自明清以来,中医一直认为肾为脏腑之本,十二经之根,先天之本在于肾。肾与命门本同一气,为人身阴阳消长之枢纽。肾阳主导一身之阳气,火衰其本则阳虚之证迭出。肺肾阴虚是指因外邪入里化热伤阴,或内伤喘嗽,久咳不已,阴液受损,或房事过度,情志不遂,阴液暗伤等均可导致肺肾阴虚证。肺肾阴虚证可以先由肺阴不足,由肺及肾渐至肺肾阴虚者,有肾阴先伤,由肾及肺而致肺肾阴虚者,也有外感内伤同时损伤肺肾之阴者。肺肾阴虚提示肺肾阴液不足,功能紊乱,主要衷现为肺脏清肃失职,肾脏滋养全身无权,而虚热内扰,脏腑功能失调。
肺肾阴液互相滋养,有“金水相生”之称。上由肺阴不足、虚热上扰、清肃失职,故咳嗽痰少,虚热灼伤肺络则痰中带血。下因肾阴亏虚,筋骨失于濡养而腰膝酸软。肺肾阴虚,津不上承故口燥咽干,虚火灼伤声带而音哑,薰蒸咽喉而咽痛。肌肉失于濡养故形体日渐消瘦。阴液亏乏,虚火蒸腾,骨蒸潮’热。阴虚内热、相火上炎则颧红,内逼营阴为盗汗。若火扰精室则遗精,阴血亏虚而经少,虚火损及阴络则崩漏。舌红少苔,脉细数皆为阴虚内热之象。
本病往往呈现乾咳,短气、咽喉乾燥,腰酸腿软,骨蒸潮热,遗精盗汗等症状。症见面色潮红,耳鸣、腰酸、足跟痛、盗汗,五心烦热,月经量多而红、舌红、脉虚数。
西医对功能失调性子宫出血病因,治疗的研究现状功能失调性子宫出血(dysfunctionaluterinebleeding,DUB)简称功血,是指下丘脑一垂体一卵巢轴系神经内分泌调节机制失常导致的子宫异常出血,而全身包括生殖器官无器质性病变″DUB是妇科常见疾病,发病率约为10%,文献报道约有50%的患者发生于绝经前期,于育龄期占30%。青春期占20%。DUB可分为无排卵型功血和排卵型功血两大类,其中无排卵型功血约占85%。多发于青春期和绝经期妇女,但也可发生于生育期妇女。绝经期功血多是无排卵型功血,约占80%-90%,多发生在44-55岁之间,常造成患者不同程度的贫血,是影响围绝经期妇女健康最常见的疾病之一。临床上最常见的症状是子宫不规则出血,特点是月经周期紊乱,经期长短不一,经量不定,甚至大量出血。据WHO统计,功血在育龄妇女的发病率为22%以月经失调和子宫异常出血为主要临床特征,不仅给患者造成了极大的精神负担,严重威胁妇女身心健康,还增加了社会卫生负担。
功能失调性子宫出血疾病分类:妇产科功能失调性子宫出血病是现代医学的病名,是指由于卵巢功能失调而引起的子宫出血,简称“功血”。常表现为月经周期失去正常规律,经量过多,经期延长,甚至不规则阴道流血等。机体内外任何因素影响了丘脑下~垂体~卵巢轴任何部位的调节功能,均可导致月经失调。多数学者认为,“瘀血阻滞胞宫、冲任”是子宫内膜异位症痛经的基本病机,活血化瘀是基本治疗原则。然而,单用活血化瘀药堆砌的方药治疗,效果并不令人满意。结合临床实践,子宫内膜异位症痛经与肝经功能失调关系密切,在治疗时采用从肝论治,以辨证论治为基础,在应用活血化瘀药同时分别加疏肝、温肝、养肝、清肝等调肝药物,临床取得很好的疗效。
无排卵性功血患者可有各种不同的临床表现。临床上最常见的症状是子宫不规则出血,特点是月经周期紊乱。经期长短不一,出血量时多时少,甚至大量出血。有时先有数周或数月停经,然后发生阴道不规则流血,血量往往较多,持续2~4周或更长时间,不易自止;有时则一开始即为阴道不规则流血,也可表现为类似正常月经的周期性出血。出血期无下腹疼痛或其他不适,出血多或时间长者常伴贫血。妇科检查子宫大小在正常范围,出血时子宫较软。
西医药物治疗,一般采用内分泌治疗极有效,但对不同年龄的对象应采取不同方法。使用性激素治疗时应周密计划,制定合理方案,尽可能使用最低有效剂量,并作严密观察,以免性激素应用不当而引起出血。中医认为疾病之所以发生,是由于内因和外因共同造成的。一方面在于人体的正气是否充实,另一方面还在于邪气的强弱。而人体的正气是否充实,受到多种因素的影响,如先天禀赋,气候变化,饮食劳倦,情志变化等等。因此,中医治疗疾病必须因人,因时,因地治疗。这也符合中医的“天人相应”学说。中医本病病因复杂,病机多变,证型错综复杂。目前尚存在着中医辨证分型不规范,诊断及疗效评价标准不统一。
发明内容
本发明所要解决的技术问题在于,目前西医药物治疗,一般采用内分泌治疗极有效,使用性激素治疗时应周密计划,制定合理方案,尽可能使用最低有效剂量,并作严密观察,以免性激素应用不当而引起出血。中医认为疾病之所以发生,是由于内因和外因共同造成的。中医治疗疾病必须因人、因时、因地治疗。中医本病病因复杂,病机多变,证型错综复杂。目前尚存在着中医辨证分型不规范,诊断及疗效评价标准不统一。
为解决这些技术难题,本发明提供了一种治疗肾阴虚型功能失调性子宫出血的中药制剂,其所述中药制剂中包括以下重量份数的原料药:黄精10~20份,白芍10~20份,覆盆子10~20份,黄芪10~20份,山茱萸10~20份,天门冬10~20份,贯众炭10~20份,生地10~20份,麦冬10~20份,延胡索10~20份,益母草10~20份,血余炭10~20份,当归10~20份,五灵脂10~20份,地骨皮10~20份,远志10~20份,香附10~20份,五味子10~20份,茯苓10~20份,仙鹤草10~20份,炒地榆10~20份,旱莲草10~20份,仙茅10~20份。

为解决上述技术问题,本发明还提供一种治疗肾阴虚型功能失调性子宫出血的中药制剂的制备方法,其所述制剂剂型为口服液剂时,制备方法包括:
a、取所有原料放入乙醇浸泡,热提取2次,为组分1;
b、将上述提取的药渣加水提取1次,为组分2;
c、将组分1与组分2合并浓缩;干燥成粉末罐装胶囊。
