技术领域
本发明涉及一种治疗高血压复方制剂的制备方法,属于中药制剂技术领域。
背景技术
高血压(hypertension)是指以体循环动脉血压(收缩压和/或舒张压)增高为主要特征(收缩压≥140毫米汞柱,舒张压≥90毫米汞柱),可伴有心、脑、肾等器官的功能或器质性损害的临床综合征。高血压是最常见的慢性病,也是心脑血管病最主要的危险因素。正常人的血压随内外环境变化在一定范围内波动。在整体人群,血压水平随年龄逐渐升高,以收缩压更为明显,但50岁后舒张压呈现下降趋势,脉压也随之加大。近年来,人们对心血管病多重危险因素的作用以及心、脑、肾靶器官保护的认识不断深入,高血压的诊断标准也在不断调整。引起高血压的原因主要有遗传因素、精神和环境因素、年龄因素、生活习惯因素、药物的影响或其他疾病的影响。
因此,目前高血压的治疗,往往需要多种药物配合使用,不但疗效难以多方面兼顾,并且患者适应性差。而多种药物的一次性用药难以实现,即使能够实现效果也不够理想。
发明内容
发明目的:为了克服现有技术中存在的不足,本发明提供一种治疗高血压复方制剂的制备方法。
技术方案:为实现上述目的,本发明提供了一种治疗高血压复方制剂的制备方法,包括以下步骤:
(1)取环糊精,加水研磨成糊状;取姜黄素,加入足量乙醇,超声溶解,然后滴加入上述糊状环糊精中,边滴边研磨,滴加完后继续研磨,干燥,得姜黄素包合物;
(2)按比例称取黄芪、怀牛膝、荞麦、夏枯草和郁金香,加入乙醇提取,得醇提残渣和提取液,提取液浓缩,得醇提液,备用;
(3)去上述醇提残渣,加入纯水提取,提取液浓缩,得水提液,备用;
(4)将骨架材料溶解,加入上述醇提液和水提液,高速分散,浓缩,干燥,得合成物;
(5)将上述姜黄素包合物和合成物混合均匀,即得。
作为优选,步骤(1)中所述滴加完后继续研磨,研磨时间为20-40min。
作为另一种优选,步骤(2)中所述乙醇提取为:加入6-10倍体积的60-80%乙醇回流提取2次,每次2小时。
作为另一种优选,步骤(3)中所述纯水提取为:加入8-12倍体积的水回流提取2次,每次2小时。
作为另一种优选,步骤(4)中所述高速分散的条件为:5000-7000r/min,分散30-60分钟。

处方:
姜黄素0.02份、β-环糊精10份、黄芪20份、怀牛膝10份、
荞麦5份、夏枯草15份、郁金香10份、壳聚糖60份。
制备工艺:
(1)取β-环糊精,加水研磨成糊状;取姜黄素,加入足量乙醇,超声溶解,然后滴加入上述糊状β-环糊精中,边滴边研磨,滴加完后继续研磨,研磨时间20-40min,干燥,得姜黄素包合物;
(2)按比例称取黄芪、怀牛膝、荞麦、夏枯草和郁金香,加入6-10倍体积的60-80%乙醇回流提取2次,每次2小时,得醇提残渣和提取液,提取液浓缩,得醇提液,备用;
(3)去上述醇提残渣,加入8-12倍体积的水回流提取2次,每次2小时,提取液浓缩,得水提液,备用;

(1)取或α-环糊精,加水研磨成糊状;取姜黄素,加入足量乙醇,超声溶解,然后滴加入上述糊状α-环糊精中,边滴边研磨,滴加完后继续研磨,研磨时间20-40min,干燥,得姜黄素包合物;
(2)按比例称取黄芪、怀牛膝、荞麦、夏枯草和郁金香,加入6-10倍体积的60-80%乙醇回流提取2次,每次2小时,得醇提残渣和提取液,提取液浓缩,得醇提液,备用;
(3)去上述醇提残渣,加入8-12倍体积的水回流提取2次,每次2小时,提取液浓缩,得水提液,备用;

