Abstract
本发明公开了一种双金胃疡制剂的制作方法,所述双金胃疡制剂主要由雪胆、金荞麦、大血藤、紫珠、麻布袋、延胡索、仙鹤草、白及、凤凰衣、土木香及核桃仁制成。本发明根据双金胃疡制剂组方中各药物的性质,以及该药物在组方中所起的作用,对双金胃疡制剂的具体制作工艺及参数进行了改进,使得雪胆的有效成分雪胆甲素容易测出,且测出的雪胆甲素量高,使得本药物更加精炼,药效更加显著,临床疗效好。

技术领域
本发明属于中药领域,具体涉及一种双金胃疡制剂的制作方法。
背景技术
中药提取是中药制药中常用的一种方法,中药化学成份极其复杂,需从复杂的混合物中提取对人体有效的中药活性成分。以提高药效。
双金胃疡制剂主要由雪胆、金荞麦、大血藤、紫珠、麻布袋、延胡索、仙鹤草、白及、凤凰衣、土木香及核桃仁制成;具有舒肝理气,健胃止痛,收敛止血的功效,用于肝胃气滞血淤所致的胃脘刺痛,呕吐吞酸,脘腹胀痛,胃及十二指肠溃疡见上述症候者。
现有技术中,双金胃疡制剂是这样制作的:取原料中的所有药材,粉碎成细粉,混匀,装入胶囊,即得。但是发明人在研究过程中发现,采用现有提取工艺制成双金胃疡制剂中,药物中的有效成分不易测出,尤其是雪胆中的有效成分雪胆甲素不易测出,使得双金胃疡制剂的临床疗效还不够理想。
发明内容
本发明提供了一种双金胃疡制剂的制作方法。本发明根据双金胃疡制剂组方中各药物的性质,以及该药物在组方中所起的作用,对双金胃疡制剂的具体制作工艺及参数进行了改进,使得雪胆的有效成分雪胆甲素容易测出,且测出的雪胆甲素量高,使得本药物更加精炼,药效更加显著,临床疗效好。
为了实现上述目的,本发明提供如下技术方案:
一种双金胃疡制剂的制作方法,所述双金胃疡制剂按重量份计算,主要由雪胆32-52份、金荞麦22-42份、大血藤22-42份、紫珠15-26份、麻布袋22-42份、延胡索22-42份、仙鹤草22-42份、白及55-70份、凤凰衣32-52份、土木香22-42份及核桃仁30-50份制成;所述双金胃疡制剂的制作方法为:取雪胆,加水煎煮后,过滤,滤液浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加醇提取,回收乙醇至净,提取液与上述浓缩液混合,浓缩成浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,冷水浸泡后,再温浸2-4次,过滤,合并滤液备用,药渣加入乙醇溶液,用盐酸调pH,浸泡后,回流提取,过滤,得提取液,提取液与上述合并滤液混合,浓缩成浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,加入辅料或不加辅料制成药物制剂。
前述的双金胃疡制剂的制作方法,其特征在于:所述双金胃疡制剂按重量份计算,主要由雪胆42份、金荞麦32份、大血藤32份、紫珠21份、麻布袋32份、延胡索32份、仙鹤草32份、白及63份、凤凰衣42份、土木香32份及核桃仁40份制成;所述双金胃疡制剂的制作方法为:取雪胆,加6-10倍量水煎煮2-3次,每次2-3h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加8-10倍量浓度为70-80%的乙醇提取2-3h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加6-10倍量1-5℃的冷水浸泡0.5-1.5h后,75-80℃,0.07-0.08MPa下温浸2-4次,每次2-3h,过滤,合并滤液备用,药渣加8-12倍量浓度为70-80%的乙醇,用盐酸调pH1-2,浸0.5-1.5h后,回流提取2-3h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,加入辅料或不加辅料制成药物制剂。