所述步骤a中,取所有原料药泡入占其质量10倍量乙醇中,浸泡1-2小时,加热提取2次,每次1-2小时,合并提取液,静置后去上清液,然后放入减压浓缩罐内,减压回收乙醇,浓缩至药液浓度为0.6g生药/mL,抽滤至滤液的相对密度为80℃时1.36的浸膏,得到组分1。
所述步骤b中,将上述提取过的原料药的药渣加水,加热提取2次,每次1-2小时,合并提取液,静置后去上清液,然后放入减压浓缩罐内,减压,浓缩至药液浓度为0.6g生药/mL,抽滤至滤液的相对密度为80℃时1.36的浸膏,得到组分2。
所述步骤c中,将上述两种提取液合并,减压浓缩相对密度为80℃时1.35的滤液,回收乙醇,将上述两种提取液置入双效真空浓缩器中,浓缩至90℃时相对密度为1.60的浓缩液,置0~5℃低温冷藏24小时;将冷藏液加0.3%的助滤剂硅藻土,过滤,滤液再置入双效真空浓缩器中,浓缩至每1ml含0.2g生药量,加糊精调和后杀菌装瓶。
为解决上述技术问题,本发明再提供一种治疗肾阴虚型功能失调性子宫出血的中药制剂的制备方法,其所述制剂剂型为胶囊剂的制备方法包括:
a.将所述原料放入粉碎机粉碎后过筛;
b.将过筛后剩下的颗粒物放入乙醇中加热回流提取2次;
c.再取上述乙醇提取后的药渣,加水加热回流提取3次;
d.将上述两种提取液合并,减压回收乙醇并喷雾干燥,得药粉;混合过筛得到的细粉罐装胶囊。
所述步骤a中,将所述原料放入粉碎机粉碎后过120目细筛,使能通过120目筛的细粉量在达到30%,收集通过120目筛的原料细粉备用;所述步骤b中,将过筛后剩下的颗粒物放入乙醇中加热回流提取2次,每次1~2小时,将2次提取液合并静置。
所述步骤c中,将上述乙醇提取过的药渣加10倍量水,然后放入提取罐加热煮沸1小时,过滤,滤液备用;滤渣加水,第二次加热,煮沸45分钟,过滤,滤液备用;滤渣再加水,第三次加热煮沸半小时,过滤;将三次滤液合在一起,用纱布过滤,残渣中加入50-60%乙醇,60℃-70℃继续浸提2h,每10min搅拌一次,纱布过滤,合并浸提液,浓缩成糊状;所述步骤d中,将上述提取液合并,减压浓缩相对密度为80℃时1.08的滤液,回收乙醇,将所得滤液调整比重到1.06,通入喷雾干燥机进行喷雾干燥,过喷雾干燥机喷头的压缩空气压力为4kg/m2,干燥室热气流温度是80℃,得原料药粉;将过筛后得到的原料细粉与乙醇提炼药粉混合,罐装胶囊。
本发明秉承中医标本兼治之原则,深入病灶治疗,本发明采取益气养阴,补肺滋肾之法,以治其因;疏肝健脾,养血和营,以治其本;坚固冲任,固经止崩而治其标。本发明一方面大量补充雌激素,止血敛血,使血停或血量减少,治其标;另一方面,着手温补肾阳、益气生血,滋补肝肾,补肾调和冲任,使经血有固,周期有序而崩漏痊愈,治其本;标本兼治,从此告别功能失调性子宫出血。
具体实施方式
与西医不同,中医所说的“肾”,不仅指解剖学上的脏器,而是指以解剖学上的肾为代表的多个系统功能的集合,涵盖了西医的神经系统、内分泌系统、生殖系统、泌尿系统、免疫系统、以及呼吸系统等多个系统的部分功能,包括了人体的生长、发育、生殖、水液代谢以及精神情志等多方面的功能。
中医强调补肾应该辨证,首先要确定是肾阳虚还是肾阴虚。阳虚补阳,阴虚滋阴,调反了非但达不到补肾的效果,还会适得其反。
肾阴虚主要表现为:腰膝酸软、两腿无力,眩晕耳鸣,脱发齿松,盗汗,失眠,梦呓磨牙,口干,尿黄,大便干燥,男子阳强易举或阳痿、早泄、遗精,妇女经少经闭,或见崩漏,形体消瘦,潮热盗汗,五心烦热,咽干颧红,溲黄便干,舌红少津,脉细数。
肾阴虚,是肾脏阴气、阴液不足表现的证候。多由久病伤肾,或禀赋不足房事过度,或过服温燥劫阴之品所致。肾阴虚的治疗包括药物治疗及辅助治疗。不管是阴虚还是阳虚也好,匮乏到一定程度,就是阴损及阳,或阳损及阴,比如说肾阳虚时间长了会有肾阴虚的症状,就是两种症状都有很多。所以补肾阳的时候,一定要补肾阴,而且要阴中求阳,而且在补肾阴的时候也不是一味的只用补肾阴的药物。
在于,肾阴是指肾本脏的阴气阴液,是一身之阴的根本,对全身起着凉润、濡养、制约过度阳热的作用,肾阳是指肾脏中的阳气,对全身起着温煦、推动、气化的作用。无论肾阴、肾阳都是肾脏精气功能活动对立统一的两个方面,肾阴虚、肾阳虚的本质都是肾脏精气功能活动的减退,其本质都是肾精气虚了,因此肾阴虚到一定程度会阴损及阳使肾阳亦虚,肾阳虚到一定程度会阳损及阴,使肾阴亦虚。肾阴、肾阳都是物质与功能的结合,不存在一种非物质的纯功能之气。不能说肾阴虚就是物质的减少,肾阳虚是功能的减少,不论肾阴虚、肾阳虚实质都是相关物质和功能的减少。阳气是一种活力很强的精微物质。比如肾上腺皮质激素、甲状腺激素、性激素分泌不足中医辨证多属肾阳虚,要通过补肾阳治疗。
从物质与功能来分,物质属阴,功能属阳,人的一切功能活动都属于阳气,人的一切构成物质都属于阴精,但人体内不存在一种非物质的纯功能之气,在中医学里气是一种活力很强的精微物质。因此气是物质与功能的辩证统一。肾气由肾精化生,广义肾气指的是肾脏的功能活动,包括了肾阴、肾阳。狭义肾气指的是肾脏中起固摄、封藏作用的部分,一般讲的肾气不固,指的是狭义肾气。肾阴是肾脏功能活动的一部分,起着濡润、收敛等作用。平常所说的肾阴的含义其实还包含了肾精在内,有时用肾阴专指肾精。因此阴阳的含义在不同的语境是不同的。有时用阳气表示人体的所有功能活动,有时单指的是人体功能活动中起温煦、推动作用的部分,因此要注意分辨。阴阳可以指脏腑之气中发挥相反作用的气,也可以指正气和邪气,也可以指清气和浊气。总之阴气并不总是代表坏的、不利的,阳气也并不总是好的。提到阴气不要总是认为就是指的人体的病气、邪气,它还可以是指人体生理功能活动中必不可少的一部分。年四十,阴气自半也,指的是人的五脏之气衰少,仅是原来的一半。
