发明内容

RNA干扰是一个功能强大的工具,其利用短RNA双链来抑制细胞中的特定蛋白质的形成(1-3)。在自然界中,沉默RNA分子由更大的转录物被Dicer复合物切割而产生(4)。然而对于生物技术应用,RNA分子(siRNA)是化学制备并施用的。将siRNA用作治疗剂的想法(5)在过去十年里被强烈地追求,但最主要的障碍是RNA双链体的差细胞摄取不能被克服(6)。目前,集中研究RNA递送系统,不同的如纳米粒子(7,8)、脂质体(9,10)或聚阳离子聚合物(11)。尽管在该领域有重大进展,然而,经常仍然是高毒性(12-14)和低细胞特异性,这代表未解决的问题。

缀合物包含至少一个核苷组分,特别是1-25,更特别是2-20或2-10,和甚至更特别是2-8,即2、3、4、5、6、7或8个核苷组分。如果缀合物包含多于一个核苷组分,核苷组分可以是相同的或不同的。
在进一步的具体实施方式中,缀合物包含1个多不饱和脂肪酸残基,例如大麻素残基和2-20,特别是2-8,即2、3、4、5、6、7或8个核苷组分。
在一个实施方式中,缀合物可以具有线性结构。因此,核苷组分可以连接在线性链中,其中多不饱和脂肪酸残基可以存在于链中,在链的一端或链的两端。
在另一个实施方式中,缀合物具有分枝的结构,其中通过分枝的连接体将核苷组分结合至多不饱和脂肪酸残基,所述连接体例如树枝状连接体。

在一个实施方式中,核酸分子是其包含至少一个经修饰的构建区块的DNA分子。术语“DNA分子”包括单链或双链DNA分子。在双链DNA分子中,各链可以以单独的分子存在或者通过单链环或通过异源连接体而共价连接。
术语“DNA分子”包括由天然DNA构建区块即2'-脱氧核苷酸构建区块构成的分子,和包含至少一个经修饰的构建区块的分子。
在进一步的实施方式中,核酸分子是RNA分子,其可包含至少一个经修饰的构建区块。术语“RNA分子”包括单链或双链RNA分子,其中双链RNA分子具有至少一个突出,例如至少一个3'-突出。在双链RNA分子,各链可以以单独的分子存着或者通过单链环或通过异源连接体而共价连接。