前述的双金胃疡制剂的制作方法中,所述胶囊剂这样制备:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,装入胶囊,即得。
前述的双金胃疡制剂的制作方法中,所述丸剂这样制备:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,每100g粉末用炼蜜25-40g加适量水制成水蜜丸,干燥,或每100g粉末加炼蜜100-130g,制成小蜜丸或大蜜丸,即得。
前述的双金胃疡制剂的制作方法中,所述片剂这样制作:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,加入制成量10%的淀粉,用乙醇制粒,压片,干燥,即得。
前述的双金胃疡制剂的制作方法中,所述颗粒剂这样制作:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,再加入制成量40%的蔗糖粉混匀,制粒,干燥,即得。
本发明主要由雪胆、金荞麦、大血藤、紫珠、麻布袋、延胡索、仙鹤草、白及、凤凰衣、土木香及核桃仁制成;具有舒肝理气,健胃止痛,收敛止血的功效,用于肝胃气滞血淤所致的胃脘刺痛,呕吐吞酸,脘腹胀痛,胃及十二指肠溃疡见上述症候者。
现有双金胃疡制剂的制作方法方法为:取原料中的所有药材,粉碎成细粉,混匀,装入胶囊,即得。但是发明人在研究过程中发现,采用这种方法制成的双金胃疡制剂中,药物中的有效分不易测出。使得双金胃疡制剂的临床疗效还不够理想。发明人在工艺研究中还发现,采用本发明方法将雪胆和仙鹤草进行单独提取后,再与其他药材细粉混合,制成药物制剂,这样制成的双金胃疡制剂中,药物中的有效成分容易测出,且测出的药物中的有效成分含量高。这就使得使得本药物更加精炼,药效更加显著,临床疗效好。因此该工艺促进了药物中的有效成分的溶出率,特别是雪胆中雪胆甲素的溶出率,雪胆甲素是雪胆的重要的活性成分,具有很强的药理活性,雪胆甲素的溶出率提高后,可有效提高双金胃疡制剂的药效。
本发明进行了大量的实验研究,以下为本发明实验研究的结果:
实验例1:雪胆甲素含量考察
1仪器与试药
1.1本发明双金胃疡胶囊,按实施例1的方法进行制作。
1.2现有双金胃疡胶囊:按下述工艺进行制作:
处方:雪胆42g、金荞麦32g、大血藤32g、紫珠21g、麻布袋32g、延胡索32g、仙鹤草32g、白及63g、凤凰衣42g、土木香32g及核桃仁40g。
工艺:取上述药物,粉碎成细粉,混匀,装入胶囊,即得。
1.3仪器岛津LC-2010A高效液相色谱仪,岛津AEG-45SM十万分之一电子分析天平(日本);FA2004分析天平(上海天平仪器厂制造);SK250LH超声仪(上海科导超声仪器有限公司)。雪胆甲素对照品(1518-200101,供含量测定用,中国药品生物制品检定所);甲醇(色谱纯),乙醇(分析纯)。
2方法与结果
2.1色谱条件色谱柱Diamonsil(钻石)柱4.6mm×150mm,5μm(C18柱,迪马公司),柱温:25℃,流动相:甲醇-水(80∶20),流速:1.0ml/min,检测波长:210nm,灵敏度0.05AUFS,进样量为10μl。
2.2对照品溶液的制备
精密称取经五氧化二磷减压干燥至12小时以上的雪胆甲素对照品适量,以甲醇为溶剂,配成浓度为0.427mg/ml的对照品溶液。
2.3供试品溶液的制备
2.3.1供试品溶液1的制备
取本发明双金胃疡胶囊适量,研细,过20目筛,精密称定1.2g,置50ml容量瓶中,加甲醇约40ml,超声提取30分钟,静置冷却,再用甲醇稀释至刻度,摇匀,即得供试品溶液1。
2.3.2供试品溶液2的制备
取现有双金胃疡胶囊适量,研细,精密称定1.2g,研细,过20目筛,精密称定1.2g,置50ml容量瓶中,加甲醇约40ml,超声提取30分钟,静置冷却,再用甲醇稀释至刻度,摇匀,即得供试品溶液2。
2.4线性关系取对照品溶液,用色谱仪的自动进样器分别进样对照品溶液1、2、4、6、8、10μl,测定峰面积,以雪胆甲素(μg)为横坐标X,峰面积为纵坐标Y,得回归方Y=1230.65+380007.3012X,r=0.9999。
2.5精密度试验精密吸取上述对照品溶液10μl,重复进样6次,按上述色谱条件测定雪胆甲素峰面积。结果,RSD=0.36%,表明仪器精密度良好。
2.6重复性试验分别精密称取本发明双金胃疡胶囊和现有双金胃疡胶囊各6份,照“2.3”项下方法制备供试品溶液1和供试品溶液2,按上述色谱条件进样,测定。结果RSD=1.05%和RSD=1.08%,表明本方法重复性良好。
2.7稳定性试验分别取供试品溶液1和供试品溶液2,分别于0h、3h、5h、7h、11h、16h各精密吸取10μl,按上述色谱条件测定峰面积积分值。结果RSD=0.91%和RSD=0.95%,表明供试品溶液在12h内稳定。
2.8加样回收率试验分别取已知含量的本发明双金胃疡胶囊和现有双金胃疡胶囊粉末各9份,每份约1.2g,精密称重,分别精密加入1ml的雪胆甲素对照品溶液,照“2.3”项下方法制备供试品溶液1和供试品溶液2,按上述色谱条件进样10μl,测定含量,计算回收率和RSD。结果平均回收率为98.67%和98.55%,RSD=0.99%和RSD=0.10%。
2.9样品含量测定
分别取本发明双金胃疡胶囊和现有双金胃疡胶囊,粉碎后,分别精密称取粉末1g,平行分成7份,按照上述方法制备供试品溶液1和供试品溶液2,在分别精密吸取10μl,注入高效液相色谱仪,分别计算样品中雪胆甲素含量。结果见表1。