功能失调性子宫出血(dysfunctional uterine bleeding)是患者由于卵巢功能失调而导致一些列机体功能发生的障碍,从而导致患者出现的子宫出血,临床上简称“功血”。功能失调性子宫出血的临床症状主要有患者的月经周期不规律,患者在月经期间的经血量出现过多现象,以及患者经期不同程度发生延长现象,甚至有患者出现不规则的阴道流血现象等。患者一旦发病,将会对其生活质量和身心健康产生一定的影响,因此对于功能失调性子宫出血的研究具有重要的理论意义和临床实践意义。
女子以肝为先天,女子一生都与肝存在着密切的联系。故妇科疾病几乎都与肝有关。“女子以血为主,以肝为养”。肝体阴而用阳,肝为女子的先天。妇女以血为本,临经之期阴血下注,肝血偏虚,厥阴督脉失去濡养,并令全身阴阳、气血升降调节失常所致。治疗用养血柔肝之法。

功血属于中医崩漏、经量过多、月经失调、月经先期、经期延长、等范畴。病机为冲任损伤,不能制约经血,多责之于气虚、气滞、血虚、血癖。失血较多则多气血两虚之证。祖国医学治疗崩漏是以无周期的阴道出血为辩证要点,辩证时结合出血的量、色、质的变化和全身证候辨明,寒、热、虚、实,依据病情的缓急,出血的久暂采用急则治其标,缓则治其本的原则,灵活运用止血的塞流,求因治本的澄源和调理以后的复旧之法辩证论治。
中医本病病因复杂,病机多变,证型错综复杂。目前尚存在着中医辨证分型不规范,诊断及疗效评价标准不统一。然而报道的病案居多,前瞻性随机对照试验少;以临床研究居多,实验研究较少。主观性很强,缺乏科学性;多侧重于临床疗效的观察,基础研究和实验研究者少等不足。
通过对功血现代文献的研究,拟找出其中医证候及规律,分析功血中医证候研究现状与存在的问题,为进一步研究功血中医证候规范化研究提供思路,使中医辨证论治具有可操作性,灵活性,准确性及先进性的特点,从而对临床诊治具指导意义。
方中以远志清热除烦,旱莲草清热凉血祛瘀;仙鹤草清泄三焦之热,清热止血;生地散热疏肝解郁;五灵脂养血和血;白芍敛阴和营;白术健脾统血;益母草行血祛瘀,地榆清热止血,二者相互调制,行血不溢血,止血不留瘀;五味子兼调冲任,益肾固经止崩,以治血乱。全方共收清肝降火、疏肝泄热、健脾统血、养血止血、固经止崩之效。
益母草味辛苦、凉。活血、祛淤、调经、消水。治疗妇女月经不调,胎漏难产,胞衣不下,产后血晕,瘀血腹痛,崩中漏下,尿血、泻血,痈肿疮疡。益母草含益母草碱、延胡索酸等物质,有活血调经、降血压、利尿消肿等作用。
覆盆子性温;甘、酸;归肾、膀胱经。主治:益肾,固精,缩尿。用于肾虚遗尿,小便频数,阳痿早泄,遗精滑精。《本草经疏》:″覆盆子,其主益气者,言益精气也。肾藏精、肾纳气,精气充足,则身自轻,发不白也。苏恭主补虚续绝,强阴建阳,悦泽肌肤,安和脏腑。甄权主男子肾精虚竭,阴痿,女子食之有子。大明主安五脏,益颜色,养精气,长发,强志。皆取其益肾添精,甘酸收敛之义耳。″《药性论》载其:″主男子肾精虚竭,女子食之有子。主阴痿。″
白芍入肝、脾经,养血柔肝,缓中止痛,敛阴收汗。治胸腹胁肋疼痛,泻痢腹痛,自汗盗汗,阴虚发热,月经不调,崩漏,带下,妇人血闭不通,消瘀血,能蚀脓,益女子血。《日华子本草》:治风、补劳,主女人一切病,并产前后诸疾,通月水,退热,除烦,益气,天行热疾,瘟瘴,惊狂,妇人血运,及肠风,泻血,痔瘘。发背,疮疥,头痛,明目,目赤努肉。赤色者多补气,白者治血。
当归,其味甘而重,故专能补血,其气轻而辛,故又能行血,补中有动,行中有补,诚血中之气药,亦血中之圣药。大约佐之以补则补,故能养营养血,补气生精,安五脏,强形体,益神志,凡有形虚损之病,无所不宜。佐之以攻则通,故能祛痛通便,利筋骨,治拘挛、瘫痪、燥、涩等证。营虚而表不解者,佐以柴、葛、麻、桂等剂,大能散表卫热,而表不敛者,佐以大黄之类,又能固表。惟其气辛而动,故欲其静者当避之,性滑善行,大便不固者当避之。凡阴中火盛者,当归能动血,亦非所宜,阴中阳虚者,当归能养血,乃不可少。
五灵脂为复齿鼯鼠的干燥粪便,可用于瘀血内阻、血不归经之出血,如妇女崩漏经多,色紫多块,少腹刺痛。既可单味炒研末,温酒送服,又可与其他药物配合使用。活血散瘀,炒炭止血。用于心腹淤血作痛,痛经,血瘀经闭,产后淤血腹痛;炒炭治崩漏下血;外用治跌打损伤,蛇、虫咬伤。五灵脂苦泄温通,“通利气脉”,“通则不痛。”故《本草经疏》谓之“血滞经脉,气不得行,攻刺疼痛等证,在所必用。”
黄芪具有补气固表、利水退肿、托毒排脓、生肌等功效。黄芪的药用迄今已有2000多年的历史,现代研究,黄芪含皂甙、蔗糖、多糖、多种氨基酸、叶酸及硒、锌、铜等多种微量元素。有增强机体免疫功能、保肝、利尿、抗衰老、抗应激、降压和较广泛的抗菌作用。但表实邪盛,气滞湿阻,食积停滞,痈疽初起或溃后热毒尚盛等实证。黄芪具有补气固表,利尿托毒,排脓,敛疮生肌的功效。用于气虚乏力,食少便溏,中气下陷,久泻脱肛,便血崩漏,表虚自汗,痈疽难溃,久溃不敛,血虚萎黄,内热消渴。《本经》记载:“主痈疽,久败疮,排脓止痛。补虚,小儿百病。”《日华子本草》记载:“助气壮筋骨,长肉补血。”
延胡索(Corydalis yanhusuo W.T.Wang ex Z.Y.Su et C.Y.Wu),罂粟科、紫堇属多年生草本,别名:延胡、玄胡索、元胡索、元胡。中国传统药材。主要产于浙江和江苏。其块茎(夏天无)含紫堇碱、原阿片碱等多种生物碱,经制成粉剂或醋制流浸膏等,主治心腹腰膝诸痛;月经不调;症瘕;崩中;产后血晕;恶露不尽;跌打损伤。有镇痛、镇静、催眠作用。