经修饰的构建区块可以选自糖-、主干-和/或核碱基-修饰的构建区块。糖-修饰的脱氧核糖核苷酸包含与脱氧核糖不同的糖基团,例如经修饰脱氧核糖基团,其中2'-H基团被选自OH、R、OR、卤素、SH、SR、NH2、NHR,NR2或CN的基团取代,其中R是C1-C6烷基或烷氧基,或者C2-C6烯基或炔基,并且卤素是F、Cl、Br、I。2'-H修饰的具体例子都是2'-F和2'-O甲基。糖修饰的核苷酸包含不同于核糖的糖基团,例如经修饰的核糖基团,其中2'-OH基团被选自H、R、OR、卤素、SH、SR、NH2、NHR,NR2或CN的基团取代,其中R是C1-C6烷基或烷氧基,或者C2-C6烯基或炔基,并且卤素是F、Cl、Br、I。2'-OH修饰的具体例子是2'-F和2'-O甲基。在主干-修饰的构建区块中,连接相邻的构建区块的磷酸酯基团可以被经修饰的连接基团例如硫代磷酸酯基团所取代。在核碱基-修饰的构建区块中,可以存在非天然发生的核碱基而不是天然发生的核碱基。嘌呤或嘧啶核碱基的相应类似物是本领域所熟知的。应指出的是可以组合上述修饰。
核酸分子优选地选自适合于药学应用的核酸分子,特别是反义分子,或者选自能够调节RNA干扰的RNA分子如siRNA分子或其前体。其他合适的RNA分子包括miRNA分子、antagomirs、核酶及其前体。
术语“核苷组分”还包括核苷或核苷酸及其类似物。核苷是包含核碱基和糖基团的化合物。核苷酸是包含核碱基、糖基团和磷酸基团的化合物组。糖-、磷酸-和核碱基-修饰的化合物也包括在本发明中,特别是适用于治疗癌症和/或病毒感染的核苷或核苷酸的类似物治疗剂,例如AZT、阿昔洛韦、更昔洛韦、伐昔洛韦、吉西他滨、阿糖胞苷等。
核苷组分可以通过分子的核碱基、糖或磷酸基团连接至脂肪酸残基。如果这种化合物是核酸,它可通过核酸分子中存在的构建区块进行连接,特别是通过末端构建区块,即位于核酸链的5'或3'-末端的构建区块,更特别是通过核酸链的3'-末端构建区块。在优选的实施例中,通过存在于核酸分子特别是RNA分子中的经修饰的末端核碱基而发生连接。
在优选的实施例中,与多不饱和脂肪酸残基的共价连接可以附着至核苷组分中存在的核碱基,例如DNA或RNA分子的构建区块,例如至嘌呤碱基的位置8或嘧啶碱基的位置5。
核酸分子,如DNA或RNA的分子,通常具有长度为从5、10、12、15或18个构建区块和多至25、30、50或100个构建区块或更多。核酸分子可以通过化学合成法或通过酶法从核酸模板来制备,例如通过转录,由RNA聚合酶的催化,如由T3、T7或SP6 RNA聚合酶,或通过DNA复制或通过逆转录。优选地,化学或酶促合成过程中,并入构建区块包含功能基团,例如Click-功能基团,如末端炔烃基团或叠氮化物基团,受限的烯烃基团,例如降冰片烯基团、腈氧化物基团、丁腈亚胺基团或四嗪基团。在特定的实施方式中,并入通过(任选地通过连接体)包括末端炔烃基团而修饰的构建区块。WO2006/117161和WO2008/052775描述了将Click修饰的构建区块并入核酸分子的方法,其内容通过引用并入本文。根据已知的方法,核苷组分上的功能基团可以缀合至互补的功能基团,其附着至多不饱和脂肪酸残基。优选地,通过例如Click-反应与互补的Click-功能反应基团,例如叠氮化物组进行缀合。
