Description
技术领域
本发明涉及一种微生物的耐酸性调节方法及具有耐酸性的微生物的筛选方法。
背景技术
近年来,在微生物中,以活菌的状态到达肠道并显示对宿主有效的保健效果的有用微生物备受关注。在这些有用微生物中包括某种乳酸杆菌属细菌及双歧杆菌属细菌。迄今为止,关于乳酸杆菌属细菌,除便秘及腹泻的改善等肠道调节作用以外,还报告有乳腺癌的发病风险的降低作用(专利文献1)及白细胞介素12产生诱导能力(专利文献2);关于双歧杆菌属细菌,报告有胆甾醇吸收抑制作用(专利文献3)及弹性蛋白酶活性抑制作用(专利文献4)等这样的许多的保健效果。
为了使这些有用微生物在消化道内有效地起作用发挥保健效果,需要保持活菌的状态到达消化道内,然而直至到达消化道内时,对有用微生物而言存在温度、pH、氧、浸透压、酸等各种抑制繁殖的环境及抑制繁殖的物质。因此,在利用这些微生物时,该微生物是否具有对抑制繁殖的环境及抑制繁殖的物质的耐受性是重要的。特别是酸成为这些菌是否可到达消化道内的较大的因素。因此,具有耐酸性可以说是有用微生物的有利的性质之一。
在有用微生物中,特别是双歧杆菌属细菌为专性厌氧菌,对氧、低pH、高酸度不耐受,在发酵饮食品的制造时的增殖及保存时的存活性等操作困难的情况较多。为了得到双歧杆菌属细菌的保健效果,可以认为需要使尽量多的细菌以活菌的状态到达肠道,特别是提高饮食品中的细菌的存活性、即饮食后向肠道的到达率成为重要的因子。因此,可以说增强耐酸性且提高向肠道的到达度的双歧杆菌属细菌的必要性较高。
作为具有耐酸性的有用微生物,已知有乳酸杆菌属细菌的干酪乳杆菌YIT9029(专利文献5)、双歧杆菌属细菌的短双歧杆菌YIT12272(专利文献6)等。这些具有耐酸性的有用微生物以各种发酵乳制品及活菌制剂等形态出售有多种市售品。特别是发酵乳饮食品具有优异的适口性,因此,易于持续饮用,适合这些有用微生物的投予。
另一方面,也存在减弱微生物的耐酸性的必要性,以期望的形式调节微生物的耐酸性极其重要。
作为微生物的耐酸性调节机制,可以列举利用ATP依赖性的质子泵将质子抽出到菌体外。但是,推测利用依赖ATP的本机制在低温保存微生物时微生物的细胞内ATP耗尽而不起作用,期望利用在低温状态下也起作用的耐酸性调节机制来调节微生物的耐酸性。
fadD基因在若干微生物中确认到其存在,是推测其作为将脂肪酸转化为酰基CoA的酶即long-chain-fatty-acid CoA ligase起作用的基因。
然而,迄今为止未知fadD基因和耐酸性的关系。

发明内容
发明所要解决的课题
本发明涉及提供一种通过在微生物中抑制fadD基因的表达来调节耐酸性的方法、以及以fadD基因的表达量等为指标的具有耐酸性的微生物的筛选方法。
用于解决课题的方法
本发明的发明人在为专性厌氧菌且通常不具有耐酸性的双歧杆菌属细菌中对具有耐酸性的特定的短双歧杆菌株及其菌株的基因进行网罗式解析,结果发现,在具有耐酸性的株中,可以显著抑制fadD基因的转录量,通过控制fadD基因的表达,能够调节微生物的耐酸性,能够以fadD基因的表达量等为指标进行具有耐酸性的微生物的筛选。
即,本发明涉及以下的1)~16)。
1)一种微生物的耐酸性调节方法,其特征在于,在具有fadD基因的微生物中,控制fadD基因的表达。
2)根据1)的耐酸性调节方法,其中,耐酸性为在低温状态维持功能的耐酸性。
3)根据1)或2)的耐酸性调节方法,其中,阻碍或抑制fadD基因的表达,增强耐酸性。
4)根据3)的耐酸性调节方法,其中,相对转录量为1%以下。
5)根据3)或4)的耐酸性调节方法,其中,相对转录量为0.1%以下。