血余炭为中药材,原名乱发,是止血药中的收敛止血药,始载于《神农本草经》。血余炭是人发煅烧之后的炭化物。本品性苦、平,入肝、胃经。具有收敛止血、化瘀利尿的功效。血余炭有一定的止血作用.血余炭水煎液或醇提取液对大鼠或小鼠腹腔给药,能明显缩短出血时间。血余炭粗晶液对大鼠腹腔给药,可加速大鼠的血凝作用;并可诱发大鼠血小板聚集;缩短狗和兔的凝血时间。有报告指出,血余炭粗晶对ADP诱导的大鼠血小板聚集有较显著增强作用,尚能明显地降低大鼠血浆cAMP的含量。.提示血余炭粗晶具有促内源性系统凝血功能,其止血原理与血浆中cAMP含量有关.亦有报告指出,血余炭粗晶液对ADP诱导的大鼠血小板聚集有增强作用,对血小板黏附率有增加趋势,能显著缩短白陶土部分凝血活酶时间,能明显降低血小板内环核苷酸含量,并有一定的抗炎作用。尚有报告指出,由于炮制条件不同,血余之药理活性和作用性质亦有变化。血余炭(350℃炮制品)口服止血作用最强,而煎剂(300℃以下炮制品)注射则表现为中枢兴奋作用。
仙鹤草:西洋龙芽草又名仙鹤草,高约五尺,每年六到九月间开黄色的花,生长于美国、加拿大及欧洲的树林和草原中;含有丰富的单宁酸,具有良好的收敛效果。《履巉岩本草》:“味辛涩,温,无毒。”《滇南本草》:“性微温,味苦涩。”《生草药性备要》:“味甜,性平。”功用主治:收敛止血,止痢,杀虫。广泛用于各种出血之证,例如:吐血、尿血、便血、崩漏、咯血、衄血,赤白痢疾,劳伤脱力,痈肿,跌打,创伤出血等证。《滇南本草》:“治妇人月经或前或后,赤白带下,面寒腹痛,日久赤白血痢。“《生草药性备要》:”理跌打伤,止血,散疮毒。“《百草镜》:”下气活血,理百病,散痞满;跌打吐血,血崩,痢,肠风下血。“
炒地榆:地榆苦酸;寒;无毒,归经肝;肺;肾;大肠经。功效:凉血止血,清热解毒,消肿敛疮。止血药;凉血止血药。主治:吐血,咯血,衄血,尿血,便血,痔血,血痢,崩漏,赤白带下,疮痈肿痛,湿疹,阴痒,水火烫伤,蛇虫咬伤。《纲目》:地榆,除下焦热,治大小便血证。止血,取上截切片炒用,其梢则能行血,不可不知。杨士瀛云:诸疮痛者加地榆,痒者加黄芩。《本草求真》:地榆,诸书皆言因其苦寒,则能入于下焦血分除热,俾热悉从下解。又言性沉而涩,凡人症患吐衄崩中肠风血痢等症,得此则能涩血不解。按此不无两歧,讵知其热不除,则血不止,其热既清,则血自安,且其性主收敛,既能清降,又能收涩,则清不虑其过泄,涩亦不虑其或滞,实力解热止血药也。
淫羊藿辛、甘、温,归肝、肾经。能补肾助阳,强筋健骨,祛风除湿,善治肾阳不足,阳痿遗精,遗尿尿频;风湿痹痛,骨痿瘫疾。《本草述》云“淫羊火,《本经》药主阴痿绝伤,《日华子》亦首言其疗男子绝阳,女子绝阴,则谓入命门、补真阳者是也。盖命门为肾中之真阳,即人身之元气也,其所谓绝阳绝阴,不本之元气,何以嘘之于既槁。所谓益气力,强志,并治冷气劳气,筋骨挛急等证,皆其助元气之故。老人昏耄,中年健忘,皆元阳衰败而不能上升者也。须知此味以降为升,其升。
贯众炭:贯众炭性味和功用苦、涩,微寒。归肝、胃经。清热解毒,止血,杀虫。用于风热感冒,乙型脑炎,痄腮,血痢,肠风便血,血崩,带下,产后血气胀痛,驱蛔虫和蛲虫,贯众炭制备方法:取净贯众片炒至焦黑色为度,喷洒清水,放凉。性苦味凉。《纲目》:″治下血崩中,带下,产后血气胀痛,斑疹毒,漆毒,骨哽。″《会约医镜》:″治邪热腹痛,解时行疫气。″
中药何首乌有生首乌与制首乌之分,直接切片入药为生首乌,用黑豆煮汁拌蒸后晒干入药为制首乌。二者的功用有所不同:生首乌功能解毒、消痈、润肠通便,常用于治疗瘰疬疮痈、风疹瘙痒、肠燥便秘;制首乌功能补肝肾、益精血、乌须发、强筋骨,用于血虚萎黄、眩晕耳鸣、须发早白、腰膝酸软、肢体麻木、崩漏带下、久疟体虚等。本发明所采用的制首乌是补肾益肾的佳品。
黄精:黄精以根茎入药。具有补气养阴,健脾,润肺,益肾功能。用于治疗脾胃虚弱,体倦乏力,口干食少,肺虚燥咳,精血不足,内热消渴等症。对于糖尿病很有疗效。黄精具有降血压,降血糖,降血脂,防止动脉粥样硬化,延缓衰老和抗菌等作用,黄精多糖具有免疫激活作用。用于阴虚肺燥,干咳少痰,及肺肾阴虚的劳嗽久咳等。用于脾胃虚弱。既补脾阴,又益脾气,用于肾虚精亏得头晕,腰膝酸软,须发早白及消渴等。黄精壮筋骨,益精髓,变白发;适宜用于肺阴不足所致的咳嗽痰少,干咳无痰,咳血等症。
麦冬性味功能:甘、微苦、凉、滋阴生津、润肺止咳、清心除烦。主治热病伤津、心烦、口渴、咽干肺热、咳嗽、肺结核。配沙参,肺胃同治,具有清肺凉胃,养阴生津之良好效用,用于阴虚肺燥或热伤肺阴所致的干咳少痰,咽喉干燥等症。配五味子,酸甘化阴,守阴所以留阳,阳留汗自止。功能养阴敛汗,用治阴虚汗多,心悸,肺虚久咳,少痰或痰粘不爽等。
山茱萸味酸、涩性,微温。归肝、肾经。补益肝肾,涩精固脱。用于眩晕耳鸣,腰膝酸痛,阳痿遗精,遗尿尿频,崩漏带下,大汗虚脱,内热消渴,为固涩药,它含有山茱萸甙、莫罗甙、7-O-甲基莫罗甙、山茱萸新甙、当药甙、番木鳖甙等环烯醚萜甙类及皂甙、熊果酸、酒石酸、没食子酸、苹果酸、山茱萸鞣质1、2、3等多种有效成份。有明显的利尿、降脂、抗菌与升高血液白细胞的作用,对治疗肝肾亏损所致的眩晕耳鸣、腰酸等症及由于肾阴不足所致的遗精、尿频等症有明显疗效。又因其具有收敛止血功效被常用于妇女体虚所致的月经过多,崩漏带下等症,对男科、妇科的常见病均有很好的治疗作用,大量用于补益药物的生产,故被列为国药四十种大宗品种之一,在中医药界有着广泛的市场。
