技术领域
本发明涉及一种中药制剂的制备方法,具体为一种用于提高小儿感冒疗效的药物制剂的制备工艺。
背景技术
小儿感冒,即小儿急性上呼吸道感染。目前,主要采用西药和中药治疗。西药一般以抗生素、激素、非甾体类抗炎药为主,虽起效较快,但副作用较大,对患儿的健康成长会造成严重影响;而治疗小儿感冒的相关中药制剂,大多存在提取制备工艺相对简单粗放,药物起效较慢,治疗效果不理想,且患儿服药的顺应性差,制约临床的用药需求。
发明内容
本发明的目的是针对上面所述缺陷,提供一种能够起效迅速,且副作用小的用于提高小儿感冒疗效的药物制剂的制备工艺。
本发明的目的是通过以下技术方案予以实现的。
一种用于提高小儿感冒疗效的药物制剂的制备工艺,由下列重量份的原料制成,薄荷80-240份、荆芥穗67-201份、黄芩80-240份、苦杏仁80-240份、栀子(炒)40-120份、桔梗67-201份、前胡80-240份、白芷27-81份、山楂(焦)27-81份、六神曲(焦)27-81份、牛蒡子80-240份、麦芽(焦)27-81份、芦根120-360份、金银花120-360份、连翘80-240份,其特征在于,采用以下步骤制备:
①分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材粉碎成细粉,备用;
②将步骤①中粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,提取出薄荷油和荆芥油等挥发油,备用;
③将步骤①中粉碎的苦杏仁制取杏仁水,备用;
④将步骤①中粉碎的金银花、连翘、牛蒡子、黄芩、栀子(炒)、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十二味药材粉末一并加入6倍药材重量的水超声提取,提取后过滤;
⑤将步骤④过滤的滤液合并,低温静置48h;
⑥将步骤⑤静置后的药液过滤,减压浓缩成60℃时相对密度为1.10-1.20的清膏;
⑦将步骤⑥的清膏加入各种辅料,溶解,再加入步骤③的杏仁水混匀,静置,过滤,取上清液;
⑧将步骤⑦的上清液加入步骤②的薄荷油和荆芥油,并加入食用香精调节口感和气味,随加随搅拌,加蒸馏水稀释,滤过,分装,即得。
步骤②所述的超临界CO2流体萃取技术的萃取压力为30-60Mpa,萃取温度为25-45℃,萃取时间为0.5-2h。
步骤④所述的超声提取次数为2次,每次0.75-1.5h,超声波功率为40-80kw,温度为室温。
步骤⑦所述的加入的辅料为蔗糖、甜菊素、尼泊金乙酯的一种或多种。
所述治疗小儿感冒的药物还可制备成颗粒剂、散剂或糖衣片。
本发明的有益效果是:本发明通过超临界CO2流体萃取技术和超声波技术提取,萃取分离效果好,避免了药材中热敏性成分的降解,提取率高,有利于药材的充分利用,节约能源和成本,具有提取时间短、提取率高和无需加热等特点,且该产品制成口服合剂,起效较迅速,加入香精改善了口感,提高了患儿服药的顺应性,能够迅速有效地疏散风热,清热止咳,可常年用于小儿感冒发烧,汗出不爽,鼻塞流涕,咳嗽咽痛等症的治疗。
具体实施方式
以下通过具体实施例对本发明作进一步说明,应该理解的是,这些实施例仅用于说明本发明,并不限制本发明的范围。
实施例1。
处方组成:荆芥穗134kg、薄荷160kg、苦杏仁160kg、牛蒡子160kg份、芦根240kg、金银花240kg、黄芩160kg、桔梗134kg、前胡160kg、白芷54kg、栀子(炒)80kg、山楂(焦)54kg、六神曲(焦)54kg、麦芽(焦)54kg、连翘160kg,蔗糖80kg,甜菊素1.5kg,尼泊金乙酯1kg,共制成20000瓶(100ml/瓶)。
采用以下步骤制备:分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材粉碎成细粉,备用;将粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,萃取压力为50Mpa,萃取温度为40℃,萃取时间为1.5h,提取出薄荷油和荆芥油等挥发油,备用;将粉碎的苦杏仁制取杏仁水,备用;将粉碎的金银花、连翘、牛蒡子、黄芩、栀子(炒)、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十二味药材粉末一并加入6倍药材重量的水超声提取,声提取次数为2次,每次1h,超声波功率为60kw,温度为室温,提取后过滤;将过滤的滤液合并,4℃静置48h;将静置后的药液过滤,减压浓缩成60℃时相对密度为1.10-1.20的清膏;将清膏加入蔗糖、甜菊素和尼泊金乙酯,溶解,再加入杏仁水混匀,静置,过滤,取上清液;将上清液加入薄荷油和荆芥油,并加入食用香精调节口感和气味,随加随搅拌,加蒸馏水稀释,滤过,分装,即得。