从表1可知,本发明双金胃疡胶囊中雪胆甲素的平均含量为0.523mg/g,现有双金胃疡丸中雪胆甲素的平均含量为0.399mg/g。
结论:与现有双金胃疡胶囊相比,本发明双金胃疡胶囊中雪胆有效成分雪胆甲素的含量更高。同理,按照本发明提取方法制成的其他剂型的制剂,雪胆甲素含量也应增加。
实验例2:药效考察
1实验材料

1.1.3西咪替丁片,北京第四制药厂生产,批号:010908;胃复安片,北京太洋药业有限公司生产,批号:020304。
1.2动物ICR小鼠,♀♂兼用,♀,体重(26±3)g;SD大鼠,♂,体重(240±10)g。
1.3仪器与试剂704—1型双筒立体显微镜,中国计量科学研究院计量标准仪器厂产品;722分光光度计,上海精密科学仪器有限公司产品;HZQ-C空气浴振荡器,哈尔滨市东明医疗仪器厂产品;OHAUS天平,TP200S型。
以上药物用前均以0.5%CMC溶液配成所需浓度;硫酸阿托品注射液,山东新华制药厂生产,批号:960816;甲基硫酸新斯的明注射液,上海信谊药厂生产,批号:930602;盐酸吗啡,青海制药厂生产,批号:850802;阿尔新兰为E.M.Chemicais进口分装,柠檬酸、磷酸氢二钠、氢氧化钠等均为分析纯。
2方法
2.1镇痛作用
筛选合格的♀小鼠,随机分为对照组、本发明组(2g/kg),现有制剂组(2g/kg),盐酸吗啡组(10mg/kg)。各组小鼠ig等容量的药,对照组给予0.5%CMC溶液,连续3d,于末次给药后15min,30min,60min,120min测定痛阈值。盐酸吗啡组于实验前15min ip给药。结果见表2.

由表可知,本发明双金胃疡胶囊和现有双金胃疡胶囊在给药后30min内能明显提高小鼠痛阈值,具有镇痛作用,且本发明双金胃疡胶囊优于现有双金胃疡胶囊。
2.2对利血平所致小鼠胃溃疡的影响
筛选合格的♀小鼠,随机分为对照组、本发明组(2g/kg),现有制剂组(2g/kg),西咪替丁组(10mg/kg)。各组小鼠ig等容量的药,对照组给予0.5%CMC溶液,连续3d,于末次给药后1h,小鼠sc利血平10mg/kg,6h后处死动物。取胃溃疡程度评价。结果见表3.

由表可知,本发明双金胃疡胶囊和现有双金胃疡胶囊均能明显降低小鼠利血平性胃溃疡模型的溃疡指数,且本发明双金胃疡胶囊优于现有双金胃疡胶囊。
与现有技术相比,本发明所述方法制作的双金胃疡制剂中,药物中的有效成分雪胆甲素容易测出,且测出的雪胆甲素含量高。可以有效的提高双金胃疡制剂的药效。
具体实施方式:
实施例1:
配方:雪胆42g、金荞麦32g、大血藤32g、紫珠21g、麻布袋32g、延胡索32g、仙鹤草32g、白及63g、凤凰衣42g、土木香32g及核桃仁40g。
工艺:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,装入胶囊,即得胶囊剂。
用法用量:口服,一次3粒,一日3次。
规格:0.4g/粒。
注意:肾脏病患者,孕妇、新生儿禁用。儿童及老人慎用;在医生指导下服用。
实施例2:
配方:雪胆52g、金荞麦42g、大血藤42g、紫珠26g、麻布袋42g、延胡索42g、仙鹤草42g、白及70g、凤凰衣52g、土木香42g、核桃仁50g及辅料。
工艺:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,每100g粉末用炼蜜34g加适量水制成水蜜丸,干燥,制成小蜜丸,即得丸剂。
用法用量:口服,一次3粒,一日3次。
规格:0.4g/丸。
注意:肾脏病患者,孕妇、新生儿禁用。儿童及老人慎用;在医生指导下服用。
实施例3:
配方:雪胆32g、金荞麦22g、大血藤22g、紫珠15g、麻布袋22g、延胡索22g、仙鹤草22g、白及55g、凤凰衣32g、土木香22g、核桃仁30g及辅料。
工艺:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,加入制成量10%的淀粉,用乙醇制粒,压片,干燥,即得片剂。
用法用量:口服,一次3粒,一日3次。
规格:0.4g/片。

