实施例2、谷氨酸棒杆菌(Corynebacterium glutamicum)重组菌株SY12在生产D-阿洛酮糖中的应用
1、谷氨酸棒杆菌重组菌株SY12的培养及诱导
选用CGXII培养基(配方:(NH4)2SO4(5g/L),
urea (5g/L), KH2PO4 (1g/L), K2HPO4
(1g/L), MgSO4∙7H2O (0.25g/L), CaCl2
(10mg/L), FeSO4∙7H2O (10mg/L), MnSO4∙H2O
(0.1mg/L), ZnSO4∙7H2O (1mg/L), CuSO4∙5H2O
(0.2mg/L), NiCl2∙6H2O (20μg/L), 生物素 (0.4mg/L), MOPS (42g/L) (pH 7.4)),培养基中添加葡萄糖(10g/L)和氯霉素(12.5mg/L),在30℃、200rmp条件下对谷氨酸棒杆菌重组菌株SY12进行培养,当OD600达到0.6-0.8时,添加IPTG,终浓度为1mM,降低摇床转速为120rmp,诱导约12h。
2、谷氨酸棒杆菌重组菌株SY12的收集和浓缩
将诱导得到的谷氨酸棒杆菌重组菌株SY12菌液(100mL),4℃、8000rmp离心15min收集菌体,用CGXII培养基洗涤菌液两次,最终用CGXII培养基浓缩菌液至10mL。
3、D-阿洛糖的发酵生产
取10mL谷氨酸棒杆菌重组菌株SY12的浓缩菌液,放置于50mL的锥形瓶中,加入终浓度为2%(质量/体积(W/V,单位g/100mL)百分比浓度)的葡萄糖和1%(质量/体积(W/V,单位g/100mL)百分比浓度)的D-甘油醛进行发酵,发酵条件为:温度30℃,pH 7.0,菌体浓度(OD600)为30。
发酵结束后,对样品进行
14000rmp离心20min,并用0.22μm的微孔滤膜过滤,滤液做高效液相分析。高效液相色谱分析按如下条件进行:仪器为安捷伦高效液相色谱仪1200,分析柱:Xtimate Sugar-Ca,流动相:EDTA-Ca(0.1mM),流速:0.4
mL/min,柱温:70℃,检测器:示差折光检测器。以Sigma公司生产的D-阿洛酮糖纯品为标准品,上样量为20μl。
结果如图3((a)表示D-阿洛酮糖标品;(b)表示发酵液)所示,可以看出,经过24小时反应,发酵液中D-阿洛糖的浓度为10.6
g/L, D-甘油醛转化率为53%,发酵液中未检测到剩余葡萄糖和D-甘油醛,表明谷氨酸棒杆菌重组菌株SY12可以以D-甘油醛为底物合成单一产物D-阿洛酮糖。
目前通过异构酶以果糖为底物合成阿洛酮糖,仅有25-30%的转化率,且产物不容易分离,本发明的谷氨酸棒杆菌重组菌株SY12在生物合成D-阿洛酮糖方面,具有较高转化率,且整个发酵过程采用基本盐培养基,成本低廉,产物便于分离纯化,具有广阔的应用前景和竞争力。
