本发明涉及一种用于治疗小儿感冒的药物制剂的制备方法。该小儿感冒颗粒是以金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材为原料,通过超临界流体萃取、超声波提取和微孔滤膜过滤等现代提取分离技术经环糊精包合、制粒、整粒、分装而成。本发明萃取分离效果好,提取率高,节约能源和成本,且该产品采用包合物制备技术,避免了挥发油类功效成分的挥发和掩盖了产品的不良气味,进一步提升了产品的品质和患儿服药的顺应性。

本发明涉及一种中药制剂的制备方法,具体为一种用于治疗小儿感冒的药物制剂的制备方法。
背景技术
小儿感冒,即小儿急性上呼吸道感染,指喉部以上,上呼吸道鼻咽部的急性感染。小儿感冒以病毒为主,此外可能有支原体或细菌感染。幼儿期发病最多,大多表现为流清水样鼻涕、打喷嚏、鼻塞、咳嗽,常伴有发热,体温可高达39℃-40℃,个别患儿还会因发热而引起抽风,或出现腹泻、腹痛、呕吐等症状。目前,对于小儿感冒的治疗主要采用西医和中医治疗。西医一般采用抗生素、激素、非甾体类抗炎药为主,虽起效较快,但副作用较大,严重影响患儿的身心健康;而目前上市的治疗小儿感冒的相关中药制剂,大多提取制备工艺相对简单粗放,药物起效慢,治疗效果不理想,且患儿服药的顺应性差,难以满足临床的用药需求。
鉴于此,本发明选用金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材为原料,通过现代提取制备工艺,制成小儿感冒颗粒,能够迅速有效地疏散风热,清热止咳,可用于春、秋、冬季小儿感冒发烧,汗出不爽,鼻塞流涕,咳嗽咽痛等症。
发明内容
本发明的目的是针对目前产品存在的一些缺陷,提供一种能够迅速有效,且副作用小的治疗小儿感冒药物制剂的制备方法。
本发明的目的是通过以下技术方案予以实现的。
一种用于治疗小儿感冒的药物制剂的制备方法,由下列重量份的原料制成,薄荷80-320份、荆芥穗67-268份、苦杏仁80-320份、牛蒡子80-320份、黄芩80-320份、桔梗67-268份、前胡80-320份、白芷27-108份、栀子(炒)40-160份、山楂(焦)27-108份、六神曲(焦)27-108份、麦芽(焦)27-108份、芦根120-480份、金银花120-480份、连翘80-320份,其特征在于,采用以下步骤制备:
①分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材粉碎成细粉,备用;
②将步骤①中粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,提取出薄荷油和荆芥油等挥发油;
③将步骤②中的挥发油用8倍量的β-环糊精,再加10倍量的水溶解,并于40℃搅拌2h进行包合,制成环糊精包合物,备用;
④将步骤①中粉碎的金银花、连翘、牛蒡子、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十三味药材粉末一起加入8倍药材重量的水超声提取,提取后过滤;
⑤将步骤④过滤的滤液合并,低温静置48h;
⑥将步骤⑤静置后的清膏用微孔滤膜过滤,减压浓缩成60℃时相对密度为1.25-1.35的稠膏;
⑦将步骤③制备的环糊精包合物和步骤⑥的稠膏混合,再加入辅料,一并混合均匀,制粒、干燥、整粒,分装,即得。
步骤②所述的超临界CO2流体萃取技术的萃取压力为20-45Mpa,萃取温度为25-60℃,萃取时间为1-3h。
步骤④所述的超声提取次数为2次,每次0.5-1h,超声波功率为40-80kw,温度为25℃。
步骤⑦所述的加入的辅料为蔗糖、糊精、淀粉、甜菊素、微晶纤维素、硬脂酸镁的一种或多种。
所述治疗小儿感冒的药物还可制备成口服液、散剂或片剂。
本发明的有益效果是:本发明通过超临界CO2流体萃取技术和超声波技术提取,萃取分离效果好,避免了药材中热敏性成分的降解,提取率高,有利于药材的充分利用,节约能源和成本,具有提取时间短、提取率高和无需加热等特点,且该产品采用包合物制备技术,避免了挥发油类功效成分的挥发和掩盖了产品的不良气味,进一步提升了产品的品质和患儿服药的顺应性。
具体实施方式
以下通过具体实施例对本发明作进一步说明,应该理解的是,这些实施例仅用于说明本发明,并不限制本发明的范围。
实施例1
处方组成:薄荷80kg、荆芥穗67kg、苦杏仁80kg、牛蒡子80kg份、黄芩80kg、桔梗67kg、前胡80kg、白芷27kg、栀子(炒)40kg、山楂(焦)27kg、六神曲(焦)27kg、麦芽(焦)27kg、芦根120kg、金银花120kg、连翘80kg,蔗糖40kg,糊精200kg,甜菊素1.5kg,共制成200000袋(2.5g/袋)。
采用以下步骤制备:分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材粉碎成细粉,备用;将粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,萃取压力为20Mpa,萃取温度为30℃,萃取时间为1.5h,提取出薄荷油和荆芥油等挥发油;将提取的挥发油用8倍量的β-环糊精,再加10倍量的水溶解,并于40℃搅拌2h进行包合,制成环糊精包合物,备用;将粉碎的金银花、连翘、牛蒡子、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十三味药材粉末一起加入8倍药材重量的水超声提取,声提取次数为2次,每次0.5h,超声波功率为45kw,温度为25℃,提取后过滤;将滤液合并,低温静置48h;静置后的清膏用微孔滤膜过滤,减压浓缩成60℃时相对密度为1.30的稠膏;将制备的环糊精包合物加入到该稠膏中混合,再加入辅料,一并混合均匀,制粒、干燥、整粒,分装,即得。
鉴别:(1)取本品10g,研细,用20ml甲醇超声提取20分钟,滤过,滤液作为供试品溶液。分别取薄荷脑加醋酸乙酯制成每2ml含0.5mg的溶液,取荆芥油加乙酸乙酯制成每5ml含0.5ml的溶液,作为对照品溶液。照薄层色谱法(中国药典2015版)试验,吸取上述供试品溶液15μl、对照品溶液各2μl,分别点于同一含羧甲基纤维素钠为粘合剂的硅胶G薄层板上,以环己烷-醋酸乙酯(6:1)为展开剂,展开,取出,晾干,喷以硫酸-5%香草醛乙醇溶液(1:18)的混合溶液,在105℃烘约5分钟,日光下检识。供试品色谱中,在与对照品色谱相应的位置上,显相同颜色的斑点。

