述物料架2的顶面设有盛放板13,在所述盛放板13与所述震动凹槽12之间设有震动装置14。
所述震动装置14包括驱动电机141,设置在所述驱动电机141主轴上的偏心轮142,所述偏心轮142的轮边抵压在所述盛放板13的底面中心,在所述盛放板13的底部与所述震动凹槽12的底壁之间设有拉簧15,所述拉簧以所述偏心轮为中心对称设置,在使用时通过偏心轮的抵压使盛放板上升,当偏心轮的凹部抵压盛放板时,盛放板在拉簧的作用下复位,在下落过程中使物材脱离盛放板之后再跌落在盛放板上实现物料的震动。
在所述箱体1内设有温度传感器16,在所述箱体的侧壁内设有与所述温度传感器相连接的驱动器17,所述驱动器与所述电加热块的控制端相连接。温度传干器和驱动器为市场上已有的现有技术,其在使用连接时只需要按照对应的连接说明连接即可。
在所述箱体上还设有箱门18。
取上述方法制备得到的中药提取物、美沙酮、洛非西丁的重量比为12:3:4,按常规方法制备成片剂。
实施例2

实施例4:按实施例1中制备不添加美沙酮的中西药复方戒毒药。

实施例6:本发明中西药复方戒毒药中延胡索乙素含量测定实验研究资料
1.1实验药物:本发明中西药复方戒毒药:按实施例1-3方法制备。对照组为中药提取物采用普通浓缩和干燥方法制备,取上述方法制备得到的中药提取物、美沙酮、洛非西丁的重量比为12:3:4,按常规方法制备成片剂。
1.2仪器:高效液相色谱仪系统包括高效液相色谱仪(Waters 515);P200高压泵;Waters 2487紫外检测器及GJ605型高压六通进样阀;色谱柱为BDS HYPERSIL-C18(4.6mm×250mm,5μm);数据采集及处理采用HS色谱数据工作站V4.0+(杭州英谱科技)
1.3测定条件:照高效液相色谱法(通则0512)测定。色谱条件与系统适用性试验以十八烷基硅烷键合硅胶为填充剂;以甲醇-水(60:40)为流动相;检测波长为250nm;理论板数按延胡索乙素峰计算应不低于2000。取延胡索乙素对照品适量,精密称定,加甲醇制成每lm l含5ug的溶液,即得。供试品溶液的制备取装量差异项下的本品,研细,取约0.5g,精密称定,置25ml量瓶中,加甲醇20ml,超声处理(功率250W,频率40kHz)30分钟,放冷,加甲醇至刻度,摇匀,滤过,取续滤液,即得。分别精密吸取对照品溶液与供试品溶液各20ul注入液相色谱仪,测定,即得。

根据上表的试验结果可知,本发明实施例1-3制备的中西药复方戒毒药中延胡索乙素含量明显高于对照组。
实施例7:本发明抑制海洛因依赖小白鼠跳跃反应的实验研究
试验材料:按制备实施例1、4、5方法制备得到中西药复方戒毒药。海洛因,山东省滨州市公安局提供,纯度为90.1%。盐酸纳洛酮,北京四环制药厂,批号:20130802。
实验动物:昆明种小鼠,雌雄各半,体重20±2g,由北京维通利华实验动物技术有限公司心提供。
实验方法:小鼠随机分成4组,每组12只,各组均皮下注射海洛因成瘾,按剂量递增法从6.25mg/kg开始给小鼠给药,每天两次,每次递增3.75mg/kg,连续给药5天,末次注射后3小时,用盐酸纳洛酮5mg/kg催瘾,跳跃发生率达100%,证明造模成功,继续进行海洛因成瘾实验,在末次注射海洛因1小时后,海洛因组灌胃生理盐水,其他三组分别灌胃实施例1、4、5,剂量为6g/kg,在灌胃给药2小时后,即末次注射海洛因3小时后,腹腔注射盐酸纳洛酮20mg/kg催瘾,立即将小白鼠放入一直径为10cm,高40cm的量筒中,观察10分钟内小白鼠的跳跃反应,所有组在注射纳洛酮前及注射1小时候后称体重,观察催瘾前后体重变化。

