Abstract

本发明提供了南瓜藤总甾醇的提取分离方法,它包括如下操作步骤:(1)取新鲜南瓜藤70℃烘干,粉碎过60目筛;(2)以质量浓度90%乙醇70℃加热回流,提取物干燥后得乙醇提取物;(3)乙醇提取物加水分散后,用石油醚多次萃取,合并石油醚层,除去溶剂后得石油醚萃取物;(4)取石油醚萃取物上硅胶柱分离,先用体积比为100:1的二氯甲烷和甲醇洗脱除去杂质,再用体积比为10:1的二氯甲烷和甲醇作为洗脱剂进行洗脱,收集洗脱液,减压浓缩,干燥得南瓜藤总甾醇提取物。本发明的工艺简单,提取率高,重现性好,稳定可行。

技术领域
本发明涉及南瓜藤总甾醇的提取分离方法,属于天然药物化学领域。
背景技术
南瓜[Cucurbitamoschata(Duch).Poiret]是葫芦科南瓜属的植物。南瓜藤,别名番瓜藤,盘肠草,南瓜苗,是一年生蔓生草本,茎条达2-5m,原产墨西哥到中美洲一带,世界各地普遍栽培,明代传入我国,现南北各地广泛种植。南瓜藤含丰富的胡萝卜素,粗纤维素等营养成分,具有清肺、和胃、通络,治肺结核低热,胃痛,月经不调,烫伤等功效。植物甾醇具有多种不同的生物学功能,如作为细胞膜的成分及构成肾上腺皮质激素和性激素等。不少植物甾醇还具有很强的药理或毒理效应,如增强心肌的收缩,治疗心力衰竭等。目前,从本属植物中已分离得到的化学成分包括糖类、多肽、萜类、黄酮及有机酸等。然而,目前关于南瓜藤甾醇提取工艺的研究少见报道。
发明内容
本发明的目的在于:提供一种南瓜藤总甾醇的提分离取方法,该提取分离方法简单,得率高。
本发明的技术解决方案是该南瓜藤总甾醇的提取分离方法包括如下步骤:
(1)取新鲜南瓜藤70℃烘干,粉碎过60目筛,得南瓜藤粉末;
(2)将南瓜藤粉末置于加热回流装置中,用体积分数90%乙醇70℃加热回流2-3小时,加热回流后过滤,滤渣再重复回流3-4次,合并滤液,减压回收乙醇后真空干燥得乙醇提取物;其中南瓜藤粉末与90%乙醇的质量体积比为1:20(g/mL);
(3)称取乙醇提取物,按乙醇提取物质量与蒸馏水体积1:1(g/mL)加入蒸馏水;然后再按乙醇提取物质量与石油醚体积1:1(g/mL)加入石油醚萃取,激烈混匀后常温静置24小时除去石油醚层,萃余相以相同的方式再萃取3-4次,收集合并石油醚萃取液,减压蒸馏,浓缩蒸干,得到石油醚萃取物;
(4)取石油醚萃取物上硅胶层析柱(200-300目)分离,南瓜藤石油醚萃取物采用干法上样,先使用体积比为100:1的二氯甲烷与甲醇的混合液作为洗脱液去除脂肪酸类杂质,然后以体积比为10:1二氯甲烷与甲醇的混合液作为洗脱剂,收集洗脱液,减压浓缩,干燥得南瓜藤总甾醇。
本发明的优点在于:工艺简单,成本低,易操作,得到的提取物中总甾醇含量高,具有进一步开发价值。
具体实施方式
下面结合具体实施例进一步说明本发明的技术解决方案,这些实施例不能理解为是对技术方案的限制。
实施例1:依以下步骤提取分离南瓜藤总甾醇
(1)取新鲜南瓜藤70℃烘干,粉碎过60目筛,得南瓜藤粉末;
(2)将南瓜藤粉末置于加热回流装置中,用体积分数90%乙醇70℃加热回流2小时,加热回流后过滤,滤渣再重复回流4次,合并滤液,减压回收乙醇后真空干燥得乙醇提取物;其中南瓜藤粉末与90%乙醇质量体积比为1:20(g/mL);
(3)称取乙醇提取物,按乙醇提取物质量与蒸馏水体积1:1(g/mL)加入蒸馏水;然后再按乙醇提取物质量与石油醚体积1:1(g/mL)加入石油醚萃取,激烈混匀后常温静置24小时除去石油醚层,萃余相以相同的方式再萃取3次,将石油醚萃取液合并,减压蒸馏,浓缩蒸干,得到石油醚萃取物;
(4)取石油醚萃取物上硅胶层析柱(200-300目)分离,南瓜藤石油醚萃取物采用干法上样,先使用体积比为100:1的二氯甲烷与甲醇的混合液作为洗脱液去除脂肪酸类杂质,然后以体积比为10:1二氯甲烷与甲醇的混合液作为洗脱剂,收集洗脱液,减压浓缩,干燥得南瓜藤总甾醇,总甾醇得率为1.36g总甾醇/100g干南瓜藤。
实施例2:依以下步骤提取分离南瓜藤总甾醇
(1)取新鲜南瓜藤70℃烘干,粉碎过60目筛,得南瓜藤粉末;
(2)将南瓜藤粉末置于加热回流装置中,用体积分数90%乙醇在70℃加热回流2.5小时,加热回流后过滤,滤渣再重复回流4次,合并滤液,减压回收乙醇后真空干燥得乙醇提取物;其中南瓜藤粉末与90%乙醇质量体积比为1:20(g/mL);
(3)称取乙醇提取物,按乙醇提取物质量与蒸馏水体积1:1(g/mL)加入蒸馏水;然后再按乙醇提取物质量与石油醚体积1:1(g/mL)加入石油醚萃取,激烈混匀后常温静置24小时除去石油醚层,萃余相以相同的方式再萃取4次,将石油醚萃取液合并,减压蒸馏,浓缩蒸干,得到石油醚萃取物;
(4)取石油醚萃取物上硅胶层析柱(200-300目)分离,南瓜藤石油醚萃取物采用干法上样,先使用体积比为100:1的二氯甲烷与甲醇的混合液作为洗脱液去除脂肪酸类杂质,然后以体积比为10:1二氯甲烷与甲醇的混合液作为洗脱剂,收集洗脱液,减压浓缩,干燥得南瓜藤总甾醇,总甾醇得率为1.52g总甾醇/100g干南瓜藤。

Claims (1)

1.南瓜藤总甾醇的提取分离方法,其特征是该提取分离方法包括如下步骤:
(1)取新鲜南瓜藤70℃烘干,粉碎过60目筛,得南瓜藤粉末;
(2)将南瓜藤粉末置于加热回流装置中,用体积分数90%乙醇70℃加热回流2-3小时,加热回流后过滤,滤渣再重复回流3-4次,合并滤液,减压回收乙醇后真空干燥得乙醇提取物;其中南瓜藤粉末与90%乙醇的质量体积比为1:20(g/mL);
(3)称取乙醇提取物,按乙醇提取物质量与蒸馏水体积1:1(g/mL)加入蒸馏水;然后再按乙醇提取物质量与石油醚体积1:1(g/mL)加入石油醚萃取,激烈混匀后常温静置24小时除去石油醚层,萃余相以相同的方式再萃取3-4次,收集合并石油醚萃取液,减压蒸馏,浓缩蒸干,得到石油醚萃取物;