天门冬,学名:Asparagus cochinchinensis(Lour.)Merr,为百合科天门冬属植物,别名天冬、丝东。甘、苦,寒。养阴生津,润肺清心。用于肺燥干咳,虚劳咳嗽,津伤口渴,心烦失眠,内热消渴,肠燥便秘,白喉。阴虚发热,咳嗽吐血,肺痿,肺痈,咽喉肿痛,消渴,便秘,小便不利。内服:煎汤,2-4钱;熬膏或入丸、散。虚寒泄泻及外感风寒致嗽者忌。
茯苓味甘、淡、性平,入药具有利水渗湿、益脾和胃、宁心安神之功用。现代医学研究:茯苓能增强机体免疫功能,茯苓多糖有明显的抗肿瘤及保肝脏作用。茯苓性味甘淡平,入心、肺、脾经。具有渗湿利水,健脾和胃,宁心安神的功效。可治小便不利,水肿胀满,痰饮咳逆,呕逆,恶阻,泄泻,遗精,淋浊,惊悸,健忘等症。茯苓之利水,是通过健运脾肺功能而达到的,与其它直接利水的中药不同。用于脾虚泄泻,带下茯苓既能健脾,又能渗湿,对于脾虚运化失常所致泄泻、带下,应用茯苓有标本兼顾之效,常与党参、白术、山药等配伍。有可用为补肺脾,治气虚之辅佐药。
旱莲草为菊科植物鳢肠的全草,因为搓揉它的茎叶时,可见黑色的汁液流出,所以又叫“墨旱莲”。中医认为旱莲草是补肝肾药.性寒.有养阴功效。旱莲草和女贞子都有保肝、解毒、降低转氨酶的作用,能用来治疗慢性肝病。凉血,止血,补肾,益阴。治吐血,咳血,衄血,尿血,便血,血痢,刀伤出血,须发早白,白喉,淋浊,带下,阴部湿痒。《唐本草》:主血痢。针灸疮发,洪血不可止者敷之;汁涂发眉,生速而繁。《日华子本草》:排脓,止血,通小肠,敷一切疮并蚕瘑。《滇南本草》:固齿,乌须,洗九种痔疮
仙茅具有补肾助阳、益精血、强筋骨和行血消肿的作用,主要用于肾阳不足、阳痿遗精、虚痨内伤和筋骨疼痛等病症。补肾阳,强筋骨,散寒湿。治阳萎精冷,小便失禁,崩漏,心腹冷痛,腰脚冷痹,痈疽,瘰疬,阳虚冷泻。阳痿精寒,腰膝风冷,筋骨痿痹等症。古方中有仙茅丸,能壮筋骨、益精神、明目、黑须发。《海药本草》:主风,补暖腰脚,清安五脏,强筋骨,消食。宣而复补,主丈夫七伤,明耳目,益筋力,填骨髓,益阳。《日华子本草》:治一切风气,补五劳七伤,开胃下气。
地骨皮性寒,味甘。功能主治:凉血除蒸,清肺降火。用于阴虚潮热、骨蒸盗汗、肺热咳嗽、咯血、衄血。清热,凉血。治虚劳潮热盗汗,肺热咳喘,吐血,衄血,血淋,消渴,高血压,痈肿,恶疮。《本经》:″主五内邪气,热中消渴,周痹。″《别录》:″主风湿,下胸胁气,客热头痛,补内伤大劳嘘吸,坚筋,强阴,利大小肠,耐寒暑。″《药性论》:″细锉,面拌熟煮吞之,主治肾家风。″《食疗本草》:″去骨热消渴。″
远志,学名:Polygala tenuifolia Willd.别名:葽绕、蕀蒬、棘菀、小草、细草、线儿茶,小草根、神砂草。为远志科、远志属。产东北、华北、西北和华中以及四川。性温,味苦、辛,具有安神益智、祛痰、消肿的功能,用于心肾不交引起的失眠多梦、健忘惊悸,神志恍惚,咳痰不爽,疮疡肿毒,乳房肿痛。
香附辛微苦甘,平功能主治:理气解郁,调经止痛。用于肝郁气滞,胸、胁、脘腹胀痛,消化不良,月经不调,经闭痛经,寒疝腹痛,乳房胀痛。《纲目》载其:散时气寒疫,利三焦,解六郁,消饮食积聚,痰饮痞满,跗肿,腹胀,脚气,止心腹、肢体、头、目、齿、耳诸痛,痈疽疮疡,吐血,下血,尿血,妇人崩漏带下,月候不调,胎前产后百病。
生地:生地也叫生地黄,玄参科多年生草本植物地黄Rehmannia glutinosaLibosch.的新鲜或干燥的块根。清热、生津、润燥、滑肠、破瘀、生新、止痛、调经、金疮瘀、凉血、止血;鲜地黄和生地黄的作用是有所不同的。鲜地黄清热生津,可以止血和凉血,经常是用于热病伤阴,吐血、咽喉肿痛、舌绛烦渴、发斑发疹以及衄热等病症。相对于鲜地黄,生地黄是凉血清热,也可以生津,还可以养阴,多用于治疗阴虚内热、骨蒸劳热、舌绛烦渴、发斑发疹等问题。生地性寒,功能为滋阴补肾。不只生津止渴和凉血清热,还对血崩、月经不调、胎动不安、便秘等有作用,凡是血分有热伤阴的人,都可以经常服用。如果想要治疗温热病,可以用生地做清营汤;如果是治疗温病的后期,由于余热还没消尽,应该把生地用来煮青蒿鳖甲汤;另外,针对血热毒盛,斑疹紫黑,温热病严重热入营血的症状,可以服用生地制成的四生丸;一般要养阴生津来治疗内热消渴和便秘的,应该食用生地煮的曾液汤。
本发明蜜炼丸剂的制作过程为:取黄精1500g,白芍1500g,覆盆子1500g,黄芪1500g,山茱萸1500g,天门冬1500g,贯众炭1500g,生地1500g,麦冬1500g,延胡索1500g,益母草1500g,血余炭1500g,当归1500g,五灵脂1500g,地骨皮1500g,远志1500g,香附1500g,五味子1500g,茯苓1500g,仙鹤草1500g,炒地榆1500g,旱莲草1500g,仙茅1500g,取所有原料药泡入水中,浸泡2小时以上,分别加原料药总重量的8倍量、6倍量的水提取2次,提取时间为2小时、1小时;合并2次提取液,提取液通过管式离心,得离心液,离心液过D 101大孔树脂进行吸附,先用水洗除杂,然后用70%的乙醇进行洗脱,收集洗脱液,减压浓缩,成浸膏,为组分成1;将上述提取过的原料药的药渣加乙醇,分别加药渣总重量的10倍量、8倍量的乙醇温浸2次,温浸时间为2小时、1.5小时,温度为85℃,合并2次温浸液;温浸液减压浓缩至密度为60℃时1.25浸膏,将浸膏加入其4倍量的乙酸乙酯萃取二次,萃取液回收完乙酸乙酯,成浸膏,得到组分2;将上述两种提取液合并,减压浓缩相对密度为80℃时1.35的滤液,回收乙醇,将上述两种提取液置入双效真空浓缩器中,浓缩至90℃时相对密度为1.60的浓缩液,置0~5℃低温冷藏24小时;将冷藏液加0.3%的助滤剂硅藻土,过滤,滤液再置入双效真空浓缩器中,浓缩成浸膏,然后干燥成粉末,加蜂蜜搓成丸剂。