备选地,根据标准方法在固相合成中,例如使用亚磷酰胺构建区块,可将连接至多不饱和脂肪酸残基的经修饰的核酸构建区块引入核酸,例如RNA分子。

接触步骤(a)可以包括将缀合物引入靶细胞,例如分离的靶细胞,其可以是存在于细胞培养物中,单细胞的微生物,或者在多细胞生物中的靶细胞或多个靶细胞。靶细胞优选地是哺乳动物细胞,包括人细胞。靶生物体优选地是哺乳动物生物体,如人生物体。引入生物体可以包括胃肠外施用,例如通过注射或输液,经粘膜施用或经皮施用。
调节步骤(b)包括抑制靶核酸,例如当使用siRNA缀合物时通过RNA干扰,或当使用反义分子缀合物时通过抑制mRNA转录,或通过使用治疗性核苷/核苷酸缀合物对病毒或肿瘤细胞复制的抑制。
优选地通过受体-介导的内吞作用,更优选地通过大麻素受体-介导的内吞作用将缀合物引入靶细胞。因此,可以在缺少递送媒介物和/或转染试剂的情况下将缀合物可以引入靶。
在一个实施方式中,本发明的缀合物是用于体外转染细胞,特别是用于体外转染哺乳动物细胞,包括人细胞。惊奇地发现本发明的缀合物是特别适合于转染免疫细胞如B细胞、T细胞、巨噬细胞、自然杀伤细胞及其前体,以及神经元细胞、星形胶质细胞或其他表达大麻素受体的细胞。表达大麻素受体CB2的免疫细胞的实例描述在(49、50和51)中,其内容通过引用并入本文。表达大麻素受体CB1的神经元细胞的实例描述在(52、53、54和55)中,其内容通过引用并入本文。
在进一步的具体实施方式中,本发明的缀合物用于医学,特别是人类医学,但也用于兽医学。因此,本发明还提供了药物组合物,其包含如上文所述的缀合物作为活性成分以及合适的载体。对于诊断或治疗应用,所述药物组合物可以为溶液的形式,例如用于输液或注射的溶液、乳剂、软膏、片剂、悬浮液等等。可以以任何合适的方式施用组合物,例如通过胃肠外施用,如注射或输液,通过透粘膜应用,或通过透皮应用,或通过口服、局部、鼻、直肠应用等。
所述药物组合物可包含以非封装形式的所述缀合物作为活性剂,例如没有递送媒介物如脂质体和/或没有转染试剂。
本发明的缀合物可以用于细胞或生物体中基因的下调,例如病毒基因或细胞疾病相关基因,如癌基因,或者自身免疫或过敏性疾病相关基因。优选的细胞靶基因是例如syk基因,其是一种自身免疫或过敏性疾病相关的基因,编码脾酪氨酸激酶(SYK),参与IgE依赖性炎症信号传导级联。人SYK直系同源物描述在UniProt P 43405,小鼠SYK直系同源物描述在UniProt P 48025。另一个优选的靶基因是APP基因,其编码淀粉样前体蛋白(APP)。人APP直系同源物描述在UniProt P 05067。APP被β-或γ分泌酶切割成神经毒性碎片,与阿尔茨海默氏症的发展相关联。优选的病毒靶基因是编码单股反链病毒目的病毒例如埃博拉病毒、麻疹病毒和狂犬病病毒的N或P蛋白的基因。

