Abstract

本发明提供了紫藤花总黄酮的提取分离方法,它包括如下步骤:(1)取新鲜紫藤花在70℃烘干,粉碎过60目筛;(2)以体积分数90%乙醇在70℃加热回流,提取物干燥后得乙醇提取物;(3)乙醇提取物加水分散后,依次用石油醚、乙酸乙酯萃取,合并乙酸乙酯层,除去溶剂后,得乙酸乙酯提取物;(4)取乙酸乙酯提取物上D101大孔吸附树脂柱分离,先用体积分数30%乙醇洗脱除去杂质,再用体积分数80%乙醇作为洗脱剂进行洗脱,收集80%乙醇洗脱液,减压浓缩,干燥得紫藤花总黄酮提取物。本发明所述的方法工艺简单,提取率高,重现性好,稳定可行。

技术领域
本发明涉及紫藤花总黄酮的提取分离方法,属于食品添加剂及其制备方法领域。
背景技术
紫藤花为豆科植物紫藤的花序,是一种常见的可食性花,具有利水消瘕之功效,紫藤花中含有挥发油成分、酚类和黄酮类等多种活性成分,经研究显示,黄酮类化合物具有抗菌、消炎、提高人体免疫力、镇静、抗抑郁、抗氧化自由基等作用,而且黄酮类化合物可以作为天然食品添加剂、抗氧化剂,食用色素等,很多以黄酮类化合物作为功能因子的保健食品被开发。然而,目前关于紫藤花成分的研究报道主要是挥发油成分,对于紫藤花黄酮提取工艺的研究少见报道。
发明内容
本发明的目的在于:提供一种紫藤花总黄酮的提分离取方法,该提取分离方法简单,得率高。
本发明的技术解决方案是该紫藤花总黄酮的提取分离方法包括如下步骤:
(1)取新鲜紫藤花在70℃烘干,粉碎过60目筛,得紫藤花粉末;
(2)将紫藤花粉末置于加热回流装置中,用体积分数90%乙醇在70℃加热回流2-3小时,加热回流后过滤,滤渣再重复回流3-4次,合并滤液,减压回收乙醇后真空干燥得乙醇提取物;其中紫藤花粉末与90%乙醇质量体积比为1:15(g/mL);
(3)称取乙醇提取物,按乙醇提取物质量与蒸馏水体积1:1(g/mL)加入蒸馏水;然后再按乙醇提取物质量与石油醚体积1:1(g/mL)加入石油醚萃取,激烈混匀后常温静置24小时除去石油醚层,萃余相以相同的方式再萃取3-4次,除去乙醇提取物中脂溶性成分;将除去脂溶性成分的萃余液再加入乙酸乙酯进行萃取,乙酸乙酯加入体积与乙醇提取物质量比为1:1(g/mL),激烈混匀后常温静置24小时,萃取3-4次,合并乙酸乙酯萃取液,减压蒸馏,浓缩蒸干,得到乙酸乙酯萃取物浸膏;
(4)取乙酸乙酯萃取物浸膏上D101大孔吸附树脂柱分离,紫藤花乙酸乙酯萃取物浸膏上样浓度为1-4g/mL,先用3-5倍树脂床体积的体积分数30%乙醇洗脱除去杂质,再用3-5倍树脂床体积的体积分数80%乙醇洗脱,收集洗脱液,回收乙醇,减压浓缩,干燥得紫藤花总黄酮。
本发明的优点在于:采用大孔树脂富集纯化紫藤花总黄酮,工艺简单,成本低,易操作,得到的提取物中总黄酮含量高,具有进一步开发价值。
具体实施方式
下面结合具体实施例进一步说明本发明的技术解决方案,这些实施例不能理解为是对技术方案的限制。
实施例1:依以下步骤提取分离紫藤花总黄酮
(1)取新鲜紫藤花在70℃烘干,粉碎过60目筛,得紫藤花粉末;
(2)将紫藤花粉末置于加热回流装置中,用体积分数90%乙醇在70℃加热回流2小时,加热回流后过滤,滤渣再重复回流4次,合并滤液,减压回收乙醇后真空干燥得乙醇提取物;其中紫藤花粉末与90%乙醇质量体积比为1:15(g/mL);
(3)称取乙醇提取物,按乙醇提取物质量与蒸馏水体积1:1(g/mL)加入蒸馏水;然后再按乙醇提取物质量与石油醚体积1:1(g/mL)加入石油醚萃取,激烈混匀后常温静置24小时除去石油醚层,萃余相以相同的方式再萃取3次,除去乙醇提取物中脂溶性成分;将除去脂溶性成分的萃余液再加入乙酸乙酯进行萃取,乙酸乙酯加入体积与乙醇提取物质量比为1:1(g/mL),激烈混匀后常温静置24小时,萃取3次,合并乙酸乙酯萃取液,减压蒸馏,浓缩蒸干,得到乙酸乙酯萃取物浸膏;
(4)取乙酸乙酯萃取物浸膏上D101大孔吸附树脂柱分离,紫藤花乙酸乙酯萃取物浸膏上样浓度为1g/mL,先用3倍树脂床体积的体积分数30%乙醇洗脱除去杂质,再用3倍树脂床体积的体积分数80%乙醇洗脱,收集洗脱液,回收乙醇,减压浓缩,干燥得紫藤花总黄酮,总黄酮得率为0.98g总黄酮/100g干紫藤花。
实施例2:依以下步骤提取分离紫藤花总黄酮
(1)取新鲜紫藤花在70℃烘干,粉碎过60目筛,得紫藤花粉末;
(2)将紫藤花粉末置于加热回流装置中,用体积分数90%乙醇在70℃加热回流2.5小时,加热回流后过滤,滤渣再重复回流4次,合并滤液,减压回收乙醇后真空干燥得乙醇提取物;其中紫藤花粉末与90%乙醇质量体积比为1:15(g/mL);
(3)称取乙醇提取物,按乙醇提取物质量与蒸馏水体积1:1(g/mL)加入蒸馏水;然后再按乙醇提取物质量与石油醚体积1:1(g/mL)加入石油醚萃取,激烈混匀后常温静置24小时除去石油醚层,萃余相以相同的方式再萃取4次,除去乙醇提取物中脂溶性成分;将除去脂溶性成分的萃余液再加入乙酸乙酯进行萃取,乙酸乙酯加入体积与乙醇提取物质量比为1:1(g/mL),激烈混匀后常温静置24小时,萃取4次,合并乙酸乙酯萃取液,减压蒸馏,浓缩蒸干,得到乙酸乙酯萃取物浸膏;
(4)取乙酸乙酯萃取物浸膏上D101大孔吸附树脂柱分离,紫藤花乙酸乙酯萃取物浸膏上样浓度为2.5g/mL,先用4倍树脂床体积的体积分数30%乙醇洗脱除去杂质,再用4倍树脂床体积的体积分数80%乙醇洗脱,收集洗脱液,回收乙醇,减压浓缩,干燥得紫藤花总黄酮,总黄酮得率为1.02g总黄酮/100g干紫藤花。
实施例3:依以下步骤提取分离紫藤花总黄酮
(1)取新鲜紫藤花在70℃烘干,粉碎过60目筛,得紫藤花粉末;
(2)将紫藤花粉末置于加热回流装置中,用体积分数90%乙醇在70℃加热回流3小时,加热回流后过滤,滤渣再重复回流3次,合并滤液,减压回收乙醇后真空干燥得乙醇提取物;其中紫藤花粉末与90%乙醇质量体积比为1:15(g/mL);
(3)称取乙醇提取物,按乙醇提取物质量与蒸馏水体积1:1(g/mL)加入蒸馏水;然后再按乙醇提取物质量与石油醚体积1:1(g/mL)加入石油醚萃取,激烈混匀后常温静置24小时除去石油醚层,萃余相以相同的方式再萃取4次,除去乙醇提取物中脂溶性成分;将除去脂溶性成分的萃余液再加入乙酸乙酯进行萃取,乙酸乙酯加入体积与乙醇提取物质量比为1:1(g/mL),激烈混匀后常温静置24小时,萃取4次,合并乙酸乙酯萃取液,减压蒸馏,浓缩蒸干,得到乙酸乙酯萃取物浸膏;
(4)取乙酸乙酯萃取物浸膏上D101大孔吸附树脂柱分离,紫藤花乙酸乙酯萃取物浸膏上样浓度为4g/mL,先用5倍树脂床体积的体积分数30%乙醇洗脱除去杂质,再用5倍树脂床体积的体积分数80%乙醇洗脱,收集洗脱液,回收乙醇,减压浓缩,干燥得紫藤花总黄酮,总黄酮得率为1.12g总黄酮/100g干紫藤花。