本发明片剂的制作过程为:取黄精1200g,白芍1200g,覆盆子1100g,黄芪1100g,山茱萸1200g,天门冬1100g,贯众炭1200g,生地1200g,麦冬1200g,延胡索1200g,益母草1200g,血余炭1100g,当归1100g,五灵脂1200g,地骨皮1100g,远志1200g,香附1200g,五味子1200g,茯苓1100g,仙鹤草1200g,炒地榆1100g,旱莲草1200g,仙茅1200g,取所述原料药浸泡于70%-80%乙醇中,加热回流提取三次,时间1.5小时、1小时、1小时,合并提取液,滤过,滤液回收乙醇,浓缩至干,于60℃下真空干燥,取出,粉碎,过100目筛,细粉备用;加淀粉压片。
本发明胶囊剂的制作过程为:取黄精1200g,白芍1300g,覆盆子1100g,黄芪1100g,山茱萸1200g,天门冬1100g,贯众炭1200g,生地1200g,麦冬1200g,延胡索1200g,益母草1200g,血余炭1100g,当归1300g,五灵脂1200g,地骨皮1200g,远志1200g,香附1300g,五味子1200g,茯苓1300g,仙鹤草1200g,炒地榆1300g,旱莲草1200g,仙茅1300g,加入10倍量乙醇中加热提取2次,每次1~2小时,将2次提取液合并静置;将上述乙醇提取过的组份原料药材的药渣加10倍量水加热回流提取2次,每次1~2小时,将2次提取液合并静置。将上述两种提取液合并,减压回收乙醇并浓缩至药液浓度为0.6g生药/mL,抽滤后,滤液的相对密度约为20℃时1.08;减压至0.03-0.08MPa,温度保持在60-80℃,浓缩至相对密度为1.20,温度至60℃-70℃的浸膏,放入烘箱干燥得到组份药粉,装入胶囊制为胶囊剂。每粒胶囊装0.3g。可以每12粒为一版。

本发明粉剂制备步骤为:取黄精1200g,白芍1300g,覆盆子1300g,黄芪1100g,山茱萸1200g,天门冬1200g,贯众炭1200g,生地1200g,麦冬1200g,延胡索1200g,益母草1200g,血余炭1200g,当归1300g,五灵脂1200g,地骨皮1200g,远志1200g,香附1100g,五味子1200g,茯苓1100g,仙鹤草1200g,炒地榆1300g,旱莲草1200g,仙茅1300g,药材加水浸泡30分钟至1小时,加热煮沸30~40分钟,过滤,滤液备用;滤渣加水第二次加热,煮沸30~40分钟,过滤,滤液备用;滤渣再加水,第三次加热煮沸30~40分钟,过滤;将三次滤液合在一起,加热浓缩至糊状药液,放入烘箱80度烘干至粉末状。
毒理学试验:
试验例1:本发明急性毒性试验
一、试验材料:
动物:昆明种小鼠,体重21-24g,雌雄各半,山东大学生物试验室育种。药物:本发明内服胶囊剂。
二、方法:
1、LD50计算:采用改良寇氏法,将小鼠随机分成5组,每组10只,雌雄各半,将栓剂溶解,配成最大浓度,按小鼠最大允许容量给药,所给剂量按生药量依次为18,14.4,11.5,9.2,7.4(g.kg-1),在动物禁食(不禁水)18小时后,一日内分两次给药(间隔半小时),每次0.5ml,治疗动物死亡情况。
2、最大耐受剂量测定(MTD值):取小鼠20只,雌雄各10只。将栓剂加蒸馏水溶解,配成最高浓度,按动物的最大耐受量,以注射灌喂器能抽动为准。在动物禁食(不禁水)18小时后,一日内分两次给药(间隔半小时),每次0.5ml(每ml含生药0.36g),总药量为18g生药/kg.d,相当临床成人50Kg体重用量的300倍。给药后连续治疗7天。
三、试验结果:
在LD50计算中当用最大允许浓度和最大允许容量给予小鼠时(18g/Kg.d),未见小鼠死亡,即未测出LD50,只可求最大耐受剂量,在7天治疗期中,动物其食欲、活动、毛色、精神状态等皆正常,发育正常,未见有死亡。即选用相当于临床剂量的300倍药量,并无不良反应发生,表明急性毒性极小,MTD>18g/Kg.d。
试验例2:本发明急性皮肤刺激试验报告
1、实验目的:检测本发明栓剂对实验动物皮肤的刺激/腐蚀作用和强度
2、材料和方法:
2.1.受试物:名称:本发明丸剂(实施例3方法制得)
2.2.实验动物:壹级大耳白种家兔,10只,雌性,体重2.1-2.3kg;由青岛大学医学院生物系实验动物中心提供;
2.3.试验方法:
选用皮肤完好的健康家兔10只,于试验前24小时将其背部脊柱两侧被毛剪掉,去毛面积左、右各约4cm×4cm。试验时取受试物0.2ml涂于2.5cm×2.5cm的二层纱布上,敷贴于一侧去毛皮肤表面,油纸覆盖,胶布固定。另一侧皮肤作为空白对照。封闭4小时后,去除覆盖物,并用温水清洗去残留受试物,于去除受试物后的1、24和48小时治疗涂抹部位皮肤反应,并评分。
3、实验结果:实验动物皮肤未出现任何红斑,红肿,发炎现象。
4、小结:根据《消毒技术规范》(1999)中皮肤刺激反应评分标准判定,本发明软膏剂对动物皮肤刺激强度属无刺激性。
实施例3:阴道粘膜刺激试验结果
1.试验方法:
实验动物检疫3天后,选用18只上述健康家兔,随机分为三组,每组各6只,分别为:空白对照、赋形剂对照、本发明丸剂组(按应用液5倍浓度)。分别用适当大小的灭菌棉条浸以灭菌生理盐水和受试物置于赋形剂对照组和受试物组动物阴道内,与阴道粘膜接触6小时,24小时后处死动物。取出局部阴道组织,治疗病变情况。
2、试验结果:阴道粘膜刺激试验结果
实施例4:皮肤过敏试验结果