6)根据1)~5)的耐酸性调节方法,其中,微生物为双歧杆菌属细菌。
7)根据1)~6)的耐酸性调节方法,其中,微生物为短双歧杆菌。
8)一种由1)~7)的方法调节了耐酸性的突变微生物。
9)根据8)的突变微生物,其具有如下性质:在培养到1×108cell/mL以上的菌数后,在37℃用胃酸对低温保存后的突变微生物处理60分钟的情况下,该突变微生物的存活率比突变前的微生物的存活率高5倍以上。
10)根据9)的突变微生物,其具有如下性质:在培养到1×108cell/mL以上的菌数后,在37℃用胃酸对低温保存后的突变微生物处理60分钟后,再在37℃用胆汁酸处理60分钟的情况下,该突变微生物的存活率比突变前的微生物的存活率高10倍以上。

发明的效果
根据本发明,不是利用育种改良获得具有耐酸性的突变株,能够容易地构建耐酸性得到增强的突变微生物。这里所得到的耐酸性为通过fadD基因的表达调节获得的,与依赖微生物的培养后的ATP的耐酸性不同,即使在低温状态也能够发挥其功能。另外,该突变微生物在活体内的存活性得到改善,因此,能够更可靠地发挥微生物所具有的保健效果。并且,该突变微生物的低温保存下的存活性得到改善,因此,能够延长制品的保存期间。并且,由于在制品的制造时能够培养至酸度更高的状态,因此,能够使初期培养的菌数增加,能够以1次培养回收更多的菌数。
另外,根据本发明,能够容易地构建耐酸性减弱的突变微生物。这样的突变微生物由于胃酸而大量死亡,因此,能够用作在胃中不发挥微生物所具有的保健效果的微生物。
另外,根据本发明的筛选方法,能够容易地筛选具有耐酸性的微生物。

具体实施方式
本发明中的fadD基因的核苷酸序列(序列号1)及与其具有高的相似性的多个核苷酸序列在NCBI数据库中已有登录,推测其作为将脂肪酸转化为酰基CoA的酶即long-chain-fatty-acid CoA ligase起作用。
在本发明中,控制fadD基因的表达包括阻碍或抑制fadD基因的表达、或者增强或重新导入fadD基因的表达。通过阻碍或抑制fadD基因的表达能够增强耐酸性,通过增强或重新导入fadD基因的表达能够减弱耐酸性。
在本发明中,作为控制fadD基因的表达的对象微生物,没有特别限定,可以优选列举要求增强耐酸性的有用的革兰氏阳性菌、革兰氏阴性菌、酵母等。其中,优选革兰氏阳性菌,特别优选已确认在活体中的安全性的乳酸杆菌属细菌、双歧杆菌属细菌。
作为乳酸杆菌属细菌,优选利用干酪乳杆菌、类干酪乳杆菌、玉米乳杆菌、鼠李糖乳杆菌等属于干酪乳杆菌群的细菌,能够特别优选利用干酪乳杆菌、鼠李糖乳杆菌。
另外,作为双歧杆菌属细菌,可以列举短双歧杆菌、长双歧杆菌、婴儿双歧杆菌、青春双歧杆菌、双歧双歧杆菌、链状双歧杆菌、假小链双歧杆菌、角双歧杆菌等,能够特别优选利用短双歧杆菌。
显示对人有用的生理效果的双歧杆菌属细菌为专性厌氧菌,对氧、低pH、高酸度不耐受,制造时的增殖及保存时的存活性等操作困难的情况较多,因此,优选作为成为本发明的应用对象的微生物。
在本发明中,典型而言,阻碍或抑制fadD基因的表达可以列举:(i)阻碍或抑制fadD基因向fadD mRNA的转录、(ii)阻碍或抑制fadD mRNA向fadD蛋白质的翻译,但并不限定于这些情况。
为了阻碍fadD基因的表达,只要破坏该基因、使其缺损即可,作为该方法,可以列举将完全不同的DNA片段插入基因的内部的插入失活法及通过阶段性的同源重组使靶基因的一部分或全部缺失的阶段性双交叉法,特别优选阶段性双交叉法。