Claims (1)

1.紫藤花总黄酮的提取分离方法,其特征是它包括如下步骤:
(1)取新鲜紫藤花在70℃烘干,粉碎过60目筛,得紫藤花粉末;
(2)将紫藤花粉末置于加热回流装置中,用体积分数90%乙醇在70℃加热回流2-3小时,加热回流后过滤,滤渣再重复回流3-4次,合并滤液,减压回收乙醇后真空干燥得乙醇提取物;其中紫藤花粉末与90%乙醇质量体积比为1:15(g/mL);
(3)称取乙醇提取物,按乙醇提取物质量与蒸馏水体积1:1(g/mL)加入蒸馏水;然后再按乙醇提取物质量与石油醚体积1:1(g/mL)加入石油醚萃取,激烈混匀后常温静置24小时除去石油醚层,萃余相以相同的方式再萃取3-4次,除去乙醇提取物中脂溶性成分;将除去脂溶性成分的萃余液再加入乙酸乙酯进行萃取,乙酸乙酯加入体积与乙醇提取物质量比为1:1(g/mL),激烈混匀后常温静置24小时,萃取3-4次,合并乙酸乙酯萃取液,减压蒸馏,浓缩蒸干,得到乙酸乙酯萃取物浸膏;
(4)取乙酸乙酯萃取物浸膏上D101大孔吸附树脂柱分离,紫藤花乙酸乙酯萃取物浸膏上样浓度为1-4g/mL,先用3-5倍树脂床体积的体积分数30%乙醇洗脱除去杂质,再用3-5倍树脂床体积的体积分数80%乙醇洗脱,收集洗脱液,回收乙醇,减压浓缩,干燥得紫藤花总黄酮。