测定法分别精密吸取对照品溶液与供试品溶液各10μl,注入液相色谱仪,测定,即得。
本品含牛蒡子以牛蒡子苷(C27H34O11)计,每袋不得少于2.5mg。

采用以下步骤制备:分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材粉碎成细粉,备用;将粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,萃取压力为30Mpa,萃取温度为45℃,萃取时间为2h,提取出薄荷油和荆芥油等挥发油;将提取的挥发油用8倍量的β-环糊精,再加10倍量的水溶解,并于40℃搅拌2h进行包合,制成环糊精包合物,备用;将粉碎的金银花、连翘、牛蒡子、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十三味药材粉末一起加入8倍药材重量的水超声提取,声提取次数为2次,每次0.75h,超声波功率为60kw,温度为25℃,提取后过滤;将滤液合并,低温静置48h;静置后的清膏用微孔滤膜过滤,减压浓缩成60℃时相对密度为1.35的稠膏;将制备的环糊精包合物加入到该稠膏中混合,再加入辅料,一并混合均匀,制粒、干燥、整粒,分装,即得。
实施例3
处方组成:薄荷320kg、荆芥穗268kg、苦杏仁320kg、牛蒡子320kg份、黄芩320kg、桔梗268kg、前胡320kg、白芷108kg、栀子(炒)160kg、山楂(焦)108kg、六神曲(焦)108kg、麦芽(焦)108kg、芦根480kg、金银花480kg、连翘320kg,蔗糖110kg,糊精500kg,甜菊素4kg,共制成800000袋(2.5g/袋)。
采用以下步骤制备:分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材粉碎成细粉,备用;将粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,萃取压力为40Mpa,萃取温度为60℃,萃取时间为3h,提取出薄荷油和荆芥油等挥发油;将提取的挥发油用8倍量的β-环糊精,再加10倍量的水溶解,并于40℃搅拌2h进行包合,制成环糊精包合物,备用;将粉碎的金银花、连翘、牛蒡子、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十三味药材粉末一起加入8倍药材重量的水超声提取,声提取次数为2次,每次1h,超声波功率为80kw,温度为25℃,提取后过滤;将滤液合并,低温静置48h;静置后的清膏用微孔滤膜过滤,减压浓缩成60℃时相对密度为1.35的稠膏;将制备的环糊精包合物加入到该稠膏中混合,再加入辅料,一并混合均匀,制粒、干燥、整粒,分装,即得。

Claims (3)

1.一种用于治疗小儿感冒的药物制剂的制备方法,由下列重量份的原料制成,薄荷80-320份、荆芥穗67-268份、苦杏仁80-320份、牛蒡子80-320份、黄芩80-320份、桔梗67-268份、前胡80-320份、白芷27-108份、栀子(炒)40-160份、山楂(焦)27-108份、六神曲(焦)27-108份、麦芽(焦)27-108份、芦根120-480份、金银花120-480份、连翘80-320份,其特征在于,采用以下步骤制备:
①分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材粉碎成细粉,备用;
②将步骤①中粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,提取出薄荷油和荆芥油等挥发油;
③将步骤②中的挥发油用8倍量的β-环糊精,再加10倍量的水溶解,并于40℃搅拌2h进行包合,制成环糊精包合物,备用;
④将步骤①中粉碎的金银花、连翘、牛蒡子、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十三味药材粉末一起加入8倍药材重量的水超声提取,提取后过滤;
⑤将步骤④过滤的滤液合并,低温静置48h;
⑥将步骤⑤静置后的清膏用微孔滤膜过滤,减压浓缩成60℃时相对密度为1.25-1.35的稠膏;
⑦将步骤③制备的环糊精包合物和步骤⑥的稠膏混合,再加入辅料,一并混合均匀,制粒、干燥、整粒,分装,即得;
步骤②所述的超临界CO2流体萃取技术的萃取压力为20-45Mpa,萃取温度为25-60℃,萃取时间为1-3h;
步骤④所述的超声提取次数为2次,每次0.5-1h,超声波功率为40-80kw,温度为25℃。
2.根据权利要求1所述的一种用于治疗小儿感冒的药物制剂的制备方法,其特征在于:步骤⑦所述的加入的辅料为蔗糖、糊精、淀粉、甜菊素、微晶纤维素、硬脂酸镁的一种或多种。
3.根据权利要求1所述的一种用于治疗小儿感冒的药物制剂的制备方法,其特征在于:所述治疗小儿感冒的药物还可制备成口服液、散剂或片剂。