鉴别:(1)取本品10ml,加水40ml稀释,用水饱和正丁醇提取2次,每次50ml,合并提取液,以浓氨水洗涤2次,每次50 ml,弃去浓氨水,正丁醇液再以正丁醇饱和的水洗涤2次,每次50 ml,正丁醇液水浴蒸干,残渣加甲醇1ml使溶解,作为供试品溶液。另取牛蒡子苷加甲醇制成每1ml含5mg的溶液和栀子苷加甲醇制成每1ml含2mg的溶液,作为对照品溶液。照薄层色谱法(中国药典2015版)试验,吸取上述供试品溶液和对照品溶液各5μl,分别点于同一以含羧甲基纤维素钠为黏合剂的硅胶G薄层板上,以三氯甲烷-甲醇(9 : 2)为展开剂,展开,取出,晾干,喷以10%硫酸乙醇溶液,在105℃加热至斑点显色清晰。供试品色谱中,在与对照品色谱相应的位置上,显相同颜色的斑点。

供试品溶液的制备 取本品,精密量取2ml,置50ml量瓶中,加甲醇稀释至刻度,摇匀,取溶液高速离心15分钟(15000转/分),取上清液,即得。
测定法 分别精密吸取对照品溶液与供试品溶液各10μl,注入液相色谱仪,测定,即得。
本品每1ml含黄芩以黄芩苷(C21H18O11)计,不得少于1.2mg。
实施例2。
处方组成:荆芥穗67kg、薄荷80kg、苦杏仁80kg、牛蒡子80kg份、芦根120kg、金银花120kg、黄芩80kg、桔梗67kg、前胡80kg、白芷27kg、栀子(炒)40kg、山楂(焦)27kg、六神曲(焦)27kg、麦芽(焦)27kg、连翘80kg,蔗糖45kg,甜菊素1kg,尼泊金乙酯0.5kg,共制成10000瓶(100ml/瓶)。
采用以下步骤制备:分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材粉碎成细粉,备用;将粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,萃取压力为30Mpa,萃取温度为25℃,萃取时间为0.5h,提取出薄荷油和荆芥油等挥发油,备用;将粉碎的苦杏仁制取杏仁水,备用;将粉碎的金银花、连翘、牛蒡子、黄芩、栀子(炒)、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十二味药材粉末一并加入6倍药材重量的水超声提取,声提取次数为2次,每次0.75h,超声波功率为40kw,温度为室温,提取后过滤;将过滤的滤液合并,4℃静置48h;将静置后的药液过滤,减压浓缩成60℃时相对密度为1.10-1.20的清膏;将清膏加入蔗糖、甜菊素和尼泊金乙酯,溶解,再加入杏仁水混匀,静置,过滤,取上清液;将上清液加入薄荷油和荆芥油,并加入食用香精调节口感和气味,随加随搅拌,加蒸馏水稀释,滤过,分装,即得。
实施例3。
处方组成:荆芥穗201kg、薄荷240kg、苦杏仁240kg、牛蒡子240kg份、芦根360kg、金银花360kg、黄芩240kg、桔梗201kg、前胡240kg、白芷81kg、栀子(炒)120kg、山楂(焦)81kg、六神曲(焦)81kg、麦芽(焦)81kg、连翘240kg,蔗糖1250kg,甜菊素2kg,尼泊金乙酯1.5kg,共制成31000瓶(100ml/瓶)。
采用以下步骤制备:分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材粉碎成细粉,备用;将粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,萃取压力为60Mpa,萃取温度为45℃,萃取时间为2h,提取出薄荷油和荆芥油等挥发油,备用;将粉碎的苦杏仁制取杏仁水,备用;将粉碎的金银花、连翘、牛蒡子、黄芩、栀子(炒)、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十二味药材粉末一并加入6倍药材重量的水超声提取,声提取次数为2次,每次1.5h,超声波功率为80kw,温度为室温,提取后过滤;将过滤的滤液合并,4℃静置48h;将静置后的药液过滤,减压浓缩成60℃时相对密度为1.10-1.20的清膏;将清膏加入蔗糖、甜菊素和尼泊金乙酯,溶解,再加入杏仁水混匀,静置,过滤,取上清液;将上清液加入薄荷油和荆芥油,并加入食用香精调节口感和气味,随加随搅拌,加蒸馏水稀释,滤过,分装,即得。
