水解动物蛋白(HAP)是一种优质的蛋白源,蛋白质含量高达90%以上,主要成分为低分子多肽。HAP是良好的天然生理活性物,水溶性好,营养价值高,不含胆固醇,具有很好的耐酸、耐碱、耐热性和保水性,乳化性,不易产生沉淀;有良好的抗氧化性,不易褐变和变性,可以稳定和保护易变质的食品,能够有效改善和调整食品结构、风味和品质。HAP氨基酸组成和阿胶类似,能够迅速提供能量,促进新陈代谢和生长发育,加速血红蛋白合成,促进细胞生长和伤口愈合。
牛鲜骨是一种十分宝贵的资源,内含有丰富的营养物质,含蛋白质35.7%、脂肪10.3%、灰分45%、水分4.8%,其它物质4.2%。可以通过酶解牛骨获取水解蛋白,并通过控制蛋白质的降解程度,从而HAP的色泽、风味、口感以及营养价值,但是采用酶解法水获取水解动物蛋白HAP只利用了鲜骨中的部分蛋白质,鲜骨中尚还有脂肪、矿物质(如磷、钙、铁等)、骨胶原、软骨素以及维生素A、B1、B2等,其进一步利用的新途径值得做进一步的探讨和研究,而骨中含量很高的磷酸盐和骨髓中营养价值很高的磷脂肪和磷蛋白等的利用情况更亟需改进。

一种用于老人保健的高钙大豆牛骨蛋白饮料,其特征在于,包括以下重量份组分:鲜牛骨82-102、大豆分离蛋白粉40-50、植脂末8-10、异麦芽低聚糖3-4、维生素A0.6-1、维生素E0.5-0.9、维生素C0.8-1.2、香精0.3-0.5、木瓜蛋白酶1-1.4、胰蛋白酶1.1-1.5、酵母菌0.6-0.8、乳酸菌0.5-0.7、抗坏血酸0.4-0.8、适量的乙醇溶液、适量的去离子水和适量的水。
一种用于老人保健的高钙大豆牛骨蛋白饮料制备方法,其特征在于,包括以下步骤:
(1))取鲜牛骨用水洗干净后,放入破碎机中破碎至2-3cm长得牛骨段,之后放入高压锅中,加入总重量3-5倍的水后,在低压2.5-3.5KPa作用下,小火蒸煮3-4h,冷却后,分离得牛骨段、牛骨汤和牛骨油脂备用;
(2)取牛骨段放入真空微波炉中在100W微波中干燥30-40min,之后放入粉碎机中,粉碎后过100目得牛骨颗粒备用;将牛骨颗粒中加入胰蛋白酶以及总重量5-6倍的去离子水后,在40-50℃下快速搅拌酶解20-30min,之后过滤得牛骨蛋白酶解液一和牛骨残渣备用;将牛骨汤、木瓜蛋白酶混合并加入总重量3-4倍的去离子水后,在30-40℃下快速搅拌酶解30-40min,得酶解液二备用;
(3)将牛骨蛋白酶解液一、大豆分离蛋白粉、植脂末混合,并加入总重量6-8倍的去离子水搅拌均匀后,在92-95℃下灭酶5-15min,之后接入酵母菌在40-50℃下避光发酵10-12d,得发酵液一备用;
(4)将酶解液二、异麦芽低聚糖混合,并加入总重量6-8倍的去离子水搅拌均匀后,在92-95℃下灭酶5-15min,之后接入乳酸菌菌在30-35℃下避光发酵10-12d,得发酵液二备用;
(5)取牛骨残渣在真空微波炉中采用1100W微波加热9-18min后,放入乙醇溶液在120℃下加热活化3-4min,之后放入球磨机中超微粉碎为粒径90um得骨活化矿质粉备用;
(6)将发酵液一、发酵液二混合之后,静置2-3d,之后加入骨活化矿质粉、维生素A、维生素E、维生素C、香精以及抗坏血酸混合均质,经巴氏消毒装灌即得。

(3)将牛骨蛋白酶解液一、大豆分离蛋白粉、植脂末混合,并加入总重量6-8倍的去离子水搅拌均匀后,在92-95℃下灭酶5-15min,之后接入酵母菌在40-50℃下避光发酵10-12d,得发酵液一备用;
(4)将酶解液二、异麦芽低聚糖混合,并加入总重量6-8倍的去离子水搅拌均匀后,在92-95℃下灭酶5-15min,之后接入乳酸菌菌在30-35℃下避光发酵10-12d,得发酵液二备用;
(5)取牛骨残渣在真空微波炉中采用1100W微波加热9-18min后,放入乙醇溶液在120℃下加热活化3-4min,之后放入球磨机中超微粉碎为粒径90um得骨活化矿质粉备用;
(6)将发酵液一、发酵液二混合之后,静置2-3d,之后加入骨活化矿质粉、维生素A、维生素E、维生素C、香精以及抗坏血酸混合均质,经巴氏消毒装灌即得。