1.一种双金胃疡制剂的制作方法,其特征在于:所述双金胃疡制剂按重量份计算,主要由雪胆32-52份、金荞麦22-42份、大血藤22-42份、紫珠15-26份、麻布袋22-42份、延胡索22-42份、仙鹤草22-42份、白及55-70份、凤凰衣32-52份、土木香22-42份及核桃仁30-50份制成;所述双金胃疡制剂的制作方法为:取雪胆,加水煎煮后,过滤,滤液浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加醇提取,回收乙醇至净,提取液与上述浓缩液混合,浓缩成浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,冷水浸泡后,再温浸2-4次,过滤,合并滤液备用,药渣加入乙醇溶液,用盐酸调pH,浸泡后,回流提取,过滤,得提取液,提取液与上述合并滤液混合,浓缩成浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,加入辅料或不加辅料制成药物制剂。
2.如权利要求1所述的双金胃疡制剂的制作方法,其特征在于:所述双金胃疡制剂按重量份计算,主要由雪胆42份、金荞麦32份、大血藤32份、紫珠21份、麻布袋32份、延胡索32份、仙鹤草32份、白及63份、凤凰衣42份、土木香32份及核桃仁40份制成;所述双金胃疡制剂的制作方法为:取雪胆,加6-10倍量水煎煮2-3次,每次2-3h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加8-10倍量浓度为70-80%的乙醇提取2-3h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加6-10倍量1-5℃的冷水浸泡0.5-1.5h后,75-80℃,0.07-0.08MPa下温浸2-4次,每次2-3h,过滤,合并滤液备用,药渣加8-12倍量浓度为70-80%的乙醇,用盐酸调pH1-2,浸0.5-1.5h后,回流提取2-3h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,加入辅料或不加辅料制成药物制剂。
3.如权利要求2所述的双金胃疡制剂的制作方法,其特征在于:所述双金胃疡制剂的制作方法为:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,加入辅料或不加辅料制成药物制剂。
4.如权利要求1-3中任一项所述的双金胃疡制剂的制作方法,其特征在于:所述药物制剂为口服制剂。
5.如权利要求4所述的双金胃疡制剂的制作方法,其特征在于:所述口服制剂为胶囊剂、丸剂、片剂或颗粒剂。
6.如权利要求5所述的双金胃疡制剂的制作方法,其特征在于:所述胶囊剂这样制备:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,装入胶囊,即得。
7.如权利要求5所述的双金胃疡制剂的制作方法,其特征在于:所述丸剂这样制备:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,每100g粉末用炼蜜25-40g加适量水制成水蜜丸,干燥,或每100g粉末加炼蜜100-130g,制成小蜜丸或大蜜丸,即得。
8.如权利要求5所述的双金胃疡制剂的制作方法,其特征在于:所述片剂这样制作:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,加入制成量10%的淀粉,用乙醇制粒,压片,干燥,即得。
9.如权利要求5所述的双金胃疡制剂的制作方法,其特征在于:所述颗粒剂这样制作:取雪胆,加8倍量水煎煮3次,每次2.5h,过滤,合并滤液,浓缩成常温下相对密度为1.1-1.2g/ml的浓缩液,备用;药渣加9倍量浓度为75%的乙醇提取2.5h,过滤,回收乙醇至净,提取液与上述浓缩液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得雪胆干膏粉,备用;取仙鹤草,加8倍量1-5℃的冷水浸泡1h后,75-80℃,0.07-0.08MPa下温浸3次,每次2.5h,过滤,合并滤液备用,药渣加10倍量浓度为75%的乙醇溶液,用盐酸调pH 1.5,浸1h后,回流提取2.5h,过滤,得提取液,提取液与上述合并滤液混合,浓缩成50-60℃下相对密度为1.1-1.2的浸膏,干燥,粉碎,得仙鹤草干膏粉,备用;其余药材粉碎成细粉后,与上述雪胆干膏粉和仙鹤草干膏粉混匀,再加入制成量40%的蔗糖粉混匀,制粒,干燥,即得。