具体而言,在使fadD基因的一部分或全部缺失的情况下,将夹着缺失区域的两侧的区域从染色体DNA上分离,或者通过PCR法扩增分离,例如在如pYSSE3这样的大肠杆菌中增殖但在对象微生物中在无法复制的质粒载体上以使这些DNA片段以沿与本来的方向相同的方向排列的状态进行克隆。接着,将所得到的重组质粒DNA使用电穿孔法等导入想要引起缺失的微生物中,从产生的抗生素抗性的克隆中选择通过PCR法等在与目标缺失区域的上游或下游的克隆区域的相同区域中发生重组而使质粒插入染色体的克隆。将这样得到的克隆在不含抗生素的培养基中重复传代培养,由此,质粒再次接近,通过存在的同源区域间的重组从染色体上脱离,随着细菌的增殖而消失,从而选择失去抗生素抗性的克隆。能够通过PCR法等从这样得到的克隆中分离缺失了fadD基因区域的克隆。
为了抑制fadD基因的表达,能够使用合成与该基因的mRNA的5′末端区域互补的短RNA、即所谓的RNA干扰的方法、对控制该基因的表达的区域或控制基因进行破坏或者缺损等而突变的方法,特别优选控制该基因的表达的区域的突变。具体而言,通过对控制fadD基因的转录的启动子的序列进行突变,能够增加或者减少该基因向mRNA中的转录量。这里,减少fadD基因向mRNA中的转录量是指相对转录量为1%以下,优选为0.1%以下。相对转录量是指以在同种微生物中fadD基因突变型的微生物的fadD基因的表达量(例如mRNA的表达量)除以fadD基因野生型的微生物的fadD基因的表达量得到的值。这里,fadD基因野生型的微生物是指fadD基因或其启动子的碱基序列未变异,不增强或导入、或者阻碍或抑制fadD基因的表达的微生物,fadD基因突变型的微生物是指通过对fadD基因或其启动子的碱基序列进行突变等来增强或导入、或者阻碍或者抑制fadD基因的表达的微生物。
另外,这里,对启动子的序列进行突变是指在启动子区域中使构成DNA片段的一部分的碱基(例如1~20个左右,优选为1~10个,进一步优选为1~5个碱基)取代、缺失的情况、或者添加或插入1~多个碱基(例如1~10个,优选为1~5个碱基)的情况,例如,在fadD基因的启动子中将距起始密码子碱基68bp的上游的碱基序列从胸腺嘧啶(T)取代为胞嘧啶(C)。
另一方面,为了增强fadD基因的表达,可以列举将导入了该基因的重组质粒对对象微生物导入,将该基因通过部位特异重组导入染色体的另一位置从而使微生物内的该基因的拷贝数增加,对该基因的启动子序列进行突变来增多向mRNA中的转录量从而使表达增加等方法,特别优选使该基因的拷贝数增加的方法。
作为向其它的微生物导入fadD基因的方法,可以采用利用了DNA的导入能力的感受态细胞法、利用了原生质体的原生质体PEG法及利用了高电压脉冲的电穿孔法等。另外,在将fadD基因导入微生物染色体的情况下,能够使用利用了同源重组的方法、部位特异性导入的方法。
另外,作为使fadD基因的拷贝数增加的方法的具体例,可以列举如下方法:向每1个微生物细胞具有多个拷贝数的质粒中,对fadD基因以含有该基因的本来的启动子序列和核糖体结合位点的方式进行克隆,或者在从其它的基因中分离或通过化学合成构建的启动子和核糖体结合部位的下游仅连接编码该基因的多肽的区域,进行克隆,构建重组质粒,通过电穿孔法等导入微生物细胞内。由此,能够提高微生物细胞内的该基因的拷贝数。
在本发明中,控制fadD基因的表达、调节为增强耐酸性的突变微生物的耐酸性增强,因此,能够作为有效发挥该微生物原来所具有的多种生理效果的饮食品及医药品利用。另外,以耐酸性减弱的方式调节的突变微生物可以用作在到达胃之前发挥例如该微生物原来具有的各种生理效果,在胃中不起作用的微生物。
