实施例3. E. coli/pETPp-asd全细胞高密度转化生产L-丙氨酸
将富马酸加入转化罐中,加入一定量的水,然后边搅拌边加入氨水,并以pH计测定溶液pH变化情况,待富马酸完全溶解,再以氨水调节pH为7.0 – 8.5,控制富马酸终浓度0.5 – 1.2 摩尔/升。将发酵培养好的E. coli/pETPp-asd菌液离心收集,生理盐水悬浮,加入转化罐中,至菌体密度OD600 = 10 – 20。调节温度40 – 50oC,搅拌转速50 – 200 rpm进行转化。定期取样,高效液相色谱定量检测L-丙氨酸的生产(附图3)。9小时,L-丙氨酸的产量可达112.7 g/L,生产速率12.5 g/(L·h),转化率93.9%。