实施例
1.RNA-配体缀合物的合成
如图1所示进行大麻素修饰的RNA链的合成。合成的中心内容是炔烃修饰的RNA链和相应配体叠氮化合物1之间的Cu-催化的炔烃-叠氮化物click反应(34-39)。为了比较大麻素修饰的RNA链与其他系统,我们利用click方法也用于使用叶酸叠氮化物2制备叶酸-RNA(40)缀合物和用于使用胆固醇叠氮化物3制备胆固醇修饰的RNA链。我们在所有情况下引入RNA链和各自配体之间的短四乙二醇间隔体。click技术在所有情况下能够有效连接疏水性的和常常是非常难溶的(叶酸)配体分子至RNA。此外,该方法使得能够以更难地有效缀合至siRNA双链体的3'-末端。3'-修饰的siRNA链通常更耐受RNAi机制(41)。为了实现3'-末端附着,我们在RNA合成中使用在C5具有辛二烯(octadiine)柄的脱氧尿嘧啶亚磷酰胺。
在仅仅一步中从花生四烯酸4以及叠氮基-和氨基-官能化的寡聚乙二醇5制备大麻素叠氮化物配体1。采用相同的策略用于合成胆固醇叠氮化物3。通过更精细的合成来制备叶酸衍生物2,起始于受保护的谷氨酸衍生物6,其使用氨基-叠氮化物四乙二醇化合物5进行缩合。在叶酸2去保护之后,提供Fmoc基团的切割以及与蝶酸的缀合。化合物2以这种方式包含附着至γ-羧基基团的乙二醇间隔体,其提供了具有优越的受体结合特性的叶酸化合物(42)。三个叠氮化物随后以优异的产率与含有RNA正义链的炔烃进行click。在HPLC纯化后,配体修饰的RNA杂交到反义的相对链从而获得siRNA双链体,如图2中所示。
2.递送RNA-配体缀合物至细胞中
为了显示递送RNA双链体至活细胞中,我们最初将大麻素-和叶酸-修饰的RNA正义链杂交至含有荧光素标签的反义链。图3显示了使用两个不同细胞系执行的共聚焦显微镜研究的结果。对于大麻素修饰的RNA双链体,我们利用RBL-2H3细胞,其作为用于免疫细胞功能的模型(43)。Barker等人能够显示RBL-2H3细胞对大麻素的摄取与神经元细胞和星形胶质细胞是功能上等同的(44)。因此该细胞系是免疫细胞和神经元中大麻素摄取的极好模型。
用已知过表达叶酸受体的HeLa癌细胞研究对叶酸修饰的RNA双链体的摄取。显微镜研究表明,未修饰的siRNA按预期无法进入这两个细胞系。然而在各细胞中容易地检测到大麻素和叶酸修饰的siRNA证明存在摄取。使用经修饰的dsDNA也观察到相同的结果,这表明摄取完全是配体依赖的(图3)。
为了证明递送的siRNA分子表现出预期的RNAi作用,我们利用商业上可得的双荧光素酶报告子测定法。包含两个荧光素酶(海肾和萤火虫)的质粒转染入细胞。通过靶向海肾荧光素酶的表达来评价RNAi,而萤火虫荧光素酶作为内部标准。对于这些研究,我们使用配体修饰的siRNA而无进一步的荧光素修饰。使用未修饰的RNA双链体(无配体,无荧光素)的初始对照实验表明海肾表达不受影响。与此相反的是,我们观察到在两种细胞系中存在配体修饰的siRNA时,海肾表达的剂量依赖性沉默(图4)。最重要的是,即使相对少量的siRNA-配体缀合物已经显示相当大的影响,最终导致大约60%左右的相对沉默。
用混杂的siRNA进行最后的对照实验。再一次地,我们无法检测到任何发光减少,表明观察到的沉默是由大麻素修饰的siRNA与mRNA靶的特异性结合所引起的。
下一步评价与胆固醇-siRNA缀合物相比,大麻素修饰的siRNA的沉默功效(24)。此比较的结果如图4所示。令我们吃惊的是,我们注意到新的大麻素1修饰的siRNA一直比广泛利用的胆固醇系统明显更有效,这确立了大麻素配体作为一个强大的新的递送工具。
为了研究大麻素-siRNA缀合物是否能够向下调节治疗重要的内源基因产物,我们下一步试图抑制脾酪氨酸激酶(SYK)的表达,脾酪氨酸激酶是参与IgE依赖性炎症信号传导级联的一种关键蛋白。因此,该蛋白是治疗过敏性和炎症性疾病(45,46)的预期靶标。对于实验,我们制备大麻素修饰的siRNA,其序列如Sanderson等人描述(47)。将siRNA缀合物添加至RBL-2H3细胞后,我们使用实时PCR监视表达水平。事实上,SYK蛋白的表达成功减少了约55%(图4),并且再次发现大麻素缀合物比胆固醇修饰的siRNA显著地更有活性。
总之,我们在这里报告了使用Cu-催化的炔烃-叠氮化物化学来构建新的大麻素siRNA缀合物。所述化学使得能够以优异的产率和纯度在3'-末端有效构建不同配体修饰的RNA。对于很难获得的经叶酸修饰的寡核苷酸,这是特别值得一提的。大麻素缀合物允许转染免疫细胞,并提供优异的沉默数据。我们认为大麻素配体,与click化学组合,具有成为转染神经细胞和免疫细胞的新的黄金标准的潜力。
3.对狂犬病病毒蛋白质的调节
显示大麻素siRNA调节狂犬病病毒蛋白质。将siRNA针对病毒株SAD L16的N-和P-蛋白(...)。通过病毒滴定法和生长图,显示mRNA敲减对狂犬病病毒的整个生命周期产生负面影响。
用狂犬病病毒感染细胞及随后转染不同的大麻素-siRNA构建体后,测定细胞培养基中的病毒滴度(ffu/mL)。发现病毒滴度的减少高达99%。
用BJAB细胞(人伯基特淋巴瘤B细胞)、白血病细胞(人T细胞白血病细胞)和E14细胞(小鼠皮层神经元)进行了实验。
4.对蛋白质FKBP51的调节
在分离的人PBMC细胞中,测试针对蛋白FKBP51的mRNA的大麻素-siRNA缀合物进行测试。FKBP51调节甾体激素受体的信号转导,并且其尤其与某些精神疾病以及阿尔茨海默病相关。
实验显示了70%的蛋白质敲减。