这里,耐酸性是指对于对象微生物所具有的各种酸、特别是胃酸及胆汁酸的耐受性,更具体而言,是指在低温状态下也起作用的耐酸性。即,与推测为在低温状态下不起作用的ATP依赖的耐酸性明显不同。
这里,低温状态是指处于0~10℃的状态。具体而言,可以列举在低温状态下保存(低温保存),例如,可以列举在10℃以下保存7天以上、在5℃以下保存14天以上、在4℃下保存14天以上。
耐酸性的增强是指通过对微生物的fadD基因及其启动子进行突变等来阻碍或抑制fadD基因的表达,由此突变微生物的耐酸性相对于突变前的微生物增强。更具体而言,可以列举在将培养到1×108cell/mL以上的菌数的突变微生物在4℃、7天的条件下低温保存后,在37℃用胃酸处理60分钟的情况下,突变微生物的存活率(胃酸处理后的活菌数相对于胃酸处理前的活菌数的比例)比突变前的微生物高5倍以上、优选高10倍以上的性质。或者,可以列举在4℃、14天的条件下保存时高30倍以上,优选高50倍以上,在4℃、19天的条件下保存时高100倍以上、优选高150倍以上的性质。另外,可以列举在将培养到1×108cell/mL以上的菌数的突变微生物在4℃、7天的条件下低温保存后,在37℃用胃酸处理60分钟后,再在37℃用胆汁酸处理60分钟的情况下,突变微生物的存活率(胃酸处理后的活菌数相对于胃酸处理前的活菌数的比例)比突变前的微生物高10倍以上、优选高30倍以上的性质,或者,可以列举在4℃、14天的条件下保存时高100倍以上、优选高200倍以上的性质。这里,作为胃酸及胆汁酸,例如能够使用国际公开第2011/105335号中记载的人工胃液(pH3.3)、人工胆汁(1.0%牛胆汁(Oxgall))。
在将本发明的突变微生物制成饮食品及医药品的情况下,菌体可以为活菌或加热菌体(死菌)的任一种,或者也可以为冻干的菌体,或者也可以作为含有这些微生物的培养物、菌体处理物利用,优选以活菌的状态利用。
在用作医药品的情况下,能够将本发明的突变微生物与固体或液体的医药用无毒性载体混合,以惯用的医药品制剂的形态给药。作为这样的制剂,例如,可以列举片剂、颗粒剂、散剂、胶囊剂等固体制剂;溶液剂、悬浮剂、乳剂等液剂;冻干制剂等。这些制剂能够利用制剂上的常规方法制备。作为上述的医药用无毒性载体,例如,可以列举葡萄糖、乳糖、蔗糖、淀粉、甘露醇、糊精、脂肪酸甘油酯、聚乙二醇、羟乙基淀粉、乙二醇、聚氧乙烯山梨醇酐脂肪酸酯、氨基酸、明胶、白蛋白、水、生理盐水等。另外,也能够根据需要适当添加稳定化剂、润湿剂、乳化剂、偶联剂、等渗剂、赋形剂等惯用的添加剂。
另外,本发明的突变微生物不仅能够制成如上所述的制剂,而且也能够在饮食品中配合使用。在饮食品中配合的情况下,直接含有或与各种营养成分一同含有即可。该饮食品以期望的形式调节微生物的耐酸性,因此,能够作为有效发挥该微生物原来所具有的各种生理作用的保健用食品或食品原料利用。具体而言,在将利用本发明的方法所得到的突变微生物在饮食品中配合的情况下,可以适当使用能够用作饮食品的添加剂,采用惯用的方法成型为适于食用的形态、即颗粒状、粒状、片剂、胶囊、膏状等,另外,也可以在各种食品、例如火腿、香肠等肉食加工食品、鱼糕、竹轮等水产加工食品、面包、点心、黄油、奶粉中添加使用,或者在水、果汁、牛奶、清凉饮料、茶饮料等饮料中添加使用。此外,饮食品中也包括动物的饲料。
另外,作为饮食品,可以列举利用了本发明的突变微生物的发酵乳、乳酸菌饮料、发酵豆浆、发酵果汁、发酵植物液等发酵饮食品的例子。这些发酵饮食品的制造只要按照常规方法即可。例如在制造发酵乳的情况下,首先,将本发明的突变微生物在杀菌后的乳培养基中单独或与其它的微生物同时接种培养,将其均质化处理而得到发酵乳基料。接着,添加另行制备的糖浆溶液进行混合,用均质机等均质化,再添加风味剂加工成最终制品即可。这样得到的发酵乳也能够制成不含糖浆(甜味料)的普通型、软型、水果风味型、固体状、液状等任一形态的制品。