用本发明片剂,进行诱导和激发接触试验,在24小时和48小时后治疗试验小鼠,均不见皮肤有红斑及水肿等现象,与阴性对照组无差异。结果表明,该片剂对皮肤反应强度近于零,即无皮肤变态反应。
实施例5:长期毒性实验资料
1、实验方法
将豚鼠随机份成4组,每组15只。在豚鼠背部脊柱两侧将脱毛剂均匀涂上,使其脱毛范围约40平方厘米。洗净脱毛剂,治疗24小时后每组豚鼠分别涂本发明栓剂溶液0.2、0.4和0.8ml,分别含生药92、184和368mg,另一组涂溶媒0.8ml每日二次,连续30天,治疗豚鼠的一般情况,实验结束后处死动物进行血液学、血液生化及病理学检查。
2、结果
上述三组用药豚鼠躯干去毛区,未见局部皮肤有水肿、充血、红斑、出血点及溃疡。用药组与对照组动物毛发色泽、摄食、四肢活动等对照组无明显差异,血液生化检查,用药组与对照组均在正常范围。病理组织学检查,实验各组心、肝、肾及局部皮肤均未见明显病变。提示,本发明中药片剂长期用药对局部皮肤及全身重要脏器均未见明显的毒性作用。
试验结果表明:本发明中药片剂实验安全。该药对乙型链球菌、金黄色葡萄球菌、大肠杆菌的最小抑制浓度为1∶150,淋球菌为1∶150,白色念珠菌为1∶300,对1∶20稀释度时的杀灭时间为5分钟,1∶50时为10分钟,1∶100时为20分钟,1∶200时为40分钟,1∶400时为8小时。
药理学实验说明:
1资料与方法
本病往往呈现乾咳,短气、咽喉乾燥,腰酸腿软,骨蒸潮热,遗精盗汗等症状。症见面色潮红,耳鸣、腰酸、足跟痛、盗汗,五心烦热,月经量多而红、舌红、脉虚数。本文选取了我院2009年12月至2012年12月间入院治疗的70例肾阴虚型功能失调性子宫出血患者为研究对象。症见:出血量多,气短无力、面色苍白、腹胀便溏、纳食减少,舌淡,面色萎黄、心悸失眠、头晕目眩、出血量多而色淡,舌淡苔白,脉细。一般治疗中患者体质往往较差,呈贫血貌。患者中年龄最大的为54岁,年龄最小的为42岁,平均年龄为(44.13±6.11)岁;病程最长的为9个月,最短的为6个月,平均病程为(6.43±2.89)月。根据实施治疗方法的不同,将患者随机分为本发明治疗组和西医治疗组,每组32例患者,两组患者在年龄、病程等一般资料比较差异无统计学意义,具有可比性。
1.2治疗方法
所有患者于月经期服用安宫黄体酮片8mg/d,连用10d以调节周期,共3个周期为1个疗程,如出血多时可加用甲基辜丸素10mg/d,连用1周。
本发明治疗组在上述治疗基础上,方为:黄精12g,白芍13g,覆盆子12g,黄芪11g,山茱萸12g,天门冬12g,贯众炭13g,生地11g,麦冬12g,延胡索13g,益母草12g,血余炭13g,当归12g,五灵脂13g,地骨皮13g,远志13g,香附14g,五味子12g,茯苓13g,仙鹤草12g,炒地榆11g,旱莲草12g,仙茅13g。水煎服,1次/d。
1.3统计学方法
采用SPSS 17.0统计学软件,计量资料采用t检验,以“x±s”表示;计数资料用χ2检验,以P<0.05为差异具有统计学意义。
2结果
2.1两组患者实施不同治疗后总体疗效比较
两组患者实施不同治疗后总体疗效组间比较,本发明治疗组优于西医治疗组,且差异具有统计学意义(P<0.05)。见表1。

讨论:中医强调补肾应该辨证,首先要确定是肾阳虚还是肾阴虚。阳虚补阳,阴虚滋阴,调反了非但达不到补肾的效果,还会适得其反。
肾阴虚主要表现为:腰膝酸软、两腿无力,眩晕耳鸣,脱发齿松,盗汗,失眠,梦呓磨牙,口干,尿黄,大便干燥,男子阳强易举或阳痿、早泄、遗精,妇女经少经闭,或见崩漏,形体消瘦,潮热盗汗,五心烦热,咽干颧红,溲黄便干,舌红少津,脉细数。
肾阴虚,是肾脏阴气、阴液不足表现的证候。多由久病伤肾,或禀赋不足房事过度,或过服温燥劫阴之品所致。肾阴虚的治疗包括药物治疗及辅助治疗。不管是阴虚还是阳虚也好,匮乏到一定程度,就是阴损及阳,或阳损及阴,比如说肾阳虚时间长了会有肾阴虚的症状,就是两种症状都有很多。所以补肾阳的时候,一定要补肾阴,而且要阴中求阳,而且在补肾阴的时候也不是一味的只用补肾阴的药物。
传统中医理论认为补肾调经可以有效的改善功能失调性子宫出血患者的相关临床症状,所谓补肾调经是指用具有补益肾之精气,或滋肾阴、温肾阳等作用的方药,治疗肾虚所致女性月经不调病证的治法。本文所用的中医治疗方法符合传统中医理论。方中以远志清热除烦,旱莲草清热凉血祛瘀;仙鹤草清泄三焦之热,清热止血;柴胡散热疏肝解郁;五灵脂养血和血;白芍敛阴和营;白术健脾统血;益母草行血祛瘀,地榆清热止血,二者相互调制,行血不溢血,止血不留瘀;五味子兼调冲任,益肾固经止崩,以治血乱。全方共收清肝降火、疏肝泄热、健脾统血、养血止血、固经止崩之效。
已有研究报道临床上治疗功能失调性子宫出血的过程中,采用中西医结合的治疗方法效果显著。本文的研究结果也进一步证实了以上观点:两组患者实施治疗后总体疗效组间比较,本发明治疗组优于西医治疗组,同时本发明治疗组FSH、LH、E2指标改善情况均优于西医治疗组。综上所述,本发明治疗功能失调性子宫出血的临床疗效显著,与传统西医疗效相比优势明显,值得推广应用。
实施例:顾女,32岁,已婚。行经七八天后止,但三两天又复潮,持续七天以上,甚则半月不停。血量多,色鲜红。今又发生一次大出血,请当地医生与药打针治疗不效。刻症:主诉月经不调6月余,本次以大出血七天不止,伴头晕而眩,口苦而干;面色潮红,耳鸣、腰酸、足跟痛、盗汗,五心烦热,月经量多而红、舌红、脉虚数。方用:黄精12g,白芍13g,覆盆子12g,黄芪11g,山茱萸12g,天门冬12g,贯众炭13g,生地11g,麦冬12g,延胡索13g,益母草12g,血余炭13g,当归12g,五灵脂13g,地骨皮13g,远志13g,香附14g,五味子12g,茯苓13g,仙鹤草12g,炒地榆11g,旱莲草12g,仙茅13g。笔者以上方7剂见效,20剂痊愈。之后月经恢复正常,随访半年未复发。