Claims (24)
1.包含至少一个多不饱和脂肪酸残基和与其共价结合的至少一个核苷组分的缀合物,其中所述核苷组分选自核酸、核苷和核苷酸。

L是连接体,
N是选自核酸、核苷和核苷酸的核苷组分,
n是1-10,优选地1-5,更优选地1的整数,
m是0或1,
r是1-25,优选地2-20和更优选地2-8的整数。
3.权利要求1的缀合物,其包含其他配体,所述配体共价结合至至少一个核苷组分。
4.权利要求3的缀合物,其具有通式(II)
Fn—(Lm—N)r—Lm—Zs (II)
其中
F是多不饱和脂肪酸残基,
L是连接体,
n是1-10,优选地1-5,更优选地1的整数,
m是0或1,
N是选自核酸、核苷和核苷酸的核苷组分,
r是1-25,优选地2-20和更优选地2-8的整数
Z是其他受体配体,和
s是1-10,优选地1-5,更优选地1的整数。
5.权利要求1-4中任一项的缀合物,其中所述多不饱和脂肪酸残基是具有3、4、5或6个并且优选地4个C=C键的多不饱和C16-C24脂肪酸的残基,其中优选地至少1-5,更优选地4个C=C键是以Z构型,优选地选自顺-5,8,11,14-二十碳四烯酸(花生四烯酸)残基,顺-5,8,11,14,17-二十碳五烯酸(EPA)残基,顺-4,7,10,13,16,19-二十二碳六烯酸(DHA)残基、顺-6,9,12-十八碳三烯酸(GLA)残基和顺-8,11,14-二十碳三烯酸(DGLA)残基。
6.权利要求5的缀合物,其中所述多不饱和脂肪酸残基是花生四烯酸残基。
7.权利要求1-6中任一项的缀合物,其中所述脂肪酸残基选自游离脂肪酸残基,脂肪酸酯或脂肪酸酰胺,脂肪酸磺酸酯,脂肪酸硫酸酯、脂肪酸膦酸酯或脂肪酸磷酸酯的残基。
8.权利要求1-7中任一项的缀合物,其中所述多不饱和脂肪酸残基通过连接体共价结合至至少一个核苷组分。
9.权利要求8的缀合物,其中所述连接体包含通过Click反应形成的环基团,特别是1,2,3-三氮唑基团,通过Click反应在炔烃和叠氮化物之间形成,或者通过Click反应在降冰片烯和腈亚胺之间形成的基团,氧化腈或四嗪。
10.权利要求1-9中任一项的缀合物,其中所述核苷组分是核酸分子,特别是单链或双链RNA分子,任选地具有至少一个3'-突出,更特别地是siRNA分子。
11.权利要求1-10中任一项的缀合物,其中所述核苷组分是包含至少一个经修饰的构建区块的核酸分子。
12.权利要求1-11中任一项的缀合物,其中所述核苷组分通过核碱基、糖或磷酸基团,特别是存在于核酸分子中的构建区块,特别是通过存在于核酸分子中的末端构建区块连接至脂肪酸残基。
13.权利要求1-12中任一项的缀合物,用于体外转染细胞,特别是用于体外转染哺乳动物细胞包括人细胞的用途。
14.权利要求1-12中任一项的缀合物,用于根据权利要求13的用途,其中所述细胞是表达大麻素受体的细胞,特别是免疫细胞、神经元细胞和星形胶质细胞。
15.权利要求1-12中任一项的缀合物,用于医学的用途,特别是人类医学的用途。
16.权利要求1-12中任一项的缀合物,用于根据权利要求13、14或15的用途,其中用于下调基因,例如病毒基因或细胞疾病相关基因,例如原癌基因或自免疫疾病相关基因。
17.用于制备具有通式(V)的RNA-配体缀合物的试剂:

其中
F、n、m和r是如权利要求1-5中任一项所定义的,
L'是连接体,和
RG1是反应基团,特别是Click反应基团,例如叠氮化物基团。
18.用于制备具有通式(VI)的核酸缀合物的试剂:
BB—(L)m—F (VI)
其中
F、L、n和m是如权利要求1-5中任一项所定义的,和
BB是用于合成核酸分子的构建区块如核苷三磷酸,或适用于固相合成的构建区块如亚磷酰胺。
19.用于制备权利要求1-12中任一项的缀合物的方法,包括
(i)权利要求18的试剂与至少一个经修饰的RNA分子(VII)进行缀合
(N)r—(L")m-RG2 (VII)
其中
N、r和n是如权利要求1-5中任一项所定义的,
L"是连接体,
m是0或1,和
RG2是这样的反应基团,能够与RG1,特别是Click反应基团如炔烃基团反应,从而形成缀合物,或者
(ii)权利要求17的试剂与至少一个经修饰的核酸构建区块(VIII)进行缀合

23.权利要求21、22或23的缀合物,具有如权利要求2-12中任一项定义的结构,条件是F(如权利要求2和4所定义的)是任意C=C不饱和化合物。
24.权利要求20-23任一项的缀合物用于如权利要求13-16任一项定义的用途。