利用本发明的方法得到的调节为增强耐酸性的微生物即使在低温状态下也具有高耐酸性,因此,在存在酸的制品中的细菌的存活性高。因此,抑制了低温保存中的活菌数的降低及死亡速度的增加,易于维持制品规格,能够有效地发挥乳酸杆菌属细菌等微生物所具有的肠道调节作用等一般的生理效果。另外,针对原来具有抗癌作用、幽门螺杆菌的除菌作用等特异性的生理效果的乳酸杆菌属细菌及双歧杆菌属细菌,通过利用本发明的方法增强耐酸性,由此,能够将该菌株利用于各种饮食品,同时,能够通过改善该菌株的存活性来增强该菌株所具有的生理效果。
另外,利用本发明的方法得到的调节为耐酸性减弱的微生物因胃酸大量死亡,因此,能够作为在到达胃之前发挥该微生物原来所具有的各种生理效果、在胃中不起作用的微生物利用。
如上所述,在阻碍或抑制fadD基因的表达的情况下增强了微生物的耐酸性,因此,能够以fadD基因和/或其表达产物的表达量等为指标进行具有耐酸性的微生物的筛选。即,通过对fadD基因和/或其表达产物的有无和/或表达量进行测定,能够筛选该具有耐酸性的微生物。作为基因的表达产物,可以列举mRNA及多肽等,作为该多肽,可以列举由序列号9所示的氨基酸序列构成的多肽。
这里,fadD基因和/或其表达产物的测定能够采用能够检测fadD基因及源自该基因的mRNA的探针或引物,利用Southern杂交法、Northern杂交法、DNA微阵列、RT-PCR法对微生物中的靶基因的有无、拷贝数及表达量进行测定。另外,能够利用紫外吸收法、BCA法(二喹啉甲酸法)、Lowry法这样的分光光度法及电泳法对多肽的量进行测定。接着,根据fadD基因及其表达产物的有无、它们的表达量选择目标微生物(具有耐酸性的微生物)即可。
另外,为了高效地进行上述的基因的突变及微生物的筛选,优选使用含有序列号1所示的多核苷酸或其一部分的重组载体、由序列号1所示的多核苷酸的一部分片段构成的PCR及RT-PCR用引物、能够扩增序列号1所示的多核苷酸或其一部分的PCR及RT-PCR用引物、由与序列号1所示的多核苷酸特异性杂交的多核苷酸或其一部分构成的杂交用的核酸片段。
这里,作为所使用的引物等核酸片段,通常能够使用基于本发明的基因的碱基序列的信息化学合成的核苷酸等,作为该核苷酸,优选为与序列号1所示的碱基序列对应的部分核苷酸序列,优选为10~50个连续的碱基,更优选为15~35个连续的碱基。
下面,通过实施例对本发明的内容进一步详细地进行说明。
实施例
实施例1fadD基因的启动子区域的变异部位的确认
按照常规方法利用染料终止(dye-terminator)法对具有耐酸性的短双歧杆菌YIT12272(FERM BP-11320)、作为其菌株的短双歧杆菌YIT 4008(FERM BP-4538)及短双歧杆菌YIT 4065(FERM BP-6223)的fadD基因的周边的碱基序列进行解码、比较。其结果,在YIT12272中,在fadD基因的启动子中距起始密码子碱基68bp的上游的碱基序列从胸腺嘧啶(T)突变为胞嘧啶(C),认为存在其转录量变化的可能性。将fadD基因的启动子中的变异部位示于图1。
实施例2fadD基因的转录量解析
采用MILS培养基(Iwata and Morishita,Letter in Applied Microbiology,vol9,165-168,1989)在37℃对具有耐酸性的短双歧杆菌YIT 12272、作为其菌株的YIT 4008及YIT 4065进行厌氧培养。对厌氧培养而言,对气相进行氮置换并盖上丁基橡胶瓶塞,静置进行培养。