实施例4。
处方组成:荆芥穗134kg、薄荷160kg、苦杏仁160kg、牛蒡子160kg份、芦根240kg、金银花240kg、黄芩160kg、桔梗134kg、前胡160kg、白芷54kg、栀子(炒)80kg、山楂(焦)54kg、六神曲(焦)54kg、麦芽(焦)54kg、连翘160kg,淀粉150kg,微晶纤维素100kg,硬脂酸镁30kg,蔗糖80kg,共制成2100000糖衣片(0.5g/片)。
采用以下步骤制备:分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、栀子(炒)、苦杏仁、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十五味中药材粉碎成细粉,备用;将粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,萃取压力为50Mpa,萃取温度为40℃,萃取时间为1.5h,提取出薄荷油和荆芥油等挥发油,备用;将粉碎的苦杏仁制取杏仁水,备用;将粉碎的金银花、连翘、牛蒡子、黄芩、栀子(炒)、桔梗、前胡、白芷、山楂(焦)、六神曲(焦)、麦芽(焦)、芦根等十二味药材粉末一并加入6倍药材重量的水超声提取,声提取次数为2次,每次1h,超声波功率为60kw,温度为室温,提取后过滤;将过滤的滤液合并,4℃静置48h;将静置后的药液过滤,减压浓缩成60℃时相对密度为1.10-1.20的清膏;再加入杏仁水混匀,静置,过滤,取上清液;将上清液加入薄荷油和荆芥油,加入淀粉、微晶纤维素,一并混合均匀,湿法制粒,加入硬脂酸镁,压片,包糖衣,即得糖衣片。

Claims (5)

1.一种用于提高小儿感冒疗效的药物制剂的制备工艺,由下列重量份的原料制成,薄荷80-240份、荆芥穗67-201份、黄芩80-240份、苦杏仁80-240份、炒栀子40-120份、桔梗67-201份、前胡80-240份、白芷27-81份、焦山楂27-81份、焦六神曲27-81份、牛蒡子80-240份、焦麦芽27-81份、芦根120-360份、金银花120-360份、连翘80-240份,其特征在于,采用以下步骤制备:
①分别将金银花、连翘、牛蒡子、薄荷、荆芥穗、黄芩、炒栀子、苦杏仁、桔梗、前胡、白芷、焦山楂、焦六神曲、焦麦芽、芦根十五味中药材粉碎成细粉,备用;
②将步骤①中粉碎的薄荷、荆芥穗细粉,采用超临界CO2流体萃取技术,提取出薄荷油和荆芥油等挥发油,备用;
③将步骤①中粉碎的苦杏仁制取杏仁水,备用;
④将步骤①中粉碎的金银花、连翘、牛蒡子、黄芩、炒栀子、桔梗、前胡、白芷、焦山楂、焦六神曲、焦麦芽、芦根十二味药材粉末一并加入6倍药材重量的水超声提取,提取后过滤;
⑤将步骤④过滤的滤液合并,低温静置48h;
⑥将步骤⑤静置后的药液过滤,减压浓缩成60℃时相对密度为1.10-1.20的清膏;
⑦将步骤⑥的清膏加入各种辅料,溶解,再加入步骤③的杏仁水混匀,静置,过滤,取上清液;
⑧将步骤⑦的上清液加入步骤②的薄荷油和荆芥油,并加入食用香精调节口感和气味,随加随搅拌,加蒸馏水稀释,滤过,分装,即得。
2.根据权利要求1所述的一种用于提高小儿感冒疗效的药物制剂的制备工艺,步骤②所述的超临界CO2流体萃取技术的萃取压力为30-60Mpa,萃取温度为25-45℃,萃取时间为0.5-2h。
3.根据权利要求1所述的一种用于提高小儿感冒疗效的药物制剂的制备工艺,步骤④所述的超声提取次数为2次,每次0.75-1.5h,超声波功率为40-80kw,温度为室温。
4.根据权利要求1所述的一种用于提高小儿感冒疗效的药物制剂的制备工艺,其特征在于:步骤⑦所述的加入的辅料为蔗糖、甜菊素、尼泊金乙酯的一种或多种。
5.根据权利要求1所述的一种用于提高小儿感冒疗效的药物制剂的制备工艺,其特征在于:所述治疗小儿感冒的药物还可制备成颗粒剂、散剂或糖衣片。