姜黄素0.02份、α-环糊精12-18份、黄芪22-28份、怀牛膝12-18份、
荞麦8-12份、夏枯草18-22份、郁金香15-25份、聚乳酸80-100份。
制备工艺:
(1)取α-环糊精,加水研磨成糊状;取姜黄素,加入足量乙醇,超声溶解,然后滴加入上述糊状α-环糊精中,边滴边研磨,滴加完后继续研磨,研磨时间20-40min,干燥,得姜黄素包合物;
(2)按比例称取黄芪、怀牛膝、荞麦、夏枯草和郁金香,加入6-10倍体积的60-80%乙醇回流提取2次,每次2小时,得醇提残渣和提取液,提取液浓缩,得醇提液,备用;
(3)去上述醇提残渣,加入8-12倍体积的水回流提取2次,每次2小时,提取液浓缩,得水提液,备用;
(4)将聚乳酸溶解,加入上述醇提液和水提液,5000-7000r/min,高速分散30-60分钟,浓缩,干燥,得合成物;
(5)将上述姜黄素包合物和合成物混合均匀,即得。
实验例本发明制备方法所得复方制剂治疗高血压实验研究
选取75只实验用大鼠(已成功建立高血压模型),随机分为5组,每组15只,分别为阴性对照组、阳性对照组、实施例3-5组。
阴性对照组正常饮食;
阳性对照组在阴性对照组正常饮食的基础上,在饲料中加入姜黄素(姜黄素用量同实施例3、4和5组),进行喂养;
实施例3、4和5组,在阴性对照组正常饮食的基础上,给予本发明实施例3、4和5所得产品(其中姜黄素用量与阳性对照组均相同),同样掺入饲料中喂养。
所有组均每天给药1次,连续给药7天后,考察各组大鼠的平均血压。
实验结果见下表1:
表1各组大鼠平均血压和存活率(n=15)
注:与阴性对照组相比较,*P<0.05;与阳性对照组相比较,#P<0.05。
由上表1结果可得,与阴性对照组相比较,阳性对照组和本发明实施例3、4和5组大鼠经治疗,其平均血压显著降低;同时,与阳性对照组相比较,本发明实施例3、4和5组大鼠经治疗后,其平均血压也显著降低,并且其中实施例3组的降压效果最优。
Claims (10)
1.一种治疗高血压复方制剂的制备方法,其特征在于,包括以下步骤:
(1)取环糊精,加水研磨成糊状;取姜黄素,加入足量乙醇,超声溶解,然后滴加入上述糊状环糊精中,边滴边研磨,滴加完后继续研磨,干燥,得姜黄素包合物;
(2)按比例称取黄芪、怀牛膝、荞麦、夏枯草和郁金香,加入乙醇提取,得醇提残渣和提取液,提取液浓缩,得醇提液,备用;
(3)去上述醇提残渣,加入纯水提取,提取液浓缩,得水提液,备用;
(4)将骨架材料溶解,加入上述醇提液和水提液,高速分散,浓缩,干燥,得合成物;
(5)将上述姜黄素包合物和合成物混合均匀,即得。
2.根据权利要求1所述的制备方法,其特征在于,步骤(1)中所述滴加完后继续研磨,研磨时间为20-40min。
3.根据权利要求1所述的制备方法,其特征在于,步骤(2)中所述乙醇提取为:加入6-10倍体积的60-80%乙醇回流提取2次,每次2小时。
4.根据权利要求1所述的制备方法,其特征在于,步骤(3)中所述纯水提取为:加入8-12倍体积的水回流提取2次,每次2小时。
5.根据权利要求1所述的制备方法,其特征在于,步骤(4)中所述高速分散的条件为:5000-7000r/min,分散30-60分钟。
6.根据权利要求1所述的制备方法,其特征在于,其中各原料的重量份比例如下:
姜黄素0.02份、环糊精10-20份、黄芪20-30份、怀牛膝10-20份、
荞麦5-15份、夏枯草15-25份、郁金香10-30份、骨架材料60-120份。
7.根据权利要求1所述的制备方法,其特征在于,其中各原料的重量份比例如下:
姜黄素0.02份、环糊精12-18份、黄芪22-28份、怀牛膝12-18份、
荞麦8-12份、夏枯草18-22份、郁金香15-25份、骨架材料80-100份。
8.根据权利要求1所述的制备方法,其特征在于,所述环糊精为环糊精为β-环糊精或α-环糊精。
9.根据权利要求1所述的制备方法,其特征在于,所述骨架材料为壳聚糖、羧甲基纤维素钠、乙基纤维素、羟丙甲纤维素或者聚乳酸。
10.根据权利要求1所述的制备方法,其特征在于,所述高血压为原发性高血压或者继发性高血压。