采用RNeasy Mini Kit(QIAGEN公司制)由对数增殖期的各菌株制备Total RNA。接着,采用本RNA 1μg,利用PrimeScript 1st strand cDNA Synthesis Kit(TaKaRa公司制)制备cDNA溶液。以由各菌株制备的cDNA溶液为模板,采用SYBR Premix Ex Taq(TaKaRa公司制)和表1所示的引物,利用ABI PRISM 7500(Applied Biosystems公司制)进行实时PCR(在95℃、30秒的反应后,进行95℃、5秒及60℃、34秒的反应40个循环),对fadD基因及16SrRNA基因的转录量进行测定。此外,以16SrRNA基因的转录量为内部标准进行样品间的校正。

将各菌株中的fadD基因的转录量以相对于fadD基因为野生型的YIT 4008的相对值的形式示于图2。YIT 4065中的fadD基因的转录量与YIT 4008为同等程度,但在fadD基因的上游发生变异的YIT 12272的转录量大幅降低,与YIT 4008相比的相对转录量为0.1%以下,即使与YIT 4065相比也为0.1%以下。
实施例3导入了野生型的fadD基因的转化体的获得及转化体的fadD基因的相对转录量
为了研究fadD基因对耐酸性造成的影响,制作在YIT 12272中导入了源自YIT4008的fadD基因启动子及fadD基因的转化体HM0102株。
首先,以YIT 4008的基因组DNA为模板,采用KOD-Plus-(TOYOBO公司制)和表2所示的引物,利用iCycler(Bio-Rad公司制)进行PCR(进行96℃·15秒、60℃·30秒、72℃·150秒的反应30个循环),对包括fadD基因的全长和其上游300bp及下游100bp的DNA片段(序列号6)进行扩增。

下划线部分表示限制酶的识别序列
将扩增得到的DNA片段和日本特开平10-262670号公报中记载的pBE 4载体按照同一文献中记载的常规方法用限制酶EcoRI消化。使用两消化片段,利用DNA Ligation KitVeR.2.1(TaKaRa公司制)进行连接反应,制作原始启动子支配下的fadD基因表达用质粒DNA。将本质粒DNA的构建图示于图3。
将本质粒DNA利用电穿孔法导入短双歧杆菌YIT 12272。电穿孔采用GENE PULSERII(Bio-Rad公司制)在200Ω、25μF、18kV/cm的条件下进行。然后,在添加了3μg/mL的红霉素的MILS琼脂培养基上涂抹电穿孔反应液,采用Anaero Pack(三菱气体化学公司制)在37℃进行72小时的厌氧培养。作为转化体HM0102株得到生成的菌落,按照常规方法利用碱法提取质粒DNA,确认到保持有导入的质粒DNA。
另一方面,作为HM0102株的对照,也以同样的方法构建仅将pBE 4载体导入YIT12272而成的HM0101株。
关于YIT 12272、HM0101株及HM0102株的3株,用实施例2中记载的方法对fadD基因的表达量进行测定。此外,作为转化体的HM0101株及HM0102株的培养采用MILS培养基的使酵母抽提物浓度设为1.5%的Y-MILS培养基,以最终浓度为3μg/mL的方式添加红霉素进行。
对各株的fadD基因的转录量进行测定,结果表明,与YIT 12272及HM0101株相比,在导入了野生型的fadD基因启动子及fadD基因的HM0102株中,fadD的转录量较高。将各株中的fadD基因的转录量以相对于YIT 4008中的转录量的相对值的形式示于图4。