1.一种治疗肾阴虚型功能失调性子宫出血的中药制剂,其特征在于,所述中药制剂中包括以下重量份数的原料药:黄精10~20份,白芍10~20份,覆盆子10~20份,黄芪10~20份,山茱萸10~20份,天门冬10~20份,贯众炭10~20份,生地10~20份,麦冬10~20份,延胡索10~20份,益母草10~20份,血余炭10~20份,当归10~20份,五灵脂10~20份,地骨皮10~20份,远志10~20份,香附10~20份,五味子10~20份,茯苓10~20份,仙鹤草10~20份,炒地榆10~20份,旱莲草10~20份,仙茅10~20份。
2.根据权利要求1所述治疗肾阴虚型功能失调性子宫出血的中药制剂,其特征在于,所述中药制剂中各种原料的重量份数为:黄精10~15份,白芍10~15份,覆盆子10~15份,黄芪10~15份,山茱萸10~15份,天门冬10~15份,贯众炭10~15份,生地10~15份,麦冬10~15份,延胡索10~15份,益母草10~15份,血余炭10~15份,当归10~20份,五灵脂10~20份,地骨皮10~20份,远志10~20份,香附10~20份,五味子10~20份,茯苓10~20份,仙鹤草10~20份,炒地榆10~20份,旱莲草10~20份,仙茅10~20份。
3.根据权利要求1所述治疗肾阴虚型功能失调性子宫出血的中药制剂,其特征在于,所述中药制剂中各种原料的重量份数为:黄精10~20份,白芍10~20份,覆盆子10~20份,黄芪10~20份,山茱萸10~20份,天门冬10~20份,贯众炭10~20份,生地10~20份,麦冬10~20份,延胡索10~20份,益母草10~20份,血余炭10~20份,当归10~15份,五灵脂10~15份,地骨皮10~15份,远志10~15份,香附10~15份,五味子10~15份,茯苓10~15份,仙鹤草10~15份,炒地榆10~15份,旱莲草10~15份,仙茅10~15份。
4.一种如权利要求1~3中任一项所述治疗肾阴虚型功能失调性子宫出血的中药制剂的制备方法,其特征在于,所述制剂剂型为口服液剂时,制备方法包括:
a、取所有原料放入乙醇浸泡,热提取2次,为组分1;
b、将上述提取的药渣加水提取1次,为组分2;
c、将组分1与组分2合并浓缩;干燥成粉末罐装胶囊。
5.根据权利要求4所述治疗肾阴虚型功能失调性子宫出血的中药制剂的制备方法,其特征在于,所述步骤a中,取所有原料药泡入占其质量10倍量乙醇中,浸泡1-2小时,加热提取2次,每次1-2小时,合并提取液,静置后去上清液,然后放入减压浓缩罐内,减压回收乙醇,浓缩至药液浓度为0.6g生药/mL,抽滤至滤液的相对密度为80℃时1.36的浸膏,得到组分1。
6.根据权利要求4所述治疗肾阴虚型功能失调性子宫出血的中药制剂的制备方法,其特征在于,所述步骤b中,将上述提取过的原料药的药渣加水,加热提取2次,每次1-2小时,合并提取液,静置后去上清液,然后放入减压浓缩罐内,减压,浓缩至药液浓度为0.6g生药/mL,抽滤至滤液的相对密度为80℃时1.36的浸膏,得到组分2。
7.根据权利要求4所述治疗肾阴虚型功能失调性子宫出血的中药制剂的制备方法,其特征在于,所述步骤c中,将上述两种提取液合并,减压浓缩相对密度为80℃时1.35的滤液,回收乙醇,将上述两种提取液置入双效真空浓缩器中,浓缩至90℃时相对密度为1.60的浓缩液,置0~5℃低温冷藏24小时;将冷藏液加0.3%的助滤剂硅藻土,过滤,滤液再置入双效真空浓缩器中,浓缩至每1ml含0.2g生药量,加糊精调和后杀菌装瓶。
8.一种根据权利要求1~4中任一项所述治疗肾阴虚型功能失调性子宫出血的中药制剂的制备方法,其特征在于,所述制剂剂型为胶囊剂的制备方法包括:
a.将所述原料放入粉碎机粉碎后过筛;
b.将过筛后剩下的颗粒物放入乙醇中加热回流提取2次;
c.再取上述乙醇提取后的药渣,加水加热回流提取3次;
d.将上述两种提取液合并,减压回收乙醇并喷雾干燥,得药粉;混合过筛得到的细粉罐装胶囊。
9.根据权利要求8所述治疗肾阳虚型功能失调性子宫出血的中药制剂的制备方法,其特征在于,所述步骤a中,将所述原料放入粉碎机粉碎后过120目细筛,使能通过120目筛的细粉量在达到30%,收集通过120目筛的原料细粉备用;所述步骤b中,将过筛后剩下的颗粒物放入乙醇中加热回流提取2次,每次1~2小时,将2次提取液合并静置。
10.根据权利要求8所述治疗肾阳虚型功能失调性子宫出血的中药制剂的制备方法,其特征在于,所述步骤c中,将上述乙醇提取过的药渣加10倍量水,然后放入提取罐加热煮沸1小时,过滤,滤液备用;滤渣加水,第二次加热,煮沸45分钟,过滤,滤液备用;滤渣再加水,第三次加热煮沸半小时,过滤;将三次滤液合在一起,用纱布过滤,残渣中加入50-60%乙醇,60℃-70℃继续浸提2h,每10min搅拌一次,纱布过滤,合并浸提液,浓缩成糊状;所述步骤d中,将上述提取液合并,减压浓缩相对密度为80℃时1.08的滤液,回收乙醇,将所得滤液调整比重到1.06,通入喷雾干燥机进行喷雾干燥,过喷雾干燥机喷头的压缩空气压力为4kg/m2,干燥室热气流温度是80℃,得原料药粉;将过筛后得到的原料细粉与乙醇提炼药粉混合,罐装胶囊。