2.根据权利要求1所述中西药复方戒毒药,其特征在于中药提取物的制备方法中所述硅胶比表面为300~500m2/g,孔容为0.70~0.90ml/g,所述稀释后浓缩液通过反相硅胶柱分离、洗脱,至出水无色止。
3.根据权利要求1所述中西药复方戒毒药,其特征在于上述中药提取物的提取方法中浓缩使用装置,包括罐体,设置在所述罐体上的进料口和设置在所述罐体底部的出料管,在所述出料管上设有出料阀门和过滤器,在所述罐体内设有搅拌轴,在所述搅拌轴上设有搅拌桨,在所述罐体的顶部设有与所述搅拌轴相连接的搅拌电机,在所述罐体内的底壁设有震动腔,在所述震动腔内设有震动板,在所述震动板与所述震动腔之间设有震动器,在所述罐体内的侧壁上设有凹腔,在所述震动板的顶面设有刮板,在所述刮板与所述凹腔之间设有伸缩气缸,所述刮板在所述伸缩气缸的作用下实现对震动板表面的刮擦,在所述震动板的中心设有与所述出料管相连通的出料口。
4.根据权利要求3所述中西药复方戒毒药,其特征在于上述中药提取物的提取方法中浓缩装置所述震动板与所述震动腔的连接处设有密封金属条,在所述罐体上设有与所述震动器相连接的震动控制器,浓缩装置所述刮板的底部设有金属刮丝层,所述金属刮丝层抵压在所述震动板上,在所述罐体上设有与所述伸缩气缸相连接的伸缩控制按钮,浓缩装置所述罐体内设有水份测试仪,在所述出料管内设有冷却器。
5.根据权利要求1所述中西药复方戒毒药,其特征在于上述中药提取物的提取方法中干燥使用装置,包括箱体,设置在所述箱体内的物料架和设置在所述箱体顶部的进风管和出风管,其特征在于:在所述箱体的侧壁内设有与所述进风管相连通的进风通道,在所述箱体的侧壁上设有与所述进风通道相连通的风孔,所述出风管穿过所述箱体的顶壁连通箱体内与箱体外部,在所述箱体的侧壁内还设有调温腔和灭菌腔,在所述调温腔内设有电加热块,所述进风通道穿过所述调节腔和灭菌腔设置,在所述箱体的底壁内设有旋转腔,在所述旋转腔内设有旋转电机,所述旋转电机的主轴与所述物料架相连接,在所述物料架内设有震动凹槽,在所述物料架的顶面设有盛放板,在所述盛放板与所述震动凹槽之间设有震动装置。
6.根据权利要求5所述中西药复方戒毒药,其特征在于上述中药提取物的提取方法中干燥使用装置所述震动装置包括驱动电机,设置在所述驱动电机主轴上的偏心轮,所述偏心轮的轮边抵压在所述盛放板的底面中心,在所述盛放板的底部与所述震动凹槽的底壁之间设有拉簧,所述拉簧以所述偏心轮为中心对称设置。
7.根据权利要求5所述中西药复方戒毒药,其特征在于上述中药提取物的提取方法中干燥使用装置所述箱体内设有温度传感器,在所述箱体的侧壁内设有与所述温度传感器相连接的驱动器,所述驱动器与所述电加热块的控制端相连接,在所述箱体上还设有箱门。
8.根据权利要求1所述中西药复方戒毒药,其特征在于由权利要求1制备得到的中药提取物和美沙酮、洛非西丁组成。
9.根据权利要求8所述中西药复方戒毒药,其特征在于中药提取物和美沙酮、洛非西丁的重量比为10-15:1-4:2-5。
10.根据权利要求1所述中西药复方戒毒药在制备戒毒药中的应用。