实施例4转化体的耐酸性测定
为了更明确地确认转化体的耐酸性,对低温保存后的耐酸性进行测定。将在上述实施例3中记载的培养基中厌氧培养一晩使其为1×108cell/mL以上的菌数的HM0101株(培养后菌数:8.5×108cell/mL)及HM0102株(培养后菌数:8.8×108cell/mL)的各培养液在4℃厌氧静置保存。在保存第0天、第7天、第14天、第19天及第29天进行胃酸、胆汁酸连续处理,对两株的耐酸性进行比较。
胃酸、胆汁酸连续处理中采用专利文献6中记载的人工胃液及人工胆汁,如下进行。首先,在预先在37℃保温的人工胃液(pH3.3)10mL中添加在4℃保存的培养液0.5mL并搅拌,在37℃进行60分钟的胃酸处理。再在胃酸处理后的溶液2mL中添加人工胆汁(1.0%Oxgall)1mL和反应缓冲液(含有0.5%的氯化钠、0.1%的氯化钾、0.3%的碳酸氢钠的pH8.0的缓冲液)5mL并搅拌,在37℃连续地进行60分钟的胆汁酸处理。将胃酸处理0分钟后、60分钟后、胃酸、胆汁酸连续处理120分钟后的各处理液1mL适当稀释后,涂抹在TOS丙酸琼脂培养基(Yakult药品工业公司制)上,采用Anaero Pack(三菱气体化学公司制)在37℃厌氧培养72小时。将出现的菌落数乘以总稀释倍率求出每培养液1mL的活菌数,将各处理后的活菌数相对于酸处理0分钟后的活菌数的比例作为存活率。
这里,HM0102株具有野生型的fadD基因,HM0101株具有变异型的fadD基因,除此以外的基因完全相同,因此,HM0102株相当于fadD基因突变前微生物,HM0101株相当于fadD基因突变微生物。
将胃酸处理后及胃酸、胆汁酸连续处理后的两株的活菌数及存活率的测定结果示于图5,将HM0101株相对于HM0102株的存活率的比示于表3。其结果,随着低温保存的天数增加,在导入了野生型的fadD基因启动子的HM0102株中,胃酸处理及胃酸、胆汁酸连续处理后的存活率大幅降低,但在HM0101株中,存活率的降低幅度较小,耐酸性与HM0102株相比增强。在存活率之比中也显示HM0101株的耐酸性增强。这样在处于低温状态的情况下更显著地观察到耐酸性的增强。

由以上的结果可知,fadD基因的转录增大朝微生物、特别是短双歧杆菌中的耐酸性降低的方向发挥作用,相反,阻碍或抑制fadD基因的转录朝微生物、特别是短双歧杆菌中的耐酸性增强的方向发挥作用。因此,可以认为降低fadD基因的转录量不依赖于培养后的细胞内ATP量,能够用于微生物的耐酸性的增强。
Claims (4)
1.fadD基因在调节微生物的耐酸性中的用途,其特征在于:
在具有fadD基因的微生物中,通过改变控制fadD基因的转录的启动子的序列来阻碍或抑制fadD基因的表达,以使其相对转录量为0.1%以下,增强耐酸性,
具有fadD基因的微生物为短双歧杆菌。
2.如权利要求1所述的用途,其特征在于:
耐酸性为在低温状态维持功能的耐酸性。
3.如权利要求1或2所述的用途,其特征在于:
在控制fadD基因的转录的启动子中,距起始密码子碱基68bp的上游的碱基序列从胸腺嘧啶(T)突变为胞嘧啶(C)。
4.一种具有耐酸性的微生物的筛选方法,其特征在于:
在具有fadD基因的微生物中,通过改变控制fadD基因的转录的启动子的序列来阻碍或抑制fadD基因的表达,并通过对fadD基因和/或其表达产物的有无和/或表达量进行测定,筛选fadD基因的表达被阻碍或被抑制,以使其相对转录量为0.1%以下的微生物,作为具有耐酸性的微生物,
具有fadD基因的微生物为短双歧杆菌。
